Abstract

Protein kinase such as the casein kinase Ialpha and MEKK1 have synergic roles in suppressing NF-AT. MEKK1 protein kinase indirectly suppresses NF-AT4 nuclear import while casein kinase Ialpha directly binds and phosphorylates the transcription factor NF-AT4 which resulted to the inhibition of the transcription factor's nuclear translocation. These transcriptional factors have important function in maintaining and enhancing the masking activity responsible in controlling NF-AT4.

Abstract

A study was conducted on the gene encoding protein p73. p73 maintains a similar homology to p53, a tumor suppressing protein, and maps to 1p36. Coding sequence mutations in p73 alleles were not observed in examinations of neuroblastoma cell lines with p73 and 1p heterozygosity losses. The monoallelical expression of p73, however, suggests that it a candidate gene in neuroblastoma and that it may interact with p53 and activate p53 target genes.

Abstract

The structure of the capsular polysaccharide of Escherichia coli K5 is similar to that of N-acetyl-heparosan, a nonsulfated precursor of heparin. This makes the E. coli antigen as a good starting point for the chemical synthesis of analogs of low-molecular-weight heparin. This polysaccharide is synthesized as a high-molecular-weight molecule that can be depolymerized by an enzyme with endo-beta-eliminase activity. The eliminase-encoding gene (elmA) was cloned from E. coli K5 by expression in E. coli K-12. The elmA gene product wsa 820 amino acids long. The C terminus and the N terminus were found to be essential for activity and secretion.

Abstract

A heptapeptide coded by rat brain cDNA shows properties of an endogenous agonist of the ORL(sub 1) orphan receptor in cell cultures. The peptide suppresses the release of adenylate cyclase in the CHO(ORL1+) cell line and causes hypoglycemia. This neuropeptide contains additional Lys-Arg residues. Structure of this neuropeptide is similar to that of the peptide dynorphin A. Its effect on pain sensitivity is the reverse of the effect of OTL1 suppression. The suggested name for this peptide is nociceptin.

Abstract

The secondary structure of some protein segments may vary between α-helix and β-strand. To predict these switchable segments, we have developed an algorithm, Switch-P, based solely on the protein sequence. This algorithm was used on the extracellular parts of FGF receptors. For FGFR2, it predicted that β4 and β5 strands of the third Ig-like domain were highly switchable. These two strands possess a high number of somatic mutations associated with cancer. Analysis of PDB structures of FGF receptors confirmed the switchability prediction for β5. We thus evaluated if compound-driven α-helix/β-strand switching of β5 could modulate FGFR2 signaling. We performed the virtual screening of a library containing 1.4 million of chemical compounds with two models of the third Ig-like domain of FGFR2 showing different secondary structures for β5, and we selected 32 compounds. Experimental testing using proliferation assays with FGF7-stimulated SNU-16 cells and a FGFR2-dependent Erk1/2 phosphorylation assay with FGFR2-transfected L6 cells, revealed activators and inhibitors of FGFR2. Our method for the identification of switchable proteinic regions, associated with our virtual screening approach, provides an opportunity to discover new generation of drugs with under-explored mechanism of action.

Abstract

The role of the centrosomes in microtubule nucleation remains largely unknown at the molecular level.[Gamma]-Tubulin and the two associated proteins h103p (hGCP2) and h104p (hGCP3) are essential. These proteins are also present in soluble complexes containing additional polypeptides. Partial sequencing of a 76-kD polypeptide band from these complexes allowed the isolation of a cDNA encoding for a new protein (h76p = hGCP4) expressed ubiquitously in mammalian tissues. Orthologues of h76p have been characterized in Drosophila and in the higher plant Medicago. Several pieces of evidence indicate that h76p is involved in microtubule nucleation. (1) h76p is localized at the centrosome as demonstrated by immunofluorescence. (2) h76p and [Gamma]-tubulin are associated in the [Gamma]-tubulin complexes. (3) [Gamma]-tubulin complexes containing h76p bind to microtubules. (4) h76p is recruited to the spindle poles and to Xenopus sperm basal bodies. (5) h76p is necessary for aster nucleation by sperm basal bodies and recombinant h76p partially replaces endogenous 76p in oocyte extracts. Surprisingly, h76p shares partial sequence identity with human centrosomal proteins h103p and h104p, suggesting a common protein core. Hence, human [Gamma]-tubulin appears associated with at least three evolutionary related centrosomal proteins, raising new questions about their functions at the molecular level.
