Polyaluminum chloride (PAC) is claimed to be superior to conventional coagulants because of higher removal of particulate and/or organic matters as well as inherent advantages of lower alkalinity consumption and lesser sludge production. 1000 mL of the reaction mixture was examined using parameters, including PAC dose (5–10 mg/L), pH (4–9), and turbidity (1.9 NTU). The content was stirred at 120 rpm for 1 min. Thereafter, the turbidity of water samples was measured using a P2100 turbidimeter. Data indicated that the maximum removal efficiency of turbidity (97.74%) obtained under the PAC doses of 4 and 10, and the pH of 8. There is not a significant relationship between the different dosages of PAC (P-value > 0.05), but the influence of pH on the removal of turbidity was significant (P-value < 0.05). Based on the dataset, the removal efficiency of turbidity was depended on PAC and pH. Keywords: Polyaluminum chloride, Turbidity, Coagulation, Floculation

The heat stability of whey protein emulsions remains a real challenge due to the rapid denaturation/aggregation of native whey proteins on heating. The use of heat-stable Pickering-like whey protein microgels (WPM) makes it possible to develop heat-stable emulsions in a large range of whey protein concentrations. In this study, emulsion heat stability was evaluated with a special focus on the contribution of WPM adsorbed at the fat droplet surface and in the continuous phase of the emulsion. Dairy emulsions were prepared with 30% milk fat and 70% suspension of WPM in the dispersed phase of milk. The protein interfacial load and the composition of the fat droplet surface were determined immediately after emulsion formation, and the heat stability of the emulsions at 120 °C was assessed visually and at microscopic scale. WPM are heat stable in the continuous phase of the emulsion, but the presence of WPM at the surface of the fat droplets is responsible for a rapid gelation of the emulsions. In the heated emulsions, the fat droplets seemed to be crosslinked by WPM. The presence of caseins instead of WPM at the fat droplet surface allowed the heat stability of the emulsion to recover at low and high whey protein concentrations. This study shows that it is possible to prepare heat-stable whey protein–rich emulsions by using whey proteins previously aggregated as heat-stable WPM and a sufficient amount of caseins in order to fully cover the fat droplet surface. These results will contribute to the development of heat-stable whey protein–rich emulsions.

International audience; Arthrospira platensis, or spirulina, is a cyanobacterial species mainly exploited for its nutritional qualities and able to produce exopolysaccharides (EPS) under specific conditions. In this work we identify that the bicarbonate concentration is a key parameter that induces EPS production by the cells. Using flocculation/flotation harvesting experiments, in addition to atomic force microscopy to characterize the biophysical properties of EPS, we show that EPS produced by A. platensis cells form a soft, adhesive gel in the medium that forms aggregates in which cells are entrapped and thus bioflocculated. This results in the creation of microalgae flocs in which ascending microbubbles produced in the flotation process get trapped, thus enhancing the efficiency of the microalgae separation from water. This interdisciplinary study provides new insights into culture conditions that positively influence the EPS production, while allowing both a high final biomass concentration and a harvesting efficiency superior to 90% to be reached.

Mineralization of flocculated bitumen emulsions allows producing bitumen powders, also referred to as Bitumen@SiO2 hybrid particles, that can resist storage in hard conditions, with a load up to 8 kPa at 50 °C during 20 h, without alteration of their morphology or release of their bitumen core. Thanks to the flocculation, final Bitumen@SiO2 powders are not highly pulverulent and allow transportation and handling of bitumen at ambient temperature without the need of water as it is the case for bitumen emulsions. This “water free” storage particularity exhibits the double advantage of reducing the cost of transport and of avoiding inhibition of bitumen adhesion at mineral aggregates through the presence of water, therefore rendering the bitumen its high quality of binder. The silica shell is responsible for the high storage stability and, when mixed with aggregates for end use of asphalt, as for example in pavement applications, serves as mineral charge to the bitumen, modifying the bitumen flow properties. Indeed, when heated at 100 °C and crushed, the capsules break releasing thus the bitumen. This suspension made of bitumen and silica shell fragments exhibits an interesting shear-thinning behavior that can be attributed to the presence of the remnant fragments. Indeed, when shell fragments have been transferred into a silicon oil, initially exhibiting a Newtonian behavior, the mixture of fragments and oil also exhibited a shear thinning rheogram demonstrating the effect of the silica shell fragments. The benefit of such a behavior modification is the possibility to reduce or avoid the usual addition of fillers. Passive adhesion has been tested by generating asphalt concretes with Bitumen@SiO2 powders and showed high stability. Mechanical tests of cylindrically shaped concretes demonstrate the outstanding mechanical resistance and the high potentiality of the proposed bitumen powders.

The knowledge of suspended particulate matter (SPM) and turbidity dynamics in coastal waters is essential for studying marine ecosystems. Flocculation/deflocculation processes are crucial mechanisms controlling the dynamics of SPM physical characteristics and thus, the fate of these SPM in the environment. This PhD thesis focuses on the SPM characteristics and their dynamics in coastal waters, from tidal to annual scales, in response to hydrodynamic, hydrological and biological forcing that take place at the interface between estuaries and coastal seas. To this end, optical and acoustic sensors were deployed in situ through 6 field campaigns in 2016, in order to characterize and quantify SPM in the water column, near the mouth of the Seine estuary (France). A critical analysis of LISST-100X measurements in coastal waters is presented. The quantification of uncertainties on SPM concentration measurements is investigated: it highlights the crucial effect of salt retention, and the need for a minimum mass to filter, around 10 mg. A method for estimating an optimal filtration volume based on a reference turbidity measurement is proposed. High frequency measurements show that the dynamics of SPM and median diameter are controlled by the advection-flocculationsedimentation- resuspension cycle. These results allow to classify the factors controlling flocculation processes. Turbulence is identified as the main factor at the semi-diurnal and semi-lunar tidal scales, as the maximum median size of flocs decreases when the tidal currents intensify. At the seasonal scale, the variation of SPM characteristics (size, density, settling velocity) is correlated to the variability of the organic matter (OM) content: in particular, flocculation is enhanced by an increase of the particular OM fraction. The resulting larger and lesser dense flocs are also more resistant to the fragmentation induced by shear. This work also investigates the fractal approach applied to flocs. The fractal dimension variability, calculated by combining in situ data of SPM concentration and particle size distribution, can be associated to variations of the SPM composition, but can also result from uncertainties linked to instrument limitations. The latter are discussed in this PhD thesis. The seasonal variability of SPM characteristics is more pronounced offshore than at the mouth of the Seine estuary. From short-term observations in the Seine Bay and from long-term series in the Belgian coastal zone provided by the RBINS, optical turbidity and acoustic backscatter measurements are combined. They highlight an increase of the acoustic backscatter intensity when mean floc density decreases., La connaissance de la dynamique des matières en suspension (MES) et des turbidités en milieu côtier est un élément clé pour comprendre les écosystèmes marins. Les processus de floculation/défloculation représentent des mécanismes essentiels contrôlant la dynamique des caractéristiques physiques des MES et, par conséquent, leur devenir dans l’environnement. Cette thèse a pour objectif de mieux comprendre la caractérisation des MES et leur variabilité de l’échelle tidale à l’échelle annuelle en mer côtière, en réponse aux différents forçages hydrodynamiques, hydrologiques et biologiques ayant lieu à l’interface entre estuaire et baie. À cette fin, des capteurs optiques et acoustiques ont été déployés in situ, lors de 6 campagnes en mer réalisées en 2016, en vue de caractériser et de quantifier les MES dans la colonne d’eau, à la sortie de l’embouchure de l’estuaire de Seine (France). Une analyse critique des mesures granulométriques issues du LISST- 100X est proposée. Un travail de quantification des incertitudes de mesure associées à l’estimation de la concentration en MES a mis en avant le risqué lié à un rinçage insuffisant des filtres et a conduit à définir une masse minimum à filtrer, de l'ordre de 10 mg, quelle que soit la concentration. Ce travail a notamment permis de proposer une méthode d’estimation d’un volume optimal de filtration basée sur une mesure préalable de turbidité de référence. Les mesures à haute fréquence ont montré que la dynamique des caractéristiques physiques des MES en termes de concentration et de taille médiane sont contrôlées par le cycle advection-floculation-sédimentation-remise en suspension. Ces résultats ont permis de hiérarchiser les paramètres de contrôle des processus de floculation. La turbulence a été identifiée comme le principal paramètre de contrôle à l’échelle tidale et semi-lunaire, la taille médiane maximale des flocs étant inversement corrélée à l’intensité des courants. À l’échelle saisonnière, la variation des caractéristiques des MES (taille, densité, vitesse de chute) est liée à la variabilité du contenu en matière organique (MO), avec une floculation favorisée quand le taux de MO particulaire augmente. Ces flocs plus grands et moins denses sont également plus résistants à la fragmentation induite par cisaillement turbulent. Ces travaux ont également permis de discuter le concept de structuration fractale des flocs. La dynamique de la dimension fractale, déterminée en combinant des données in situ de concentration en MES et de distribution en classe de taille, peut traduire des variations de composition des MES, mais peut aussi résulter d'incertitudes de mesure associées aux instruments. Ces dernières sont discutées dans cette thèse. La variabilité saisonnière des caractéristiques des MES est plus prononcée au large que dans la zone sous influence de l’estuaire de Seine. À partir des observations ponctuelles en Baie de Seine et d’une série de mesure long terme dans la zone côtière belge acquise par le laboratoire RBINS, les méthodes acoustiques et optiques sont combinées. Ces dernières ont mis en avant une augmentation de la rétrodiffusion acoustique lorsque la densité moyenne des flocs diminue.

International audience; In the context of climate change, the interest for sustainable sources to produce energy is growing. One promising resource for biofuel production is microalgae, but their industrial use is limited by the lack of efficient harvesting techniques. In this study, we use a multi-scale approach to understand the magnesium hydroxide-mediated flocculation/flotation mechanism of Phaeodactylum tricornutum, an effective oil-producer diatom, under high pH. While flotation experiments give a population-scale quantification of the efficiency of flocculation/flotation using magnesium or calcium hydroxide, or at increased pH, AFM allows probing the mechanical properties of the cells at different pH values. Finally we develop an original strategy to functionalize AFM tips with hydroxide particles that we use in multiparametric imaging experiments to understand at the molecular scale the forces driving the adhesion of hydroxide particles to cells. Altogether, our results give a better understanding of the molecular mechanism underlying alkaline flocculation/flotation, paving the way towards the development of low-cost flocculant-free flotation harvesting processes.

Medienos plokščių gamyba užsiimanti įmonė naudoja šlapio veikimo elektrostatinį filtrą, kuriame susidaro nuotekos (15 m3 / dieną) . Nuotekos surenkamos į specialias talpas ir išvežamos apdoroti. Atsižvelgiant į įmonės žiedinės ekonomikos politiką, norima iš nuotekų atskirti vandenį nuo esančių skendinčių medžiagų (dumblo) ir panaudoti jį kitiems procesams. Darbo tikslas – parinkti medienos drožlių plokščių bei baldų gamybos įmonės šlapio veikimo elektrostatinio filtro nuotekų valymo metodą. Darbo metu nustatyta, kad naudojant flokuliacijos metodą (esant 1,67 mg/l koncentracijos SP73A flokuliantą) pasiekiamas 99,7 % nuotekų išvalymo efektyvumas. Susidarantis dumblo tūris yra 19,4 %. Naudojant koaguliacijos metodą (esant 600 mg/l geležies (III) chlorido koncentracijai) pasiekiamas 96,9 % nuotekų išvalymo efektyvumas. Susidarantis dublo tūris yra 12,3 %. Naudojant flokuliacijos metodą ir įvertinus sunaudojamų reagentų kiekį pH korekcijai nuotekų valymui, 1 m3 išvalymas kainuotų 0,34 Eur/m3 nuotekų, o naudojant koaguliacijos metodą su nuotekų pH korekcija 1 m3 išvalymas kainuotų 2,42 €/m3 nuotekų, tad taikant flokuliacijos metodą nuotekoms valyti yra sutaupoma 7 kartus. Taip pat flokuliacijos metodas sunaudoja mažiau aplinkai kenksmingų cheminių medžiagų. Nuotekos iš šlapio veikimo elektrostatinio filtro gali būti panaudojamos klijų gamybai. Naudojant flokuliacijos metodą, vandens antriniam panaudojimui, būtų galima gražinti 4409,2 m3, o naudojant koaguliacijos metodą vandens antriniam panaudojimui, būtų galima gražinti 4799,75 m3., The wood-based manufacturing company uses a wet electrostatic percipitator, which produces wastewater (15 m3/day). Wastewater is collected in special containers and exported to be treated. Depending on the company's circular economy policy, it is desirable to separate water from existing sediment materials (sludge) from waste water and use it for other processes. The purpose of the study is to modify the process of cleaning the wet electrostatic precipitator from the chipboard and furniture manufacturing company. The study showed that the effluent treatment efficiency of 99.7 % was achieved using the flocculation method (at 1.67 mg/l SP73A flocculant concentration). The volume of sludge is 19.4%. Using a coagulation method (600 mg/l for iron (III) chloride concentration) wastewater cleaning efficiency is 96.9 % . The volume of sludge is 12.3 %. The flocculation method costs 0.34 Eur/m3 of wastewater and the coagulation method costs 2.42 Eur/m3 of wastewater, which means that the flocculation method saves 7 times its cost to clean up the wastewater. The flocculation method also uses less environmentally harmful chemical compounds. Wastewater from a wet electrostatic precipitator can be used for adhesives manufacturing. Using the flocculation method, 4409,2 m3 of water can be recycled for secondary reuse, and using a coagulation method, 4799,75 m3 of water can be reuse.

National audience; La réalisation de travaux de génie maritime et portuaire et la conservation des profondeurs de navigation dans les voies d'accès maritimes, des canaux et des fleuves ou des hauteurs d'eau de production dans les retenues hydroélectriques sont des opérations indispensables pour l'économie, l'environnement et le développement d'un pays et/une région. Ces opérations génèrent des volumes de sédiments de dragage à teneur en eau élevée. Se posent alors les questions de leur transport et de leur devenir. En effet, d'une part, ils ont des teneurs en eau si élevée qu'ils restent difficilement transportables et d'autre part, toute valorisation ou stockage des sédiments implique de réduire la teneur en eau. Une déshydratation mécanisée en continu répondrait à cette attente. En développant des machines qui permettent de déshydrater en continu (24h/24h), cette technique paraît très avantageuse. Des presses à boues ont été testées pour des sédiments de dragage en laboratoire. Un modèle de presse KDS ® , dérivée du principe de la presse à boues Volute ® a fait l'objet d'essais de faisabilité et d'optimisation. La présente communication porte sur la définition de cet outil de déshydratation, sur la recherche des paramètres fondamentaux et l'analyse de résultats d'essais effectués sur des argiles et sédiments à teneur en eau élevée.

During winemaking Saccharomyces cerevisiae strains are exposed continuously to environmental changes and this microorganism responds modifying its transcriptional profile. Yeast flocculation is considered a social trait that allows the cells to escape from hostile conditions by sedimentation. This behaviour is based on the self-interaction of flocculins, proteins encoded by FLO family genes. These are considered responsible of the facultative helping type cooperation and were designed as green-beard genes. In order to understand the role of flocculation to stress response, the genome wide expression analysis of a wine flocculent S. cerevisiae F6789A strain and its FLO5 deleted strain (F6789A-Delta flo5) were determined, using DNA microarray technology. Results highlighted that F6789A strain showed a shorter lag phase in winemaking condition. The comparison of transcriptomic profiles underlined that, while F6789A-Delta fio5 strain seemed engaged in the re-organization of the cell wall and in finding different adhesion ways, F6789A strain presented an up-regulation of genes involved in stress response and higher alcohol production.

The objective of this study is to analyze a model of the chemostat involving the attachment and detachment dynamics of planktonic and aggregated biomass in the presence of a single resource. Considering the mortality of species, we give a complete analysis for the existence and local stability of all steady states for general monotonic growth rates. The model exhibits a rich set of behaviors with a multiplicity of coexistence steady states, bi-stability, and occurrence of stable limit cycles. Moreover, we determine the operating diagram which depicts the asymptotic behavior of the system with respect to control parameters. It shows the emergence of a bi-stability region through a saddle-node bifurcation and the occurrence of coexistence region through a transcritical bifurcation. Finally, we illustrate the importance of the mortality on the destabilization of the microbial ecosystem by promoting the washout of species., L'objectif de cette étude est d'analyser un modèle du chémostat impliquant la dynamique d'attachement et de détachement de la biomasse planctonique et agrégée en présence d'une seule ressource. En considérant la mortalité des espèces, nous donnons une analyse complète de l'existence et de la stabilité locale de tous les équilibres pour des taux de croissance monotones. Le modèle pré-sente un ensemble riche de comportements avec multiplicité d'équilibres de coexistence, bi-stabilité et apparition des cycles limites stables. De plus, nous déterminons le diagramme opératoire qui dé-crit le comportement asymptotique du système par rapport aux paramètres de contrôle. Il montre l'émergence d'une région de bi-stabilité via une bifurcation noeud col et l'occurrence d'une région de coexistence via une bifurcation transcritique. Enfin, nous illustrons l'importance de la mortalité sur la déstabilisation de l'écosystème microbien en favorisant le lessivage des espèces.

Flocculation is a key process in numerous environmental and industrial technologies such as purification of waste-water or paper making. It is necessary to understand the formation and structure of the aggregates to control and optimize such a process. Most of the studies on flocculation involve spherical particles, but there is a clear need to understand the flocculation of anisotropic particles such as clay colloids, which are platelets. I studied the flocculation of montmorillonite clay suspensions in water by well-defined cationic polyelectrolytes called ionenes. By using this model system, it is possible to study the effect of relative charge densities and sizes of ionene chains and clay platelets, on the mode of flocculation and their arrangement inside the aggregates formed. Initially, I studied the effect of charge density of polyelectrolytes on the flocculation efficiency of clay platelets, by combining measurements of turbidity and zeta potential. While the aggregates are always ionene-deficient, the charge density of ionenes leads to variations in the mode of flocculation and in the optimum flocculation conditions. The conformation of the ionene chains inside the aggregates depends on the charge density of ionenes and this is why I continued the study by small-angle scattering experiments in order to correlate the conformation of the chains with the structure of aggregates on different spatial scales. Small-angle X-ray scattering showed the presence of platelet stacks in a face-face configuration, demonstrating itself as a clear stacking peak in the scattering spectra. Further, this technique allowed to highlight the effect of the charge density on the interlamellar spacing inside these stacks. For the most weakly charged ionenes, loops of the ionene chains between the faces of adjacent platelets induce a higher interlamellar spacing inside the stacks. The differences of conformation of ionenes are confirmed by small-angle neutron scattering. This technique, under the condition of contrast matching, allows to probe only ionene chains within the aggegates aggregates. On a larger spatial scale, I have observed that by matching closely the charge density on ionene chains and on clay platelets the most compact aggregates are produced. I have also studied the behaviour of the different ionenes in aqueous solution in absence of clay particles by small-angle neutron scattering and NMR. At high chain concentrations, these systems clearly show differences in inter-chain interactions, depending on the nature of their counterions. The evolution of these interactions follows closely the Hofmeister series.; La floculation est un procédé que l’on peut retrouver dans plusieurs applications au niveau industriel comme la fabrication du papier ou le traitement de l’eau. Il est nécessaire de comprendre la formation et la structure des agrégats floculés pour maîtriser et optimiser un tel procédé. La plupart des études sur la floculation implique des particules sphériques, impliquant ainsi un manque de connaissance sur la floculation de particules anisotropes comme par exemple des plaquettes de colloïdes argileux. C’est pour cela que dans cette étude, j’ai étudié la floculation d’une suspension d’argile type Montmorillonite dans l’eau par des polyélectrolytes cationiques modèles appelés ionènes. En utilisant ce système modèle, il est possible d’étudier l’effet de la densité de charges des ionènes, la taille des plaquettes d’argile ou encore la longueur des chaînes sur le mode de floculation et l’arrangement des chaînes et des plaquettes au sein des agrégats. Dans un premier temps, l’effet de la densité de charge des polyélectrolytes sur l’efficacité de floculation a été étudié en combinant des mesures de turbidité et de potentiel zêta. Ces mesures ont permis de montrer que la densité de charge des ionènes influence le mode de floculation et les conditions de floculation optimale. La conformation des chaînes dépend de la densité de charge des ionènes et c’est pour cela que j’ai poursuivi l’étude par des mesures de diffusion de rayonnement aux petits angles afin de pouvoir corréler l’effet de la conformation des chaînes avec la structure des agrégats à différentes échelles. La diffusion des rayons X aux petits angles a permis de montrer la présence d’organisation des plaquettes sous forme d’empilement face-face. Ensuite, cette technique a permis de souligner l’effet de la densité de charge sur la distance entre plaquettes dans les empilements. Pour les ionènes les moins chargés, la formation de boucles de chaînes des ionènes entre les faces d’argile induit les distances entre plaquettes les plus grandes au sein des agrégats. Les différences de conformations des différents ionènes ont été confirmées par diffusion de neutrons aux petits angles. Cette technique, sous la condition de variation de contraste, permet de sonder uniquement les chaînes dans les agrégats. J’ai également étudié le comportement des différents ionènes en solution aqueuse en absence des particules d’argile par diffusion de neutrons aux petits angles et RMN. Dans le domaine des hautes concentrations, ces systèmes montrent clairement des différences d’interaction entre les chaînes en fonction de la nature des contre-ions. L’évolution de ces interactions suit parfaitement la série de Hofmeister.

International audience; Les boues urbaines, déchets issus du traitement des eaux usées, sont généralement déshydratées mécaniquement. Afin de les déshydrater une étape de floculation est quasi-systématiquement nécessaire. Or l'optimisation de cette floculation reste très empirique. Dans ce travail, nous utilisons un nouvel indice, l'indice d'égouttage, qui permet de quantifier la qualité d'une floculation pour le procédé d'égouttage, pour déterminer les paramètres optimaux (choix du polymère, dose de polymère, vitesse et durée de mélange du polymère avec la boue) de floculation lors d'une déshydratation par pressage en cellule de filtration-compression (CFC). Par ailleurs, nous montrons que lors des essais de compression de la boue urbaine étudiée en CFC, la quantité de matière introduite dans la cellule (qui impacte l'épaisseur finale du gâteau) peut avoir une importance bien plus importante que la pression appliquée pour améliorer la siccité finale de la boue.

The destabilization mechanism of the high stable cutting-oil emulsion by aluminum sulfate (Al-2(SO4)(3)) was investigated since it can affect properties of aggregates and following separation units. Al-2(SO4)(3) dosage and pH were key factors in the destabilization. The effective separation occurred when precipitated Al(OH)(3) is dominated at the neutral pH of 6.5-7.0. The best separation can be achieved when solid flocs were formed at 1.0mM, which exceeded the dosage from the critical coagulation concentration (CCC) of 0.75mM. Two different mechanisms were proved for the emulsion destabilization depending upon the Al3+ concentration under this pH range. The first mechanism was the adsorption of Al(OH)(3) on surface of oil droplets, which led to the droplet coalescence. By increasing the Al3+ dosage, the sweep flocculation by Al(OH)(3) precipitates occurred. Al3+ dosage for effective destabilization was increased in accordance with oil concentration. The formation of aluminum hydroxide precipitates in bayerite structure was affirmed by analyzing elemental composition and crystalline structure of flocs from the destablization.

We study this flocculation model with monotonic growth rates and same removal rates. We show that this model has at most one positive steady state that is stable as long as it exists where planktonic and attached bacteria can coexist. According to control parameters, the operating diagram illustrates the washout and coexistence regions that depend only on the growth rate of planktonic bacteria. Indeed, flocculation process affects only the density of isolated and attached bacteria at steady state.; Dans ce travail, on considère un modèle du chémostat incluant une seule espèce composée de bactéries planctoniques et attachées en compétition pour une seule ressource. Nous étudions ce modèle de floculation avec des taux de croissance monotones et le même taux de disparition. Nous montrons que ce modèle admet au plus un équilibre positif qui est stable dès qu'il existe où les bactéries planctoniques et attachées peuvent coexister. Selon les paramètres de contrôle, le diagramme opératoire illustre les régions de lessivage et de coexistence qui dépendent que du taux de croissance des bactéries planctoniques. En effet, le processus de floculation affecte seulement la densité des bactéries isolées et attachées à l'équilibre.

Flocculation is an important feature for yeast survival in adverse conditions. The natural diversity of flocculating genes in Saccharomyces cerevisiae can also be exploited in several biotechnological applications. Flocculation is mainly regulated by the expression of genes belonging to the FLO family. These genes have a similar function, but their specific contribution to flocculation ability is still unclear. In this study, the distribution of FLO1, FLO5 and FLO8 genes in four S. cerevisiae wine strains was investigated. Subsequently, both FLO1 and FLO5 genes were separately deleted in a flocculent S. cerevisiae wine strain. After gene disruption, flocculation ability and agar adhesion were evaluated. FLO1 and FLO5 genes inheritance was also monitored. All strains presented different lengths for FLO1 and FLO5 genes. Results confirm that in S. cerevisiae strain F6789, the FLO5 gene drives flocculation and influences adhesive properties. Flocculation ability monitoring after a cross with a non-flocculent strain revealed that FLO5 is the gene responsible for flocculation development.

This paper concerns experimental investigation of the sequenced flocculation of latex particles in a Taylor-Couette reactor. The aim of this work was to investigate the evolution of both the size and the shape of aggregates under sequenced hydrodynamics. A number of studies have focused on the evolution of the aggregate size or size distribution during steps of growth-breakage-regrowth, but aggregates generally experience steps of breakage-regrowth on repeated occasions in real operating conditions (passages near the impeller or during the transfer processes, for example). The experiments conducted in this work consisted thus of an alternation of six steps with alternately low and high shear rates under turbulent conditions. The particle size distributions were monitored throughout the sequencing, and the circularity and convexity (shape parameters) distributions were measured, enabling a more precise description of the entire floc population, rather than a fractal dimension. While the aggregate size distribution was clearly controlled by hydrodynamics, the shape distributions continuously evolved during the sequencing. The main new finding of our work notes the independence between the aggregate shape and hydrodynamics. Indeed, after multiples steps of breakage-regrowth, regardless of the aggregate size distribution and hydrodynamics, the aggregate shape seemed to reach a unique steady-state morphological distribution.
