Abstract

Research into the effects of soundscapes on aquaculture species in key production systems is sparse, despite potential impacts of sound on animal welfare and commercial yields. In the following study, 2 high-value global aquaculture species, whiteleg shrimp Litopenaeus vannamei and Atlantic salmon Salmo salar, were exposed to aquaculture production system soundscapes. For shrimp, sound recordings of a commercial recirculating aquaculture system (RAS) were played back at a sound pressure level (SPL) of 128 dB re 1 µPa, and for salmon, recordings from a commercial sea pen production system were played back at an SPL of 127 dB re 1 µPa for an 8 wk period. Effects of exposure on growth, survival, and indications of metabolic stress were measured as parameters of interest for aquaculture production. Mean growth performance and survival rates did not differ significantly between sound and control treatments for either species. Blood and haemolymph parameters from both species indicated no measurable change in metabolic status or stress levels. Slight, but non-significant, increases in total haemocyte count and, in particular, hyaline cell count were recorded in shrimp exposed to sound. Slight, but non-significant reductions in overall weight gain were recorded in sound-exposed salmon. Overall, the results indicate that sound exposure in current production systems does not negatively affect the early grow-out stage of these key species, either due to rapid habituation or higher hearing thresholds of hatchery-produced individuals, and that no measurable stress response occurs in sound-exposed animals. In future studies, response of the studied species to acute sound exposure and the response of earlier, and potentially more sensitive, life-stages will need to be determined to ensure optimal welfare and production performance.

Abstract

Background Radium-223 improves overall survival and preserves quality of life in patients with metastatic castration-resistant prostate cancer (mCRPC) and symptomatic bone metastases and no known visceral metastases. Radium-223 can be used in combination with a luteinizing hormone releasing hormone (LHRH) analogue and as part of a sequential treatment scheme if disease progresses after at least two prior lines of systemic mCRPC therapies or if no other available systemic treatment is eligible. Objectives Today physicians are faced with a previously unknown multitude and complexity of options for the treatment of mCRPC. An increasing number of clinical trials contribute to the dynamics of the therapeutic landscape. Radium-223 was approved for mCRPC treatment in 2013. Up to now the recommendations of use have been adjusted several times. Highlighting recent clinical trials and practice, this paper explores the position of radium-223 within the therapeutic sequence and outlines key elements for the interdisciplinary cooperation between uro-oncologists and nuclear medicine specialists. Results The mode of action of radium-223 does not depend on the androgen receptor (AR) pathway. Thus, it is an option in the therapeutic sequence when the efficacy of other agents is reduced by resistance. Furthermore, the efficacy of prior or subsequent medications are neither reduced nor enhanced by radium-223. The opportunity of an AR-independent and survival-prolonging medication should be taken as soon as the indication criteria are met because the incidence of visceral metastases increases during disease progression. According to current mCRPC guidelines, the osteoprotective use of bisphosphonates or denosumab is recommended, before treatment with radium-223 is started or resumed.

Abstract

Whole genome analysis can be performed by next-generation-sequencing (NGS) techniques, microarrays, or parallel real-time PCR addressing multiple genomic regions. These analyses require a minimal amount of gDNA from 100–1000 ng, equivalent to 16,000–160,000 cells. For analysis of genomic differences between individual cells, accurate replication of the single-cell genome is required. Here, we describe the reliability of single-cell whole genome amplification (WGA) and its application in NGS and real-time PCR. Single cells were obtained by picking cells under the microscope or by dilution. Amplification was performed for 8 hours at 30°C, generating up to 40 μg of DNA. WGA methods from other suppliers were applied to single-cell samples in parallel. For NGS, 2 μg of DNA was used for shearing and library preparation was done using the TruSeq® DNA Sample Preparation Kit. The libraries were quantified and sequenced (paired-end) on a MiSeq® Instrument. For real-time PCR, 100 pg (bacterial cells) or 10 ng (mammalian cells) of WGA DNA was analyzed using SYBR-Green reagents or RT2 qPCR Arrays. A real-time PCR analysis of 267 loci across the entire genome was performed using 10 ng of WGA DNA for each primer assay. Results showed low and consistent CT-values in real-time PCR for all loci, with no dropout from any marker, indicating successfully amplified DNA from all areas of the genome and high suitability for single-cell genomics. Whole genome sequencing of the Bacillus subtilis genome was performed on the MiSeq from 2 μg of non-amplified genomic DNA or DNA amplified by REPLI-g Single Cell WGA from cells. Comparable sequence coverage was observed for gDNA and REPLI-g Single Cell amplified DNA. A comparison of non-amplified and REPLI-g amplified DNA revealed error rates in a similar, very low, percentage range. The representation of regions of different GC content matches the representation of the genome.

Abstract

Recent studies on grammaticalization in sign languages have shown that, for the most part, the grammaticalization paths identified in sign languages parallel those previously described for spoken languages. Hence, the general principles of grammaticalization do not depend on the modality of language production and perception. However, in addition to these modality-independent paths, some modality-specific paths have been described. Of special interest in this context is the grammaticalization of gestures. Since sign language and co-speech gesture share the visual-gestural modality, sign languages, unlike spoken languages, have the potential to integrate manual and non-manual gestures into their linguistic system. Co-speech gestures cannot only be integrated as lexical items (lexicalization of gestures), but more importantly also as grammatical markers (grammaticalization of gestures). This chapter focuses on the latter type of change, and presents two case studies which illustrate the grammaticalization of manual and non-manual gestures in sign languages.

Abstract

Understanding the factors that affect life cycle costs in the earliest stages of product development bring valuable insights for vehicle architecture decision making. This study presents a life cycle costing model used to compare hybrid electric vehicle architectures with varying levels of electrification to a reference conventional car. The work presented highlights the importance of considering total costs of ownership (COO) and operation alongside manufacturing costs in making strategic business decisions. Results from the life cycle cost model scenarios set for 2015-2024 show that car architectures with increased electric range capability allow for significant customer fuel cost savings. These savings can offset increased manufacturing costs within the first three years of ownership based on US Energy Information Agency (EIA) current fuel and electricity pricing forecasts. If fuel prices or annual vehicle use remain low, electrification becomes less attractive as payback periods are extended beyond 10 years time. Regardless of future energy pricing scenarios, the fixed costs of vehicle ownership remain the largest costs to the end user. Manufacturers can use this information to their advantage in creating new business models and designing cars that deliver increased value to the end customer. Because electrification significantly reduces CO2 tailpipe emissions, government incentives and taxing schemes are expected to play a positive roll in offsetting a large part of the additional manufacturing costs. Finally, optimization methods are used to determine sensitivities of variables that affect total cost of ownership. Of these variables the user’s city/highway driving profiles and the price of fuel/Electricity have the greatest sensitivity to cost of ownership.

The automotive industry is going through fundamental change as new car architectures begin to compete in the market. If the new hybrid and electric vehicle offerings prove to gain traction, established firms that are slow to join in the market can be facing potential failure. However, if conventional cars outperform the new entrant vehicle architectures, these can disappear as rapidly as they gained popularity as explained in informational cascades theory. Automotive manufacturers can mitigate this uncertainty by focusing on vehicle architecture platforms alongside the traditional development process in order to filter new technology offerings that become relevant.

Abstract

This work presents a methodology to analyze various hybrid electric vehicle (HEV) architectures by means of Multiple Domain Matrices (MDM), with the goal of exploring how functions and components relate for any chosen HEV configuration. The analysis of dependencies between and amongst the functional and component domains of eight different HEV architectures developed a basis of knowledge to define a total of 76 sensible possible architectures out of a field of more than 432 different combinations. 38. The Design of Future Cars in a New Age of Architectural Competition

Abstract

This paper presents how complex system architecture lifecycles, such as that of cars, follow a similar S-curve shaped path as that of individual technological innovations. By applying this theory we show that today’s automotive industry has started a new chapter of architectural competition with similarities to its early history from 1885-1915 when steam, electric and internal combustion engine cars were competing to dominate the automotive market. Taking a historical perspective, we find that firms that organize their development activities to focus on bringing about architectural innovation are better placed in succeeding in the future market until a new dominant architecture emerges. The architecture lifecycle framework used in this study is constructed by means of a performance index. The index scores the performance of 91 cars of various architectures based on five overall system variables: power, weight, maximum velocity, fuel efficiency and the manufacturer’s suggested retail price. Depicting architectural performance over time helps identify periods of architecture competition and dominance where historical agents to change can be identified. The key factors that brought about architectural competition in the early 1900’s involved a series of innovation breakthroughs in engine and fuel technologies. Today, a new wave of power train innovations is being triggered primarily by environmental regulatory demands to reduce vehicle emissions. Future research lies in presenting a methodology for selecting vehicle architectures early on in the product development cycle that are best suited for the market going forward based on a manufacturer’s goals and a cost-benefit analysis.

40. Tumour specificity of 11C-acetate and 18F-FDG positron emission tomography in prostate cancer patients

Abstract

Visualisation of primary prostate cancer and its metastases remains a clinical important problem, state-of-the-art methods like CT, MRI, TRUS (transrectal ultrasound) and 18F-FDG PET notwithstanding. The difficulties are even more pronounced in case of local recurrence as 18F-FDG PET, while useful in the detection of metastases, is not considered helpful in the investigation of relapse. The aim of this study was to evaluate the efficacy of 11C-acetate and 18F-FDG-PET in the detection of prostate cancer and its metastases in patients treated with or without hormonal ablation. 25 patients (age: 54 to 82 years) were investigated during follow-up of a primary prostate cancer, suspected relapse, or metastatic disease using 11C-acetate PET and 15 with 18F-FDG PET additionally. 14 Patients had antiandrogen treatment at the time of the investigation, the other 11 patients were drug-naive. Primary tumours were detected in 20/24 (83%) patients using 11C-acetate PET and in 10/15 (75%) patients using 18F-FDG PET. Based on the results of both PET scans, one patient was diagnosed with a lung cancer as second malignancy. Median 18F-FDG uptake exceeded that 11C-acetate in distant metastases (SUV=3.2 vs. 2.3, respectively). 11C-acetate uptake, however, was higher in local recurrence and in local lymph node metastases (median SUVs = 2.9 and 3.8) compared to 11C-acetate (median SUVs = 1.0 and 1.1, respectively). 11C-acetate and 18F-FDG uptake was lower in patients on antiandrogen therapy compared with the drug-naïve patients but 11C-acetate uptake was not significantly impaired. A close correlation of serum PSA level and Gleason score on the one hand and both 11C-acetate uptake and 18F-FDG uptake on the other hand was observed. These data confirm that 11C-acetate is more useful than 18F-FDG in the detection of local recurrences and localised lymph node metastases. 18F-FDG, however, appears to be more accurate in visualising distant metastases. Antiandrogen therapy reduces tumour metabolic activity and therefore overall PET accuracy but detection of local recurrences by 11C-acetate is not severly compromised. Ultimately, there may be a role for combined 11C-acetate/18F-FDG PET in the follow-up of patients with prostate cancer and persisting or increasing PSA.

41. Quantitative myocardial perfusion PET combined with coronary anatomy derived from CT angiography: Validation of a new fusion and visualisation software

Abstract

Aim Dynamic perfusion PET offers a clinical relevant advantage over myocardial perfusion scintigraphy due to its ability to measure myocardial blood flow quantitatively. This leads to an improved detection of multivessel disease and the possibility to assess not only the culprit lesion but lower grade stenoses as well. For appropriate revascularization, perfusion defects must be matched to coronary lesions. It has been shown that image fusion of morphological and functional images is superior to side-by-side analysis. Still, software for quantitative perfusion PET combined with CT angiography is rare. In this paper we present a new software tool for image fusion and visualization of quantitative perfusion PET and coronary morphology derived from CT angiography. Methods In our software, a PET uptake image is used for manual co-registration. Co-registration results are then applied to the functional data derived from compartment modelling. To evaluate the reproducibility of the manual co-registration, we calculated the deviation between a series of manual co-registrations performed on nine pairs of unregistered PET and CT datasets by five trained participants. Two dimensional transfer functions were used to highlight the coronary arteries from the CT study in the combined data sets. Results The average Euclidian distances for three references points were between 3.7 and 4.1 mm. The maximum distance was 10.6 mm. By the use of the two dimensional transfer functions, coronary anatomy could be easily visualised either by user-interaction or automatically by use of neuronal networks. Conclusions With this approach it is possible to combine quantitative perfusion PET with coronary anatomy derived from CT angiography. Our first experiences indicate that manual image fusion with our tool is reproducible and that visualisation of the combined datasets is achieved within short time. 43. Koregistrierung parametrischer PET-Daten mit CT-Datensätzen in der kardialen Diagnostik

Abstract

Ziel/Aim: Die kardiale PET zeichnet sich gegenüber der SPECT u.a. durch die Möglichkeit der absolut quantifizierenden Messung der myokardialen Perfusion aus. In der Diagnostik der KHK wird zunehmend eine Kombination von SPECT oder PET mit einer morphologischen Darstellung der Koronargefäße durch eine CT-Angiographie eingesetzt. Auch bei Einsatz von Hybridgeräten machen Unterschiede in der Datenakquisition und die Thoraxbewegung bei der Atmung eine Koregistrierung der Datensätze erforderlich. Von Seiten der Gerätehersteller wird derzeit jedoch nur eine Koregistrierung statischer Datensätze unterstützt. Der wesentliche Vorteil der PET, die Einschätzung einer koronaren 3-Gefäßerkrankung durch Beurteilung der absoluten Flusswerte, geht dadurch verloren. Ziel war daher die Koregistierung von CT und funktionellem PET-Datensatz. Methodik/Methods: Verwendet wurden dynamische PET-Studien mit N-13-Ammoniak und CT-Angiographien der Koronargefäße, die an unabhängigen Geräten akquiriert wurden. Die Koregistrierung von CT und PET erfolgte anhand des rekonstruierten CT-Datensatzes und des PET-Uptake-Bildes (letztes Frame der dynamischen Studie, 9.-16. Minute). Die Koregistrierung erfolgte manuell sowie durch automatische Detektion von Grenzflächen über Schwellenwerte. Im Rahmen der Kompartmentmodellierung dynamischer PET-Datensätze findet eine Reorientierung entsprechend der Herzachsen statt und die funktionellen Parameter können nur für Volumenelemente innerhalb des Myokards bestimmt werden. Daher gehen die ursprüngliche Orientierung der Datensätze und weitergehende anatomische Informationen zunächst verloren. Für eine kombinierte Darstellung der Morphologie der Koronargefäße und der quantitativen Flussinformation wurde daher in einem nächsten Schritt eine Rückprojektion der Flussinformation in die anatomisch korrekte Lokalisation durchgeführt.
