3. Restoring process of sunspot penumbra

We describe the disappearance of a sector of sunspot penumbra and its restoring process observed in the preceding sunspot of active region NOAA 12348. The evolution of the magnetic field and the plasma flows support the idea that the penumbra forms due to a change of inclination of the magnetic field of the canopy. Moving magnetic features have been observed during the disintegration phase of that sector of sunspot penumbra. During the restoring phase we have not observed any magnetic flux emergence around the sunspot. The restoring process of the penumbra sector completed in about 72 hours and it was accompanied by the transition from the counter-Evershed flow to the classical Evershed flow. The inversion of photospheric spectropolarimetric measurements taken by IBIS allowed us to reconstruct how the uncombed configuration of the magnetic field forms during the new settlement of the penumbra, i.e., the vertical component of the magnetic field seems to be progressively replaced by some horizontal field lines, corresponding to the intra-spines., Comment: 34 pages, 11 figures, accepted for ApJ

5. Solar flare forecasting using morphological properties of sunspot groups

We describe a new tool developed for solar flare forecasting on the base of some sunspot group properties. Assuming that the flare frequency follows the Poisson statistics, this tool uses a database containing the morphological characteristics of the suspot groups daily observed by the Equatorial Spar of INAF $-$ Catania Astrophysical Observatory since January 2002 up today. By means of a linear combination of the flare rates computed on the base of some properties of the sunspot groups, like area, number of pores and sunspots, Zurich class, relative importance between leading spot and density of the sunspot population, and type of penumbra of the main sunspot, we determine the probability percentages that a flare of a particular energy range may occur. Comparing our forecasts with the flares registered by GOES satellites in the 1$-$8 \AA{} X$-$ray band during the subsequent 24 hrs we measured the performance of our method. We found that this method, which combines some morphological parameters and a statistical technique, has the best performances for the strongest events, which are more interesting for their implications in the Earth environment., Comment: 15 pages. accepted by the "Journal of Space Weather and Space Climate"

In this paper we describe observations acquired by satellite instruments (Hinode/SOT and IRIS) and ground-based telescopes (ROSA@DST) during two consecutive C7.0 and X1.6 flares occurred in active region NOAA 12205 on 2014 November 7. The analysis of these data show the presence of continuum enhancements during the evolution of the events, observed both in ROSA images and in IRIS spectra. Probably, the long-term monitoring of the solar atmosphere started in Italy with the first telescopic observations of the Sun made by Galileo Galilei in the early $17^{\mathrm{th}}$ century. His recorded observations and science results, as well as the work carried out by other following outstanding Italian astronomers inspired the start of institutional programs of regular solar observations at the Arcetri, Catania, and Rome Observatories. These programs have accumulated daily images of the solar photosphere and chromosphere taken at various spectral bands over a time span greater than 80 years. In the last two decades, regular solar observations were continued with digital cameras only at the Catania and Rome Observatories, which are now part of the INAF National Institute for Astrophysics. At the two sites, daily solar images are taken at the photospheric G-band, Blue ($\lambda=409.4$ nm), and Red ($\lambda=606.9$ nm) continua spectral ranges and at the chromospheric Ca II K and H$\alpha$ lines, with a $2^{\prime\prime}$ spatial resolution. Solar observation in Italy, which benefits from over 2500 hours of yearly sunshine, currently aims at the operational monitoring of solar activity and long-term variability and at the continuation of the historical series as well. Existing instruments will be soon enriched by the SAMM double channel telescope equipped with magneto-optical filters that will enable the tomography of the solar atmosphere with simultaneous observations at the K I 769.9 nm and Na I D 589.0 nm lines. 9. Homologous white light solar flares driven by photosperic shear motions

We describe the peculiarity of two recurrent white light flares occurred on Sept. 06, 2017, in the super Active Region (SAR) NOAA 12673, with a time interval, between their peaks, of about 3 hours. These events of X2.2 and X9.3 GOES class are very important not only for their high level of emission and for the visible effects on the lower layers of the solar atmosphere, discernible as clear white light ribbons, but as well for the strong horizontal photospheric motions which seemed to drive them. In fact, we observed for several hours before the flare occurrence the displacement of a negative umbra located in the main delta spot of the Active Region. We measured velocities up to 0.6 km s$^{-1}$. The strong and persistent shear motion of the photospheric structures together with the high intensity of the magnetic flux involved by these events can be considered responsible for the new energy continuously supplied into the magnetic system. From the timing of the emissions at different wavelengths we were able to provide some constraints for the modeling of such events., Comment: 19 pages, 5 figures

10. Observation of a 3D magnetic null point

We describe high resolution observations of a GOES B-class flare characterized by a circular ribbon at chromospheric level, corresponding to the network at photospheric level. We interpret the flare as a consequence of a magnetic reconnection event occurred at a three-dimensional (3D) coronal null point located above the supergranular cell. The potential field extrapolation of the photospheric magnetic field indicates that the circular chromospheric ribbon is cospatial with the fan footpoints, while the ribbons of the inner and outer spines look like compact kernels. We found new interesting observational aspects that need to be explained by models: 1) a loop corresponding to the outer spine became brighter a few minutes before the onset of the flare; 2) the circular ribbon was formed by several adjacent compact kernels characterized by a size of 1"-2"; 3) the kernels with stronger intensity emission were located at the outer footpoint of the darker filaments departing radially from the center of the supergranular cell; 4) these kernels start to brighten sequentially in clockwise direction; 5) the site of the 3D null point and the shape of the outer spine were detected by RHESSI in the low energy channel between 6.0 and 12.0 keV. Taking into account all these features and the length scales of the magnetic systems involved by the event we argued that the low intensity of the flare may be ascribed to the low amount of magnetic flux and to its symmetric configuration., Comment: accepted for the publication in apj

11. Kinematics and Magnetic Properties of a Light Bridge in a Decaying Sunspot

We present the results obtained by analyzing high spatial and spectral resolution data of the solar photosphere acquired by the CRisp Imaging SpectroPolarimeter at the Swedish Solar Telescope on 6 August 2011, relevant to a large sunspot with a light bridge (LB) observed in NOAA AR 11263. These data are complemented by simultaneous Hinode Spectropolarimeter (SP) observation in the Fe I 630.15 nm and 630.25 nm lines. The continuum intensity map shows a discontinuity of the radial distribution of the penumbral filaments in correspondence with the LB, which shows a dark lane (about 0.3" wide and about 8.0" long) along its main axis. The available data were inverted with the Stokes Inversion based on Response functions (SIR) code and physical parameters maps were obtained. The line-of-sight (LOS) velocity of the plasma along the LB derived from the Doppler effect shows motions towards and away from the observer up to 0.6 km/s, which are lower in value than the LOS velocities observed in the neighbouring penumbral filaments. The noteworthy result is that we find motions toward the observer up to 0.6 km/s in the dark lane where the LB is located between two umbral cores, while the LOS velocity motion toward the observer is strongly reduced where the LB is located between an umbral core at one side and penumbral filaments on the other side. Statistically, the LOS velocities correspond to upflows/downflows andcomparing these results with Hinode/SP data, we conclude that the surrounding magnetic field configuration (whether more or less inclined) could have a role in maintaining the conditions for the process of plasma piling up along the dark lane. The results obtained from our study support and confirm outcomes of recent magnetohydro-dynamic simulations showing upflows along the main axis of a LBs.

The present day universe consists of galaxies, galaxy clusters, one-dimensional filaments and two-dimensional sheets or pancakes, all of which combine to form the cosmic web. The so called "Zeldovich pancakes", are very difficult to observe, because their overdensity is only slightly greater than the average density of the universe. Falco et al (2014) presented a method to identify Zeldovich pancakes in observational data, and these were used as a tool for estimating the mass of galaxy clusters. Here we expand and refine that observational detection method. We study two pancakes on scales of 10 Mpc, identified from spectroscopically observed galaxies near the Coma cluster, and compare with twenty numerical pancakes. We find that the observed structures have velocity dispersion about 100 km/sec, which is relatively low compared to typical groups and filaments. These velocity dispersions are consistent with those found for the numerical pancakes. We also confirm that the identified structures are in fact two-dimensional structures. Finally, we estimate the stellar to total mass of the observational pancakes to be $2 \times 10^{-4}$, within one order of magnitude, which is smaller than that of clusters of galaxies., Comment: 19 pages, 11 figures. When measuring the mass profile of any given cosmological structure through internal kinematics, the distant background density is always ignored. This trick is often refereed to as the "Jeans Swindle". Without this trick a divergent term from the background density renders the mass profile undefined, however, this trick has no formal justification. We show that when one includes the expansion of the Universe in the Jeans equation, a term appears which exactly cancels the divergent term from the background. SMS is caused by interstitial 17p11.2 deletions, encompassing multiple genes and including the retinoic acid-induced 1 gene (RAI1), or by mutations in RAI1 itself. About 10% of all the SMS patients, in fact, carry an RAI1 mutation responsible for the phenotype. RAI1 (OMIM *607642) is a dosage-sensitive gene expressed in many tissues and highly conserved among species. Over the years, several studies have demonstrated that RAI1 (or its homologs in animal models) acts as a transcriptional factor implicated in embryonic neurodevelopment, neuronal differentiation, cell growth and cell cycle regulation, bone and skeletal development, lipid and glucose metabolisms, behavioral functions, and circadian activity. Patients with RAI1 pathogenic variants show some phenotypic differences when compared to those carrying the typical deletion. They usually have lower incidence of hypotonia and less cognitive impairment than those with 17p11.2 deletions but more frequently show the behavioral characteristics of the syndrome and overeating issues. These differences reflect the primary pathogenetic role of RAI1 without the pathogenetic contribution of the other genes included in the typical 17p11.2 deletion. The better comprehension of physiological roles of RAI1, its molecular co-workers and interactors, and its contribution in determining the typical SMS phenotype will certainly open a new path for therapeutic interventions. - Abstract
Natural killer (NK) cells represent a promising cell type in antitumor immunotherapy for efficacy and safety, particularly in the treatment of hematologic malignancies. NK cells have been shown to exert antileukemia activity in the context of haploidentical hematopoietic stem cell transplantation (haplo-HSCT). Products have been developed to boost the activation of NK cells only when cross-linked by tumor cells, avoiding any off-target effect. Here, we tested the in vitro effect of different NK-cell engagers (NKCE), which trigger either NKp46 or NKp30 together with CD16A, and target either CD19 or CD20 to induce killing of pediatric B-cell precursor acute lymphoblastic leukemia (BCP-ALL). Target cells were NALM-16 and MHH-CALL-4 cell lines and four primary leukemias, while effector cells were resting NK cells derived from healthy donors and pediatric patients with leukemia after abT/Bdepleted haplo-HSCT. The NK cell-resistant MHH-CALL-4 was efficiently killed using all NKCEs. Boosting of NK activity against MHH-CALL-4 was also evident by degranulation and IFNg production. Because of the lack of CD20 and high expression of CD19 on primary BCP-ALL, we focused on NKCEs targeting CD19. NKp46- and NKp30-based NKCEs displayed similar potency at inducing NK-cell activity, even when challenged with primary BCPALL blasts. Their efficacy was shown also using NK cells derived from transplanted patients. NKCE-induced activation against BCPALL can override HLA-specific inhibitory interactions, although the strongest response was observed by the alloreactive NK-cell subset. These data support the therapeutic use of NKp46/CD16A/ CD19-NKCE to fight refractory/relapsed leukemia in pretransplantation or posttransplantation settings.

- Abstract
Several nonmalignant disorders (NMDs), either inherited or acquired, can be cured by allogeneic hematopoietic stem cell transplantation (HSCT). Between January 2012 and April 2020, 70 consecutive children affected by primary immunodeficiencies, inherited/acquired bone marrow failure syndromes, red blood cell disorders, or metabolic diseases, lacking a fully matched donor or requiring urgent transplantation underwent TCRab/CD19-depleted haploidentical HSCT from an HLA-partially matched relative as part of a prospective study. The median age at transplant was 3.5 years (range 0.3-16.1); the median time from diagnosis to transplant was 10.5 months (2.7 for SCID patients). Primary engraftment was obtained in 51 patients, while 19 and 2 patients experienced either primary or secondary graft failure (GF), the overall incidence of this complication being 30.4%. Most GFs were observed in children with disease at risk for this complication (eg, aplastic anemia, thalassemia). All but 5 patients experiencing GF were successfully retransplanted. Six patients died of infectious complications (4 had active/ recent infections at the time of HSCT), the cumulative incidence of transplant-related mortality (TRM) being 8.5%. Cumulative incidence of grade 1-2 acute GVHD was 14.4% (no patient developed grade 3-4 acute GVHD). Only one patient at risk developed mild chronic GVHD. With a median follow-up of 3.5 years, the 5-year probability of overall and disease-free survival was 91.4% and 86.8%, respectively. In conclusion, TCRab/CD19-depleted haploidentical HSCT from an HLA-partially matched relative is confirmed to be an effective treatment of children with NMDs. Prompt donor availability, low incidence of GVHD, and TRM make this strategy an attractive option in NMDs patients. The study is registered at ClinicalTrial.gov as NCT01810120.

Although many efforts have been dedicated to the development of sustainable biorefineries, there are still some gaps to cover, especially to make chemicals production energetically efficient and rentable. The proposed route combines biotechnological and thermochemical processes, departing from low-cost raw materials to produce valuable chemicals via the activity of environmental microbial communities. This work study the use of bio-oil (obtained from pyrolysis of lignocellulosic waste biomass from agricultural local activities) as carbon source for microbial communities from hydrocarbon contaminated environments. The aim is that these microbial communities (resistant to hydrocarbons) use the bio-oil as carbon source to produce polyhydroxyalkanoates (PHAs), which are biocompatible polymers with similar properties to polyethylene and polypropylene. Sediments were sampled from two different hydrocarbon contaminated environments and microbial communities were isolated in batch cultures under feast-famine regime. Bio-oil was produced by intermediate pyrolysis of lignocellulosic biomass in a batch reactor at 500°C. The bio-oil was used as carbon source for microbial cultures. Preliminary results showed that the microbial communities isolated from the sediments can grow in presence of bio-oil and use it as carbon source. The presence of PHA granules was observed by fluorescence under microscopy.
