Cytochrome P450 (CYP) 2A enzymes are involved in the metabolism of numerous drugs and hormones and activate different carcinogens. Human CYP2A6, mouse CYP2A5 and rat CYP2A3 are orthologous enzymes that present high similarity in their amino acid sequence and share substrate specificities. However, different from the human and mouse enzyme, CYP2A3 is not expressed in the rat liver. There are limited data about expression of CYP2A3 in extrahepatic tissues and its regulation by typical CYP inducers. Therefore, the objective of the present study was to analyze CYP2A3 mRNA expression in different rat tissues by RT-PCR, and to study the influence of 3-methylcholanthrene, pyrazole and ß-ionone treatment on its expression. Male Wistar rats were divided into four groups of 5 rats each, and were treated ip for 4 days with 3-methylcholanthrene (25 mg/kg body weight), pyrazole (150 mg/kg body weight), ß-ionone (1 g/kg body weight), or vehicle. Total RNA was extracted from tissues and CYP2A3 mRNA levels were analyzed by semiquantitative RT-PCR. CYP2A3 mRNA was constitutively expressed in the esophagus, lung and nasal epithelium, but not along the intestine, liver, or kidney. CYP2A3 mRNA levels were increased in the esophagus by treatment with 3-methylcholanthrene and pyrazole (17- and 7-fold, respectively), in lung by pyrazole and ß-ionone (3- and 4-fold, respectively, although not statistically significant), in the distal part of the intestine and kidney by 3-methylcholanthrene and pyrazole, and in the proximal part of the intestine by pyrazole. CYP2A3 mRNA was not induced in nasal epithelium, liver or in the middle part of the intestine. These data show that, in the rat, CYP2A3 is constitutively expressed in several extrahepatic tissues and its regulation occurs through a complex mechanism that is essentially tissue specific.

Cytochrome P450 monooxygenases of the CYP2A subfamily play important roles in xenobiotic disposition in the liver and in metabolic activation in extrahepatic tissues. Many of the CYP2A transcripts and enzymes are inducible by xenobiotic compounds, and the expression of at least some of the CYP2A genes is influenced by physiological status, such as circadian rhythm, and pathological conditions, such as inflammation, microbial infection, and tumorigenesis. Variability in the expression of the CYP2A genes, which differs by species, animal strain, gender, and organ, may alter the risks of chemical toxicity for numerous compounds that are CYP2A substrates. The mechanistic bases of these variabilities are generally not well understood. However, recent studies have yielded interesting findings in several areas, such as the role of nuclear factor 1 in the tissue-selective expression of CYP2A genes in the olfactory mucosa (OM); the roles of constitutive androstane receptor, pregnane X receptor (PXR), and possibly, peroxisome proliferator-activated receptors in transcriptional regulation of the Cyp2a5 gene; and the involvement of heterogeneous nuclear ribonucleoprotein A1 in pyrazole-induced stabilization of CYP2A5 mRNA. The aims of this minireview are to summarize current knowledge of the regulation of the CYP2A genes in rodents and humans, and to stimulate further mechanistic studies that will ultimately improve our ability to determine, and to understand, these variabilities in humans. The effect of chronic ethanol feeding on the status of α- and γ- tocopherol in plasma, liver, lung, and testes of Sprague-Dawley rats was characterized. Rats were pair-fed liquid diets containing 36% of total calories either as ethanol or isocaloric carbohydrates. After 3 weeks, ethanol ingestion resulted in a significant ( p≤ 0.05) increase in liver weight and induced fatty liver without affecting total body weight. Ethanol feeding did not affect the plasma concentration of α-tocopherol but doubled that of γ-tocopherol. When expressed per milligram of tissue, liver α-tocopherol did not vary with ethanol ingestion, whereas γ-tocopherol concentration increased 2.5 times that of control animals. However, the concentration of α-tocopherol expressed per milligram of total lipids was significantly ( p≤ 0.01) decreased in the liver with ethanol feeding. In contrast to the liver, ethanol feeding significantly increased α- and γ-tocopherol levels per milligram of total lipids in the testes. The concentration of γ- tocopherol (but not α-tocopherol) per milligram of lung tissue and per total lung was significantly ( p≤ 0.05) increased with ethanol feeding. These data indicate that chronic ethanol ingestion significantly alters the distribution of α-tocopherol and γ-tocopherol in hepatic and extrahepatic tissues of the rat. [ABSTRACT FROM AUTHOR]

Lipogenic activation was studied in mice that had been restricted to a single large meal once a day rather than being allowed to eat at frequent intervals throughout the night. Mice were injected intravenously with [U-14C]glucose, and the flux of glucose C to total lipid fatty acids (TLFA) and to all “end products” was estimated from serial plasma glucose specific activities and measurements of incorporation of 14C into TLFA of hepatic and extrahepatic tissues. Tracer studies were carried out in mice fasted for 1 day and at various times after the mice ate one or two small test meals or a single large test meal. Test meals consisted of a fat-free, 58% glucose diet. The flux of glucose C to TLFA increased by an order of magnitude within an hour after mice nibbled a test meal for several minutes. After ingestion of two small test meals or a single large test meal, the flux of glucose C to TLFA increased from a fasting rate of 0.5 to 35 and 87 μg of glucose C/min/20 g body wt, respectively. Although trained meal eaters are thought to have abnormally increased lipogenesis, their lipogenic response to a single test meal was the same as that previously reported for untrained nibbling mice. Most of the newly synthesized fatty acids were found in extrahepatic tissues. Ingestion of a first test meal completely prevented the expected hyperglycemic response following ingestion of a second test meal even though the latter contained over 10 times more glucose than that in the total body glucose pool.
