Euryops pectinatus‘Viridae’ weed belonging to the family Asteraceae is commonly known as Golden Euryops or African daisy. Present investigation includes phytochemical analysis of methanol extract ofEuryops pectinatusweed for the presence of various secondary metabolites. Presence of various secondary metabolites was observed including phenols, flavonoids, alkaloids, steroids, saponins, glycosides, etc. These secondary metabolites are well known for their biological activities like antioxidation, anti-cancerous, anti-microbial, cardiovascular, and anti-aging. After determination of rich secondary metabolites in the methanol extract, mass spectrometric analysis was carried out for the identification of compounds present inEuryops pectinatus. Eight compounds were identified in the methanol extract reported for nematicide, antioxidant, analgesic, antitumor, anticancerous and several other important activities. Thus phytochemical and mass spectrometric analysis would open new doors to natural product research that would be cost effective and safe for the mankind. Helianthus tuberosus L. (Jerusalem artichoke) is an herbaceous perennial plant in the Asteraceae that is native to North America but introduced to China in the 17th century. The tubers of H. tuberosus are used as a vegetable, for the pharmaceutical production of inulin and as a source of ethanol biofuel, several B vitamins and minerals。From June to September 2009, severe powdery mildew infection was observed on H. tuberosus in a vegetable garden at Shangqiu Normal University, Shangqiu, China. Approximately 60% of leaves on individual plants were symptomatic, and almost 70% of the plants were infected. Initially, discrete powdery mildew colonies appeared on the upper surface of the leaves and on stems of the plants. Later, mycelial growth was amphigenous, thick, forming irregular white patches, covering the whole leaf surface. Eventually, leaves turned yellow, withered, and abscissed. Microscopic observations showed that conidia on infected leaves were ellipsoid-ovoid to barrel-shaped, with distinct fibrosin bodies visible in their cytoplasm, measuring 28 to 38 × 15 to 22 μm (n = 40). Conidiophores were unbranched, straight, 80 to 210 × 8 to 14 µm (n = 40) in size, and produced two to six immature conidia in chains with intercellular diaphragms. Foot cells of conidiophores were cylindrical and 45 to 65 μm × 8 to 14 µm (n = 40), with slight constrictions at basal septa, and followed by one to three short cells. Fungal hyphae were septate, branched, and flexuous to straight and 5 to 8 µm wide with indistinct to slightly nipple-shaped appressoria. These structures are typical of the genus Podosphaera, although chasmothecia were not observed. The sequence of ITS1-5.8s-ITS2 region of rDNA were amplified from conidia collected from infected leaves with universal primers ITS1F and ITS4 (White et al. 1990), sequenced and analyzed using the BLASTn search of GenBank. Amplicons were 603 bp (GQ927254) and exhibited 99.83% sequence identity with sequence of P. fusca from Cucurbita pepo (KJ698669) in Italy (Pirondi et al. 2015). The fungal species was identified as P. fusca (synonym P. xanthii ) by morphological characteristics and molecular analysis (Braun and Takamatsu 2000; Braun and Cook 2012). Pathogenicity tests were conducted by gently pressing the infected leaves onto leaves of six healthy H. tuberosus plants while six noninoculated plants served as controls. Plants were maintained in a greenhouse at 25 ± 2°C. Eight days after inoculation, symptoms similar to those observed under natural conditions developed on the inoculated leaves of H. tuberosus plants, whereas the control plants remained symptomless. The fungus on inoculated leaves was morphologically identical to that first observed in the field. P. fusca parasitizes a large number of asteraceous species including Euryops pectinatus (Saenz et al. 2000), Coreopsis lanceolata (Garibaldi et al. 2007), Cosmos caudatus (Siddiqui et al. 2011), Herba eupatorii (Ding et al. 2013) and etc. Powdery mildew caused by P. fusca has been reported on H. tuberosus in Russia (Farr and Rossman 2019). Golovinomyces ambrosiae was previously recorded on H. tuberosus in China (Huang et al. 2017; Radisek et al. 2018). This is the first report to our knowledge of powdery mildew P. fusca on H. tuberosus in China. It could cause significant yield losses and become a threat to production of H. tuberosus .

Euryops pectinatus is a South African ornamental plant belonging to family Asteraceae. The present work evaluates the cytotoxic activity and phytochemical profile of the flower extract. Metabolite profiling was performed using HPLC-PDA-ESI-MS/MS. Total phenolics and flavonoids content were assessed. Cytotoxicity was evaluated against 6 different cancer cell lines using MTT assay. The possible underlying mechanism was proposed. We analyzed whether the extract could overcome the resistance of multidrug-resistant cancer cells for doxorubicin. The effect of combination of E. pectinatus with doxorubicin was also studied. Additionally, the potential inhibitory activity of the identified phytochemicals to PB1 protein was analyzed using in silico molecular docking. Twenty-five compounds were tentatively identified. Total phenolic and flavonoid contents represented 49.41 &plusmn, 0.66 and 23.37 &plusmn, 0.23 &micro, g/mg dried flower extract, respectively. The extract showed selective cytotoxicity against Caco2 cells but its main effect goes beyond mere cytotoxicity. It showed strong inhibition of P-glycoprotein, which helps to overcome multidrug resistance to classical chemotherapeutic agents. In silico molecular docking showed that dicaffeoyl quinic acid, kaempferol-O-rutinoside, rutin, and isorhamnetin-O-rutinoside exhibited the most potent inhibitory activity to PB1 involved in tumor progression. Euryops pectinatus flower heads could have promising selective cytotoxicity alone or in combination with other chemotherapeutic agents to counteract multidrug resistance.

Farfugium japonicum (L.) Kitam., known as Japanese silver leaf, is native to Japan, Korea, and Taiwan. It is grown as an ornamental plant for garden plantings and containers not only in East Asia but more recently also in Europe and North America. Since 2003, powdery mildew infections of F. japonicum ‘Gigantea’ have been consistently found in the southern part of Korea, including the districts of Jeju, Seogwipo, Busan, Wando, and Ulleungdo. Specimens have been deposited in the Korea University Herbarium (KUS). Signs of powdery mildew first appeared as circular to irregular white patches on both sides of the leaves. The infections were usually severe on young leaves and caused malformation and browning. Appressoria on the mycelium were nipple-shaped or nearly absent. Conidiophores, measuring 160 to 280 × 10 to 12.5 μm, were simple and produced 2 to 12 immature conidia in chains, followed by 2 to 3 cells. Foot-cells in conidiophores were relatively short, 50 to 95 μm long, and constricted at the base. Conidia were hyaline, ellipsoid to ovate, 32 to 48 × 17.5 to 25 μm (length/width ratio = 1.4 to 2.3), had distinct fibrosin bodies, and produced germ tubes on the lateral position. No chasmothecia were observed. The morphology and dimentions of reproductive structures were compatible with those of Podosphaera xanthii (Castagne) U. Braun & Shishkoff (1). To confirm the identity of the causal fungus, the complete ITS region of rDNA from isolate KUS-F26469 was amplified with primers ITS5 and P3 (4) and directly sequenced. The resulting sequence of 475 bp was deposited in GenBank (Accession No. KC155426). A GenBank BLAST search of this sequence revealed 100% identity (475/475 bp) with those of many P. fusca isolates on plants in the Aster family plants including Calendula officinalis, Euryops pectinatus, Syneilesis palmata, and F. japonicum from Japan (e.g., AB040346). The P. fusca isolates listed above are now placed in P. xanthii (1). Pathogenicity was confirmed through inoculation by gently pressing diseased leaves onto leaves of three healthy potted plants of the same cultivar. Three non-inoculated plants served as controls. Plants were maintained in a greenhouse at 25 ± 2°C. Inoculated plants developed typical signs and symptoms of powdery mildew after 8 days, whereas the control plants remained symptomless. The fungus present on the inoculated leaves was morphologically identical to that originally observed on diseased plants. Powdery mildew infections of F. japonicum caused by P. fusca (syn. P. fuliginea) have been reported previously in both Japan and Korea (2). In Korea, it was listed simply as a host fungus of Ampelomyces quisqualis, which is hyperparasitic to powdery midlews, without any data on its identity (3). A hexane extract of the flower-heads of Euryops pectinatus L. (Cass.) was formulated into local anti-inflammatory implantation patches with controlled release. Cross-linked sodium hyaluronate patches (F1-F3) and chitosan patches (F4-F6) were prepared by a casting/solvent evaporation technique. Morphological and mechanical characterizations including the components ratio, surfactant and the loaded amount of the hexane extract (50, 100, and 200 mg/kg b.wt.) were investigated. Release studies were performed during 24 h using a diffusion cell. Films with optimum in vitro release rate have been investigated for testing the anti-inflammatory activity and the sustaining effect of the formulations. The sustained anti-inflammatory effect of the hexane extract of E. pectinatus flower-heads from the selected films was studied by inducing paw edema in rats with 1% (w/v) carrageenan solution. The results indicated the compatibility of hexane extract with both sodium hyaluronate and chitosan patches forming yellowish transparent films. Based on variations in drug release profiles throughout the 24-h among the formulations (F1-F6) studies, F3 and F6 were selected for further investigation. When the films were applied 1 h before the subplantar injection of carrageenan in the hind paw of male Albino rats, formulation (F3) provided its maximum inhibition of paw edema in rats (91.3%) 4 h after edema induction whereas, formulation (F6) showed less inhibition after 4 h (70.6%). The previous two formulations (F3 and F6) produced potent results (95.3 and 89.5%, respectively) after 24 h when compared with a local market preparation containing 25% β-sitosterol used as positive control. Histophathological investigation was conducted for 1, 4, and 12 weeks to study the tissue response for the two formulations (F3 and F6) at the implantation site. Chemical investigation of the hexane extract was achieved for both unsaponifiable matter (USM) and fatty acid methyl esters (FAME) using gas liquid chromatography (GLC). The USM was dominated by n-pentacosane (14.40%), phytosterols (Cholesterol, Campesterol, Stigmasterol, β-sitosterol, α-amyrin) reached 33.44% and the FAME was dominated by Linoleinic (49.97%). Quality control of the local implantation was evaluated by GLC using cholesterol as an analytical marker and phytosterols as an active marker compared to the plain extract.

Euryops pectinatus is grown in Italy for landscape use in parks and gardens. In 2001, severe outbreaks of a previously unknown powdery mildew were observed in commercial farms located in Albenga (northern Italy). All green parts (leaves, stems, and petioles) became covered with a white mycelium. Infections were particularly severe on the upper leaf surface. With progress of the disease, infected leaves turned yellow and died. The presence of powdery mildew infections on leaves and stems only rarely was linked to growth reduction. Conidia were hyaline, cylindric to slightly doliform, did not show fibrosin bodies, borne in chains, and measured 24 to 41 × 12 to 20 μm. Cleistothecia were not observed. The pathogen was identified as Oidium sp. subgen. Fibroidium (1). Pathogenicity was confirmed by gently pressing diseased leaves on leaves of healthy E. pectinatus plants. Inoculated plants were maintained in a growth chamber at 20 to 24°C. After 12 to 14 days, powdery mildew symptoms developed. A similar disease of E. pectinatus was observed in 1999 in California and identified as being caused by Podosphaera (Sphaerotheca) fusca (2). It is possible that the powdery mildew observed in Italy belongs to the same species, also considering that recently the two genera, Podosphaera and Sphaerotheca, have been unified in the genus Podosphaera (1). Euryops pectinatus Cass. is an evergreen shrub that is planted extensively in Spain for landscape use. In 2007, powdery mildew outbreaks were observed on E. pectinatus in several nurseries located in the Axarquia area (Malaga, southern Spain). Fungal growth appeared as typical, white, powdery mildew colonies that were restricted to upper leaf surfaces. Initially, individual colonies were small and nearly circular, but later enlarged and coalesced to cover the whole leaf surface. With progress of the disease, all green parts (leaves, stems, and petioles) were covered with a white mycelium. Newly developed leaves especially became rapidly infected. Diseased leaves ultimately dried up and senesced, making nursery plants aesthetically unattractive and unsaleable. Conidiophores were erect, had crenate edge lines, cylindrical foot cells that measured 37.5 to 45.0 × 10.0 to 12.5 μm, and were followed by one to three shorter cells. Conidia were hyaline, ellipsoid to ovoid, borne in chains, and measured 27.5 to 35.0 × 12.5 to 17.5 μm. Conidial length-to-width ratios ranged from 1.6 to 2.4. Conidia possessed conspicuous fibrosin bodies and from their sides produced short germ tubes. No chasmothecia were found. The nuclear rDNA internal transcribed spacer (ITS) regions were amplified by PCR and sequenced (GenBank Accession No. EU424056). On the basis of morphological characteristics of the imperfect state and ITS sequence data, this powdery mildew was identified as Podosphaera fusca (Fr.) U. Braun & N. Shishkoff (1), this isolate belonging to ITS haplotype 15 (group III) (3); this group is considered a separate species, P. xanthii (Castagne) U. Braun & N. Shishkoff by some authors (2). Pathogenicity was confirmed by gently pressing diseased leaves onto leaves of healthy E. pectinatus plants. Plants were incubated in a growth chamber at 25°C, and after 14 days, powdery mildew colonies developed. A similar disease of E. pectinatus was observed in 1999 in California (4). P. fusca parasitizes a large number of asteraceous species including field marigold (Calendula arvensis) and fleabane (Erigeron sp.) weeds, as previously reported in the same area, and ornamentals such as Calendula officinalis, Chrysanthemum spp., and Gerbera spp., which are also grown in the same nurseries and frequently attacked by powdery mildew. Gray-leaved Euryops (Euryops pectinatus Cass., Asteraceae) is an evergreen shrub that is widely planted in landscapes in the United States. In the fall of 1999, powdery mildew was observed on E. pectinatus planted in landscapes in Redlands (San Bernardino County), CA. Symptoms consisted only of slight cupping of leaves. Fungal growth was observed on stems, leaves, petioles, and pedicels and was ectophytic and amphigenous. The white mycelium was patchy to effuse. Hyphal appressoria were indistinct (1). Conidiophore foot cells were cylindric and sometimes were tapered toward or constricted at the base. Foot cells measured 30 to 50 by 10 to 12 μm and were followed by one to two shorter cells. Conidia were cylindric to slightly doliform, borne in chains of two to three, and measured 26 to 38 by 14 to 18 μm. Conidial length to width ratios ranged from 1.7 to 2.4. Catenate conidia had crenate edge lines (3). Conidia possessed conspicuous fibrosin bodies and from their sides produced short germ tubes without appressoria. Cleistothecia were not observed. Based on these characters, the fungus was identified as Podosphaera fusca (Fr.) U. Braun & N. Shishkoff (Podosphaera sect. Sphaerotheca) (1,2). Pathogenicity was confirmed by gently pressing diseased leaves onto leaves of healthy E. pectinatus plants. Plants were incubated in a humidity chamber at 22 to 24°C and after 12 to 14 days powdery mildew colonies developed. E. pectinatus cv. Viridis, a cultivar that lacks the extensive pubescence of E. pectinatus, also developed disease when inoculated. This appears to be the first report of powdery mildew on E. pectinatus in North America. A voucher specimen has been deposited into the University of California Herbarium (accession # UC1738635). P. fusca was also observed on cv. Viridis in a nursery in New York in 1999. It is unclear where this pathogen originated. P. fusca parasitizes a large number of asteraceous species including dandelion (Taraxacum officinalis) and sowthistle (Sonchus spp.) weeds, which occur in the area and sometimes are infected with powdery mildew. The Euryops powdery mildew pathogen may be a race that is different than those found on other composites in the United States.
