Abstract In the present day, water treatment has emerged as a significant global concern, particularly due to the proliferation of pollution sources. The utilization of dyes, such as Evans blue, in several industries is among the most significant contributors to these pollutants. Forsterite nanoparticles were synthesized by the sol-gel technique and calcined at different temperatures to determine the optimum temperature at which pure nanoforsterite was obtained. Then, it was analyzed using X-ray diffraction (XRD), atomic force microscope (AFM), transmission electron microscope (TEM), Brunauer-Emmett-Teller (BET) , contact angle, and zero-point charge. The adsorption capability of forsterite nanoparticles (Nps) was evaluated by a batch adsorption experimental method to remove Evans blue dye (EBD). Parameters such as agitation speed, dosage of forsterite Nps, pH, and contact time were considered at ambient temperature. At pH = 3, dose of Nps = 1 g/L, and 600 rpm within 10 min, the results indicated a removal rate of around 100%. Furthermore, it was shown that the material may be employed for 3 cycles with a removal rate of 90%. Multiple kinetic and isotherm models, including Langmuir, Temkin, and Freundlich models, were used to analyze the results and clarify the mechanism of the adsorption phenomena. The findings from the isotherm and kinetic studies indicated that the system conforms to Langmuir and pseudo-second-order, respectively.

The Zirconium Oxide-Zinc Oxide (2ZrO2-7ZnO) nanoparticles (NPs) were successfully synthesized by solution combustion method using Basella alba raw juice at 600oC. We investigated the properties of the ZrO2-ZnO NPs via characterizations like X-Ray powder Diffraction, Energy Dispersive X-ray analysis, Scanning Electron Microscopy, Transmission Electron Microscopy, Fourier Transform Infrared Spectroscopy and Brunauer–Emmett–Teller analyses. X-ray powder diffraction studies revealed that the cubic ZrO2 and hexagonal wurtzite ZnO having the average particle size was found to be 12.6 nm and was 10.8 nm. The influence of contact time, initial dye concentration, and pH were among the independent variables used in the study. At a fixed dye concentration and catalytic load, a series of photocatalytic tests were carried out at an identical experimental condition. ZrZn4 has a high degradation capability, and it degraded 100% of the dye in 90 min. The Response Surface Model (RSM) was used to optimize and describe the interdependencies of the different variables. The method was evaluated using the Box-Behnken design (BBD). Under the constraints, the best conditions for EB elimination are 1.25 mg/L dye concentration at pH 12.0 and a contact time of 60 min, with a 99% response. A second-order polynomial model was used to understand the experimental results, and the effectiveness of the chosen model was verified by the strong agreement in determination coefficient values. The possible use of the 2ZrO2-7ZnO NPs as electrode modifier for EB sensing purposes was then evaluated using carbon paste electrodes, using cyclic voltammetry. [ABSTRACT FROM AUTHOR]

Membrane isolation, ion exchange, precipitation, transformation, and biosorption are all viable methods for pollutant removal. Adsorption is a common commercial method to concentrate precious molecules or eliminate contaminants, and it is a cost-effective method of treating industrial wastewater. A novel method for increasing their removal effectiveness has been developed for this purpose, using a low-cost biosorbent made from durian husk and Evans blue (EB) dye as a subject. The EB dye adsorption percentage was determined to be 95.95% with 72.0 mg/g adsorption amount at optimal conditions of pH 2 and 40 °C. The second-order kinetic model fit the experimental data the best. Additionally, the results indicated that the Sips isotherm model fits the experimental data better and that the experiment involved single-layer adsorption on the adsorbent surface. A non-linear regression of the van't Hoff plot gave negative values of Gibbs free energy (−39.38 to −41.48 kJ/mol) at all the temperatures studied (from 20 to 60 °C), indicating that the adsorption process is spontaneous and feasible. A negative value for the enthalpy (ΔHo=−23.37kJ/mol) indicated that the adsorption was exothermic, and the positive value of (ΔSo=54.55kJ/mol·K) concludes the nature of adsorption of Evans blue dye by Durian husk likely to follow a physisorption adsorption mechanism. Methods for removing pollutants include membrane isolation, ion exchange, precipitation, transformation, and biosorption. Adsorption is a cost-effective method of treating industrial wastewater and a common commercial method for concentrating valuable molecules or eliminating contaminants. Banana peel is one of the largest underutilized agricultural wastes in Malaysia. A novel method of using a low-cost biosorbent made from banana peel and Evans blue (EB) dye as a target is the target of this study. The optimal conditions for EB dye adsorption occurred at a dye concentration of 200 mg/L, adsorbent dosages between 10 and 20 g/L, temperature of 25 °C, incubation time of 180 min, and agitation speeds of 100 rpm. Statistical discriminatory analysis showed that the pseudo-second-order kinetic model and the Redlich–Petersen isotherm model were the best models. The maximum adsorption capacity based on Langmuir's isotherm prediction was 58.51 mg g−1. A non–linear regression of the thermodynamic van't Hoff plot based on a dimensionless equilibrium constant resulted in negative values for Gibb's free energy and enthalpy, indicating that the adsorption process is spontaneous and exothermic. In this work, the tetragonal Zirconium oxide (ZrO2) nanoparticles (NPs) were successfully synthesized by solution combustion method using Zirconium (IV) oxynitrate hydrate as the metal precursor and an oxidizer, Basella alba raw extract at 6000C. In this study, natural fuel is used to avoid harmful chemical fuels that may pollute the environment during combustion. The impact of the fuel-to-oxidant molar ratio on the surface morphological features of nanocrystalline zirconia particles has been documented. We investigated the Physico-chemical properties of the ZrO2 NPs via thorough characterizations like XRD, EDS, SEM, TEM, FTIR, UV-Vis, and BET. ZrO2 NPs exhibit perfect photocatalytic degradation activity towards Evans blue, a toxic dye. The influence of contact time, initial dye concentration, and pH were among the independent variables used in the study. The Response Surface Model (RSM) was used to optimize and describe the interdependencies of the different variables. The method was evaluated using the Box-Behnken design (BBD). A second-order polynomial model was used to properly understand the experimental results, and the effectiveness of the chosen model was verified by the strong agreement in determination coefficient values. ZrO2 NPs also exhibit good antibacterial activity on Gram-negative Klebsiella pneumoniae and Gram-positive bacteria, Bacillus subtilis.

Pulsed ultrasound combined with microbubbles use can disrupt the blood–brain barrier (BBB) temporarily; this technique opens a temporal window to deliver large therapeutic molecules into brain tissue. There are published studies to discuss the efficacy and safety of the different ultrasound parameters, microbubble dosages and sizes, and sonication schemes on BBB disruption, but optimal the paradigm is still under investigation. Our study is aimed to investigate how different sonication parameters, time, and microbubble dose can affect BBB disruption, the dynamics of BBB disruption, and the efficacy of different sonication schemes on BBB disruption. Method: We used pulsed weakly focused ultrasound to open the BBB of C57/B6 mice. Evans blue dye (EBD) was used to determine the degree of BBB disruption. With a given acoustic pressure of 0.56 MPa and pulse repetitive frequency of 1 Hz, burst lengths of 10 ms to 50 ms, microbubbles of 100 μL/kg to 300 μL/kg, and sonication times of 60 s to 150 s were used to open the BBB for parameter study. Brain EBD accumulation was measured at 1, 4, and 24 h after sonication for the time–response relationship study; EBD of 100 mg/kg to 200 mg/kg was administered for the dose–response relationship study; EBD injection 0 to 6 h after sonication was performed for the BBB disruption dynamic study; brain EBD accumulation induced by one sonication and two sonications was investigated to study the effectiveness on BBB disruption; and a histology study was performed for brain tissue damage evaluation. Results: Pulsed weakly focused ultrasound opens the BBB extensively. Longer burst lengths and a larger microbubble dose result in a higher degree of BBB disruption; a sonication time longer than 60 s did not increase BBB disruption; brain EBD accumulation peaks 1 h after sonication and remains 81% of the peak level 24 h after sonication; the EBD dose administered correlates with brain EBD accumulation; BBB disruption decreases as time goes on after sonication and lasts for 6 h at least; and brain EBD accumulation induced by two sonication increases 74.8% of that induced by one sonication. There was limited adverse effects associated with sonication, including petechial hemorrhages and mild neuronal degeneration. Conclusions: BBB can be opened extensively and reversibly by pulsed weakly focused ultrasound with limited brain tissue damage. Since EBD combines with albumin in plasma to form a conjugate of 83 kDa, these results may simulate ultrasound-induced brain delivery of therapeutic molecules of this size scale. Background and objective: Leaves of plant Dolichandrone falcata Seem. Are traditionally used by the healers of “India” for the treatment of piles and diabetes. Therefore, the main objective of the present investigation was to evaluate the anti-hemorrhoidal potential of ethanolic leaf extract from the plant D. falcata (EDF). Methods: D. falcata leaf extract obtained after maceration process was initially subjected to phytochemical standardization with the help of HPLC using rutin as a marker compound. Hemorrhoid was induced in the rats using croton oil rat model, and the treatment was carried out with EDF suspension prepared using carboxymethyl cellulose for 5 days using 100, 200 and 300 mg/kg p. o. The scoring of inflammation was calculated on the basis of the clinical hemorrhoidal area and severity index, which was followed by estimating the level of Evans blue dye as an inflammatory marker. The hemorrhoidal tissues were also subjected to cytokine profiling, biochemical evaluation and histopathology. Results: EDF demonstrated the presence of mainly polyphenols, tannins, flavonoids, alkaloids, and saponins, while rutin was calculated to be 3.08% w/w in EDF following HPLC analysis. The results depicted more potent anti-hemorrhoidal activity of EDF at 200 mg/kg orally which was evident through the grading of inflammatory index. The result demonstrated significant decline in the levels of IL-1β, IL-6, TNF-α expression and also showed restoration of altered antioxidants and liver enzymes. Histopathological study showed minimal inflammation and reduced dilated blood vessels in treated animals, thereby confirming the tissue recovery. Conclusion: Thus, we have successfully explored and substantiated the traditional use of the leaves from the plant D. falcata for the first time in the treatment of hemorrhoids, which may be attributed to its anti-inflammatory and its antioxidant potential.

Abstract Background Plasma volume expansion is an important physiologic change across gestation. High or low expansion has been related to adverse pregnancy outcomes, yet there is a limited understanding of normal/healthy plasma volume expansion. We aimed to evaluate the pattern of plasma volume expansion across healthy pregnancies from longitudinal studies. Methods We conducted a systematic review and meta-analysis to identify original studies that measured plasma volume in singleton pregnancies of healthy women. Specifically, we included studies that measured plasma volume at least two times across gestation and one time before or after pregnancy in the same women. PubMed, Web of Science, Cochrane, CINAHL, and clinicaltrials.gov databases were searched from the beginning of each database to February 2019. We combined data across studies using a random effects model. Results Ten observational studies with a total of 347 pregnancies were eligible. Plasma volume increased by 6% (95% CI 3–9) in the first trimester compared to the nonpregnant state. In the second trimester, plasma volume was increased by 18% (95% CI 12–24) in gestational weeks 14–20 and 29% (95% CI 21–36) in weeks 21–27 above the nonpregnant state. In the third trimester, plasma volume was increased by 42% (95% CI 38–46) in weeks 28–34 and 48% (95% CI 44–51) in weeks 35–38. The highest rate of increase occurred in the first half of the second trimester. Included studies were rated from moderate to high quality; 7 out of 10 studies were conducted over 30 years ago. Conclusions In healthy pregnancies, plasma volume begins to expand in the first trimester, has the steepest rate of increase in the second trimester, and peaks late in the third trimester. The patterns observed from these studies may not reflect the current population, partly due to the changes in BMI over the last several decades. Additional longitudinal studies are needed to better characterize the range of normal plasma volume expansion across maternal characteristics.

Fe3O4@catechol formaldehyde resin coated @Graphene Oxide nanocomposite (Fe3O4@CFR@GO) and Fe3O4@catechol formaldehyde resin coated @TiO2 (Fe3O4@CFR@TiO2) nanocomposite were fabricated by hydrothermal method. Particularly, catechol bunches on the highest layer of nanospheres to play a mussel-inspired chemistry to assist combined with graphene oxide (GO) to wrap the Fe3O4@ coated nanosphere. The prepared catalyst was proven to be very efficient with less than a minute and vey less dosage (15–17 mg) in the adsorptive degradation of Evans blue dye. The adsorptive degradation of Evans blue dye with Fe3O4@CFR@GO and Fe3O4@CFR@TiO2 nanocomposites are studied by several variables like the dye concentration, dosage, pH, contact time and temperature. It shows maximum adsorption capacity of 0.1435 mg/g (Fe3O4@CFR@GO) and 9.345 mg/g (Fe3O4@CFR@TiO2) nanocomposites. The equilibrium concentration and the adsorption capacity were evaluated using three different isothermal models. The kinetic study determined that Evans blue dye adsorption was in good analogy with the pseudo-first-order kinetic model.

Abstract Introduction Although the hippocampus (HIP) is thought impermeable to blood‐borne proteins because of the integrity of the blood–brain barrier (BBB), it was recently suggested to be susceptible to hydrophilic hormones. The present study determined the accessibility of blood‐borne signal molecules such as hormones to hippocampal neurons in physiologically normal rats. Methods As a probe for accessibility, Evans blue dye (EB) that rapidly binds to albumin (Alb), which is impermeable to the BBB, was injected intravenously. To increase the vascular permeability of the BBB, a daily single administration of angiotensin II (Ang II) was applied intravenously for seven consecutive days. Results Fifteen minutes after the injection of EB, histological observation revealed that a number of neurons had entrapped and accumulated EB into their cell bodies in the hippocampal dentate gyrus in all rats. Of these, relatively large oval neurons (>15 µm) in the hilus and molecular layer showed parvalbumin immunopositivity, indicating they are GABAergic interneurons. The population of EB‐accumulating neurons (approximately 10 µm) were localized in the inner margin of the granule cell layer, suggesting they were granule cells. However, the number of EB‐positive neurons did not change in rats treated with Ang II compared with vehicle injection. Conclusions These findings suggest an intriguing possibility that blood‐derived proteins such as hormones have access to hippocampal neurons constitutively in the absence of stimuli that increase the vascular permeability of the BBB in a physiologically normal state.

Purpose: AL2106 is a new medical device based on a mixture of chondroitin sulphate in a xyloglucan and glycerol solution made to maximize its bioadhesive capability to the esophageal mucosa. The aim of the present study was twofold to evaluate the AL2106 protective effect on the esophageal mucosa when exposed to an acidic solution mimicking gastric reflux and to assess the resilience of this effect to saline washing. Materials and Methods: A porcine ex vivo model was used and the effects of the new medical device were compared to a sodium alginate suspension (SAS) already present on the market which was assumed as reference. Mucosal damage was induced in 19 porcine esophagi by perfusion with an acidic solution added with pepsin, and Evans blue dye (EBD) tissue uptake was used as an indicator of mucosal permeability. The EBD penetration, expressed as EBD μg/g of dry tissue, was assessed in specimens of untreated damaged mucosa and in specimens treated with AL2106 or SAS. The same evaluation was carried out after washing with normal saline. Results: Both topical agents tested significantly reduced the EBD uptake by more than 60% (AL2106 8.4± 4.5, SAS 3.6± 2.7 vs control 23.2± 13.1, p< 0.01). The saline washing did not cause any significant reduction in the protective effect of AL2106 (8.6± 5.9), while it significantly reduced that of SAS (5.9± 4.3, p< 0.05). Conclusion: The new AL2106 medical device showed a good barrier effect against a reflux-like damaging solution and preserved this effect after the mucosal washing test, thus suggesting its possible relevance for the treatment of gastroesophageal reflux disease. Objective: To prove whether the cannabinoid 2 receptor agonist has an anti-inflammatory effect in the model of Ischemia/Reperfusion (I/R) injury. Methods: Intestinal ischemia and reperfusion model was created in rats. The cannabinoid 2 receptor agonist (AM-1241) was given through the abdominal vein (iv) just before creating ischemia and reperfusion. Afterwards, evans blue was applied iv. The transition of evans blue to the tissues was seen with the naked eye. Immediately after this stage, the systemic blood circulation pool was emptied properly. Tissues were incubated at formamide for 48 hours after weighing, and measurements were made at a wavelength of 620 nm on a spectrophotometer. Results: About 803% evans blue escape was observed in the I/R group compared to the sham control group. The difference between the I/R and the I/R+CB2 agonist is that the agonist holds proteins and reduces the transition of protein and evans blue to tissue fluid. Conclusions: The cannabinoid 2 receptor agonist was found to inhibit the escape of protein from both the ileum and the distant organ (lung) capillaries, and thus showed antiinflammatory effect in the ileum I/R injury. In this work, Norway spruce bark was used as a precursor to prepare activated biochars (BCs) via chemical activation with potassium hydroxide (KOH) as a chemical activator. A Box–Behnken design (BBD) was conducted to evaluate and identify the optimal conditions to reach high specific surface area and high mass yield of BC samples. The studied BC preparation parameters and their levels were as follows: pyrolysis temperature (700, 800, and 900 °C), holding time (1, 2, and 3 h), and ratio of the biomass: chemical activator of 1: 1, 1.5, and 2. The planned BBD yielded BC with extremely high SSA values, up to 2209 m2·g−1. In addition, the BCs were physiochemically characterized, and the results indicated that the BCs exhibited disordered carbon structures and presented a high quantity of O-bearing functional groups on their surfaces, which might improve their adsorption performance towards organic pollutant removal. The BC with the highest SSA value was then employed as an adsorbent to remove Evans blue dye (EB) and colorful effluents. The kinetic study followed a general-order (GO) model, as the most suitable model to describe the experimental data, while the Redlich–Peterson model fitted the equilibrium data better. The EB adsorption capacity was 396.1 mg·g−1. The employment of the BC in the treatment of synthetic effluents, with several dyes and other organic and inorganic compounds, returned a high percentage of removal degree up to 87.7%. Desorption and cyclability tests showed that the biochar can be efficiently regenerated, maintaining an adsorption capacity of 75% after 4 adsorption–desorption cycles. The results of this work pointed out that Norway spruce bark indeed is a promising precursor for producing biochars with very promising properties.

The treatment of CNS disorders suffers from the inability to deliver large therapeutic agents to the brain parenchyma due to protection from the blood-brain barrier (BBB). Herein, we investigated high-frequency pulsed electric field (HF-PEF) therapy of various pulse widths and interphase delays for BBB disruption while selectively minimizing cell ablation. Eighteen male Fisher rats underwent craniectomy procedures and two blunt-tipped electrodes were advanced into the brain for pulsing. BBB disruption was verified with contrast T1W MRI and pathologically with Evans blue dye. High-frequency irreversible electroporation cell death of healthy rodent astrocytes was investigated in vitro using a collagen hydrogel tissue mimic. Numerical analysis was conducted to determine the electric fields in which BBB disruption and cell ablation occur. Differences between the BBB disruption and ablation thresholds for each waveform are as follows: 2-2-2 μs (1028 V/cm), 5-2-5 μs (721 V/cm), 10-1-10 μs (547 V/cm), 2-5-2 μs (1043 V/cm), and 5-5-5 μs (751 V/cm). These data suggest that HF-PEFs can be fine-tuned to modulate the extent of cell death while maximizing peri-ablative BBB disruption. Furthermore, numerical modeling elucidated the diffuse field gradients of a single-needle grounding pad configuration to favor large-volume BBB disruption, while the monopolar probe configuration is more amenable to ablation and reversible electroporation effects.

Abstract—The Miles assay using Evans Blue dye is a conventional method to assess vascular permeability. The penetration Evans dye into intestinal tissue was studied in the early reperfusion phase of ischemia–reperfusion injury to the small intestine. Significant destruction of villus microvessels and an increase in vascular permeability to plasma proteins were observed in the early reperfusion phase, impairing the blood flow at the microvasculature level based on evidence from histology and laser Doppler flowmetry. Evans Blue accumulated to the highest concentration in intestinal tissue after being injected into the bloodstream prior to 60-min ischemia; the lowest level was detected when the dye was injected after 15-min reperfusion. Changes in the dye content in intestinal tissue therefore depended on the number of vessels that remain intact at the time of dye injection at various stages of ischemia–reperfusion. Evans Blue observed in tissue was assumed to reflect the degree of vessel preservation in a study of ischemia–reperfusion injury of the small intestine, rather than reporting the degree of vascular permeability, as commonly believed. Electrical skin measurements at acupuncture points (acupoints) have been utilized as a diagnostic and therapeutic aid for more than 50 years. Although acupoints are described as having distinct electrical properties, such as high conductance and low impedance, the underlying mechanisms are currently unknown. The present study investigated in a rat model of hypertension whether the high conductance at acupoints is a result of the release of the neuropeptides substance P (SP) and calcitonin gene-related peptide (CGRP) during neurogenic inflammation in the referred pain area. When plasma extravasation from neurogenic inflammation was examined by exploring the leakage of intravenously injected Evans blue dye (EBD) to the skin, extravasated EBD was found most frequently in acupoints on the wrist. The increased conductance and temperature at these acupoints occurred during the development of hypertension. The increase in conductance and plasma extravasation at acupoints in hypertensive rats was ablated by cutting median and ulnar nerves, blocking small diameter afferent fibers with resiniferatoxin (RTX) injection into median and ulnar nerves, or antagonizing SP or CGRP receptors in acupoints. In turn, intradermal injection of SP or CGRP resulted in increased conductance and plasma extravasation in naïve rats. Elevated levels of SP and CGRP were found in the acupoints of hypertensive rats. These findings suggest that the high conductance at acupoints is due to vascular leakage following local release of SP and CGRP during neurogenic inflammation.

The effects of light‐driven enhancement of Evans Blue dye complexes with blood plasma proteins were observed for the first time, both in vitro and in vivo. The possible background of the effect concerns the photochemical cis‐trans isomerization of the azo dye molecules. The effect was induced in the solution with a red laser with a wavelength of 638 nm, which corresponds to the peak of the dye absorption. The lifetime of the enhanced fluorescence is approximately 1 second and enables its use as an optically tagged molecular flow tracer for blood flow velocity measurements. Utilizing the effect, we performed for the first time the intravital molecular tagging velocimetry of the blood velocity in blood vessels in a living animal. Ischemic edema can alter the structure and permeability of the blood-brain barrier. Recent studies have reported that progesterone reduces cerebral edema after cerebral ischemia. However, the underlying mechanism of this effect has not yet been elucidated. In the present study, progesterone effectively reduced Evans blue extravasation in the ischemic penumbra, but not in the ischemic core, 48 hours after cerebral ischemia in rats. Progesterone also inhibited the down-regulation of gene and protein levels of occludin and zonula occludens-1 in the penumbra. These results indicate that progesterone may effectively inhibit the down-regulation of tight junctions, thereby maintaining the integrity of the blood-brain barrier and reducing cerebral edema.

Background: Blood volume measurement though important in management of critically ill-patients is not routinely estimated in clinical practice owing to labour intensive, intricate and time consuming nature of existing methods. Aims: The aim was to compare blood volume estimations using trivalent chromium [51 Cr(III)] and standard Evans blue dye (EBD) method in New Zealand white rabbit models and establish correction-factor (CF). Materials and Methods: Blood volume estimation in 33 rabbits was carried out using EBD method and concentration determined using spectrophotometric assay followed by blood volume estimation using direct injection of 51Cr(III). Twenty out of 33 rabbits were used to find CF by dividing blood volume estimation using EBD with blood volume estimation using 51Cr(III). CF is validated in 13 rabbits by multiplying it with blood volume estimation values obtained using 51Cr(III). Results: The mean circulating blood volume of 33 rabbits using EBD was 142.02 ± 22.77 ml or 65.76 ± 9.31 ml/kg and using 51Cr(III) was estimated to be 195.66 ± 47.30 ml or 89.81 ± 17.88 ml/kg. The CF was found to be 0.77. The mean blood volume of 13 rabbits measured using EBD was 139.54 ± 27.19 ml or 66.33 ± 8.26 ml/kg and using 51Cr(III) with CF was 152.73 ± 46.25 ml or 71.87 ± 13.81 ml/kg (P = 0.11). Conclusions: The estimation of blood volume using 51Cr(III) was comparable to standard EBD method using CF. With further research in this direction, we envisage human blood volume estimation using 51Cr(III) to find its application in acute clinical settings.

In a 2 × 2 factorial design, 60 male Ross-308 broilers were fed either a control or 1 g/kg betaine diet and housed under thermoneutral (TN) or heat stress (HS) conditions. Broilers were acclimated to diets for 1 week under TN (25 °C), then either kept at TN or HS, where the temperature increased 8 h/day at 33 °C and 16 h/day at 25 °C for up to 10 days. Respiration rate (RR) was measured at four time points, and on each of 1, 2, 3, 7 and 10 days of HS, 12 broilers were injected with 0.5 mg/kg of Evans Blue Dye (EBD) solution to quantify regional changes in tissue damage. Betaine was quantified in tissues, and ileal damage was assessed via morphometry and transepithelial resistance (TER). Heat stress elevated RR (p < 0.001) and resulted in reduced villous height (p = 0.009) and TER (p < 0.001), while dietary betaine lowered RR during HS (p < 0.001), increased betaine distribution into tissues, and improved ileal villous height (p < 0.001) and TER (p = 0.006). Heat stress increased EBD in the muscle and kidney of chickens fed the control diet but not in those receiving betaine. Overall, these data indicate that supplemented betaine is distributed to vital organs and the gastrointestinal tract, where it is associated with improved tolerance of HS. Furthermore, EBD markers help reveal the effects of HS on organs dysfunction.

Objective: The popularity of contemporary microsurgical techniques in treatment of lower-limb lymphedema calls for better visualization of the lymphatic system, both preoperatively and intra-operatively. The aim of this prospective study was to investigate the feasibility of a novel combination of Ga-NEB positron emission tomography (PET) with magnetic resonance lymphography (MRL) in evaluating lymphedema and guiding surgical intervention. Methods: A total of 11 patients (F 9, M 2, age range 29-69 y) with lower-limb lymphedema classified into stage I to III were recruited. PET acquisition was performed at 30, 60 and 90 min after subcutaneous injection of the albumin-binding radiotracer Ga-NEB into the bilateral first web spaces of the feet. All the patients were also subjected to Tc-sulfur colloid (SC) lymphoscintigraphy for comparison. Gd-DTPA-enhanced magnetic resonance imaging (MRI) was performed using sequences specialized for lymphatic vessel scans. All the patients underwent surgical interventions within a week. The surgical approach includes the use of a linear marker for edema localization and indocyanine green (ICG) lymphography with a near-infrared surgical navigation system intra-operatively. Results: Lymph transport in lymphatic channels was clearly observed by visualization of Ga-NEB activity in the lymphatic vessels and within lymph nodes for all 11 patients as well as the visualization of the edema section plane with dermal backflow (DB), abnormally increased and disconnected uptake along the lymphatic channels. Preoperative Ga-NEB PET combined with MRL provides advantageous three-dimensional images, higher temporal resolution, significantly shorter time lapse before image acquisition after tracer injection and more accurate pathological lymphatic vessel distribution than Tc-SC lymphoscintigraphy combined with MRI. Conclusion: This study documented an effective imaging pattern to combine Ga-NEB PET and MRL in patients with lower-limb lymphedema. This strategy demonstrated significant advantage over Tc-SC lymphoscintigraphy/MRL in the evaluation of lymphedema severity, staging and pathological location of lymph vessels to make an individualized treatment plan. Aims In the present study, a novel mice model of chronic kidney disease (CKD) was developed, and psycho-motor behavioural abnormalities, blood-brain barrier (BBB) integrity and brain histology were studied. Main methods Swiss albino female mice were given high adenine diet (0.3% w/w mixed with feed) for 4 weeks. Serum urea and creatinine levels and renal histological studies were performed to validate the model. Psycho-motor behavioural abnormalities and neurological severity were studied. BBB integrity was assessed using Evans blue extravasation method. Nissl staining was performed to see possible morphological aberrations in brain. Key findings There was a significant increase in serum urea and creatinine levels in mice given high adenine diet, and the mice had abnormal kidney morphology. Deposition of adenine and 2,8-dihydroxyadenine crystals, and increased collagen deposits in the renal tissues were found, which validate induction of CKD in the mice. Motor behavioural abnormalities, depression-like and anxiolytic behaviour and increase in neurological severity were prevalent in mice with CKD. Evans Blue dye extravasation was found to occur in the brain, which signifies disruption of BBB. However, Nissl staining did not reveal any morphological aberration in brain tissue. Significance The present study puts forward a highly reproducible mice model of CKD validated with serum parameters and renal histopathological changes. The mice showed psycho-motor behavioural abnormalities and BBB disruption. Background and aims Transport of macromolecules between plasma and the arterial wall plays a key role in atherogenesis. Scattered hotspots of elevated endothelial permeability to macromolecules occur in the aorta; a fraction of them are associated with dividing cells. Hotspots occur particularly frequently downstream of branch points, where lesions develop in young rabbits and children. However, the pattern of lesions varies with age, and can be explained by similar variation in the pattern of macromolecule uptake. We investigated whether patterns of hotspots and mitosis also change with age. Methods Evans’ Blue dye-labeled albumin was injected intravenously into immature or mature rabbits and its subsequent distribution in the aortic wall around intercostal branch ostia examined by confocal microscopy and automated image analysis. Mitosis was detected by immunofluorescence after adding 5-bromo-2-deoxiuridine to drinking water. Results Hotspots were most frequent downstream of branches in immature rabbits, but a novel distribution was observed in mature rabbits. Neither pattern was explained by mitosis. Hotspot uptake correlated spatially with the much greater non-hotspot uptake ( p < 0.05), and the same pattern was seen when only the largest hotspots were considered. Conclusions The pattern of hotspots changes with age. The data are consistent with there being a continuum of local permeabilities rather than two distinct mechanisms. The distribution of the dye, which binds to elastin and collagen, was similar to that of non-binding tracers and to lesions apart from a paucity at the lateral margins of branches that can be explained by lower levels of fibrous proteins in those regions. Background Systemic inflammation and oxidative stress are crucial in mediating blood-brain barrier (BBB) integrity loss during sepsis. Simvastatin possess potent anti-inflammation and antioxidation capacity. We sought to elucidate whether an acute bolus of simvastatin could mitigate BBB integrity loss in a rodent model of polymicrobial sepsis. Methods A total of 96 adult male rats (200-250 g) were randomized to receive cecal ligation and puncture (CLP), CLP plus simvastatin, sham operation, or sham operation plus simvastatin (n = 24 in each group). After maintaining for 24 h, BBB integrity in the surviving rats was determined. Results CLP significantly induced BBB integrity loss, as grading of Evans blue staining of the brains, BBB permeability to Evans blue dye, and brain edema levels in rats receiving CLP were significantly higher than those receiving sham operation. In contrast, grading of Evans blue staining (P = 0.020), BBB permeability to Evans blue dye (P = 0.031), and brain edema levels (P = 0.009) in rats receiving CLP plus simvastatin were significantly lower than those receiving CLP alone. Tight junction proteins claudin-3 and claudin-5 in endothelial cells are major structural components of BBB. Our data revealed that concentrations of claudin-3 and claudin-5 in rats receiving CLP were significantly lower than those receiving CLP plus simvastatin (P = 0.010 and 0.007). Immunohistochemistry further revealed significant fragmentation of claudin-3 and claudin-5 in rats receiving CLP. Moreover, levels of claudin-3 and claudin-5 fragmentation in rats receiving CLP plus simvastatin were significantly lower than those receiving CLP. Exercise involving lengthening contraction (LC) often results in delayed myofiber damage and functional deficits over the ensuing days. The present study examined whether the stretch speed of LC is a determinant of damage severity. Under isoflurane anesthesia, LC was repeatedly induced in rat ankle extensor muscles at different stretch speeds (angular velocities of 50, 100, 200, and 400 deg/sec) over a fixed stretch range of motion (90°). The number of muscle fibers labeled with Evans blue dye, a marker of muscle fiber damage associated with increased membrane permeability, increased with the angular velocity of LC (by 20% of all myofibers at 400 deg/sec). Muscle fibers with cross-sectional areas in the range of 3600-4800 μm2, corresponding to type IIb fiber size, exhibited the most severe damage as revealed by the largest decrease in the number of fibers 3 days after LC at 200 deg/sec, suggesting that muscle damage occurred preferentially in type IIb myofibers. Isometric torque of dorsiflexion measured 2 days after LC decreased progressively with LC angular velocity (by 68% reduction at 400 deg/sec). The angular velocity of muscle stretch during LC is thus a critical determinant of the degree of damage, and LC appears to damage type IIb fibers preferentially, resulting in a disproportionate reduction in isometric torque. Diabetic nephropathy is a major cause of end-stage renal disease worldwide. The current studies were performed to determine the later stages of the progression of renal disease in type II diabetic mice (BKS; db/db). Methodology was developed for determining glomerular filtration rate (GFR) in conscious, chronically instrumented mice using continuous intravenous infusion of FITC-labeled inulin to achieve a steady-state plasma inulin concentration. Obese diabetic mice exhibited increased GFR compared with control mice. GFR averaged 0.313 ± 0.018 and 0.278 ± 0.007 ml/min in 18-wk-old obese diabetic (n = 11) and control (n = 13) mice, respectively (P < 0.05). In 28-wk-old obese diabetic (n = 10) and control (n = 15) mice, GFR averaged 0.348 ± 0.030 and 0.279 ± 0.009 ml/min, respectively (P < 0.05). GFR expressed per gram BW was significantly reduced in 18- and 28-wk-old obese diabetic compared with control mice (5.9 ± 0.3 vs. 9.0 ± 0.3; 6.6 ± 0.6 vs. 7.8 ± 0.3 μl·min−1·g body wt−1), respectively (P < 0.05). However, older nonobese type II diabetic mice had significantly reduced GFR (0.179 ± 0.023 ml/min; n = 6) and elevated urinary albumin excretion (811 ± 127 μg/day) compared with obese diabetic and control mice (514 ± 54, 171 ± 18 μg/day), which are consistent with the advanced stages of renal disease. Abstract: Entry of most compounds into the CNS is impeded by the blood–brain barrier (BBB). Because vascular endothelial growth factor (VEGF) is important in the formation and maintenance of the BBB and is known to modulate BBB permeability in newborn rodents, we tested the hypothesis that VEGF may enhance BBB permeability in adult mice. We examined the effect of VEGF on the CNS distribution of three different agents: a small molecule (Evans blue dye) that is known to bind plasma proteins, an exogenous protein (tetanus toxin fragment C; TTC), and a viral vector (recombinant adeno-associated virus serotype 2/5 marked with lacZ; rAAV2/5-lacZ). Pretreatment with VEGF (20 μg; i.v.) increased permeability of the BBB to Evans blue dye and TTC as detected by augmented concentrations of these substances in the cerebrum, brainstem, and spinal cord. By contrast, VEGF did not alter BBB permeability to AAV2/5-lacZ, as defined by β-galactosidase activity assay. These data demonstrate the potential utility of VEGF for pharmacological modulation of the BBB, and indicate that the increase in BBB permeability mediated by VEGF is limited by the size of the delivered substance. Isehemia-reperfusion (I/R) lung injury is characterized by increased pulmonary endothelial permeability and edema, but the genetic basis for this injury is unknown. We utilized an in vivo mouse preparation of unilateral lung I/R to evaluate the genetic determinants of I/R lung injury. An index of pulmonary vascular protein permeability was measured by the ratio of left-to-right lung Evans blue dye of eight inbred mouse strains after 30 min of left lung isehemia and 150 min of reperfusion. The order of strain-specific sensitivity to I/R lung injury was BALB/c < SJL/J < CBA/J < C57BL/6J < 129/J < A/J < C3H/H3J < SWR/J. The reciprocal FI offspring of the BALB/c and SWR/J progenitor strains had intermediate phenotypes but a differing variance. A similar pattern of right lung Evans blue dye content suggested the presence of contralateral injury because baseline vascular permeability was not different. Lung I/R injury was attenuated by NADPH oxidase inhibition, indicating a role for NADPH oxidase-derived reactive oxygen species (ROS). There was no strain-dependent difference in lung NADPH oxidase expression. Strain-related differences in zymosan-stimulated neutrophil ROS production did not correlate with I/R lung injury in that neutrophil ROS production in SWR/J mice was greater than C57BL/6J but not different from BALB/c mice. These data indicate the presence of a genetic sensitivity to lung I/R injury that involves multiple genes including a maternal-related factor. We have shown that left pulmonary artery ligation (LPAL) in mice causes a prompt angiogenic response, with new systemic vessels from inter-costal arteries penetrating the pleura within 6 days. Because angiogenic vessels in other organs have been shown to exhibit increased permeability, we studied vascular permeability (Evans blue dye extravasation, lung wet weight-to-dry weight ratio, and lavaged protein) in naive C57BL/6 mice and 4 h, and 14 and 21 days after LPAL (4–6 mice/time point). We also measured radiolabel clearance as an index of functional perfusion after LPAL. Tracer clearance from the left lung was maximal by 6 days after LPAL and not different from right lungs. Thus a functional vasculature is established before 6 days of LPAL that results in normal tracer clearance. By 21 days after LPAL, Evans blue-albumin was significantly increased in the left lung relative to both 4 h (no vasculature) and 14 days after LPAL. Only after 21 days of LPAL was left lung wet weight-to-dry weight ratio significantly different from naive lungs. Additionally, lavaged protein was significantly increased both 4 h and 21 days after LPAL relative to control mice. Thus, using three different methods, results consistently demonstrated increased permeability to protein and water 21 days after LPAL. Although changes in surface area of perfusion might affect the interpretation of these results, blood flow measured with labeled microspheres indicated no change in left lung perfusion between 14 and 21 days of LPAL. Abstract: Hypercholesterolemia is associated with increased cardiovascular mortality and is known to promote the advancement of atherosclerotic lesions in experimental animal models. Juvenile swine were fed a normal or high-cholesterol diet, and the transendothelial macromolecular permeability of the external iliac arteries of these animals was assessed by measuring the uptake rate of circulating Evans blue dye (EBD). The extent and patterns of lipid-containing lesions were also determined using en face staining with Oil Red O (ORO). Sites of ORO staining often excluded EBD, possibly via the fragmentation of the internal elastic lamina, to which EBD binds. By spatially averaging the EBD uptake in arterial segments relatively free of ORO-positive lesions, it was found that endothelial permeability to albumin was greater in hypercholesterolemic pigs than in those on a normal diet (p =0.056). The volume of red blood cells (VRBC) is used routinely in the diagnostic workup of polycythemia, in assessing the efficacy of erythropoietin administration, and to study factors affecting oxygen transport. However, errors of various methods of measurement of VRBC and related parameters are not well characterized. We meta-analyzed 346 estimates of error of measurement of VRBC for techniques based on Evans blue (VRBC,Evans), 51chromium-labeled red blood cells (VRBC,51Cr), and carbon monoxide (CO) rebreathing (VRBC,CO), as well as hemoglobin mass with the carbon-monoxide method (MHb,CO), in athletes and active and inactive subjects undergoing various experimental and control treatments lasting minutes to months. Subject characteristics and experimental treatments had little effect on error of measurement, but measures with the smallest error showed some increase in error with increasing time between trials. Adjusted to 1 day between trials and expressed as coefficients of variation, mean errors for MHb,CO (2.2%; 90% confidence interval 1.4-3.5%) and VRBC,51Cr (2.8%; 2.4-3.2%) were much less than those for VRBC,Evans (6.7%; 4.9-9.4%) and VRBC,CO (6.7%; 3.4-14%). Most of the error of VRBC,Evans was due to error in measurement of volume of plasma via Evans blue dye (6.0%; 4.57.8%), which is the basis of VRBC,Evans. Most of the error in VRBC,CO was due to estimates from laboratories with a relatively large error in MHb,CO, the basis of VRBC,51Cr and MHb,CO are the best measures for research on blood-related changes in oxygen transport. A better understanding of how hemodynamic factors affect the integrity and function of the vascular endothelium is necessary to appreciate more fully how atherosclerosis is initiated and promoted. A novel technique is presented to assess the relation between fluid dynamic variables and the permeability of the endothelium to macromolecules. Fully anesthetized, domestic swine were intravenously injected with the albumin marker Evans blue dye, which was allowed to circulate for 90 min. After the animals were euthanized, silicone casts were made of the abdominal aorta and its lilac branches. Pulsatile flow calculations were subsequently made in computational regions derived from the casts. The distribution of the calculated time-dependent wall shear stress in the external iliac branches was directly compared on a point-by-point basis with the spatially varying in vivo uptake of Evans blue dye in the same arteries. The results indicate that in vivo endothelial permeability to albumin decreases with increasing time-average shear stress over the normal range. Plasma extravasation (PE) was measured in adult Wistar rats by injecting Evans blue dye (EB) (20 mg kg–1) intravenously in the absence or presence of human urotensin II (U-II) (0.1–10 nmol kg–1). A consistent increase of PE was observed in specific organs (e.g., aorta, from 28.1 ± 2.4 to 74.6 ± 3.6 µg EB g–1 dry tissue; P < 0.001) after an administration of 4.0 nmol kg–1 (a preselected optimal dose) of U-II. The effects of U-II (4.0 nmol kg–1) were compared with those of endothelin-1 (ET-1) (1.0 nmol kg–1). In the thoracic aorta and pancreas, U-II was active, while ET-1 was not. The two agents were equivalent in the heart and kidney, whereas, in the duodenum, ET-1 was more active than U-II. Increases of plasma extravasation induced by U-II, but not by ET-1, were reduced after treatment with [Orn8]U-II (0.3 µmol kg–1). This latter antagonist did not show any significant residual agonistic activity in vivo in the rat. Other specific receptor antagonists for ET-1, such as BQ-123 (endothelin type A (ETA) receptor) and BQ-788 (endothelin type B (ETB) receptor), and for the platelet activating factor (PAF), such as BN50730, failed to modify the action of U-II. The present study is the first report describing the modulator roles of U-II on vascular permeability in specific organs. The genetic defect in the cardiomyopathic (CM) hamster is a mutation in the glycoprotein-sarcoglycan (a component of the dystrophin-glycoprotein complex). Apoptosis has been identified in skeletal muscle of dystrophin-deficient mice, and therefore the role of myocardial apoptosis in relation to oncosis in causing myocardial necrosis was assessed at the onset of left ventricular (LV) dysfunction in CM hamsters. LV size and function were evaluated in normal and CM hamsters (CHF147 line) by echocardiography at 1, 2, 3, and 5 months (mo) of age. The decrease of LV fractional shortening was found to be most marked (45 %) between 1 and 2 mo of age. Apoptotic nuclei were identified at each time point using in situ end-labeling of DNA strand breaks (TUNEL), together with immunolabeling of myocytes; DNA fragmentation (laddering) and nuclear morphology were also assessed. Myocyte oncotic necrosis was assessed at 2 mo by Evans blue dye (EBD), wheat germ agglutinin, hematoxylin/eosin staining, and electron microscopy. Apoptotic nuclei were not detected in age-matched normal hamsters. In the CM hamsters apoptotic myocyte nuclei comprised an average of 0.041 % of myocyte nuclei between 1 and 5 mo, an increase at 2 mo (to 0.076 %) was not significant, and DNA laddering was not detected. The number of myocyte nuclei per unit area decreased by 32 % between 1 and 2 mo, and in 2 mo old CM hamsters myocardial staining with EBD was positive in 9.82 % of the myocardial cross sectional areas examined, most of which was consistent with sarcolemmal rupture and oncosis with inflammatory cell infiltration. It is concluded that myocyte oncosis provides the major mechanism for the decreased number of myocyte nuclei and the early decrease of cardiac function between 1 and 2 mo of age in the CM hamster, with only a small contribution of myocyte apoptosis. In this work, Norway spruce bark was used as a precursor to prepare activated biochars (BCs) via chemical activation with potassium hydroxide (KOH) as a chemical activator. A Box–Behnken design (BBD) was conducted to evaluate and identify the optimal conditions to reach high specific surface area and high mass yield of BC samples. The studied BC preparation parameters and their levels were as follows: pyrolysis temperature (700, 800, and 900 °C), holding time (1, 2, and 3 h), and ratio of the biomass: chemical activator of 1: 1, 1.5, and 2. The planned BBD yielded BC with extremely high SSA values, up to 2209 m2·g−1. In addition, the BCs were physiochemically characterized, and the results indicated that the BCs exhibited disordered carbon structures and presented a high quantity of O-bearing functional groups on their surfaces, which might improve their adsorption performance towards organic pollutant removal. The BC with the highest SSA value was then employed as an adsorbent to remove Evans blue dye (EB) and colorful effluents. The kinetic study followed a general-order (GO) model, as the most suitable model to describe the experimental data, while the Redlich–Peterson model fitted the equilibrium data better. The EB adsorption capacity was 396.1 mg·g−1. The employment of the BC in the treatment of synthetic effluents, with several dyes and other organic and inorganic compounds, returned a high percentage of removal degree up to 87.7%. Desorption and cyclability tests showed that the biochar can be efficiently regenerated, maintaining an adsorption capacity of 75% after 4 adsorption–desorption cycles.
