Ganoderma lucidum, known as the "mushroom of immortality," is a white rot fungus renowned for its medicinal properties, attributed to its bioactive compounds. Although species with similar morphological traits to G. lucidum are found across the globe, precise identification is made possible through DNA barcoding and molecular phylogenetic analysis. Global cultivation and wild harvesting of G. lucidum are both done in response to the growing market needs. Artificial cultivation is typically performed on sawdust, but other woody substrates and the wood log method are also employed. This cultivation leverages the fungus's ecological role in converting industrial and agricultural solid wastes into biomass, thereby producing functional food and potential pharmaceutical sources. The review consolidates research on various aspects of, including cultivation methods (sawdust, agricultural waste, wood logs, and submerged fermentation), and the current global market conditions. Agro-residues, including banana peel, pineapple peel, mango peel, apple peel, and coconut husk, were utilized to produce extracellular polysaccharide (EPS) via solid state fermentation(SSF) by the fungus, Trichoderma hamatum. EPS production was highest in pineapple peel (84.2 ± 0.4 mg/g), followed by mango peel (70.3 ± 0.41 mg/g) (p<0.01). The SSF bioprocess was optimized via a two-level full factorial design and response surface methodology. The effects of five selected variables on EPS biosynthesis, namely, the concentrations of glucose (10 to 30%), ammonium sulphate (0.1 to 1%), yeast extract (0.5 to 2%), MgSO4 (0.01 to 0.1%), and medium pH (4.5 to 6.5), were analyzed via a full factorial design (FFD). The EPS production ranged widely from 15.3 to 576.2 mg/g substrate. Three significant variables affecting EPS production were assessed in central composite design (CCD) to optimize concentrations of MgSO4, and glucose, and the pH. The designed CCD model was fitted to the quadratic model and was significant (p<0.0001). For 50 μL of EPS, the scavenging ratio was 43.4 ± 4.1% at a concentration of 200 μL (78.5 ± 6.9 μL) (p<0.01). The extracted EPS exhibited an alpha-glucosidase inhibitory effect (p<0.001). Solid-state fermentation allows the utilization of low-cost biomass for EPS production and the application of T. hamatum EPS as a natural antioxidant and α-glucosidase inhibitor. Interest in natural extracts for managing oral biofilms is increasing, with black cumin seed oil (BCSO) demonstrating efficacy against Streptococcus mutans. The effectiveness of antibacterial agents should be evaluated using multi-species oral biofilm models that closely mimic actual conditions. This study aimed to compare the antibacterial effects of BCSO and chlorhexidine gluconate (CHX) on oral microcosm biofilms. Biofilms using human saliva as the inoculum were cultured for 2 days and subsequently treated with 0.5% dimethyl sulfoxide, 0.5% BCSO, or 0.12% CHX once daily for 6 days. Following treatment, the red fluorescence intensity (RatioR/G) of the oral biofilm; biomass, including extracellular polymeric substance (EPS) levels and live bacteria counts; and colony-forming units (CFUs) of aciduric bacteria were evaluated. RatioR/G after BCSO treatment (1.26 ± 0.03) was not significantly different from that after CHX treatment (p = 0.552). The EPS levels were also not significantly different between the two groups (p = 0.743). The live bacteria count was 0.55 times lower in the BCSO-treated group than in the CHX-treated group (p = 0.018). No significant between-group difference was observed in the CFUs of aciduric bacteria (p = 0.935). Abstract Cyanobacteria possess special defense mechanisms to protect themselves against ultraviolet (UV) radiation. This study combines experimental and computational methods to identify the role of protective strategies in Nostoc species against UV-C radiation. To achieve this goal, various species of the genus Nostoc from diverse natural habitats in Iran were exposed to artificial UV-C radiation. The results indicated that UV-C treatment significantly reduced the photosynthetic pigments while simultaneously increasing the activity of antioxidant enzymes. Notably, N. sphaericum ISB97 and Nostoc sp. ISB99, the brown Nostoc species isolated from habitats with high solar radiations, exhibited greater resistance compared to the green-colored species. Additionally, an increase in scytonemin content occurred with a high expression of key genes associated with its synthesis (scyF and scyD) during the later stages of UV-C exposure in these species. The molecular docking of scytonemin with lipopolysaccharides of the cyanobacteria that mainly cover the extracellular matrix revealed the top/side positioning of scytonemin on the glycans of these lipopolysaccharides to form a UV-protective shield. These findings pave the way for exploring the molecular effects of scytonemin in forming the UV protection shield in cyanobacteria, an aspect that has been ambiguous until now.

This study aimed to enhance extracellular polysaccharide (EPS) production in Cordyceps militaris by constructing a quorum sensing (QS) system to regulate the expression of biosynthetic enzyme genes, including phosphoglucomutase, hexokinase, phosphomannomutase, polysaccharide synthase, and UDP‐glucose 4‐epimerase genes. The study found higher EPS concentrations in seven recombinant strains compared to the wild‐type C. militaris, indicating that the overexpression of key enzyme genes increased EPS production. Among them, the CM‐pgm‐2 strain exhibited the highest EPS production, reaching a concentration of 3.82 ± 0.26 g/L, which was 1.52 times higher than the amount produced by the wild C. militaris strain. Additionally, the regulatory effects of aromatic amino acids on the QS system of the CM‐pgm‐2 strain were investigated. Under the influence of 45 mg/L tryptophan, the EPS production in CM‐pgm‐2 reached 4.75 ± 0.20 g/L, representing a 1.90‐fold increase compared to wild C. militaris strains. This study provided an effective method for the large‐scale production of EPSs in C. militaris, and opened up new avenues for research into fungal QS mechanisms. [ABSTRACT FROM AUTHOR]

Cordyceps militaris is a valuable medicinal fungus which has been widely used as a traditional medicine in East Asia. Compared to the well-known medicinal fungus C. sinensis, C. militaris can produce similar fermented metabolites with various biological activities, but it requires a shorter culture time and simpler culture conditions, and therefore, it has attracted increasing attention in recent years. The purpose of this review was to organize the current studies regarding metabolite production from C. militaris relative to their biological functions. We combined findings of metabolite production to correlate with different fermentation modes to obtain a full view of production processes used to yield the product. While research on C. militaris fermentation is not uncommon to date, its high value still highlights the importance of developing more modern fermentation processes for industrial production. A novel, gelatinous, colony-forming, rod-shaped bacterial strain, designated IK01T was isolated from biofilms formed on the membrane surface of a sewage-treating membrane bioreactor (MBR). Strain IK01T produced gelatinous and almost transparent colonies at lower medium concentrations. Fourier transform infrared analysis of the gelatinous colony matrix showed that the matrix could be a biofilm substance. This suggests that strain IK01T is a fouling-causing bacteria in the MBR. Furthermore, 16S rRNA gene sequence analysis showed that strain IK01T was phylogenetically placed in the genus Novosphingobium. The average nucleotide identity values for IK01T and the other 50 species of the genus Novosphingobium ranged from 78.5 to 83.9 %. Correspondingly, the estimated digital DNA-DNA hybridization values ranged from 20.8 to 24.4 %. The genomic DNA G + C content was 66.0 %. The predominant fatty acids were summed feature 8 (C18:1 ω7c and/or C18:1 ω6c), summed feature 3 (C16:1 ω7c and/or C16:1 ω6c), and C14:0 2-OH. A polar lipid profile revealed phosphatidylethanolamine, two unidentified phospholipids, and three aminoglycophospholipids as major compounds. The major respiratory quinone was ubiquinone Q-10. Genotypic, chemotaxonomic, and phenotypic analyses characterized the newly identified strain IK01T, as a novel species of the genus Novosphingobium, for which we propose the name Novosphingobium pituita sp. nov. The type strain is IK01T (NBRC 116408T = DSM 116658T).

The newly identified Sanghuangporus alpinus species of the Sanghuang mushroom genus has been found to possess significant medical benefits. However, the current artificial cultivation technology has not reached the requisite maturity. The response-surface methodology (RSM) was used to optimize the Sanghuangporus alpinus culture medium formulation and evaluate the functional activity of S. alpinus exopolysaccharides. First, a single-factor experiment was conducted to screen for optimal carbon and nitrogen sources for S. alpinus. Then, using Box–Behnken's central composite design, a response-surface experiment was conducted to determine optimal culture parameters. Finally, the rationality of those parameters was assessed in a shaking flask experiment. The optimal culture parameters, determined through regression analysis, were 20.20 ± 0.17 g/L fructose (carbon source), 7.29 ± 0.10 g/L yeast extract (nitrogen source), and 0.99 ± 0.01 g/L dandelion. With optimization, the S. alpinus yield increased to 12.79 ± 1.41 g/L, twice that obtained from the initial culture medium. The S. alpinus exopolysaccharide exhibited an excellent antioxidant capacity, with the strongest scavenging effect noted on ABTS free radicals (lowest half-inhibitory concentration: 0.039 mg/mL). Additionally, this exopolysaccharide effectively inhibited various cancer cells, exhibiting the strongest activity against human glioma cells U251 (half-inhibitory concentration: 0.91 mg/mL). The RSM used to optimize the fermentation culture parameters of S. alpinus significantly increased the mycelial biomass. The improvement of Sanghuangporus alpinus yield through liquid fermentation and optimizing the fermentation medium could fill the existing gap in the cultivation of Sanghuangporus alpinus, as well as provide valuable data for the large-scale production of S. alpinus. Porphyridium aerugineum is a unicellular freshwater red microalga that synthesizes and secretes into the culture medium an extracellular polysaccharide (EPS). In this study, algal growth and polysaccharide production, as well as the antioxidant capacity and antitumor effect of Porphyridium aerugineum EPS (PaEPS), were investigated. Cultivation of the microalgae was carried out in a photobioreactor under controlled conditions. Algal growth and the amount of EPS were monitored daily. The accumulated polysaccharide was extracted and lyophilized. At the end of cultivation, the concentration of microalgal biomass and PaEPS reached 3.3 and 1.2 g L−1, respectively. To examine the antioxidant capacity of PaEPS, FRAP and ABTS assays were performed. The cytotoxic activity of PaEPS was evaluated on the tumor cell lines MCF-7 (breast cancer) and HeLa (cervical adenocarcinoma) and on BJ (a non-tumor human skin fibroblast cell line), using MTT assay. The results obtained indicated that P. aerugineum polysaccharide exhibited a high ABTS radical-scavenging activity reaching up to 55%. The cytotoxic effect was best expressed in MCF-7 cells treated for 72 h with 1000 µg/mL PaEPS, where tumor cell proliferation was inhibited by more than 70%. Importantly, the PaEPS treatments did not significantly affect the viability of BJ cells. These findings promote the biotechnological production of P. aerugineum extracellular polysaccharide and reveal its potential as an anticancer and antioxidant agent for future applications. As biofilms mature, biomass and extracellular polysaccharide (EPS) content increases, enhancing pathogenicity. Therefore, this study aimed to evaluate the antibacterial efficacy of cold atmospheric plasma (CAP) against oral microcosm biofilms and the influence of biofilm maturity on treatment. Oral microcosm biofilms were cultured on hydroxyapatite disks for 2 and 6 days. Based on the treatment and biofilm maturity, these were subsequently allocated into six groups (N = 19 each): Groups 1 and 2 were incubated with distilled water for 1 min; Groups 3 and 4 were treated with CAP for 2 min, and Groups 5 and 6 were treated with 0.12% chlorhexidine gluconate for 1 min. Groups 1, 3, and 5 represent 2-day biofilms, and Groups 2, 4, and 6 represent 6-day biofilms. Treatments were repeated daily for 5 days. Antibacterial efficacy was analyzed by measuring oral biofilms' red fluorescence intensity (RatioR/G) and quantifying EPS content and bacterial viability. The RatioR/G was 1.089-fold and 1.104-fold higher in Groups 4 and 6 than in Groups 3 and 5 following antibacterial treatment, respectively (p < 0.001). EPS content increased by 1.71-fold in Group 6 than in Group 5 (p < 0.001). Bacterial survival rate was the lowest in Group 3 (p = 0.005). These findings underscore the relevance of CAP treatment in maintaining antibacterial efficacy regardless of the biofilm development stage, highlighting its potential utility in oral care. Citrocin is an anti-microbial peptide that holds great potential in animal feed. This study evaluates the anti-microbial and anti-biofilm properties of Citrocin and explores the mechanism of action of Citrocin on the biofilm of P. aeruginosa. The results showed that Citrocin had a significant inhibitory effect on the growth of P. aeruginosa with a minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of 0.3 mg/mL. All five concentrations (1/4MIC, 1/2MIC, MIC, 2MIC, and 4MIC) of Citrocin inhibited P. aeruginosa biofilm formation. Citrocin at the MIC, 2MIC and 4MIC removed 42.7%, 76.0% and 83.2% of mature biofilms, respectively, and suppressed the swarming motility, biofilm metabolic activity and extracellular polysaccharide production of P. aeruginosa. Metabolomics analysis indicated that 0.3 mg/mL of Citrocin up- regulated 26 and down-regulated 83 metabolites, mainly comprising amino acids, fatty acids, organic acids and sugars. Glucose and amino acid metabolic pathways, including starch and sucrose metabolism as well as arginine and proline metabolism, were highly enriched by Citrocin. In summary, our research reveals the anti-biofilm mechanism of Citrocin at the metabolic level, which provides theoretical support for the development of novel anti-biofilm strategies for combatting P. aeruginosa. Extracellular polysaccharide producers from Kursunlu (Manisa) thermal spring were identified and a total of nine isolates were selected. One isolate was selected based on its strong mucoid structure and identified as Anoxybacillus flavithermus (AAG61) by MALDI-TOF MS. A. flavithermus AAG-61 had multiple antibiotic resistance to both Sulfamethoxazole, and Ofloxacin. The polysaccharide produced by A. flavithermus AAG-61 was purified and characterized. It was an anionic heteropolysaccharide containing rhamnose, mannose, arabinose, and glucose. The polysaccharide was water-soluble and had a concentration of 3.1 g/L carbohydrate and 0.11 g/L protein. The EPS consisted of 32% carbon, 7% nitrogen, 5% hydrogen, and traces of sulfur (0.6%). The presence of protein and sulfur in the polysaccharide was confirmed by FT-IR. Zeta potential, mobility, and conductivity values were -8.3 (± 0.5) mV, -0.65 (± 0.04) µmcm/Vs, and 1.57 (± 0.08) mS/cm. The crystallinity pattern was also determined by XRD analysis. No effect on the viability of the human fibroblast cell line was observed.

The formation of soil in karst ecosystem has always been a scientific problem of great concern to human beings. Algae can grow on the exposed and nonnutrition carbonate surface, inducing and accelerating weathering of rock substrates, thus promoting soil formation. Yet the actual contribution of algae to solutional weathering intensity remains unclear. In this study, we performed weathering simulation experiment on two algae species (Klebsormidium dissectum (F.Gay) H.Ettl & G.Gärtner and Chlorella vulgaris Beijerinck), which were screened from carbonated rock surfaces from a typical karst region in South China. The results showed: (1) both algae have solutional weathering effect on carbonate rock, (2) there is no difference of solutional intensity observed, yet the solutional modes are different, suggesting different ecological adaptative strategies, (3) algae on carbonate rocks have higher carbonic anhydrase activity (CAA) and secrete more extracellular polysaccharide (EPS), accelerating rock weathering. (4) The absolute dissolution amount of carbonate rock with algae participation is 3 times of that of without algae. These results indicate the significant impact of terrestrial algae on carbonate rock solutional weathering and provides quantitative evidence that terrestrial algae are pioneer species. Seven photosynthethic microbiomes were collected from field environmental samples to test their potential in polyhydroxybutyrate (PHB) and exopolysaccharides (EPS) production, both alternatives to chemical-based polymers. Microscope observations together with microbial sequence analysis revealed the microbiome enrichment in cyanobacteria after culture growth under phosphorus limitation. PHB and EPS production were studied under three culture factors (phototrophy, mixotrophy and heterotrophy) by evaluating and optimizing the effect of three parameters (organic and inorganic carbon and days under light:dark cycles) by Box-Behnken design. Results showed that optimal conditions for both biopolymers synthesis were microbiome-dependent; however, the addition of organic carbon boosted PHB production in all the tested microbiomes, producing up to 14 % dcw PHB with the addition of 1.2 g acetate·L−1 and seven days under light:dark photoperiods. The highest EPS production was 59 mg·L−1 with the addition of 1.2 g acetate·L−1 and four days under light:dark photoperiods. The methodology used is suitable for enriching microbiomes in cyanobacteria, and for testing the best conditions for bioproduct synthesis for further scale up. ● Growth under phosphorus limitation leaded to microbiomes rich in cyanobacteria. ● Up to 14 % dcw polyhydroxybutyrate (PHB) produced under photoheterotrophic regime. ● 59 mg·L−1 exopolysaccharides (EPS) obtained with acetate and four days under light. ● PHB and EPS production showed to be microbiome-dependent. ● Acetate boosted PHB synthesis in all microbiomes tested. Microcystis possesses the capacity to form colonies and blooms in lakes and reservoirs worldwide, causing significant ecological challenges in aquatic ecosystems. However, little is known about the determining factors of physicochemical surface properties that govern the competitive advantage of Microcystis. Here, The physico-chemical surface properties of Microcystis wesenbergii and Microcystis aeruginosa, including specific surface area (SSA), hydrophobicity, zeta potential, and functional groups were investigated. Additionally, the extracellular polysaccharide (EPS) were analyzed. Laboratory-cultured Microcystis exhibited hydrophilic, a negative zeta potential and negatively charged. Furthermore, no significant relationship was shown between these properties and the cultivation stage. Microcystis wesenbergii exhibited low free energy of cohesion, high surface free energy, high growth rate, and high EPS content during the logarithmic phase. On the other hand, M. aeruginosa displayed lower free energy of cohesion, high surface free energy, high EPS content, and high growth rate during the stationary phase. These characteristics contribute to their respective competitive advantage. Furthermore, the relationship between EPS and surface properties was investigated. The polysaccharide component of EPS primarily influenced the SSA and total surface energy of Microcystis. Likewise, the protein component of EPS influenced hydrophobicity and surface tension. The polysaccharide composition, including glucuronic acid, xylose, and fructose, mainly influenced surface properties. Additionally, hydrophilic groups such as O-H and P-O-P played a crucial role in determining hydrophobicity in Microcystis. This study elucidates that EPS influenced the SSA, hydrophobicity, and surface free energy of Microcystis cells, which in turn impact the formation of Microcystis blooms and the collection. The signalling effects of microbial biofilms and extracellular polysaccharides (EPS) determine the stability of microbial communities. As messenger molecules, extracellular vesicles (EVs) mediate the interactions of microbial populations by carrying cargo. Therefore, this study took biofilm and EPS as screening factors, preliminarily explored the basic characteristics of strains with strong biofilm formation ability and high EPS production. The results showed that the EVs concentration of three lactic acid bacteria (KLK29, KLH2 and KEF14) were high. The concentration of their EVs was proportional to the EPS production, indicating that EVs were directly involved in the formation and regulation of EPS, but it did not mean that EVs could directly affect other biological characteristics of the strains. This study offers crucial information about kefir-derived strains and their EVs, and provides a preliminary research basis for EVs-mediated expression of probiotic characteristics of strains in kefir.

To evaluate the immune enhancement effect of the extracellular polysaccharide of Lactobacillus plantarum on oprH recombinant subunit vaccine from Pseudomonas aeruginosa, a recombinant subunit vaccine of oprH (rOprH vaccine) was developed. The EP-rOprH vaccine was prepared with the extracellular polysaccharide of L. plantarum as an adjuvant. Mice were vaccinated with the rOprH and EP-rOprH vaccines, and the outer membrane protein (OMP) and inactivated vaccines were used as controls. The levels of serum antibody, interferon-γ (IFN-γ), interleukin (IL-2), and IL-4 were determined after vaccination. Finally, the protective efficacy of the vaccine was evaluated after challenge with virulent P. aeruginosa. Following vaccination, the serum antibody levels were significantly higher in mice vaccinated with the EP-rOprH vaccine than in those vaccinated with the rOprH vaccine (P<0.05). Moreover, the serum antibody levels detected in the EPrOprH vaccine group were similar to those detected in the OMP vaccine group when P. aeruginosa suspension was used as the coating antigen. However, the levels in the EP-rOprH vaccine group were higher than those in the OMP vaccine and inactivated vaccine groups when the purified rOprH protein was used as the coating antigen (P<0.05). The level of IFN-γ, IL-2, and IL-4 in mice vaccinated with the EP-rOprH vaccine was significantly higher than that in mice vaccinated with the rOprH vaccine (P<0.05) and comparable to that in mice vaccinated with the OMP vaccine. The protective rates were 65%, 80%, 80%, and 95% with the rOprH, EP-rOprH, OMP, and inactivated vaccines, respectively. Thus, the extracellular polysaccharide of L. plantarum significantly enhanced the immune response and protection provided by the recombinant subunit vaccine of oprH. [ABSTRACT FROM AUTHOR]

The biofilm lifestyle is critical for bacterial survival and proliferation in the fluctuating marine environment. Cyclic diguanylate (c-di-GMP) is a key second messenger during bacterial adaptation to various environmental signals, which has been identified as a master regulator of biofilm formation. However, little is known about whether and how c-di-GMP signaling regulates biofilm formation in Vibrio alginolyticus, a globally dominant marine pathogen. Here, a large set of 63 proteins were predicted to participate in c-di-GMP metabolism (biosynthesis or degradation) in a pathogenic V. alginolyticus strain HN08155. Guided by protein homology, conserved domains and gene context information, a representative subset of 22 c-di-GMP metabolic proteins were selected to determine which ones affect biofilm-associated phenotypes. By comparing phenotypic differences between the wild-type and mutants or overexpression strains, we found that 22 c-di-GMP metabolic proteins can separately regulate different phenotypic outputs in V. alginolyticus. The results indicated that overexpression of four c-di-GMP metabolic proteins, including VA0356, VA1591 (CdgM), VA4033 (DgcB) and VA0088, strongly enhanced rugose colony morphotypes and strengthened Congo Red (CR) binding capacity, both of which are indicators of biofilm matrix overproduction. Furthermore, rugose enhanced colonies were accompanied by increased transcript levels of extracellular polysaccharide (EPS) biosynthesis genes and decreased expression of flagellar synthesis genes compared to smooth colonies (WTpBAD control), as demonstrated by overexpression strains WTp4033 and AVA4033p4033. Overall, the high abundance of c-di-GMP metabolic proteins in V. alginolyticus suggests that c-di-GMP signaling and regulatory system could play a key role in its response and adaptation to the ever-changing marine environment. Staphylococcus aureus is a commensal skin bacterium and a causative agent of infectious diseases. Biofilm formation in S. aureus is a mechanism that facilitates the emergence of resistant strains. This study proposes a mechanism for the regulation of biofilm formation in S. aureus through strain-specific physiological changes induced by the plant steroid diosgenin. A comparison of diosgenin-induced changes in the expression of regulatory genes associated with physiological changes revealed the intracellular regulatory mechanisms involved in biofilm formation. Diosgenin reduced biofilm formation in S. aureus ATCC 6538 and methicillin-resistant S. aureus (MRSA) CCARM 3090 by 39% and 61%, respectively. Conversely, it increased biofilm formation in S. aureus ATCC 29213 and MRSA CCARM 3820 by 186% and 582%, respectively. Cell surface hydrophobicity and extracellular protein and carbohydrate contents changed in a strain-specific manner in response to biofilm formation. An assessment of the changes in gene expression associated with biofilm formation revealed that diosgenin treatment decreased the expression of icaA and spa and increased the expression of RNAIII, agrA, sarA, and sigB in S. aureus ATCC 6538 and MRSA CCARM 3090; however, contrasting gene expression changes were noted in S. aureus ATCC 29213 and MRSA CCARM 3820. These results suggest that a regulatory mechanism of biofilm formation is that activated sigB expression sequentially increases the expression of sarA, agrA, and RNAIII. This increased RNAIII expression decreases the expression of spa, a surface-associated adhesion factor. An additional regulatory mechanism of biofilm formation is that activated sigB expression decreases the expression of an unknown regulator that increases the expression of icaA. This in turn decreases the expression of icaA, which decreases the synthesis of polysaccharide intercellular adhesins and ultimately inhibits biofilm formation. By assessing strain-specific contrasting regulatory signals induced by diosgenin in S. aureus without gene mutation, this study elucidated the signal transduction mechanisms that regulate biofilm formation based on physiological and gene expression changes. A novel aluminum-tolerant bacterial strain CA42 was isolated from the aquatic plant Eleocharis dulcis, which grows in a highly acidic swamp in Vietnam. Inoculation with CA42 allowed Oryza sativa to grow in the presence of 300  μM AlCl3 at pH 3.5, and biofilms were observed around the roots. Using 16S rRNA gene sequencing analysis, the strain was identified as Pullulanibacillus sp. CA42. This strain secreted large amounts of an extracellular polysaccharide (CA42 EPS). Results from structural analyses on CA42 EPS, namely methylation analysis and nuclear magnetic resonance (NMR), indicated that the chemical structure of CA42 EPS was a glycogen-like α-glucan. Purified CA42 EPS and the commercially available oyster glycogen adsorbed aluminum ions up to 15–30  μmol/g dry weight. Digestion treatments with α-amylase and pullulanase completely attenuated the aluminum ion-adsorbing activity of purified CA42 EPS and oyster glycogen, suggesting that the glycogen-like structure adsorbed aluminum ions and that its branching structure played an important role in its aluminum adsorbing activity. Furthermore, the aluminum tolerance of CA42 cells was attenuated by pullulanase treatment directly on the live CA42 cells. These results suggest that CA42 EPS adsorbs aluminum ions and is involved in the aluminum tolerance mechanism of Pullulanibacillus sp. CA42. Thus, this strain may be a potential plant growthpromoting bacterium in acidic soils. In addition, this study is the first to report a glycogen-like polysaccharide that adsorbs aluminum ions. Cordyceps militaris is a valuable medicinal fungus which has been widely used as a traditional medicine in East Asia. Compared to the well-known medicinal fungus C. sinensis, C. militaris can produce similar fermented metabolites with various biological activities, but it requires a shorter culture time and simpler culture conditions, and therefore, it has attracted increasing attention in recent years. The purpose of this review was to organize the current studies regarding metabolite production from C. militaris relative to their biological functions. We combined findings of metabolite production to correlate with different fermentation modes to obtain a full view of production processes used to yield the product. While research on C. militaris fermentation is not uncommon to date, its high value still highlights the importance of developing more modern fermentation processes for industrial production.

Porphyridium aerugineum is a unicellular freshwater red microalga that synthesizes and secretes into the culture medium an extracellular polysaccharide (EPS). In this study, algal growth and polysaccharide production, as well as the antioxidant capacity and antitumor effect of Porphyridium aerugineum EPS (PaEPS), were investigated. Cultivation of the microalgae was carried out in a photobioreactor under controlled conditions. Algal growth and the amount of EPS were monitored daily. The accumulated polysaccharide was extracted and lyophilized. At the end of cultivation, the concentration of microalgal biomass and PaEPS reached 3.3 and 1.2 g L−1, respectively. To examine the antioxidant capacity of PaEPS, FRAP and ABTS assays were performed. The cytotoxic activity of PaEPS was evaluated on the tumor cell lines MCF-7 (breast cancer) and HeLa (cervical adenocarcinoma) and on BJ (a non-tumor human skin fibroblast cell line), using MTT assay. The results obtained indicated that P. aerugineum polysaccharide exhibited a high ABTS radical-scavenging activity reaching up to 55%. The cytotoxic effect was best expressed in MCF-7 cells treated for 72 h with 1000 µg/mL PaEPS, where tumor cell proliferation was inhibited by more than 70%. Importantly, the PaEPS treatments did not significantly affect the viability of BJ cells. These findings promote the biotechnological production of P. aerugineum extracellular polysaccharide and reveal its potential as an anticancer and antioxidant agent for future applications.

The newly identified Sanghuangporus alpinus species of the Sanghuang mushroom genus has been found to possess significant medical benefits. However, the current artificial cultivation technology has not reached the requisite maturity. The response-surface methodology (RSM) was used to optimize the Sanghuangporus alpinus culture medium formulation and evaluate the functional activity of S. alpinus exopolysaccharides. First, a single-factor experiment was conducted to screen for optimal carbon and nitrogen sources for S. alpinus. Then, using Box–Behnken’s central composite design, a response-surface experiment was conducted to determine optimal culture parameters. Finally, the rationality of those parameters was assessed in a shaking flask experiment. The optimal culture parameters, determined through regression analysis, were 20.20 ± 0.17 g/L fructose (carbon source), 7.29 ± 0.10 g/L yeast extract (nitrogen source), and 0.99 ± 0.01 g/L dandelion. With optimization, the S. alpinus yield increased to 12.79 ± 1.41 g/L, twice that obtained from the initial culture medium. The S. alpinus exopolysaccharide exhibited an excellent antioxidant capacity, with the strongest scavenging effect noted on ABTS free radicals (lowest half-inhibitory concentration: 0.039 mg/mL). Additionally, this exopolysaccharide effectively inhibited various cancer cells, exhibiting the strongest activity against human glioma cells U251 (half-inhibitory concentration: 0.91 mg/mL). The RSM used to optimize the fermentation culture parameters of S. alpinus significantly increased the mycelial biomass. The improvement of Sanghuangporus alpinus yield through liquid fermentation and optimizing the fermentation medium could fill the existing gap in the cultivation of Sanghuangporus alpinus, as well as provide valuable data for the large-scale production of S. alpinus.

As biofilms mature, biomass and extracellular polysaccharide (EPS) content increases, enhancing pathogenicity. Therefore, this study aimed to evaluate the antibacterial efficacy of cold atmospheric plasma (CAP) against oral microcosm biofilms and the influence of biofilm maturity on treatment. Oral microcosm biofilms were cultured on hydroxyapatite disks for 2 and 6 days. Based on the treatment and biofilm maturity, these were subsequently allocated into six groups (N = 19 each): Groups 1 and 2 were incubated with distilled water for 1 min; Groups 3 and 4 were treated with CAP for 2 min, and Groups 5 and 6 were treated with 0.12% chlorhexidine gluconate for 1 min. Groups 1, 3, and 5 represent 2-day biofilms, and Groups 2, 4, and 6 represent 6-day biofilms. Treatments were repeated daily for 5 days. Antibacterial efficacy was analyzed by measuring oral biofilms’ red fluorescence intensity (RatioR/G) and quantifying EPS content and bacterial viability. The RatioR/G was 1.089-fold and 1.104-fold higher in Groups 4 and 6 than in Groups 3 and 5 following antibacterial treatment, respectively (p < 0.001). EPS content increased by 1.71-fold in Group 6 than in Group 5 (p < 0.001). Bacterial survival rate was the lowest in Group 3 (p = 0.005). These findings underscore the relevance of CAP treatment in maintaining antibacterial efficacy regardless of the biofilm development stage, highlighting its potential utility in oral care.

Armed with an arsenal of protein secretion systems, antibiotic efflux pumps, and the occasional proclivity for explosive self-destruction, Pseudomonas aeruginosa has become a model for the study of bacterial pathogenesis and biofilm formation. There is accruing evidence to suggest that the biofilm matrix—the bioglue that holds the structure together—acts not only in a structural capacity, but is also a molecular “net” whose function is to capture and retain certain secreted products (including proteins and small molecules). In this perspective, we argue that the biofilm matrixome is a distinct extracellular compartment, and one that is differentiated from the bulk secretome. Some of the points we raise are deliberately speculative, but are becoming increasingly accessible to experimental investigation.

Extracellular polysaccharide (EPS) produced by five lactic acid bacteria (Limosilactobacillus fermentum B55, Limosilactobacillus fermentum B62, Lactiplantibacillus plantarum 7830, Pediococcus acidilactici B30, and Lactobacillus helveticus K2) were purified and identified, and their effects on the texture of fermented goat milk were evaluated. The purified EPS fractions EPS 1a, EPS 2b, EPS 3c, EPS 4d, and EPS 5e of strains B62, 7830, K2, B55, and B30 were obtained with ion exchange chromatography, and their molecular weights were 2.41 × 104, 1.62 × 104, 6.42 × 103, 6.45 × 103, and 1.26 × 104 Da, found using gel permeation chromatography. The infrared spectrum results showed that these substances all contained polysaccharide characteristic absorption peaks, most of which contained O-H bonds, C-H bonds, hydroxyl and carboxyl bonds, and groups. The analysis of monosaccharide composition presented that EPS1a was composed of guluronic acid, rhamnose, and galactose, with a molar ratio of 2.7:1:2.4; EPS2b and EPS3c were composed of guluronic acid; EPS4d was composed of guluronic acid, glucose, and galactose, with a molar ratio of 1:1.1:1.2; and EPS5e was composed of glucose and galactose, with a molar ratio of 1.6:1, indicating the differences in the composition and structure of EPS produced by various strains. Compared with the control group that only had the starter added, adding EPS-producing strains could promote acid production and improve the texture of fermented goat milk, and its acidity, hardness, consistency, viscosity, and viscosity index were higher. This study explored the antibiofilm efficacy of 3,3'-diindolylmethane (DIM) on Staphylococcus aureus and its disinfection on common food-contact surfaces. The minimum biofilm inhibitory concentration (MBIC) of DIM on S. aureus was 62.5 μmol/L, while it did not impede the bacterial growth evaluated by growth curve and XTT reduction assay. DIM in the concentration range of 31.2−62.5 μmol/L demonstrated a dose-dependent antibiofilm activity to S. aureus, as confirmed by light microscopic (LM), confocal laser scanning microscopic (CLSM), and scanning electron microscopic (SEM) analyses. At DIM of 62.5 μmol/L, the biomass of S. aureus biofilm was significantly reduced by 97 % and its average thickness by 58 % (P < 0.05). DIM of 62.5 μmol/L inhibited the bacterial initial adhesion and proliferation, as well as cell motility; the release of extracellular DNA (eDNA) and extracellular polysaccharide (EPS) were reduced by 75 % and 69 %, respectively. DIM exhibited a strong inhibition to S. aureus biofilm formation on common food-contact surfaces, including 304 stainless steel, glass, and polyvinyl chloride (PVC) but not disperse the mature biofilm. Overall, our investigation identified DIM as a promising antibiofilm agent and its suitability to prevent the biofilm formation of S. aureus on common food-contact surfaces utilized during food processing.

Abstract Background Enterococcus faecalis (E. faecalis) plays an important role in the failure of root canal treatment and refractory periapical periodontitis. As an important virulence factor of E. faecalis, extracellular polysaccharide (EPS) serves as a matrix to wrap bacteria and form biofilms. The homologous rnc gene, encoding Ribonuclease III, has been reported as a regulator of EPS synthesis. In order to develop novel anti-biofilm targets, we investigated the effects of the rnc gene on the biological characteristics of E. faecalis, and compared the biofilm tolerance towards the typical root canal irrigation agents and traditional Chinese medicine fluid Pudilan. Methods E. faecalis rnc gene overexpression (rnc+) and low-expression (rnc−) strains were constructed. The growth curves of E. faecalis ATCC29212, rnc+, and rnc− strains were obtained to study the regulatory effect of the rnc gene on E. faecalis. Scanning electron microscopy (SEM), confocal laser scanning microscopy (CLSM), and crystal violet staining assays were performed to evaluate the morphology and composition of E. faecalis biofilms. Furthermore, the wild-type and mutant biofilms were treated with 5% sodium hypochlorite (NaOCl), 2% chlorhexidine (CHX), and Pudilan. The residual viabilities of E. faecalis biofilms were evaluated using crystal violet staining and colony counting assays. Results The results demonstrated that the rnc gene could promote bacterial growth and EPS synthesis, causing the EPS-barren biofilm morphology and low EPS/bacteria ratio. Both the rnc+ and rnc− biofilms showed increased susceptibility to the root canal irrigation agents. The 5% NaOCl group showed the highest biofilm removing effect followed by Pudilan and 2% CHX. The colony counting results showed almost complete removal of bacteria in the 5% NaOCl, 2% CHX, and Chinese medicine agents’ groups. Conclusions This study concluded that the rnc gene could positively regulate bacterial proliferation, EPS synthesis, and biofilm formation in E. faecalis. The rnc mutation caused an increase in the disinfectant sensitivity of biofilm, indicating a potential anti-biofilm target. In addition, Pudilan exhibited an excellent ability to remove E. faecalis biofilm.

A strain, 3EQS1, was isolated from a salt sample taken from Lake Qarun (Fayoum Province, Egypt). On the basis of physiological, biochemical, and phylogenetic analyses, the strain was classified as Chromohalobacter salexigens. By 72 h of growth at 25 °C, strain 3EQS1 produced large amounts (15.1 g L−1) of exopolysaccharide (EPS) in a liquid mineral medium (initial pH 8.0) containing 10% sucrose and 10% NaCl. The EPS was precipitated from the cell-free culture medium with chilled ethanol and was purified by gel-permeation and anion-exchange chromatography. The molecular mass of the EPS was 0.9 × 106 Da. Chemical analyses, Fourier transform infrared spectroscopy, and nuclear magnetic resonance spectroscopy showed that the EPS was a linear β-d-(2 → 6)-linked fructan (levan). In aqueous solution, the EPS tended to form supramolecular aggregates with a critical aggregation concentration of 240 µg mL−1. The EPS had high emulsifying activity (E24, %) against kerosene (31.2 ± 0.4%), sunflower oil (76.9 ± 1.3%), and crude oil (98.9 ± 0.8%), and it also had surfactant properties. A 0.1% (w/v) aqueous EPS solution reduced the surface tension of water by 11.9%. The levan of C. salexigens 3EQS1 may be useful in various biotechnological processes. [ABSTRACT FROM AUTHOR]

RpoN is an alternative sigma factor (sigma 54) that recruits the core RNA polymerase to promoters of genes. In bacteria, RpoN has diverse physiological functions. In rhizobia, RpoN plays a key role in the transcription of nitrogen fixation (nif) genes. The Bradyrhizobium sp. DOA9 strain contains a chromosomal (c) and plasmid (p) encoded RpoN protein. We used single and double rpoN mutants and reporter strains to investigate the role of the two RpoN proteins under free-living and symbiotic conditions. We observed that the inactivation of rpoNc or rpoNp severely impacts the physiology of the bacteria under free-living conditions, such as the bacterial motility, carbon and nitrogen utilization profiles, exopolysaccharide (EPS) production, and biofilm formation. However, free-living nitrogen fixation appears to be under the primary control of RpoNc. Interestingly, drastic effects of rpoNc and rpoNp mutations were also observed during symbiosis with Aeschynomene americana. Indeed, inoculation with rpoNp, rpoNc, and double rpoN mutant strains resulted in decreases of 39, 64, and 82% in the number of nodules, respectively, as well as a reduction in nitrogen fixation efficiency and a loss of the bacterium's ability to survive intracellularly. Taken together, the results show that the chromosomal and plasmid encoded RpoN proteins in the DOA9 strain both play a pleiotropic role during freeliving and symbiotic states. [ABSTRACT FROM AUTHOR]

Ginkgo biloba leaf polysaccharide (GBLP) is a biomolecule with various activities such as anti-tumor and anti-inflammatory, but the natural production is low. Extracellular polysaccharide (EPS) was isolated and purified from endophytic Lysinibacillus sphaericus Ya6 of ginkgo biloba. Similarities in chemical structure and biological activity of two polysaccharides were investigated. Results showed that both polysaccharides are acidic polysaccharides containing pyran rings with characteristic absorption peaks for polysaccharides and carboxylates. The molecular weights varied slightly, with 13.20 kDa and 12.08 kDa for EPS Ya6 and GBLP, respectively. GCMS– showed that fucose was present only in EPS Ya6 , and rhamnose and arabinose were higher in GBLP than in EPS Ya6. These gaps above may result in higher antioxidant activity of GBLP than EPS Ya6 , as verified in free radical scavenging capacity. EPS Ya6 and GBLP also have antibacterial effects, and EPS Ya6 has a wide antibacterial spectrum. Overall, EPS Ya6 has similar chemical structure and same biological activity to GBLP, which provides a theoretical basis for subsequent product development. This study can alleviate the shortage of polysaccharide resources caused by the long growth cycle of ginkgo and the complex natural polysaccharide extraction process, which is conducive to the sustainable development of the ecological environment. Both the rnc+ and rnc− biofilms showed increased susceptibility to the root canal irrigation agents. The 5% NaOCl group showed the highest biofilm removing effect followed by Pudilan and 2% CHX. The colony counting results showed almost complete removal of bacteria in the 5% NaOCl, 2% CHX, and Chinese medicine agents' groups. Conclusions: This study concluded that the rnc gene could positively regulate bacterial proliferation, EPS synthesis, and biofilm formation in E. faecalis. The rnc mutation caused an increase in the disinfectant sensitivity of biofilm, indicating a potential anti-biofilm target. Dental caries, the most prevalent oral infectious disease, is closely associated with Streptococcus mutans . This study investigates the antimicrobial properties of the temporin-GHb peptide and its derivatives (GHbR, GHbK, and GHb3K) against S. mutans . These peptides exhibited potent anti - S. mutans activity through a membrane-disruptive mechanism, confirmed by flow cytometry and fluorescence staining assays while showing lower bactericidal effects on beneficial probiotic bacteria. Additionally, they inhibited the biofilm matrix formation by disrupting extracellular polysaccharide (EPS) synthesis, as demonstrated by zymography, qRT-PCR, and sucrose metabolism experiments. In a rat model of S. mutans -induced dental caries, treatment with these peptides significantly reduced the incidence of dental lesions. H&E staining analysis of rat oral tissues confirmed the biosafety of GHb and GHb3K. These findings suggest that temporin-derived peptides effectively target EPS, inhibiting biofilm formation and virulence, offering a promising strategy for preventing dental caries and promoting oral health. The findings suggest potential applications for peptide-based interventions to mitigate biofilm-related issues across various fields, including agriculture, food processing, and healthcare.

Stenotrophomonas rhizophila JC1 and its extracellular polysaccharides (EPS) have been shown to effectively adsorb heavy metals in previous studies. The fermentation conditions of EPS by S. rhizophila JC1 were optimized using the Box-Behnken design (BBD). The composition, structural characteristics, and heavy metal adsorption capacity of EPS were systematically evaluated. The alleviation mechanism of Pb 2+ stress on alfalfa was investigated through EPS inoculation. The maximum EPS yield reached 0.313 %. EPS consisted of glucose, glucosamine, galactose, and mannose in a molar ratio of 12.20:1:22.29:1.68. EPS also contained four distinct polymers with molecular weights of 623,683.71 Da, 144,072.27 Da, 105,892.21 Da, and 51,094.79 Da. The adsorption processes conformed to the pseudo-second-order model and Langmuir isotherm model. High Pb 2+ concentrations significantly reduced germination percentage, germinative force, root length, fresh weight, and soluble protein, inhibited photosynthesis, exacerbated oxidative stress, and caused damage to the antioxidant system, thereby inhibiting seedling growth. EPS at low concentrations can promote alfalfa seed germination and mitigate Pb 2+ stress by reducing the aforementioned damage. The unique tissue structure of pathogenic bacteria biofilm plays an important role in its pathogenicity and bactericide resistance. Inhibition or destruction of biofilm formation of pathogenic bacteria is of great significance for the control of plant bacterial diseases. In this study, Paracidovorax citrulli was inoculated into KB medium containing acetic acid, and after shaking at 28°C and 55 r/min for 48 h, it was found that the content of extracellular polysaccharide, extracellular protein and extracellular DNA (eDNA) decreased with the increase of acetic acid concentration, which resulted in the decrease of biofilm formation, it is not even possible to form biofilms on plastic slides. When the final concentration of acetic acid in the culture medium was greater than or equal to 0.5 mg/mL, there was no biofilm on the plastic slides. Streptococcus mutans is considered a major cariogenic bacterium. Most anti-cariogenic dentifrices are limited by a short exposure time. The aim of the present study was to test the hypothesis that adding a mucoadhesive agent to the formulation may increase its bioavailability and efficacy. We tested the effect of adding hydroxyethyl cellulose (HEC) to an herbal extract solution containing lavender, echinacea, sage, and mastic gum, which have been previously shown to be effective against Streptococcus mutans. Mucin-coated wells were treated with four test solutions: saline, herbal extracts, herbal extracts with HEC, and chlorhexidine. The wells were incubated with Streptococcus mutans and studied for biofilm formation (Crystal violet assay), acid production (lactate assay), acid tolerance (ATPase assay), and exopolysaccharide (EPS) production using fluorescent microscopy. The results showed that the addition of HEC to the herbal extract solution caused a significant reduction in Streptococcus mutans biofilm formation, lactic acid production, and EPS quantity (p < 0.001). BACKGROUND: Escherichia coli can cause difficult refractory infections, which seriously threaten human health, due to the formation of "biofilms." The pgaABCD opero is one of the important regulatory genes for biofilm formation. OBJECTIVE: To construct gene deletion strain MG1655/Δpga of Escherichia coli, construct the complementation strains of Δpga/pgaA, Δpga/pgaB, Δpga/pgaC, Δpga/pgaD by complementing pgaA, pgaB, pgaC and pgaD, respectively, observe the biofilm formation ability, and exopolysaccharide and morphological characteristics of deletion and complementation strains, and analyze the effects of gene pga on the regulation and characteristics of biofilm. METHODS: Based on the Lambda Red recombination system, helper plasmid pKD46 was transferred into competent cells MG1655 to obtain MG1655/pKD46 monoclones. Targeting fragment of pgaABCD gene was transformed to competent cells MG1655/pKD46 to construct gene deleted mutant of MG1655/Δpga. Using plasmid pBR322 as vector, pBR322-pgaA, pBR322-pgaB, pBR322-pgaC, and pBR322-pgaD were constructed and transformed into MG1655/Δpga to obtain the corresponding complementation strains. Congo red-broth agar plate, crystal violet semi-quantitative method, and transmissionelectronmicroscopy were utilized to detect property changes of biofilm formation capacity of construction strains. At the same time, extracellular polysaccharide was detected using phenol-sulfuric acid method. RESULTS AND CONCLUSION: The biofilm formation capacity and extracellular polysaccharide of deletion strains were significantly decreased (P < 0.05). Some cells were damaged and collapsed. The section showed vacuolardegeneration in bacteria. To conclude, deletion of gene pgaABCD can affect the biofilm characteristics for Escherichia coli, and the genes pgaA, pgaB, pgaC and pgaD, but not the only regulatory gene, are involved in the biofilm formation.
