Abstract: Abstract mHealth, such as apps running on consumer smart devices is becoming increasingly popular and has the potential to profoundly affect healthcare and health outcomes. However, it may be disruptive and results achieved are not always reaching the goals. Allergic Rhinitis and its Impact on Asthma (ARIA) has evolved from a guideline using the best evidence-based approach to care pathways suited to real-life using mobile technology in allergic rhinitis (AR) and asthma multimorbidity. Patients largely use over-the-counter medications dispensed in pharmacies. Shared decision making centered around the patient and based on self-management should be the norm. Mobile Airways Sentinel networK (MASK), the Phase 3 ARIA initiative, is based on the freely available MASK app (the Allergy Diary, Android and iOS platforms). MASK is available in 16 languages and deployed in 23 countries. The present paper provides an overview of the methods used in MASK and the key results obtained to date. These include a novel phenotypic characterization of the patients, confirmation of the impact of allergic rhinitis on work productivity and treatment patterns in real life. Most patients appear to self-medicate, are often non-adherent and do not follow guidelines. Moreover, the Allergy Diary is able to distinguish between AR medications. The potential usefulness of MASK will be further explored by POLLAR (Impact of Air Pollution on Asthma and Rhinitis), a new Horizon 2020 project using the Allergy Diary.

Abstract: The rapid expansion of the aviation sector raises concerns about air quality impacts within and around airports. Ultrafine particles (UFP, diameter 2-fold) of Fe, Al, Cr, Cu, Mo, Mn, Pb, Ti, and Sb at the airport compared to the UB, with Al exhibiting the most pronounced disparity. Generally, PAH levels were low at both sites, although concentrations were higher at the airport relative to the UB. Overall, this study provides a comprehensive understanding of UFP within a major European airport, identifying the different sources contributing to PNC and PNSD.

This is the only therapy with an aetiological aim that modifies the course of the disease by ensuring remission after the interruption of the procedure. The prevention of new sensitizations by immunotherapy is still under discussion. In this review we will consider the main immunological mechanisms and indications for immunotherapy in children and adolescents.

Abstract: The pressing need for multifunctional materials in medical settings encompasses a wide array of scenarios, necessitating specific tissue functionalities. A critical challenge is the occurrence of biofouling, particularly by contamination in surgical environments, a common cause of scaffolds impairment. Beyond the imperative to avoid infections, it is also essential to integrate scaffolds with living cells to allow for tissue regeneration, mediated by cell attachment. Here, we focus on the development of a versatile material for medical applications, driven by the diverse time-definite events after scaffold implantation. We investigate the potential of incorporating graphene oxide (GO) into polycaprolactone (PCL) and create a composite for 3D printing a scaffold with time-controlled antibacterial and anti-adhesive growth properties. Indeed, the as-produced PCL-GO scaffold displays a local hydrophobic effect, which is translated into a limitation of biological entities-attachment, including a diminished adhesion of bacteriophages and a reduction of E. coli and S. aureus adhesion of ∼81% and ∼69%, respectively. Moreover, the ability to 3D print PCL-GO scaffolds with different heights enables control over cell distribution and attachment, a feature that can be also exploited for cellular confinement, i.e., for microfluidics or wound healing applications. With time, the surface wettability increases, and the scaffold can be populated by cells. Finally, the presence of GO allows for the use of infrared light for the sterilization of scaffolds and the disruption of any bacteria cell that might adhere to the more hydrophilic surface. Overall, our results showcase the potential of PCL-GO as a versatile material for medical applications.

Abstract: Aerosols are one of the most important pollutants in the atmosphere and have been monitored for the past few decades by remote sensing and in situ observation platforms to assess the effectiveness of government-managed reduction emission policies and assess their impact on the radiative budget of the Earth's atmosphere. In fact, aerosols can directly modulate incoming short-wave solar radiation and outgoing long-wave radiation and indirectly influence cloud formation, lifetime, and precipitation. In this study, we quantitatively evaluated long-term temporal trends and seasonal variability from a climatological point of view of the optical and microphysical properties of atmospheric particulate matter at the Universitat Politècnica de Catalunya (UPC), Barcelona, Spain, over the past 17 years, through a synergy of lidar, sun photometer, and in situ concentration measurements. Interannual temporal changes in aerosol optical and microphysical properties are evaluated through the seasonal Mann–Kendall test. Long-term trends in the optical depth of the recovered aerosol; the Ångström exponent (AE); and the concentrations of PM10, PM2.5, and PM1 reveal that emission reduction policies implemented in the past decades were effective in improving air quality, with consistent drops in PM concentrations and optical depth of aerosols. The seasonal analysis of the 17-year average vertically resolved aerosol profiles obtained from lidar observations shows that during summer the aerosol layer can be found up to an altitude of 5 km, after a sharp decay in the first kilometer. In contrast, during the other seasons, the backscatter profiles fit a pronounced exponential decay well with a well-defined scale height. Long-range transport, especially dust outbreaks from the Sahara, is likely to occur throughout the year. During winter, the dust aerosol layers are floating above the boundary layer, while during the other seasons they can penetrate the layer. The analysis also revealed that intense, short-duration pollution events during winter, associated with dust outbreaks, have become more frequent and intense since 2016. This study sheds some light on the meteorological processes and conditions that can lead to the formation of haze and helps decision makers adopt mitigation strategies to preserve large metropolitan areas in the Mediterranean basin.

Abstract: For a Positive Matrix Factorization (PMF) aerosol source apportionment (SA) studies there is no standard procedure to select the most appropriate chemical components to be included in the input dataset for a given site typology, nor specific recommendations in this direction. However, these choices are crucial for the final SA outputs not only in terms of number of sources identified but also, and consequently, in the source contributions estimates. In fact, PMF tends to reproduce most of PM mass measured independently and introduced as a total variable in the input data, regardless of the percentage of PM mass which has been chemically characterized, so that the lack of some specific source tracers (e.g. levoglucosan) can potentially affect the results of the whole source apportionment study. The present study elaborates further on the same concept, evaluating quantitatively the impact of lacking specific sources’ tracers on the whole source apportionment, both in terms of identified sources and source contributions. This work aims to provide first recommendations on the most suitable and critical components to be included in PMF analyses in order to reduce PMF output uncertainty as much as possible, and better represent the most commons PM sources observed in many sites in Western countries. To this aim, we performed three sensitivity analyses on three different datasets across EU, including extended sets of organic tracers, in order to cover different types of urban conditions (Mediterranean, Continental, and Alpine), source types, and PM fractions. Our findings reveal that the vehicle exhaust source resulted to be less sensitive to the choice of analytes, although source contributions estimates can deviate significantly up to 44 %. On the other hand, for the detection of the non-exhaust one is clearly necessary to analyze specific inorganic elements. The choice of not analysing non-polar organics likely causes the loss of separation of exhaust and non-exhaust factors, thus obtaining a unique road traffic source, which provokes a significant bias of total contribution. Levoglucosan was, in most cases, crucial to identify biomass burning contributions in Milan and in Barcelona, in spite of the presence of PAHs in Barcelona, while for the case of Grenoble, even discarding levoglucosan, the presence of PAHs allowed identifying the BB factor. Modifying the rest of analytes provoke a systematic underestimation of biomass burning source contributions. SIA factors resulted to be generally overestimated with respect to the base case analysis, also in the case that ions were not included in the PMF analysis. Trace elements were crucial to identify shipping emissions (V and Ni) and industrial sources (Pb, Ni, Br, Zn, Mn, Cd and As). When changing the rest of input variables, the uncertainty was narrow for shipping but large for industrial processes. Major and trace elements were also crucial to identify the mineral/soil factor at all cities. Biogenic SOA and Anthropogenic SOA factors were sensitive to the presence of their molecular tracers, since the availability of OC alone is unable to separate a SOA factor. Arabitol and sorbitol were crucial to detecting fungal spores while odd number of higher alkanes (C27 to C31) for plant debris.

Abstract: Introduction L’omalizumab est une biotherapie indiquee chez l’enfant presentant un asthme allergique severe. Il est donc important de definir plus clairement la population cible qui peut beneficier au mieux de cette biotherapie. Objectifs Le but de cette etude est de definir les facteurs predictifs de reponse complete a l’omalizumab a 4 mois de traitement. Methodes Nous avons inclus retrospectivement des enfants presentant un asthme severe ou refractaire traite par omalizumab. La reponse complete a l’omalizumab etait evaluee a 4 mois lors d’une reunion multidisciplinaire (en tenant compte de l’ACT, des exacerbations severes, des EFR) et classee en reponse partielle, complete ou non reponse. Resultats Pour les 45 inclus dans l’etude, 22 patients (49 %) avaient une reponse complete, 20 (44 %) une reponse partielle, et 3 (7 %) etaient non repondeurs. Dans l’analyse par regression logistique la presence d’un eczema et un VEMS eleve etaient des facteurs predictifs de reponse complete a l’omalizumab a 4 mois (en comparaison aux non-repondeurs ou aux repondeurs partiels) avec un OR de 1,50 (IC 95 % [1,09 ; 1,9], p = 0,005), et 1,02 (IC 95 % [1,01 ; 1,20], p = 0,001), respectivement. Nous avons identifie 3 clusters independants de reponse a l’omalizumab. Cluster 1, 16 patients avec un asthme eosinophilique (eosinophilie > 300/mm3, n = 15 (94 %), p = 0,001), VEMS moyen 65,4 ± 10,2, (p = 0,006) avec une faible reponse a l’omalizumab (n = 1 (6 %), p Conclusion De multiples morbidites allergiques (dont l’eczema) avec des sensibilisations allergiques multiples et une fonction pulmonaire normale s’associent a une reponse precoce a l’omalizumab.

Abstract: Allergic Rhinitis and its Impact on Asthma (ARIA) has evolved from a guideline by using the best approach to integrated care pathways using mobile technology in patients with allergic rhinitis (AR) and asthma multimorbidity. The proposed next phase of ARIA is change management, with the aim of providing an active and healthy life to patients with rhinitis and to those with asthma multimorbidity across the lifecycle irrespective of their sex or socioeconomic status to reduce health and social inequities incurred by the disease. ARIA has followed the 8-step model of Kotter to assess and implement the effect of rhinitis on asthma multimorbidity and to propose multimorbid guidelines. A second change management strategy is proposed by ARIA Phase 4 to increase self-medication and shared decision making in rhinitis and asthma multimorbidity. Abstract: L’asthme de l’enfant est une des premieres causes de consultation aux urgences pediatriques. L’evaluation de la gravite de la crise, le choix du traitement et l’orientation du patient ameliorent le devenir a court terme des enfants asthmatiques. Apres un traitement adequat, certains pourront rentrer au domicile, d’autres necessiteront d’etre hospitalises. Les protocoles de soins permettent d’aider a la decision afin d’optimiser la prise en charge aux urgences. L’efficacite de ces protocoles depend entre autres de l’evaluation de la gravite de la crise, de la qualite du protocole et de son implementation en pratique. L’utilisation des scores cliniques simples peut etre une aide a l’evaluation de l’enfant des l’admission. L’objectif de cet article est de proposer un protocole de soins base sur l’experience et les pratiques d’une equipe multidisciplinaire a la lecture des dernieres publications. Le traitement des exacerbations et leurs applications en pratique clinique a fait l’objet de nombreuses publications recentes. Les β2-agonistes administres par voie inhalee constituent dans tous les cas le traitement prioritaire. Lorsqu’elle est indiquee, la corticotherapie orale doit etre administree precocement.

Abstract: International audience; The water at the proposed site of the CHIPS water Cherenkov detector has been studied to measure its attenuation length for Cherenkov light as a function of filtering time. A scaled model of the CHIPS detector filled with water from the Wentworth 2W pit, proposed site of the CHIPS deployment, in conjunction with a 3.2 m vertical column filled with this water, was used to study the transmission of 405 nm laser light. Results consistent with attenuation lengths of up to 100 m were observed for this wavelength with filtration and UV sterilization alone.

Abstract: Introduction La securite et l’efficacite de l’immunotherapie allergenique sous-cutanee (ITSC) dans l’asthme reste debattue. Nous avons evalue la securite et l’efficacite de l’ITSC sur l’asthme et les comorbidites allergiques chez des enfants atteints d’asthme allergique modere a severe. Methodes Nous avons collecte de facon retrospective des donnees sur les effets indesirables, le nombre annuel d’exacerbations severes, la dose de corticosteroides inhales (CSI) et l’activite des comorbidites allergiques, dans une cohorte d’enfants beneficiant d’une ITSC acceleree envers Dermatophagoides pteronyssinus. Resultats Soixante-douze enfants, ayant en majorite un asthme severe (56 %), ont ete inclus Les reactions systemiques severes representaient 0,3 % des 3455 injections realisees. L’utilisation d’adrenaline a ete necessaire pour un patient. Le taux d’exacerbations de l’asthme diminuait de facon significative apres 1 an d’ITSC (p Discussion Les donnees sur la securite de l’ITSC dans cette cohorte de patients atteints d’asthme persistant modere a severe presentent un interet pour la prise en charge de ce type de malades. Une des principales limites de cette etude est sa methodologie retrospective, sans groupe temoin. Conclusion Cette etude montre un taux faible de RSS au vu du nombre total d’injections realisees et une efficacite significative de l’ITSC sur l’asthme, particulierement en cas de phenotype exacerbateur.

Abstract: Acute sinusitis in children is a controversial issue in terms of its diagnostic criteria, classification and therapeutic management. A therapeutic delay can lead to complications if the cause is bacterial. Guidelines have been set, but they are not consensual in pediatrics. Complications of acute bacterial sinusitis are uncommon in children, but they can be extremely severe and cause high morbidity and mortality. Because of their rarity, they often are not identified early, exposing the patient to an unfavorable outcome. We report on a case of acute bacterial pan-sinusitis complicated with thrombophlebitis of the cavernous sinuses and meningitis in a 9-year-old child, in spite of early and adapted antibiotic therapy. The bacterial agent was Staphylococcus aureus, which had no resistance or toxin profile. The progression was favorable under intravenous antibiotic therapy and after bilateral sphenoidectomy. This case raises the question of the best therapy for acute bacterial sinusitis in pediatrics and the management of complications.

Abstract: Purpose Vernal keratoconjunctivis (VKC) is a severe form of pediatric ocular allergy, characterized by acute and chronic corneoconjunctival inflammation that may lead to visual sequelae. Although topical cyclosporine is usually effective, severe forms may be refractory and require prolonged steroid therapy. Omalizumab is a monoclonal anti IgE antibody, administered systematically and authorized for severe asthma. We report our clinical experience with omalizumab in severe VKC children. Methods We retrospectively reviewed the files of 4 boys treated with omalizumab because of severe VKC, defined as persistent corneal inflammation despite continuous topical 2% cyclosporine and steroid eye drops. Results Four boys, aged 7 to 13 years old, were treated. All children had asthma and 1 had severe lid eczema. Two patients had required supratarsal steroid injections. Omalizumab was administered every 2 weeks by subcutaneous injections, at doses varying from 450 to 600 mg per injection. Three patients out of 4 responded to the treatment, with a decrease in frequency and in duration of the inflammatory flares, and also a decreased need for topical steroid. However, the response was incomplete and they still had inflammatory corneoconjunctival flares despite continuous topical cyclosporine. On the other hand, asthma and lid eczema were completely controlled in these 3 patients. The fourth child did not respond to o and needed oral steroids for his VKC and his asthma. Noticeably, this patient did not have detectable sensitization to any allergen, contrary to the other cases. The treatment was stopped in this refractory case, but is still ongoing in all other cases, with a median duration of 16 months (6 to 26 months). Conclusions Omalizumab is an interesting treatment in severe refractory forms of VKC, but its efficacy is incomplete in these very severe cases.

Abstract: To clarify the evolution of pyrimidine fungicides under natural aquatic environmental conditions, the visible-light-promoted degradation of the model fungicide 2-amino-4-hydroxy-6-methylpyrimidine (AHMPD) has been studied in air-equilibrated pH 6 aqueous solutions in the presence of riboflavin (Rf), employing time-resolved and stationary kineticspectroscopic methods. AHMPD, a compound practically inert towards the attack of singlet molecular oxygen (O2(1Δg)), quenches excited singlet and triplet states of Rf with rate constants of 2.7 × 109 M1 s1 and 2.7 × 107 M1 s1, respectively. In the presence of AHMPD, the photodecomposition of Rf, which occurs from the excited triplet state of the pigment, depends on the concentration of the fungicide: at [Formula: see text]4050 mM a limited photochemistry occurs due to the quenching of excited singlet Rf, while at ca. 510 mM triplet Rf is largely photogenerated and subsequently quenched either by oxygen, giving rise to O2(1Δg), or by AHMPD, yielding semireduced Rf through an electron transfer process. Flash photolysis experiments and indirect auxiliary tests confirm the presence of superoxide anion generated by the reaction of Rf anion radical with the dissolved oxygen. The neat result of this intricate scheme of competitive reactions is the photodegradation of both AHMPD and Rf, mainly through a superoxide anion-mediated oxidation, although some contribution of O2(1Δg)-mediated photooxidations cannot be disregarded.Key words: photooxidation, pyrimidine derivatives, riboflavin, singlet molecular oxygen, superoxide anion.

28. Reproductive cycle of the sand lizard (Lacerta agilis) in its southwestern range

Abstract: The reproductive cycle of Lacerta agilis was studied in the Pyrenees at the southwestern boundary of the species range. The activity period is short (about 4.5 months). Males attain sexual maturity at smaller sizes than females and show mixed type spermatogenesis and spring spermiogenesis. Vitellogenesis in females is rapid (45 days for the whole population) with a long period of oviductal retention. Clutches are laid at the beginning of summer. During the reproductive period males mobilise reserves during the mating activities. Females deplete more fat reserves but later in the season than males and they are at their lowest after egg production. In both sexes, energy resources are mobilised from the fat bodies and also from the liver and the proximal part of the tail. Clutch size increases with female size but also depends on other factors. The short activity period does not appear to affect the reproductive cycle of this species. The gonad cycles and clutch traits do not show substantial differences from other European populations, perhaps with the exception of slight variation in the timing of reproductive events and, to a lesser extent, clutch frequency. This evidence strongly suggests that the limited southwestern range of Lacerta agilis is not due to environmental constraints but to very recent colonization.

Abstract: The frequency of emergency department visits for asthma is a major public health problem in pediatrics. The aim of this study is to establish the characteristics of children visiting pediatric emergency departments for acute asthma and to assess their therapeutic management prior to admission.A prospective clinical study performed during 3 months at the pediatric emergency department of the university teaching hospital of Clermont-Ferrand, of children aged 1 to 16 years admitted to the department with a clinical diagnosis of asthma exacerbation.One hundred and forty-three patients were included in the study. Asthma crises were moderate to severe in 69.2% of cases (n=99). Initial therapeutic management prior to the admission to the emergency department was appropriate in 17.5% of cases (n=25). Most of the known asthmatic patients had not been followed up by a pediatric pulmonologist (n=56). A crisis protocol had been set up in 16.5% of cases (n=20). Exacerbations were more severe among younger patients (P=0.002) and economically disadvantaged children (P=0.025).This study uncovers poor knowledge of the disease among asthmatic children and their families, and an insufficient awareness among health practitioners of current recommendations for the treatment of asthmatic children. Admissions to the emergency department for asthma could be partly avoided by improving diagnosis and therapeutic education.

32. Characterization of polymorphic microsatellite markers in the brine shrimp Artemia (Branchiopoda, Anostraca)

Abstract: The brine shrimp Artemia is a complex genus containing sexual species and parthenogenetic lineages. Artemia franciscana is native to America and its cysts (diapausing eggs) are used worldwide as a food source in aquaculture. As a consequence, this anostracan has become an invasive species in many hypersaline aquatic ecosystems of other continents. Parthenogenetic Artemia lineages occur only in the Old World. Ten and five microsatellite markers were developed to characterize two populations for A. franciscana and two populations for diploid parthenogenetic Artemia, respectively. For A. franciscana the number of alleles ranged from 11 to 58 per locus, while for parthenogens the number of alleles ranged from three to 10. The levels of heterozygosity in A. franciscana and in parthenogens ranged from 0.115 to 0.976 and from 0.000 to 0.971, respectively. These microsatellite loci showed a high population assignment power, which will be useful for future studies of population genetics and invasive processes in Artemia.

Abstract: Soluble polymeric photosensitizers, with various contents of the 4-nitro-1-naphthylcarbamoyl chromophore, and the corresponding low molecular weight model compound have been tested as triplet energy donors against trans -stilbene in dioxane solution. Stern-Volmer constants show that the higher the chromophore content in the polymer the lower is its sensitizing efficiency. This effect can be explained by efficient intramolecular self-quenching processes which take place in the functionalized polymers because of the large local concentrations of chromophore.

Abstract: We report the synthesis of novel derivatives of thioxanthone with built-in tertiary amino groups. The aim of tailoring these new structures was to have photoinitiators which could react into the photocurable formulation so that the chromophore becomes attached to the polymer matrix, so reducing the possibility of it being extracted by solvents. The general photochemistry was studied by measurements of u.v. absorption, fluorescence and phosphorescence; a photoreduction study was carried out. Luminescence properties have shown the importance of the built-in amino group in the behaviour of the singlet and triplet excited states. Photoreduction quantum yields of aminopropoxythioxanthones are low, confirming that the amino group acts as a quencher of the excited state as well as a hydrogen atom donor. Polymerization quantum yields, determined by photocalorimetry in the presence of oxygen, are slightly higher with aminopropoxythioxanthones than with simple 2-propoxythioxanthone.

Abstract: Acute sinusitis in children is a controversial issue in terms of its diagnostic criteria, classification and therapeutic management. A therapeutic delay can lead to complications if the cause is bacterial. Guidelines have been set, but they are not consensual in pediatrics. Complications of acute bacterial sinusitis are uncommon in children, but they can be extremely severe and cause high morbidity and mortality. Because of their rarity, they often are not identified early, exposing the patient to an unfavorable outcome. We report on a case of acute bacterial pan-sinusitis complicated with thrombophlebitis of the cavernous sinuses and meningitis in a 9-year-old child, in spite of early and adapted antibiotic therapy. The bacterial agent was Staphylococcus aureus, which had no resistance or toxin profile. The progression was favorable under intravenous antibiotic therapy and after bilateral sphenoidectomy. This case raises the question of the best therapy for acute bacterial sinusitis in pediatrics and the management of complications.

Abstract: The photophysical properties of multichromophoric dyes with borondipyrromethene (BODIPY) and poly- p-phenylene (di- p-phenylene and tri- p-phenylene) groups in the same molecule are studied in detail. The excitation of the polyphenylene moiety in the UV region leads to a strong visible fluorescent emission of the BODIPY chromophore, via intramolecular excitation energy transfer between both groups. Consequently, these multichromophoric dyes are characterized by a large "virtual" Stokes shift, with a high fluorescence capacity and an efficient laser emission. On the other hand, the photophysical properties of a related dichromophoric dye with a hydroxy end group at the di- p-phenylene moiety show an important decrease in the fluorescent emission due to a photoinduced electron transfer process in basic media. Therefore, its photophysical properties are sensitive to the environmental acidity/basicity and could be applied as a proton sensor.

Abstract: The history of molecular fluorescence is closely associated with the emission from plant extracts.N. Monardes, in his Historia Medicinal (Seville, 1565), was the firstto describe the blue opalescence of the water infusion of the wood of a Mexican tree used to treatkidney ailments. The strange optical properties of the wood, known as Lignum nephriticum(kidney wood), were later investigated by Kircher, Grimaldi, Boyle, Newton and many other scientists andnaturalists in the ensuing centuries. However, when G.G. Stokes published in 1852 the first correct relationshipbetween light absorption and fluorescence, his observations were based on the emission of quinine sulphatesolution, because in Europe the wood of Lignum nephriticum was no longeravailable and its botanic origin was unknown. An inspection of the works of sixteenth century Spanish missionariesand scholars who compiled information on the Aztec culture, such as Fr. Bernardino de Sahagun and FranciscoHernandez, indicates that pre-Hispanic Indian doctors had already noticed the blue color (fluorescence)of the infusion of coatli, a wood used to treat urinary diseases.Coatli wood was obtained from Eyserhardtia, a tree of the familyof Leguminosae, and is the most likely source of the exotic Lignum nephriticum. The wood of Eysenhardtia polystachyacontains large quantities of Coatline B, a rare C-glucosyl-α-hydroxydihydrochalcone. This compound gives rise to a fluorescentreaction product, in slightly alkaline water at room temperature, which is responsible for the blue emissionof Lignum nephriticum infusion.

Abstract: The photophysical, lasing and thermostability properties of newly synthesized analogs of the commercial dye pyrromethene 567 (PM567) have been measured in polymeric matrices of poly(methyl methacrylate) both when used as a dopant and when covalently bounded to the polymeric chain. These analogs have an acetoxy or a polymerizable methacryloyloxy group at the end of a polymethylene chain at Position 8 of the PM567 chromophore core. Clear correlations between photophysical and lasing characteristics are observed. Linking chain lengths with three or more methylene units give the highest fluorescence quantum yields (as high as 0.89) and lasing efficiencies (as high as 41%). The covalent linkage of the chromophore to the polymeric chain via the methacryloyloxy group improves the photostability of the PM567 chromophore.

Abstract: Microtubules offer a very large local concentration of binding sites for cytotoxic taxoids or for hypothetical endogenous regulators. Several compounds from diverse sources stabilize microtubules and arrest cell division similarly to the antitumour drug Taxol. We have investigated the subcellular location of the Taxol binding sites, employing a fluorescent taxoid (FLUTAX) that reversibly interacts with the Taxol binding sites of microtubules and induces cellular effects similar to Taxol. The specific binding of FLUTAX to a fraction of the available cellular binding sites effectively inhibits division of cultured human tumour cells at G(2)/M, and triggers apoptotic death. The loci of reversible binding, directly imaged in intact U937 cells treated with cytotoxic doses of fluorescent taxoid are the centrosomes, with a few associated microtubules in interphase cells, and the spindle pole microtubules in mitotic cells, instead of uniformly labelling the microtubule cytoskeleton. Cytoskeletal lesions induced and visualized with FLUTAX consist of microtubule bundles and abnormal mitoses, including monopolar spindles and highly fluorescent multipolar spindles. The multiple asters and monopolar spindles mark arrested U937 leukaemia and OVCAR-3 ovarian carcinoma cells on their path to apoptosis (as well as K562, HeLa, and MCF-7 cells). Depending on the FLUTAX treatment, OVCAR-3 cells died from abnormal mitosis or from a subsequent interphase with double chromatin content and lobulated nuclei (micronuclei), indicating impairment of centrosome separation. Fragmented centrosomes could be observed in FLUTAX-treated non-transformed 3T3.A31 cells, which developed micronuclei but were resistant to apoptosis. These results strongly suggest that centrosomal impairment by taxoid binding during interphase, in addition to the suppression of the kinetochore microtubule dynamics in the mitotic spindle, is a primary cause of cell cycle de-regulation and cell death.

Abstract: Microtubules are specifically and efficiently visualized with the new fluorescent taxoids 7-O-[N-(4'-fluoresceincarbonyl)-L-alanyl]taxol (FLUTAX) and 7-O-[N-(4'-tetramethylrhodaminecarbonyl)-L-alanyl]taxol (ROTAX). Similarly to taxol, FLUTAX and ROTAX are able to drive inactive GDP-liganded tubulin into microtubule assembly. One molecule of FLUTAX binds per alphabeta-tubulin dimer assembled, competing with taxol for the same microtubule binding site with an eightfold smaller relative affinity. FLUTAX-induced microtubule elongation is markedly Mg2+-dependent, encompassing the binding of one Mg2+ ion more per tubulin dimer polymerized than in the case of taxol. A small perturbation of the absorption spectrum of bound FLUTAX is consistent with a cationic microenvironment relative to the solution. The fluorescence anisotropy of FLUTAX increases by an order of magnitude upon binding to microtubules and time-resolved measurements indicate that the fluorescein moiety remains considerably mobile on a protein surface. The rate of labeling suggests that this is the outer microtubule wall. Alternatively, the microtubule lumen would be functional. FLUTAX- and ROTAX-induced microtubules, radial structures, and organized microtubule bundles are readily observed under the fluorescence microscope. Rapid and accurate visualization of native (or very mildly fixed) cytoplasmic and spindle microtubules of a variety of permeabilized cells is simply obtained with micromolar FLUTAX, with an advantage over immunofluorescence. In addition, FLUTAX labels the centrosomes of PtK2 cells more intensely than antibodies to alpha- or beta-tubulin, and co-localizing with antibodies to gamma-tubulin. Two brightly fluorescent spots, probably separating or duplicating centrioles, can be resolved in the centrosomes of interphase cells. This finding indicates that centrosomes may well be additional targets of action of taxoids. FLUTAX strongly labels microtubules near the spindle poles, as well as microtubules at the telophase spindle equator and the central part of the midbody in cytokinesis (instead of the dark zone frequently observed with immunofluorescence), suggesting a predominant interaction of FLUTAX with sites at which tubulin is newly polymerized. Nanomolar concentrations of FLUTAX also permit specific imaging of centrosomes, half-spindles and midbodies in growing U937 cells.

47. CALIOPE-Urban v1.0: coupling R-LINE with a mesoscale air quality modelling system for urban air quality forecasts over Barcelona city (Spain)

Abstract: The NO2 annual air quality limit value is systematically exceeded in many European cities. In this context, understanding human exposure, improving policy and planning, and providing forecasts requires the development of accurate air quality models at the urban (street level) scale. We describe CALIOPE-Urban, a system coupling CALIOPE – an operational mesoscale air quality forecast system based on the HERMES (emissions), WRF (meteorology) and CMAQ (chemistry) models – with the urban roadway dispersion model R-LINE. Our developments have focused on Barcelona city (Spain), but the methodology may be replicated for other cities in the future. WRF drives pollutant dispersion and CMAQ provides background concentrations to R-LINE. Key features of our system include the adaptation of R-LINE to street canyons, the use of a new methodology that considers upwind grid cells in CMAQ to avoid double counting traffic emissions, a new method to estimate local surface roughness within street canyons, and a vertical mixing parameterisation that considers urban geometry and atmospheric stability to calculate surface level background concentrations. We show that the latter is critical to correct the night-time overestimations in our system. Both CALIOPE and CALIOPE-Urban are evaluated using two sets of observations. The temporal variability is evaluated against measurements from five traffic sites and one urban background site for April–May 2013. While both systems show a fairly good agreement at the urban background site, CALIOPE-Urban shows a better agreement at traffic sites. The spatial variability is evaluated using 182 passive dosimeters that were distributed across Barcelona during 2 weeks for February–March 2017. In this case, the coupled system also shows a more realistic distribution than the mesoscale system, which systematically underpredicts NO2 close to traffic emission sources. Overall CALIOPE-Urban improves mesoscale model results, demonstrating that the combination of both scales provides a more realistic representation of NO2 spatio-temporal variability in Barcelona.

Abstract: Diatomaceous earth, or diatomite, is produced through the accumulation of diatom (Bacillariophyceae) skeletons (i.e. cell walls called frustules) made of amorphous silica. The porous, highly symmetrical structure and microscopic size of diatom cell walls make them ideal constituents of sensing devices and analytical chips. Here, we propose chemical methods to purify diatom frustules extracted from diatomaceous earth. Using photo deposition techniques, we grow gold nanoparticles on the surface of diatom skeletons and within the pores of the skeletons, where the size and density of nanoparticles can be controlled by changing the parameters of the synthesis. Resulting devices have an internal porous structure that can harvest molecules from a solution, and an external shell of gold nanoparticles that amplifies the electromagnetic field generated by the measurement laser in Raman or other spectroscopies. The combination of these effects enables the analysis of biological specimens, chemical analytes and pollutants in extremely low abundance ranges. The devices were demonstrated in the analysis of Bovine Serum Albumin in water with a concentration of 100 aM, and mineral oil with a concentration of 50 ppm. Abstract: In this study, the performance of two types of source apportionment models was evaluated by assessing the results provided by 40 different groups in the framework of an intercomparison organised by FAIRMODE WG3 (Forum for air quality modelling in Europe, Working Group 3). The evaluation was based on two performance indicators: z-scores and the root mean square error weighted by the reference uncertainty (RMSEu), with pre-established acceptability criteria. By involving models based on completely different and independent input data, such as receptor models (RMs) and chemical transport models (CTMs), the intercomparison provided a unique opportunity for their cross-validation. In addition, comparing the CTM chemical profiles with those measured directly at the source contributed to corroborate the consistency of the tested model results. The most commonly used RM was the US EPA- PMF version 5. RMs showed very good performance for the overall dataset (91% of z-scores accepted) while more difficulties were observed with the source contribution time series (72% of RMSEu accepted). Industrial activities proved to be the most difficult sources to be quantified by RMs, with high variability in the estimated contributions. In the CTMs, the sum of computed source contributions was lower than the measured gravimetric PM10 mass concentrations. The performance tests pointed out the differences between the two CTM approaches used for source apportionment in this study: brute force (or emission reduction impact) and tagged species methods. The sources meeting the z-score and RMSEu acceptability criteria tests were 50% and 86%, respectively. The CTM source contributions to PM10 were in the majority of cases lower than the RM averages for the corresponding source. The CTMs and RMs source contributions for the overall dataset were more comparable (83% of the z-scores accepted) than their time series (successful RMSEu in the range 25% - 34%). The comparability between CTMs and RMs varied depending on the source: traffic/exhaust and industry were the source categories with the best results in the RMSEu tests while the most critical ones were soil dust and road dust. The differences between RMs and CTMs source reconstructions confirmed the importance of cross validating the results of these two families of models. 50. Intercomparison of aerosol measurements performed with multi-wavelength Raman lidars, automatic lidars and ceilometers in the framework of INTERACT-II campaign

Abstract: Following the previous efforts of INTERACT (INTERcomparison of Aerosol and Cloud Tracking), the INTERACT-II campaign used multi-wavelength Raman lidar measurements to assess the performance of an automatic compact micro-pulse lidar (MiniMPL) and two ceilometers (CL51 and CS135) in providing reliable information about optical and geometric atmospheric aerosol properties. The campaign took place at the CNR-IMAA Atmospheric Observatory (760 m a. s. l. ; 40.60° N, 15.72° E) in the framework of ACTRIS-2 (Aerosol Clouds Trace gases Research InfraStructure) H2020 project. Co-located simultaneous measurements involving a MiniMPL, two ceilometers and two EARLINET multi-wavelength Raman lidars were performed from July to December 2016. The intercomparison highlighted that the MiniMPL range-corrected signals (RCSs) show, on average, a fractional difference with respect to those of CNR-IMAA Atmospheric Observatory (CIAO) lidars ranging from 5 to 15 % below 2.0 km a.s.l. (above sea level), largely due to the use of an inaccurate overlap correction, and smaller than 5 % in the free troposphere. For the CL51, the attenuated backscatter values have an average fractional difference with respect to CIAO lidars Finally, additional tests performed during the campaign using the CHM15k ceilometer operated at CIAO showed the clear need to investigate the CHM15k historical dataset (2010–2016) to evaluate potential effects of ceilometer laser fluctuations on calibration stability. The number of laser pulses shows an average variability of 10 % with respect to the nominal power which conforms to the ceilometer specifications. Nevertheless, laser pulses variability follows seasonal behavior with an increase in the number of laser pulses in summer and a decrease in winter. This contributes to explain the dependency of the ceilometer calibration constant on the environmental temperature hypothesized during INTERACT.
