Increasing evidence indicated that metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) acted as a key regulator in the proliferation and invasion of several cancers. However, the function of MALAT1 in the development of cholangiocarcinoma has not been experimentally established. In the present study, the expression levels of MALAT1 in cholangiocarcinoma cell lines were detected by quantitative real-time PCR. The effects of MALAT1 knockdown on the cell proliferation and invasion of cholangiocarcinoma cells were detected with Cell Counting Kit-8 (CCK-8), colony formation assay and Trans-well assay, respectively. The expressions of epithelial-mesenchymal transition (EMT)-related proteins (E-cadherin, Vimentin) were evaluated to discover whether the process of EMT was involved. We also evaluated the expression of phos-phatidylinositol-3-kinase/serine/threonine kinase (PI3K/Akt) signaling pathway proteins (PI3K, p-PI3K, Akt, p-Akt) to determine the associated molecular mechanism. And we discovered that MALAT1 was up-regulated in cholangiocarcinoma cancer cells. CCK-8, colony formation and trans-well assay showed that the proliferation and invasion of QBC-939 and RBE with MALAT1 knockdown were inhibited. Moreover, MALAT1 could promote EMT in cholangiocarcinoma cells. In addition, MALAT1 may activate PI3K/Akt pathway. These results indicated that MALAT1 promoted cholangiocarcinoma cell proliferation and invasion. The effects of MALAT1 on cholangiocarcinoma cells might be through activating the PI3K/Akt signaling pathway. These investigations may facilitate a better understanding of MALAT1 and it might be a potential therapeutic target for the treatment of cholangiocarcinoma.

The purpose of this study was to explore the role of cordycepin in human cholangiocarcinoma (CCA) cell growth and apoptosis. In the present study, colony formation assay, cell-counting kit-8 (CCK-8) assay and tumor xenograft experiment were performed to evaluate the effect of cordycepin on human CCA cell growth in vitro and in vivo; flow cytometric analysis was performed to evaluate the effect of cordycepin on cell apoptosis; quantitative real-time reverse transcription PCR (qRT-PCR) and western blot assays were performed to evaluate the expression levels of Caspase-3, Bcl-2 and Bax. The results showed that cordycepin inhibited cell growth in QBC939 and RBE cells in vitro and it could also inhibit QBC939 cells growth in vivo. Furthermore, the flow cytometric analysis, qRT-PCR and western blot assays showed that cordycepin could trigger QBC939 and RBE cells apoptosis by regulating the expression levels of Caspase-3, Bcl-2 and Bax. And we proposed that cordycepin could inhibit human CCA cell growth in vitro and in vivo, while, this function is related to the induction of cell apoptosis.

MicroRNAs are a group of gene expression regulators and some of which have been confirmed to be associated with acute viral myocarditis (VM). This study aims to find new biomarkers for VM diagnosis and explore the roles of miRNAs during the pathogenesis of VM.23 patients with acute myocarditis and 12 controls were included in this research. The expression of 10 candidate miRNAs in the serum exosome was examined by qRT-PCR. The direct targets were predicted using bioinformatics tools and then confirmed by dual luciferase assay and immunoblotting. Levels IL-6 of cell culture supernatants were determined by enzyme-linked immunosorbent assay. Six weeks old male mice were injected intraperitoneally with Coxsackievirus B3 (CVB3) and then treated by miRNA inhibitors through tail vein injection.Five miRNAs were found to have disturbed expression in the exosome and may have the potential to be used as biomarker for VM diagnosis. Meanwhile, the expression of miR-30a and -181d was also altered in the cells after CVB3 infection. We identified SOCS3 as a direct target of miR-30a and -181d. Furthermore, during CVB3 infection, up-regulated miR-30a and -181d are related to enhanced IL-6 level via modulating SOCS3 expression. miRNA inhibitors injection increased mice survival rate after CVB3 infection.miR-30a and -181d contribute to the over-activated inflammatory response to viral infection of the heart during coxsackievirus infection.

SUMMARYA field experiment was conducted to quantify changes in soil aggregation and aggregate-associated soil organic carbon (SOC) concentration 1, 3, 5 and 10 years after abandoned, salinized land in the Manasi River Basin was reclaimed for cotton (Gossypium hirsutum L.). Results showed that reclamation significantly increased SOC concentrations and SOC stocks. Specifically, 10 years of cotton production increased SOC concentrations by 45% in the 0–60 cm depth and SOC stocks by 35%. The SOC concentrations and stocks decreased as soil depth increased. Reclamation time, season and soil depth had significant interaction effects on SOC. The SOC concentrations were significantly and positively correlated with available soil nitrogen and available soil phosphorus. Compared with abandoned farmland, macro-aggregate-associated (>250 µm) SOC concentrations in the 0–60 cm depth increased by 47% after 5 years of cotton production and by 53% after 10 years of cotton production. The contribution of macro-aggregate-associated SOC to total SOC in the 0–60 cm depth increased by 87% after 5 years of cotton production and by 69% after 10 years of cotton production. The findings indicate that soil aggregates were more stable after abandoned, salinized farmland was reclaimed for cotton production. Furthermore, cotton production can increase SOC concentrations and sequester C in this arid area.

Potassium dihydrogen phosphate (KDP) is an important nonlinear optical crystal material for light frequency converters and Pockels photoelectric switches in laser systems. However, KDP is apt to fracture, is deliquescent, and can suffer from microstructural changes under a temperature variation. As such, KDP has been one of the most difficult-to-handle materials, but its properties have not been well understood. This paper aims to explore the mechanical properties of KDP crystals in detail with the aid of the nanoindentation technique using a Berkovich diamond indenter. It was found that the mechanical properties of KDP can be easily altered by machining-induced subsurface damage. It was also discovered that a KDP crystal is a visco-elasto-plastic material during micro/nanoscale deformation, although it is very brittle macroscopically.

The coalescence of drops is a fundamental process in the coarsening of emulsions. However, counter-intuitively, this coalescence process can produce a satellite, approximately half the size of the original drop, which is detrimental to the overall coarsening. This also occurs during the coalescence of bubbles, while the resulting satellite is much smaller, approximately 10 %. To understand this difference, we have conducted a set of coalescence experiments using xenon bubbles inside a pressure chamber, where we can continuously raise the pressure from 1 up to 85 atm and thereby vary the density ratio between the inner and outer fluid, from 0.005 up to unity. Using high-speed video imaging, we observe a continuous increase in satellite size as the inner density is varied from the bubble to emulsion-droplet conditions, with the most rapid changes occurring as the bubble density grows up to 15 % of that of the surrounding liquid. We propose a model that successfully relates the satellite size to the capillary wave mode responsible for its pinch-off and the overall deformations from the drainage. The wavelength of the primary wave changes during its travel to the apex, with the instantaneous speed adjusting to the local wavelength. By estimating the travel time of this wave mode on the bubble surface, we also show that the model is consistent with the experiments. This wavenumber is determined by both the global drainage as well as the interface shapes during the rapid coalescence in the neck connecting the two drops or bubbles. The rate of drainage is shown to scale with the density of the inner fluid. Empirically, we find that the pinch-off occurs when 60 % of the bubble fluid has drained from it. Numerical simulations using the volume-of-fluid method with dynamic adaptive grid refinement can reproduce these dynamics, as well as show the associated vortical structure and stirring of the coalescing fluid masses. Enhanced stirring is observed for cases with second-stage pinch-offs. Numerous sub-satellites are observed when the length of the top protrusion of the drop exceeds the Rayleigh instability wavelength. We also find a parameter regime where the focusing of more than one capillary wave can pinch-off satellites. One realization shows a sequence of three pinch-offs, where the middle one pinches off a toroidal bubble.

Breast cancer (BC) is one of the most common malignant tumors occurred in women. There is no sensitive and specific marker for early diagnosis, treatment and prognosis of breast cancer. It is suggested that miRNA may be a potential tumor marker for breast cancer. Mir-520g is considered to be associated with many tumors. This study aims to test the expression of mir-520g in peripheral blood of BC patients and healthy control. We also explored the relationship between mir-520g and several prognostic factors in breast cancer patients.The peripheral blood of 86 cases with breast cancer (including 18 cases with stage 0, 24 cases of phase I, 20 cases of stage II, 24 cases of stage III) and 26 cases of healthy subjects were collected. The miR-520g level was measured by real-time quantitative PCR (RT qPCR) method. The correlation between plasma miR-520g level and the clinical stage, molecular subtype, receptors' expression and other factors related to the prognosis of the patients were examined.Plasma mir-520g expression levels were significantly higher in BC patients with lymph node metastatic and low differentiation degree grade (p = 0.033 and 0.016), and plasma miR-520g expression was significantly higher in breast cancer patients with mammary gland invasion (p0.01) and low expressed p53 (p = 0.0039).Highly expressed mir-520g is associated with lymph node metastasis and low differentiation of breast cancer, and also is associated with mammary gland invasion in breast cancer. This study suggests that mir-520g may be associated with some important prognostic factors in breast cancer patients, and may have a potential value for breast cancer marker.

Based on the computational fluid dynamics technology, a cold flow model is built to study the characteristics of an entrained-flow gasifier with downdraft multi-injector, regarding the flow field, the distribution of solid particles, and their residence time under different injector arrangement conditions. Effects of the injector structures are also discussed. The simulation model is verified via experimental results of a cold flow gasifier test platform, where the diagnostic method including particle image velocimetry (PIV) is used to measure the flow fields. It is found that both an improper injector arrangement and an improper spacing between adjacent injectors could lead to a turbulent flow field. Such unstable flow field is mainly caused by the interference between adjacent injectors and the wall. Further study shows that the interference is intensified with the wrong injector arrangement and the decrease in the injector spacing. In addition, it is observed that a stable flow field distribution could be reached when six injectors are annularly arranged at the top of gasifier, which could establish a stable low-pressure zone at the center of the gasifier dome. The residence time distribution of solid particles is also obtained from this study. It is observed that certain amount of solid particles takes the short circuit under the condition of single injector, and the particles in the gasifier tend to be totally mixed flow under the seven-injector condition. The six-injector annular arrangement gasifier is found to have the most average particle residence time distribution which indicates less occurrence of backmixing and short circuit.

The Daya Bay experiment consists of functionally identical antineutrino detectors immersed in pools of ultrapure water in three well-separated underground experimental halls near two nuclear reactor complexes. These pools serve both as shields against natural, low-energy radiation, and as water Cherenkov detectors that efficiently detect cosmic muons using arrays of photomultiplier tubes. Each pool is covered by a plane of resistive plate chambers as an additional means of detecting muons. In-situ observations on the initiation and propagation behaviour of low-cycle fatigue small cracks in cast magnesium–aluminium alloys (AM50 and AM60B) were carried out with scanning electron microscopy (SEM) to elucidate the resistance to fatigue cracking and to evaluate the fatigue small crack growth rate accurately and quantitatively. The results indicate that the fatigue small cracks formed preferentially on β-phase (Mg17Al12) boundaries at room temperature. In addition, the effects of the parameters of stress levels in low-cycle fatigue and temperatures as well as microstructure on fatigue small crack propagation behaviour are revealed. The variation of crack open displacement (COD) with stress levels and cycles at elevated temperature shows that it is unsuitable to estimate the fatigue small crack growth rate of cast magnesium alloys using conventional measurement methods such as the plastic-replica technique due to the obvious difference between microscopic cracks in the open and closed states. A new measurement of the θ_(13) mixing angle has been obtained at the Daya Bay Reactor Neutrino Experiment via the detection of inverse beta decays tagged by neutron capture on hydrogen. The antineutrino events for hydrogen capture are distinct from those for gadolinium capture with largely different systematic uncertainties, allowing a determination independent of the gadolinium-capture result and an improvement on the precision of the θ_(13) measurement. With a 217-day antineutrino data set obtained with six antineutrino detectors and from six 2.9 GW_(th) reactors, the rate deficit observed at the far hall is interpreted as sin^2 2θ_(13) = 0.083±0.018 in the three-flavor oscillation model. When combined with the gadolinium-capture result from Daya Bay, we obtain sin^2 2θ_(13) = 0.089±0.008 as the final result for the six-antineutrino-detector configuration of the Daya Bay experiment.

A measurement of the energy dependence of antineutrino disappearance at the Daya Bay Reactor Neutrino Experiment is reported. Electron antineutrinos ($\overline{\nu}_{e}$) from six $2.9$ GW$_{\rm th}$ reactors were detected with six detectors deployed in two near (effective baselines 512 m and 561 m) and one far (1579 m) underground experimental halls. Using 217 days of data, 41589 (203809 and 92912) antineutrino candidates were detected in the far hall (near halls). An improved measurement of the oscillation amplitude $\sin^{2}2\theta_{13} = 0.090^{+0.008}_{-0.009} $ and the first direct measurement of the $\overline{\nu}_{e}$ mass-squared difference $|\Delta m^{2}_{ee}|= (2.59_{-0.20}^{+0.19}) \times 10^{-3}\ {\rm eV}^2 $ is obtained using the observed $\overline{\nu}_{e}$ rates and energy spectra in a three-neutrino framework. This value of $|\Delta m^{2}_{ee}|$ is consistent with $|\Delta m^{2}_{\mu\mu}|$ measured by muon neutrino disappearance, supporting the three-flavor oscillation model., Comment: As accepted for publication by Phys. A search for light sterile neutrino mixing was performed with the first 217 days of data from the Daya Bay Reactor Antineutrino Experiment. The experiment's unique configuration of multiple baselines from six 2.9~GW$_{\rm th}$ nuclear reactors to six antineutrino detectors deployed in two near (effective baselines 512~m and 561~m) and one far (1579~m) underground experimental halls makes it possible to test for oscillations to a fourth (sterile) neutrino in the $10^{\rm -3}~{\rm eV}^{2} < |\Delta m_{41}^{2}| < 0.3~{\rm eV}^{2}$ range. The relative spectral distortion due to electron antineutrino disappearance was found to be consistent with that of the three-flavor oscillation model. The derived limits on $\sin^22\theta_{14}$ cover the $10^{-3}~{\rm eV}^{2} \lesssim |\Delta m^{2}_{41}| \lesssim 0.1~{\rm eV}^{2}$ region, which was largely unexplored., Comment: 7 pages, 4 figures

The Daya Bay Reactor Neutrino Experiment has measured a non-zero value for the neutrino mixing angle $\theta_{13}$ with a significance of 5.2 standard deviations. Antineutrinos from six 2.9 GW$_{\rm th}$ reactors were detected in six antineutrino detectors deployed in two near (flux-weighted baseline 470 m and 576 m) and one far (1648 m) underground experimental halls. With a 43,000 ton-GW_{\rm th}-day livetime exposure in 55 days, 10416 (80376) electron antineutrino candidates were detected at the far hall (near halls). The ratio of the observed to expected number of antineutrinos at the far hall is $R=0.940\pm 0.011({\rm stat}) \pm 0.004({\rm syst})$. The Daya Bay Reactor Neutrino Experiment is designed to determine precisely the neutrino mixing angle $\theta_{13}$ with a sensitivity better than 0.01 in the parameter sin$^22\theta_{13}$ at the 90% confidence level. To achieve this goal, the collaboration will build eight functionally identical antineutrino detectors. The first two detectors have been constructed, installed and commissioned in Experimental Hall 1, with steady data-taking beginning September 23, 2011. This paper considers how to design coordination strategies to achieve the coordination in decentralized supply chain, considering pricing, inventory and transportation cost simultaneously under the price sensitive environment. Based on a supplier-Stackelberg game structure, we analyze the non-cooperation decision model and design the algorithm. Then we propose the cooperation decision model and design the algorithm. Revenue sharing and price discount strategies are used to coordinate the decentralized supply chain. The experimental results show that both revenue sharing and price discount strategy can efficiently make the coordination of the supplier and the retailer.

OBJECTIVE:This study was to establish cephalometric normals for Shangdong Chinese based on the Tweed-Merrifield analysis. MEHTODS: Lateral cephalograms of 80 young adults(40 female and 40 male) with normal occlusions were selected.The ages was 18-24. RESULTS: To establish cephalometric normals for Shangdong Chinese based on the Tweed-Merrifield analysis,and discover FMA femalemale,AFH,PFH,IND(P/A) femalemale. CONCLUSION: These normals will be used to orthodontic diagnosis and treatment planning for Shangdong adult patients.

To provide a series of surgical approaches for treatment of talus neck fracture, ischemic necrosis of talus body, and other bone lesions in ankle and foot.The major blood supply to cuboid bone, medial cuneiform bone and navicular bone was observed in 30 adult cadavers, by infiltration of red emulsion via major arteries of the lower limbs. Based on these anatomical investigations, 3 types of vascularized tarsal bone grafting were designed for repair of bone lesions in the area of ankle and foot, and applied in 49 clinical cases, ranging from 10 to 58 years in age, and 43 cases of which were followed up for 4 years and 3 months in average.Primary healing was achieved in 40 cases, and secondary healing achieved after further surgical intervention in other 3 cases. The function of all ankle joints recovered satisfactorily.The designed three types of vascularized tarsal bone flaps are easy and reliable for dissection because of their superficial pedicles, and they are available for different clinical cases with various bone lesions in ankle and foot.

The suppression of tumor formation, first demonstrated by somatic cell hybrid and microcell fusion experiments, suggests the existence of a class of genes that selectively suppress the growth of tumor cells but not normal cells. The reintroduction of these genes into tumor cells presumably renders the cells responsive to in vivo growth inhibitory environment. As the inheritance of a defective retinoblastoma gene (Rb-1) allele results in a predisposition to the development of various cancers, and since inactivation of both alleles are observed in tumor cells, the Rb gene has been suspected to have the ability to suppress tumor growth. Data presented here demonstrated that different types of normal cells, which have a limited life span, were also growth arrested by a transfected Rb gene. Cell lines which are resistant to the growth suppression effect of the Rb gene in vitro, retain the ability to form tumors in nude mice even in the presence of a stable and highly expressed wild type Rb protein. We conclude that while the Rb gene can suppress the growth of many tumor cell lines, its growth suppression effect is not tumor specific.

Most of the 19 cases of high myopia with esotropia fixus had the refractive error in childhood while esotropia fixus set in progressively during middle age. The eyeball was fixed in a position of marked adduction, and the forced duction test was positive. Electromyography showed normal innervation of the medial and lateral recti. Contracture of the medial rectus and thinning of the lateral rectus were observed during surgery. Histopathological examination revealed degeneration and fibrosis in the medial rectus. Etiology was not clear. Tenotomy of the medial rectus or that of the inferior rectus in addition, with fixation of the lateral rectus tendon to the lateral orbital rim produced satisfactory corrective results.

We grow epitaxial (La0.8Pr0.2)0.67Ca0.33MnO3 films simultaneously on the lattice-closely-matched substrates, cubic (LaAlO3)0.3(Sr2AlTaO6)0.7 [LSAT(001)] and orthorhombic NdGaO3 [NGO(001) and NGO(110)]. While all as-grown films show a ferromagnetic-metallic (FM) ground state as observed for the bulk target, the annealed films show quite different magnetotransport behavior as follows: on NGO(110) they show a robust FM ground state, on LSAT(001) and NGO(001) however, they show surprisingly a coexisted antiferromagnetic insulating state with high phase instability in a wide temperature range. The phase coexistence being easily induced via the control of anisotropic epitaxial strain suggests that the phase separation in manganites could be elastically driven, and thus can be strain-engineered for devices applications.

Free radical generating substances, including fulvic acids, mycotoxins, Fe(II), etc attack chondrocytes, causing dedifferentiation. The dedifferentiated cells synthesize and secrete abnormal collagens rich in type I instead of the normal type II. These substances also directly attack collagens in the extracellular matrix. The abnormal collagens make the matrix change from hydroxyapatite crystallization inhibiting to promoting, and give crystals of low crystallinity, high aggregation and small size. This study leads to a new concept of the development of Kaschin-Beck's Disease (KBD).

We experimentally study the formation of a satellite bubble, during the coalescence of two larger bubbles. The daughter bubble is generated by capillary waves which propagate from the growing neck, connecting the two bubbles and focus on the opposite apex. As the successive waves converge on the top, their amplitude grows until a small daughter bubble is pinched off. The mechanism is robust over a large range of parameters. We have observed it in water for bubble diameters from 5.2mm down to 175μm. For equally sized parent bubbles, the size of the daughter bubble is 0.10±0.03 times that of the mother bubble, but the daughter size also depends weakly on the approach velocity of the two initial bubbles. The effects of viscosity is to dampen the capillary waves, preventing the pinch-off for Ohnesorge number >6.5×10−3, which is significantly smaller than the critical value observed for the coalescence cascade of a drop. The effect of albendazole on the body wall and gut of Clornorchis sinensis was studied with transmission and scanning electron microscopes after albendazole administration to rats infected with Chonorchis sinensis at a single dose of 150 mg/kg. The results showed that swelling and adhesion of the projections of the tegument and gut microvilli occurred 1h after medication. Necrosis and disruption of the projections and the gut microvilli were seen at the 24th h. By the 72nd h, detachment of the partial projections were seen. The dynamic process of the damages observed on the tegument was identical with that of the gut microvilli (Figs. 1-10).

By using highly conductive p-type La0.7Sr0.3MnO3 (LSMO) and n-type La0.07Sr0.93SnO3 (LSSO) as electrodes, all-oxide Pb(Zr0.52Ti0.48)O3 (PZT) capacitors, LSMO/PZT/LSMO, LSSO/PZT/LSSO, LSSO/PZT/LSMO, and LSMO/PZT/LSSO, have been grown epitaxially on (001)SrTiO3 substrates, and their structure, switching, fatigue, and optical properties were investigated. Strikingly, contrary to the LSMO/PZT/LSMO capacitors, those having the n-type electrode show poor fatigue resistance especially at lower driving frequencies, which was further confirmed by using another n-type oxide electrode, SrTi0.9Sb0.1O3. The results suggest that with a depletion layer at the PZT/LSSO interface, charge accumulation and injection during switching may be responsible for the fatigue.

The scleral encircling or explant procedures for treatment of retinal detachment usually lead to post-operative strabismus, due to traumatic muscular adhesion to the globe, explant interference with muscular function, nerve damage and muscular paralysis, excessive or prolonged stretching of the muscle and surgical muscular reattachment at an improper site. In addition, there may be muscular atrophy or cicatrizing transformation due to compression. In 3 patients, the authors made use of the cicatricial tissue in place of the original muscle with or without surgical adjustment of the other ocular muscles. Satisfactory cosmetic result and ocular motility were obtained in all 3 cases.

Sequence analysis of the human retinoblastoma gene cDNA revealed the presence of repeated elements in the form of direct repeats, inverted repeats and dyad symmetries. The clustering of the dyad symmetrical elements in some exons, #16 and #17, coincides with the hot spots for structural aberrations of the RB-1 locus previously observed in tumors. The RB-1 gene is divided into at least 27 exons distributed over 200 kbp. Three potential Sp1 binding sites are presented within 600 bp upstream of the translation start site. A DNA fragment containing these Sp1 sites ligated to a promotorless CAT gene can promote its transcription in transfected cell culture.
