18. Transfection of entomopathogenic Metarhizium species with a mycovirus confers hypervirulence against two lepidopteran pests.

Abstract

Although most known viruses infecting fungi pathogenic to higher eukaryotes are asymptomatic or reduce the virulence of their host fungi, those that confer hypervirulence to entomopathogenic fungus still need to be explored. Here, we identified and studied a novel mycovirus in Metarhizium flavoviride, isolated from small brown planthopper (Laodelphax striatellus). Based on molecular analysis, we tentatively designated the mycovirus as Metarhizium flavoviride partitivirus 1 (MfPV1), a species in genus Gammapartitivirus, family Partitiviridae. MfPV1 has two double-stranded RNAs as its genome, 1,775 and 1,575 bp in size respectively, encapsidated in isometric particles. When we transfected commercial strains of Metarhizium anisopliae and Metarhizium pingshaense with MfPV1, conidiation was significantly enhanced (t test; P-value < 0. 01), and the significantly higher mortality rates of the larvae of diamondback moth (Plutella xylostella) and fall armyworm (Spodoptera frugiperda), two important lepidopteran pests were found in virus-transfected strains (ANOVA; P-value < 0.05). Transcriptomic analysis showed that transcript levels of pathogenesis-related genes in MfPV1-infected M. anisopliae were obviously altered, suggesting increased production of metarhizium adhesin-like protein, hydrolyzed protein, and destruxin synthetase. Further studies are required to elucidate the mechanism whereby MfPV1 enhances the expression ofpathogenesis-related genes and virulence ofMetarhizium to lepidopteran pests. This study presents experimental evidence that the transfection of other entomopathogenic fungal species with a mycovirus can confer significant hypervirulence and provides a good example that mycoviruses could be used as a synergistic agent to enhance the biocontrol activity of entomopathogenic fungi. Abstract

Silver nanoparticles (AgNPs) are present in biosolids from wastewater treatment facilities, a common soil amendment. Exposing earthworms (Eisenia fetida) to AgNP and AgNO3 in soil with 0 and 7.5 g biosolids kg−1 for 28 days showed AgNO3 was more lethal to earthworms in artificial soil (LC20 ≤ 325 mg Ag kg−1) than natural soil (LC20 ≥ 573 mg Ag kg−1). In contrast, AgNPs were more lethal in natural soil (LC20 ≤ 425 mg Ag kg−1) than artificial soil (LC20 ≥ 653 mg Ag kg−1). Earthworm response to silver in artificial soil may not reflect toxicity in biosolids-amended natural soils. [ABSTRACT FROM AUTHOR]

Abstract

BACKGROUND: Facultative bacterial endosymbionts have the potential to influence the interactions between aphids, their natural enemies, and host plants. Among the facultative symbionts found in populations of the grain aphid Sitobion avenae in central Chile, the bacterium Regiella insecticola is the most prevalent. In this study, we aimed to investigate whether infected and cured aphid lineages exhibit differential responses to wheat cultivars containing varying levels of the benzoxazinoid DIMBOA (2,4‐dihydroxy‐7‐methoxy‐2H‐1,4‐benzoxazin‐3(4H)‐one), which is a xenobiotic compound produced by plants. Specifically, we examined the reproductive performance responses of the most frequently encountered genotypes of Sitobion avenae when reared on wheat seedlings expressing low, medium, and high concentrations of DIMBOA. RESULTS: Our findings reveal that the intrinsic rate of population increase (rm) in cured lineages of Sitobion avenae genotypes exhibits a biphasic pattern, characterized by the lowest rm and an extended time to first reproduction on wheat seedlings with medium levels of DIMBOA. In contrast, the aphid genotypes harbouring Regiella insecticola display idiosyncratic responses, with the two most prevalent genotypes demonstrating improved performance on seedlings featuring an intermediate content of DIMBOA compared to their cured counterparts. CONCLUSION: This study represents the first investigation into the mediating impact of facultative endosymbionts on aphid performance in plants exhibiting varying DIMBOA contents. Abstract

The edible caterpillar Aegocera rectilinea Boisduval 1836, locally called Mikombidila, is collected from its wild habitat for consumption by people in the western region of the Democratic Republic of Congo (DRC). Its production by farming would represent a valuable source of animal protein for local populations. Working toward this end, the present study examined the life cycle of A. rectilinea, fed on Boerhavia diffusa L. leaves in an uncontrolled breeding room (26±2 °C, 77±6% relative humidity), starting with caterpillars previously harvested in Kinshasa, DRC. The fecundity of the females was 340.8 eggs on average, with an egg viability of 89.7%. Developmental phase durations of 19.8±1.6 and 12.5±1.9 days were observed for the larval and pupal stages, respectively. The mean total life-cycle duration, from egg to adult emergence, was 36.2±2.3 days, with a proportion of female of 0.785%. Adult longevity was 8.2±0.8 days for males and 8.0±2.0 days for females. The results of this study validate the interest for A. rectilinea rearing program to reduce food insecurity and protein deficiencies, particularly in western areas of the DRC.
