Lactose is the main carbohydrate in milk and its absorption occurs due to enzymatic hydrolysis, generating glucose and galactose. Lactose intolerance is the reduction of the intestinal hydrolysis capacity due to hypolactasia, promoving the need to consume dairy foods with low content of this carbohydrate. The beta-galactosidase enzymes are used in dairy industries to hydrolyze lactose, providing the possibility of eating dairy products without harm to health to the intolerant consumer. Alternative and official analytical methods are used to quantify the carbohydrate content resulting from the enzymatic hydrolysis. The objective of this study was to evaluate the enzymatic hydrolysis of two distinct industrial enzymes produced by the microorganisms Bacillus licheniformis and Kluyveromyces lactis using three analytical methods: enzymatic method, cryoscopy and HPLC using artificial intelligence to improve the control of industrial processes. After adding the enzymes to the skim milk, time kinetics was performed collecting samples at time 0, every 10 minutes until completing 1hour of reaction, and every 30 minutes until the closure of 05 hours of hydrolysis reaction. In 97% of the cases, the decrease in lactose concentration were observed by HPLC followed (by) the deepening of the cryoscopic point. Glucose measurements by absorbance and HPLC quantification were correlated (r = 0.79; p < 0.01), but not concordant (p < 0.01). It was concluded that by means of the artificial intelligence it is possible to indirectly estimate the lactose concentration from an algorithm that associates cryoscopy and glucose concentration. O principal carboidrato do leite é a lactose e a sua absorção ocorre devido à hidrólise enzimática, gerando glicose e galactose. A intolerância à lactose é a redução da capacidade de hidrólise intestinal devido à hipolactasia, gerando a necessidade do consumo de alimentos lácteos com baixo teor deste carboidrato. As enzimas beta-galactosidase são utilizadas nas indústrias de laticínios para hidrolisar a lactose, proporcionando ao consumidor intolerante a possibilidade de ingerir os produtos lácteos sem prejuízos à saúde. Para quantificar o conteúdo de carboidratos resultante da hidrólise enzimática, são utilizados métodos analíticos alternativos e oficiais. O objetivo deste estudo foi avaliar a hidrólise enzimática de duas enzimas industriais distintas produzidas pelos microrganismos Bacillus licheniformis e Kluyveromyces lactis, através de três métodos analíticos: método enzimático, crioscopia e HPLC. A inteligência artificial foi utilizada para melhorar o controle dos processos industriais. Após a adição das enzimas ao leite desnatado, foi realizada a cinética de tempo coletando as amostras no tempo 0, a cada 10 minutos até completar 1 hora de reação e a cada 30 minutos até o fechamento das 5 horas de reação de hidrólise. Em 97% dos casos observados a diminuição da concentração de lactose por HPLC acompanhou o aprofundamento do ponto crioscópico. As medições de glicose por absorbância e HPLC foram correlacionadas (r = 0,79; p < 0,01), mas não concordantes (p < 0,01). Concluiu-se que, por meio da inteligência artificial, é possível estimar indiretamente a concentração de lactose a partir de um algoritmo que associa a crioscopia e a concentração de glicose.

In humans, it's known that there is a significant correlation between obesity and the development of hypertension and insulin resistance, a clinical picture called metabolic syndrome. In dogs, however, there is only anecdotal about it. The objectives of the study werel to determine serum levels of adiponectin, leptin, triglycerides, cholesterol, insulin, glucose, HOMA-B, HOMA-IR, and systolic blood pressure in dogs with different body conditions (BC) without endocrine disease. Seventy six healthy dogs submitted to evaluation of BC and morphometry. The dogs were separated into four groups: optimal BC (ECC 3,4 or 5 of 9 and GC% of 13 to 27-G1), overweight (ECC 6 and 7 of 9 and GC% of 14 to 38- G2), obese (ECC 8 and 9 of 9 and GC% greater than or equal to 34-G3). G3 presented higher serum levels of total protein, triglycerides, glucose, insulin, and HOMA-IR. Adipokines didn't correlate to any other parameter, but the occurrence of hyperinsulinemia was higher in G3. The results have shown that obese dogs presented IR and alterations in fat metabolism. The seasons have a major influence on milk production, especially when animals are exposed to climatic variations. This situation is very common in Brazil and further investigation is needed to understand the losses caused by oscillations in milk constituents. The aim was to evaluate how seasonality can affect the physicochemical and microbiological quality of raw milk in Ivaiporã region, state of Paraná, Brazil. In this research, the milk from properties in northern Paraná was evaluated during 24 months for lactose, protein, fat, total solids, total bacteria count and somatic cell count. The regional rainfall and average temperature recorded at each station were analyzed for determination of Thermal Comfort Index and evaluation of heat stress. This study concluded that seasonality have influence on microbiological and physicochemical quality of milk in Ivaiporã region, with higher percentages of solids in the fall and inferior microbiological quality in the spring. As estações do ano têm grande influência na produção leiteira, principalmente quando os animais lactentes ficam expostos às suas variações climáticas. No Brasil, essa situação é recorrente, sendo necessários maiores esclarecimentos a fim de amenizar os prejuízos decorrentes de oscilações em componentes do leite. O objetivo foi avaliar como a sazonalidade influencia os componentes físico-químicos e a qualidade microbiológica do leite cru da região de Ivaiporã – PR. Assim, o leite produzido em propriedades do norte do Paraná foi analisado durante o decorrer de 24 meses. As amostras foram quantificadas quanto à porcentagem de lactose, proteínas, gordura, sólidos totais, contagem de bactérias totais e contagem de células somáticas. O índice pluviométrico regional e a temperatura média registrada em cada estação foram analisados para a determinação de Índice de Conforto Térmico e avaliação do estresse térmico sofrido pelos animais. Este estudo concluiu que a sazonalidade tem influência nas características microbiológicas e físico-químicas do leite na região de Ivaiporã, com maiores porcentagens de sólidos no outono e pior qualidade microbiológica na primavera.

Considerando que o leite é um alimento susceptível a fraudes, o presente trabalho teve como objetivo avaliar o efeito da adição de diferentes concentrações de citrato de sódio e fortificante na densidade e crioscopia do leite. A adição de 0,1% de citrato de sódio foi suficiente para diminuir a média do ponto de congelamento em 0,021° H e aumentar a média da densidade em 0,0008 g cm-3. Nessas amostras foi possível adicionar até 5,6% de água sem ultrapassar os padrões brasileiros de densidade e crioscopia. O fortificante adicionado ao leite em concentrações iguais ou maiores que 0,056% diminuiu a média da crioscopia em 0,0024o H quando comparada ao tratamento controle. A média da densidade diferiu apenas no grupo com adição de 0,44% de fortificante, com aumento de 0,0034 g cm-3 em relação ao tratamento controle. Na diluição recomendada pelo fabricante (0,007%), o fortificante não alterou as médias de densidade e crioscopia do leite. Os resultados mostraram que a adição de citrato de sódio, mesmo dentro da porcentagem permitida pela legislação, foi capaz de alterar o ponto de congelamento e a densidade do leite. Considerando que no processo UAT direto, a evaporação do excesso da água do leite com 0,1% de citrato de sódio é baseada em 0,530º H, é possível que a retirada da água incorporada no beneficiamento não seja completa, podendo estar presente em porcentagens superiores a 5%. Por sua vez, a adição de fortificante na concentração recomendada pelo fabricante não deixa margens para a adição fraudulenta de água.

Resumo Vários fatores extratireoidianos como idade, raça, doenças concomitantes e fármacos podem influenciar na função e no diagnóstico do hipotireoidismo em cães. As alterações nas concentrações dos hormônios tireoidianos decorrentes de doenças não tireoidianas é denominada de síndrome do eutireoideo doente. O objetivo deste estudo foi avaliar a influência da leishmaniose visceral na função da tireoide em cães sem sinais de hipotireoidismo, com ou sem azotemia. Os animais positivos para leishmaniose foram divididos em seis grupos (albumina nornal ou diminuída, creatinina normal ou elevada, uréia normal ou elevada) e foi avaliado o efeito nesses grupos das concentrações séricas do hormônio estimulante da tireoide (TSH), tiroxina total (TT4) e da tiroxina livre (FT4). No. of bitstreams: 0 Previous issue date: 2014-01-01 Several non-thyroid factors such as age, breed, concomitant diseases and use of certain drugs might influence the diagnosis of hypothyroidism in dogs. Changes in the concentrations of thyroid hormones due to non-thyroid illnesses or drugs are named euthyroid sick syndrome. The aim of this study is to evaluate the thyroid function in dogs with musculoskeletal diseases or without musculoskeletal diseases receiving carprofen and chondroitin sulphate, glucosamine, collagen, ascorbic acid and manganese sulphate. A total of 43 dogs were used, divided into three groups: 20 dogs in the Carprofen Group (GC), 20 dogs in the Chondroitin Sulphate Group (GSC) and 15 dogs in the placebo group (GP). The animals included in the GC group were previously diagnosed with degenerative joint disease. Dogs in the GSC and GP groups did not present joint diseases or a diagnosis of systemic disease. Dogs in the GC group received daily 2.2 mg kg-1 carprofen during 60 days, twice a day. The dogs in the GSC group received daily chondroitin sulphate (30 mg kg-1) and association (glucosamine, collagen, ascorbic acid, manganese sulphate) during 60 days, twice a day. Dogs in the GP group received daily 10 mg kg-1 corn starch in capsule during 60 days, twice a day. The animals were evaluated in five Moments (M), the initial Moment (M0), before the first administration, resulting in basal values within the parameters studied; at 15 days (Ml), at 30 days (M2), at 45 days (M3) and at 60 days (M4). The measurements of thyroid function (total thyroxine-TT4, free thyroxine-FT4, thyrotropin-TSH) were performed in the five moments. In the control group the M's had an effect on TT4 and FT4 with reduced concentrations of TT4 below the reference values. In the SC group, the M's had an effect only on the FT4 and the highest average was observed in M4. Carprofen in dose for 60 days promoted the reduction of serum TT4 in dogs with musculoskeletal diseases. Therefore, the strain caused the TT4 would be consistent with hypothyroidism. Recommend discontinuation of therapy with carprofen for a minimum period of 15-30 days before the tests of thyroid function to be interference. While chondroitin sulfate at the dose and duration used does not alter thyroid function.
