Objective: To determine the lead bioactive compound in kernel extract of Mangifera pajang and its anti-cancer activity against human breast cancer cell lines with positive estrogen receptor (MCF-7). Methods: The methanolic extract of dried powder kernel of Mangifera pajang was exposed to column chromatography for isolation. The structural elucidation of the isolated compound was characterized using infrared, nuclear magnetic resonance, mass spectrometry. Furthermore, cytotoxicity, morphological changes, flow cytometry and cell cycle arrest analyses were performed to examine the mechanism of anti-proliferation and apoptosis induced by methyl gallate against MCF-7. Results: One compound was isolated from the methanolic extract of Mangifera pajang kernel and identified as methyl gallate. Phytosterols (PSs), classified into plant sterols and stanols, are bioactive compounds found in foods of plant origin. PSs have been proposed to exert a wide number of pharmacological properties, including the potential to reduce total and low‐density lipoprotein (LDL) cholesterol levels and thereby decreasing the risk of cardiovascular diseases. Other health‐promoting effects of PSs include anti‐obesity, anti‐diabetic, anti‐microbial, anti‐inflammatory, and immunomodulatory effects. Also, anticancer effects have been strongly suggested, as phytosterol‐rich diets may reduce the risk of cancer by 20%. The aim of this review is to provide a general overview of the available evidence regarding the beneficial physiological and pharmacological activities of PSs, with special emphasis on their therapeutic potential for human health and safety. Abstract: Humans are exposed to harmful pathogens and a wide range of noxious substances every day. The immune system reacts to, and destroys, these pathogens and harmful substances. The immune system is composed of innate and adaptive immunity, which liaise to protect the host and maintain health. Foods, especially cereals, have been reported to modulate the immune response. Arabinoxylans are nonstarch polysaccharides that have been shown to possess immune‐modulatory activities. This review article discusses the fundamentals of the immune system and provides an overview of the immunomodulatory potential of arabinoxylans in conjunction with their structural characteristics and proposed similarities with lipopolysaccharides. Practical applications: Understanding how the immune system works is of vital importance to prevent unnecessary or excessive inflammatory responses. Consumption of arabinoxylans has been shown to possess immunomodulatory potential. However, their mechanism of action has not been elucidated. Arabinoxylans share some similarities with lipopolysaccharides (LPS), a molecule that induces substantial and sometimes excessive immune responses such as fever following infection by pathogens. Thus, we propose that arabinoxylans might possibly act on the same receptor as LPS. Competition between dietary‐derived arabinoxylans and LPS at a shared receptor would then have the potential to inhibit or attenuate excessive LPS‐induced inflammatory responses that are typical of infection/fever. In the absence of infection and consequently no competition at the LPS receptor, consumption of dietary arabinoxylans may protect against the risk of infection by moderately activating the receptor and heightening natural (background) levels of immunity. Context: Postprandial lipemia (PPL) is associated with increased risk of endothelial dysfunction (ED), a precursor of atherosclerotic cardiovascular disease (ASCVD). The effects of low-carbohydrate, high-fat (LCHF) diets on ASCVD risk are uncertain; therefore, gaining a greater understanding of LCHF meals on PPL may provide valuable insights., Objective: The current systematic review investigated the effects of single LCHF meal consumption on PPL and markers of ED., Data Sources: CINAHL Plus, PubMed, Web of Science, and Cochrane Central Register of Controlled Trials (CENTRAL) were searched for key terms related to endothelial function, cardiovascular disease, glycemia, lipemia, and the postprandial state with no restriction on date., Data Extraction: Full-text articles were independently screened by 2 reviewers, of which 16 studies were eligible to be included in the current review. All trials reported a minimum analysis of postprandial triglycerides (PPTG) following consumption of an LCHF meal (<26% of energy as carbohydrate). Results were reported according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement., Data Analysis: Single-meal macronutrient composition was found to play a key role in determining postprandial lipid and lipoprotein responses up to 8 hours post-meal. Consumption of LCHF meals increased PPTG and may contribute to ED via reduced flow-mediated dilation and increased oxidative stress; however, energy and macronutrient composition varied considerably between studies., Conclusion: Consumption of an LCHF meal had a negative impact on PPL based on some, but not all, single-meal studies; therefore, the contribution of LCHF meals to cardiometabolic health outcomes remains unclear.
