Aim: To develop delivery systems for efficient siRNA delivery to breast cancer. Methods: Poly(ethylene oxide)-block-poly(ϵ-caprolactone-grafted-spermine) (PEO-b-P(CL-g-SP)) micelles were modified with cholesterol group in their core and with RGD4C peptide on their shell. Transfection efficiency of complexed MCL-1 siRNA in MDA-MB-435 was investigated, in vitro and in vivo following intratumoral and intravenous injection. Results: Cholesteryl modification of the core significantly increased the transfection efficiency of PEO-b-P(CL-g-SP)-complexed siRNA, in vitro, but not following intratumoral or intravenous administration, in vivo. Instead, RGD4C modification of the micellar shell enhanced transfection efficiency of complexed MCL-1 siRNA in tumor upon intravenous administration. Conclusion: RGD4C-PEO-b-P(CL-g-SP) micelles, without or with cholesterol modification, can provide efficient delivery of siRNA to breast tumors following systemic administration.

Post-transcriptional gene silencing mechanism using small interfering RNA (siRNA) as a potential therapeutic modality has generated great interest in recent years. However, significant hurdles such as off-target effects, possible immunogenicity, in vivo instability and effective delivery to the cellular and intracellular site of action following systemic administration should be overcome before synthetic siRNA can be used as an effective therapeutic agent. Development of several siRNA delivery systems as well as chemical modification of the siRNA structure has been pursued to combat problems associated with therapeutic use of siRNA. Among different structures, polymeric micelles have attracted tremendous attention in recent years as promising carriers for targeted siRNA delivery. The aim of this manuscript is to review different polymeric micelles of siRNA developed to date and provide an overview on the extent of in vitro/in vivo success in siRNA delivery, targeted gene silencing or therapeutic activity by these siRNA delivery systems.

The aim of this study was to develop methoxy poly(ethylene oxide)-b-poly(epsilon-caprolactone) (MePEO-b-PCL) containing stearyl (St) substituents on PCL, and assess the efficacy of nanocarriers formed from this structure in comparison to unmodified MePEO-b-PCL and those with carboxyl substitutes on PCL on the solubilization and delivery of Amphotericin B (AmB). Prepared block copolymers were characterized for their molecular weight by (1)H NMR and gel permeation chromatography. The self-assembly of synthesized MePEO-b-PStCL to spherical particles of nanometer size range was shown by dynamic light scattering as well as electron and atomic force microscopy. Encapsulated AmB showed a reduction in its hemolytic activity against rat red blood cells in comparison to the commercial formulation of AmB, Fungizone.

A sensitive and specific reversed phase HPLC method was developed to quantitate plasma levels of cisapride in order to conduct comparative bioavailability studies. The drug and internal standard was extracted from plasma with heptane-isoamyl alcohol (95:5 v/v) and back extracted with sulfuric acid. The acidic layer was then re-extracted with the same extracting solvent. The separated organic layer was evaporated to dryness under nitrogen and the residue reconstituted with acetonitrile. Analysis was performed on a C-8 Sil-X-10 HPLC column, with a mobile phase of acetonitrile, water, and triethylamine (75:25:0.01) and UV detection at 215 nm. The standard curve covering the concentration range 5-160 ng/ml was linear (r(2)=0.9992), relative errors were within +/-10% and the CV% ranged from 1.34 to 11.82. The in vivo study was carried out in 12 healthy volunteers according to a single dose, two-sequence, cross over randomized design. The bioavailability was compared using the total area under the plasma level versus time curve (AUC(0-34,) AUC(0- infinity )), peak plasma concentration (C(max)) and time to C(max) (T(max)). No statistically significant difference was found between the AUC(0- infinity ) or C(max) values of the test (cisapride) and reference (Propulsid). It was, therefore, concluded that the generic cisapride was bioequivalent with the innovator formulation.

Considering the narrow therapeutic index of theophylline and the low range between the safe and toxic serum concentrations of this drug, the study of its pharmacokinetic properties is necessary. However, considering the time consuming and expensive in vivo tests, quantitative correlation between in vivo bioavailability and in vitro dissolution tests can be used routinely in quality control tests of these drug products to predict the in vivo pharmacokinetic parameters. For this reason healthy human volunteers were used for in vivo studies and serum samples were analyzed by a fluorescence polarization immunoassay analysis (FPIA) method. The results showed that an open one compartmental model could best describe the pharmacokinetic properties of orally administered theophylline and aminophylline tablets. Linear regression analysis by least-square method showed a good correlation between some in vivo and in vitro parameters obtained from dissolution studies by rotating basket and paddle methods. D(30)% (percentage of drug dissolved in vitro after 30 min) and F(0.5)% (drug absorbed in vivo after half an hour calculated by Wagner-Nelson equation) showed best correlation (r=0. 99036). C(max) (maximum serum concentration) of this drug also correlates well with t(25%) (time required to dissolve 25% of the drug). The calculated correlation coefficients could best predict the actual values of some pharmacokinetic parameters; AUC(0-->infinity), AUC(0-->1), F(0.5)% and C(max).

Signal transducer and activator of transcription 3 (STAT3) is a transcription factor that plays a major role in the development of resistance to conventional anti-cancer drugs in many types of cancer, when constitutively activated. Inhibition of STAT3 is considered as a promising strategy for inhibition of tumor growth and overcoming the drug resistance manifested. In this study, the capability of STAT3 knockdown by lipid substituted low molecular weight (2 kDa) polyethyleneimine (PEI2) complexes of STAT3-siRNA was assessed. The efficiency of PEI/STAT3-siRNA polyplexes in the induction of STAT3 associated cell death in wild type and drug-resistant MDA-MB-435 breast cancer cells as monotherapy and upon combination with chemotherapeutic agents, doxorubicin and paclitaxel, was also investigated. Our results identified linoleic acid-substituted (PEI-LA) polymer as the most efficient carrier among different lipid substituted PEI2 for siRNA delivery, leading to most STAT3 associated loss of cell viability in MDA-MB-435 cells. STAT3-siRNA delivery by the PEI-LA polymer resulted in efficient down-regulation of STAT3 at both mRNA and protein levels. Furthermore, pre-treatment of cancer cells with STAT3-siRNA formulation increased the cytotoxic effect of doxorubicin and paclitaxel in both wild type and drug resistant MDA-MB-435 cells. The results of this study point to the potential of PEI-LA polyplexes of STAT3-siRNA as inhibitors of STAT3 expression in breast tumor cells. The results also demonstrate an improved efficacy for chemotherapeutic drugs in combination with lipid-substituted low molecular weight PEI-LA/STAT3-siRNA complexes in comparison to drug therapy alone.

Ethyl cellulose encapsulated aspirin particles, suitable for preparation of direct compression tablets were prepared by the solvent evaporation method. Ethyl acetate was used as a solvent for the polymer in combination with a saturated solution of aspirin as the dispersing medium to prevent partitioning and drug loss. This resulted in a high yield of free-flowing, non-aggregated particles. In vitro-in vivo evaluations of the experimental aspirin tablets (made by direct compression of ethyl cellulose encapsulated particles) and three different commercial aspirin products (a conventional tablet, a timed-release tablet, and a timed-release caplet) were undertaken. Comparison of the dissolution in acidic media at pH 1.2 showed different release profiles for these products. While the conventional tablet and the timed-release caplet showed the highest and the lowest rate of release, respectively; the timed-release tablet and the experimentally made tablet revealed an intermediate rate and very similar release profiles. The cumulative urinary excretion data collected in a complete crossover study, using five healthy subjects further indicated that the experimental tablet has an in vivo availability identical to that of the timed-release tablet.

Methylmethacrylate/dimethylaminoethyl methacrylate cross-linked with divinylbenzene is a pH-sensitive hydrogel. We have studied the diffusion mechanisms of drugs with different water solubilities through this hydrogel. A water-soluble model drug (aminopyrine) was used to study the diffusion coefficient changes in different pHs. The results showed a water-content dependent diffusion for this pH-sensitive polycationic hydrogel. However, decreasing the solubility of the drug and increasing the hydrophobic character of the polymer by changing the pH caused a greater affinity (or partition coefficient) between the hydrogel and the drug. Aminopyrine diffusion was shown to follow the free-volume theory, suggesting the 'pore' type mechanism for water soluble drugs, while the 'partition' or 'solution-diffusion' mechanism better described the slow diffusion of water insoluble solutes through this pH-sensitive hydrogel. Comparing the swelling interface number for aminopyrine release through this pH-sensitive hydrogel showed a non-Fickian mechanism in the hydrated form of the hydrogel (pH 1.2), while Fickian in the dehydrated form (pH 7.4).

The effect of pendent side-chain length and crosslinking agent concentration in methyl methacrylate/dimethylaminoethyl methacrylate as a polybasic/hydrophobic pH-sensitive hydrogel was studied. Increasing both side-chain length and crosslinking agent concentration decreased the sharpness of response to pH and water-uptake capacity of the polymer. A case-II water transport mechanism and a nonlinear swelling kinetic was observed for the homologues of this hydrogel.

In relation to the new pharmaceutical system in Iran, the in vitro release of five brands of 100 mg phenytoin sodium capsules, namely A,B,C,D & E were determined in distilled water. using three dissolution methods, i.e. Rotating basket, Magnetic basket and Levy beaker method. Also the average amount of phenytoin content of each brand was measured.The results showed that although the dissolution rate of each product is different by each method, but the pattern of drug release is more or less similar.The dissolution time for products C and D is much longer than those of products A,B & C with all methods, but the dissolution behaviour of capsules C & D is not equivalent to those of standard “slow release” phenytoin capsules. The release pattern of products A & E are similar to those of standard “fast release” phenytoin capsules. The dissolution of product B is poor and not acceptable.

The word-association technique was used to evaluate public awareness of texture in medicines. The results indicated that texture is an important attribute of medicines. The degree of association with texture varied with the type of medicine. Externally used semi-solid dosage forms, such as creams, pastes and ointments, and also viscous materials, such as medicinal syrups, elicited more texture responses than the other types of medicines. The largest percentage of responses belonged to a ‘visual aspects’ category, while texture responses were the second largest group.
