In species richness studies, citizen-science surveys where participants make individual decisions regarding sampling strategies provide a cost-effective approach to collect a large amount of data. However, it is unclear to what extent the bias inherent to opportunistically collected samples may invalidate our inferences. Here, we compare spatial predictions of forest ground-floor bryophyte species richness in Limburg (Belgium), based on crowd- and expert-sourced data, where the latter are collected by adhering to a rigorous geographical randomisation and data collection protocol. We develop a log-Gaussian Cox process model to analyse the opportunistic sampling process of the crowd-sourced data and assess its sampling bias. We then fit two geostatistical Poisson models to both data-sets and compare the parameter estimates and species richness predictions. We find that the citizens had a higher propensity for locations that were close to their homes and environmentally more valuable. The estimated effects of ecological predictors and spatial species richness predictions differ strongly between the two geostatistical models. Unknown inconsistencies in the sampling process, such as unreported observer’s effort, and the lack of a hypothesis-driven study protocol can lead to the occurrence of multiple sources of sampling bias, making it difficult, if not impossible, to provide reliable inferences.

The present manuscript aims to discuss the implications of sequential knowledge integration of small preclinical trials in a Bayesian pharmacokinetic and pharmacodynamic (PK-PD) framework. While, at first sight, a Bayesian PK-PD framework seems to be a natural framework to allow for sequential knowledge integration, the scope of this paper is to highlight some often-overlooked challenges while at the same time providing some guidances in the many and overwhelming choices that need to be made. Challenges as well as opportunities will be discussed that are related to the impact of (1) the prior specification, (2) the choice of random effects, (3) the type of sequential integration method. In addition, it will be shown how the success of a sequential integration strategy is highly dependent on a carefully chosen experimental design when small trials are analyzed.

Most social and health science data are longitudinal and additionally multilevel in nature, which means that response data are grouped by attributes of some cluster. Ignoring the differences and similarities generated by these clusters results to misleading estimates, hence motivating for a need to assess variance components (VCs) using multilevel models (MLMs) or generalised linear mixed models (GLMMs). This study has explored and fitted teenage pregnancy census data that were gathered from 2011 to 2015 by the Africa Centre at Kwa-Zulu Natal, South Africa. The exploration of these data revealed a two level pure hierarchy data structure of teenage pregnancy status for some years nested within female teenagers. To fit these data, the effects that census year (year) and three female characteristics (namely age (age), number of household membership (idhhms), number of children before observation year (nch) have on teenage pregnancy were examined. Model building of this work, firstly, fitted a logit gen eralised linear model (GLM) under the assumption that teenage pregnancy measurements are independent between females and secondly, fitted a GLMM or MLM of female random effect. A better fit GLMM indicated, for an additional year on year, a 0.203 decrease on the log odds of teenage pregnancy while GLM suggested a 0.21 decrease and 0.557 increase for each additional year on age and year, respectively. A GLM with only year effect uncovered a fixed estimate which is higher, by 0.04, than that of a better fit GLMM. The inconsistency in the effect of year was caused by a significant female cluster variance of approximately 0.35 that was used to compute the VCs. Given the effect of year, the VCs suggested that 9.5% of the differences in teenage pregnancy lies between females while 0.095 similarities (scale from 0 to 1) are for the same female. Bayesian hierarchical models are attractive structures for conducting regression analyses when the data are subject to missingness. However, the requisite probability calculus is challenging andMonte Carlo methods typically are employed.We develop an alternative approach based on deterministic variational Bayes approximations. Both parametric and nonparametric regression are considered. Attention is restricted to the more challenging case of missing predictor data.We demonstrate that variational Bayes can achieve good accuracy, but with considerably less computational overhead. The main ramification is fast approximate Bayesian inference in parametric and nonparametric regression models with missing data. Also regional differences can be observed. This study aims at explaining the characteristics of the spatio-temporal changers in the geographical distance between the places of birth of spouses in the Netherlands in the period 1812-1922.
