Abstract Background Neoadjuvant chemotherapy (NAC) is an effective treatment for locally advanced breast cancer (BC). However, there are no effective biomarkers for evaluating its efficacy. CDR1-AS, well known for its important role in tumorigenesis, is a famous circular RNA involved in the chemosensitivity of cancers other than BC. However, the predictive role of CDR1-AS in the efficacy and prognosis of NAC for BC has not been fully elucidated. We herein aimed to clarify this role. Methods The present study included patients treated with paclitaxel-cisplatin-based NAC. The expression of CDR1-AS was detected by real-time quantitative reverse transcription polymerase chain reaction testing. The predictive value of CDR1-AS expression was examined in pathological complete response (pCR) after NAC using logistic regression analysis. The relationship between CDR1-AS expression and survival was demonstrated using the Kaplan–Meier method, and tested by log-rank test and Cox proportional hazards regression model. Results The present study enrolled 106 patients with BC. Multivariate logistic regression analysis revealed that CDR1-AS expression was an independent predictive factor for pCR (odds ratio [OR] = 0.244; 95% confidence interval [CI] 0.081–0.732; p = 0.012). Furthermore, pCR benefits with low CDR1-AS expression were observed across all subgroups. The Kaplan–Meier curves and log-rank test suggested that the CDR1-AS high-expression group showed significantly better disease-free survival (DFS; log-rank p = 0.022) and relapse-free survival (RFS; log-rank p = 0.012) than the CDR1-AS low-expression group. Multivariate analysis revealed that CDR1-AS expression was an independent prognostic factor for DFS (adjusted HR = 0.177; 95% CI 0.034–0.928, p = 0.041), RFS (adjusted HR = 0.061; 95% CI 0.006–0.643, p = 0.020), and distant disease-free survival (adjusted HR = 0.061; 95% CI 0.006–0.972, p = 0.047). Conclusions CDR1-AS may be a potential novel predictive biomarker of pCR and survival benefit in patients with locally advanced BC receiving NAC. This may help identify specific chemosensitive individuals and build personalized treatment strategies.

Abstract Non-alcoholic fatty liver disease (NAFLD) is becoming a significant global public health threat. Seabuckthorn (Hippophae rhamnoides L.) has been used in traditional Chinese medicine (TCM). The hypolipidemic effects of Seabuckthorn polysaccharides (SP) against high-fat diets (HFD)-induced NAFLD were systematically explored and compared with that of Bifidobacterium lactis V9 (B. Lactis V9). Results showed that HFD-induced alanine transaminase (ALT) and aspartate aminotransferase (AST) levels decreased by 2.8-fold and 4.5-fold, respectively, after SP supplementation. Abstract In forensic genetics, utilizing massively parallel sequencing (MPS) to analyze short tandem repeats (STRs) has demonstrated several advantages compared to conventional capillary electrophoresis (CE). Due to the current technical limitations, although flanking region polymorphisms had been mentioned in several previous studies, most studies focused on the core repeat regions of STRs or the variations in the adjacent flanking regions. In this study, we developed an MPS system consisting of two sets of multiplex PCR systems to detect not only the STR core repeat regions but also to observe variants located at relatively distant positions in the flanking regions. The system contained 42 commonly used forensic STRs, including 21 autosomal STRs (A-STRs) and 21 Y-chromosomal STRs (Y-STRs), and a total of 350 male individuals from a Chinese Han population were genotyped. The length and sequence variants per locus were tallied and categorized based on length (length-based, LB), sequence without flanking region (core repeat regions sequence-based, RSB), and sequence with flanking region (core repeat and flanking regions sequence-based, FSB), respectively. Allele frequencies, Y-haplotype frequencies, and forensic parameters were calculated based on LB, RSB, and FSB, respectively, to evaluate the improvement in discrimination power, heterozygosity, and effectiveness of forensic systems. The results suggested the sequence variations have more influence on A-STRs and could improve the identification ability of MPS-STR genotyping. Concordance between MPS and CE methods was confirmed by using commercial CE-based STR kits. The impact of flanking region variations on STR genotype analysis and potential factors contributing to discordances were discussed. A total of 58 variations in the flanking regions (53 SNPs/SNVs and 5 InDels) were observed and most variations (48/58) were distributed in A-STRs. In summary, this study delved deeper into the genetic information of forensic commonly used STR and advanced the application of massively parallel sequencing in forensic genetics.

Abstract Real-time location tracking is essential for personal security in industrial scenes, e.g. monitoring worker safety in power substations. Ultra wideband (UWB) technology is suitable for indoor positioning thanks to its high penetration capability, high ranging accuracy and low power consumption. However, UWB based trilateration positioning requires high workload for field deployment of base stations. Indoor complex topology results in multipath and non-line of sight (NLOS) conditions of UWB signals, and degrades the positioning performance in terms of accuracy and reliability. This paper proposes a three-dimensional (3D) area recognition solution by integrating UWB time of flight (TOF) ranging and barometer measurements. The proposed solution utilizes a multi-tier distributed joint probabilistic inference model, which accomplishes the indoor 3D area recognition exploiting multiple clustering and prediction algorithms of machine learning. The field experiments showed that the proposed method can achieve an accuracy of 3D area recognition of more than 99.2%. The proposed method improves the computing efficiency by 93%. The errors of improved differential barometric height estimation method are less than 1 m, which means a success rate of 100% for floor identification, given a floor separation of 3–4 m. The proposed solution is suitable for personnel security applications of industrial scenes, which requires reliable real-time area information rather than just coordinates.

The interfacial behaviors between protective coatings and substrate single-crystal superalloys significantly impact their service performances. This work focuses on the elements interdiffusion and microstructural evolution at the interface between a NiCrAlY coating and a series of high-Mo Ni3Al-based single-crystal superalloys, through the coupled phase-field simulations and DICTRA kinetics calculations of their model systems. The critical roles of Al and Cr played in the microstructural evolution at coating/superalloy interface have been confirmed, which requires the appropriate content decreases of Al and Cr in the coating at 1100 °C. However, the addition of Mo reduces the driving force of element diffusion from coating to substrate by increasing the activities of Al and Cr in superalloy. Moreover, increasing the Mo content in superalloy from 8 wt% to 10 wt% could also counteract the promoting effect of Al on Ni mobility and mitigate the γ′ coarsening and detrimental TCP precipitation. Therefore, the effects of Mo enable the reasonable increase of Al content in coating to better balance the interfacial stability and oxidation resistance of coated superalloy, besides its advantage in reducing the anisotropy of TCP precipitate. The obtained interfacial evolution mechanisms resulting from the phase transformation thermodynamics and element interdiffusion kinetics are expected to aid the coating design for advanced single-crystal superalloys servicing at ultra-high temperature.

The inferior creep ductility of Inconel 718 (IN718) fabricated by selective laser melting (SLM), which is unable to meet aerospace material specification (≥4%), limits its engineering application. Despite persistent optimization efforts in the SLM process and post-heat treatment, this issue remains unresolved. This study involved SLM fabrication of the modified IN718 with trace amounts of LaB6 and/or P, followed by homogenization and double aging heat treatment. The addition of trace LaB6 notably influenced the recrystallized grain structure and carbide precipitation at grain boundaries, while trace P addition had minimal impact. All samples exhibited similar γ′ and γ″ strengthening precipitates. Creep tests conducted at 650°C/690 MPa revealed that the individual modification of LaB6 showed limited improvement in creep performance and P showed no improvement, but the combined addition of LaB6 and P led to a substantial enhancement in creep performance, especially reaching as high as 8.6% creep strain. This remarkable improvement in creep ductility is unlikely to arise from the altered grain structure and grain boundary precipitates induced by LaB6 but predominantly arises from the synergistic effect of P and B in enhancing resistance to crack propagation during the third stage of creep.

Abstract The role of learner emotions in language learning has long been observed and researched. Of various emotions, foreign language anxiety, enjoyment and boredom have been the most often researched, while research on other emotions like pride and sadness is hardly available in the current literature. Research on emotions in students with special characteristics can be hardly found as well. Hence, this study collected interview data from 26 and survey data from 520 top students in two universities in China to examine their emotions in English language class. Analyses of the data revealed the following major findings: (a) The participants experienced a diversity of emotions in their English language class due to both learner-internal and external reasons, but the most often reported emotions were anxiety, enjoyment and boredom, (b) significant correlations existed among the students’ English language classroom anxiety, enjoyment and boredom, (c) English language classroom anxiety and boredom significantly negatively while enjoyment significantly positively predicted the students’ English test performance, and (d) English language classroom anxiety debilitated English learning but motivated students to study harder as well; enjoyment facilitated English learning but students might forget much of what had been learned after class; though boredom caused some students to be absent-minded in class, most students would study on their own when feeling bored. These findings further pinpoint the important role and complex nature of learner emotions in second/foreign language learning. Based on these findings, specific suggestions for language teachers and learners are discussed.

Abstract Objectives Newly detected hepatic nodules during follow-up of cancer survivors receiving chemotherapy may pose a diagnostic dilemma. We investigated a series of hepatic focal nodular hyperplasia (FNH) diagnosed by either typical MRI features and follow-up or pathology in cancer survivors. Methods This retrospective study evaluated 38 patients with tumours who developed new hepatic FNH after cyclophosphamide-based (n = 19) and oxaliplatin-based (n = 19) chemotherapies. The main tumour types were breast cancer (n = 18) and colorectal cancer (n = 17). MRI findings, clinical features, and temporal evolution of all target hepatic lesions (n = 63) were reported. In addition, the two chemotherapy drug groups were compared. Results The median interval between chemotherapy completion and FNH detection was 30.4 months (12.9, 49.4). Six patients underwent biopsy or surgery, while the remaining patients were diagnosed based on typical MRI features and long-term follow-up. Among the patients, 60.5% (23/38) presented with multiple nodules and 63 target lesions were detected. The median size of target lesions was 11.5 mm (8.4, 15.1). The median follow-up time was 32.5 months (21.2, 48.6), and 15 patients experienced changes in their lesions during the follow-up period (11 increased and 4 decreased). The cyclophosphamide-based treatment group had a younger population, a greater proportion of females, and a shorter time to discovery than the oxaliplatin-based chemotherapy group (all p ≤ 0.016). Conclusions FNH may occur in cancer survivors after cyclophosphamide- or oxaliplatin-based chemotherapy. Considering a patient’s treatment history and typical MRI findings can help avoid misdiagnosis and unnecessary invasive treatment. Clinical relevance statement When cancer survivors develop new hepatic nodules during follow-up, clinicians should think of the possibility of focal nodular hyperplasia in addition to liver metastasis, especially if the cancer survivors were previously treated with cyclophosphamide or oxaliplatin. Key Points Cancer survivors, after chemotherapy, can develop hepatic focal nodular hyperplasia. Cyclophosphamide and oxaliplatin are two chemotherapeutic agents that predispose to focal nodular hyperplasia development. Objective: Liver cancer is a major health concern globally and in China. This analysis investigated deaths and disability-adjusted life years (DALYs) with respect to etiologies and risk factors for liver cancer in China and worldwide. Methods: Global and China-specific data were collected on liver cancer deaths, DALYs, and age-standardized rates (ASRs) from the Global Burden of Disease Study 2019 database. Liver cancer etiologies were classified into five groups and risk factors were categorized into three levels. Each proportion of liver cancer burden was calculated in different geographic regions. The joinpoint regression model were used to assess the trends from 1990–2019. Results: Liver cancer accounted for 484,577 deaths worldwide in 2019 with an ASR of 5.9 per 100,000 population. China had an elevated liver cancer death ASR in 2019 and males had an ASR 1.7 times the global rate. The global ASR for DALYs peaked at 75–79 years of age but peaked earlier in China. Hepatitis B virus was the prominent etiology globally (39.5%) and in China (62.5%), followed by hepatitis C virus and alcohol consumption. In high sociodemographic index countries, non-alcoholic steatohepatitis has gained an increasing contribution as an etiologic factor. The liver cancer burden due to various etiologies has decreased globally in both genders. However, metabolic risk factors, particularly obesity, have had a growing contribution to the liver cancer burden, especially among males. Conclusions: Despite an overall decreasing trend in the liver cancer burden in China and worldwide, there has been a rising contribution from metabolic risk factors, highlighting the importance of implementing targeted prevention and control strategies that address regional and gender disparities.

Objective: The burden of gastric cancer (GC) across different age groups needs updating. We determined the GC global, regional, and national burden profiles and changes in incidence for 3 sequential 5-year intervals from 2003 to 2017. Methods: The latest incidence and mortality estimates of GC from 185 countries and regions were extracted from the GLOBOCAN 2022 database. The 5-year interval age-standardised incidence rates (ASIRs) were evaluated using cancer registry data from volumes X–XII of the Cancer Incidence in Five Continents (CI5). Correlation analysis was used to evaluate the relationship between ASIR or the age-standardised mortality rate (ASMR) and the Human Development Index (HDI). Results: There was an estimated global 968,000 new GC cases and 660,000 deaths in 2022, with male predominance. GC ASIRs and ASMRs were 9.2 and 6.1 per 100,000 persons, respectively. East Asia had the highest burden, with 53.8% of cases and 48.2% of deaths among all geographic regions. There was a significant correlation between ASIR and HDI. Over three 5-year intervals from 2003 to 2017, the incidence of GC notably decreased in most countries but peaked at 2008–2012 in New Zealand, Turkey, and South Africa. Several countries in Europe, Oceania, and America suggest an increasingly concerning trend among younger individuals, especially females. Conclusions: GC is a significant health issue, especially among males and in geographic regions with an HDI, such as eastern Asia. While the incidence of GC is decreasing in many countries due to prevention efforts and improved treatments, a rising trend persists among younger individuals. Comprehensive prevention strategies tailored to different age patterns are clearly needed.

Objective: This study aimed to provide a comprehensive overview of the global burden of esophageal cancer (EC) and determine the temporal trends and factors influencing changes in the global burden. Methods: The latest incidence and mortality data for EC worldwide were obtained from GLOBALCAN 2022. The mortality and disability-adjusted life years (DALYs) rates for EC from 1990–2019 were sourced from the 2019 Global Burden of Diseases. Trends in EC mortality and DALYs attributable to 11 risk factors or clusters of risk were analyzed using the joinpoint regression model. The trends in age-related EC burden were assessed using a decomposition approach. Results: An estimated 511,054 new cases of EC were diagnosed in 2022 with 445,391 deaths worldwide. Approximately 75% of cases and deaths occurred in Asia. Nearly 50% of global EC deaths and DALYs were attributed to tobacco use in men in 2019, while 20% were attributed to high body mass index (BMI) in women. From 1990–2019, EC deaths and DALYs attributable to almost all risk factors had declining trends, while EC deaths and DALYs attributed to high BMI in men had upward trends. The age-related EC burden exhibited an upward trend driven by population growth and aging, which contributed to 307.4 thousand deaths and 7.2 million DALYs due to EC. Conclusions: The EC burden remains substantial worldwide. Effective tobacco and obesity control measures are critical for addressing the risk-attributable burden of EC. Population growth and aging pose challenges for EC prevention and control efforts.

Abstract Backgroup The pathogenesis of shoulder impingement syndrome (SIS) is still unclear, and its questionable causal relationship with rotator cuff (RC) injury has led to confusion in treatment. The purpose of this study was to explore the bidirectional causal relationship between SIS and RC injury. Methods SIS and RC injury datasets downloaded from the IEU Open GWAS project and GWAS catalog databases. Inverse variance weighted (IVW), MR Egger, Weighted median, and Weighted mode were used in this Mendelian randomization (MR) analysis. Cochran’s Q test, leave-one-out, and funnel plot method were used to evaluate heterogeneity between single nucleotide polymorphisms (SNPs). MR-Egger regression was used to test the horizontal pleiotropy of this study. Results The IVW method (OR = 1.189, P = 0.0059) suggest the putative causal effect of RC injury on SIS. The results of MR Egger method (OR = 1.236, P = 0.2013), weighted median method (OR = 1.097, P = 0.2428) and weighted mode method (OR = 1.013, P = 0.930) showed no statistically significant (OR = 1.069071, P = 0.6173). Heterogeneity test and horizontal pleiotropy analysis suggested that there was no significant heterogeneity and horizontal pleiotropy in the results of this MR analysis. The reverse MR analysis showed heterogeneity, and the conclusion needs to be further explored. Conclusions The results of MR analysis support that RC injury may be causally associated with SIS.

Abstract Background Tissue plasminogen activator (tPA) is recommended as the preferred thrombolytic therapy for acute myocardial infarction (AMI). This study aimed to explore tPA-related adverse events (AEs) reported in the United States Food and Drug Administration Adverse Event Reporting System (FAERS), assess the potential safety of three preferred tPA therapies for treating AMI, and provide guidance for selecting tPA for prehospital thrombolysis. Method Four algorithms, including ROR, PRR, BCPNN, and MGPS, were used to quantify the signals of Tenecteplase, Reteplase, and Alteplase related AEs and compare the differential degrees of the three tPA-associated AEs in the actual data. Result We detected 18 signals of Tenecteplase-induced AE, 29 signals of Reteplase-induced AE, and 22 signals of Alteplase-induced AE. Among the three drugs, Tenecteplase had the highest signal intensity for intracranial hemorrhage-related AE, followed by Alteplase. Besides, Reteplase had the highest signal intensity for procedure-related AE and Alteplase had the highest signal intensity for arrhythmia-related AE. The time-onset analysis indicates that we should be vigilant for AEs, especially within the first week and the first 1–2 days after medication. Conclusion This study identified and compared the signals of AE related to Tenecteplase, Reteplase, and Alteplase in AMI patients.

Abstract The free-field switching of the perpendicular magnetization by the out-of-plane polarized spin current induced spin-orbit torque makes it a promising technology for developing high-density memory and logic devices. The materials intrinsically with low symmetry are generally utilized to generate the spin current with out-of-plane spin polarization. However, the generation of the out-of-plane polarized spin current by engineering the symmetry of materials has not yet been reported. Here, we demonstrate that paramagnetic CaRuO3 films are able to generate out-of-plane polarized spin current by engineering the crystal symmetry. The non-uniform oxygen octahedral tilt/rotation along film’s normal direction induced by oxygen octahedral coupling near interface breaks the screw-axis and glide-plane symmetries, which gives rise to a significant out-of-plane polarized spin current. This spin current can drive field-free spin-orbit torque switching of perpendicular magnetization with high efficiency. Our results offer a promising strategy based on crystal symmetry design to manipulate spin current and could have potential applications in advanced spintronic devices.

Abstract Maltotetraose amylase (Mta) catalyzes the hydrolysis of amylaceous polysaccharides into maltotetraose, which is an important functional sugar used in the food industry. However, the lack of efficient expression systems for recombinant Mta has hindered its scale-up production and application. In this study, a codon-optimized mta gene from Pseudomonas saccharophila was efficiently produced in Bacillus subtilis by optimizing the regulatory elements. First, a plasmid library containing 173 different signal peptide sequences placed upstream of mta gene was constructed, and transformed into B. subtilis strain WB800N(amyEΔ1) for high-throughput screening. The signal peptide yhcR was found to significantly enhance the secretion of Mta, reaching an activity of 75.4 U/mL in the culture medium. After optimization of the promoters, the Mta activity was further increased to 100.3 U/mL using a dual-promoter PHpaIIPamyE. Finally, the carbon sources and nitrogen sources for recombinant Mta production were optimized, yielding a highest Mta activity of 288.9 U/mL under the optimal culture conditions. The crude enzyme solution containing recombinant Mta produced a highest maltotetraose yield of 70.3% with 200 g/L of maltodextrin as the substrate. Therefore, the present study have demonstrated a high yield of Mta produced in B. subtilis, laying the foundation for large-scale Mta production and application.

In order to explore a new way for the application of phosphogypsum in road materials, phosphogypsum powder and asphalt mortar were mixed to prepare phosphogypsum modified asphalt. The effects of mixing temperature, powder-binder ratio and polyphosphoric acid content on the three major indicators of phosphogypsum modified asphalt were studied. The research results show that the softening point increases as the mixing temperature increases, and the ductility increased first and then kept constant. Considering energy efficiency and aging temperature, the processing temperature of asphalt should be in the range 135 °C and 145 °C. increasing the powder gum ratio can increase the softening point of the modified asphalt and decrease the ductility of the asphalt. Therefore, the ratio of filler bitumen should not be greater than 0.9 using phosphogypsum as the modifier. The addition of polyphosphoric acid greatly increases the softening point of modified asphalt, but reduces its ductility and penetration. The optimal amount of polyphosphoric acid should be around 1.5%. Infrared spectroscopy (FTIR) and thermogravimetric analysis (TG-DSC) showed that phosphogypsum physically functioned with Asphalt. When polyphosphoric acid was mixed into asphalt, it would react chemically with asphalt. The research results could provide references for the engineering application and theoretical research of phosphogypsum in road materials.

The study of the hydro-chemical characteristics of burnt rock aquifers helps to distinguish the hydraulic connections between different aquifers. Taking Zhangjiamao Coal Mine in Shenfu Mining Area as an example, 29 groups of water samples were collected from weathered bedrock, bedrock, 2−2 coal burned rock and 3−1 coal burned rock aquifers, and the hydro-chemical characteristics and control factors in the study area were analyzed by comprehensive use of statistical analysis, Piper trilinear diagram, Gibbs model and hybrid model, and the source of main ions was determined, based on which the contact relationship of each aquifer was identified. The results indicated that the total dissolved solids (TDS) of the weathered bedrock and the 2−2 coal burnt rock aquifer were close, slightly lower than the TDS of the bedrock aquifer, while the TDS of the 3−1 coal burnt rock aquifer was the highest, showing a trend of gradually increasing TDS with the increase of aquifer depth. The cations in the weathered bedrock and the 2−2 coal burnt rock aquifer were mainly Ca2+, while the cations in the bedrock and 3−1 coal burnt rock aquifer were mainly Na+; the anions in the weathered bedrock, 2−2 coal burnt rock, and 3−1 coal burnt rock aquifers were mainly HCO3−, while the bedrock aquifers were mainly HCO3−, and SO42−. The hydro-chemical types of weathered bedrock, 2−2 coal burnt rock, and 3−1 coal burnt rock aquifers were mainly HCO3−Ca type, HCO3−Ca and HCO3−Ca·Mg type, HCO3−Ca and HCO3−Na·Mg type. The hydro-chemical composition of weathered bedrock and 2−2 coal burnt rock aquifer was mainly controlled by water rock interaction, while the hydro-chemical composition of bedrock and 3−1 coal burnt rock aquifer was mainly influenced by both water rock interaction and concentration. The main ions of 3−1 coal burned rock, bedrock, weathered bedrock and 2−2 coal burned rock aquifer were respectively from the dissolution of silicate rock salt, the dissolution of silicate rock and evaporite rock salt, and the weathering dissolution of silicate rock and carbonate rock salt. At the same time, the reverse cation exchange interaction effect occurred in the groundwater disasters of each aquifer. Multiple indicator results showed that the ion composition of the weathered bedrock and the aquifer of the 2−2 coal fired rock was similar, and there was a strong hydraulic connection. There may be a direct contact relationship locally, which is different from the ion composition of the aquifer of the 3−1 coal fired rock. There is a certain barrier layer between them, and the hydraulic connection between them is weak.

Abstract Abdominal adhesions manifests following abdominal infections triggered by intestinal fistulas. The severity of such adhesions depends on the extent of fiber deposition and peritoneal fibrinolysis following peritoneal injury, which may be influenced by sustained inflammation within the abdominal cavity. In this regard, the visceral-to-subcutaneous fat area (VFA/SFA) ratio has been implicated as a potential marker of inflammation. This study aimed to explore the relationship between VFA/SFA and abdominal adhesions. This multicenter study was conducted across four tertiary institutions and involved patients who had undergone definitive surgery (DS) for intestinal fistula from January 2009 and October 2023. The presence of abdominal adhesions was determined intraoperatively. VFA/SFA was investigated as a potential risk factor for severe adhesions. The study comprised 414 patients with a median age of 50 [interquartile range (IQR) 35–66] years and a median body mass index of 20.0 (IQR 19.2–22.4) kg/m2, including 231 males with a median VFA/SFA of 1.0 (IQR 0.7–1.2) and 183 females a median VFA/SFA of 0.8 (0.6–1.1). VFA/SFA was associated with severe abdominal adhesions in males [odds ratio (OR) = 3.34, 95% CI 1.14–9.80, p = 0.03] and females (OR = 2.99, 95% CI 1.05–8.53, p = 0.04). J-shaped association between VFA/SFA ratio and severe adhesions was revealed in both sex. The increasing trend can be revealed when OR more than 0.8, and 0.6 in males and females respectively. Preoperative VFA/SFA demonstrates predictive value for statues of severe abdominal adhesions in DS for anastomotic fistula after small intestine resection.

Abstract Background Numerous insect species undertake long-distance migrations on an enormous scale, with great implications for ecosystems. Given that take-off is the point where it all starts, whether and how the external light and internal circadian rhythm are involved in regulating the take-off behaviour remains largely unknown. Herein, we explore this issue in a migratory pest, Cnaphalocrocis medinalis, via behavioural observations and RNAi experiments. Results The results showed that C. medinalis moths took off under conditions where the light intensity gradually weakened to 0.1 lx during the afternoon or evening, and the take-off proportions under full spectrum or blue light were significantly higher than that under red and green light. The ultraviolet-A/blue light–sensitive type 1 cryptochrome gene (Cmedcry1) was significantly higher in take-off moths than that of non-take-off moths. In contrast, the expression of the light-insensitive CRY2 (Cmedcry2) and circadian genes (Cmedtim and Cmedper) showed no significant differences. After silencing Cmedcry1, the take-off proportion significantly decreased. Thus, Cmedcry1 is involved in the decrease in light intensity induced take-off behaviour in C. medinalis. Conclusions This study can help further explain the molecular mechanisms behind insect migration, especially light perception and signal transmission during take-off phases.

Abstract Background This study aimed to investigate whether persistent human papillomavirus integration at the same loci (PHISL) before and after treatment can predict recurrent/residual disease in women with CIN2-3. Methods A total of 151 CIN2-3 women treated with conization between August 2020 and September 2021 were included. To investigate the precision of HPV integration, we further analyzed HPV integration-positive patients. Sensitivity, specificity, positive and negative predictive values (PPV and NPV, respectively), and the Youden index for predicting recurrence/residual disease were calculated. Results Among the 151 enrolled CIN2-3 women, 56 were HPV integration-positive and 95 had HPV integration-negative results. Six (10.7%) experienced recurrence among 56 HPV integration-positive patients, which was more than those in HPV integration-negative patients (one patient, 1.1%). In the 56 HPV integration-positive patients, 12 had positive HPV results after treatment, seven had PHISL, and two had positive cone margin. Among the seven patients who tested with PHISL, six (85.7%) had residual/recurrent disease. PHISL was a prominent predictor of persistent/recurrent disease. The HPV test, the HPV integration test, and PHISL all had a sensitivity of 100% and a NPV of 100% for residual/recurrent disease. PHISL showed better specificity (98.0% vs. 82.0%, p = 0.005) and PPV (85.7% vs. 40.0%, p = 0.001) than the HPV test for predicting recurrence. Conclusions The HPV-integration-positive CIN2-3 women had much higher relapse rates than HPV-integration-negative CIN2-3 women. The findings indicate that PHISL derived from preoperative and postoperative HPV integration tests may be a precise biomarker for the identification of residual/recurrent CIN 2/3.

Abstract Graphene growth on widely used dielectrics/insulators via chemical vapor deposition (CVD) is a strategy toward transfer-free applications of CVD graphene for the realization of advanced composite materials. Here, we develop graphene-skinned alumina fibers/fabrics (GAFs/GAFFs) through graphene CVD growth on commercial alumina fibers/fabrics (AFs/AFFs). We reveal a vapor-surface-solid growth model on a non-metallic substrate, which is distinct from the well-established vapor-solid model on conventional non-catalytic non-metallic substrates, but bears a closer resemblance to that observed on catalytic metallic substrates. The metalloid-catalytic growth of graphene on AFs/AFFs resulted in reduced growth temperature (~200 °C lower) and accelerated growth rate (~3.4 times faster) compared to that obtained on a representative non-metallic counterpart, quartz fiber. The fabricated GAFF features a wide-range tunable electrical conductivity (1-15000 Ω sq−1), high tensile strength (>1.5 GPa), lightweight, flexibility, and a hierarchical macrostructure. These attributes are inherited from both graphene and AFF, making GAFF promising for various applications including electrical heating and electromagnetic interference shielding. Beyond laboratory level preparation, the stable mass production of large-scale GAFF has been achieved through a home-made roll-to-roll system with capacity of 468-93600 m2/year depending on product specifications, providing foundations for the subsequent industrialization of this material, enabling its widespread adoption in various industries.

Abstract The ClC-3 chloride/proton exchanger is both physiologically and pathologically critical, as it is potentiated by ATP to detect metabolic energy level and point mutations in ClC-3 lead to severe neurodegenerative diseases in human. However, why this exchanger is differentially modulated by ATP, ADP or AMP and how mutations caused gain-of-function remains largely unknow. Here we determine the high-resolution structures of dimeric wildtype ClC-3 in the apo state and in complex with ATP, ADP and AMP, and the disease-causing I607T mutant in the apo and ATP-bounded state by cryo-electron microscopy. In combination with patch-clamp recordings and molecular dynamic simulations, we reveal how the adenine nucleotides binds to ClC-3 and changes in ion occupancy between apo and ATP-bounded state. We further observe I607T mutation induced conformational changes and augments in current. Therefore, our study not only lays the structural basis of adenine nucleotides regulation in ClC-3, but also clearly indicates the target region for drug discovery against ClC-3 mediated neurodegenerative diseases.

Abstract Breast cancer represents the most prevalent tumor type and a foremost cause of mortality among women globally. The complex pathophysiological processes of breast cancer tumorigenesis and progression are regulated by protein post-translational modifications (PTMs), which are triggered by different carcinogenic factors and signaling pathways, with small ubiquitin-like modifier (SUMOylation) emerging as a particularly pivotal player in this context. Recent studies have demonstrated that SUMOylation does not act alone, but interacts with other PTMs, such as phosphorylation, ubiquitination, acetylation, and methylation, thereby leading to the regulation of various pathological activities in breast cancer. This review explores novel and existing mechanisms of crosstalk between SUMOylation and other PTMs. Typically, SUMOylation is regulated by phosphorylation to exert feedback control, while also modulates subsequent ubiquitination, acetylation, or methylation. The crosstalk pairs in promoting or inhibiting breast cancer are protein-specific and site-specific. In mechanism, alterations in amino acid side chain charges, protein conformations, or the occupation of specific sites at specific domains or sites underlie the complex crosstalk. In summary, this review centers on elucidating the crosstalk between SUMOylation and other PTMs in breast cancer oncogenesis and progression and discuss the molecular mechanisms contributing to these interactions, offering insights into their potential applications in facilitating novel treatments for breast cancer.

Progranulin (PGRN), a multifunctional growth factor-like protein expressed by a variety of cell types, serves an important function in the physiologic and pathologic processes of fibrotic diseases, including wound healing and the inflammatory response. PGRN was discovered to inhibit pro-inflammation effect by competing with tumor necrosis factor-alpha (TNF-α) binding to TNF receptors. Notably, excessive tissue repair in the development of inflammation causes tissue fibrosis. Previous investigations have indicated the significance of PGRN in regulating inflammatory responses. Recently, multiple studies have shown that PGRN was linked to fibrogenesis, and was considered to monitor the formation of fibrosis in multiple organs, including liver, cardiovascular, lung and skin. This paper is a comprehensive review summarizing our current knowledge of PGRN, from its discovery to the role in fibrosis. This is followed by an in-depth look at the characteristics of PGRN, consisting of its structure, basic function and intracellular signaling. Finally, we will discuss the potential of PGRN in the diagnosis and treatment of fibrosis.

Abstract The reconstruction of critical-size calvarial defects remains a fundamental challenge. Recombinant collagen has gained significant attention in bone tissue engineering owing to its remarkable bioactivity and non-immunogenicity. Herein, we have for the first time developed a bioactive poly(ethylene glycol)-chondroitin sulfate-triple helical recombinant collagen (PEG-ChS-THRC) hydrogel for enhanced bone regeneration in cranial defects. A simple and mild crosslinking reaction of two-arm polyethylene glycol active ester (NHS-PEG-NHS), adipic dihydrazide modified chondroitin sulfate (ChS-ADH) and triple helical recombinant collagen (THRC) leads to the formation of the PEG-ChS-THRC hydrogel. The hydrogel demonstrates interconnected porous structures, enhanced mechanical strength, diminished swelling ratios and adjustable biodegradability. It possesses exceptional biocompatibility and bioactivity, significantly facilitating cell proliferation, adhesion, migration, and osteogenic differentiation of BMSCs. Micro-computed tomography (micro-CT), magnetic resonance imaging (MRI) and histological characterization of rat models with critical-size cranial defects have consistently demonstrated that the PEG-ChS-THRC hydrogel significantly promotes bone tissues regeneration. The innovative bioactive scaffold provides a remarkably improved remedy for critical-size cranial defects, holding greatly promising applications in the fields of bone tissue regeneration. Abstract Pseudorabies have caused enormous economic losses in China’s pig industry and have recurred on many large pig farms since late 2011. The disease is caused by highly pathogenic, antigenic variant pseudorabies virus (vPRV) strains. Our laboratory isolated a pseudorabies virus in 2015 and named it XJ5. The pathogenic ability of this mutant strain was much stronger than that of the original isolate. After we sequenced its whole genome (GenBank accession number: OP512542), we found that its overall structure was not greatly changed compared with that of the previous strain Ea (KX423960.1). The whole genome alignment showed that XJ5 had a strong genetic relationship with the strains isolated in China after 2012 reported in GenBank. Based on the isolation time of XJ5 and the mutation and recombination analysis of programs, we found that the whole genome homology of XJ5 and other strains with Chinese isolates was greater than 95%, while the homology with strains outside Asia was less than 94%, which indicated that there may be some recombination and mutation patterns. We found that virulent PRV isolates emerged successively in China in 2011 and formed two different evolutionary clades from foreign isolates. At the same time, this may be due to improper immunization and the presence of wild strains in the field, and recent reports have confirmed that Bartha vaccine strains recombine with wild strains to obtain new pathogenic strains. We performed genetic evolution analysis of XJ5 isolated and sequenced in our laboratory to trace its possible mutations and recombination. We found that XJ5 may be the result of natural mutation of a virus in a branch of mutant strains widely existing in China.

Liver fibrosis is an important step in the transformation of chronic liver disease into cirrhosis and liver cancer, and structural changes and functional disorders of liver sinusoidal endothelial cells (LSECs) are early events in the occurrence of liver fibrosis. Therefore, it is necessary to identify the key regulatory genes of endothelial dysfunction in the process of liver fibrosis to provide a reference for the diagnosis and treatment of liver fibrosis. In this study, we identified 230 common differentially expressed genes (Co-DEGs) by analyzing transcriptomic data of primary LSECs from three different liver fibrosis mouse models (carbon tetrachloride; choline-deficient, l-amino acid-defined diet; and nonalcoholic steatohepatitis). Enrichment analysis revealed that the Co-DEGs were mainly involved in regulating the inflammatory response, immune response, angiogenesis, formation and degradation of the extracellular matrix, and mediating chemokine-related pathways. A Venn diagram analysis was used to identify 17 key genes related to the progression of liver cirrhosis. Regression analysis using the Lasso–Cox method identified genes related to prognosis among these key genes: SOX4, LGALS3, SERPINE2, CD52, and LPXN. In mouse models of liver fibrosis (bile duct ligation and carbon tetrachloride), all five key genes were upregulated in fibrotic livers. This study identified key regulatory genes for endothelial dysfunction in liver fibrosis, namely SOX4, LGALS3, SERPINE2, CD52, and LPXN, which will provide new targets for the development of therapeutic strategies targeting endothelial dysfunction in LSECs and liver fibrosis.

Background Insulin resistance (IR) is the key pathological basis of many metabolic disorders. Lack of asialoglycoprotein receptor 1 (ASGR1) decreased the serum lipid levels and reduced the risk of coronary artery disease. However, whether ASGR1 also participates in the regulatory network of insulin sensitivity and glucose metabolism remains unknown. Methods The constructed ASGR1 knockout mice and ASGR1-/- HepG2 cell lines were used to establish the animal model of metabolic syndrome and the IR cell model by high-fat diet (HFD) or drug induction, respectively. Then we evaluated the glucose metabolism and insulin signaling in vivo and in vitro. Results ASGR1 deficiency ameliorated systemic IR in mice fed with HFD, evidenced by improved insulin intolerance, serum insulin, and homeostasis model assessment of IR index, mainly contributed from increased insulin signaling in the liver, but not in muscle or adipose tissues. Meanwhile, the insulin signal transduction was significantly enhanced in ASGR1-/- HepG2 cells. By transcriptome analyses and comparison, those differentially expressed genes between ASGR1 null and wild type were enriched in the insulin signal pathway, particularly in phosphoinositide 3-kinase-AKT signaling. Notably, ASGR1 deficiency significantly reduced hepatic gluconeogenesis and glycogenolysis. Conclusion The ASGR1 deficiency was consequentially linked with improved hepatic insulin sensitivity under metabolic stress, hepatic IR was the core factor of systemic IR, and overcoming hepatic IR significantly relieved the systemic IR. It suggests that ASGR1 is a potential intervention target for improving systemic IR in metabolic disorders.

Abstract Background Bipolar depression (BPD) is often misdiagnosed as a major depressive disorder (MDD) in clinical practice, which may be attributed to a lack of robust biomarkers indicative of differentiated diagnosis. This study analysed the differences in various hormones and inflammatory markers to explore peripheral biomarkers that differentiate BPD from MDD patients. Methods A total of 2,048 BPD and MDD patients were included. A panel of blood tests was performed to determine the levels of sex hormones, stress hormones, and immune-related indicators. Propensity score matching (PSM) was used to control for the effect of potential confounders between two groups and further a receiver operating characteristic (ROC) curve was used to analyse the potential biomarkers for differentiating BPD from MDD. Results Compared to patients with MDD, patients with BPD expressed a longer duration of illness, more hospitalisations within five years, and an earlier age of onset, along with fewer comorbid psychotic symptoms. In terms of biochemical parameters, MDD patients presented higher IgA and IgM levels, while BPD patients featured more elevated neutrophil and monocyte counts. ROC analysis suggested that combined biological indicators and clinical features could moderately distinguish between BPD and MDD. In addition, different biological features exist in BPD and MDD patients of different ages and sexes. Conclusions Differential peripheral biological parameters were observed between BPD and MDD, which may be age-sex specific, and a combined diagnostic model that integrates clinical characteristics and biochemical indicators has a moderate accuracy in distinguishing BPD from MDD.

Abstract This study aimed to evaluate the value of low-dose dual-input computed tomography perfusion (CTP) imaging in the differential diagnosis of benign and malignant pulmonary ground-glass opacity nodules (GGO). A retrospective study was conducted in patients with GGO who underwent CTP in our hospital from January 2021 to October 2023. All nodules were confirmed via pathological analysis or disappeared during follow-up. Postprocessing analysis was conducted using the dual-input perfusion mode (pulmonary artery and bronchial artery) of the body perfusion software to measure the perfusion parameters of the pulmonary GGOs. Abstract This study aimed to investigate body physical parameters as substitutes for water equivalent diameter (Dw) while calculating size-specific dose estimates (SSDEs) during adult chest computed tomography (CT). A retrospective analysis was conducted on 776 patients. Patients were divided into training set (542 patients) and validation set (234 patients) according to a ratio of 7:3. The correlations between physical parameters and Dw were analyzed. The differences between SSDEsubstitutes and the reference SSDE (SSDEreference) were compared. Strong positive correlations were observed between body mass index (BMI) and Dw as well as between weight and Dw in overall, male, and female patients (all p 0.05). The RMSEs of overall patients between SSDEweight and SSDEreference as well as between SSDEBMI and SSDEreference were 0.237 and 0.2, respectively. The use of sex-specific regression equations for BMI caused a slightly reduction in RMSE. Weight and BMI can be used as surrogate parameters for Dw when calculating SSDE in adult chest CT exams, with BMI being the preferred substitute parameter.

Abstract Background Blacks/African American (BAA) patients diagnosed with head and neck squamous cell carcinoma (HNSCC) have worse survival outcomes than White patients. However, the mechanisms underlying racial disparities in HNSCC have not been thoroughly characterized. Methods Data on gene expression, copy number variants (CNVs), gene mutations, and methylation were obtained from 6 head and neck cancer datasets. Comparative bioinformatics analysis of the above genomic features was performed between BAAs and Whites. The expression pattern of GSTM1 was validated by immunohistochemistry using tumor tissue microarray (TMA). Effect of GSTM1 knockdown were assessed by cell proliferation, colony formation, and tumor development in an orthotopic mouse model. The changes in protein kinases were determined using the Proteome Profiler Human Phospho-Kinase Array Kit in HNSCC cells with or without GSTM1 knockdown. Results We identified ancestry-related differential genomic profiles in HNSCC. Specifically, in BAA HNSCC, FAT1 mutations were associated with its gene expression, SALL3 gene expression correlated with its gene CNVs, and RTP4 gene expression showed an inverse correlation with its methylation. Notably, GSTM1 emerged as a prognostic risk factor for BAA HNSCC, with high gene CNVs and expression levels correlating with poor overall survival in BAA patients. Immunohistochemistry results from newly developed in-house TMA validated the expression pattern of GSTM1 between BAA HNSCC and White HNSCC. In an orthotopic mouse model, GSTM1 knockdown significantly inhibited malignant progression in tumors derived from BAAs. In contrast, loss of GSTM1 did not affect the development of HNSCC originating in Whites. Mechanistically, GSTM1 knockdown suppressed HSP27 phosphorylation and β-catenin in BAA HNSCC cells, but not in White HNSCC cells. This differential effect at least partially contributes to tumor development in BAA patients. Conclusion This study identifies GSTM1 as a novel molecular determinant of survival in HNSCC patients of African descent. It also provides a molecular basis for future research focused on identifying molecular determinants and developing therapeutic interventions to improve outcomes for BAA patients with HNSCC.

Abstract Previous research on the association between Family Doctor Contract Services (FDCS) and health has only considered a single indicator of health and has not considered the endogeneity of independent variables. This study aimed to evaluate the association from a multidimensional perspective of the health of middle-aged and older people using the instrumental variables method and determine the underlying mechanisms. Using data from the 2018 China Health and Retirement Longitudinal Study surveys, a total of 19,438 sample was obtained. Health was measured by health related-quality of life (HR-QoL), subjective well-being, and cognitive function. The instrumental variables method was used to estimate the association. Abstract Accelerating the design of Ni-based single crystal (SX) superalloys with superior creep resistance at ultrahigh temperatures is a desirable goal but extremely challenging task. In the present work, a deep transfer learning neural network with physical constraints for creep rupture life prediction at ultrahigh temperatures is constructed. Transfer learning enables deep learning model breaks through the generalization performance barrier in the extrapolation space of ultrahigh temperature creep properties in the case of a very small dataset, which is the key to achieving the above design goal. Transfer learning is demonstrated to be effective in utilizing the prior compositional sensitivities information contained in the pre-trained model, and motivates the fine-tuned model to capture the particular relationship between composition and creep rupture life at ultrahigh temperature. Aiming to find advanced SX superalloys applied at 1200 °C, the proposed transfer learning-based model guides us to design a superalloy with a verified creep rupture life of ~170 h at 80 MPa, which exceeds the state-of-art value by 30%. The improved γ/γ′ interface strengthening, which is effectively regulated by the Mo/Ta ratio to form γ′ rafting with longer, flatter interfaces and achieve stronger interfacial bonding, is revealed as the dominant mechanism behind combining experiments and first-principles calculations. Moreover, the excellent extrapolation ability of the proposed model is further confirmed to enhance the efficiency of active learning by reducing its dependence on the initial dataset size. This study provides a pioneering AI-driven approach for the rapid development of Ni-based SX superalloys applied in advanced aero-engine blades.

Abstract Background Treatment of non-small lung cancer (NSCLC) has evolved in recent years, benefiting from advances in immunotherapy and targeted therapy. However, limited biomarkers exist to assist clinicians and patients in selecting the most effective, personalized treatment strategies. Targeted next-generation sequencing–based genomic profiling has become routine in cancer treatment and generated crucial clinicogenomic data over the last decade. This has made the development of mutational biomarkers for drug response possible. Methods To investigate the association between a patient’s responses to a specific somatic mutation treatment, we analyzed the NSCLC GENIE BPC cohort, which includes 2,004 tumor samples from 1,846 patients. Results We identified somatic mutation signatures associated with response to immunotherapy and chemotherapy, including carboplatin-, cisplatin-, pemetrexed- or docetaxel-based chemotherapy. The prediction power of the chemotherapy-associated signature was significantly affected by epidermal growth factor receptor (EGFR) mutation status. Therefore, we developed an EGFR wild-type–specific mutation signature for chemotherapy selection. Conclusion Our treatment-specific gene signatures will assist clinicians and patients in selecting from multiple treatment options.

In order to sort out the research process and hotspots of honey safety, this paper was based on the China national knowledge infrastructure (CNKI) database and the Web of Science (WOS) core collection, utilizing CiteSpace software to analyze and visualize publication trends, journals, authors, institutions, countries (regions), keywords, and other relevant information. The aforementioned databases yielded a total of 169 Chinese and 281 English literatures. The analysis revealed that the number of publications in Chinese literature on honey safety had been steadily increasing since 2011. The number of publications in English literature had displayed an oscillating upward trend since 2007, with a higher volume of publications in English literature over the past fifteen years compared to Chinese literature. ZHANG Q and GUNDUZ A were the most prolific authors in Chinese and English literature, respectively. While there were established cooperative relationships between researchers in both fields, collaborations between different groups remained relatively limited. Journal of Bee and Food Chemistry were the leading journals for publications on honey safety in English and Chinese, respectively. Both English and Chinese publishing journals primarily focused on the food industry. Yunnan Province Center for Disease Control and Prevention and Karadeniz Technical University were the research institutions with the highest publication output in Chinese and English, respectively. However, there was limited collaboration between research institutes in China. Turkey was the most influential country among the study countries (regions), followed by China, Poland, and New Zealand. During the initial stages of research, Chinese literature primarily explored topics such as "botulinum toxin" "pollen plants" "Triperygium wilfordii Hook. f." and "honey poisoning". However, after 2000, advancements in technologies such as chromatography, spectroscopy, and scanning electron microscopy led to the emergence of new keywords like "melissopalynology" and "gas chromatography". Consequently, gas chromatography, liquid chromatography, liquid-mass spectrometry, and scanning electron microscopy gained popularity as methods for detecting honey safety in research. During the early stages of research, English literature primarily delved into topics such as "pyrrolizidine alkaloids" "mad honey" "grayanotoxin" and "natural toxicants". This indicates that during this time, the primary emphasis was on studying toxic honey and natural toxins found in honey, specifically pyrrolizidine alkaloids and grayanotoxin. As technologies such as chromatography advanced, keywords like "mass spectrometry" began to emerge. It could be seen that the research in Chinese and English literature was focused on similar topics, such as toxic honey, honey poisoning, and the detection and tracing of toxic components in honey. However, there were slight differences in emphasis. In China, the main toxic nectar plants were Triperygium wilfordii Hook. f., Gelsemium elegans (Gardn. & Champ.) Benth. and Macleaya cordata (Willd.) R. Br., while the main toxic components detected in honey were triptolide and koumine. In foreign countries, honey poisoning incidents were mostly caused by grayanotoxin, pyrrolizidine alkaloids or tutin. This study provides reference for analyzing the research content and development process of honey safety and identifying research hotspots.

To explore the structural dynamic characteristics of the impeller and shaft system of the axial flow pump as turbine, the variation law of impeller transient equivalent stress, deformation, and the impeller and shaft system mode are analyzed by the transient fluid-structure interaction method. Findings indicate that the equivalent stress is centralized at the blade root around the hub and tends to decrease as it moves towards the shroud. The maximum equivalent stress at 1.0Qbep and 1.1Qbep is 1.26 times and 2.0 times higher than that at 0.8Qbep. Blade deformation is significantly influenced by flow rate. The maximum deformation of the blade at 1.0Qbep and 1.1Qbep is 1.6 times and 3.6 times higher than that at 0.8Qbep. The impeller and shaft system with prestress in dry mode has an average natural frequency of the first 10 orders that is 16.7 Hz higher than without prestress. However, with the fluid adding mass and flow damping, the average natural frequency of the first 10 orders of the impeller and shaft system in wet mode is reduced by 278.08 Hz compared with dry mode. In fact, the impeller and shaft system of the axial flow pump as turbine is not prone to vibration.

The synchronized development of mineralized bone and blood vessels is a fundamental requirement for successful bone tissue regeneration. Adequate energy production forms the cornerstone supporting new bone formation. ETS variant 2 (ETV2) has been identified as a transcription factor that promotes energy metabolism reprogramming and facilitates the coordination between osteogenesis and angiogenesis. In vitro molecular experiments have demonstrated that ETV2 enhances osteogenic differentiation of dental pulp stem cells (DPSCs) by regulating the ETV2- prolyl hydroxylase 2 (PHD2)- hypoxia-inducible factor-1α (HIF-1α)- vascular endothelial growth factor A (VEGFA) axis. Notably, ETV2 achieves the rapid reprogramming of energy metabolism by simultaneously accelerating mitochondrial aerobic respiration and glycolysis, thus fulfilling the energy requirements essential to expedite osteogenic differentiation. Furthermore, decreased α-ketoglutarate release from ETV2-modified DPSCs contributes to microcirculation reconstruction. Additionally, we engineered hydroxyapatite/chitosan microspheres (HA/CS MS) with biomimetic nanostructures to facilitate multiple ETV2-DPSC functions and further enhanced the osteogenic differentiation. Animal experiments have validated the synergistic effect of ETV2-modified DPSCs and HA/CS MS in promoting the critical-size bone defect regeneration. In summary, this study offers a novel treatment approach for vascularized bone tissue regeneration that relies on energy metabolism activation and the maintenance of a stable local hypoxia signaling state.

ABSTRACTLivestock behavior is related to the healthy breeding and welfare level. Therefore, monitoring the behavior of sheep is helpful to predict the health status of sheep and thus safeguard the production performance of sheep. Taking semi-housed sheep as the research object, a behavior identification method for housed sheep based on spatio-temporal information is proposed. Firstly, video acquisition of housed sheep is carried out, and sheep detection and tracking is implemented based on the YOLOv5 coupled with a Deep-SORT algorithm, which detects sheep from the flock and labels their identity information; then, sheep posture estimation is done based on the Alphapose algorithm, which estimates the keypoints in the skeleton; finally, the detected keypoints are inputted into the trained spatio-temporal graph convolutional network model, and the spatio-temporal graph constructed from the keypoints and edge information is used for sheep behavior recognition. In the study, two keypoints selection proposals were made to implement sheep behavior recognition for non-contacted detection of ruminating, lying down and other behaviors. The results show the feasibility of sheep behavior recognition based on spatio-temporal information in semi-housed farming. The method provides a new solution idea for intelligent monitoring of sheep behavior and health assessment.

Federated learning, capable of training models by sharing gradient parameters, faces the risk of dishonest data aggregation by malicious servers during the model aggregation process. Untrusted users participating in federated learning may also pose a threat by poisoning the global model, thereby compromising the reliability and security of the model training process. To address these issues, homomorphic proxy re-authentication was introduced for the first time to propose a bi-directional authentication method suitable for multi-party aggregation computation. Additionally, a privacy-preserving, efficient, and trustworthy federated learning aggregation method was constructed, combining the double mask technique. This method not only enables users to verify the correctness of global model aggregation results but also allows the aggregation server to assess the trustworthiness and model integrity of the model sources uploaded by the users. It prevents attackers from maliciously manipulating users to disrupt secure aggregation, without leaking users’ private data during the verification process. The security of the verifiable federated aggregation method was demonstrated through formal security analysis, which effectively resists forgery attacks and Sybil attacks, exhibiting good robustness. Simulation experiments further illustrated that the proposed method can achieve credible verification of the aggregation results without impacting federated training. Moreover, the verification process remains unaffected even if users quit in the middle.

Abstract Disulfidptosis is a recently identified mode of regulated cell death. Regulating disulfidptosis in carcinoma is a promising therapeutic approach. Long non-coding RNAs (lncRNAs) have been reported to be related to the occurrence and development of many cancers. Disulfidptosis-related lncRNAs (DRLs) in HPV-negative oral squamous cell carcinoma (OSCC) have not been studied. Based on The Cancer Genome Atlas (TCGA) database, least absolute shrinkage selection operator (LASSO) analysis and Cox regression analysis were used to identify overall survival related DRLs and construct the signature. Kaplan–Meier, time-dependent receiver operating characteristics (ROC) and principal component analyses (PCA) were explored to demonstrate the prediction potential of the signature. Subgroup analysis stratified by different clinicopathological characteristics were conducted. Nomogram was established by DRLs signature and independent clinicopathological characteristics. The calibration plots were performed to reveal the accuracy of nomogram. Immune cell subset infiltration, immunotherapy response, drug sensitivity analysis, and tumor mutation burden (TMB) were conducted. Underlying functions and pathways were explored by Gene Set Enrichment Analysis (GSEA) analysis. Previous lncRNA signatures of OSCC were retrieved from PubMed for further validation. Gene expression omnibus (GEO) datasets (GSE41613 and GSE85446) were merged as an external validation for DRLs signature. Consensus clustering analysis of DRLs signature and experimental validation of DRLs were also explored. This research sheds light on the robust performance of DRLs signature in survival prediction, immune cell infiltration, immune escape, and immunotherapy of HPV-negative OSCC.

Abstract Synthetic hexaploid wheats (SHWs) are effective genetic resources for transferring agronomically important genes from wild relatives to common wheat (Triticum aestivum L.). Dozens of reference-quality pseudomolecule assemblies of hexaploid wheat have been generated, but none is reported for SHW-derived cultivars. Here, we generated a chromosome-scale assembly for the SHW-derived cultivar ‘Chuanmai 104’ based on PacBio HiFi reads and chromosome conformation capture sequencing. The total assembly size was 14.81 Gb with a contig N50 length of 58.25 Mb. A BUSCO analysis yielded a completeness score of 99.30%. In total, repetitive elements comprised 81.36% of the genome and 122,554 high-confidence protein-coding gene models were predicted. In summary, the first chromosome-level assembly for a SHW-derived cultivar presents a promising outlook for the study and utilization of SHWs in wheat improvement, which is essential to meet the global food demand.

Abstract Oral squamous cell carcinoma (OSCC) associated pain commonly predicts adverse events among patients. This clinical feature indicates the engagement of nociceptors on sensory neurons during the development of malignancy. However, it is yet to be determined if targeting oncometabolite-associated nociception processes can hinder OSCC progression. In this study, we reported that nociceptive endings infiltrating both clinical samples and mouse tumor xenografts were associated with poorer clinical outcomes and drove tumor progression in vivo, as evidenced by clinical tissue microarray analysis and murine lingual denervation. We observed that the OSCC microenvironment was characteristic of excessive adenosine due to CD73 upregulation which negatively predicted clinical outcomes in the TCGA-HNSC patient cohort. Notably, such adenosine concentrative OSCC niche was associated with the stimulation of adenosine A2A receptor (A2AR) on trigeminal ganglia. Antagonism of trigeminal A2AR with a selective A2AR inhibitor SCH58261 resulted in impeded OSCC growth in vivo. We showed that trigeminal A2AR overstimulation in OSCC xenograft did not entail any changes in the transcription level of CGRP in trigeminal ganglia but significantly triggered the release of CGRP, an effect counteracted by SCH58261. We further demonstrated the pro-tumor effect of CGRP by feeding mice with the clinically approved CGRP receptor antagonist rimegepant which inhibited the activation of ERK and YAP. Finally, we diminished the impact of CGRP on OSCC with istradefylline, a clinically available drug that targets neuronal A2AR. Therefore, we established trigeminal A2AR-mediated CGRP release as a promising druggable circuit in OSCC treatment.

Abstract Latent bioreactive unnatural amino acids (Uaas) have been widely used in the development of covalent drugs and identification of protein interactors, such as proteins, DNA, RNA and carbohydrates. However, it is challenging to perform high-throughput identification of Uaa cross-linking products due to the complexities of protein samples and the data analysis processes. Enrichable Uaas can effectively reduce the complexities of protein samples and simplify data analysis, but few cross-linked peptides were identified from mammalian cell samples with these Uaas. Here we develop an enrichable and multiple amino acids reactive Uaa, eFSY, and demonstrate that eFSY is MS cleavable when eFSY-Lys and eFSY-His are the cross-linking products. An identification software, AixUaa is developed to decipher eFSY mass cleavable data. We systematically identify direct interactomes of Thioredoxin 1 (Trx1) and Selenoprotein M (SELM) with eFSY and AixUaa.

Abstract. Background:. Hepatitis B virus (HBV) infection is the primary cause of hepatocellular carcinoma (HCC) in China. The target population for HCC screening comprises individuals who test positive for hepatitis B surface antigen (HBsAg). However, current data on the prevalence of HBV infection among individuals who are eligible for HCC screening in China are lacking. We aimed to assess the seroepidemiology of HBV infection among Chinese individuals eligible for HCC screening to provide the latest evidence for appropriate HCC screening strategies in China. Methods:. Questionnaires including information of sex, age, ethnicity, marital status, educational level, source of drinking water, as well as smoking and alcohol consumption history and serum samples were collected from females aged 45–64 years and males aged 35–64 years in 21 counties from 4 provinces in eastern and central China between 2015 and 2023. Enzyme-linked immunosorbent assay methods were used to detect the serum HBV marker HBsAg. Results:. A total of 603,082 individuals were enrolled, and serum samples were collected for analysis from January 1, 2015 to December 31, 2023. The prevalence of HBsAg positive in the study population was 5.23% (31,528/603,082). Based on the latest results of near-source exploration in the Middle and Lower Jurassic of the Tuha Basin, a new understanding of the source rocks, reservoir conditions, and source-reservoir-cap rock combinations of the Jurassic Shuixigou Group in the Taibei Sag is established using the concept of the whole petroleum system, and the coal-measure whole petroleum system is analyzed thoroughly. The results are obtained in three aspects. First, the coal-measure source rocks of the Badaowan Formation and Xishanyao Formation and the argillaceous source rocks of the Sangonghe Formation in the Shuixigou Group exhibit the characteristics of long-term hydrocarbon generation, multiple hydrocarbon generation peaks, and simultaneous oil and gas generation, providing sufficient oil and gas sources for the whole petroleum system in the Jurassic coal-bearing basin. Second, multi-phase shallow braided river delta–shallow lacustrine deposits contribute multiple types of reservoirs, e.g. sandstone, tight sandstone, shale and coal rock, in slope and depression areas, providing effective storage space for the petroleum reservoir formation in coal-measure strata. Third, three phases of hydrocarbon charging and structural evolution, as well as effective configuration of multiple types of reservoirs, result in the sequential accumulation of conventional-unconventional hydrocarbons. From high structural positions to depression, there are conventional structural and structural-lithological reservoirs far from the source, low-saturation structural-lithological reservoirs near the source, and tight sandstone gas, coal rock gas and shale oil accumulations within the source. Typically, the tight sandstone gas and coal rock gas are the key options for further exploration, and the shale oil and gas in the depression area is worth of more attention. The new understanding of the whole petroleum system in the coal measures could further enrich and improve the geological theory of the whole petroleum system, and provide new ideas for the overall exploration of oil and gas resources in the Tuha Basin.

Abstract Direct chemical vapor deposition (CVD) growth of graphene on dielectric/insulating materials is a promising strategy for subsequent transfer-free applications of graphene. However, graphene growth on noncatalytic substrates is faced with thorny issues, especially the limited growth rate, which severely hinders mass production and practical applications. Herein, graphene glass fiber fabric (GGFF) is developed by graphene CVD growth on glass fiber fabric. Dichloromethane is applied as a carbon precursor to accelerate graphene growth, which has a low decomposition energy barrier, and more importantly, the produced high-electronegativity Cl radical can enhance adsorption of active carbon species by Cl–CH2 coadsorption and facilitate H detachment from graphene edges. Consequently, the growth rate is increased by ~3 orders of magnitude and carbon utilization by ~960-fold, compared with conventional methane precursor. The advantageous hierarchical conductive configuration of lightweight, flexible GGFF makes it an ultrasensitive pressure sensor for human motion and physiological monitoring, such as pulse and vocal signals.
