Purpose: This study aims to explore the feasibility and performance of three-dimensional ultrasound (3DUS) imaging in ophthalmology using commercially available ultrasound probes adapted to a slit lamp., Significance: Despite ultrasound's long-standing application in eye care for visualizing ocular components, the evolution of 3DUS technology has remained inactive, with limited development and commercial availability. This study introduces a novel method that could potentially enhance ophthalmic diagnostics and treatment planning by providing comprehensive 3D views of ocular structures using existing ultrasound probes adapted to the conventional slit lamp., Methods: A custom system was designed for mounting a linear actuator to a slit lamp and enabling the horizontal actuation of any ultrasound probe. Ophthalmic and non-ophthalmic ultrasound probes were tested for their ability to reconstruct 3DUS images of the posterior pole. The study involved designing and evaluating three US phantoms ex vivo and performing in vivo imaging on human subjects to assess the system's applicability., Results: The system successfully acquired 3D volume scans of phantoms and live human eyes, demonstrating the system's potential for detailed ocular imaging. The adaptability of the device allowed for compatibility with various ultrasound probes. In vivo imaging revealed the system's capability to produce high-resolution 3D reconstructions of ocular structures, including eyes with pathological conditions., Conclusions: The introduction of a slit lamp-mounted 3DUS system represents a significant advancement in ophthalmic ultrasonography, offering a practical and accessible solution for comprehensive 3D ocular assessments. The device's modularity and compatibility with existing ultrasound probes make it a versatile tool for a wide range of ophthalmic applications. The human cortex has a rich fiber structure as revealed by myelin-staining of histological slices. Myelin also contributes to the image contrast in Magnetic Resonance Imaging (MRI). Recent advances in Magnetic Resonance (MR) scanner and imaging technology allowed the acquisition of an ex-vivo data set at an isotropic resolution of 100 µm. This study focused on a computational analysis of this data set with the aim of bridging between histological knowledge and MRI-based results. This work highlights: (1) the design and implementation of a processing chain that extracts intracortical features from a high-resolution MR image; (2) a demonstration of the correspondence between MRI-based cortical intensity profiles and the myelo-architectonic layering of the cortex; (3) the characterization and classification of four basic myelo-architectonic profile types; (4) the distinction of cortical regions based on myelo-architectonic features; and (5) the segmentation of cortical modules in the entorhinal cortex., (© 2023 The Authors. The rapid evolution of image acquisition and data analytic methods has established in vivo whole-brain tractography as a routine technology over the last 20 years. Imaging-based methods provide an additional approach to classic neuroanatomical studies focusing on biomechanical principles of anatomical organization and can in turn overcome the complexity of inter-individual variability associated with histological and tractography studies. In this work we propose a novel, reliable framework for determining brain tracts resolving the anatomical variance of brain regions. We distinguished 4 region types based on anatomical considerations: (i) gyral regions at borders between cortical communities; (ii) gyral regions within communities; (iii) sulcal regions at invariant locations across subjects; and (iv) other sulcal regions. Region types showed strikingly different anatomical and connection properties. Genetic influences that govern the spatial patterning of the human cortex and its structural variability are still incompletely known. We analyzed structural MR images in twins, siblings, and pairs of unrelated subjects. A comprehensive set of methods was employed to quantify properties of cortical features at different spatial scales. Measures were used to assess the influence of genetic similarity on structural patterning. Results indicated that: (1) Genetic effects significantly influence all structural features assessed here at all spatial resolutions, albeit at different strengths. (2) While strong genetic effects were found at the whole-brain and hemisphere level, effects were weaker at the regional and vertex level, depending on the measure under study. (3) Besides cortical thickness, sulcal (geodesic) depth was found to be under strong genetic control. The local pattern indicated that two axes along (a) the anterior-posterior direction (insula to parieto-occipital sulcus), and (b) superior-inferior direction (central sulcus to callosal sulcus) presumably determine the segregation of four quadrants in each hemisphere early in development. This study aimed at uncovering mechanisms that govern the spatio-temporal patterning of the human cortex and its structural variability, and drawing links between fetal brain development and variability in adult brains. A data-driven analytic approach based on structural MR images revealed the following findings: (1) The cortical surface can be subdivided into 13 independent regions ("communities") based on macroscopic features. (2) Thirty centers of low inter-subject variability were found in major sulci on the cortical surface. Their variability showed a strong positive correlation with the known time points at which they appear in fetal development. Centers forming early induce a higher inter-subject regularity in a larger local vicinity, while those forming later result in smaller regions of higher variability. (3) The layout of sulcal and gyral patterns within a community is governed typically by two centers. Depending on the relative variability of each center, communities can be classified into structural sub-types. (4) Sub-types across ipsi-lateral communities are independent, but associated with the sub-type of the same community on the contra-lateral side. An automatic and objective assessment of image quality is important in an era, where large-scale processing of imaging data from multi-center studies becomes commonplace. Based on a comprehensive statistical image model that includes noise and blur, a measure for image acuity is derived here as the ratio of the maximal gradient magnitude and the intensity difference at a boundary. Acuity may be affected by the object under study, the image acquisition, reconstruction processes, and any post-processing steps. The acuity measure presented here is post-hoc, intuitive to understand, simple to compute, and easily integrates with other standard measures of image quality. Three applications in medical imaging are included where our acuity measure is useful in the objective and automatic assessment of image quality.

The human neocortex shows a considerable individual structural variability. While primary gyri and sulci are found in all normally developed brains and bear clear-cut gross structural descriptions, secondary structures are highly variable and not present in all brains. The blend of common and individual structures poses challenges when comparing structural and functional results from quantitative neuroimaging studies across individuals, and sets limits on the precision of location information much above the spatial resolution of current neuroimaging methods. This work aimed at quantifying structural variability on the neocortex, and at assessing the spatial relationship between regions common to all brains and their individual structural variants. Based on structural MRI data provided as the "900 Subjects Release" of the Human Connectome Project, a data-driven analytic approach was employed here from which the definition of seven cortical "communities" emerged. Apparently, these communities comprise common regions of structural features, while the individual variability is confined within a community. Similarities between the community structure and the state of the brain development at gestation week 32 lead suggest that communities are segregated early. Background: The importance of energy homeostasis brain circuitry in the context of obesity is well established, however, the developmental ontogeny of this circuitry in humans is currently unknown. Here, we investigate the prospective association between newborn gray matter (GM) volume in the insula, a key brain region underlying energy homeostasis, and change in percent body fat accrual over the first six months of postnatal life, an outcome that represents among the most reliable infant predictors of childhood obesity risk., Methods: A total of 52 infants (29 male, 23 female, gestational age at birth=39(1.5) weeks) were assessed using structural MRI shortly after birth (postnatal age at MRI scan=25.9(12.2) days), and serial Dual X-Ray Absorptiometry shortly after birth (postnatal age at DXA scan 1=24.6(11.4) days) and at six months of age (postnatal age at DXA scan 2=26.7(3.3) weeks)., Results: Insula GM volume was inversely associated with change in percent body fat from birth to six-months postnatal age and accounted for 19% of its variance (β=-3.6%/S.D., P=0.001). This association was driven by the central-posterior portion of the insula, a region of particular importance for gustation and interoception. The direction of this effect is in concordance with observations in adults, and the results remained statistically significant after adjusting for relevant covariates and potential confounding variables., Conclusions: Altogether, these findings suggest an underlying neural basis of childhood obesity that precedes the influence of the postnatal environment. The identification of plausible brain-related biomarkers of childhood obesity risk that predate the influence of the postnatal obesogenic environment may contribute to an improved understanding of propensity for obesity, early identification of at-risk individuals, and intervention targets for primary prevention.

Background: This is one of the first studies to examine the structural brain anatomy and connectivity associated with an ADHD diagnosis and child as well as adult ADHD symptoms in young adults. It was hypothesized that an adult ADHD diagnosis and in particular childhood symptoms, are associated with widespread changes in the brain macro- and microstructure, which can be used to develop a morphometric biomarker for ADHD., Methods: Voxel-wise linear regression models were used to examine structural and diffusion-weighted MRI data in 72 participants (31 young adults with ADHD and 41 controls without ADHD) in relation to diagnosis and the number of self-reported child and adult symptoms., Results: Findings revealed significant associations between ADHD diagnosis and widespread changes to the maturation of white matter fiber bundles and gray matter density in the brain, such as structural shape changes (incomplete maturation) of the middle and superior temporal gyrus, and fronto-basal portions of both frontal lobes. ADHD symptoms in childhood showed the strongest association with brain macro- and microstructural abnormalities. At the brain circuitry level, the superior longitudinal fasciculus (SLF) and cortico-limbic areas are dysfunctional in individuals with ADHD. The morphometric findings predicted an ADHD diagnosis correctly up to 83% of all cases., Conclusion: An adult ADHD diagnosis and in particular childhood symptoms are associated with widespread micro- and macrostructural changes. The SLF and cortico-limbic findings suggest complex audio-visual, motivational, and emotional dysfunctions associated with ADHD in young adults. The sensitivity of the morphometric findings in predicting an ADHD diagnosis was sufficient, which indicates that MRI-based assessments are a promising strategy for the development of a biomarker.

Background & New Method: The widely used framework of voxel-based morphometry for analyzing neuroimages is extended here to model longitudinal imaging data by exchanging the linear model with a linear mixed-effects model. The new approach is employed for analyzing a large longitudinal sample of 756 diffusion-weighted images acquired in 177 subjects of the Alzheimer's Disease Neuroimaging initiative (ADNI)., Results and Comparison With Existing Methods: While sample- and group-level results from both approaches are equivalent, the mixed-effect model yields information at the single subject level. Interestingly, the neurobiological relevance of the relevant parameter at the individual level describes specific differences associated with aging. In addition, our approach highlights white matter areas that reliably discriminate between patients with Alzheimer's disease and healthy controls with a predictive power of 0.99 and include the hippocampal alveus, the para-hippocampal white matter, the white matter of the posterior cingulate, and optic tracts. In this context, notably the classifier includes a sub-population of patients with minimal cognitive impairment into the pathological domain., Conclusion: Our classifier offers promising features for an accessible biomarker that predicts the risk of conversion to Alzheimer's disease. Human birth presents an abrupt transition from intrauterine to extrauterine life. Here we introduce a novel Maturation Index (MI) that considers the relative importance of gestational age at birth and postnatal age at scan in a General Linear Model. The MI is then applied to Diffusion Tensor Imaging (DTI) in newborns for characterizing typical white matter development in neonates. DTI was performed cross-sectionally in 47 neonates (gestational age at birth=39.1±1.6 weeks [GA], postnatal age at scan=25.5±12.2days [SA]). Radial diffusivity (RD), axial diffusivity (AD) and fractional anisotropy (FA) along 27 white matter fiber tracts were considered. The MI was used to characterize inflection in maturation at the time of birth using GLM estimated rates of change before and after birth. It is proposed that the sign (positive versus negative) of MI reflects the period of greatest maturation rate. Two general patterns emerged from the MI analysis. First, RD and AD (but not FA) had positive MI on average across the whole brain (average MI AD =0.31±0.42, average MI RD =0.22±0.34). Second, significant regions of negative MI in RD and FA (but not AD) were observed in the inferior corticospinal regions, areas known to myelinate early. Observations using the proposed method are consistent with proposed models of the white matter maturation process in which pre-myelination is described by changes in AD and RD due to oligodendrocyte proliferation while true myelination is characterized by changes in RD and FA due to myelin formation., (Copyright © 2016 ISDN. Purpose: This article presents a simple method for estimating the effective diffusion coefficients parallel and perpendicular to the axons unconfounded by the intravoxel fiber orientation distribution. We also call these parameters the per-axon or microscopic diffusion coefficients., Theory and Methods: Diffusion MR imaging is used to probe the underlying tissue material. The key observation is that for a fixed b-value the spherical mean of the diffusion signal over the gradient directions does not depend on the axon orientation distribution. By exploiting this invariance property, we propose a simple, fast, and robust estimator of the per-axon diffusion coefficients, which we refer to as the spherical mean technique., Results: We demonstrate quantitative maps of the axon-scale diffusion process, which has factored out the effects due to fiber dispersion and crossing, in human brain white matter. These microscopic diffusion coefficients are estimated in vivo using a widely available off-the-shelf pulse sequence featuring multiple b-shells and high-angular gradient resolution., Conclusion: The estimation of the per-axon diffusion coefficients is essential for the accurate recovery of the fiber orientation distribution. In addition, the spherical mean technique enables us to discriminate microscopic tissue features from fiber dispersion, which potentially improves the sensitivity and/or specificity to various neurological conditions. Noninvasive diagnostic tests for Alzheimer's disease (AD) are limited. Postmortem diagnosis is based on density and distribution of neurofibrillary tangles (NFTs) and amyloid-rich neuritic plaques. In preclinical stages of AD, the cells of origin for the perforant pathway within the entorhinal cortex are among the first to display NFTs, indicating its compromise in early stages of AD. We used diffusion tensor imaging (DTI) to assess the integrity of the parahippocampal white matter in mild cognitive impairment (MCI) and AD, as a first step in developing a noninvasive tool for early diagnosis. Subjects with AD (N = 9), MCI (N = 8), or no cognitive impairment (NCI; N = 20) underwent DTI-MRI. Fractional anisotropy (FA) and mean (MD) and radial (RD) diffusivity measured from the parahippocampal white matter in AD and NCI subjects differed greatly. Discriminant analysis in the MCI cases assigned statistical membership of 38% of MCI subjects to the AD group. Preliminary data 1 year later showed that all MCI cases assigned to the AD group either met the diagnostic criteria for probable AD or showed significant cognitive decline. Voxelwise analysis in the parahippocampal white matter revealed a progressive change in the DTI patterns in MCI and AD subjects: whereas converted MCI cases showed structural changes restricted to the anterior portions of this region, in AD the pathology was generalized along the entire anterior-posterior axis. The use of DTI for in vivo assessment of the parahippocampal white matter may be useful for identifying individuals with MCI at highest risk for conversion to AD and for assessing disease progression., (Copyright © 2013 Wiley Periodicals, Inc.)

A single case study recently documented one woman's ability to recall accurately vast amounts of autobiographical information, spanning most of her lifetime, without the use of practiced mnemonics (Parker, Cahill, & McGaugh, 2006). The current study reports findings based on eleven participants expressing this same memory ability, now referred to as Highly Superior Autobiographical Memory (HSAM). Participants were identified and subsequently characterized based on screening for memory of public events. They were then tested for personal autobiographical memories as well as for memory assessed by laboratory memory tests. Additionally, whole-brain structural MRI scans were obtained. Results indicated that HSAM participants performed significantly better at recalling public as well as personal autobiographical events as well as the days and dates on which these events occurred. However, their performance was comparable to age- and sex-matched controls on most standard laboratory memory tests. Neuroanatomical results identified nine structures as being morphologically different from those of control participants. Diffusion MR imaging provides a unique tool to probe the microgeometry of nervous tissue and to explore the wiring diagram of the neural connections noninvasively. Generally, a forward model is established to map the intra-voxel fiber architecture onto the observable diffusion signals, which is reformulated in this article by adopting a measure-theoretic approach. However, the inverse problem, i.e., the spherical deconvolution of the fiber orientation density from noisy MR measurements, is ill-posed. We propose a nonparametric representation of the tangential distribution of the nerve fibers in terms of a Dirichlet process mixture. Given a second-order approximation of the impulse response of a fiber segment, the specified problem is solved by Bayesian statistics under a Rician noise model, using an adaptive reversible jump Markov chain Monte Carlo sampler. The density estimation framework is demonstrated by various experiments with a diffusion MR dataset featuring high angular resolution, uncovering the fiber orientation field in the cerebral white matter of the living human brain., (Copyright © 2012 Elsevier B.V. Careful selection of the reference region for non-quantitative positron emission tomography (PET) analyses is critically important for Region of Interest (ROI) data analyses. We introduce an empirical method of deriving the most suitable reference region for computing neurodegeneration sensitive (18)fluorodeoxyglucose (FDG) PET ratios based on the dataset collected by the Alzheimer's Disease Neuroimaging Initiative (ADNI) study. Candidate reference regions are selected based on a heat map of the difference in coefficients of variation (COVs) of FDG ratios over time for each of the Automatic Anatomical Labeling (AAL) atlas regions normalized by all other AAL regions. Visual inspection of the heat map suggests that the portion of the cerebellum and vermis superior to the horizontal fissure is the most sensitive reference region. Analyses of FDG ratio data show increases in significance on the order of ten-fold when using the superior portion of the cerebellum as compared with the traditionally used full cerebellum. The approach to reference region selection in this paper can be generalized to other radiopharmaceuticals and radioligands as well as to other disorders where brain changes over time are hypothesized and longitudinal data is available. Based on the empirical evidence presented in this study, we demonstrate the usefulness of the COV heat map method and conclude that intensity normalization based on the superior portion of the cerebellum may be most sensitive to measuring change when performing longitudinal analyses of FDG-PET ratios as well as group comparisons in Alzheimer's disease. Reconstruction of the cerebral cortex from magnetic resonance (MR) images is an important step in quantitative analysis of the human brain structure, for example, in sulcal morphometry and in studies of cortical thickness. Existing cortical reconstruction approaches are typically optimized for standard resolution (~1 mm) data and are not directly applicable to higher resolution images. A new PDE-based method is presented for the automated cortical reconstruction that is computationally efficient and scales well with grid resolution, and thus is particularly suitable for high-resolution MR images with submillimeter voxel size. The method uses a mathematical model of a field in an inhomogeneous dielectric. This field mapping, similarly to a Laplacian mapping, has nice laminar properties in the cortical layer, and helps to identify the unresolved boundaries between cortical banks in narrow sulci. The pial cortical surface is reconstructed by advection along the field gradient as a geometric deformable model constrained by topology-preserving level set approach. The method's performance is illustrated on exvivo images with 0.25-0.35 mm isotropic voxels. The method is further evaluated by cross-comparison with results of the FreeSurfer software on standard resolution data sets from the OASIS database featuring pairs of repeated scans for 20 healthy young subjects.

Diffusion magnetic resonance (MR) imaging has enabled us to reveal the white matter geometry in the living human brain. The Q-ball technique is widely used nowadays to recover the orientational heterogeneity of the intra-voxel fiber architecture. This article proposes to employ the Funk-Radon transform in a Hilbert space with a reproducing kernel derived from the spherical Laplace-Beltrami operator, thus generalizing previous approaches that assume a bandlimited diffusion signal. The function estimation problem is solved within a Tikhonov regularization framework, while a Gaussian process model allows for the selection of the smoothing parameter and the specification of confidence bands. Shortcomings of Q-ball imaging are discussed.

Registration of histology to three-dimensional (3D) magnetic resonance (MR) images is often required for the analysis of brain structure and investigation of brain pathologies. A novel algorithm for deformable registration of an individual histological section to a brain MR image is described. The cost function uses a novel hybrid intensity- and boundary surface-based measure that reflects the contrast of histological slice intensities across the boundary of the pial and inner cortical surface. The algorithm relies on implicit representation of cortical surfaces reconstructed from an anatomical MR image, and computes the cost function in a level set framework. The algorithm is evaluated on cross-modality registration of myelin-stained histological sections to a high-resolution MR image of the human brain.

Registration of histological images to three-dimensional imaging modalities is an important step in quantitative analysis of brain structure, in architectonic mapping of the brain, and in investigation of the pathology of a brain disease. Reconstruction of histology volume from serial sections is a well-established procedure, but it does not address registration of individual slices from sparse sections, which is the aim of the slice-to-volume approach. This study presents a flexible framework for intensity-based slice-to-volume nonrigid registration algorithms with a geometric transformation deformation field parametrized by various classes of spline functions: thin-plate splines (TPS), Gaussian elastic body splines (GEBS), or cubic B-splines. Algorithms are applied to cross-modality registration of histological and magnetic resonance images of the human brain. Registration performance is evaluated across a range of optimization algorithms and intensity-based cost functions. For a particular case of histological data, best results are obtained with a TPS three-dimensional (3D) warp, a new unconstrained optimization algorithm (NEWUOA), and a correlation-coefficient-based cost function.

Morphometry of brain structures based on magnetic resonance imaging (MRI) data has become an important tool in neurobiology. Recent multicenter studies in neurodegenerative diseases raised the issue of the precision of volumetric measures, and their dependence on the scanner properties and imaging protocol. A large dataset consisting of 1073 MRI examinations in 843 subjects, acquired on 90 scanners at 58 sites, is analyzed here. A comprehensive set of image quality and content measures is used to describe the influence of the scanner hardware and imaging protocol on the variability of morphometric measures. Scanners equipped with array coils show a remarkable advantage over conventional coils in terms of image quality measures. The signal- and contrast-to-noise ratio in similar systems is equal or slightly better at 1.5 T than 3.0 T, while the white/grey matter tissue contrast is generally better on high-field systems. Repeated MRI investigations on the same scanner were available in 41 subjects, on different scanners in 172 subjects. The retest reliability of repeated volumetric measures under the same conditions was found as sufficient to track changes in longitudinal examinations in individual subjects. Using different acquisition conditions in the same subject, however, the variance of volumetric measures was up to 10 times greater. The topologically correct and geometrically accurate reconstruction of the cerebral cortex from magnetic resonance (MR) images is an important step in quantitative analysis of the human brain structure, e.g. in cortical thickness measurement studies. Limited resolution of MR images, noise, intensity inhomogeneities, and partial volume effects can all contribute to geometrical inaccuracies and topological errors in the model of cortical surfaces. For example, unresolved touching banks of gray matter (GM) in narrow sulci pose a particular challenge for an automated algorithm, requiring specific steps for the recovery of separating boundaries. We present a method for the automated reconstruction of the cortical compartment from MR images. The method is based on several partial differential equation (PDE) modelling stages. First, a potential field is computed in an electrostatic model with GM posing as an insulating dielectric layer surrounding a charged conductive white matter (WM) object. Second, geodesic distances from WM along the streamlines of the potential field are computed in a Eulerian framework PDE. Third, a digital skeleton surface separating GM sulcal banks is derived by finding shocks in the distance field. At the last stage, a geometric deformable model based on the level set PDE is used to reconstruct the outer cortical surface by advection along the gradient of the distance or potential field. The rule preserving the digital topology, and the skeleton of the distance field resolving fused adjacent banks in sulci, constrain the deformable model evolution. In addition, the deformable model may use the distance field as a constraint on thickness of the reconstructed cortical layer.

Background: With the exception of APOE epsilon4 allele, the common genetic risk factors for sporadic Alzheimer's Disease (AD) are unknown., Methods and Findings: We completed a genome-wide association study on 381 participants in the ADNI (Alzheimer's Disease Neuroimaging Initiative) study. Samples were genotyped using the Illumina Human610-Quad BeadChip. 516,645 unique Single Nucleotide Polymorphisms (SNPs) were included in the analysis following quality control measures. The genotype data and raw genetic data are freely available for download (LONI, http://www.loni.ucla.edu/ADNI/Data/). Two analyses were completed: a standard case-control analysis, and a novel approach using hippocampal atrophy measured on MRI as an objectively defined, quantitative phenotype. A General Linear Model was applied to identify SNPs for which there was an interaction between the genotype and diagnosis on the quantitative trait. The case-control analysis identified APOE and a new risk gene, TOMM40 (translocase of outer mitochondrial membrane 40), at a genome-wide significance level of < or =10(-6) (10(-11) for a haplotype). TOMM40 risk alleles were approximately twice as frequent in AD subjects as controls. The quantitative trait analysis identified 21 genes or chromosomal areas with at least one SNP with a p-value < or =10(-6), which can be considered potential "new" candidate loci to explore in the etiology of sporadic AD. These candidates included EFNA5, CAND1, MAGI2, ARSB, and PRUNE2, genes involved in the regulation of protein degradation, apoptosis, neuronal loss and neurodevelopment. Thus, we identified common genetic variants associated with the increased risk of developing AD in the ADNI cohort, and present publicly available genome-wide data. Supportive evidence based on case-control studies and biological plausibility by gene annotation is provided. Currently no available sample with both imaging and genetic data is available for replication., Conclusions: Using hippocampal atrophy as a quantitative phenotype in a genome-wide scan, we have identified candidate risk genes for sporadic Alzheimer's disease that merit further investigation.

Functional neuroimaging studies have reported that the neural correlates of retrieval success (old>new effects) are larger and more widespread in older than in young adults. In the present study we investigated whether this pattern of age-related 'over-recruitment' continues into advanced age. Using functional magnetic resonance imaging (fMRI), retrieval-related activity from two groups (N=18 per group) of older adults aged 84-96 years ('old-old') and 64-77 years ('young-old') was contrasted. Subjects studied a series of pictures, half of which were presented once, and half twice. At test, subjects indicated whether each presented picture was old or new. Recognition performance of the old-old subjects for twice-studied items was equivalent to that of the young-old subjects for once-studied items. Old>new effects common to the two groups were identified in several cortical regions, including medial and lateral parietal and prefrontal cortex. There were no regions where these effects were of greater magnitude in the old-old group, and thus no evidence of over-recruitment in this group relative to the young-old individuals. In one region of medial parietal cortex, effects were greater (and only significant) in the young-old group. The failure to find evidence of over-recruitment in the old-old subjects relative to the young-old group, despite their markedly poorer cognitive performance, suggests that age-related over-recruitment effects plateau in advanced age. The findings for the medial parietal cortex underscore the sensitivity of this cortical region to increasing age.

The architectonic analysis of the human cerebral cortex is presently based on the examination of stained tissue sections. Recent progress in high-resolution magnetic resonance imaging (MRI) promotes the feasibility of an in vivo architectonic analysis. Since the exact relationship between the laminar fine-structure of a cortical MRI signal and histological cyto-and myeloarchitectonic staining patterns is not known, a quantitative study comparing high-resolution MRI to histological ground truth images is necessary for validating a future MRI based architectonic analysis. This communication describes an ongoing study comparing post mortem MR images to a myelin-stained histology of the brain cortex. After establishing a close spatial correspondence between histological sections and MRI using a slice-to-volume nonrigid registration algorithm, transcortical intensity profiles, extracted from both imaging modalities along curved trajectories of a Laplacian vector field, are compared via a cross-correlational analysis.

The neocortical surface has a rich and complex structure comprised of folds (gyri) and fissures (sulci). Sulci are important macroscopic landmarks for orientation on the cortex. A precise segmentation and labeling of sulci is helpful in human brain mapping studies relating brain anatomy and function. Due to their structural complexity and inter-subject variability, this is considered as a non-trivial task. An automatic algorithm is proposed to accurately segment neocortical sulci: vertices of a white/gray matter interface mesh are classified under a Bayesian framework as belonging to gyral and sulcal compartments using information about their geodesic depth and local curvature. Then, vertices are collected into sulcal regions by a watershed-like growing method. Experimental results demonstrate that the method is accurate and robust.

Complex shapes - such as the surface of the human brain - may be represented and analyzed in frequency space by means of a spherical harmonics transformation. A key step of the processing chain is introducing a suitable parametrization of the triangular mesh representing the brain surface. This problem corresponds to mapping a surface of topological genus zero on a unit sphere. An algorithm is described that produces an optimal combination of an area- and angle-preserving mapping. A multi-resolution scheme provides the robustness required to map the highly detailed and convoluted brain surface. More than 1000 datasets were successfully processed by this mature and robust approach.

Diffuse lesions of the white matter of the human brain are common pathological findings in magnetic resonance images of elderly subjects. These lesions are typically caused by small vessel diseases (e.g., due to hypertension, diabetes), and related to cognitive decline. Because these lesions are inhomogeneous, unsharp, and faint, but show an intensity pattern that is different from the adjacent healthy tissue, a segmentation based on texture properties is proposed here. This method was successfully applied to a set of 116 image data sets of elderly subjects. Quantitative measures for the lesion load are derived that compare well with results from experts that visually rated lesions on a semiquantitative scale. Texture-based segmentation can be considered as a general method for lesion segmentation, and an outline for adapting this method to similar problems is presented.

The purpose of this work was to study specific texture properties of the brain's white matter (WM) based on conventional high-resolution T1-weighted magnetic resonance imaging (MRI) datasets. Quantitative parameters anisotropy and laminarity were derived from 3-D texture analysis. Differences in WM texture associated with gender were evaluated on an age-matched sample of 210 young healthy subjects (mean age 24.8, SD 3.97 years, 103 males and 107 females). Changes of WM texture with age were studied using 112 MRI-T1 datasets of healthy subjects aged 16 to 70 years (57 males and 55 females). Both texture measures indicated a "more regular" WM structure in females (p < 10(-6)). An age-related deterioration of WM structure manifests itself as a remarkable decline of both parameters (p < 10(-6)) that is more prominent in females (p < 10(-6)) than in males (p = 0.02). Texture analysis of anatomical MRI-T1 brain datasets provides quantitative information about macroscopic WM characteristics and helps discriminating between normal and pathological aging.

The size of head compartments (head and brain volume, intracranial volume, gray and white matter volume, cerebrospinal fluid volume) and their ratios were determined on the basis of magnetic resonance images of the head acquired in a reference population of 502 healthy subjects. Age-matched subgroups were selected to reveal gender-related differences and changes with age. Normative data are provided in the form of simple equations that allow transforming measured compartment volumes into z scores, offering the possibility to relate individual data to a larger population.

One problem of providing time-critical medical services over the grid is always its dependency on the Internet. It cannot be assumed that transfer ofa certain amount of data over the Internet is always achieved during a specified period. Such a requirement cannot be fulfilled by the infrastructure of the Web. There is always the risk of a network delay or even an overload. Because of this, another goal of this project is the evaluation of grid services versus the use of local services. A further point for future research related to the chain has to deal with the optimization approach for the linear registration step. Because the optimization uses the downhill simplex algorithm in a nine-dimensional search space, the number of iterations needed to find the optimum can vary dramatically. This makes linear registration the most unpredictable step of the chain in terms of execution time. It cannot be assured that the global optimum is found. Additional work has to be done in validating the registration accuracy, including the examination of the influence of intensity variations between intraoperative images as well as the influence of tumor resection and the presence of the opened skull versus the closed skull in the fluid-based registration.

The structural correlates of mild cognitive impairment (MCI) were examined in 105 elderly subjects whose cognitive function ranged from intact to demented, including 38 subjects with MCI. Hippocampal volumes (left and right HcV), brain volume (BV), and grey matter volume (GMV) and white matter volume (WMV) were segmented from high resolution magnetic resonance data sets and normalised to intracranial volume (ICV). Hippocampal volume reductions, but not global brain, white or grey matter atrophy, were associated with MCI. White matter lesion severity did not differ over cognitive states. In multiple logistic regression models, normalised HcV and ICV (indicating premorbid brain volume) were significant predictors of MCI versus normality. Normalised BV and ICV significantly predicted dementia versus MCI. Absolute volumetric measures of HcV and BV yielded comparable classification accuracies. Hippocampal atrophy may be the crucial step for the transition from normality to MCI. Widespread brain atrophy may be the step to determine the transition from MCI to dementia. Brain volume reserve effects appear to be involved in both of these steps.

This paper is concerned with the detection of multiple small brain lesions from magnetic resonance imaging (MRI) data. A model based on the marked point process framework is designed to detect Virchow-Robin spaces (VRSs). These tubular shaped spaces are due to retraction of the brain parenchyma from its supplying arteries. VRS are described by simple geometrical objects that are introduced as small tubular structures. Their radiometric properties are embedded in a data term. A prior model includes interactions describing the clustering property of VRS. A Reversible Jump Markov Chain Monte Carlo algorithm (RJMCMC) optimizes the proposed model, obtained by multiplying the prior and the data model. Example results are shown on T1-weighted MRI datasets of elderly subjects.

The pattern of callosal atrophy might be useful for the differentiation between frontotemporal dementia (FTD) and Alzheimer's disease (AD) in advanced cases. However, it is unclear whether the pattern of callosal atrophy differs between patients with FTD and patients with AD in mild to moderate stages. Volumetric MR images were recorded from 48 probands (12 with FTD, 12 with late-onset AD, and 24 controls). All patients were in a mild or in a moderate stage. The corpus callosum was divided into five segments. A repeated-measures analysis of variance showed that there was no difference in the pattern of callosal atrophy between the groups. We provide evidence that patients with FTD and patients with late-onset AD do not differ in the pattern of callosal atrophy on condition that: (1) FTD patients and AD patients are in a mild to moderate stage and (2) FTD patients and AD patients differ in age., (Copyright 2004 S. Cytoarchitectonic fields of the human neocortex are defined by characteristic variations in the composition of a general six-layer structure. It is commonly accepted that these fields correspond to functionally homogeneous entities. Diligent techniques were developed to characterize cytoarchitectonic fields by staining sections of post-mortem brains and subsequent statistical evaluation. Fields were found to show a considerable interindividual variability in extent and relation to macroscopic anatomical landmarks. With upcoming new high-resolution magnetic resonance imaging (MRI) protocols, it appears worthwhile to examine the feasibility of characterizing the neocortical fine-structure from anatomical MRI scans, thus, defining neocortical fields by in vivo techniques. A fixated brain hemisphere was scanned at a resolution of approximately 0.3 mm. After correcting for intensity inhomogeneities in the dataset, the cortex boundaries (the white/grey matter and grey matter/background interfaces) were determined as a triangular mesh. Radial intensity profiles following the shortest path through the cortex were computed and characterized by a sparse set of features. A statistical similarity measure between features of different regions was defined, and served to define the extent of Brodmann's Areas 4, 17, 44 and 45 in this dataset.

Effects of gender and age on structural brain asymmetry were studied by 3D texture analysis in 380 adults. Asymmetry is detected by comparing the complex 3D gray-scale image patterns in the left and right cerebral hemispheres as revealed by anatomical T1-weighted MRI datasets. The Talairach and Tournoux parcellation system was applied to study the asymmetry on five levels: the whole cerebrum, nine coronal sections, 12 axial sections, boxes resulting from both coronal and axial subdivisions, and by a sliding spherical window of 9 mm diameter. The analysis revealed that the brain asymmetry increases in the anterior-posterior direction starting from the central region onward. Male brains were found to be more asymmetric than female. This gender-related effect is noticeable in all brain areas but is most significant in the superior temporal gyrus, Heschl's gyrus, the adjacent white matter regions in the temporal stem and the knee of the optic radiation, the thalamus, and the posterior cingulate. The brain asymmetry increases significantly with age in the inferior frontal gyrus, anterior insula, anterior cingulate, parahippocampal gyrus, retrosplenial cortex, coronal radiata, and knee region of the internal capsule. Asymmetry decreases with age in the optic radiation, precentral gyrus, and angular gyrus. The texture-based method reported here is based on extended multisort cooccurrence matrices that employ intensity, gradient, and anisotropy features in a uniform way. It is sensitive, simple to reproduce, robust, and unbiased in the sense that segmentation of brain compartments and spatial transformations are not necessary. Thus, it should be considered as another tool for digital morphometry in neuroscience.
