Filamentous fungi represent a diverse group of microorganisms, encompassing industrially significant species such as Aspergillus oryzae and Aspergillus niger , as well as biologically active, edible, and medicinal fungi such as Ganoderma lucidum (G. lucidum) and Auricularia auricula. The morphology of filamentous fungal mycelia is crucial for species development, natural product synthesis, and environmental adaptation. Therefore, accurate qualitative and quantitative assessment of mycelial morphology is imperative. Mathematical modeling is a useful tool for investigating microbial morphology, aiding in experimental research on mycelial development. However, owing to the inherent complexity of mycelial development, it cannot be fully captured by existing models. This paper presents a two-dimensional, lattice-free mathematical model using MATLAB (R2023b). The model incorporates hyphal tip extension, branching, anastomosis, and energy translocation, while introducing continuous growth, range anastomosis, and variable growth length behaviors. These enhancements allow more accurate simulation of real mycelial growth patterns. The model was used to simulate and optimize G. lucidum growth, and its accuracy was verified. Our findings indicate that the proposed model effectively simulates filamentous fungi mycelial growth and provides a generalized framework for describing the morphology of other filamentous fungi. Many microbial pathogens form biofilms, assemblages of polymeric compounds that play a crucial role in establishing infections. The biofilms of Fusarium species also contribute to high antifungal resistance. Using our collection of 29 clinical Fusarium isolates, we focused on characterizing differences in thermotolerance, anaerobic growth, and biofilm formation across four Fusarium species complexes commonly found in clinical settings. We investigated the role of carbon sources, temperature, and fungal morphology on biofilm development. Using fluorescence microscopy, we followed the stages of biofilm formation. Biofilms were screened for sensitivity/resistance to the antifungals voriconazole (VOR), amphotericin B (AmB), and 5-fluorocytosine (5-FC). Our findings revealed generally poor thermotolerance and growth under anaerobic conditions across all Fusarium species. VOR was more effective than AmB in controlling biofilm formation, but the combination of VOR, AmB, and 5-FC significantly reduced biofilm formation across all species. Additionally, Fusarium biofilm formation varied under non-glucose carbon sources, highlighting the species' adaptability to different nutrient environments. Notably, early stage biofilms were primarily composed of lipids, while polysaccharides became dominant in late-stage biofilms, suggesting a dynamic shift in biofilm composition over time. The Ganoderma lucidum complex includes fungi with similar morphologies but which are thought to represent different species. The lack of available type material and associated absence of multiple locus sequence data has complicated identification of these fungi. The aim of this study was to clarify the identity of the laccate Ganoderma species occurring in Finland by inferring a phylogeny using DNA sequences from available boreal-temperate material. DNA from Finnish isolates together with an older G. lucidum isolate originating from the United Kingdom was sequenced, and the morphological features of the Finnish specimens were examined. The phylogenetic analysis of the internal transcribed spacer region (ITS), the elongation factor 1-α (tef1), RNA polymerase II subunit (rpb2), and partial β-tubulin (β-tub) genes revealed that the G. lucidum isolate from the United Kingdom did not fall within a well-supported clade with other G. lucidum sequences or related species. The Finnish isolates were closely related to the G. tsugae lineage in tef1, rpb2, and β-tub phylogenies. However, G. tsugae appears morphologically distinct from the Finnish material. The results suggest that G. tsugae, or a species phylogenetically closely related to it, may occur in Finland. But further investigation into the relationship between G. tsugae and G. lucidum from Europe will be needed to clarify the identity of the laccate Ganoderma species in Finland. Stipitate Thelephorales are basidiomycetous, mostly hydnoid, ectomycorrhizal fungi. Some species have declined considerably, and some are threat-listed as vulnerable or endangered. These ecological concerns require a well-resolved taxonomy to understand diversity in this group of fungi and facilitate conservation. However, phylogenetic studies have mostly neglected Southern Hemisphere representatives. This study examines the fleshy species of stipitate Thelephorales from native forests in Australia and New Zealand, using morphological analyses and phylogenetic analyses of nuc rDNA internal transcribed spacer region ITS1-5.8S-ITS2 (ITS barcode) and D1–D2 domains at the 5′ end of nuc 28S rDNA (28S) sequences amplified from DNA isolated from fungarium collections and environmental DNA (eDNA) sequences from the Australian Microbiome initiative. Five new species, Sarcodon austrofibulatus, Hydnellum gatesiae, H. nothofagacearum, H. pseudoioeides, and H. variisporum, are described, Sarcodon carbonarius is transferred to Neosarcodon, and a key is provided for the six named species in the region. Boletopsis and Neosarcodon are reported from Australia for the first time based on detections from eDNA in soil samples taken from native forests. The Australasian species of Hydnellum occupy a highly derived position with the phylogeny of the genus, the members of which are otherwise all from the Northern Hemisphere, suggestive of a long-distance dispersal origin for the Australasian species. The treatment of carbendazim-contaminated effluents is a challenge because of its complex composition and toxicity. A promising solution lies in biodegradation and the fungus Actinomucor elegans LBM 290 shows significant potential in this regard. Thus, the aim of this study was to biodegrade MBC by A. elegans LBM 290 in a liquid medium addressing the changes in the fungal morphology and protein production. The fungus A. elegans LBM 290 efficiently remove the fungicide carbendazim, with 86.6% removal within 8 days. This degradation is a combination of biodegradation (24.54%) and adsorption (62.08%). Exposure to carbendazim negatively affected the fungus, causing a decrease in biomass and morphological changes. Proteomic analysis revealed the fungal response to carbendazim stress through increased production of Cu–Zn superoxide dismutase, an antioxidant enzyme that combats oxidative stress, and the presence of a G protein subunit, suggesting participation in stress signaling pathways. This study reported the synthesis and characterization of chitosan-copper nanoparticles (Ch-CuNPs) using a 1% copper sulfate solution in 0.2% w/v chitosan. The Ch-CuNPs, displaying a stable brick-red hue, showed an absorption peak at 572 nm, indicative of monodisperse nanoparticle formation and surface plasmon resonance. X-ray diffraction confirmed the face-centered cubic structure with peaks at 36.78°, 43.38°, 50.56°, and 74.26°, and an average particle size of 87–89 nm. FTIR analysis showed interactions between chitosan and copper, particularly around 3370 –3226 cm⁻¹, 1633 cm⁻¹, and 680 cm⁻¹. In vitro assays revealed that Ch-CuNPs inhibited Macrophomina phaseolina growth by 18–71% at 0.03–0.09% concentrations, achieving complete inhibition at 0.12–0.15%, with PCA analysis confirming that growth peaked at lower concentrations and sharply declined at higher levels. Ch-CuNPs also altered fungal morphology and enzyme activity, with notable degradation at higher concentrations. The Cu uptake by the fungus peaked at 29.9% with 0.03% Ch-CuNPs and decreased at higher concentrations. FTIR analysis showed shifts and disappearance of peaks in fungal biomass treated with Ch-CuNPs, indicating molecular interactions and potential structural changes. Abstract\nHIGHLIGHTSIn this study, the ability of live mycelia of Schizophyllum commune to remove heavy metals from aqueous solution was investigated. The influence of various experimental parameters, such as pH, temperature, and contact time on the biosorption process was investigated. The maximum biosorption capacity using live biomass was found as 81.5, 90.71, 70.18, and 66.18 mg/g for Pb(II), Sr(II), Zn(II), and Cd(II), respectively, at optimized conditions. The biosorption process reaches equilibrium between 180 and 240 min for the tested metal ions. The experimental data were used to study biosorption isotherm and kinetics models. The thermodynamic parameters, ΔG°, ΔH°, and ΔS°, showed that the biosorption process was endothermic and spontaneous at 303–343 K. SEM images revealed that the presence of metal ions altered the morphology of fungal hyphae. The presence of Pb(II), Sr(II), Zn(II), and Cd(II)on the fungal mycelia was confirmed using SEM-EDX. FTIR spectroscopy of biomass showed the involvement of functional groups like, -CH, amino acid, fatty acid, carboxyl group, and alkane and alkyl groups in adsorption of heavy metal ions. The adsorption capacity of S. commune for removal of heavy metals under the optimized conditions was Sr(II) > Pb(II) > Zn(II) > Cd(II). Fusarium verticillioides is an important pathogen of maize and causes serious yield losses and food safety issues worldwide. F. verticillioides produces highly toxic mycotoxin Fumonisin B1 (FB1) in infested commodities which makes these food and feeds unsafe for humans and animals. For pathogenic fungi to successfully penetrate its plant hosts, the pathogen secretes hydrolytic enzymes that can facilitate penetration into the plant cutin layer. However, there is limited information on how cutinases transcriptionally regulated to impact F. verticillioides pathogenicity. In this study, our aim is to functionally characterize cutinase transcription factors that regulate key cutinase activities that are directly associated with F. verticillioides pathogenicity and FB1 biosynthesis. Gene deletion of cutinase transcription factor FvCTF1α did not affect the growth and morphology of the fungal mycelia on CMII medium, whereas the conidiation, utilization of sodium acetate and sodium oleate, stress tolerance against cell wall interfering agent, and the cutinase and pectinase activities in the ΔFvctf1α mutant were negatively impacted. FvCtf1α regulates the expression of induced cutinase genes FvCUT1 and FvCUT4 by binding to their GC-rich promoters. In addition, FvCtf1α, containing a novel function in regulating FB1, interacts with the promoter of FvFUM1 and FvFUM6 to down-regulate the expression of FvFUM1 and FvFUM6, resulting in decreased production of FB1 in the ΔFvctf1α strain. ΔFvctf1α exhibited decreased pathogenicity in maize due to the down-regulation of pathogenicity-related genes as well as key downstream cutinase genes FvCUT3 and FvCUT4 in F. verticillioides. We also demonstrated that FvCtf1α regulated FvCUT3 and FvCUT4 differently; FvCUT4 via direct regulation while FvCUT3 via indirect regulation by interacting with FvFarB, a homologous protein of FvCtf1α. Moreover, RNA-seq analysis showed that FvCtf1α was associated with many pathways, such as fatty acid metabolism, carbon source utilization, cell wall integrity, oxidative stress, and fumonisin synthesis in F. verticillioides. Our study demonstrated that FvCtf1α was not only involved in the regulation of cutinases but also a broad spectrum of pathways that ultimately affect F. verticillioides virulence and mycotoxin biosynthesis. Two new species, Chlorocillium sinense and Calcarisporium guizhouense, isolated from a spider and fruiting body of Cordyceps sp., are introduced. Morphological comparisons and phylogenetic analyses based on multigene datasets (ITS + LSU +RPB2+tef-1alpha) support the establishment of the new species. A combined dataset of ITS , LSU , RPB2, and tef-1alpha showed the taxonomic placement of Chlorocillium in Clavicipitaceae for the first time. Pseudometarhizium is regarded as a synonym of Chlorocillium and two Pseudometarhizium species are transferred into the latter based on the phylogenetic analysis and morphological characteristics. The purpose of this study is to investigate the species diversity of Malmidea in China and further identify its species composition and distribution. The lichen specimens collected from Yunnan Province were classified based on the investigation of lichen resources in the field and the collection of lichen specimens, and combined with the methods of morphology, anatomy and chemistry. Three new records of Malmidea were reported from China, M. indica (D. D. Awasthi & M. R. Agarwal) Hafellner & T. Sprib, M. reunionis Kalb and M. hechicerae Kalb. In this paper, detailed descriptions, morphology and anatomical pictures of the recorded species were provided, and comparison and discussion were made with similar species. A key to all known Chinese Malmidea species was also provided. This study enriches the diversity of Malmidea in China, and has some significance for conservation and study of lichen diversity of China. [ABSTRACT FROM AUTHOR]

The treatment of fungal infections presents significant challenges due to the lack of standardized diagnostic procedures, a restricted range of antifungal treatments, and the risk of harmful interactions between antifungal medications and the immunosuppressive drugs used in anti-inflammatory treatment for critically ill patients with COVID-19. Mucormycosis and aspergillosis are the primary invasive fungal infections in patients with severe COVID-19, occurring singly or in combination. Histopathological examination is a vital diagnostic technique that details the presence and invasion of fungi within tissues and blood vessels, and the body's response to the infection. However, the pathology report omits information on the most common fungi associated with the observed morphology, as well as other potential fungi and parasites that ought to be included in the differential diagnosis. This research marks significance in diagnosing fungal infections, such as mucormycosis and aspergillosis associated to COVID-19 by field emission scanning electron microscopy (FESEM) imaging to examine unstained histopathology slides, allowing for a detailed morphological analysis of the fungus. FESEM provides an unprecedented resolution and detail, surpassing traditional Hematoxylin & Eosin (H&E) and Grocott's Methenamine Silver (GMS) staining methods in identifying and differentiating dual fungal infections and diverse fungal species. The findings underscore the critical need for individualized treatment plans for patients severely affected by COVID-19 and compounded by secondary fungal infections. The high-magnification micrographs reveal specific fungal morphology and reproductive patterns. Current treatment protocols largely depend on broad-spectrum antifungal therapies. However this FESEM guided diagnostic approach can help in targeted patient specific anti fungal therapies. Such precision could lead to more effective early interventions, addressing the complex management required for severe COVID-19 cases with coexisting fungal infections. Introduction: Parasites can facilitate their own spread and reproduction by manipulating insect hosts behavior, as seen in the interaction between Thitarodes xiaojinensis and Ophiocordyceps sinensis. Infection by O. sinensis leads to the mummification of T. xiaojinensis larvae, but the underlying mechanisms remain mysterious. Methods: The morphology of O. sinensis infected larvae and fungal growth were first observed. Subsequently, the metabolite changes in the larvae before and after infection with the fungus were analyzed by LC/MS and targeted metabolomics. The expression of mannitol-related genes was detected using RT-qPCR, and morphological changes in larvae were observed after injection of different concentrations of mannitol into the O. sinensis-infected larvae. Results: Significant changes were found in phenotype, fungal morphology in hemocoel, larval hardness, and mannitol metabolites in infected, mummified 0 h larvae and larvae 5 days after mummification behavior. Surprisingly, the occurrence of mummification behavior was accompanied by fungal dimorphism, as well as the absence of mannitol in both infected and non-infected larvae, until the initial accumulation of mannitol and the expression of mannitol-associated genes occurred at the time ofmummification behavior. The presence of mannitol may promote fungal dimorphism to mediate changes in fungal toxicity or resistance, leading to the end of the fungus-insect coexistence period and the incidence of mummification behavior. Furthermore, mannitol injections increase the mummification rate of the infected larvae without significant difference from the normal mummification phenotype. Onychomycosis (OM) is a widespread infection requiring prolonged treatment with potential side effects. Diagnostic certainty is therefore essential before initiating antifungal therapy. Molecular biology has already shown benefits in reducing the time to diagnosis, providing technical ease, and increasing sensitivity for the respective species that molecular tests can detect. Nevertheless, causative agents are numerous, and culture remains essential, particularly for detecting non-dermatophytes mold infections. This study compared the performance of three different diagnostic strategies: conventional culture technique, the multiplex DermaGenius® 2.0 PCR (DG), and a mixed PCR/culture algorithm guided by the result of direct examination with calcofluor (DEC). The mixed algorithm (MA) prioritizes DG PCR and DEC as the primary diagnostic tools, supplemented by selective sample inoculation when mycelial elements are visualized in DEC and when DG PCR fails to detect any fungus or identifies a fungus with morphology differing from that observed in DEC (filamentous fungi versus yeasts). With only 13% of samples requiring inoculation, MA emerged as the most effective strategy, demonstrating significantly higher sensitivity (98.18%; p < 0.001) compared to single-method approaches (78.18% for DG PCR alone and 74.55% for culture alone) while maintaining a specificity comparable to DG PCR (100%). Pointed gourd (Trichosanthes dioica Roxb) is a perennial cucurbitaceous vegetable popular in the eastern and northern parts of India. In July 2018, an occurrence of southern blight disease was observed in a pointed gourd crop grown in Odisha state located in the eastern part of India. The typical symptoms included angular necrotic patches and disintegration of leaf tissues with the vein and veinlet intact, exhibiting a diagnostic netted pattern of veins. Silky white, fan-shaped mycelial growth with sclerotial bodies was observed on the lower surface of infected leaves. Eventually, the infected leaves dried up, leaving several bare patches in the field. Associated fungi were isolated, which resembled the reference strain Athelia rolfsii based on colony characteristics. Phylogenetic analysis of the internal transcribed spacer of rRNA gene region confirmed that the isolates were A. rolfsii. Pathogenicity was successfully proved on detached leaves as well as on potted pointed gourd plants. On the basis of symptoms associated with the disease, fungal morphology, molecular identification, and pathogenicity tests on host plants, the causal fungus was identified as A. rolfsii. Paracoccidioidomycosis (PCM) is a systemic mycosis with serious clinical consequences in which the use of antifungal drugs requires long-term treatment. Therefore, we studied the effect of low-level LASER therapy (LLLT) to evaluate its prospects as a complementary treatment for PCM and improve the clinical response to the disease. Objectives: Our study focused on the resolution of lesions caused by fungal infection using a subcutaneous air pouch model of infection. Methods: We evaluated cell profile and cytokines, fungi viability, and the presence of fibroblasts and fibrocytes at the site of infection. Inoculation of P. brasiliensis (Pb) was performed using a subcutaneous air pouch model and the LLLT irradiation was performed on alternate days on the rear paws of mice for 10 days, after which the cells from the air pouch were collected and analyzed. Results: In animals irradiated with LLLT, the influx of cells to the air pouch was reduced, but they were more activated and produced pro-inflammatory (IL-12, IL-17 and TNF-α) and neutrophil (PMN) activating cytokines (IL-8, GM-CSF and γ-IFN). A better resolution of the infection, evidenced by the reduction in the number of viable fungi with preserved morphology in the air pouch, and an increase in the number of fibrocytes, indicating a healing profile were also observed. Conclusion: LLLT decreased the influx of PMN, but those presents were highly activated, with increased fungicidal activity. LLLT irradiation also resulted in earlier cicatrization at the site of infection, leading to a better outcome of the infection. The demand for common hazel (Corylus avellana) fruit increases constantly. Powdery mildew (PM) on hazels in Hungary and throughout Europe was previously caused mainly by Phyllactinia guttata. However, less than a decade ago, another fungus of Asian origin, Erysiphe corylacearum, appeared on hazels in Europe, including Hungary. Our investigation aimed to develop a species-specific PCR (ssPCR) to aid the identification of P. guttata and E. corylacearum, and to assess the presence of the latter, non-native fungus in Hungary. For this study, 59 samples were collected from Hungary between 2021 and 2023. The chasmothecial morphology of the PM fungi was observed, and the internal transcribed spacer (ITS) of ribosomal DNA was sequenced in representative samples. Morphological analysis distinguished two types of chasmothecia. Parts of the chasmothecia, typical of P. guttata, were flattened and spherical with bristle-like appendages, while other chasmothecia, characteristic of E. corylacearum, were distinctly smaller, bearing appendages with branched apices. Sequence data also verified the presence of P. guttata and E. corylacearum in our samples. The developed ssPCR revealed that E. corylacearum was present in more than three-quarters of the samples, more than a quarter of the samples contained both fungi and about one-fifth carried solely P. guttata. The alien fungus E. corylacearum was found in all but one of the sampled regions and was found on C. avellana and also on C. colurna. Erysiphe corylacearum spreads rapidly and can be considered an invasive pathogen. Its practical importance lies in its ability to infect hazelnuts, potentially causing economic losses. BACKGROUND: Potatoes, a major economic crop, are significantly impacted by Fusarium dry rot, a prevalent postharvest disease. Despite the broad‐spectrum antimicrobial properties of cinnamaldehyde, a naturally‐derived plant substance, its efficacy against the causal pathogen of potato dry rot (Fusarium oxysporum) and the underlying mechanisms have not been extensively studied. RESULTS: Our study demonstrates that cinnamaldehyde effectively inhibits the growth of Fusarium oxysporum, the pathogen responsible for potato dry rot, and increases its sensitivity to environmental stress factors such as extreme temperatures and high salt stress. Treatment with cinnamaldehyde results in altered fungal mycelium morphology, compromised cell wall stability, and disrupted cell membrane integrity, thereby reducing spore viability. Specifically, it interferes with the cell membrane and cell wall structures of the fungus, potentially disrupting fungal growth by modulating signaling pathways involved in cell wall maintenance, chitin metabolism, and GPI‐anchored protein function. Notably, we show that cinnamaldehyde induces a form of regulated cell death in F. oxysporum, which is characterized not as typical apoptosis, as evidenced by Annexin V negative staining. However, the specific cell death type and underlying mechanism still needed to be further explored. CONCLUSION: Cinnamaldehyde, an environmentally friendly plant‐based active compound, exhibits strong inhibitory effects on F. oxysporum, indicating its potential use in the prevention and control strategies for potato dry rot. Here we report on the detection and surveillance response to two Rugonectria species found in Sydney, Australia, in 2015. Both Rugonectria castaneicola and R. wingfieldii sp. nov. were found in association with cankers on Quercus robur (English oak). The fungi were initially found to be localised on amenity trees in northern Sydney, New South Wales, and as they were new detections for Australia, eradication was considered. Ongoing surveillance across the Sydney basin, regional New South Wales, and the Australian Capital Territory, however, indicated that they were already well established. Species identities were confirmed through morphological examination and molecular barcoding, with the subsequent analysis undertaken to classify R. wingfieldii sp. nov. Quantifying fungal growth underpins our ability to effectively treat severe fungal infections. Current methods quantify fungal growth rates from time-course morphology-specific data, such as hyphal length data. However, automated large-scale collection of such data lies beyond the scope of most clinical microbiology laboratories. In this paper, we propose a mathematical model of fungal growth to estimate morphology-specific growth rates from easy-to-collect, but indirect, optical density (OD600) data of Aspergillus fumigatus growth (filamentous fungus). Our method accounts for OD600 being an indirect measure by explicitly including the relationship between the indirect OD600 measurements and the calibrating true fungal growth in the model. Therefore, the method does not require de novo generation of calibration data. Our model outperformed reference models at fitting to and predicting OD600 growth curves and overcame observed discrepancies between morphology-specific rates inferred from OD600 versus directly measured data in reference models that did not include calibration. As fungal growth is complex, with fungal morphology (shape) dynamically changing over time, previous studies have quantified fungal growth by estimating growth rates during specific fungal morphologies (morphology-specific growth rates) or by mathematically modelling fungal growth. However, collecting time-series data that captures the morphological information required for mathematical model fitting or estimating morphology-specific growth rates is prohibitively time consuming for large-scale drug testing in most microbiology laboratories. Currently available antifungal therapies often have limitations such as toxicity and the development of resistance. The aim of this study was to investigate the antifungal activity and mode of action of Caraway (Carum carvi L.) essential oil (EO) and its constituent carvone against A. fumigatus, a fungal pathogen responsible for invasive aspergillosis. Caraway EO was obtained by hydrodistillation, and its antifungal activity was assessed by the broth microdilution method according to the CLSI protocol. The ergosterol content of fungal mycelia was measured using a spectrophotometric method. The mode of action of Caraway EO was assessed by electron microscopy. The interaction between Caraway EO and two known antifungals, itraconazole and caspofungin, was evaluated by determining the fractional inhibitory concentration index (FICI). At twofold dilutions ranging from 3.12 to 400 µg/mL, Caraway EO inhibited A. fumigatus growth by 10.27–100 %, with a minimum inhibitory concentration (MIC) of 200 µg/mL, while carvone inhibited fungal growth by 7.31–67.58 %, with an MIC of 400 µg/mL. The IC 50 values of the Caraway EO and carvone were 56.09 and 47.68 μg/mL, respectively. Moreover, the minimum fungicidal concentration (MFC) of the Caraway EO was reported to be 400 μg/mL. Both Caraway EO and carvone significantly inhibited ergosterol biosynthesis in A. fumigatus. Caraway EO showed synergistic antifungal activity in combination with itraconazole (FICI=0.38) and additive antifungal activity (FICI=0.75) in combination with caspofungin. Caraway EO and its constituent carvone showed strong antifungal activity against A. fumigatus by inhibiting ergosterol biosynthesis and destroying essential compartments of fungal mycelia, such as mitochondria. In addition, the combination of carvone with itraconazole or caspofungin had synergistic/additive effects, suggesting potential therapeutic efficacy for the treatment of A. fumigatus -related infections. Background: Filamentous fungi have long been recognized for their exceptional enzyme production capabilities. Among these, Trichoderma reesei has emerged as a key producer of various industrially relevant enzymes and is particularly known for the production of cellulases. Despite the availability of advanced gene editing techniques for T. reesei, the cultivation and characterization of resulting strain libraries remain challenging, necessitating well-defined and controlled conditions with higher throughput. Small-scale cultivation devices are popular for screening bacterial strain libraries. However, their current use for filamentous fungi is limited due to their complex morphology. Results: This study addresses this research gap through the development of a batch cultivation protocol using a microbioreactor for cellulase-producing T. reesei strains (wild type, RutC30 and RutC30 TR3158) with offline cellulase activity analysis. Additionally, the feasibility of a microscale fed-batch cultivation workflow is explored, crucial for mimicking industrial cellulase production conditions. A batch cultivation protocol was developed and validated using the BioLector microbioreactor, a Round Well Plate, adapted medium and a shaking frequency of 1000 rpm. A strong correlation between scattered light intensity and cell dry weight underscores the reliability of this method in reflecting fungal biomass formation, even in the context of complex fungal morphology. Building on the batch results, a fed-batch strategy was established for T. reesei RutC30. Starting with a glucose concentration of 2.5 g l - 1 in the batch phase, we introduced a dual-purpose lactose feed to induce cellulase production and prevent carbon catabolite repression. Investigating lactose feeding rates from 0.3 to 0.75 g (l h) - 1 , the lowest rate of 0.3 g (l h) - 1 revealed a threefold increase in cellobiohydrolase and a fivefold increase in β -glucosidase activity compared to batch processes using the same type and amount of carbon sources. Conclusion: We successfully established a robust microbioreactor batch cultivation protocol for T. reesei wild type, RutC30 and RutC30 TR3158, overcoming challenges associated with complex fungal morphologies. In July 2022, root rot was observed in several goji (Lycium barbarum) orchards located in Qinghai Province, China. Approximately 40% of the goji plants were affected in the orchards. Morphology of fungi isolated from affected plant, phylogenetic analyses, using internal transcribed spacer (ITS), translation elongation factor 1-alpha (TEF), and trichothecene (Tri5) sequences, as well as pathogenicity assays, were conducted to characterize and identify the causing agent of goji root rot. Isolate GQGF1-3 caused typical symptoms of L. barbarum root rot. Fungal colony characteristics and conidium morphology, combined with ITS, TEF, and Tri5 sequences showed that isolate GQGF1-3 was Fusarium sambucinum. The chemical composition, quality characteristics, antioxidant activity, and "golden flower" fungi morphology of Fu brick tea from Hunan, Hubei, Shaanxi, Guizhou, and Zhejiang provinces of China had been researched in this study. The results revealed significant differences in the contents of chemical components in different Fu brick tea (P<0.05). Among them, Fu brick tea from Guizhou had the highest contents of free amino acids and total catechins (9.03, 63.12 mg/g, respectively), while Zhejiang Fu brick tea standed out with the most elevated content of tea polyphenols and flavonoids (132.93, 8.63 mg/g, respectively). According to the electronic taste evaluation, the Fu brick tea from Hunan and Shaanxi exhibited the strongest bitterness and astringency, respectively. Meanwhile, the Zhejiang sample had the most powerful aftertaste bitterness and saltiness, whereas the Guizhou sample demonstrated the strongest umami and richness. Different Fu brick teas had different antioxidant activities, with samples from Hubei and Zhejiang showing higher levels of antioxidant activity. Correlation analysis revealed a significant positive correlation between the main chemical components and taste attributes, and tea polyphenols contributed to the antioxidant activity of Fu brick tea. Additionally, morphology indicated that the five strains of "golden flower" fungi from different origins showed slight differences in the same medium, while the Hunan strain grew faster than the others. Question: What are the extent and the possible causes of imperfect detection in lichens? Because lichens are sessile and lack seasonality, they should be easier to survey than animals that can move or plants and fungi with seasonal morphology, and one could therefore expect relatively high detection probabilities. Location: 826 standardised sampling plots across Switzerland. Methods: Using repeated detection/non‐detection data from a national lichen survey conducted by professional lichenologists, we estimated the mean and variation in detectability for 373 tree‐living species with a multi‐species occupancy model. We also quantified the effect of species conspicuousness, identifiability and observer experience on detection probability. Results: The average detection probability for a single survey was unexpectedly low with an average of 0.49 (range across species: 0.25–0.74). Conspicuous species showed higher average detectability (0.56) than inconspicuous species (0.41), and identifiability as well as previous experience with a species substantially increased the probability of a person detecting it. Accounting for experience, the mean detection probabilities of observers ranged from 0.32 to 0.69. Conclusions: Our study confirms that detection probability per survey is often far below 1 also in sessile organisms, even when a standardised survey is conducted by experts. When species are seasonal (plants, fungi, etc.), survey areas are larger, or field personnel are less experienced, as is the case for many surveys and monitoring programs, detectabilities are likely to be substantially lower. Background: Onychomycosis is a fungal nail infection of difficult treatment due to the fungal survival capacity and reduced number of effective therapies. The present study aimed to isolate fungal agents that cause onychomycosis in immunocompetent patients and evaluate how LASER treatments affect the growth and ultrastructure of isolates. Methods: In total, 21 patients with positive direct microscopic examination (DME) for onychomycosis had nail samples collected for cultivation and phenotypic identification of microorganisms. From these patients, 12 underwent LASER treatment, divided in Group 1 (n = 5) treated with Nd: YAG 1,064 nm, and Group 2 (n = 7) treated with Nd: YAG 1,064 nm + Er: YAG 2,940 nm + topical isoconazole. Transmission Electron Microscopy (TEM) was performed to evaluate ultrastructural changes after treatment. Results: DME, cultivation, and phenotypic identification showed that the most identified fungus was Trichophyton rubrum spp. After LASER therapy, sample cultivation showed alterations in the fungal morphology with reduction of hyphae, conidia, and reproductive structures. Alterations in fungal cell wall structure, cytoplasm density, and organelles were observed by TEM. Conclusion: LASER irradiation causes changes in the fungal cells, especially in the number of hyphae and the presence of conidia. Background: Weather conditions and fungal morphology may influence spatial and temporal dynamics of carbon transfer to ectomycorrhizal fungi from their tree hosts but field studies of these dynamics are challenging. Methods: Broad-leaved trees in a Swiss forest were labeled with 13C-depleted CO2 from 2001–2005 and 13C traced into ectomycorrhizal sporocarps collected at different distances from those trees. We then correlated distance (zone), year, fungal genus, fungal morphology, and solar radiation against sporocarp δ13C and loge C/N in stepwise regressions. Results: CO2-supplied trees contributed 71 ± 5%, 32 ± 6%, and 19 ± 7% of sporocarp carbon at 0–6, 6–12, and 12–18 m from these trees, respectively. Fungal morphology (rhizomorph presence and ectomycorrhizae hydrophobicity) was uncorrelated with fungal carbon acquisition. After drought in 2003, CO2-supplied trees contributed less carbon in 2004 than in other years to sporocarps and sporocarp loge C/N decreased. In contrast, sporocarp loge C/N peaked in 2005 and contributions from CO2-labeled trees to the 6–12 and 12–18 m zones increased. Solar radiation with a 14–21 day lag correlated negatively with loge C/N. Sporocarp loge C/N correlated positively with δ13C at 0–6 m and negatively elsewhere. Conclusions: These patterns indicated: 1) fungal morphology did not affect carbon transfers, 2) carbohydrates rather than amino acids were transferred across zones to sporocarps (inferred from spatial correlations of loge C/N and δ13C), 3) sporocarp δ13C and loge C/N reflected the last 2–3 weeks of photosynthesis; 4) summer drought and plant allocation influenced the quantity and spatial extent of belowground transfer. Background Modicella Kanouse (1936) is the only genus of Mortierellaceae known to produce macroscopic fruiting bodies in the form of small, whitish, round sporocarps. Specimens which belong to Modicella were collected during our field investigations in tropical karst areas of China. Based on morphological characteristics and phylogenetic analysis, a new species is introduced. New information Modicella guangxiensis is described as a new species from tropical karst areas of China. The main distinguishing characteristics of M. guangxiensis are the number of sporangiospores per sporangium (11-18), the size of sporangiospores (14-34 × 12-27.5 µm) and the surface of some hyphae with hemispherical tuber. The phylogenetic analyses, based on the internal transcribed spacer (ITS) and the large subunit (LSU) regions of rDNA sequences using Bayesian (BA) and Maximum Likelihood (ML) methods showed that the new taxon is closely related to M. reniformis. Superelongation disease (SED) is a fungal disease that affects cassava in the Caribbean. The symptoms include the appearance of dry necrotic spots and lesions on the leaves, which may severely affect the plant yield. However, the primary causal pathogen is difficult to culture and isolate in the lab because of its slow growth and potential contamination from faster-growing organisms. In addition, the leaf symptoms can be confused with those caused by other pathogens that produce similar necrotic spots and scab-like lesions. There is also little or no information on the contribution of endophytes, if any, to disease symptoms in cassava, a plant where the disease is prevalent. Therefore, this study aimed to characterize the fungal communities in cassava associated with SED symptoms by analyzing gross fungal morphology and performing metagenomics profiling. First, several individual pathogenic fungi were isolated and cultured from diseased cassava leaf tissues from seven locations in Barbados (BB). Both culture isolation and molecular community analyses showed the presence of several other fungi in the disease microenvironment of symptomatic cassava leaves. These included Fusarium, Colletotrichum, and Alternaria species and the suspected species Elsinoë brasiliensis synonym Sphaceloma manihoticola. Additionally, a community analysis using ITS2 amplicon sequencing of 21 symptomatic leaf tissues from BB, St. Vincent and the Grenadines (SVG), Trinidad and Tobago (TT), and Jamaica (JA) revealed that the disease symptoms of superelongation may also result from the interactions of fungal communities in the mycobiome, including Elsinoë species and other fungi such as Colletotrichum, Cercospora, Alternaria, and Fusarium. The globally distributed genus Trichaptum is one of the most species-rich among polypores in terms of hosting other fungi. Among Trichaptum-associates, there is a group of mazaediate lichenized fungi (Coniocybomycetes, Ascomycota) that previously had an uncertain phylogenetic position. DNA sequences - mitochondrial small subunit (mtSSU), nuclear large subunit rDNA (nuLSU), and internal transcribed spacer (ITS) - were obtained from 29 specimens collected from Europe and North America. Maximum likelihood and Bayesian inference analyses of these three gene loci were used to infer phylogenetic position and relationships among lineages. Statistical tests were used to find which phenotypical characteristics distinguish species. The molecular sequence data provide evidence that the fungicolous specimens form a distinct lineage within Coniocybomycetes sister to the combined clade of Chaenotheca s. lat. and Sclerophora. Considering its phylogenetic placement and strict specialization, we describe a new genus - Chaenotricha. This fungicolous lineage contains three species based on molecular characteristics. Morphological characters mostly overlap except for spore size and stalk length of apothecia. We provide a new combination, Chaenotricha obscura, for the only previously described species for which we designate an epitype, and introduce a new species - Chaenotricha cilians. Recent lichenological fieldwork in the Czech Republic resulted in the discovery of two previously unrecognized entities in the genus Psilolechia which are described below as new species: the saxicolous P. cretacea and the lignicolous P. torii. Both taxa regularly produce a hyphomycetous anamorph in the form of erect, elongate conidiogenous cells on the thallus surface, and both contain unidentified specific secondary metabolites. The placement of the two new species within the genus is primarily based on morphology and chemistry. In addition, three barcodes were received for P. cretacea (ITS, mtSSU) and P. torii (ITS). Agonimia gargominyi (Verrucariaceae, Verrucariales) is described and illustrated from bark samples collected from Morane atoll (Tuāmotu-Gambier Islands, French Polynesia). It is characterized by the large superficial perithecia having a pale brown-colored ostiolar region, the 8-spored asci, and the relatively large lobed squamules. Micromorphology of the ascomata and the thalline squamules matches the genus Agonimia. Additionally, to accommodate the inclusion of this newly identified species, the existing comprehensive key encompassing all known Agonimia species worldwide has been modified and updated accordingly. The Biatora vernalis- and meiocarpa-groups comprise species with pale beige to reddish brown apothecia and 0-, rarely also 1(–3)-septate ascospores. A DNA barcoding approach based on ITS sequences shows that these two groups comprise more species and phylogenetic diversity than previously known. Specimens identified as B. vernalis, the type species of the genus, appear to be paraphyletic with regard to B. chrysantha. In addition, there is a morphologically similar species belonging to the B. meiocarpa-group and tentatively named “B. orientalis” in previous publications. Biatora subduplex has for some time been known to comprise specimens from the B. vernalis-, as well as the B. meiocarpa-group. Similar to the situation in B. vernalis, samples from the meiocarpa-clade form several subclades close to B. meiocarpa. Anatomical studies reveal subtle, but recognizable morphological differences between B. subduplex s.str. and the species in the meiocarpa-clade, but not between the subclades. Here, we describe Biatora orientalis as new to science, raise B. meiocarpa var. tacomensis to species rank and provide revised identification keys for the B. vernalis- and B. meiocarpa-groups. The species described as Bacidia genuensis is transferred here to Bacidina as B. genuensis (Ramalinaceae, Lecanorales, lichenized Ascomycota). An updated morphological description is provided. The species is characterized by mostly blackish apothecia on a thick, microsquamulose thallus, a crystal-inspersed proper exciple that is mostly prosoplechtenchymatous, an ascus with a wide and dome-shaped axial body and an expanded c-layer (resulting in a thin, amyloid d-layer), a blue-green pigment in the epihymenium, proper exciple, and pycnidial wall, and an orange-brown, K+ intensifying pigment in the hypothecium and sometimes proper exciple. This combination of characters sets the species apart from its potentially close relatives Bacidina egenula and B. indigens, as well as the superficially similar, but more distantly related, Toniniopsis bagliettoana. The new combination Thalloidima squamatum is proposed for a morphologically and phylogenetically distinct species, until now included within the variation of T. sedifolium. Compared to T. sedifolium, the species is characterized by its larger squamules and apothecia, shorter ascospores, the pale color of the excipulum, and its habitat growing on moss cushions over calcareous rock or directly on rock rather than on soil. Most of the material studied originates from Norway and Sweden, but specimens are also reported from Austria, Croatia, Germany, Greenland, Italy, Poland, Russia, Scotland and Switzerland. Coriandrum sativum L. is a globally significant economic herb with medicinal and aromatic properties. While coriander leaf blight disease was previously confined to India and the USA, this study presents new evidence of its outbreak in Africa and the Middle East caused by Alternaria dauci. Infected leaves display irregular chlorotic to dark brown necrotic lesions along their edges, resulting in leaf discoloration, collapse, and eventual death. The disease also impacts inflorescences and seeds, significantly reducing seed quality. Koch's postulates confirmed the pathogenicity of the fungus through the re-isolation of A. dauci from artificially infected leaves, and its morphology aligns with typical A. dauci features. Notably, this study identified strong lytic activity (cellulase: 23.76 U, xylanase: 12.83 U, pectinase: 51.84 U, amylase: 9.12 U, and proteinase: 5.73 U), suggesting a correlation with pathogenicity. Molecular characterization using ITS (ON171224) and the specific Alt-a-1 gene (OR236142) supports the fungal morphology. This research provides the first comprehensive documentation of the pathological, lytic, and molecular evidence of A. dauci leaf blight disease on coriander. Future investigations should prioritize the development of resistant coriander varieties and sustainable disease management strategies, including the use of advanced molecular techniques for swift and accurate disease diagnosis to protect coriander from the devastating impact of A. dauci. The purpose of the study is to develop a novel peptide for caries management. Gallic-Acid-Polyphemusin-I (GAPI) was synthesised by grafting Polyphemusin I (PI) and gallic acid (GA). Biocompatibility was evaluated using a Cell Counting Kit-8 Assay. Antimicrobial properties were assessed using minimum inhibitory concentration (MIC) and minimum bactericidal/fungicidal concentration (MBC/MFC). The bacterial and fungal morphology after GAPI treatment was investigated using transmission electron microscopy (TEM). The architecture of a consortium biofilm consisting of Streptococcus mutans, Lacticaseibacillus casei and Candida albicans was evaluated using scanning electron microscopy (SEM) and confocal laser scanning microscopy. The growth kinetics of the biofilm was examined using a propidium monoazide–quantitative polymerase chain reaction. The surface and calcium-to-phosphorus molar ratio of GAPI-treated enamel after pH cycling were examined with SEM and energy-dispersive X-ray spectroscopy. Enamel crystal characteristics were analysed using X-ray diffraction. Lesion depths representing the enamel's mineral loss were assessed using micro-computed tomography. The MIC of GAPI against S. mutans, L. casei and C. albicans were 40 μM, 40 μM and 20 μM, respectively. GAPI destroyed the biofilm's three-dimensional structure and inhibited the growth of the biofilm. SEM showed that enamel treated with GAPI had a relatively smooth surface compared to that treated with water. The calcium-to-phosphorus molar ratio of enamel treated with GAPI was higher than that of the control. The lesion depths and mineral loss of the GAPI-treated enamel were less than the control. The crystallinity of the GAPI-treated enamel was higher than the control. Fungi are among the most common infectious agents affecting the skin of animals. The skin can serve as a port of entry for fungal infections, which can eventually become disseminated. In some regions of the world, oomycetes, such as Pythium and Lagenidium, are also responsible for a significant number of severe cutaneous infections. Histologic evaluation of fungal morphology, including size, shape, septation, branching, and budding characteristics, combined with the distribution of inflammatory infiltrates within different skin layers can potentially identify etiologic agents, guiding selection of antifungals and additional diagnostics. Fungal infections of the skin surface are typically caused by Malassezia and rarely Candida, with opportunistic fungi also capable of colonizing the skin surface, especially when the barrier is broken. Folliculocentric infections, caused by dermatophytes, result in mild to severe inflammation and can occasionally penetrate deep into the skin. A wide range of fungi, including agents of hyalohyphomycosis, phaeohyphomycosis, and dimorphic fungal infections, as well as oomycetes, result in nodular cutaneous and subcutaneous lesions. With the occasional exception of dimorphic fungi, fungal speciation often requires cultures performed on fresh tissues. However, molecular techniques such as pan-fungal polymerase chain reaction on paraffin blocks is becoming an increasingly useful tool to distinguish between cutaneous fungal pathogens. This review focuses on describing the clinical and histologic features of the most common fungal and oomycete infections affecting the skin of animals, divided according to distribution patterns of lesions and fungal or oomycete morphology. Biological control is referred to as the "use of natural or modified organisms, genes" to minimize the effects of undesirable pests, pathogenic microorganisms, and diseases on plant crops. This measure has become a suitable and safe alternative for chemical fungicides in plant disease management. Endophytic fungi have received much attention as biological control agents against many plant pathogens through antibiosis, parasitism, invading spores, mycelium, and cells of the pathogen, and secreting bioactive metabolites. While the therapeutic properties of Garcinia atroviridis have been studied, the existence of microbial endophytes and their properties is still less documented. In this research, G. atroviridis endophytic fungi were isolated and identified by fungal colony morphology observation combined with the PCR-amplified fungal internal transcribed spacer (ITS) sequence analyses. Fungal endophytes were assessed for their biocontrol potential against Colletotrichum gloeosporioides. In total, 111 endophytic fungal isolates harboring inside the leaf, branch, and fruit of G. atroviridis belonged to 5 different species with 3 different genera and two unidentified genera. All the endophytic fungal species isolated were evaluated using an in vitro dual culture assay against C. gloeosporioides, a common pathogen that causes anthracnose disease. The results of the present study clearly showed that seven species of isolated fungal endophytes were capable of inhibiting the mycelial colony growth of C. gloeosporioides with an inhibition percentage between 54.67% to 87.94%. Among these species, Nigrospora sphaerica recorded the highest PIRG with 87.94%. Our work indicates that endophytic fungi isolated from G. atroviridis have a biocontrol effect on C. gloeosporioides and are expected to be a potential source of bioactive metabolites. Information of dark septate endophyte (DSE), arbuscular mycorrhizal (AM), and fine root endophyte (FRE) fungal symbioses of aroids in the Western Ghats region are very low. Therefore, we assessed the endorrhizal symbiosis in 25 aroid species belonging to 16 genera of Araceae from six different locations of the Western Ghats. The results revealed co-occurrence of the DSE and AM symbiosis in all the examined aroids, and FRE presence in seven aroids (Alocasia × amazonica, Alocasia sp., Anthurium andraeanum, Epipremnum aureum, Spathiphyllum sp., Syngonium podophyllum, and Zantedeschia aethiopica). We found variance in root length having AM (inter and intracellular hyphae, arbuscules, vesicles and arbusculate coils) and DSE (melanized septate hyphae, microsclerotia, and moniliform hyphae) fungal structures. Moreover, the AM fungal morphology of Arum-Paris type was widespread, and intermediate type morphology reported for the first time in five aroids. AM fungi colonized the roots of Philodendron xanadu the most, followed by DSE in Caladium bicolor, and FRE in Spathiphyllum sp. AM fungal spores were present in all aroid soils examined. The percentage of root length comprising FRE hyphae was significantly and positively correlated root length with FRE arbuscules, AM fungal spore numbers and total colonization. Background: Research interest in plant essential oils has increased significantly due to their natural properties and consumer demand for safer methods of food preservation. Plants produce large amounts of secondary metabolites, which have potential activity against fungal pathogens. This study aimed at screening essential oils for their antifungal effects on citrus against Penicillium digitatum, morphological effect and finally determine which essential oils are the most effective. Results: The EC50 of seven selected cinnamon (0.424 μL/mL), patchouli (0.513 μL/mL), vetiver (0.612 μL/mL), dill (1.597 μL/mL), origanum (1.971 μL/mL) and ylang (2.214 μL/mL) was determined. In addition, cinnamon substantially reduced sporulation (100%) followed by patchouli (86.02%), vetiver (82.73%), and chamomile (79.04%), respectively. Our GC–MS result determined variance in concentration of essential oils compound composition. The total compound composition in all seven essential oils > 1% was found to be 3 in cinnamon, 5 in dill, 10 in origanum, 13 in ylang, 11 in patchouli, 9 in chamomile and 16 in vetiver. Addition of essential oils significantly altered fungal morphology by scanning electron cryomicroscopy. Patchouli and origanum showed broken hyphae while there was an indication of severe deformation and collapse of spores in cinnamon and chamomile. Conclusion: Based on our findings, we report that these essential oils could potentially be applicable in controlling P. digitatum with reduced concern for human health, environmental contamination and possibly replacement of synthetic treatments. This study investigated the efficacy of lauric arginate (LAE) as a potential preservative against wood deteriorating fungi. First, the antifungal properties of LAE against two brown rot fungi, Gloeophyllum trabeum (G.t.) and Rhodonia placenta (R.p.), and two white rot fungi, Trametes versicolor (T.v.) and Irpex lacteus (I.l.) were examined using malt-agar media as substrate. Then the biological resistance of LAE-treated wood specimens (10, 20, and 25 % LAE solutions) was tested following American Wood Protection Association (AWPA) E10 soil-block test procedures. The in vitro study showed LAE actively inhibited the growth of all the fungi tested compared to growth in control plates but there were no considerable changes observed in the morphology of fungi hyphae. Wood impregnated with LAE showed increased weight percent gain as a function of treatment concentration. Characterization of LAE-treated wood on a thermogravimetric analyzer showed LAE shifted the thermal degradation temperatures to lower stages but did not significantly improve the thermal stability of the treated specimen. LAE in wood significantly suppressed fungus growth, leading to decreased mass loss but it also leached out from wood specimens during fungi exposure. [ABSTRACT FROM AUTHOR]

Invasive fungal infections are an important cause of morbidity and mortality in a broad range of patients. Adequate and early diagnosis is a challenge and of importance for improved survival. New molecular-based diagnostic methods are trendsetting, yet with the drawback that conventional tests receive less attention, in the laboratory as well as in the clinical setting. We aimed to provide a useful recommendation for direct microscopy for effectively managing numerous specimens related to fungal infections, mainly covering opportunistic pathogens. A PubMed literature search covering direct fungal microscopy was performed with no restrictions on publication dates. Best practise recommendations targeting the role of direct microscopy in diagnosing fungal infections are given. This review highlights when to perform direct microscopy, displays the main fungal morphologies, discusses the pitfalls related to microscopy, and recommends how to best report the results to clinicians. In many samples, the performance of direct microscopy provides an important diagnostic benefit that is greater than culture alone. Fluorescent dyes improve sensitivity and allow a fast and rapid read. Reporting includes the presence or absence of yeast forms, septate or non-septate hyphae, pigmentation, cellular location, or any other specific structures being present. Grafting technology is applied to control the trunk canker disease of Chinese hickory (Carya cathayensis). In this study, the grafting of Chinese hickory hybrids onto American pecan (C. illinoinensis) and Hunan pecan (C. hunanensis) as rootstocks was investigated in the field. The trunk canker was significantly more serious in self-grafted C. cathayensis (CC), and the disease of C. hunanensis-grafted C. cathayensis (HC) was less serious than that of C. illinoinensis-grafted C. cathayensis (IC). The causal agent of Botryosphaeria dothidea was characterized by comparison of the fungal morphology and its combined DNA sequence data. Seedlings of CC, IC and HC were inoculated with B. dothidea, and we observed that the phloem and xylem tissues of CC were more damaged by B. dothidea mycelia than those in IC and HC at 2 and 4 days, respectively, using a scanning electron microscope (SEM). The phenol contents in the trunk bark tissues of HC, the total phenylalanine ammonia-lyase (PAL) activities of HC and IC were significantly increased, and the total polyphenol oxidase (PPO) activity of HC was higher than that of other pecans at 5 days. The expression levels of the CHS gene were significantly upregulated in HC, and the expression levels of PAL, C4H, 4CL, and CHS in IC and HC were high at 1, 3, and 5 days. In summary, the grafted pecan IC and HC showed higher phenol content, PAL activity, and C4H, CHS, and PAL gene expression levels. Filamentous fungi are widely exploited as cell-factories in industry due to their metabolic diversity, high production capacity and secretion efficiency. In stirred tank bioreactors, the filamentous fungus Aspergillus niger builds agglomerates of complex morphology. The formed morphology has a decisive influence on productivity and is significantly influenced by the hydrodynamic stress induced by the stirrer. Due to the complex interactions between the cultivation parameters, an examination solely of the influence of the stirrer is difficult. In this work, fragmentation experiments were established and conducted allowing the selective examination of the impact of stirrer type and stirring intensity on the bio-agglomerates' morphology at a reasonable effort. With the applied method, a clear order of stress intensity for different stirrer types was found. Studying the breakage mechanism, both fracture and abrasion have been found to occur, significantly depending on the employed stirrer type. The impeller swept volume model has been proven to correlate the particle size well for the employed stirrer types. In the literature, most work on the characterisation of stirrer types concerning hydrodynamic stress has been done using model systems. This study characterised and identified the causative agent of irregular necrotic leaf spots in eggplant. The culture characteristics and morphology of three fungal isolates indicate that they are from the Didymellaceae group. Using a polyphasic approach—combined morphological, cultural, pathogenicity, and molecular characterisation (concatenated sequences of three gene loci: ITS, TUB2, LSU), the fungal isolates MBELIQ03A and MBELIQ03D were identified as Epicoccum poaceicola. Isolate MBELIQ02 was identified as Epicoccum sp. due to a lack of clear grouping with a single Epicoccum species. In repeated trials, the three Epicoccum isolates were pathogenic to eggplant leaves in detached leaf assays. Similar fungi were consistently re-isolated from the inoculated leaves and thus, establishing Koch's postulates. Inoculation of the three isolates on solanaceous leaves and fruits (tomato, eggplant, and pepper) revealed their varied pathogenicity. This is the first confirmed scientific report of Epicoccum poaceicola causing eggplant leaf spots. This is also the first record of this fungal species in the Philippines.
