Abstract: PLATO (PLAnetary Transits and Oscillations of stars) is ESA's M3 mission designed to detect and characterise extrasolar planets and perform asteroseismic monitoring of a large number of stars. PLATO will detect small planets (down to <2 R_(Earth)) around bright stars (<11 mag), including terrestrial planets in the habitable zone of solar-like stars. With the complement of radial velocity observations from the ground, planets will be characterised for their radius, mass, and age with high accuracy (5 %, 10 %, 10 % for an Earth-Sun combination respectively). PLATO will provide us with a large-scale catalogue of well-characterised small planets up to intermediate orbital periods, relevant for a meaningful comparison to planet formation theories and to better understand planet evolution. It will make possible comparative exoplanetology to place our Solar System planets in a broader context. In parallel, PLATO will study (host) stars using asteroseismology, allowing us to determine the stellar properties with high accuracy, substantially enhancing our knowledge of stellar structure and evolution. The payload instrument consists of 26 cameras with 12cm aperture each. For at least four years, the mission will perform high-precision photometric measurements. Here we review the science objectives, present PLATO's target samples and fields, provide an overview of expected core science performance as well as a description of the instrument and the mission profile at the beginning of the serial production of the flight cameras. PLATO is scheduled for a launch date end 2026. This overview therefore provides a summary of the mission to the community in preparation of the upcoming operational phases.

Abstract: The discovery of gravitational wave (GW) events and the detection of electromagnetic counterparts from GW170817 has started the era of multimessenger GW astronomy.The field has been developing rapidly and in this paper,we discuss the preparation for detecting these events with the ESA Gaia satellite,during the 4th observing run of the LIGO-Virgo-KAGRA (LVK) collaboration that has started on May 24,2023. Gaia is contributing to the search for GW counterparts by a new transient detection pipeline called GaiaX. In GaiaX, a new source appearing in the field of view of only one of the two telescopes on-board Gaia is sufficient to send out an alert on the possible detection of a new transient. Ahead of O4, an experiment was conducted over a period of about two months. During the two weeks around New Moon in this period of time, the MeerLICHT (ML) telescope located in South Africa tried (weather permitting) to observe the same region of the sky as Gaia within 10 minutes. Any GaiaX detected transient was published publicly. ML and Gaia have similar limiting magnitudes for typical seeing conditions at ML. At the end of the experiment, we had 11861 GaiaX candidate transients and 15806 ML candidate transients, which we further analysed and the results of which are presented in this paper. Abstract: Gaia DR3 contains 1.8 billion sources with G-band photometry, 1.5 billion of which with BP and RP photometry, complemented by positions on the sky, parallax, and proper motion. The median number of field-of-view transits in the three photometric bands is between 40 and 44 measurements per source and covers 34 months of data collection. We pursue a classification of Galactic and extra-galactic objects that are detected as variable by Gaia across the whole sky. Supervised machine learning (eXtreme Gradient Boosting and Random Forest) was employed to generate multi-class, binary, and meta-classifiers that classified variable objects with photometric time series in the G, BP, and RP bands. Classification results comprise 12.4 million sources (selected from a much larger set of potential variable objects) and include about 9 million variable stars classified into 22 variability types in the Milky Way and nearby galaxies such as the Magellanic Clouds and Andromeda, plus thousands of supernova explosions in distant galaxies, 1 million active galactic nuclei, and almost 2.5 million galaxies. The identification of galaxies was made possible by the artificial variability of extended objects as detected by Gaia, so they were published in the galaxy_candidates table of the Gaia DR3 archive, separate from the classifications of genuine variability (in the vari_classifier_result table). The latter contains 24 variability classes or class groups of periodic and non-periodic variables (pulsating, eclipsing, rotating, eruptive, cataclysmic, stochastic, and microlensing), with amplitudes from a few milli-magnitudes to several magnitudes., Comment: 106 pages, 513 figure panels, 4 tables. 4. Gaia-TESS synergy: Improving the identification of transit candidates

Abstract: Context: The TESS team periodically issues a new list of transiting exoplanet candidates based on the analysis of the accumulating light curves obtained by the satellite. The list includes the estimated epochs, periods, and durations of the potential transits. As the point spread function (PSF) of TESS is relatively wide, follow-up photometric observations at higher spatial resolution are required in order to exclude apparent transits that are actually blended background eclipsing binaries (BEBs). Aims: The Gaia space mission, with its growing database of epoch photometry and high angular resolution, enables the production of distinct light curves for all sources included in the TESS PSF, up to the limiting magnitude of Gaia. This paper reports the results of an ongoing Gaia-TESS collaboration that uses the Gaia photometry to facilitate the identification of BEB candidates and even to confirm on-target candidates in some cases. Methods: We inspected the Gaia photometry of the individual sources included in the TESS PSF, searching for periodic dimming events compatible with their ephemerides and uncertainties, as published by TESS. The performance of the search depends mainly on the number of Gaia measurements during transit and their precision. Results: Since February 2021, the collaboration has been able to confirm 126 on-target candidates and exclude 124 as BEBs. Since June 2021, when our search methodology matured, we have been able to identify on the order of 5% as on-target candidates and another 5% as BEBs. Conclusions: This synergistic approach is combining the complementary capabilities of two of the astronomical space missions of NASA and ESA. Abstract: Context: The space telescope Gaia is dedicated mainly to performing high-precision astrometry, but also spectroscopy and epoch photometry which can be used to study various types of photometric variability. One such variability type is exoplanetary transits. The photometric data accumulated so far have finally matured enough to allow the detection of some exoplanets. Aims: In order to fully exploit the scientific potential of Gaia, we search its photometric data for the signatures of exoplanetary transits. Methods: The search relies on a version of the Box-Least-Square (BLS) method, applied to a set of stars prioritized by machine-learning classification methods. An independent photometric validation was obtained using the public full-frame images of TESS. In order to validate the first two candidates, radial-velocity follow-up observations were performed using the spectrograph PEPSI of the Large Binocular Telescope (LBT). Results: The radial-velocity measurements confirm that two of the candidates are indeed hot Jupiters. Thus, they are the first exoplanets detected by Gaia - Gaia-1b and Gaia-2b. Conclusions: Gaia-1b and Gaia-2b demonstrate that the approach presented in this paper is indeed effective. Abstract: Entropy and its various generalizations are important in many fields, including mathematical statistics, communication theory, physics and computer science, for characterizing the amount of information associated with a probability distribution. In this paper we propose goodness-of-fit statistics for the multivariate Student and multivariate Pearson type II distributions, based on the maximum entropy principle and a class of estimators for R\'{e}nyi entropy based on nearest neighbour distances. We prove the L^2-consistency of these statistics using results on the subadditivity of Euclidean functionals on nearest neighbour graphs, and investigate their rate of convergence and asymptotic distribution using Monte Carlo methods. In addition we present a novel iterative method for estimating the shape parameter of the multivariate Student and multivariate Pearson type II distributions.

Abstract: In this paper, we provide the proof of $L^2$ consistency for the $k$th nearest neighbour distance estimator of the Shannon entropy for an arbitrary fixed $k\geq 1.$ We construct the non-parametric test of goodness-of-fit for a class of introduced generalized multivariate Gaussian distributions based on a maximum entropy principle. The theoretical results are followed by numerical studies on simulated samples.

Abstract: In the context of the ESA M5 (medium mission) call we proposed a new satellite mission, Theia, based on relative astrometry and extreme precision to study the motion of very faint objects in the Universe. Theia is primarily designed to study the local dark matter properties, the existence of Earth-like exoplanets in our nearest star systems and the physics of compact objects. Furthermore, about 15 $\%$ of the mission time was dedicated to an open observatory for the wider community to propose complementary science cases. With its unique metrology system and "point and stare" strategy, Theia's precision would have reached the sub micro-arcsecond level. This is about 1000 times better than ESA/Gaia's accuracy for the brightest objects and represents a factor 10-30 improvement for the faintest stars (depending on the exact observational program). In the version submitted to ESA, we proposed an optical (350-1000nm) on-axis TMA telescope. Due to ESA Technology readiness level, the camera's focal plane would have been made of CCD detectors but we anticipated an upgrade with CMOS detectors. Abstract: In this article we present an overview of the ESA Gaia mission and of the unprecedented impact that Gaia will have on the field of variable star research. We summarise the contents and impact of the first Gaia data release on the description of variability phenomena, with particular emphasis on pulsating star research. The Tycho-Gaia astrometric solution, although limited to 2.1 million stars, has been used in many studies related to pulsating stars. Abstract: We present the discovery of stellar tidal tails around the Large and the Small Magellanic Clouds in the Gaia DR1 data. In between the Clouds, their tidal arms are stretched towards each other to form an almost continuous stellar bridge. Our analysis relies on the exquisite quality of the Gaia's photometric catalogue to build detailed star-count maps of the Clouds. We demonstrate that the Gaia DR1 data can be used to detect variable stars across the whole sky, and in particular, RR Lyrae stars in and around the LMC and the SMC. Additionally, we use a combination of Gaia and Gale to follow the distribution of Young Main Sequence stars in the Magellanic System. Viewed by Gaia, the Clouds show unmistakable signs of interaction. Around the LMC, clumps of RR Lyrae are observable as far as ~20 degrees, in agreement with the most recent map of Mira-like stars reported in Deason et al (2016). The SMC's outer stellar density contours show a characteristic S-shape, symptomatic of the on-set of tidal stripping. Beyond several degrees from the center of the dwarf, the Gaia RR Lyrae stars trace the Cloud's trailing arm, extending towards the LMC. This stellar tidal tail mapped with RR Lyrae is not aligned with the gaseous Magellanic Bridge, and is shifted by some ~5 degrees from the Young Main Sequence bridge. Abstract: This thesis develops and applies novel methods to understand water mass variability and change in the global ocean. A water mass framework is developed that determines the diathermal and diahaline transformations of water between water mass classes from the temporal variations in their volumetric distribution. Water masses are defined in terms of their temperature and salinity. This reveals the roles of air--sea fluxes, mixing and transport changes. The second chapter explores the drivers of interannual variability in the overturning circulation in the North Atlantic subtropical gyre using the water mass framework. Variations in the volumetric distribution of water masses reveal that transport anomalies at the gyre boundaries predominantly set the volume and heat budget and that these transport anomalies are governed by Ekman pumping over the gyre. In the third and fourth chapters of this thesis the water mass framework is applied to observations of temperature and salinity in the Southern Ocean. Seasonal variations in the distribution of water masses reveal the imprint of the Southern Ocean overturning. This highlights the importance of seasonally varying air-sea fluxes in the formation of intermediate water at the expense of deep water, winter water and surface water. This reveals a diabatic pathway for the upwelling and conversion of deep water into intermediate water. Deep water is first cooled and freshened during the winter by mixing with overlying winter water triggered by a cabbeling instability. Sea ice-melt and surface heating then warm and freshen this seasonally formed water mass to create intermediate water during the summer months. These results suggest that the process of cabbeling could be a rate determining step in the global overturning circulation and the upwelling of deep waters. The fifth chapter of this thesis explores an alternative method to determine a volumetric distribution using individual Argo profiles. The volumetric distribution determined using this profile based estimate is compared to the distribution calculated using a geographically interpolated dataset. This comparison reveals that the interpolation scheme used to geographically grid Argo appears to artificially mix water masses toward the centre of the distribution.

Abstract: Objective To compare colorectal cancer (CRC) incidences in carriers of pathogenic variants of the MMR genes in the PLSD and IMRC cohorts, of which only the former included mandatory colonoscopy surveillance for all participants. Methods CRC incidences were calculated in an intervention group comprising a cohort of confirmed carriers of pathogenic or likely pathogenic variants in mismatch repair genes (path_MMR) followed prospectively by the Prospective Lynch Syndrome Database (PLSD). All had colonoscopy surveillance, with polypectomy when polyps were identified. Comparison was made with a retrospective cohort reported by the International Mismatch Repair Consortium (IMRC). This comprised confirmed and inferred path_MMR carriers who were first- or second-degree relatives of Lynch syndrome probands. Results In the PLSD, 8,153 subjects had follow-up colonoscopy surveillance for a total of 67,604 years and 578 carriers had CRC diagnosed. Average cumulative incidences of CRC in path_MLH1 carriers at 70 years of age were 52% in males and 41% in females; for path_MSH2 50% and 39%; for path_MSH6 13% and 17% and for path_PMS2 11% and 8%. In contrast, in the IMRC cohort, corresponding cumulative incidences were 40% and 27%; 34% and 23%; 16% and 8% and 7% and 6%. Comparing just the European carriers in the two series gave similar findings. Numbers in the PLSD series did not allow comparisons of carriers from other continents separately. Cumulative incidences at 25 years were < 1% in all retrospective groups. Conclusions Prospectively observed CRC incidences (PLSD) in path_MLH1 and path_MSH2 carriers undergoing colonoscopy surveillance and polypectomy were higher than in the retrospective (IMRC) series, and were not reduced in path_MSH6 carriers. These findings were the opposite to those expected. CRC point incidence before 50 years of age was reduced in path_PMS2 carriers subjected to colonoscopy, but not significantly so. We highlight a few of Gaia's properties that will benefit the study of variable objects, and illustrate with two examples the work being done in the preparation of the data processing and object characterization. The first example relates to the analysis of the nearly simultaneous multi-band data of Gaia with the Principal Component Analysis techniques, and the second example concerns the classification of Gaia time series into variability types. Abstract: The overturning streamfunction as measured at the OSNAP (Overturning in the Subpolar North Atlantic Program) mooring array represents the transformation of warm, salty Atlantic Water into cold, fresh North Atlantic Deep Water (NADW). The magnitude of the overturning at the OSNAP array can therefore be linked to the transformation by air–sea buoyancy fluxes and mixing in the region north of the OSNAP array. Here, we estimate these water mass transformations using observational-based, reanalysis-based and model-based datasets. Our results highlight that air–sea fluxes alone cannot account for the time-mean magnitude of the overturning at OSNAP, and therefore a residual mixing-driven transformation is required to explain the difference. A cooling by air–sea heat fluxes and a mixing-driven freshening in the Nordic Seas, Iceland Basin and Irminger Sea precondition the warm, salty Atlantic Water, forming subpolar mode water classes in the subpolar North Atlantic. Mixing in the interior of the Nordic Seas, over the Greenland–Scotland Ridge and along the boundaries of the Irminger Sea and Iceland Basin drive a water mass transformation that leads to the convergence of volume in the water mass classes associated with NADW. Air–sea buoyancy fluxes and mixing therefore play key and complementary roles in setting the magnitude of the overturning within the subpolar North Atlantic and Nordic Seas. This study highlights that, for ocean and climate models to realistically simulate the overturning circulation in the North Atlantic, the small-scale processes that lead to the mixing-driven formation of NADW must be adequately represented within the model's parameterisation scheme.

Abstract: The North Atlantic meridional overturning circulation and its variability are examined in terms of the overturning in density space and diapycnal water mass transformation. The magnitude of the mean overturning is similar to the surface water mass transformation, but the density and properties of these waters are modified by diapycnal mixing. Surface waters are progressively densified while circulating cyclonically around the subpolar gyre, with the densest waters and deepest convection occurring in the Labrador Sea and Nordic Seas. The eddy-driven interaction between the convective interior and boundary currents is a key to the export of dense waters from marginal seas. Due to the multitude of pathways of dense waters within the subpolar gyre, as well as mixing with older waters, waters exiting the subpolar gyre have a wide range of ages, with a mean age on the order of a decade. As a result, interannual changes in water mass transformation are mostly balanced locally and do not result in changes in export to the subtropics. Only persistent changes in water mass transformation result in changes in export to the subtropics. The dilution of signals from upstream water mass transformation suggests that variability in export of dense waters to the subtropics may be controlled by other processes, including interaction of dense waters with the energetic upper ocean. Abstract: Background The Prospective Lynch Syndrome Database (PLSD) collates information on carriers of pathogenic or likely pathogenic MMR variants (path_MMR) who are receiving medical follow-up, including colonoscopy surveillance, which aims to the achieve early diagnosis and treatment of cancers. Here we use the most recent PLSD cohort that is larger and has wider geographical representation than previous versions, allowing us to present mortality as an outcome, and median ages at cancer diagnoses for the first time. Methods The PLSD is a prospective observational study without a control group that was designed in 2012 and updated up to October 2022. Data for 8500 carriers of path_MMR variants from 25 countries were included, providing 71,713 years of follow up. Cumulative cancer incidences at 65 years of age were combined with 10-year crude survival following cancer, to derive estimates of mortality up to 75 years of age by organ, gene, and gender. Findings Gynaecological cancers were more frequent than colorectal cancers in path_MSH2, path_MSH6 and path_PMS2 carriers [cumulative incidence: 53.3%, 49.6% and 23.3% at 75 years, respectively]. Endometrial, colon and ovarian cancer had low mortality [8%, 13% and 15%, respectively] and prostate cancers were frequent in male path_MSH2 carriers [cumulative incidence: 39.7% at 75 years]. Pancreatic, brain, biliary tract and ureter and kidney and urinary bladder cancers were associated with high mortality [83%, 66%, 58%, 27%, and 29%, respectively]. 39. Seasonal overturning variability in the eastern North Atlantic subpolar gyre: a Lagrangian perspective

Abstract: Both observations and ocean reanalyses show a pronounced seasonality in the strength of the Atlantic meridional overturning circulation (MOC) within the eastern North Atlantic subpolar gyre (eSPG). However, attributing this overturning seasonality to seasonal dense water formation remains challenging owing to the wide distribution of recirculation timescales within the Iceland and Irminger basins. Here, we investigate the nature of seasonal overturning variability using Lagrangian water parcel trajectories initialised across the Overturning in the Subpolar North Atlantic Program (OSNAP) East section within an eddy-permitting ocean sea ice hindcast. By adopting a Lagrangian perspective, we show that the seasonal minimum of the Eulerian overturning at OSNAP East in autumn results from a combination of enhanced stratification and increased southward transport within the upper East Greenland Current. This convergence of southward transport within the MOC upper limb is explained by decreasing water parcel recirculation times in the upper Irminger Sea, consistent with a gyre-scale response to seasonal wind forcing. To account for the diversity of recirculation times within the eSPG, we also quantify the Lagrangian overturning (LMOC) as the total dense water formation along water parcel trajectories. The majority of water parcels, sourced from the central and southern branches of the North Atlantic Current, fail to return to OSNAP East prior to experiencing wintertime diapycnal transformation into the lower limb, and thus they determine the mean strength of the LMOC within the eSPG (8.9 ± 2.2 Sv). The strong seasonality of the LMOC is explained by a small collection of upper-limb water parcels, circulating rapidly (≤ 8.5 months) in the upper Irminger and central Iceland basins, whose along-stream transformation is determined by their month of arrival at OSNAP East.

Numerical simulations in which a period search is performed on Gaia time series with a scan-angle-dependent signal qualitatively reproduce the general structure observed in the spurious period distribution of photometry and astrometry, and the associated spatial distributions on the sky. A variety of statistics allows for the deeper understanding and identification of affected sources., [Conclusions] The origin of the scan-angle-dependent signals and subsequent spurious periods is well understood and is mostly caused by fixed-orientation optical pairs with a separation < 0.5″ (including binaries with P ≫ 5 y) and (cores of) distant galaxies. Although most of the sources with affected derived parameters have been filtered out from the Gaia archive nss_two_body_orbit and several vari-tables, Gaia DR3 data remain that should be treated with care (no sources were filtered from gaia_source). Finally, the various statistics discussed in the paper can be used to identify and filter affected sources and also reveal new information about them that is not available through other means, especially in terms of binarity on sub-arcsecond scale.

42. The vitality assessment of traumatised permanent anterior teeth using laser Doppler flowmetry

Abstract: The clinical management of traumatised permanent anterior teeth is complicated by the unreliability of current methods of assessing dental pulp vitality. This study investigated the reliability of laser doppler flowmetry as a method of assessing the pulpal status of traumatised permanent anterior teeth. The study found that although laser doppler flowmetry could record pulpal blood flow, non-pulpal signals were an unavoidable component of the signal obtained. The optimum non-pulpal/pulpal flux signal ratio was obtained when the laser doppler flowmeter probe was supported by a two-stage elastomeric impression jig, perpendicular to the tooth surface, at a distance of between 2-3 mm from the gingival margin. This method resulted in only 10% of the flux signal obtained from a vital dental pulp being of non-pulpal origin. Other recording methods reported in the literature were found to have between 15-45% of the flux signal of non-pulpal origin. Flux signals from vital permanent anterior teeth were found to have two characteristics which distinguished them from flux signals originating from non-vital anterior teeth; a minimum Mean Flux value of 7 perfusion units (P.U.) and a rhythmical variation in the flux signal (termed Slow Wave Vasomotion) with a frequency of between 1-10 cycles a minute, and a mean amplitude of at least 1.6 P.U. Diagnostic criteria based on these variables were used to discriminate between flux signals from 84 vital anterior teeth and 67 non-vital anterior teeth, and were found to have a diagnostic sensitivity and specificity of 1.0. All currently used methods of assessing dental pulp vitality were found to be less reliable (have a lower sensitivity and specificity) than laser doppler flowmetry. The sensitivity and specificity of the majority of the tests remained relatively unchanged whether applied to traumatised or untraumatised teeth. The exception were tests of pulpal sensibility, which showed a large fall in specificity when applied to traumatised incisors as compared with untraumatised incisors. For example, it was found that vital luxated incisors were more likely than not to fail to respond positively to sensibility testing. Although laser doppler flowmetry was found to have a sensitivity and specificity in excess of other pulpal diagnostic tests in current use, it was also found that the pulpal status of teeth was not always determined at the time of injury but could change subsequent to the injury. Both pulpal necrosis and pulpal revascularisation were possible late consequences of dental trauma, occurring months or even years following the injury. Therefore, the reliable diagnosis of pulpal vitality provided by laser doppler flowmetry should only be regarded as an aid to treatment planning the dental care of traumatised incisors. It would appear, however, that the technique of laser doppler flowmetry has potential in increasing current knowledge regarding the pathogenesis of dental pulp disease following traumatic injury.

Abstract: The biological characteristics of the adult sea trout stock in the River Tywi were examined between 1988 and 1992 by sampling catches from a temporary main-river trap, and the commercial seine, commercial coracle and recreational rod fisheries. Radio tracking of adults during the spawning season investigated behavioural activities and their possible effects on stock characteristics. Catch effort data from angler log books assessed the annual performance of the rod fishery between 1990 and 1993. The effects of ameliorating the impact of acidification on water quality, benthic macro-invertebrates and juvenile salmonids, through reservoir liming, was studied.

Abstract: Intrahepatic cholestasis of pregnancy (ICP) is a pregnancy-specific liver disorder affecting 0.5-2% of pregnancies. The majority of cases present in the third trimester with pruritus, elevated serum bile acids and abnormal serum liver tests. ICP is associated with an increased risk of adverse outcomes, including spontaneous preterm birth and stillbirth. Whilst rare mutations affecting hepatobiliary transporters contribute to the aetiology of ICP, the role of common genetic variation in ICP has not been systematically characterised to date. Here, we perform genome-wide association studies (GWAS) and meta-analyses for ICP across three studies including 1138 cases and 153,642 controls. Eleven loci achieve genome-wide significance and have been further investigated and fine-mapped using functional genomics approaches. 48. Obtaining an Evidence-Base for Clinical Dentistry through Clinical Trials

Abstract: Ideally, all clinical practice should be based on the best available evidence; however, in general dental practice even a very routine issue, such as the appropriate management of the carious primary dentition, is hampered by the lack of a sound evidence-base from high quality clinical trials run in general dental practices. Furthermore, the use of the evidence that is available may be confounded by inadequate reporting. This paper reviews the difficulties reported in successfully completing such clinical trials, and reports on the authors’ own experiences. Suggestions are made for overcoming the challenge of patient recruitment in randomised controlled clinical trials in primary dental care. These include: regular contact with a principal investigator, who is viewed as a peer; ensuring the relevance of the research topic to the dentists’ daily practice; appropriate remuneration; acknowledgement of the contribution made and recognition of the contribution of research activity to continuing professional development. Use of the CONSORT statement to improve planning and reporting of clinical trials is recommended.
