Sperm heterogeneity creates challenges for successful artificial insemination. Seminal plasma (SP) surrounding sperm is an excellent source for detecting reliable non-invasive biomarkers of sperm quality. Here, we isolated microRNAs (miRNAs) from SP-derived extracellular vesicles (SP-EV) of boars with divergent sperm quality statuses. Raw semen from sexually mature boars was collected for eight weeks. Sperm motility and normal morphology were analyzed, and the sperm was classified as poor- or good-quality based on standard cutoffs of 70% for the parameters measured. SP-EVs were isolated by ultracentrifugation and confirmed by electron microscopy, dynamic light scattering, and Western immunoblotting. The SP-EVs were subjected to total exosome RNA isolation, miRNA sequencing, and bioinformatics analysis. The isolated SP-EVs were round spherical structures approximately 30–400 nm in diameter expressing specific molecular markers. miRNAs were detected in both poor- (n = 281) and good (n = 271)-quality sperm, with fifteen being differentially expressed. Only three (ssc-miR-205, ssc-miR-493-5p, and ssc-miR-378b-3p) allowed gene targeting associated with cellular localization (nuclear and cytosol) and molecular functions (acetylation, Ubl conjugation, and protein kinase binding), potentially impairing sperm quality. PTEN and YWHAZ emerged as essential proteins for protein kinase binding. We conclude that SP-EV-derived miRNAs reflect boar sperm quality to enable therapeutic strategies to improve fertility.

Interaction exists in lung cancer and microbiota. Lung microecological homeostasis can improve the immune tolerance, enhance immune suppression, and inhibit inflammatory responses, to reduce the lung cancer; while lung cancer can lead to pulmonary microecological imbalance, change the lung environment, and promote tumor cell proliferation. Therefore, modulating microbial flora and microecological immunotherapy may be a potential and preventive treatment for lung cancer, to restore tumor immunosuppression and improve patient survival. However, the individual differences in the lung microecology, because of different genetics, ethnic characteristics, and dietary habits, increasing the difficulty of precise diagnosis and treatment, which is also the current bottleneck in the application of microecological immunotherapy. Otherwise, the effectiveness of regulatory measures such as probiotics, prebiotics or antimicrobials is questionable. The research on microbial flora is still in its infancy, and further exploration is needed to form a standardized, effective, and precise treatment plan. Dyslipidemia is an important risk factor of atherosclerotic cardiovascular disease (ASCVD). Statins delay the occurrence and development of ASCVD, and reduce the risk of cardiovascular events and death. Due to safety concerns, there exist insufficient use of lipid-lowering agents and a high withdrawal rate of the agents in the elderly. Background While massive transfusion protocols (MTP) are associated with decreased mortality in adult trauma patients, there is limited research on the impact of MTP on pediatric trauma patients. The purpose of this study was to compare pediatric trauma patients requiring massive transfusion to all other pediatric trauma patients to identify triggers for MTP activation in injured children. Methods Using our level I trauma center's registry, we retrospectively identified all pediatric trauma patients from January 2015 to January 2018. Massive transfusion (MT) was defined as infusion of 40 mL/kg of blood products in the first 24 hours of admission. Patients missing prehospital vital sign data were excluded from the study. We retrospectively collected data including: demographics, blood utilization, variable outcome data, prehospital vital signs, prehospital transport times, and injury severity scores (ISS). Statistical significance was determined using Mann-Whitney U test and chi-square test. P values less than 0.05 were considered significant. Results Thirty-nine of the 2,035 pediatric patients (1.9%) met criteria for MT. All-cause mortality in MT patients was 49% (19/39) versus 0.01% (20/1996) in Non-MT patients. The two groups significantly differed in ISS, prehospital vital signs, and outcome data.Both systolic blood pressure (SBP) 1.4 were found to be highly specific for massive transfusion with specificities of 86% and 92%, respectively. The combination of SBP 1.4 had a specificity of 94%. The positive and negative predictive values of SBP 1.4 in predicting massive transfusion were 18% and 98%, respectively. Based on positive likelihood ratios, patients with both SBP 1.4 were 7.2 times more likely to require massive transfusion than patients who did not meet both of these vital sign criteria. Conclusions Pediatric trauma patients requiring early blood transfusion present with lower blood pressures and higher heart rates, as well as higher shock indexes and lower pulse pressures. We found that shock index and systolic blood pressure are highly specific tools with promising likelihood ratios that could be used to identify patients requiring early transfusion. Using alternative feedstuffs can be a feasible measure to reduce swine production cost. GuarPro F-71 (GP71), a newly developed guar meal product, is considered as a potential protein source for pigs. This preliminary study was conducted to evaluate the effects of GP71 inclusion in feed on growth performance and nutrient metabolism of growing pigs. Sixty crossbred barrows (31.9±1.84 kg body weight (BW)) were allotted to one of five dietary treatments in a completely randomised experiment design, with four pens/treatment and three pigs/pen. Diet 1 was a maize- and soybean-based basal control diet formulated to meet NRC recommendations. GP71 was used to replace 25, 50, 75, and 100% soybean meal in diet 1 (on an equivalent crude protein (CP) basis) to generate diets 2, 3, 4, and 5, respectively. Crystalline amino acids were used to balance the essential amino acid concentrations across the five diets. Inclusion of GP71 quadratically reduced average daily feed intake (ADFI) (P=0.01) with no observed clinical signs of unhealthiness of pigs. The blood plasma concentrations of six metabolites, including urea nitrogen, total protein, albumin, glucose, total cholesterol and total triglycerides, were not affected (P>0.30), indicating that the pigs’ metabolic utilisation of nutrients was not compromised. This study aims to investigate public protest slogans in response to the Biden Administration's announcement of two COVID-19 vaccination regulations on November 4, 2021. We identified three protest themes: (1) support for individual freedom/rights, (2) opposition to government's control, and (3) anti-science reasoning/misinformation/disinformation. Major policy recommendations include implementing efforts to dispel unscientific misinformation/disinformation and to emphasize individuals' civic responsibilities for vaccination during the COVID-19 pandemic, especially considering the current spreading of the Omicron variants and the relaxing of mask mandates across the US.

Multidomain/multichannel sequence analysis has become widely used in social science research to uncover the underlying relationships between two or more observed trajectories in parallel. For example, life-course researchers use multidomain sequence analysis to study the parallel unfolding of multiple life-course domains. In this article, the authors conduct a critical review of the approaches most used in multidomain sequence analysis. The parallel unfolding of trajectories in multiple domains is typically analyzed by building a joint multidomain typology and by examining how domain-specific sequence patterns combine with one another within the multidomain groups. The authors identify four strategies to construct the joint multidomain typology: proceeding independently of domain costs and distances between domain sequences, deriving multidomain costs from domain costs, deriving distances between multidomain sequences from within-domain distances, and combining typologies constructed for each domain. The second and third strategies are prevalent in the literature and typically proceed additively. The authors show that these additive procedures assume between-domain independence, and they make explicit the constraints these procedures impose on between-multidomain costs and distances. Regarding the fourth strategy, the authors propose a merging algorithm to avoid scarce combined types. As regards the first strategy, the authors demonstrate, with a real example based on data from the Swiss Household Panel, that using edit distances with data-driven costs at the multidomain level (i.e., independent of domain costs) remains easily manageable with more than 200 different multidomain combined states. Recent developments in the instrumentation and data analysis of synchrotron small-angle X-ray scattering (SAXS) on biomolecules in solution have made biological SAXS (BioSAXS) a mature and popular tool in structural biology. This article reports on an advanced endstation developed at beamline 13A of the 3.0 GeV Taiwan Photon Source for biological small- and wide-angle X-ray scattering (SAXS–WAXS or SWAXS). The endstation features an in-vacuum SWAXS detection system comprising two mobile area detectors (Eiger X 9M/1M) and an online size-exclusion chromatography system incorporating several optical probes including a UV–Vis absorption spectrometer and refractometer. The instrumentation and automation allow simultaneous SAXS–WAXS data collection and data reduction for high-throughput biomolecular conformation and composition determinations. The performance of the endstation is illustrated with the SWAXS data collected for several model proteins in solution, covering a scattering vector magnitude q across three orders of magnitude. The crystal-model fittings to the data in the q range ∼0.005–2.0 Å−1 indicate high similarity of the solution structures of the proteins to their crystalline forms, except for some subtle hydration-dependent local details. These results open up new horizons of SWAXS in studying correlated local and global structures of biomolecules in solution.

Semiconducting substances form one of the most important families of functional materials. However, semiconductors containing only metals are very rare. The chemical mechanisms behind their ground‐state properties are only partially understood. Our investigations have rather unexpectedly revealed the semiconducting behaviour (band gap of 190 meV) for the intermetallic compound Be5Pt formed at a very low valence‐electron count. Quantum‐chemical analysis shows strong charge transfer from Be to Pt and reveals a three‐dimensional entity of vertex‐condensed empty Be4 tetrahedrons with multi‐atomic cluster bonds interpenetrated by the framework of Pt‐filled vertex‐condensed Be4 tetrahedrons with two‐atomic polar Be−Pt bonds. The combination of strong Coulomb interactions with relativistic effects results in a band gap., Progressive metals: Be5Pt is a rare representative of semiconductors that are formed by metallic elements only. The gap in the band structure appears due to both relativistic effects and a unique chemical‐bonding situation with polar two‐atom and multi‐atom Pt–Be interactions.

Maternal separation (MS) has been developed as a model for inducing stress and depression in studies using rodents. The concept of the gut-brain axis suggests that gut health is essential for brain health. Here, we present the effects of administration of a probiotic, Lactobacillus paracasei PS23 (PS23), to MS mice against psychological traits including anxiety and depression. The relationship between migrant workers’ migratory moves and their fertility is complex. The literature on international migration contains three main explanations of the relationship—the disruption hypothesis, the adaptation hypothesis, and the selectivity hypothesis. The research reported in the literature has so far focused on international migrants, many of whom have the prospect of becoming legal immigrants, while there is little research on the relation between migration and fertility for Southeast Asian labor migrants who are on employment contracts and cannot gain permanent residency. Of the three hypotheses, only the disruption hypothesis is relevant for temporary migrant workers, yet its findings have been mixed. This research note aims to fill in the gap by focusing on the disruption hypothesis in an examination of Filipina and Indonesian migrant domestic workers’ migration and fertility trajectories, using the 2017 survey of Filipina and Indonesian migrant domestic workers in Hong Kong. The findings from the analysis suggest a disruptive effect of migration on fertility for the women in their earlier reproductive years from both Indonesia and the Philippines but not for the women in their later childbearing ages, thus shedding light on the divergent disruptive mechanism of migration on fertility.
