Speleothems found in caves worldwide are considered the natural libraries of paleontology. Bacteria found in these ecosystems are generally limited to Proteobacteria and Actinomycetota, but rare microbiome and “Dark Matter” is generally under-investigated and often neglected. This research article discusses, for the first time to our knowledge, the diachronic diversity of Actinomycetota entrapped inside a cave stalactite. The planet’s environmental microbial community profile of different eras can be stored in these refugia (speleothems). These speleothems could be an environmental “Microbial Ark” storing rare microbiome and “Dark Matter” bacterial communities evermore.

In sole aquaculture production, consistency in the quality of produced eggs throughout the year is unpredictable. Hox genes have a crucial role in controlling embryonic development and their genetic variation could alter the phenotype dramatically. In teleosts genome duplication led paralog hox genes to become diverged. Direct association of polymorphism in hoxa1a, hoxa2a & hoxa2b of Solea solea with egg viability indicates hoxa2b as a potential genetic marker. High Resolution Melt (HRM) analysis was carried out in 52 viable and 61 non-viable eggs collected at 54±6 hours post fertilization (hpf). Allelic and genotypic frequencies of polymorphism were analyzed and results illustrated a significantly increased risk for non-viability for minor alleles and their homozygous genotypes. Haplotype analysis demonstrated a significant recessive effect on the risk of non-viability, by increasing the odds of disrupting embryonic development up to three-fold. Phylogenetic analysis showed that the paralog genes hoxa2a and hoxa2b, are separated distinctly in two clades and presented a significant ω variation, revealing their diverged evolutionary rate.

The Mediterranean monk seal (Monachus monachus) is classified as an endangered species by the IUCN, with a global population that does not exceed 800 individuals. There is limited understanding around the biology and health status of the species, rendering possible parasitic infections grave for its conservation efforts. The aim of the current study was the molecular identification of a parasitic nematode found in the digestive system of a sub-adult Mediterranean monk seal individual, that was found stranded in the area of Pagasitikos Gulf, Greece in 2019. Analysis of the stomach contents revealed the presence of two intact female nematode individuals. Standard protocols were followed as DNA extraction of the parasites was conducted and PCR amplification of the cytochrome oxidase subunit I (COI) mitochondrial gene was implemented. Sequencing analysis of a 585 bp-amplified product displayed a 96% similarity of the screened nematodes to the Pseudoterranova bulbosa species. Bayesian inference was implemented for the subsequent tree reconstruction. The phylogenetic tree revealed a clear genetic similarity between our parasitic nematode individuals named as Pseudoterranova spp. and Pseudoterranova bulbosa (bootstrap value: 82%), which is indicated for the first and only time as such, to be found in the waters of the Mediterranean Sea and also in the stomach of a Mediterranean monk seal.

Allometric data of marine fish species from Rhodes, southeastern Aegean Sea are scarce. Their collection is crucial as they provide important information on the ongoing changes of small-scale fisheries in Rhodian coastal marine waters, a highly affected region of the Eastern Mediterranean by biological invasions. Monthly experimental trials and random samplings with static nets were conducted from April 2021 to March 2022 in the coastal marine waters of eastern Rhodes, Levantine Sea. Experimental boat seining was deployed in November 2022 in the coastal waters of northwest Rhodes, Aegean Sea. Total length and total weight of several individuals of 21 fish species were measured and the length-weight relationships of 12 indigenous and nine nonindigenous fish species were examined. The allometric coefficient (b) did not differ significantly from 3.00 for most of the species (70%), demonstrating isometric growth. Positive and negative allometry were found for three indigenous and three non-indigenous species. The largest and heaviest species were the bluespotted cornetfish (Fistularia commersonii), with a maximum TL of 117.60 cm and the silver-cheeked toadfish (Lagocephlaus sceleratus), with a maximum weight of 4640.90 g. Statistically significant positive allometric relationships were found for three non-indigenous fish, namely the bluespotted cornetfish, the devil firefish (Pterois miles) and the dusky spinefoot (Siganus luridus), suggesting that these species are thriving in the under-study area. Recently, we could show that scallops show limitations of muscular performance like a reduced force under ocean warming and acidification. However, the underlying mechanisms at the cellular level are not completely understood. Metabolomics has become a valuable tool to evaluate the responses of marine organisms to various stressors. In the present study we therefore used a semi-targeted, multi tissue NMR based metabolomic approach to analyze metabolite patterns in the Atlantic king scallop, Pecten maximus, that were long-term acclimated to different end of century conditions of ocean warming (OW), ocean acidification (OA) and their combination (OWA). We investigated tissue specific metabolic profiles and metabolite concentrations in frozen tissues from gills, mantle and phasic and tonic adductor muscle of P. maximus under present conditions using ¹H-HR-MAS NMR spectroscopy. A set of 33 metabolites revealed a clear tissue-specific pattern which can be attributed to the individual functions of the respective tissue type. We then evaluated the impact of OW, OA and OWA on the metabolic profiles of the different tissues. OW was the main driver of the changes in metabolites. In particular, energy-related metabolites seem to play an important role in the physiological response of scallops to OW and OWA. In combination with pathway analysis and network exploration we propose a possible correlation between metabolic changes in the adductor muscle and limited swimming performance of P. maximus under future climate. While the metabolic response of the phasic muscle seems to mainly depend on net consumption of energy related metabolites such as ATP and phospho-L-arginine, the tonic muscle seems to rely on metabolizing specific amino acids and beta-oxidation to account for the elevated energetic requirements under ocean warming and acidification. This manuscript explores the role of European anchovies (Engraulis encrasicolus) in the central Mediterranean Region, shedding light on their ecological significance, conservation challenges, and sustainable utilization. The European anchovy is one of Europe's most important fish resources in the Mediterranean basin, and it is considered a keystone species, playing a pivotal role in both ecological and socio-economic dimensions. However, in recent decades, European anchovy, together with Sardina pilchardus (commonly known as European sardine), has suffered a population decline for several reasons. Consequently, it is necessary to improve the management of anchovy fisheries by understanding the reproductive modes and characteristics, the influence of currents on the passive transport of eggs and larvae, the feeding habits, the environmental adaptability (e.g., salinity), and the distribution of ecotypes along the Italian coasts. Such information is fundamental for the management of fisheries, especially artisanal ones, and to control frauds, especially in protected, geographically referred, and traditional high-quality commercial products. Various aspects, ranging from their population dynamics to their industrial processing and the ecological implications of these activities were delved, highlighting the knowledge about anchovy populations and ecotypes and its importance in maintaining ecosystem balance and sustaining human communities. The ecological interaction of anchovies within the food web, as essential data in the conservation actions and management of these resources was emphasized. In addition, the metabolic and stomach contents diversity among anchovy populations and ecotypes was discussed, enhancing our understanding of their adaptability to varying environmental conditions. The manuscript then explores the traditional and industrial processing of anchovies, encompassing aspects ranging from fishing techniques (i.e., methods of capture) to their industrial significance, sustainability concerns, issues of fraud, and the establishment of geographical traceability. Finally, the opportunities for sustainable and biotechnological utilization of anchovy discards were also further explored, demonstrating the potential for waste reduction and resource optimization. This study reports on a new translocated population of the Western Greece Goby (Economidichthys pygmaeus Holly, 1929) within the Thrace freshwater ecoregion in Northeastern Greece, approximately 370 km east of the species' native range (Ionian freshwater ecoregion; Western Greece). Specimens were identified by morphological examination and molecular confirmation. Based on a literature review and anecdotal information the Western Greece Goby was possibly introduced within the last two to five years, perhaps associated with fish farm imports from Western Greece. Population characteristics of the species in the new locations indicate that the species is fully established within an important biodiversity hotspot, the Aggitis sub-basin of the Strymon River, a transboundary river shared with Greece, Bulgaria and North Macedonia. This record represents the easternmost range expansion of the Western Greece Goby. In addition, it confirms a noticeable trend of expansion of this species since it has been also documented in the Southeastern Adriatic (Albania, North Macedonia) and the Western Aegean (southern Greece) during the last decade. Various methodologies including genetic analyses, morphometrics, proteomics, lipidomics, metabolomics, etc. are now used or being developed to authenticate fish and seafood. Such techniques usually lead to the generation of enormous amounts of data. The analysis and interpretation of this information can be particularly challenging. Statistical techniques are therefore commonly used to assist in analyzing these data, visualizing trends and differences and extracting conclusions. This review article aims at presenting and discussing statistical methods used in studies on fish and seafood authenticity and adulteration, allowing researchers to consider their options based on previous successes/failures but also offering some recommendations about the future of such techniques. Techniques such as PCA, AMOVA and FST statistics, that allow the differentiation of genetic groups, or techniques such as MANOVA that allow large data sets of morphometric characteristics or elemental differences to be analyzed are discussed. Furthermore, methods such as cluster analysis, DFA, CVA, CDA and heatmaps/Circos plots that allow samples to be differentiated based on their geographical origin are also reviewed and their advantages and disadvantages as found in past studies are given. Finally, mathematical simulations and modeling are presented in a detailed review of studies using them, together with their advantages and limitations. [ABSTRACT FROM AUTHOR]

Abstract Background Comprehensively detailed information on population dynamics for benthic species is crucial since potential admixture of individuals could shift the genetic subdivision and age structure during a full breeding period. The apparent genetic impact of the potential recruitment strategy of Norway lobster Nephrops norvegicus is still under research. For this reason the present study was focused on genetic variation of the species over a given continuous year period in a semi-enclosed gulf of the Aegean Sea. Results Analyses revealed that the relative smaller size class in females and the apparent faster growth of males may represent a key-role differential strategy for the two sexes, whereas females tend to mature slower. Heterozygosity fitness correlations (HFCs) showed substantially significant associations suggesting that inbreeding depression for females and outbreeding depression for males are the proximate fitness mechanisms, respectively. Conclusions Nephrops norvegicus uniformal genetic composition (background of high gene flow), could be attributed to potential population recolonization, due to a hypothesized passive larval movement from deeper waters, which may suggest that some offspring of local residents and potential male non-breeders from other regions admixture randomly.

Cryopreservation is a technique that offers various advantages, especially in fish, among others, that makes the reproduction of species easier through a constant supply of sperm, synchronization of the gamete availability of both sexes, storage of semen for genetic improvement programs, reduction in the cost by eliminating the need to maintain male broodstock, and conserving the gametes of endangered species. However, freezing and warming procedures for cryopreservation lead to a reduction in the quality and viability of cryopreserved sperm because of oxidative stress. For this reason, the enrichment of extender media with antioxidants is a common method of cryopreservation of the semen of several fish species. Recently, many studies have been published for the protective role of antioxidants and especially of melatonin on male fertility preservation both in farm animals and humans, demonstrating the beneficial effects of melatonin as a sperm cryoprotectant. On the other hand, very few studies were conducted using melatonin as an antioxidant in different male fish species for semen cryopreservation. We conclude that the use of moderate concentrations of melatonin are beneficial to semen preservation, and the mechanisms through which melatonin acts positively on spermatozoa need to be further investigated to establish improvement protocols for cryopreservation in fish species.

The Mediterranean monk seal (Monachus monachus) is classified by the IUCN as “endangered,” with a global population estimated to number fewer than 800 individuals. Our understanding of the biology and health status of the species is still limited, rendering every medical case a challenge for conservationists and veterinary clinicians. Although studying and managing disease in wild marine hosts is complex and challenging, studying and mitigating the effects of any disease to the Mediterranean monk seal is of utmost importance for conservation. The aim of this study was to document for the first time the presence of the hookworm Uncinaria hamiltoni in rehabilitated Mediterranean monk seal pups in Greece. A detailed examination protocol was followed for all pups that live-stranded over 30 years in 22 different locations, including physical, parasitological, and other examinations. Hookworms (adults and/or eggs) were detected in all the fecal samples, from all animals. Molecular identification using MtDNA (COI) and ribosomal DNA (D2/D3 28S and internal transcribed spacer [ITS] regions) identified the nematode species as Uncinaria hamiltoni. The clinical impacts and the benefits of anthelmintic treatment as a tool for the conservation management of the species are discussed.

In this review we propose the use of telomeric length (TL) as an authenticity marker that could provide an alternative method for differentiating fish and seafood samples or detecting fraud. Considering the ever-growing number of incidents of economically motivated fish and seafood adulteration using even more sophisticated methods to overcome current authenticity markers, the need to identify novel authenticity markers becomes essential. The TL of fish and seafood depends on individual characteristics (e.g., sex, age) and the environmental stimuli (e.g., temperature, water quality) to which these are exposed. Hence, both wild marine and freshwater populations occupying different geographical origin habitats might differ substantially because of the environmental cues affecting them. Moreover, the implementation of various rearing practices in aquaculture, such as different levels of fish and seafood density and increased ambient noise combined with site-specific environmental cues could affect TL, providing regulatory authorities with valuable information by distinguishing wild from reared populations and organic from conventional ones. In the present review the effects of both the environmental conditions and individual characteristics on the telomeric stability of fish and seafood telomeres are discussed, suggesting TL as a potential prospect authenticity marker that could be used to prevent fish and seafood adulteration. A fishery-independent survey with a commercial bottom otter trawl was conducted in the Pagasitikos Gulf, a semi-enclosed, trawl-restricted gulf in Greece. The study aimed to ecologically assess the fishery communities that are experiencing a decline in fishery landings. Twelve hauls of 30 min duration each were conducted in the eastern, western, and central parts of the gulf at depths between 62 and 97 m. In total, 57 species were captured, with the highest species number recorded in the west (35 species), followed by the center (32 species) and east (29 species). The highest numerical abundance was recorded at the center, followed by the east and west, with an opposing trend exhibited for biomass; however, GLM detected no significant spatial differences (in area or depth) or interaction. Ecological indices exhibited significantly lower values in the west–east area comparison. Linear Discriminant Analysis, mMDS, and hierarchical clustering indicated the presence of two main groups (east and west), with PERMANOVA showing a significant difference in the demersal communities among the identified groups. The results indicated moderately disturbed demersal communities and an increasing level of disturbance from east to west. Considering the economic value of Pagasitikos to the local fishing community, additional fishery management measures are deemed necessary to protect the fishery resources and mitigate the potential risks of overexploitation. Monitoring of Delphinidae species population patterns in the Mediterranean Sea was carried out in a sequence of surveys employing different approaches. Data from seven-year surveys with small catamaran sailing boats were analyzed under model-based approaches. Density Surface Models were used to produce spatial distribution prediction of three Delphinidae species (Stenella coeruleoalba, Tursiops truncatus, and Delphinus delphis) in an extended study area covering much of the Mediterranean Sea. A classical distance sampling protocol was applied in order to calculate the detection probability of clusters. Static (depth, slope, distance from the coast, and distance from isobaths of 200 m) and nonstatic (sea surface temperature and chlorophyll) variables were used to predict the species distribution/abundance in a generalized additive model context. Stenella coeruleoalba was found to be the dominant species, with an extended distribution in the study area; its abundance was significantly affected by both depth and distance. Tursiops truncatus and Delphinus delphis illustrated a significant abundance correlation with depth and chlorophyll, respectively, while both species showed a robust longitude correlation. Our model pinpoints the significance of nondesigned transect line surveys, suggesting the importance of specific habitat areas for future monitoring and conservation aspects of marine mammals.

Admixture and hybridization may play a key role in population dynamics and speciation with respect to habitat, demographic history, and adaptive selection. The present study examines the genetic diversity of two congeneric—but in different subgenera—holothurians that live in sympatry in mixed populations. Strong evidence of admixture was provided by analyzing RAD sequencing data from 90 and 67 individuals of Holothuria (Holothuria) tubulosa and Holothuria (Roweothuria) poli, respectively, from various areas of the Hellenic Seas (eastern Mediterranean). Coalescent demographic analysis revealed a recent directional gene flow from H. poli to H. tubulosa. The two species populations diverged around 13.5 thousand years ago, just after the Last Glacial Maximum. According to the pairwise sequential Markovian coalescent approach, the historical population effective sizes for both species declined during the last Pleistocene glaciations, probably due to population decline, followed by a relative rapid recovery as it is calculated using LD methods. The presented results imply a role for admixture upon secondary contact and are consistent with the recent suggestion that the genomic underpinning of ecological speciation often has an older, allopatric origin. The red porgy (Pagrus pagrus) and the common dentex (Dentex dentex) are Sparidae species of high commercial value, traded in the Greek market. In some cases, fish species identification from Greek fisheries is difficult for the consumer due to the strong morphological similarities with their imported counterparts or closely related species such as Pagrus major, Pagrus caeroleustictus, Dentex gibbosus and Pagellus erythrinus, especially when specimens are frozen, filleted or cooked. Techniques based on DNA sequencing, such as COI barcoding, accurately identify species substitution incidents; however, they are time consuming and expensive. In this study, regions of mtDNA were analyzed with RFLPs, multiplex PCR and HRM in order to develop a rapid method for species identification within the Sparidae family. HRM analysis of a 113 bp region of cytb and/or a 156 bp region of 16s could discriminate raw or cooked samples of P. pagrus and D. dentex from the aforementioned closely related species and P. pagrus specimens sampled in the Mediterranean Sea when compared to those fished in the eastern Atlantic. HRM analysis exhibited high accuracy and repeatability, revealing incidents of mislabeling. Multiple samples can be analyzed within three hours, rendering this method a useful tool in fish fraud monitoring. [ABSTRACT FROM AUTHOR]

Population dynamics in the marine realm can shape species’ spatial structure and genetic variability between given geographical areas. Connectivity is an important factor of species’ population structure. In this study, we examined the genetic diversity and structure of white seabream (Diplodus sargus, L. 1758) in the eastern Mediterranean basin, using a panel of four microsatellite markers. Recorded low FST values within the study area indicate little evidence of genetic differentiation among populations. Results suggest high gene flow which may imply near-panmixia between populations, indicating the possibility of a probable movement of adult migrants, or strong passive drift at sea in early life stages of the species. To this extent, bibliographically speaking, different species within the Sparidae family favor altered population dynamics patterns with respect to local populations and genetic divergence, in the context of the molecular marker used.

Acute toxicity and inhibition on growth of Artemia franciscana nauplii (Instar I-II) after exposure to the reference toxicants bisphenol a (BPA) and sodium dodecyl sulfate (SDS) were studied. LC50 values were calculated and differences in body growth were recorded after 24, 48, and 72 h of exposure to the toxicants. The results indicated that BPA had lower toxicity than SDS. Development of the nauplii was clearly influenced by duration of exposure. Growth inhibition was detected for both toxicants. Abnormal growth of the central eye of several Artemia nauplii after 72 h of exposure to BPA was also detected. Our results indicate that growth inhibition could be used as a valid endpoint for toxicity studies.
