Abstract: The neutron time of flight (n_TOF) facility at CERN is a neutron spallation source, its white neutron energy spectrum ranges from thermal to several GeV, covering the full energy range of interest for nuclear astrophysics, in particular for measurements of the neutron capture cross-section required in s-process nucleosynthesis. This contribution gives an overview on the astrophysical program made at n_TOF facility, the results and the implications will be considered.

Abstract: The Am241(n,γ) cross section has been measured at the n_TOF facility at CERN using deuterated benzene liquid scintillators, commonly known as C6D6 detectors, and time-of-flight spectrometry. The results in the resolved resonance range bring new constraints to evaluations below 150 eV, and the energy upper limit was extended from 150 to 320 eV with a total of 172 new resonances not present in current evaluations. The thermal capture cross section was found to be σth=678±68 b, which is in good agreement with evaluations and most previous measurements. The capture cross section in the unresolved resonance region was extracted in the remaining energy range up to 150 keV, and found to be larger than current evaluations and previous measurements.

Abstract: The CERN Neutron Time-Of-Flight (n-TOF) facility is well suited to measure small neutron-induced fission cross sections, as those of subactinides. The cross section ratios of {sup nat}Pb and {sup 209}Bi relative to {sup 235}U and {sup 238}U were measured using PPAC detectors. The fragment coincidence method allows to unambiguously identify the fission events. The present experiment provides the first results for neutron-induced fission up to 1 GeV for {sup nat}Pb and {sup 209}Bi. A good agreement with previous experimental data below 200 MeV is shown. The comparison with proton-induced fission indicates that the limiting regime where neutron-induced and proton-induced fission reach equal cross section is close to 1 GeV.

Abstract: The precise determination of the neutron capture cross sections of Os186 and Os187 is important to define the s-process abundance of Os187 at the formation of the solar system. This quantity can be used to evaluate the radiogenic component of the abundance of Os187 due to the decay of the unstable Re187 (t1/2=41.2 Gyr) and from this to infer the time duration of the nucleosynthesis in our galaxy (Re/Os cosmochronometer). The neutron capture cross sections of Os186, Os187, and Os188 have been measured at the CERN n-TOF facility from 1 eV to 1 MeV, covering the entire energy range of astrophysical interest. The measurement has been performed by time-of-flight technique using isotopically enriched samples and two C6D6 scintillation detectors for recording the prompt γ rays emitted in the capture events. Maxwellian averaged capture cross sections have been determined for thermal energies between kT=5 and 100 keV corresponding to all possible s-process scenarios. The estimated uncertainties for the values at 30 keV are 4.1, 3.3, and 4.7% for Os186, Os187, and Os188, respectively. © 2010 The American Physical Society

Abstract: Neutron resonance analyses have been performed for the capture cross sections of 186Os, 187Os, and 188Os measured at the n_TOF facility at cern. Resonance parameters have been extracted up to 5, 3, and 8 keV, respectively, using the sammy code for a full R-matrix fit of the capture yields. From these results average resonance parameters were derived by a statistical analysis to provide a comprehensive experimental basis for modeling of the stellar neutron capture rates of these isotopes in terms of the Hauser-Feshbach statistical model. Consistent calculations for the capture and inelastic reaction channels are crucial for the evaluation of stellar enhancement factors to correct the Maxwellian averaged cross sections obtained from experimental data for the effect of thermally populated excited states. These factors have been calculated for the full temperature range of current scenarios of s-process nucleosynthesis using the combined information of the experimental data in the region of resolved resonances and in the continuum. The consequences of this analysis for the s-process component of the 187Os abundance and the related impact on the evaluation of the time duration of galactic nucleosynthesis via the Re/Os cosmochronometer are discussed.

Abstract: The U233(n,f) cross section has been measured at the white neutron source n-TOF in a wide energy range with a dedicated fission ionization chamber. We report here the results from ∼30 meV to 1 MeV neutron energy. The U233(n,f) cross section has been determined relative to a reference sample of U235(n,f) measured simultaneously with the same detector. The very high instantaneous neutron flux and the intrinsically low background of the n-TOF installation result in an accuracy around 3% in the whole energy range, while the energy resolution of the neutron beam allows for an accurate description of the fission cross section by means of R-matrix analysis over a wide energy range. The results are, in general, in good agreement with the most recent high-accuracy measurement of this fission cross section, over the more limited range of the previous measurements, and indicated that even the latest evaluations underestimate the cross section in the epithermal region. The present high-quality data provide the basis for a more precise evaluation of the U233 fission cross section and for improving the reliability of databases needed for the design of new energy systems based on the Th/U cycle. © 2009 The American Physical Society

Abstract: The n TOF collaboration operates a Total Absorption Calorimeter (TAC) [1] for measuring neu- tron capture cross-sections of low-mass and/or radioactive samples. The results obtained with the TAC have led to a substantial improvement of the capture cross sections of 237Np and 240Pu [2]. The experience acquired during the rst measurements has allowed us to optimize the performance of the TAC and to improve the capture signal to background ratio, thus opening the way to more complex and demanding measurements on rare radioactive materials. The new design has been reached by a series of detailed Monte Carlo simulations of complete experiments and dedicated test measurements. The new capture setup will be presented and the main achievements highlighted.

Abstract: The successful development of advanced nuclear systems for sustainable energy production and nuclear waste management depends on high quality nuclear data libraries. Recent sensitivity stud- ies and reports [1-3] have identi ed the need for substantially improving the accuracy of neutron cross-section data for minor actinides. The n TOF collaboration has initiated an ambitious exper- imental program for the measurement of neutron capture cross sections of minor actinides. Two experimental setups have been constructed for this purpose: a Total Absorption Calorimeter (TAC) [4] for measuring neutron capture cross-sections of low-mass and/or radioactive samples and a set of two low neutron sensitivity C6D6 detectors for the less radioactive materials.

Abstract: This article analyzes the content of 1,359 police civil liability cases handed down by the United States Federal District Courts from 1978 through 1990. A data classification system was developed to enable the content analysis to determine the longitudinal trends in these cases, the types of law enforcement agencies being sued, the prevailing parties, allegations against the police, and amounts of damages and attorney fees awarded to successful plaintiffs. Issues brought before the courts were classified, and high-risk areas of police behavior were identified in a longitudinal fashion. The research focused exclusively on published federal litigation brought against the police under Title 42 United States Code, Section 1983, as opposed to civil liability under state of federal tort law.

Abstract: The nuclear resonance parameters and the capture cross section of the neutron magic isotope 139La has been measured n_TOF. The description of the experimental apparata is followed by the data analysis procedures. We extracted the resonance parameters, the main nuclear quantities such as the resonance integral, the average gamma widths for s- and p-waves, the nuclear level densities and the neutron strength functions. These results represent a signifcant improvement over previous data. The deduced Maxwellian-averaged capture cross sections are important for the interpretation of the most recent spectroscopic observations in low metallicity stars. Abstract: Further investigation of the targeting of the intracellular membrane lectin endoplasmic reticulum (ER)-Golgi intermediate compartment-53 (ERGIC-53) by site-directed mutagenesis revealed that its lumenal and transmembrane domains together confer ER retention. In addition we show that the cytoplasmic domain is required for exit from the ER indicating that ERGIC-53 carries an ER-exit determinant. Two phenylalanines at the C terminus are essential for ER-exit. Thus, ERGIC-53 contains determinants for ER retention as well as anterograde transport which, in conjunction with a dilysine ER retrieval signal, control the continuous recycling of ERGIC-53 in the early secretory pathway. In vitro binding studies revealed a specific phenylalanine-dependent interaction between an ERGIC-53 cytosolic tail peptide and the COPII coat component Sec23p. These results suggest that the ER-exit of ERGIC-53 is mediated by direct interaction of its cytosolic tail with the Sec23p.Sec24p complex of COPII and that protein sorting at the level of the ER occurs by a mechanism similar to receptor-mediated endocytosis or Golgi to ER retrograde transport.

Abstract: ERGIC-53 (former designation, p53) is a 53-kDa nonglycosylated, dimeric, and hexameric type I membrane protein that has been established as a marker protein for a tubulovesicular intermediate compartment in which protein transport from the endoplasmic reticulum to the Golgi apparatus is blocked at 15 degrees C. Although ERGIC-53 is not a resident protein of the rough endoplasmic reticulum its cDNA sequence carries a double lysine endoplasmic reticulum retention motif at the cytoplasmically exposed COOH terminus. Here we report that overexpression of ERGIC-53 in COS cells saturates its intracellular retention system leading to the appearance of ERGIC-53 at the cell surface. Cell surface ERGIC-53 is efficiently endocytosed by a mechanism that is disturbed when the two critical lysines of the endoplasmic reticulum retention motif are replaced by serines. The results suggest a mechanistic similarity of pre-Golgi retention by the double lysine motif and lysine-based endocytosis.

18. Fast polarisation and wavelength switching in quasi-index guided GaInAsP twin-stripe lasers by direct current modulation

Abstract: Current-controlled polarisation and wavelength switching in 1.3 mu m GaInAsP ridge-wavelength twin-stripe (RWTS) lasers with a properly designed effective index profile is reported. In CW operation at room temperature, an output power of 4 mW can be switched from TM- to TE-polarisation with an extinction ratio of 15 dB by a control current of only 4 mA. The polarisation switching is accompanied by a wavelength shift of 17 nm. Under fast direct current modulation, intrinsic optical response times of less than 235 ps are obtained. A self-consistent analysis of the active lateral waveguide shows that the switching effect is based on the deformation of the TE-mode profile resulting from antiguiding controlled by a small injection current. >

19. Measurement of theHe3andHe4total neutron cross sections up to 40 MeV

Abstract: The total neutron cross sections of the helium isotopes were measured in the energy range from 1.5 to 40 MeV. A continuous energy neutron source was used in connection with a 190 m long flight path and high resolution time-of-flight techniques. The energy resolution achieved enabled us to improve considerably the parameters of the narrow (3/2)/sup +/ resonance in the n+ /sup 4/He system. The position of the resonance was found to be 22 133 +- 10 keV, with a total width of 76 +- 12 keV and a neutron width of 37 +- 5 keV. This agrees well with previous analyses, which yield GAMMA/sub n/approx.(1/2)GAMMA. Absolute values for the cross sections were determined with an overall error smaller than 3%. Comparison with older data shows agreement within the total errors. Our data are 4% to 5% higher than the most recent data set; however, in the case of /sup 3/He, the data are still below the predictions from an R-matrix analysis. It was pointed out recently that a slight disagreement exists between the total neutron cross section of /sup 3/He and the sum of the partial n-/sup 3/He cross sections. This discrepancy is removed by our new data.

20. Phase and amplitude characteristics of a synchronized S-band Trapatt oscillator

Abstract: The synchronization, of an S-band Trapatt oscillator working in the fundamental mode has been investigated. The phase shift between the input and output signal has been measured in dependence upon the frequency deviation from the free-running oscillator for various input power levels. It is shown that the phase difference can be described by Adler's locking equation using a new defined figure of merit. The high injection signals required for stable locking together with the significant non-linearity of the Trapatt diode cause an output power variation, which differs from the conventional locking ellipses. As a new result, the characteristic reduction of the output power within the locking range is explained by a model using a constant current source for the Trapatt diode. The theoretical results are in good agreement with measurements.

Abstract: The Karlsruhe 4 pi BaF/sub 2/ detector is composed of 42 crystals of 1.5-l volume, shaped as hexagonal or pentagonal truncated pyramids, forming a spherical shell of 20-cm inner diameter and 15-cm thickness. Each crystal is covered by a thin reflector and viewed by a phototube, thus representing a complete gamma-ray detector. The energy resolution of the individual detector modules is 10-11% at 662- keV and 4-5% at 6.13-MeV gamma-ray energy, while the time resolution as measured with a /sup 60/Co source is 400-500 ps. The overall efficiency of the entire detector is about 100% for gamma-rays up to 10 MeV. Compared to 4 pi detectors with comparable efficiency made from NaI (TI), the present setup offers five times better time resolution at similar resolution in gamma-ray energy. The detector will be used for the accurate determination of neutron capture reactions in the neutron energy range from 3 to 200 keV primarily for astrophysical applications. The performance of the detector under experimental conditions is demonstrated in various test measurements on gold samples. >

22. ERGIC-53 and traffic in the secretory pathway.

Abstract: The ER-Golgi intermediate compartment (ERGIC) marker ERGIC-53 is a mannose-specific membrane lectin operating as a cargo receptor for the transport of glycoproteins from the ER to the ERGIC. Lack of functional ERGIC-53 leads to a selective defect in secretion of glycoproteins in cultured cells and to hemophilia in humans. Beyond its interest as a transport receptor, ERGIC-53 is an attractive probe for studying numerous aspects of protein trafficking in the secretory pathway, including traffic routes, mechanisms of anterograde and retrograde traffic, retention of proteins in the ER, and the function of the ERGIC. Understanding these fundamental processes of cell biology will be crucial for the elucidation and treatment of many inherited and acquired diseases, such as cystic fibrosis, Alzheimer's disease and viral infections.

23. The Influence of Harmonic Injection on Power, Efficiency and Stability of Trapatt Amplifiers

Abstract: It is shown by an analytical model, that power and efficiency of TRAPATT diodes are limited by the fact, that the optimum diode impedances cannot be matched with a passive load circuit at some harmonics of the oscillation. These restrictions can be overcome by injection of a small amount of power at the respective harmonics. The experimental verification of this method has been performed with an unique lumped element circuit. By injection of the third harmonic fundamental power and efficiency could almost be doubled. At 1,7 GHz a peak power of 230 Wwith a total efficiency of 46% has been achieved. The maximum gain (from the third to the first harmonic) was 23 dB. The good symmetry of the output spectrum indicates high phase and amplitude stability.

24. High CW-Power Phase-locked Semiconductor Laser Arrays

Abstract: Very high coherent optical power can be achieved from recently developed GaAlAs laser arrays consisting of 10 to 40 closely spaced laser stripes integrated on a common substrate. Phase-locked operation of such a large number of lasers has been rendered possible by the high uniformity of the expitaxial layers grown by metal-organic vapour phase epitaxy (MO VPE). The achievable output povi/er (up to 1.6 W per facet from a 40-stripe array) and the high overall conversion efficiency (up to 36 %) considerably exceed the performance of single stripe lasers. 12-stripe arrays delivering some hundred milliwatts at an emission wavelength ranging from 805 to 880 nm have been processed on a larger scale. They show very reproducible laser characteristics with a standard deviation of less than 5 % both for threshold current and differential efficiency across a 20 mm MO VPE wafer.

Abstract: The paper gives the results of an experimental and theoretical investigation of GaAlAs/GaAs oxidestripe lasers under high-pulsed-power conditions. At a pulsewidth of 18 ns the best samples delivered optical powers of more than 2 W per facet. The kink-free light/current characteristics, the well-behaved far-fields and the very-small-amplitude jitter in the optical pulse indicate a stable operation in the fundamental lateral mode up to the maximum power level. The maximum available power is found to vary less than the inverse square root of the pulse width between 20 ns and 1 ?s. Catastrophic failure of these lasers is always accompanied with damage to one or both of the Al2O3-coated mirrors. The experimental power-law dependence and the distinct levels of power limit for lasers with differently prepared mirrors can be explained by a model which assumes a heat source in the mirror plane originating from nonradiative carrier recombination at the surface.

Abstract: The excellent quality and uniformity of MO VPE-grown (Al, Ga)As heterostructures for laser diodes is demonstrated by results on narrow oxide stripe lasers (λ=870 nm). The high uniformity over the substrate area is characterized by a low scatter of significant laser parameters. The low interface roughness in MO VPE heterostructure layers facilitates good laser performance up to high pulsed power levels. The characteristic temperature ranges from 180 to 220 K which allows CW lasing operation at heat sink temperatures of up to more than 100°C even for junction-up mounted lasers. Degradation rates as low as 1×10-5 h -1 at 100°C and 5 mW optical output/facet have been measured over operating times exceeding now 16000 h.

Abstract: The neutron flux of the n TOF facility at CERN was measured, after installation of the new spallation target, with four different systems based on three neutron-converting reactions, which represent accepted cross sections standards in different energy regions. A careful comparison and combination of the different measurements allowed us to reach an unprecedented accuracy on the energy dependence of the neutron flux in the very wide range (thermal to 1 GeV) that characterizes the n TOF neutron beam. This is a pre-requisite for the high accuracy of cross section measurements at n TOF. An unexpected anomaly in the neutron-induced fission cross section of 235U is observed in the energy region between 10 and 30 keV, hinting at a possible overestimation of this important cross section, well above currently assigned uncertainties. © Società Italiana di Fisica / Springer-Verlag 2013.

Abstract: At the n\_TOF experiment at CERN a dedicated single-crystal chemical vapor deposition (sCVD) Diamond Mosaic-Detector has been developed for (n,$\alpha$) cross-section measurements. The detector, characterized by an excellent time and energy resolution, consists of an array of 9 sCVD diamond diodes. The detector has been characterized and a cross-section measurement has been performed for the $^{59}$Ni(n,$\alpha$)$^{56}$Fe reaction in 2012. Abstract: The n_TOF Collaboration has built and commissioned a high-performance detector for (n, gamma) measurements called the Total Absorption Calorimeter (TAC). The TAC was especially designed for measuring neutron capture cross-sections of low-mass and/or radioactive samples with the accuracy required for nuclear technology and stellar nucleosynthesis. We present a detailed description of the TAC and discuss its overall performance in terms of energy and time resolution, background discrimination, detection efficiency and neutron sensitivity. 33. Randomized controlled evaluation of the psychophysiological effects of social support stress management in healthy women.

Abstract: Considering the high and increasing prevalence of stress, approaches to mitigate stress-related biological processes become a matter of public health. Since supportive social interactions contribute substantially to mental and physical health, we set out to develop a social support stress management intervention and examined its effects on psychophysiological stress responses as well as self-reported stress in healthy women. In a parallel-group randomized controlled trial, registered in the DSRK (DRKS00017427), 53 healthy women were randomly assigned to a social support stress management or a waitlist control condition. All participants underwent a standardized psychosocial stress test where physiological and emotional stress responses were assessed by repeated measurements of cortisol, heart rate, heart rate variability and state anxiety. Also, all participants completed self-report questionnaires of perceived stress and social support at pre-intervention, post-intervention and follow-up four weeks later. Participants in the social support stress management showed a significantly attenuated integrated state anxiety response in comparison to those in the control condition, but conditions did not differ in any of the assessed physiological stress responses. The intervention significantly reduced perceived stress in comparison to the control condition, but perceived stress levels returned to baseline at follow-up. Our results indicated that the intervention had no effect on physiological responses to acute psychosocial stress, even though anxiety responses to stress were attenuated. Abstract: Selective export of proteins from the endoplasmic reticulum (ER) requires transport signals that have not been fully characterized. Here, we provide the first complete map of ER export determinants of a type I membrane protein, ERGIC-53, that cycles in the early secretory pathway. ER export requires a phenylalanine motif at the C-terminus, known to mediate coat protein II (COPII) interaction, that is assisted by a glutamine in the cytoplasmic domain. Disulfide bond-stabilized oligomerization is also required. Efficient hexamerization depends on the presence of a polar and two aromatic residues in the transmembrane domain (TMD). Oligomerization becomes independent on disulfide bonds when TMD hydrophobicity is increased. ER export is also influenced by TMD length, 21 amino acids being most efficient. When transferred to a signal-less construct, the established targeting motifs reconstitute full transport activity. The results suggest an ER-export mechanism in which transmembrane and luminal determinants mediate oligomerization required for efficient recruitment of ERGIC-53 into budding vesicles via the C-terminal COPII-binding phenylalanine motif.

Abstract: Export of membrane proteins from the ER is believed to be selective and require transport signals, but the identity of such signals has remained elusive. The recycling type I membrane protein ERGIC-53 carries a C-terminal diphenylalanine motif that is required for efficient ER export. Here we show that this motif can be functionally substituted by a single phenylalanine or tyrosine at position -2, two leucines or isoleucines at position -1 and -2 or a single valine at position -1. These motifs are common among mammalian type I membrane proteins. A single C-terminal valine, but none of the other motifs, accelerates transport of inefficiently exported reporter constructs and hence operates as an export signal. The valine signal is position, but not context, dependent. All transport motifs mediate COPII binding in vitro with distinct preferences for the COPII subunits Sec23p, Sec24Bp, Sec24Cp and p125. These results suggest that cytoplasmic C-terminal amino-acid motifs, either alone or in conjunction with other transport determinants, accelerate ER export of numerous type I and probably polytopic membrane proteins by mediating interaction with COPII coat components.

Abstract: The ER-Golgi intermediate compartment (ERGIC) marker ERGIC-53 is a mannose-specific membrane lectin operating as a cargo receptor for the transport of glycoproteins from the ER to the ERGIC. Lack of functional ERGIC-53 leads to a selective defect in secretion of glycoproteins in cultured cells and to hemophilia in humans. Beyond its interest as a transport receptor, ERGIC-53 is an attractive probe for studying numerous aspects of protein trafficking in the secretory pathway, including traffic routes, mechanisms of anterograde and retrograde traffic, retention of proteins in the ER, and the function of the ERGIC. Understanding these fundamental processes of cell biology will be crucial for the elucidation and treatment of many inherited and acquired diseases, such as cystic fibrosis, Alzheimer's disease and viral infections.

Abstract: Soluble secretory proteins are transported from the endoplasmic reticulum (ER) to the ER-Golgi intermediate compartment (ERGIC) in vesicles coated with COP-II coat proteins. The sorting of secretory cargo into these vesicles is thought to involve transmembrane cargo-receptor proteins. Here we show that a cathepsin-Z-related glycoprotein binds to the recycling, mannose-specific membrane lectin ERGIC-53. Binding occurs in the ER, is carbohydrate- and calcium-ion-dependent and is affected by untrimmed glucose residues. Binding does not, however, require oligomerization of ERGIC-53, although oligomerization is required for exit of ERGIC-53 from the ER. Dissociation of ERGIC-53 occurs in the ERGIC and is delayed if ERGIC-53 is mislocalized to the ER. These results strongly indicate that ERGIC-53 may function as a receptor facilitating ER-to-ERGIC transport of soluble glycoprotein cargo.

Abstract: Dilysine signals confer localization of type I membrane proteins to the endoplasmic reticulum (ER). According to the prevailing model these signals target proteins to the ER by COP I-mediated retrieval from post-ER compartments, whereas the actual retention mechanism in the ER is unknown. We expressed chimeric membrane proteins with a C-terminal -Lys-Lys-Ala-Ala (KKAA) or -Lys-Lys-Phe-Phe (KKFF) dilysine signal in Lec-1 cells. Unlike KKFF constructs, which had access to post-ER compartments, the KKAA chimeras were localized to the ER by confocal microscopy and were neither processed by cis-Golgi-specific enzymes in vivo nor included into ER-derived transport vesicles in an in vitro budding assay, suggesting that KKAA-bearing proteins are permanently retained in the ER. The ER localization was nonsaturable and exclusively mediated by the dilysine signal because mutating the lysines to alanines led to cell surface expression of the chimeras. Although the KKAA signal avidly binds COP I in vitro, the ER retention by this signal does not depend on intact COP I in vivo because it was not affected in an epsilon-COP-deficient cell line. We propose that dilysine ER targeting signals can mediate ER retention in addition to retrieval.

Abstract: The positioning and dynamics of organelles in eukaryotic cells critically depend on membrane-cytoskeleton interactions. Motor proteins play an important role in the directed movement of organelle membranes along microtubules, but the basic mechanism by which membranes stably interact with the microtubule cytoskeleton is largely unknown. Here we report that p63, an integral membrane protein of the reticular subdomain of the rough endoplasmic reticulum (ER), binds microtubules in vivo and in vitro. Overexpression of p63 in cell culture led to a striking rearrangement of the ER and to concomitant bundling of microtubules along the altered ER. Mutational analysis of the cytoplasmic domain of p63 revealed two determinants responsible for these changes: an ER rearrangement determinant near the N-terminus and a central microtubule-binding region. The two determinants function independently of one another as indicated by deletion experiments. A peptide corresponding to the cytoplasmic tail of p63 promoted microtubule polymerization in vitro. p63 is the first identified integral membrane protein that can link a membrane organelle directly to microtubules. By doing so, it may contribute to the positioning of the ER along microtubules.

Abstract: ERGIC-53, a homo-oligomeric recycling protein associated with the ER-Golgi intermediate compartment (ERGIC), has properties of a mannose-selective lectin in vitro, suggesting that it may function as a transport receptor for glycoproteins in the early secretory pathway. To investigate if ERGIC-53 is involved in glycoprotein secretion, a mutant form of this protein was generated that is incapable of leaving the ER. If expressed in HeLa cells in a tetracycline-inducible manner, this mutant accumulated in the ER and retained the endogenous ERGIC-53 in this compartment, thus preventing its recycling. Mistargeting of ERGIC-53 to the ER did not alter the gross morphology of the early secretory pathway, including the distribution of beta'-COP. However, it impaired the secretion of one major glycoprotein, identified as the precursor of the lysosomal enzyme cathepsin C, while overexpression of wild-type ERGIC-53 had no effect on glycoprotein secretion. Transport of two other lysosomal enzymes and three post-Golgi membrane glycoproteins was unaffected by inactivating the recycling of ERGIC-53. The results suggest that the recycling of ERGIC-53 is required for efficient intracellular transport of a small subset of glycoproteins, but it does not appear to be essential for the majority of glycoproteins.

Abstract: Polarized expression of most epithelial plasma membrane proteins is achieved by selective transport from the Golgi apparatus or from endosomes to a specific cell surface domain. In Madin-Darby canine kidney (MDCK) cells, basolateral sorting generally depends on distinct cytoplasmic targeting determinants. Inactivation of these signals often resulted in apical expression, suggesting that apical transport of transmembrane proteins occurs either by default or is mediated by widely distributed characteristics of membrane glycoproteins. We tested the hypothesis of N-linked carbohydrates acting as apical targeting signals using three different membrane proteins. The first two are normally not glycosylated and the third one is a glycoprotein. In all three cases, N-linked carbohydrates were clearly able to mediate apical targeting and transport. Cell surface transport of proteins containing cytoplasmic basolateral targeting determinants was not significantly affected by N-linked sugars. In the absence of glycosylation and a basolateral sorting signal, the reporter proteins accumulated in the Golgi complex of MDCK as well as CHO cells, indicating that efficient transport from the Golgi apparatus to the cell surface is signal-mediated in polarized and non-polarized cells.

Abstract: Further investigation of the targeting of the intracellular membrane lectin endoplasmic reticulum (ER)-Golgi intermediate compartment-53 (ERGIC-53) by site-directed mutagenesis revealed that its lumenal and transmembrane domains together confer ER retention. In addition we show that the cytoplasmic domain is required for exit from the ER indicating that ERGIC-53 carries an ER-exit determinant. Two phenylalanines at the C terminus are essential for ER-exit. Thus, ERGIC-53 contains determinants for ER retention as well as anterograde transport which, in conjunction with a dilysine ER retrieval signal, control the continuous recycling of ERGIC-53 in the early secretory pathway. In vitro binding studies revealed a specific phenylalanine-dependent interaction between an ERGIC-53 cytosolic tail peptide and the COPII coat component Sec23p. These results suggest that the ER-exit of ERGIC-53 is mediated by direct interaction of its cytosolic tail with the Sec23p.Sec24p complex of COPII and that protein sorting at the level of the ER occurs by a mechanism similar to receptor-mediated endocytosis or Golgi to ER retrograde transport.

Abstract: Membrane proteins often contain a sorting signal in their cytoplasmic tail that promotes their clustering into coated vesicles at a specific cellular site. ERGIC-53 contains a cytoplasmic ER-retrieval signal, KKFF. However, overexpressed ERGIC-53 is transported to the cell surface and rapidly endocytosed. Here we report that ERGIC-53 carries a previously undescribed endocytosis signal. Surprisingly, the signal was KKFF and like the ER-retrieval signal required a C-terminal position. In fact, the minimal consensus sequence determined by substitutional mutagenesis (K-K/R-F/Y-F/Y) was related to the ER-retrieval consensus (K-K-X-X). Furthermore, we provide evidence that internalization of VIP36, a protein that cycles between plasma membrane and Golgi, is mediated by a signal at its C-terminus that matches the internalization consensus sequence. The relatedness of the two signals suggests that coatomer-mediated retrieval of proteins may be mechanistically more related to clathrin-dependent sorting than previously anticipated.

Abstract: ERGIC-53 (former designation, p53) is a 53-kDa nonglycosylated, dimeric, and hexameric type I membrane protein that has been established as a marker protein for a tubulovesicular intermediate compartment in which protein transport from the endoplasmic reticulum to the Golgi apparatus is blocked at 15 degrees C. Although ERGIC-53 is not a resident protein of the rough endoplasmic reticulum its cDNA sequence carries a double lysine endoplasmic reticulum retention motif at the cytoplasmically exposed COOH terminus. Here we report that overexpression of ERGIC-53 in COS cells saturates its intracellular retention system leading to the appearance of ERGIC-53 at the cell surface. Cell surface ERGIC-53 is efficiently endocytosed by a mechanism that is disturbed when the two critical lysines of the endoplasmic reticulum retention motif are replaced by serines. The results suggest a mechanistic similarity of pre-Golgi retention by the double lysine motif and lysine-based endocytosis.
