Pathogenic yeasts and fungi are an increasing global healthcare burden, but discovery of novel antifungal agents is slow. The mycoparasitic yeast Saccharomycopsis schoenii was recently demonstrated to be able to kill the emerging multi-drug resistant yeast pathogen Candida auris. However, the molecular mechanisms involved in the predatory activity of S. schoenii have not been explored. To this end, we de novo sequenced, assembled and annotated a draft genome of S. schoenii. Using proteomics, we confirmed that Saccharomycopsis yeasts have reassigned the CTG codon and translate CTG into serine instead of leucine. Further, we confirmed an absence of all genes from the sulfate assimilation pathway in the genome of S. schoenii, and detected the expansion of several gene families, including aspartic proteases. Using Saccharomyces cerevisiae as a model prey cell, we honed in on the timing and nutritional conditions under which S. schoenii kills prey cells. We found that a general nutrition limitation, not a specific methionine deficiency, triggered predatory activity. Nevertheless, by means of genome-wide transcriptome analysis we observed dramatic responses to methionine deprivation, which were alleviated when S. cerevisiae was available as prey, and therefore postulate that S. schoenii acquired methionine from its prey cells. During predation, both proteomic and transcriptomic analyses revealed that S. schoenii highly upregulated and translated aspartic protease genes, probably used to break down prey cell walls. With these fundamental insights into the predatory behavior of S. schoenii, we open up for further exploitation of this yeast as a biocontrol yeast and/or source for novel antifungal agents. A new species Micromyces bulbosus Kadłubowska is described and illustrated from a puddle in the forest reserve Wolbórka (near Łódź~). This species differs from the others by the dimensions of the prosorus, sorus and resting spore, as well as shape and length of spines, also reticulate outer surface of prosorus.

Investigations carried out on the genus Spirogyra Link and Mougeotia Agardh revealed the following species of fungi parasiting in the Spirogyra and Mougeotia cells: Olpidium endogenum, Blyttiomyces helicus, B. spinulosus, Micromyces zygogonii and Rhizophydium ampullaceum. First information on B. helicus as parasitic on algae is presented.

Seven filamentous fungal species were isolated from individual eggs of Globodera pallida cysts collected from infested fields in Shelley Idaho, USA and identified as Chaetomium globosum, Fusarium oxysporum, Fusarium solani, Fusarium tricinctum, Microdochium bolleyi, Purpureocillium lilacinum, and Plectosphaerella cucumerina. Their ability to reduce infection by G. pallida in planta were assessed in simple, reproducible micro-rhizosphere chambers (micro-ROCs). All fungi reduced G. pallida infection in potato, but greatest reduction was observed with C. globosum at an average reduction of 76%. Further non-destructive methods were developed to rapidly assess biological control potential of putative fungal strains by staining the infectious second stage juveniles of G. pallida with the live fluorescent stain PKH26. In comparisons between the standard, invasive acid fuchsin method and use of the live stain PKH26, no significant difference in infection level of G. pallida was observed whether roots were stained with PKH26 or acid fuchsin. For both methods, a similar reduction (77% for acid fuchsin, and 78% for PKH26 stain) in invasion of infectious stage of G. pallida was observed when potato plants were inoculated with C. globosum compared to non-inoculated potato. More than 520 soil samples were surveyed for species of the mycoparasitic zygomycete genus Syncephalis using a culture-based approach. These fungi are relatively common in soil using the optimal conditions for growing both the host and parasite. Five species obtained in dual culture are unknown to science and are described here: (i) S. digitata with sporangiophores short, merosporangia separate at the apices, simple, 3-5 spored; (ii) S. floridana, which forms galls in the host and has sporangiophores up to 170 mm long with unbranched merosporangia that contain 2-4 spores; (iii) S. pseudoplumigaleta, with an abrupt apical bend in the sporophore; (iv) S. pyriformis with fertile vesicles that are long-pyriform; and (v) S. unispora with unispored merosporangia. To facilitate future molecular comparisons between species of Syncephalis and to allow identification of these fungi from environmental sampling datasets, we used Syncephalis-specific PCR primers to generate internal transcribed spacer (ITS) sequences for all five new species. The entomopathogenic fungus Lecanicillium lecanii persists in a highly dynamic network of habitat patches (i.e., a metapopulation) formed by its primary host, the green coffee scale Coccus viridis . Lecanicillium lecanii is an important biological control of both C . viridis and the coffee rust, Hemileia vastatrix . Successfully managing this biocontrol agent will depend on an increased understanding of the characteristics of its dispersal, as migration between occupied and unoccupied patches is essential for the persistence of this metapopulation. In the present study, we employ a population genetics approach, and show that in our study system, a coffee farm in the Soconusco region of southern Mexico, L . lecanii is characterized by clear spatial genetic structure among plots within the farm but a lack of apparent structure at smaller scales. This is consistent with dispersal dominated by highly localized transport, such as by insects or rain splash, and less dependence on longer distance dispersal such as wind transport. The study site was dominated by a few multi-locus microsatellite genotypes, and their identities and large-scale locations persist across both study years, suggesting that local epizootics (outbreaks) are initiated each wet season by residual propagules from the previous wet season, and not by long-distance transport of propagules from other sites. The index of association, a measure of linkage disequilibrium, indicates that epizootics are primarily driven by asexual, clonal reproduction, which is consistent with the apparent lack of a teleomorph in the study site and the presence of only a single mating type across the site (MAT-1-2-1). Although the same predominant clonal genotypes were found across years, a drastic difference in genotypic diversity was witnessed across two sites between the two years, suggesting that interclonal selection was occurring. In light of the dispersal limitation of L . lecanii , spatial structure may be an essential axis of management to ensure the persistence of L. lecanii and preserve the ecosystem services provided by this versatile biocontrol agent in this and similar coffee farms. Background: Ophiocordyceps sinensis (DongChong XiaCao (DCXC) in Chinese), a fungal parasite of caterpillars, is a traditional Chinese medicine. Bioactive components isolated from natural DCXC possess a wide range of pharmacological actions. Many efforts have been directed towards isolating the fungi based on culture-dependent methods for investigation of fungal diversity in order to determine the anamorph of natural DCXC and find new medicinal fungi resources, and the results have been varied. Results: In the present study, a total of 44,588 bacterial and 51,584 fungal sequences corresponding to 11,694 and 9297 putative operational taxonomic units (OTU) were respectively identified by a Roche/454-based, high throughput sequence analysis of 16S rRNA genes and ITS regions. The main bacterial groups were Proteobacteria, Acidobacteria, Bacteroidetes, Actinobacteria and Firmicutes, while the Ascomycota, Basidiomycota and Zygomycota were the main fungal phyla. Proteobacteria presented 68.4, 49.5, 38.9 and 35.6 % of all bacteria in the sclerotia, stromata, external mycelial cortices and soil, respectively. As the main fungi phyla, Ascomycota presented 21.0, 45.6 26.4 and 59.3 % in the sclerotia, stromata, external mycelial cortices and soil, respectively. Bacterial and fungal communities were more diverse in the environmental sample than in the natural DCXC sample. Microbial communities were obviously distinct in each sample. Several novel unclassifiable bacterial (10.41 %) and fungal (37.92 %) species were also detected. Conclusions: This study revealed an abundant endogenetic fungal and bacterial resources and a variety of genetic information in natural DCXC by high-throughput 454 sequencing technology. Background:Amazonian populations are exposed to diverse parasites and pathogens, including protozoal, bacterial, fungal and helminthic infections. Yet much knowledge of the immune system is based on industrialised populations where these infections are relatively rare. Aim:This study examines distributions and age-related differences in 22 measures of immune function for Bolivian forager-horticulturalists and US and European populations. Subjects and methods:Subjects were 6338 Tsimane aged 0–90 years. Blood samples collected between 2004–2014 were analysed for 5-part blood differentials, C-reactive protein, erythrocyte sedimentation rate (ESR) and total immunoglobulins E, G, A and M. Flow cytometry was used to quantify naïve and non-naïve CD4 and CD8 T cells, natural killer cells, and B cells. Results:Compared to reference populations, Tsimane have elevated levels of most immunological parameters, particularly immunoglobulins, eosinophils, ESR, B cells, and natural killer cells. However, monocytes and basophils are reduced and naïve CD4 cells depleted in older age groups. Conclusion:Tsimane ecology leads to lymphocyte repertoires and immunoglobulin profiles that differ from those observed in industrialised populations. Mycoparasitism is defined as the association of two fungi where one acts as parasite over the other. In October 2015, a powdery mildew infection was observed on Euphorbia hirta during the routine mycological survey in district Mandi the central region of Himachal Pradesh, India. During the course of microscopic examinations infection was found mixed with another unknown fungus. Upon morphological and microscopic examinations of infection on leaves, Podosphaera euphorbiae-hirtae was identified as powdery mildew fungus mixed with a mycoparasite Ampelomyces quisqualis. To the best of our knowledge, this is the first report of mycoparasitism of Ampelomyces quisquali on powdery mildew of Euphorbia hirta in India. [ABSTRACT FROM AUTHOR]

BACKGROUND Crops are often prone to both insect herbivory and disease, which necessitate multiple control measures. Ideally, an efficacious biological control agent must adequately control the target organism and not be inhibited by other biological control agents when applied simultaneously. Wheat seeds infected with the plant pathogen Fusarium culmorum were treated with Metarhizium brunneum or M. flavoviride and Clonostachys rosea individually and in combination, with the expectation to control both root-feeding insects and the pathogen. Emerging roots were evaluated for disease and then placed with Tenebrio molitor larvae, which were monitored for infection. RESULTS Plant disease symptoms were nearly absent for seeds treated with C. rosea, both individually and in combination with Metarhizium spp. Furthermore, roots grown from seeds treated with Metarhizium spp. caused significant levels of fungal infection in larvae when used individually or combined with C. rosea. However, cotreated seeds showed reduced virulence towards T. molitor when compared with treatments using Metarhizium spp. only. CONCLUSIONS This study clearly shows that seed treatments with both the entomopathogenic fungus M. brunneum and the mycoparasitic fungus C. rosea can protect plant roots from insects and disease. Visible light is an important source of energy and information for much of life on this planet. Though fungi are neither photosynthetic nor capable of observing adjacent objects, it is estimated that the majority of fungal species display some form of light response, ranging from developmental decision-making to metabolic reprogramming to pathogenesis. As such, advances in our understanding of fungal photobiology will likely reach the broad fields impacted by these organisms, including agriculture, industry and medicine. Parasites can profoundly affect host populations and ecological communities. Thus, it remains critical to identify mechanisms that drive variation in epidemics. Resource availability can drive epidemics via traits of hosts and parasites that govern disease spread., Here, we map resource-trait-epidemic connections to explain variation in fungal outbreaks ( Metschnikowia bicuspidata) in a zooplankton host ( Daphnia dentifera) among lakes. We predicted epidemics would grow larger in lakes with more phytoplankton via three energetic mechanisms. First, resources should stimulate Daphnia reproduction, potentially elevating host density. Secondly, resources should boost body size of hosts, enhancing exposure to environmentally distributed propagules through size-dependent feeding. Thirdly, resources should fuel parasite reproduction within hosts., To test these predictions, we sampled 12 natural epidemics and tracked edible algae, fungal infection prevalence, body size, fecundity and density of hosts, as well as within-host parasite loads., Epidemics grew larger in lakes with more algal resources. Structural equation modelling revealed that resource availability stimulated all three traits (host fecundity, host size and parasite load). However, only parasite load connected resources to epidemic size. Epidemics grew larger in more dense Daphnia populations, but host density was unrelated to host fecundity (thus breaking its link to resources)., Thus, via energetic mechanisms, resource availability can stimulate key trait(s) governing epidemics in nature. Clonostachys rosea is a mycoparasitic fungus that can control several important plant diseases. Here, we report on the genome sequencing of C. rosea and a comparative genome analysis, in order to resolve the phylogenetic placement of C. rosea and to study the evolution of mycoparasitism as a fungal lifestyle. The genome of C. rosea is estimated to 58.3 Mb, and contains 14,268 predicted genes. A phylogenomic analysis shows that C. rosea clusters as sister taxon to plant pathogenic Fusarium species, with mycoparasitic/saprotrophic Trichoderma species in an ancestral position. A comparative analysis of gene family evolution reveals several distinct differences between the included mycoparasites. Clonostachys rosea contains significantly more ATP-binding cassette (ABC) transporters, polyketide synthases, cytochrome P450 monooxygenases, pectin lyases, glucose-methanol-choline oxidoreductases, and lytic polysaccharide monooxygenases compared with other fungi in the Hypocreales. Interestingly, the increase of ABC transporter gene number in C. rosea is associated with phylogenetic subgroups B (multidrug resistance proteins) and G (pleiotropic drug resistance transporters), whereas an increase in subgroup C (multidrug resistance-associated proteins) is evident in Trichoderma virens. In contrast with mycoparasitic Trichoderma species, C. rosea contains very few chitinases. Expression of six group B and group G ABC transporter genes was induced in C. rosea during exposure to the Fusarium mycotoxin zearalenone, the fungicide Boscalid or metabolites from the biocontrol bacterium Pseudomonas chlororaphis. The plant-pathogenic fungus Sclerotinia sclerotiorum has a broad host range and a worldwide distribution. Boscalid, an inhibitor of succinate dehydrogenase in the electron transport chain of fungi, is highly effective in controlling sclerotinia stem rot caused by S. sclerotiorum. The current study characterized the S. sclerotiorum boscalid-resistant ( BR) mutants obtained by fungicide induction. Among the bioactive fungicides against S. sclerotiorum, cross-resistance was not detected between boscalid and dimethachlon, fluazinam or carbendazim; positive cross-resistance was detected between boscalid and carboxin; and negative cross-resistance was detected between boscalid and kresoxim-methyl. Compared to their parental isolates, BR mutants had slower radial growth, no ability to produce sclerotia, lower virulence and oxalic acid content but higher mycelial respiration and succinate dehydrogenase ( SDH) activity. Moreover, BR mutants had decreased sensitivity to salicylhydroxamic acid ( SHAM) but not to oxidative stress. All the results indicated that the risk of resistance to boscalid in S. sclerotiorum is low to moderate. DNA sequence analysis showed that all of the BR mutants had the same point mutation A11V ( GCA to GTA) in the iron sulphur protein subunit ( SDHB). Interestingly, expression of the cytochrome b ( cytb) gene was reduced to different degrees in the BR mutants, and this might be correlated with the negative cross-resistance between boscalid and kresoxim-methyl. Within-host competition is predicted to drive the evolution of virulence in parasites, but the precise outcomes of such interactions are often unpredictable due to many factors including the biology of the host and the parasite, stochastic events and co-evolutionary interactions. Here, we use a serial passage experiment ( SPE) with three strains of a heterothallic fungal parasite ( Ascosphaera apis) of the Honey bee ( Apis mellifera) to assess how evolving under increasing competitive pressure affects parasite virulence and fitness evolution. The results show an increase in virulence after successive generations of selection and consequently faster production of spores. This faster sporulation, however, did not translate into more spores being produced during this longer window of sporulation; rather, it appeared to induce a loss of fitness in terms of total spore production. There was no evidence to suggest that a greater diversity of competing strains was a driver of this increased virulence and subsequent fitness cost, but rather that strain-specific competitive interactions influenced the evolutionary outcomes of mixed infections. It is possible that the parasite may have evolved to avoid competition with multiple strains because of its heterothallic mode of reproduction, which highlights the importance of understanding parasite biology when predicting disease dynamics. Claviceps purpurea is an ovarian parasite infecting grasses (Poaceae) including cereals and forage plants. This fungus produces toxic alkaloids and consumption of contaminated grains can cause ergotism in humans and other mammals. Recent molecular genetics studies have indicated that it included three cryptic species (G1, G2, G3). In this study, reproductive isolation amongst these groups and among material from Phragmites and Molinia was tested using gene flow statistics for five polymorphic loci, and to support these data, phylogenetic affiliations based on gene trees and a multigene phylogeny were used. The four recognized species are characterized based on morphology and host spectrum and formal taxonomic names are proposed. Claviceps purpurea sensu stricto (G1 group) represents a typical rye ergot, but infects various other grasses. Typical hosts of Claviceps humidiphila (new name for G2 species), like Phalaris arundinacea, belong to grasses preferring humid locations. Claviceps spartinae (G3) is specific to chloridoid grasses from salt barches. The material from Phragmites and Molinia can be authenticated with the species Claviceps microcephala for which the new name Claviceps arundinis is proposed here. Despite efforts of eradication and sanitation, Phytophthora ramorum persists in the United States and abroad. Fungicides have limited effectiveness, but there are concerns that they may only inhibit pathogen growth and hasten resistance development after repeated fungicide applications. Biological control is an active control measure that can work continuously as long as the agent is alive and active. The goal of this study was to examine whether Trichoderma spp. have the potential as a biological control agent against P. ramorum . Sixteen Trichoderma spp. isolates were screened for mycoparasitism of P. ramorum in a dual culture assay. The different Trichoderma spp. isolates demonstrated variable mycoparasitic activities with some isolates showing no activity while others completely eliminated the pathogen after 4 weeks. Seven isolates of T. asperellum were consistent among replicated trials in eliminating recovery of P. ramorum from the exposed agar plugs and preventing leaf disk necrosis. Further testing of six T. asperellum isolates against two different P. ramorum isolates (A1 and A2 mating types) resulted in the same high level of mycoparasitic activity. Soil assays involving P. ramorum -infested potting mix and selected Trichoderma spp. isolates demonstrated that two isolates (04-22 and 02-64) were consistent among the repetitions to eliminate P. ramorum propagules to non-detectable levels. Based on these results, specific T. asperellum isolates have the potential to remediate P. ramorum -infested soil and have the potential to be developed into a commercially-viable product. Here we describe the first application of transient gene silencing in Saprolegnia parasitica, a pathogenic oomycete that infects a wide range of fish, amphibians, and crustaceans. A gene encoding a putative tyrosinase from S. parasitica, SpTyr, was selected to investigate the suitability of RNA-interference (RNAi) to functionally characterize genes of this economically important pathogen. Tyrosinase is a mono-oxygenase enzyme that catalyses the O -hydroxylation of monophenols and subsequent oxidation of O -diphenols to quinines. These enzymes are widely distributed in nature, and are involved in the melanin biosynthesis. Gene silencing was obtained by delivering in vitro synthesized SpTyr dsRNA into protoplasts. Expression analysis, tyrosinase activity measurements, and melanin content analysis confirmed silencing in individual lines. Silencing of SpTyr resulted in a decrease of tyrosinase activity between 38 % and 60 %, dependent on the level of SpTyr -expression achieved. The SpTyr -silenced lines displayed less pigmentation in developing sporangia and occasionally an altered morphology. Moreover, developing sporangia from individual silenced lines possessed a less electron dense cell wall when compared to control lines, treated with GFP-dsRNA. In conclusion, the tyrosinase gene of S. parasitica is required for melanin formation and transient gene silencing can be used to functionally characterize genes in S. parasitica . Fungal mycoparasitism-fungi parasitizing other fungi-is a common lifestyle in some basal lineages of the basidiomycetes, particularly within the Tremellales. Relatively nonaggressive mycoparasitic fungi of this group are in general highly host specific, suggesting cospeciation as a plausible speciation mode in these associations. Species delimitation in the Tremellales is often challenging because morphological characters are scant. Host specificity is therefore a great aid to discriminate between species but appropriate species delimitation methods that account for actual diversity are needed to identify both specialist and generalist taxa and avoid inflating or underestimating diversity. We use the Biatoropsis- Usnea system to study factors inducing parasite diversification. We employ morphological, ecological, and molecular data-methods including genealogical concordance phylogenetic species recognition (GCPSR) and the general mixed Yule-coalescent (GMYC) model-to assess the diversity of fungi currently assigned to Biatoropsis usnearum. The degree of cospeciation in this association is assessed with two cophylogeny analysis tools (ParaFit and Jane 4.0). Biatoropsis constitutes a species complex formed by at least seven different independent lineages and host switching is a prominent force driving speciation, particularly in host specialists. The new species Miriquidica invadens Hafellner, Obermayer & Tretiach is described and reported from Austria, Bulgaria, Italy, Switzerland, France and Spain. From a morphological point of view the species belongs to the M. griseoatra group, in which it is, so far, the only constantly lichenicolous taxon. Chemically it is characterized by the regular presence of miriquidic acid, stictic acid, and constictic acid (in variable amounts). It starts its ontogenetic life cycle as an obligate youth parasite on Sporastatia, usually S. polyspora.Morpho-anatomical treatments of other species of the Miriquidica griseoatra group occurring in Europe, based on the examination of type material, are also presented. According to the characters exhibited by the lectotype of Lecidea contigua var. griseoatra Flot., M. griseoatra (Flot.) Hertel & Rambold is the correct name for a montane species with subdivided areoles and immersed, laterally adnate apothecia with a pigmented hypothecium. Specimens exhibiting this set of characters were usually named M. obnubila auct. According to the characters observed on the lectotype of Lecidea (Biatora) obnubila Th. Fr. & Hellb., M. obnubila (Th. Fr. & Hellb.) Hertel & Rambold is a species with sessile apothecia with a hyaline hypothecium. Lecidea subplumbea Anzi does not constitute a heterotypic synonym of Lecidea griseoatra (Flot.) Schaer. but is the legitimate name at the species level for Miriquidica griseoatra sensu auct., and therefore M. subplumbea (Anzi) Cl. Roux is the correct name for that taxon. Lecidea inserena Nyl. is confirmed as a heterotypic synonym of Lecidea subplumbea Anzi. Lecidea plumbea Garov. ex A. Massal. is an earlier heterotypic synonym of Miriquidica limitata Hertel & Rambold and, therefore, the new combination M. plumbea (Garov. ex A. Massal.) Hafellner, Obermayer & Tretiach is introduced.Lectotypes are designated for Lecidea contigua var. griseoatra Flot., Lecidea inserena Nyl., Lecidea obnubila Th. Trichoderma spp. are used for biocontrol of several plant pathogens. However, their efficient interaction with the host needs to be accompanied by production of secondary metabolites and cell wall-degrading enzymes. Three parameters were evaluated after interaction between four Trichoderma species and plant-pathogenic fungi: Fusarium solani, Rhizoctonia solani and Sclerotinia sclerotiorum. Trichoderma harzianum and T. asperellum were the most effective antagonists against the pathogens. Most of the Trichoderma species produced toxic volatile metabolites, having significant effects on growth and development of the plant pathogens. When these species were grown in liquid cultures with cell walls from these plant pathogens, they produced and secreted β-1,3-glucanase, NAGAse, chitinase, acid phosphatase, acid proteases and alginate lyase. Abstract: ‘Candidatus Cardinium’ is an intracellular endosymbiont or parasite frequently occurring in invertebrates including mites and ticks. In this work we report Cardinium bacteria in Astigmata mites and explore their incidence in synanthropic species. Amplification of a 776bp bacterial 16S rRNA gene fragment, using specific primers, enabled identification of closely related Cardinium sequences in 13 laboratory-reared populations of mites. In addition, Cardinium sequences were identified in three wild mite populations. Large scale screening of these populations showed 100% prevalence of Cardinium, representing the highest incidence compared to other major Chelicerate groups. [Copyright &y& Elsevier]

In Rio de Janeiro State (Brazil), guava decline is a complex disease in which parasitism by Meloidogyne enterolobii predisposes Fusarium solani-immune trees to extensive root decay caused by this fungus. On the shoot, the symptoms include chlorosis, wilting, scorching of leaf margins and leaf drop, yield reduction, and plant death within months. Since in many States M. enterolobii-infested orchards have been decimated with the same symptoms, this study aimed to assess the incidence of guava decline nationwide. Root samples from declining, nematode-infested orchards were received from Southern, Northeastern and Midwestern Brazil. Of the 682 root fragments processed for fungal isolation, about half were positive for Fusarium sp., and only 5% were positive for other fungi. Sixty-nine Fusarium sp. isolates were established as representatives of those Brazilian regions. To expedite the evaluation of virulence to guava of all these isolates, a growth chamber assay was developed using as reference 11 Fusarium sp. isolates whose virulence to guava plants had been assessed in previous microplot experiments. Of the 69 Fusarium sp. isolates assessed, 87% - from all Brazilian regions - were virulent to stem cuttings used in the growth chamber assay, with 42% of these being classified as highly virulent. All virulent isolates were preliminarily identified as F. solani through morphological characterization. To confirm this, 12 representatives from different Brazilian regions were selected for molecular identification through ITS4 and ITS5 gene sequencing, which confirmed their identity as F. solani. The feasibility of short-read sequencing for genomic analysis was demonstrated for Fibroporia radiculosa, a copper-tolerant fungus that causes brown rot decay of wood. The effect of read quality on genomic assembly was assessed by filtering Illumina GAIIx reads from a single run of a paired-end library (75-nucleotide read length and 300-bp fragment size) at three different stringency levels and then assembling each data set with Velvet. A simple approach was devised to determine which filter stringency was "best." Venn diagrams identified the regions containing reads that were used in an assembly but were of a low-enough quality to be removed by a filter. By plotting base quality histograms of reads in this region, we judged whether a filter was too stringent or not stringent enough. Our best assembly had a genome size of 33.6 Mb, an N50 of 65.8 kb for a k-mer of 51, and a maximum contig length of 347 kb. Using GeneMark, 9,262 genes were predicted. TargetP and SignaiP analyses showed that among the 1,213 genes with secreted products, 986 had motifs for signal peptides and 227 had motifs for signal anchors. Blast2GO analysis provided functional annotation for 5,407 genes. We identified 29 genes with putative roles in copper tolerance and 73 genes for lignocellulose degradation. A search for homologs of these 102 genes showed that F. radiculosa exhibited more similarity to Postia placenta than Serpula lacrymans. Fungi in the genus Hypomyces are mycoparasites of various fungal groups. Studies within this genus have dealt primarily with taxonomic descriptions, and no studies have been published on the population biology of these fungi in natural ecosystems. In this study, genetic structure within and among six California (CA) bolete-infecting H. microspermus populations were analyzed with amplified fragment length polymorphism (AFLP). Four of the populations were collected south of the transverse mountain ranges of southern CA, whereas the other populations were from the central coast of CA (Pismo Beach) and from northern CA (Humboldt County). Thirty-nine genotypes were found among 68 isolates based on 30 AFLP markers. This was a higher degree of genotypic diversity than expected because no sexual fruiting bodies were observed in the field, suggesting that sexual reproduction occurs infrequently. These observations were also supported by multilocus disequilibrium analyses. Overall the populations were significantly differentiated (θ = 0.321, P < 0.001), and most population pairwise Fst values were also significantly different. The populations sampled also fit an isolation by distance model based on a Mantel test (P < 0.001). Asexuality has major theoretical advantages over sexual reproduction. Nevertheless, obligately asexual metazoan lineages seldom endure over evolutionary time. The Red Queen hypothesis posits that their limited capacity to generate genetic novelty leads to extermination by rapidly evolving parasites and pathogens. At first glance, rotifers of the class Bdelloidea appear to contradict this view: they have reproduced asexually for over 30 Myr without being overwhelmed by parasites. However, there are special ecological conditions under which Red Queen models can accommodate this unusual outcome. If hosts disperse rapidly within a structured metapopulation during a parasite-free life stage, then in principle they can become spatiotemporally decoupled from coevolving antagonists, and persist without sex. Intriguingly, bdelloid rotifers form dormant propagules when desiccated, which disperse easily by wind. In previous experiments, 7 days of desiccation and wind dispersal removed a fungal parasite from populations of one bdelloid species, allowing them to disperse independently. Here, I extend this finding to two additional bdelloid species and five more fungal parasites, and demonstrate its robustness under various desiccation regimes, and in the presence of multiple parasites. Abstract: Coniothyrium minitans is a mycoparasite and a promising biocontrol agent of Sclerotinia sclerotiorum, the causal of Sclerotinia stem rot of oilseed rape. In order to assess whether soil application of C. minitans can be combined with fertilizer application during cultivation of oilseed rape, a study was conducted to determine the compatibility of C. minitans with commercial N–P–K compound fertilizer. Results showed that the compound fertilizer at concentrations of 0.1–10% inhibited conidial germination and mycelia growth of C. minitans in a dosage–response manner in vitro. Simultaneous application of C. minitans and the compound fertilizer at various concentrations significantly (P <0.05) reduced the number of apothecia produced by sclerotia of S. sclerotiorum in both pot and field plot experiments. No negative effects of the compound fertilizer on the ability of C. minitans to infect sclerotia of S. sclerotiorum in vitro or to suppress carpogenic germination of sclerotia of S. sclerotiorum were observed. It is demonstrated that C. minitans is compatible with the compound fertilizer when applied at planting of oilseed rape. Abstract: Microsporidia are fungi-related obligate intracellular parasites with a highly reduced and compact genome, as for Encephalitozoon species which harbor a genome smaller than 3 Mbp. Genome compaction is reflected by high gene density and, for larger microsporidian genomes, size variation is due to repeat elements that do not drastically affect gene density. Furthermore, these pathogens present strong host dependency illustrated by extensive gene loss. Such adaptations associated with genome compaction induced gene size reduction but also simplification of cellular processes such as transcription. Understanding the mechanisms responsible for divergence and specialization of pathogens on different hosts is of fundamental importance, especially in the context of the emergence of new diseases via host shifts. Temporal isolation has been reported in a few plants and parasites, but is probably one of the least studied speciation processes. We studied whether temporal isolation could be responsible for the maintenance of genetic differentiation among sympatric populations of Ampelomyces, widespread intracellular mycoparasites of powdery mildew fungi, themselves plant pathogens. The timing of transmission of Ampelomyces depends on the life cycles of the powdery mildew species they parasitize. Internal transcribed spacer sequences and microsatellite markers showed that Ampelomyces populations found in apple powdery mildew ( Podosphaera leucotricha) were genetically highly differentiated from other Ampelomyces populations sampled from several other powdery mildew species across Europe, infecting plant hosts other than apple. While P. leucotricha starts its life cycle early in spring, and the main apple powdery mildew epidemics occur before summer, the fungal hosts of the other Ampelomyces cause epidemics mainly in summer and autumn. When two powdery mildew species were experimentally exposed to Ampelomyces strains naturally occurring in P. leucotricha in spring, and to strains naturally present in other mycohost species in autumn, cross-infections always occurred. Thus, the host-related genetic differentiation in Ampelomyces cannot be explained by narrow physiological specialization, because Ampelomyces were able to infect powdery mildew species they were unlikely to have encountered in nature, but instead appears to result from temporal isolation. Fusarium spp. are economically important crop pathogens and causal agents of Fusarium head blight (FHB) of cereals worldwide. Of the FHB pathogens, Fusarium graminearum 3-acetyldeoxynivalenol (3-ADON) and 15-acetyldeoxynivalenol (15-ADON) are the most aggressive mycotoxigenic chemotypes, threatening food and feed quality as well as animal and human health. The objective of the study was to evaluate host specificity and fungal-fungal interactions of Sphaerodes mycoparasitica- a recently described mycoparasite - with F. graminearum 3- and 15-ADON strains by employing in vitro, microscopic and PCR techniques. Results obtained in this study show that the germination of mycoparasite ascospore in the presence of F. graminearum 3- and 15-ADON filtrates was greatly improved compared with Fusarium proliferatum and Fusarium sporotrichioides filtrates, suggesting a compatible interaction. Using quantitative real-time PCR with Fusarium-specific (Fg16N) and trichothecene Tri5 (Tox5-1/2)-specific primer sets, S. mycoparasitica was found to reduce the amount of F. graminearum 3-ADON and 15-ADON DNAs under separate coinoculation assays. Sphaerodes mycoparasitica was not only able to germinate in the presence of F. graminearum filtrates, but also established biotrophic mycoparasitic relations with two F. graminearum chemotypes and suppressed Fusarium growth. Abstract: The aims of this study were to test the action of the fungal extract of Pochonia chlamydosporia (VC4) on the hatching of cyathostomin eggs plated in Petri dishes containing 2% water-agar (2% WA) and its enzymatic activity in fecal cultures, in two experimental assays (A and B). The fungus P. chlamydosporia (VC4) was cultured in Erlenmeyer flasks (250ml) containing 50ml of liquid minimal medium supplemented with 0.2% gelatin for production of the crude enzymatic extract. Approximately 1kg of fresh feces was collected directly from the rectum of crossbred horses naturally infected with cyathostomins. The fecal material was used to obtain eggs and prepare fecal cultures. For assay A, one thousand eggs were plated on 4.5cm diameter Petri dishes together with 5ml of VC4 fungal filtrate and incubated at 26°C in the dark for 24h. The control group consisted of 1000 eggs in Petri dishes containing 10ml of distilled water, which were incubated under the same conditions. After 24h, the total number of cyathostomin larvae present in each plate of the treated and control groups was counted. For assay B, about 20g of feces were added with 10ml of fungal extract of P. chlamydosporia (VC4) and incubated at 26°C for 8 days. Third stage larvae (L3) were recovered at the end of this period. Significant difference (p <0.01) was found for the number of larvae between the treated group and the control at end of assay A. A 72.8% reduction in the hatching of cyathostomin eggs was found in the plates of the treated group compared with the control group. At the end of 8 days, the fungal extract of P. chlamydosporia (VC4), in assay B, was effective in reducing the number of L3 cyathostomins in the treated group by 67.0% compared with the control group. Significant difference (p <0.01) was found between the means of L3 recovered from the treated group and the control group. The results of this work showed that crude enzymatic extract of P. chlamydosporia (VC4) was effective in reducing hatching of cyathostomin eggs and therefore could be used as a biological control agent of this nematode. Wood decay activity of Omphalotus guepiniformis, one of the most frequently occurring fruiting bodies on beech coarse woody debris in cool temperate forests in Japan, was estimated in situ by chronosequence with a five decay class system. The decay columns of O. guepiniformis increased from decay class 1 to decay class 2, where they occupied 20.2% of the total area of cross sections, and was estimated to be a dominant basidiomycete. The decay columns of O. guepiniformis decreased after decay class 2 and were not detected in decay class 5. The relative density of the decay columns of O. guepiniformis decreased to 0.33 g cm in decay class 2 (58.9% of fresh beech wood) but did not decrease thereafter. The lignocellulose index (LCI) of the decay columns of O. guepiniformis slightly decreased during the decay process while remaining in the range of white-rot. In contrast, the decay columns of microfungi increased in the later stages of decomposition and LCI of these decay columns decreased significantly alongside the decay process. These results suggest that O. guepiniformis has an important role in simultaneous decomposition of acid-unhydrolyzable residue (AUR, Klason lignin) and holocellulose in the early stages of beech log decomposition, while holocellulose selective decomposition by microfungi may occur in the late stages of decomposition. Plant biotrophs often establish highly specialized and localized interaction sites where sustained nutrient exchange occurs. Increased plant nuclear DNA ploidy at or adjacent to these sites has now been reported for a diverse set of interactions, including those with fungal and bacterial symbionts and parasitic fungi and nematodes. Also, novel regulators of induced endoreduplication have recently been identified. When localized host endoreduplication is reduced, so too is the growth and/or development of the biotroph, suggesting endoreduplication supports the enhanced metabolic demands imposed by these interactions. Transcriptome analyses support this function and further identify specific ploidy-impacted processes. Remarkably, notwithstanding differences in time scales, the ploidy-impacted processes are consistent with the Gene Balance Hypothesis, which can also be used to predict effector targets. As effector influence may diminish with enhanced ploidy, these interaction sites may be uniquely suited to identify effector-impacted processes as well as elucidate endocycle regulation and function. We are on the verge of a flood of new genome data from a plethora of plant pathogens. Although the data are just beginning to be analysed systematically and to be published, there are emerging patterns associated with pathogenicity and biotrophy. The expansion of protein families with functions required for host infection is found in most pathogens; in addition, we see specific adaptations of the biotrophs, as the reduction of proteins involved in plant cell-wall degradation, or a specialization in carbon utilization that avoids plant responses. The expansion of genomes by repeated elements and the resulting genome dynamics are thought to be one of the driving forces for the fast evolution of pathogenicity factors, resulting in an adaptive advantage to the changing hostile environment of the plant host. Plants can establish intracellular interactions with symbiotic as well as pathogenic microbes. Such intracellular accommodation of microbes always involves the formation of a host membrane compartment—the interface between the cytoplasm of the host and the microbe. These are the so-called perimicrobial compartments. In this review we will focus on the rhizobial legume symbiosis in which the microbes are hosted in organelle-like compartments, which are named symbiosomes. The signaling events leading to infection and symbiosome formation are discussed. Further the role of the host cell endomembrane system in symbiosome formation is described and compared with the processes involved in arbuscule and haustorium formation during the interaction of plants and biotrophic fungi. Mycoparasitic Trichoderma spp. act as potent biocontrol agents against a number of plant pathogenic fungi, whereupon the mycoparasitic attack includes host recognition followed by infection structure formation and secretion of lytic enzymes and antifungal metabolites leading to the host's death. Host-derived signals are suggested to be recognized by receptors located on the mycoparasite's cell surface eliciting an internal signal transduction cascade which results in the transcription of mycoparasitismrelevant genes. Heterotrimeric G proteins of fungi transmit signals originating from G-protein-coupled receptors mainly to the cAMP and the MAP kinase pathways resulting in regulation of downstream effectors. The order Meliolales comprises two families, namely, Armatellaceae and Meliolaceae. Except the genera Endomeliola and Pauhia, India represents rest of the nine genera of this group. The family Armatellaceae includes two genera, namely, Armatella and Basavamyces. The family Meliolaceae includes seven genera: Amazonia, Appendiculella, Asteridiella, Ectendomeliola, Irenopsis, Meliola and Prataprajella. All these nine genera represent 613 species and infraspecific taxa known till the year 2006, infected 766 host plants belonging to 349 host genera distributed among 104 families. All the host families and the fungal genera are arranged alphabetically with their corresponding parasite and the host plant. Tremella encephala is a haustorial biotrophic fungal parasite which grows both in the yeast and mycelial form during its life cycle. Biology of such non-commercial parasitic and dimorphic fungi is poorly known. We examined the effects of different nitrogen and carbon sources on growth and morphogenetic switch in T. encephala, as well as its host-specificity, host-parasite interaction and the effect of media on parasitism by culturing it alone, together with the known host Stereum sanguinolentum and with the possible host Stereum hirsutum on two different media. Tremella encephala utilized many organic and inorganic nitrogen sources and showed high variation in cellular nitrogen concentrations. Only glucose, glycerol and D-mannitol were exploited as the sole sources of carbon. Remarkable variation in dimorphism was observed between and within the strains. Parasitic interaction was not manifested in this laboratory study. Tremella encephala is not a strictly obligate parasite. Arceuthobium americanum Nuttal ex Engelmann in Gray (lodgepole pine dwarf mistletoe) causes significant losses to the timber industry. The fungus Caliciopsis arceuthobii (Peck) Barr shows high specificity to Arceuthobium spp., and therefore infection of A. americanum was monitored for 4 years to determine the effect of this fungus on fruit production. The contribution of stand-level and locally produced inoculum in the infection process was also studied to better understand the epidemiology of the pathogen. Over the period of the study, C. arceuthobii caused an average annual fruit reduction of 58%. When the contribution of local and stand-level inoculum was modelled, the results suggested that on A. americanum infections with ≥ 4 ascospore-producing immature fruit, more than half of the new C. arceuthobii infections on the same plant are derived from locally produced ascospores. The effect of the fungus on the host suggests that it has potential as a biological control agent for A. americanum, and further studies are needed to attempt to induce inoculum production in culture. Aims: To characterize the interaction of Sclerotinia sclerotiorum and S. minor with strains of the mycoparasite and commercial biocontrol agent Coniothyrium minitans using novel perfusion chamber gasket co-culture. Methods and Results: Sclerotinia were cultured in perfusion chamber gaskets and then flooded with Coniothyrium conidia. After germination, Coniothyrium failed to show any form of directed growth, making contact with Sclerotinia hyphae in a random manner. In turn, some Coniothyrium hyphae coiled round Sclerotinia counterparts and although no intracellular growth was observed, Coniothyrium proliferated, while the hyphae of Sclerotinia became vacuolated and lost the cytoplasm. When co-cultures of Sclerotinia with Coniothyrium were flooded with FITC-lectins, small difference in fluorescence between the fungi was found with FITC-Con A suggesting that cell walls of both the species exposed mannose. In contrast, Coniothyrium fluoresced poorly in comparison with Sclerotinia when FITC-wheat germ agglutinin was used, indicating a marked paucity of N-acetylglucosamine exposure by cell walls of Coniothyrium, hence reduced exposure to chitinolytic enzyme action. Three new species of Laboulbenia occurring on endogean Carabidae are described. These are L. lucifuga, parasitic on Winklerites spp. from Greece, L. magrinii, parasitic on Typloreicheia spy. from Italy, Reicheia spp. from Italy and Corsica and L. vailatii, parasitic on Coecoparvus spp. from Greece. Three recent collections of Doassansiopsis from western Cameroon are assessed taxonomically. Doassansiopsis caldesiae M. Piątek & Vánky is described as a new species from infected leaves of Caldesia reniformis (D. Don) Makino. Its diagnostic characters are flat, nonthickened sori with spore balls as blackish, slightly elevated dots, more or less globoid spores, conspicuous cortical sterile cells and parasitism on Caldesia reniformis of family Alismataceae. The species is compared to another Doassansiopsis species on host plants belonging to family Alismataceae. Doassansiopsis tomasii Vánky is described from two localities on Nymphaea nouchali Burm.f. var. caerulea (Savigny) Verdc. (Nymphaeaceae) Which represents the first report of this smut from Cameroon and western Africa. Similarities between this species and Doassansiopsis nymphaeae (Syd. & P. Syd.) Thirum. and D. ticonis M. Piepenbr. are outlined and the global distribution of the three taxa is mapped. Microsporosis is one of most frequent cases of zoonosis. We describe a case of family spreading microsporosis and it is a big probability that infection was transmitted from the new buy cat. Typical mycoses lesion occurs on smooth skin and in the hair skin in the children and only on a body in adults. Clinically in cat no changes on hair coat and any skin lesion was observed. Different interactions between soil fungi competing in the rhizosphere with each other are necessary to understand their influence on plant growth and health. The interactions between the ectomycorrhizal (ECM) fungus Laccaria laccata and soil saprotrophic fungi (T. harzianum, T. virens) were studied by transmission electron microscopy, and by gold cytochemistry to assess the potential role of cell wall lytic enzymes in mycoparasitism. Anti-β-1,3-glucan antibody, WGA/ovomucoid-gold complex and PATAg test were used to localize β-1,3-glucan, chitin and polysaccharides. Cytoplasm disorganisation of the saprotrophic fungi occurred concurrently with dissolution of β-1,3-glucan in walls of hyphae and conidia of the saprotrophic fungi. Then digestion of polysaccharides and chitin of colonised fungal structures occurred. The studies suggest sequential contribution of cell wall lytic enzymes and importance of disturbing the host's cell integrity during mycoparasitism.
