Abstract Background Monitoring hand hygiene compliance (HHC) of healthcare providers (HCPs) in healthcare facilities is critical for hand hygiene (HH) promotion. However, less is known about the cost and effectiveness of different HHC monitoring tools. In this study, we aimed to compare various health economic indicators corresponding to electronic system-based monitoring (ESM) and manual paper-based monitoring (MPM) for HHC to provide evidence-based advice for HHC monitoring measures targeted selecting. Methods A before and after study in 40 clinical departments with 4,524 healthcare providers was conducted from December 2022 to January 2023 (MPM implementation phase) and March 2023 to May 2023 (ESM implementation phase). The cost-effectiveness, cost-efficiency, the extent of the Hawthorne effect, and indirect cost-benefit of the two monitoring methods were compared. Abstract The interplay between crystals and epithelial cells forms the cornerstone of kidney stone development, communication between epithelial cells and macrophages emerging as a pivotal role in this process. We conducted next-generation sequencing on the secreted exosomes of TCMK-1 cells treated with calcium oxalate monohydrate (OX_EXO) or controls (NC_EXO), and on the macrophage cell line RAW264.7 stimulated with OX_EXO or NC_EXO, followed by validation of differentially expressed target proteins and miRNAs through Western blot and PCR. UPSET plots were employed to identify genes co-targeted by exosomal miRNAs. Various bioinformatic analyses were employed to predict potential mechanisms of the dysregulated genes. We integrated sequencing data from the GEO database, and validated findings using clinical patient urine and kidney tissues. We identified 665 differentially expressed exosomal miRNAs between OX_EXO and NC_EXO. Among the top 10 down-regulated miRNAs, the most targeted genes were AAK1 and NUFIP2, whereas PLCB1 was significantly targeted among the top 10 up-regulated miRNAs. In clinical specimens, we confirmed the differential expressions of five homologous miRNAs, as well as CNOT3, CNCNA1C, APEX1, and TMEM199. In conclusion, treatment of TCMK-1 cells with calcium oxalate significantly alerted the expression profile of exosomal miRNAs, subsequently influencing gene expression in macrophages, thereby modulating the processes of kidney stone formation.

To help with the critical nurse staffing shortages in hospitals worldwide, robotic assistants are designed to handle frequently required tasks in the digital operating room (DOR), such as the guidance of the laparoscopic camera. To enable fluent collaboration between robots and clinicians, an intuitive and efficient communication interface is needed to allow for interaction using natural language. However, the demanding requirements of the surgical domain make it challenging to develop suitable solutions. A variety of different vocabulary or phrases may be used for expressing the same command. At the same time, surgical workflows may be highly dynamic - especially in emergency situations - and thus the system must be able to grasp the user’s intent both quickly and with high accuracy. This is especially true as only some clinicians may be authorized to request certain tasks, depending on their rank or field of expertise. To solve these challenges, our proposed communication system uses the fine-tuned deep learning model to recognize the speaker information, and the robot assistant takes action only when it detects the commands from the responsible clinician. Also, our proposed conversational functions enable the finetuned large language models to understand the current natural language command given previous command history. In this work, we present a communication system to recognize the speaking person and understand the intent of conversational commands quickly and accurately.

To establish a rapid detection method for norovirus GII.2 genotype, this study employed reverse transcription recombinase polymerase amplification (RT-RPA) combined with CRISPR/Cas12a and lateral flow strip (RT-RPA-Cas12a-LFS). Here, the genome of norovirus GII.2 genotype was compared to identify highly conserved sequences, facilitating the design of RT-RPA primers and crRNA specific to the conserved regions of norovirus GII.2. Subsequently, the reaction parameters of RT-RPA were optimized and evaluated using agar-gel electrophoresis and LFS. The results indicate that the conserved sequences of norovirus GII.2 were successfully amplified through RT-RPA at 37°C for 25 minutes. Additionally, CRISPR/Cas12a-mediated cleavage detection was achieved through LFS at 37°C within 10 minutes using the amplification products as templates. Including the isothermal amplification reaction time, the total time is 35 minutes. The established RT-RPA-Cas12a-LFS method demonstrated specific detection of norovirus GII.2, yielding negative results for other viral genomes, and exhibited an excellent detection limit of 10 copies/μl. The RT-RPA-Cas12a-LFS method was further compared with qRT-PCR by analyzing 60 food-contaminated samples. The positive conformity rate was 100%, the negative conformity rate was 95.45%, and the overall conformity rate reached 98.33%. This detection method for norovirus GII.2 genotype is cost-effective, highly sensitive, specific, and easy to operate, offering a promising technical solution for field-based detection of the norovirus GII.2 genotype.

Abstract Background Tripartite motif-containing 26 (TRIM26), a member of the TRIM protein family, exerts dual function in several types of cancer. Nevertheless, the precise role of TRIM26 in clear cell renal cell carcinoma (ccRCC) has not been investigated. Methods The expression of TRIM26 in ccRCC tissues and cell lines were examined through the use of public resources and experimental validation. The impacts of TRIM26 on cell proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) process were determined via CCK-8, colony formation, EdU incorporation, wound healing, Transwell invasion, Western blot, and Immunofluorescence assays. RNA-seq followed by bioinformatic analyses were used to identify the downstream pathway of TRIM26. The interaction between TRIM26 and ETK was assessed by co-immunoprecipitation, qRT-PCR, Western blot, cycloheximide (CHX) chase, and in vivo ubiquitination assays. Results We have shown that TRIM26 exhibits a downregulation in both ccRCC tissues and cell lines. Furthermore, this decreased expression of TRIM26 is closely linked to unfavorable overall survival and diseases-free survival outcomes among ccRCC patients. Gain- and loss-of-function experiments demonstrated that increasing the expression of TRIM26 suppressed the proliferation, migration, invasion, and EMT process of ccRCC cells. Conversely, reducing the expression of TRIM26 had the opposite effects. RNA sequencing, coupled with bioinformatic analysis, revealed a significant enrichment of the mTOR signaling pathway in the control group compared to the group with TRIM26 overexpression. This finding was then confirmed by a western blot assay. Subsequent examination revealed that TRMI26 had a direct interaction with ETK, a non-receptor tyrosine kinase. This interaction facilitated the ubiquitination and degradation of ETK, resulting in the deactivation of the AKT/mTOR signaling pathway in ccRCC. ETK overexpression counteracted the inhibitory effects of TRIM26 overexpression on cell proliferation, migration, and invasion. Conclusion Our results have shown a novel mechanism by which TRIM26 hinders the advancement of ccRCC by binding to and destabilizing ETK, thus leading to the deactivation of AKT/mTOR signaling. TRIM26 shows promise as both a therapeutic target and prognostic biomarker for ccRCC patients.

ABSTRACT Background Renal ischaemia‒reperfusion injury (IRI) is the primary cause of acute kidney injury (AKI). To date, effective therapies for delaying renal IRI and postponing patient survival remain absent. Ankyrin repeat domain 1 (ANKRD1) has been implicated in some pathophysiologic processes, but its role in renal IRI has not been explored. Methods The mouse model of IRI‐AKI and in vitro model were utilised to investigate the role of ANKRD1. Immunoprecipitation‐mass spectrometry was performed to identify potential ANKRD1‐interacting proteins. Protein‒protein interactions and protein ubiquitination were examined using immunoprecipitation and proximity ligation assay and immunoblotting, respectively. Cell viability, damage and lipid peroxidation were evaluated using biochemical and cellular techniques. Results First, we unveiled that ANKRD1 were significantly elevated in renal IRI models. Global knockdown of ANKRD1 in all cell types of mouse kidney by recombinant adeno‐associated virus (rAAV9)‐mitigated ischaemia/reperfusion‐induced renal damage and failure. Silencing ANKRD1 enhanced cell viability and alleviated cell damage in human renal proximal tubule cells exposed to hypoxia reoxygenation or hydrogen peroxide, while ANKRD1 overexpression had the opposite effect. Second, we discovered that ANKRD1's detrimental function during renal IRI involves promoting lipid peroxidation and ferroptosis by directly binding to and decreasing levels of acyl‐coenzyme A synthetase long‐chain family member 3 (ACSL3), a key protein in lipid metabolism. Furthermore, attenuating ACSL3 in vivo through pharmaceutical approach and in vitro via RNA interference mitigated the anti‐ferroptotic effect of ANKRD1 knockdown. Finally, we showed ANKRD1 facilitated post‐translational degradation of ACSL3 by modulating E3 ligase tripartite motif containing 25 (TRIM25) to catalyse K63‐linked ubiquitination of ACSL3, thereby amplifying lipid peroxidation and ferroptosis, exacerbating renal injury. Conclusions Our study revealed a previously unknown function of ANKRD1 in renal IRI. By driving ACSL3 ubiquitination and degradation, ANKRD1 aggravates ferroptosis and ultimately exacerbates IRI‐AKI, underlining ANKRD1's potential as a therapeutic target for kidney IRI. Key Points/Highlights Ankyrin repeat domain 1 (ANKRD1) is rapidly activated in renal ischaemia‒reperfusion injury (IRI) models in vivo and in vitro. ANKRD1 knockdown mitigates kidney damage and preserves renal function. Ferroptosis contributes to the deteriorating function of ANKRD1 in renal IRI. ANKRD1 promotes acyl‐coenzyme A synthetase long‐chain family member 3 (ACSL3) degradation via the ubiquitin‒proteasome pathway. The E3 ligase tripartite motif containing 25 (TRIM25) is responsible for ANKRD1‐mediated ubiquitination of ACSL3.

Introduction. The emergence of new scientific and clinical evidence on the use of retrograde intrarenal surgery (RIRS) for the management of kidney stones has prompted periodic systematisation, analysis, and evaluation of outcomes to standardise its application and determine future directions for research and development.Objective. To present the Russian adaptation of a guideline on retrograde intrarenal surgery from the International Urolithiasis Alliance guideline series to provide a theoretical basis for urologists performing RIRS.Materials & Methods. A systematic review was conducted on the RIRS-associated publications available in the PubMed database to prepare a set of recommendations during the period from 1 January 1964 until 1 October 2021. The recommendations were evaluated using the GRADE (Grading of Recommendations, Assessment, Development, and Evaluation) system, which classifies, evaluates, develops, and examines recommendations. The modified Oxford Centre for Evidence-Based Medicine's (OCEBM) system for categorising the level of evidence and relevant comments have been applied to assess the strength of the conclusions.Results. The research team conducted a comprehensive analysis of 36 published clinical guidelines on the following topics: 1. Indications and Contraindications 2. Preoperative Imaging 3. Preoperative Ureteral Stenting 4. Preoperative Medications 5. Perioperative Use of Antibiotics 6. Use of Antithrombotic Therapy 7. Anesthesia Issues 8. Intraoperative Positioning 9. Equipment 10. Complications.Conclusion. A series of recommendations for RIRS, offered here should help provide safe and effective performance of RIRS.

Abstract Background Bladder cancer (BC) is the most common urinary tract malignancy. Aurora kinase B (AURKB), a component of the chromosomal passenger protein complex, affects chromosomal segregation during cell division. Mitotic arrest-deficient 2-like protein 2 (MAD2L2) interacts with various proteins and contributes to genomic integrity. Both AURKB and MAD2L2 are overexpressed in various human cancers and have synergistic oncogenic effects; therefore, they are regarded as emerging therapeutic targets for cancer. However, the relationship between these factors and the mechanisms underlying their oncogenic activity in BC remains largely unknown. The present study aimed to explore the interactions between AURKB and MAD2L2 and how they affect BC progression via the DNA damage response (DDR) pathway. Methods Bioinformatics was used to analyze the expression, prognostic value, and pro-tumoral function of AURKB in patients with BC. CCK-8 assay, colony-forming assay, flow cytometry, SA-β-gal staining, wound healing assay, and transwell chamber experiments were performed to test the viability, cell cycle progression, senescence, and migration and invasion abilities of BC cells in vitro. A nude mouse xenograft assay was performed to test the tumorigenesis ability of BC cells in vivo. The expression and interaction of proteins and the occurrence of the senescence-associated secretory phenotype were detected using western blot analysis, co-immunoprecipitation assay, and RT-qPCR. Results AURKB was highly expressed and associated with prognosis in patients with BC. AURKB expression was positively correlated with MAD2L2 expression. We confirmed that AURKB interacts with, and modulates the expression of, MAD2L2 in BC cells. AURKB knockdown suppressed the proliferation, migration, and invasion abilities of, and cell cycle progression in, BC cells, inducing senescence in these cells. The effects of AURKB knockdown were rescued by MAD2L2 overexpression in vitro and in vivo. The effects of MAD2L2 knockdown were similar to those of AURKB knockdown. Furthermore, p53 ablation rescued the MAD2L2 knockdown-induced suppression of BC cell proliferation and cell cycle arrest and senescence in BC cells. Conclusions AURKB activates MAD2L2 expression to downregulate the p53 DDR pathway, thereby promoting BC progression. Thus, AURKB may serve as a potential molecular marker and a novel anticancer therapeutic target for BC.

Drought stress impairs crop growth and development. BEL1-like family transcription factors may be involved in plant response to drought stress, but little is known of the molecular mechanism by which these proteins regulate plant response and defense to drought stress. Here we show that the BEL1-like transcription factor GhBLH5-A05 functions in cotton (Gossypium hirsutum) response and defense to drought stress. Expression of GhBLH5-A05 in cotton was induced by drought stress. Overexpression of GhBLH5-A05 in both Arabidopsis and cotton increased drought tolerance, whereas silencing GhBLH5-A05 in cotton resulted in elevated sensitivity to drought stress. GhBLH5-A05 binds to cis elements in the promoters of GhRD20-A09 and GhDREB2C-D05 to activate the expression of these genes. GhBLH5-A05 interacted with the KNOX transcription factor GhKNAT6-A03. Co-expression of GhBLH5-A05 and GhKNAT6-A03 increased the transcription of GhRD20-A09 and GhDREB2C-D05. We conclude that GhBLH5-A05 acts as a regulatory factor with GhKNAT6-A03 functioning in cotton response to drought stress by activating the expression of the drought-responsive genes GhRD20-A09 and GhDREB2C-D05.

Increasing distributed generators (DGs) and flexible loads (FLs) enable distribution systems to provide both active and reactive power reserves (P-Q reserves) in supporting the frequency and voltage regulations of transmission systems. However, such requirements at the interface between the transmission system operator (TSO) and distribution system operator (DSO) affect the distribution system operation security, considering the uncertainties of DGs and FLs. To exploit the reserve potential of distribution systems, this paper investigates the voltage-dependent P-Q reserve capacity (V-PQRC) of such types of distribution systems. V-PQRC reflects the feasible space of P-Q reserves that the DSO can provide to the TSO taking the voltage deviation limit at TSO-DSO interface into consideration, while ensuring the distribution system operation security under uncertainties of DGs and FLs. An evaluation method for V-PQRC at the TSO-DSO interface is proposed. To improve the robust performance of the evaluation method, the DG uncertainty is captured by a generalized ambiguity set and the FL uncertainty is addressed by designing a constrained sliding mode controller (CSMC). Three objectives are considered in the evaluation, i.e., P reserve capacity, Q reserve capacity, and the voltage deviation limit at the TSO-DSO interface. Then, a multi-objective optimization model integrating the generalized robust chance-constrained optimization and CSMC (GRCC-CSMC) is established for V-PQRC evaluation to obtain the Pareto optimal reserve schemes. Finally, a non-approximated selecting (NAS) method is proposed to build up a simplified V-PQRC linear model, which can be convenient to apply in the transmission-distribution system coordination. Simulation results reveal that the V-PQRC evaluation method can achieve a good performance of accuracy and robustness against uncertainties.

Abstract Epigenetic regulation is reported to play a significant role in the pathogenesis of various kidney diseases, including renal cell carcinoma, acute kidney injury, renal fibrosis, diabetic nephropathy, and lupus nephritis. However, the role of epigenetic regulation in calcium oxalate (CaOx) crystal deposition-induced kidney injury remains unclear. Our study demonstrated that the upregulation of enhancer of zeste homolog 2 (EZH2)-mediated ferroptosis facilitates CaOx-induced kidney injury. CaOx crystal deposition promoted ferroptosis in vivo and in vitro. Usage of liproxstatin-1 (Lip-1), a ferroptosis inhibitor, mitigated CaOx-induced kidney damage. Single-nucleus RNA-sequencing, RNA-sequencing, immunohistochemical and western blotting analyses revealed that EZH2 was upregulated in kidney stone patients, kidney stone mice, and oxalate-stimulated HK-2 cells. Experiments involving in vivo EZH2 knockout, in vitro EZH2 knockdown, and in vivo GSK-126 (an EZH2 inhibitor) treatment confirmed the protective effects of EZH2 inhibition on kidney injury and ferroptosis. Mechanistically, the results of RNA-sequencing and chromatin immunoprecipitation assays demonstrated that EZH2 regulates ferroptosis by suppressing solute carrier family 7, member 11 (SLC7A11) expression through trimethylation of histone H3 lysine 27 (H3K27me3) modification. Additionally, SOX4 regulated ferroptosis by directly modulating EZH2 expression. Thus, this study demonstrated that SOX4 facilitates ferroptosis in CaOx-induced kidney injury through EZH2/H3K27me3-mediated suppression of SLC7A11. Graphical Abstract

Abstract Prostate cancer (PCa) is one of the most common malignant tumors affecting the male genitourinary system. However, there is currently a lack of effective treatments for patients with advanced prostate cancer, which significantly impacts men’s overall health. Exonuclease 1 (EXO1), a protein with mismatch repair and recombination functions, has been found to play a vital role in various diseases. In our study, we discovered that EXO1 acts as a novel biomarker of PCa, which promotes prostate cancer progression by regulating lipid metabolism reprogramming in prostate cancer cells. Mechanistically, EXO1 promotes the expression of SREBP1 by inhibiting the P53 signaling pathway. In summary, our findings suggest that EXO1 regulated intracellular lipid reprogramming through the P53/SREBP1 axis, thus promoting PCa progression. The result could potentially lead to new insights and therapeutic targets for diagnosing and treating PCa.

Abstract Background Invasion and metastasis are the main causes of unfavourable prognosis in patients diagnosed with bladder cancer. The efficacy of immunotherapy in bladder cancer remains suboptimal due to the presence of an immunosuppressive microenvironment. The novel protein family with sequence similarity 171B (FAM171B) has been identified, but its precise role and mechanism in bladder cancer remain unclear. Methods In this study, we conducted an analysis to investigate the associations between FAM171B expression and the prognosis and clinicopathological stage of bladder cancer. To this end, we utilized RNA sequencing data from the TCGA and GEO databases, as well as tumor tissue specimens obtained from our clinical centre. RNA sequencing analysis allowed us to examine the biological function of FAM171B at the transcriptional level in bladder cancer cells. Additionally, we used immunoprecipitation and mass spectrometry to identify the protein that interacts with FAM171B in bladder cancer cells. The effects of FAM171B on modulating tumor-associated macrophages (TAMs) and vimentin-mediated tumor progression, as well as the underlying mechanisms, were clarified by phalloidin staining, immunofluorescence staining, ELISA, RNA immunoprecipitation, flow cytometry and a bladder cancer graft model. Results FAM171B expression exhibits strong positive correlation with poor survival outcomes and advanced clinicopathological stages in patients with bladder cancer. FAM171B significantly promoted bladder cancer growth and metastasis, accompanied by TAM accumulation in the microenvironment, in vivo and in vitro. Through studies of the molecular mechanism, we found that FAM171B contributes to tumor progression by stabilizing vimentin in the cytoplasm. Additionally, our research revealed that FAM171B enhances the splicing of CCL2 mRNA by interacting with heterogeneous nuclear ribonucleoprotein U (HNRNPU), ultimately leading to increased recruitment and M2 polarization of TAMs. Conclusions In this study, we identified FAM171B as a potent factor that promotes the progression of bladder cancer. These findings establish a solid theoretical foundation for considering FAM171B as a potential diagnostic and therapeutic biomarker for bladder cancer.

Cancers of the urinary system account for 13.1% of new cancer cases and 7.9% of cancer-related deaths. Of them, renal cancer, bladder cancer, and prostate cancer are most prevalent and pose a substantial threat to human health and the quality of life. Prostate cancer is the most common malignant tumor in the male urinary system. It is the second most common type of malignant tumor in men, with lung cancer surpassing its incidence and mortality. Bladder cancer has one of the highest incidences and is sex-related, with men reporting a significantly higher incidence than women. Tumor development in the urinary system is associated with factors, such as smoking, obesity, high blood pressure, diet, occupational exposure, and genetics. The treatment strategies primarily involve surgery, radiation therapy, and chemotherapy. Cholesterol metabolism is a crucial physiological process associated with developing and progressing urinary system tumors. High cholesterol levels are closely associated with tumor occurrence, invasion, and metastasis. This warrants thoroughly investigating the role of cholesterol metabolism in urinary system tumors and identifying novel treatment methods for the prevention, early diagnosis, targeted treatment, and drug resistance of urinary system tumors.

Optical Fabry–Perot cavity with a movable mirror is a paradigmatic optomechanical system. While usually the mirror is supported by a mechanical spring, it has been shown that it is possible to keep one of the mirrors in a stable equilibrium purely by optical levitation without any mechanical support. In this work, we expand previous studies of the nonlinear dynamics of such a system by demonstrating a possibility for mechanical parametric instability and the emergence of the “phonon laser” phenomenon.

Abstract Background The nuclear distribution E homologue 1 (NDE1) is a crucial dynein binding partner. The NDE1 protein has the potential to disrupt the normal functioning of centrosomes, leading to a compromised ability to generate spindles and ensure precise separation of chromosomes during cell division. The potential consequences of this phenomenon include genomic instability, malignant transformation and the proliferation of neoplastic growths. However, studies examining the connection between NDE1 and cancer is still very rare. Methods The expression level, prognostic impact, gene change, DNA methylation, protein interaction, mRNA m6A modification, ceRNA network, associated gene and function enrichment, and immune‐related effects of NDE1 in pan‐cancer were examined using a range of online analytic tools and the R software package. The CCK‐8 test, transwell assay, scratch assay and colony formation assay were used to confirm the effects of NDE1 on the proliferation, invasion and metastasis of bladder cancer cells. Results Numerous tumour types have elevated NDE1, which is linked to a bad prognosis. NDE1 is an excellent diagnostic tool for many different types of cancer. Numerous malignancies have been linked to genetic changes in NDE1. NDE1 was connected to TMB, MSI, several immunological checkpoint genes and immune cell infiltration. NDE1 is linked to a number of immunological subtypes. NDE1 could affect how well immunotherapy works to treat different types of cancer. NDE1 was mostly associated with cell cycle, chromosomal segregation, DNA replication and mitotic segregation, according to GO and KEGG analyses. NDE1 physically binds to PAFAH1B1 and DCTN1, respectively. The proliferation, invasion and metastasis of bladder cancer cells may be prevented by NDE1 knockdown. Furthermore, knockdown of NDE1 promoted the apoptosis of bladder cancer cells. Conclusion High expression of NDE1 is present in a variety of tumours, which is linked to a bad prognosis for cancer. Knockdown of NDE1 inhibited the proliferation, invasion and metastasis of bladder cancer cells, and promoted the apoptosis. For a number of malignancies, NDE1 may be a biomarker for immunotherapy and prognosis.

The increasing availability of building operational data has greatly encouraged the development of advanced data-driven technologies for smart building operations. Building operational data typically suffer from data quality problems, such as insufficient labeled and imbalanced data, making them incompatible with conventional machine learning algorithms. Recent advances in data science have provided novel machine learning paradigms to tackle such data challenges for practical applications, such as transfer learning, semi-supervised learning, and generative learning. This review aims to present the progress and perspectives on the effective utilization of novel machine learning paradigms for three major building energy management tasks, i.e., building energy predictions, fault detection and diagnosis, and control optimizations. In-depth discussions have been provided to demonstrate the pros and cons of different learning approaches in terms of data compatibility, modeling difficulties, and possible application scenarios, which could be helpful for enhancing the feasibility of data-driven technologies for smart building operations.

Abstract Objective Herein, we aimed at exploring the FAP expression in clear cell renal cell carcinoma (ccRCC) along with its clinical implication. Methods Using computational tools analysis of different freely accessible gene databases, the expression pattern, clinical importance, co-expressed genes, and signaling pathways of FAP in ccRCC were thoroughly investigated. FAP expression was examined in clinical ccRCC specimens through qRT-PCR, western blotting and immunohistochemistry. Furthermore, in vitro and in vivo experiments were carried out using flow cytometry, CCK-8, wound-healing and Transwell assays, as well as xenograft tumor model, respectively. Results FAP levels were found to be significantly elevated in ccRCC based on bioinformatic data from public databases. Patients who exhibited higher expression levels of FAP had poorer prognoses, according to Kaplan–Meier analysis of survival data. In addition, diagnostic and prognostic value of FAP in ccRCC was figured out by ROC curve and prognostic nomogram model. In vitro study revealed that the over-expression FAP accelerated cell proliferation, migration as well as invasion, and suppressed cell apoptosis, but silencing of FAP had the opposite effect. FAP suppression reduced the PI3K/AKT/mTOR pathway's stimulation, whereas FAP up-regulation increased the stimulation of the pathway. Blocking the PI3K/AKT/mTOR signaling pathway with the dual PI3K/mTOR inhibitor BEZ235 repressesed cancer-promoting effect of FAP. Additionally, we found that the downregulation of FAP was effective at slowing tumor progression in vivo. Conclusion It is possible that FAP could be a reliable biomarker for the diagnosis and prognosis of ccRCC because of its role in the ccRCC progression via triggering the PI3K/AKT/mTOR signaling pathway.

Abstract Bladder cancer (BCa) is one of the most common malignancies worldwide. However, the lack of accurate and effective targeted drugs has become a major problem in current clinical treatment of BCa. Studies have demonstrated that squalene epoxidase (SQLE), as a key rate-limiting enzyme in cholesterol biosynthesis, is involved in cancer development. In this study, our analysis of The Cancer Genome Atlas, The Genotype-Tissue Expression, and Gene Expression Omnibus databases showed that SQLE expression was significantly higher in cancer tissues than it was in adjacent normal tissues, and BCa tissues with a high SQLE expression displayed a poor prognosis. We then confirmed this result in qRT-PCR and immunohistochemical staining experiments, and our vitro studies demonstrated that SQLE knockdown inhibited tumor cell proliferation and metastasis through the PTEN/AKT/GSK3β signaling pathway. By means of rescue experiments, we proved that that P53 is a key molecule in SQLE-mediated regulation of the PTEN/AKT/GSK3β signaling pathway. Simultaneously, we verified the above findings through a tumorigenesis experiment in nude mice. In conclusion, our study shows that SQLE promotes BCa growth through the P53/PTEN/AKT/GSK3β axis, which may serve as a therapeutic biological target for BCa.

Abstract Influence maximization, whose aim is to maximise the expected number of influenced nodes by selecting a seed set of k influential nodes from a social network, has many applications such as goods advertising and rumour suppression. Among the existing influence maximization methods, the community‐based ones can achieve a good balance between effectiveness and efficiency. However, this kind of algorithm usually utilise the network community structures by viewing each node as a non‐overlapping node. In fact, many nodes in social networks are overlapping ones, which play more important role in influence spreading. To this end, an overlapping community‐based particle swarm optimization algorithm named OCPSO for influence maximization in social networks, which can make full use of overlapping nodes, non‐overlapping nodes, and their interactive information is proposed. Specifically, an overlapping community detection algorithm is used to obtain the information of overlapping community structures, based on which three novel evolutionary strategies, such as initialisation, mutation, and local search are designed in OCPSO for better finding influential nodes. Experimental results in terms of influence spread and running time on nine real‐world social networks demonstrate that the proposed OCPSO is competitive and promising comparing to several state‐of‐the‐arts (e.g. CGA, CMA‐IM, CIM, CDH‐SHRINK, CNCG, and CFIN).

Abstract The study aimed to assess the biocompatibility and efficacy of a prostatic urethral lift (PUL) for benign prostatic hyperplasia (BPH). Human BPH-1 cells were co-cultured with implant anchors and sutures, and cytotoxicity was measured. Scanning electron microscopy (SEM) was used to observe adhesion and growth of cells and to evaluate implant biocompatibility. Fifteen male beagle dogs were randomly assigned to the surgical (n = 9) or sham-operated (n = 6) groups. The surgical group underwent cystotomy, and PUL was used to insert two implants in each lobe of the prostate to compress the enlarged prostate and dilate the urethra; the sham group underwent cystotomy without implant insertion. Compared with the control group, no significant difference in cell viability among the groups with different co-culture times of implant anchors and sutures (P > 0.05) was observed. SEM revealed good adhesion and growth of prostate cells on the implants. Improvements in urine flow rates remained stable at 7, 28, and 180 days after surgery, and the urethral diameter in the prostate region was significantly increased compared with that before surgery. PUL is a biocompatible and effective treatment for BPH, improving the urine flow rate without causing inflammation, tissue damage, or cytotoxic effects. Here, the basis for further PUL application was provided.

Abstract Background Gremlin-1 (GREM1) is a protein closely related to tumor growth, although its function in bladder cancer (BCa) is currently unknown. Our first objective was to study the GREM1 treatment potential in BCa. Methods BCa tissue samples were collected for the detection of GREM1 expression using Western blot analysis and Immunofluorescence staining. Association of GREM1 expression with clinicopathology and prognosis as detected by TCGA (The Cancer Genome Atlas) database. The functional investigation was tested by qRT-PCR, western blot analysis, CCK-8, cell apoptosis, wound healing, and transwell assays. The interaction between GREM1 and the downstream PI3K/AKT signaling pathway was assessed by Western blot analysis. Results GREM1 exhibited high expression in BCa tissues and was linked to poor prognosis. Stable knockdown of GREM1 significantly inhibited BCa cell (T24 and 5637) proliferation, apoptosis, migratory, invasive, as well as epithelial-mesenchymal transition (EMT) abilities. GREM1 promotes the progression in BCa via PI3K/AKT signaling pathway. Conclusion Findings demonstrate that the progression-promoting effect of GREM1 in BCa, providing a novel biomarker for BCa-targeted therapy.

Abstract Offshore wind farm (OWF) is considered as a perfect zero‐carbon energy source for the future power system. However, the growing offshore distance and water depth of OWF make the OWF HVDC transmission technique a more promising solution than HVAC due to higher cost‐efficiency and reliability. In this paper, the current situation of OWF‐HVDC projects is introduced at first. Then, novel converter topologies with the higher power density and cost‐efficiency are presented, including the hybrid modular multilevel converter (MMC), alternative arm converter (AAC), and diode rectifier (DR). Next, several OWF HVDC transmission system topologies are introduced, including terminal‐hybrid, station‐hybrid and all‐DC delivered system. Furthermore, the key technologies for OWF HVDC operation and control are summarized, including grid‐forming control strategy for offshore wind turbines, stability analysis method, corresponding stability enhancement measures and frequency support control strategies. Additionally, the fault ride‐through and protection strategies for different fault locations have been presented. Finally, the main conclusions and prospects for OWF HVDC are summarized.

Chronic kidney disease (CKD) is characterized by persistent kidney dysfunction, ultimately resulting in end-stage renal disease (ESRD). Renal fibrosis is a crucial pathological feature of CKD and ESRD. However, there is no effective treatment for this condition. Despite the complex molecular mechanisms involved in renal fibrosis, increasing evidence highlights the crucial role of histone modification in its regulation. The reversibility of histone modifications offers promising avenues for therapeutic strategies to block or reverse renal fibrosis. Therefore, a comprehensive understanding of the regulatory implications of histone modifications in fibrosis may provide novel insights into more effective and safer therapeutic approaches. This review highlights the regulatory mechanisms and recent advances in histone modifications in renal fibrosis, particularly histone methylation and histone acetylation. The aim is to explore the potential of histone modifications as targets for treating renal fibrosis.

To qualify the risk of extreme weather events for power supply security during the long-term power system transformation process, this paper proposes a risk probability evaluation method based on probabilistic production simulation. Firstly, the internal relationship of extreme weather intensity and duration is depicted using the copula function, and the influences of extreme weather on power security are described using the guaranteed power output ability coefficient, which can provide the extreme scenario basis for probabilistic production simulation. Then, a probabilistic production simulation method is proposed, which includes a typical-year scenario and extreme weather events. Meanwhile, an index system is proposed to qualify the power security level, which applies the loss of load expectation (LOLE) and time of loss of load expectation (TOLE) under different scenarios and other indices to reveal the long-term power security trend. Finally, the long-term power supply risks for the Yunnan provincial power system are analyzed using the proposed method, validating that the proposed method is capable of characterizing the influences of extreme weather on power security. The security level of different long-term power transformation schemes is evaluated.

The response of plants to abiotic stress is intricately mediated by PYR/PYL/RCARs, key components within the ABA signal transduction pathway. Despite the widespread identification of PYL genes across diverse plant species, the evolutionary history and structural characteristics of these genes within the pomegranate (Punica granatum L.) remained unexplored. In this study, we uncovered, for the first time, 12 PgPYLs from the whole genome dataset of ‘Tunisia’, mapping them onto five chromosomes and categorizing them into three distinct subgroups (Group I, Group II, and Group III) through phylogenetic analysis. Detailed examination of the composition of these genes revealed similar conserved motifs and exon–intron structures among genes within the same subgroup. Fragment duplication emerged as the primary mechanism driving the amplification of the PYL gene family, as evidenced by intra-species collinearity analysis. Furthermore, inter-species collinearity analysis provided insights into potential evolutionary relationships among the identified PgPYL genes. Cis-acting element analysis revealed a rich repertoire of stress and hormone response elements within the promoter region of PgPYLs, emphasizing their putative roles in diverse signaling pathways. Upon treatment with 100 μmol/L ABA, we investigated the expression patterns of the PgPYL gene family, and the qRT-PCR data indicated a significant up-regulation in the majority of PYL genes. This suggested an active involvement of PgPYL genes in the plant’s response to exogenous ABA. Among them, PgPYL1 was chosen as a candidate gene to explore the function of the gene family, and the CDS sequence of PgPYL1 was cloned from pomegranate leaves with a full length of 657 bp, encoding 218 amino acids. Tobacco transient expression analysis demonstrated a consistent trend in the expression levels of pBI121-PgPYL1 and the related genes of the ABA signaling pathway, both of which increased initially before declining. This study not only contributes to the elucidation of the genomic and structural attributes of PgPYL genes but also provides a foundation for understanding their potential functions in stress responses. The identified conserved motifs, evolutionary relationships, and expression patterns under ABA treatment pave the way for further research into the PgPYL gene family’s role in pomegranate biology, offering valuable insights for future studies on genetic improvement and stress resilience in pomegranate cultivation.

Abstract Ionised gas, i.e., plasma, is a medium where electrons-ions dynamics are electrically and magnetically altered. Electric and magnetic fields can modify plasma’s optical loss, refraction, and gain. Still, plasma’s low pressure and large electrical fields have presented as challenges to introducing it to micro-cavities. Here we demonstrate optical microresonators, with walls thinner than an optical wavelength, that contain plasma inside them. By having an optical mode partially overlapping with plasma, we demonstrate resonantly enhanced light-plasma interactions. In detail, we measure plasma refraction going below one and plasma absorption that turns the resonator transparent. Furthermore, we photograph the plasma’s micro-striations, with 35 μm wavelength, indicating magnetic fields interacting with plasma. The synergy between micro-photonics and plasma might transform micro-cavities, and electro-optical interconnects by adding additional knobs for electro-optically controlling light using currents, electric-, and magnetic-fields. Plasma might impact microphotonics by enabling new types of microlasers and electro-optical devices.

Abstract Background Mounting evidence indicates that the gut microbiome (GMB) plays an essential role in kidney stone (KS) formation. In this study, we conducted a systematic review and meta-analysis to compare the composition of gut microbiota in kidney stone patients and healthy individuals, and further understand the role of gut microbiota in nephrolithiasis. Results Six databases were searched to find taxonomy-based comparison studies on the GMB until September 2022. Meta-analyses were performed using RevMan 5.3 to estimate the overall relative abundance of gut microbiota in KS patients and healthy subjects. Eight studies were included with 356 nephrolithiasis patients and 347 healthy subjects. The meta-analysis suggested that KS patients had a higher abundance of Bacteroides (35.11% vs 21.25%, Z = 3.56, P = 0.0004) and Escherichia_Shigella (4.39% vs 1.78%, Z = 3.23, P = 0.001), and a lower abundance of Prevotella_9 (8.41% vs 10.65%, Z = 4.49, P

ObjectivesThis research aims to assess the effectiveness and cost-effectiveness of pre-pregnancy deafness screening policies.MethodsMarried couples from Shanghai, Beijing, and Suzhou in China were enrolled. We conducted high-throughput, pre-pregnancy genetic screenings for deafness in women and their partners. We compared the cost-effectiveness of deafness genetic screening with the status quo. The two-step screening (wife then partner) and following treatments and interventions were included in the decision tree model. We conducted a cost-effectiveness analysis based on the decrease in deaf newborns, healthy newborn births, and cost-utility analysis of pre-pregnancy deafness genetic screening separately. Cost, utility, and probability data used in the three models were collected from a survey combined with literature and expert consultants. A 5% discount rate and a series of one-way sensitivity analyses along with a Monte Carlo simulation were used to test the reliability of this research.ResultsBetween Jan 1, 2019, and Dec 31, 2021, we recruited 6,200 females and 540 male spouses from community health service centers in Shanghai, Beijing, and Suzhou. The incremental cost-effectiveness ratio (ICER) for reducing deaf newborn births was USD 32,656 per case and USD 1,203,926 per case for increasing one healthy newborn birth. This gap exists because of the overall decrease of newborn births. From the perspective of the whole society, deafness genetic screening is not cost-effective for reducing the overall quality-adjusted life years (QALY) in the population.DiscussionPre-pregnancy genetic testing is effective in decreasing the occurrence of congenital deafness. It is a cost-saving measure when compared with the costs of future medical expenditure and income loss for the affected families. However, such screening and preventive avoidance of pregnancy will decrease the population size and QALY. Only post-screening ART with PGT was shown to increase the birth of healthy newborns. Focusing on key groups such as premature births or consanguineous couples may improve the societal effects of screening.

The low hilly area is a major landform in the west of Henan province, China, and it is suffering soil and water loss because of human activities. The silvopastoral system that combines trees and grasses has been widely used to restore this fragile area. We conducted in situ field experiments in 2011 in the low hilly area of Henan province involving pure forests of Populus simonii (PS; Salicaceae), Platycladus orientalis (PO; Cupressaceae), Quercusvariabilis (QV; Fagaceae), and Robinia pseudoacacia (RP; Fabaceae), and also with each forest tree species being combined with Medicago sativa (MS; Fabaceae) as silvopastoral systems, i.e., PS-MS, PO-MS, QV-MS, and RP-MS, respectively. We recorded tree diameter at breast height (DBH) and tree height (TH) in the years 2014–2016, 2018, 2020, and 2022 for all the different vegetation types. Tree biomass load (TBL, i.e., tree biomass per unit area) was estimated based on DBH and TH using allometric equations for each tree species. Generally, the results showed that the DBH, TH, and TBL were promoted in the silvopastoral systems PS-MS, PO-MS, and QV-MS. Specially, the DBH and TBL of PS-MS and PO-MS tended to be higher and were significantly higher than the pure forests of PS and MS, respectively, in 2014–2016; the silvopastoral systems were not significantly different from the pure forests for DBH in 2018 and 2022, and for TBL in 2018 and 2020. TH was lower in PS-MS and PO-MS than that in PS and PO in 2014–2016, while there was no difference of TH between PS-MS and PS in 2018 and 2022, and TH was higher in PO-MS than that in PO in 2018, 2020 and 2022. The DBH, TH, and TBL were all higher in QV-MS than QV. The TBL in RP-MS was non-significant or lower compared to that in RP. Moreover, TBL tended to be lower in upslope positions than downslope in the pure forests, while there was no difference in most silvopastoral systems. In summary, PS and PO may be suggested as the tree species selection in vegetation restoration processes for about five years, QV may be a better alternative for a long term, while RP is not recommended.

How to simultaneously improve the ionic conductivity and mechanical properties is a key problem facing currently used anion-exchange membranes (AEMs). Here, biomass-based bacterial cellulose (BC) was used as a porous template to make TiO2 localized mineralization around the surface of BC nanofibers, and constructed a TiO2-coated BC porous substrate (TiO2@BC) with hierarchical structure. Then, the coated TiO2 nanoparticles was densely grafted by quaternary ammonium groups to obtain high ionic conduction ability. After filling with a polymeric ionic liquid (PIL) with high ion exchange capacity through in situ polymerization and crosslinking, the obtained novel PIL-filled AEM possessed ultrahigh ionic conductivity of 100.5 mS cm−1 at 80 °C, which was 72.1% higher than that of the PIL-filled pure BC membrane (only 58.4 mS cm−1). Moreover, by the aid of the synergistic reinforcement effect of TiO2@BC, the membrane exhibited extremely high dry strength of 95.3 MPa and satisfactory wet strength and flexibility. When at fully hydrate state, the membrane with the size of 1 × 4 cm (width × length) can hang a bottle containing 1000 g of water. The single cell equipped with this membrane output the peak power density of 40.2 mW cm−2, showing its great potential as a high-performance biomass-based AEM.

As the filling material for the new nuclear fuel transport cask, the rigid polyurethane foam plays an important role in the safe transportation of the nuclear fuel assemblies, and therefore its reliability deserves special attention. Thermal analysis and long-term ageing test are commonly used to predict the service lifetime of rigid polyurethane foams, but they cannot fully consider the influences of mechanical loads like vibration, collision, and impact etc., which is not satisfactory for the transportation application. Herein, in addition to the conventional thermal analysis and long-term ageing methods, the random vibration testing was particularly conducted on the rigid polyurethane foam that has been used in the new nuclear fuel transport cask for the nuclear power plants in China, and its service lifetime was innovatively predicted by the intrinsic property of material, the resonance frequency. The result which can be easily obtained from the acceleration data through fast Fourier transform without additional test and treatment was in the same order of magnitude with that from the conventional methods, and is more rational and convenient, deserving recommended for the rigid polyurethane foams that are used in other applications involving mechanical loads.

Calcium oxalate (CaOx) stones are among the most common types of kidney stones and are associated with renal tubular damage, interstitial fibrosis, and chronic kidney disease. The mechanism of CaOx crystal-induced renal fibrosis remains unknown. Ferroptosis, a type of regulated cell death, is characterised by iron-dependent lipid peroxidation, and the tumour suppressor p53 is a key regulator of ferroptosis. In the present study, our results demonstrated that ferroptosis was significantly activated in patients with nephrolithiasis and hyperoxaluric mice as well as verified the protective effects of ferroptosis inhibition on CaOx crystal-induced renal fibrosis. Moreover, the single-cell sequencing database, RNA-sequencing, and western blot analysis revealed that the expression of p53 was increased in patients with chronic kidney disease and the oxalate-stimulated human renal tubular epithelial cell line, HK-2. Additionally, the acetylation of p53 was enhanced by oxalate stimulation in HK-2 cells. Mechanistically, we found that the induction of p53 deacetylation, owing to either the SRT1720-induced activation of deacetylase sirtuin 1 or the triple mutation of p53, inhibited ferroptosis and alleviated renal fibrosis caused by CaOx crystals. We conclude that ferroptosis is one of the critical mechanisms contributing to CaOx crystal-induced renal fibrosis, and the pharmacological induction of ferroptosis via sirtuin 1-mediated p53 deacetylation may be a potential target for preventing renal fibrosis in patients with nephrolithiasis.

Abstract How to allocate the existing medical resources reasonably, alleviate hospital congestion and improve the patient experience are problems faced by all hospitals. At present, the combination of artificial intelligence and the medical field is mainly in the field of disease diagnosis, but lacks successful application in medical management. We distinguish each area of the emergency department by the division of medical links. In the spatial dimension, in this study, the waitlist number in real-time is got by processing videos using image recognition via a convolutional neural network. The congestion rate based on psychology and architecture is defined for measuring crowdedness. In the time dimension, diagnosis time and time-consuming after diagnosis are calculated from visit records. Factors related to congestion are analyzed. A total of 4717 visit records from the emergency department and 1130 videos from five areas are collected in the study. Of these, the waiting list of the pediatric waiting area is the largest, including 10,436 (person-time) people, and its average congestion rate is 2.75, which is the highest in all areas. The utilization rate of pharmacy is low, with an average of only 3.8 people using it at the one time. Its average congestion rate is only 0.16, and there is obvious space waste. It has been found that the length of diagnosis time and the length of time after diagnosis are related to age, the number of diagnoses and disease type. The most common disease type comes from respiratory problems, accounting for 54.3%. This emergency department has congestion and waste of medical resources. People can use artificial intelligence to investigate the congestion in hospitals effectively. Using artificial intelligence methods and traditional statistics methods can lead to better research on healthcare resource allocation issues in hospitals.

The external windshield is a key component in high-speed trains for drag reduction. However, the installation gap between external windshields, which is intended for improving curve-passing ability, could lead to strong aerodynamic instability in train-end area and therefore significant operating risk for train. This study aims to advance this field based on detailed numerical simulation, investigating the related flow features in this area, particularly from a time-dependent perspective. Flow field around external windshields with different installation gaps (10, 20 and 30 mm) is resolved based on Improved Delayed Detached Eddy Simulation (IDDES). The aerodynamic forces on different windshield components, turbulent flow fields around the train-end area, and corresponding low-dimensional modes are analyzed. The results show that the difference in the external windshield gap can significantly affect the related aerodynamic forces, with the 20 mm case having the highest time-averaged values. The aerodynamic spectrum shows primary, secondary and tertiary peak frequencies with a relatively high power spectral density (PSD). Decreasing the gap distance generally increases the overall PSD in the Strouhal number ranges considered, particularly for the secondary and tertiary peaks. The load forms and their energy distributions under different external windshield gaps are obtained through the proper orthogonal decomposition analysis.

The battery energy storage (BES) is recognized as a key resource for the power fluctuations smoothing, peak load shaving and frequency regulation, and its performance depends heavily on the available capacity. It is meaningful to investigate the influence of the BES capacity on the power system operation cost. This paper focuses on the marginal utility of BES capacity in the day-ahead power system operation cost. Firstly, the day-ahead economic dispatch problem is formulated as a constrained quadratic programming problem, with the consideration of the BES degradation cost. Then, the relationship between the BES-related shadow prices and the marginal electricity price is derived from the KKT conditions of the dispatch problem. Based on it, the marginal utility of BES capacity is modeled to evaluate the effect of BES capacity on the system day-ahead operation cost. Finally, the numerical results demonstrate that the proposed marginal utility can manifest the influence of BSS capacity on the power system operation.

With the growth of wind power and solar power, power source clean transformation has become a feasible way for achieving the carbon peak target. As a hydropower resource-abundant province, Guizhou has already built Wujiang river cascade hydropower plants. This paper introduces the basic information of the Guizhou provincial power system and geometrical renewable resource distribution characteristics, especially the power source structure. Then, the wind and solar power complementary characteristics are analyzed by different copula conjunction functions. Then, the power structure transformation and generation expansion plan are provided with both carbon peak restriction and curtailment restrictions. At last, the future Guizhou provincial power system in 2029 operation simulation cases are provided, which can reduce the thermal power generation ratio to 42% and increase wind and solar power generation ratio to 8.5% and 13% approximately. The conclusion indicates that coal-fired power plant flexible reform and larger capacity energy storage installation effectively solve the larger power fluctuation and peak regulation demand with more renewable energy integration target.

The series–parallel DC wind farm composed of a large number of DC wind turbine (DCWT) is a potential scheme for offshore wind power collection with lower construction costs and power losses. However, enhanced coupling issues, such as energy curtailment and fault isolation, exist in the DC wind farm. To overcome the difficulties, a series–parallel DC collection system is proposed, where DCWTs based on the single active bridge (SAB) are connected in series and then are connected in parallel via DC/DC converters. The coupling mechanism among series–parallel strings is analyzed first, and a coordinate control strategy for the whole system is proposed based on the characteristics of SAB. DC fault located at different strings is analyzed and strategies are proposed to protect the whole system. Simulation results in PSCAD/EMTDC verifies the proposed control strategy with the less coupling among series–parallel DC wind farms under unequal wind speeds and fault.

Fast generation of reliable primary radiation damage plays a key role in modeling of long-term defect evolution in materials under irradiation within the multi-scale modeling framework. We have proposed a model, PradG, to fast generate reliable primary radiation damage by sampling limited molecular dynamics databases. First, MD cascade simulations for body-centered-cubic W are systematically carried out with a wide range of discrete cascade energies. Through introducing the spherical symmetry approximation of cascade volume and analyzing cascade configurations, we find that the size distribution shows a piecewise power-law formula, and the radial distribution shows the Gaussian and exponential decay types for interstitial and vacancy clusters, respectively. Second, by using a sampling method based on these fitting size and spatial distribution, regardless of the spatial correlation of interstitial and vacancy clusters, PradG is developed for generating primary defects of arbitrary cascade energies. Third, the reliability and validation of PradG are verified by comparing with cascade annealing simulations and defect evolution in fission neutron irradiated W. It provides a reliable damage source term for multi-scale modeling of long-term defect evolution with a certain PKA energy spectrum.

The sensory cortex is characterized by general organizational principles such as topography and hierarchy. However, measured brain activity given identical input exhibits substantially different patterns across individuals. Although anatomical and functional alignment methods have been proposed in functional magnetic resonance imaging (fMRI) studies, it remains unclear whether and how hierarchical and fine-grained representations can be converted between individuals while preserving the encoded perceptual content. In this study, we trained a method of functional alignment called neural code converter that predicts a target subject’s brain activity pattern from a source subject given the same stimulus, and analyzed the converted patterns by decoding hierarchical visual features and reconstructing perceived images. The converters were trained on fMRI responses to identical sets of natural images presented to pairs of individuals, using the voxels on the visual cortex that covers from V1 through the ventral object areas without explicit labels of the visual areas. We decoded the converted brain activity patterns into the hierarchical visual features of a deep neural network using decoders pre-trained on the target subject and then reconstructed images via the decoded features. Without explicit information about the visual cortical hierarchy, the converters automatically learned the correspondence between visual areas of the same levels. Deep neural network feature decoding at each layer showed higher decoding accuracies from corresponding levels of visual areas, indicating that hierarchical representations were preserved after conversion. The visual images were reconstructed with recognizable silhouettes of objects even with relatively small numbers of data for converter training. The decoders trained on pooled data from multiple individuals through conversions led to a slight improvement over those trained on a single individual. These results demonstrate that the hierarchical and fine-grained representation can be converted by functional alignment, while preserving sufficient visual information to enable inter-individual visual image reconstruction.

Bolted connections have become the most widely used connection method in steel structures. Over the long-term service of the bolts, loosening damage and other defects will inevitably occur due to various factors. To ensure the stability of bolted connections, an efficient and precise method for identifying loosened bolts in a given structure is proposed based on computer vision technology. The main idea of this method is to combine deep learning with image processing techniques to recognize and label the loosening angle from bolt connection images. A rectangular steel plate was taken as the test research object, and three grade 4.8 ordinary bolts were selected for study. The analysis was conducted under two conditions: manual loosening and simulated loosening. The results showed that the method proposed in this article could accurately locate the position of the bolts and identify the loosening angle, with an error value of about ±0.1°, which proves the accuracy and feasibility of this method, meeting the needs of structural health monitoring.

Context: Several recent randomized controlled trials (RCTs) have reported on the survival benefits of poly (ADP-ribose) polymerase inhibitors (PARPi) compared to standard-of-care (SOC) treatment (enzalutamide, abiraterone, or docetaxel) in patients with metastatic castration-resistant prostate cancer (mCRPC). However, there is a limited integrated analysis of high-quality evidence comparing the efficacy and safety of PARPi and SOC treatments in this context. Objective: This study aims to comprehensively analyze the survival benefits and adverse events associated with PARPi and SOC treatments through a head-to-head meta-analysis in mCRPC. Evidence acquisition: A systematic review search was conducted in PubMed, Embase, Clinical trials, and the Central Cochrane Registry in July 2023. RCTs were assessed following the Preferred Reporting Items for Systematic Reviews and Meta-analyses (PRISMA) guidelines. The systematic review was prospectively registered on PROSPERO (CRD42023441034). Evidence synthesis: A total of 8 studies, encompassing 2341 cases in the PARPi treatment arm and 1810 cases in the controlled arm, were included in the qualitative synthesis. The hazard ratio (HR) for radiographic progression-free survival (rPFS) and overall survival (OS) were 0.74 (95% CI, 0.61–0.90) and 0.89 (95% CI, 0.80–0.99), respectively, in the intention-to-treatment patients. For subgroup analysis, HRs for rPFS and OS in the BRCA-mutated subgroup were 0.39 (95% CI, 0.28–0.55) and 0.62 (95% CI, 0.38–0.99), while in the HRR-mutated subgroup, HR for rPFS was 0.57 (95% CI, 0.48–0.69) and for OS was 0.77 (95% CI, 0.64–0.93). The odds ratio (OR) for all grades of adverse events (AEs) and AEs with severity of at least grade 3 were 3.86 (95% CI, 2.53–5.90) and 2.30 (95% CI, 1.63–3.26), respectively. Conclusions: PARP inhibitors demonstrate greater effectiveness than SOC treatments in HRR/BRCA-positive patients with mCRPC. Further research is required to explore ways to reduce adverse event rates and investigate the efficacy of HRR/BRCA-negative patients.

Epidemiological studies have suggested a potential relationship between the transforming growth factor-β1 (TGF-β1) gene and ischemic stroke (IS) risk; however, the current results are inconsistent. Therefore, we performed this meta-analysis to assess the precise association between TGF-β1 polymorphisms and IS risk. Online databases were searched for themes related to TGF-β1 polymorphisms and ARE risk. Quantitative calculations of odds ratios (ORs) and confidence intervals (CIs) were performed using 5 genetic models of each variant locus. Heterogeneity tests, cumulative analyses, sensitivity analyses, and publication bias were conducted to examine statistical power. Moreover, changes in the secondary structure and minimum free energy (MFE) were explored using in silico analysis. Nineteen case-control studies were included in our meta-analysis on rs1800468 G>A, rs1800469 C>T, and rs1800470 T>C polymorphisms and IS risk. Overall, only a marginal association was found between the rs1800469 C>T polymorphism and IS risk (T vs C: OR = 1.12, 95%CI = 1.00-1.46, P = .05, I 2 = 77.0%). Otherwise, no significant association was observed between the rs1800468 G>A and rs1800470 T>C polymorphisms and IS risk in general and stratified analyses. Moreover, no significant changes in secondary structure and MFE were found in any of the 3 polymorphic loci. Current evidence cautiously suggests that TGF-β1 polymorphisms are not associated with IS susceptibility.

The increased activity of the mTOR pathway in bladder cancer has been extensively studied, but no satisfactory mTOR inhibitor has been found in bladder cancer. The role of AZD8055, a second-generation mTOR inhibitor, has not been reported in bladder cancer. Herein, we investigated the effects of AZD8055 on bladder cells and their interaction with macrophages in vivo and in vitro. In four bladder cancer cell lines, the phosphorylation of mTOR, AKT and S6K1 was suppressed by AZD8055. AZD8055 inhibited proliferation and induced G1 cell-cycle arrest and apoptosis of bladder cancer cells in a concentration-dependent manner. AZD8055 also inhibits the migration and invasion of bladder cancer cells by blocking EMT and MMP9. In addition, AZD8055 inhibited chemotaxis and M2 phenotype of macrophage after co-culture with bladder cancer cells. These anti-tumor effects of AZD8055 were verified in vivo. Our findings collectively demonstrated that low-dose AZD8055 induces cytotoxicity and apoptosis, and inhibits the Akt/mTOR activation, invasion and migration of bladder cancer. These findings also demonstrate that AZD8055 partially blocked the interactions of bladder cancer cells and macrophages. In conclusion, AZD8055 is a promising mTOR inhibitor for bladder cancer.

The ongoing energy revolution has led to the gradual replacement of traditional synchronous generators by renewable enecorresponding authorrgy. This transformation of power generation mode reshapes power system dynamics and poses many challenges to reliable and efficient grid operation. With the continuous improvement of the penetration level of renewable energy represented by wind power in the power system, the synchronous mode of wind turbine will gradually become one of the main modes of system synchronization. In the case of coexistence of multiple synchronization modes, the synchronization mechanism of the whole power system will be fundamentally changed; at the same time, due to the response characteristics of wind turbine to the frequency and voltage dynamics of power grid and the traditional power supply, the synchronous mode of wind turbine will gradually become one of the main modes of system synchronization The frequency and voltage stability of power system also faces new challenges. The virtual synchronous generation technology can make the grid connected inverter simulate the operation mechanism of step generator and provide an effective method to solve the above problems. In this paper, the main technical routes to realize virtual synchronous control of wind turbines are summarized. From the aspects of stability analysis, control system optimization, performance analysis and evaluation, the research and engineering status of virtual synchronization technology in wind turbines are introduced, and the problems to be solved and technical requirements are prospected.

Abstract Background The focus of students’ health concerns has gradually progressed from the single factor of physical health to comprehensive health factors, and the physical and mental health of students are now generally considered together. This study focuses on exploring the status of junior high school students’ physical health and their subjective health assessment with the major societal factors that affect students’ lives: School Life and Family involvement. In addition, we explore the main factors influencing students’ subjective health. Methods A cross-sectional survey was conducted with 190 Tibetan junior high school students in the Maozhuang Township. The intentional sampling was used to choose the research object. The structured questionnaire comprised four parts, namely social and demographic information, family condition, school life, and subjective health quality which was assessed by PROMIS (Chinese version of the Pediatric Patient-Reported Outcomes Measurement Information System). Result The average height and weight of boys and girls are statistically different (p-values of 0.026 and 0.044, respectively), but there is no statistically significant difference in BMI (Body Mass Index) between boys and girls (p-value of 0.194). The potential association between calcium oxalate stones and renal fibrosis has been extensively investigated; however, the underlying mechanisms remain unclear. Ferroptosis is a novel form of cell death characterized by iron-dependent lipid peroxidation and regulated by acyl coenzyme A synthase long-chain family member 4 (ACSL4). Yes-associated protein (YAP), a transcriptional co-activator in the Hippo pathway, promotes ferroptosis by modulating ACSL4 expression. Nevertheless, the involvement of YAP–ACSL4 axis-mediated ferroptosis in calcium oxalate crystal deposition-induced renal fibrosis and its molecular mechanisms have not been elucidated. In this study, we investigated ACSL4 expression and ferroptosis activation in the kidney tissues of patients with calcium oxalate stones and in mice using single-cell sequencing, transcriptome RNA sequencing, immunohistochemical analysis, and Western blot analysis. In vivo and in vitro experiments demonstrated that inhibiting ferroptosis or ACSL4 mitigated calcium oxalate crystal-induced renal fibrosis. Furthermore, YAP expression was elevated in the kidney tissues of patients with calcium oxalate stones and in calcium oxalate crystal-stimulated human renal tubular epithelial cell lines. Mechanistically, in calcium oxalate crystal-stimulated human renal tubular epithelial cell lines, activated YAP translocated to the nucleus and enhanced ACSL4 expression, consequently inducing cellular ferroptosis. Moreover, YAP silencing suppressed ferroptosis by downregulating ACSL4 expression, thereby attenuating calcium oxalate crystal-induced renal fibrosis. Conclusively, our findings suggest that YAP–ACSL4-mediated ferroptosis represents an important mechanism underlying the induction of renal fibrosis by calcium oxalate crystal deposition. Targeting the YAP–ACSL4 axis and ferroptosis may therefore hold promise as a potential therapeutic approach for preventing renal fibrosis in patients with kidney stones.
