The global emergence of Covid-19 has caused huge human casualties. Clinical manifestations of the disease vary from asymptomatic to lethal, and the symptomatic form can be associated with cytokine storm and non-homeostatic inflammatory response. In face of the urgent demand for effective drugs to treat Covid-19, we have searched for candidate compounds using a drug repurposing approach based on in silico analysis followed by biological validation. Here we identified celastrol, a pentacyclic triterpene isolated from Tripterygium wilfordii Hook F – a plant used in traditional Chinese medicine – as one of the best compounds out of 39 repurposed drug candidates. Celastrol reverted gene expression signature from SARS-CoV-2-infected cells; bound with high-affinity energy to viral molecular targets such as main protease (Mpro) and receptor-biding domain (RBD); inhibited SARS-CoV-2 replication in monkey (Vero and Vero-ACE2) and human (Caco-2 and Calu-3) cell lines; and decreased interleukin-6 (IL-6) secretion in SARS-CoV-2-infected human cell lines. Interestingly, celastrol acted in a concentration-dependent manner, with undetectable signs of cytotoxicity. Therefore, celastrol is a promising lead drug candidate to treat Covid-19 due to its ability to suppress SARS-CoV-2 replication and IL-6 production in infected cells, two critical events in the pathophysiology of this disease.

COVID-19 patients may exhibit neuropsychiatric and neurological symptoms. We found that anxiety and cognitive impairment are manifested by 28-56% of SARS-CoV-2-infected individuals with mild respiratory symptoms and are associated with altered cerebral cortical thickness. Using an independent cohort, we found histopathological signs of brain damage in 25% of individuals who died of COVID-19. All of the affected brain tissues exhibited foci of SARS-CoV-2 infection and replication, particularly in astrocytes. Infection of neural stem cell-derived astrocytes changed energy metabolism, altered key proteins and metabolites used to fuel neurons and for biogenesis of neurotransmitters, and elicited a secretory phenotype that reduces neuronal viability. Our data support the model where SARS-CoV-2 reaches the brain, infects astrocytes and triggers neuropathological changes that contribute to the structural and functional alterations in the brain of COVID-19 patients.

IntroductionNeutrophilia and high levels of proinflammatory cytokines and other mediators of inflammation are common finds in patients with severe acute respiratory syndrome due to COVID-19. By its action on leukocytes, we propose colchicine as an intervention worthy of being tested.ObjectiveTo evaluate whether the addition of colchicine to standard treatment for COVID-19 results in better outcomes.MethodsWe present the interim analysis of a single-center randomized, double-blinded, placebo controlled clinical trial of colchicine for the treatment of moderate to severe COVID-19, with 38 patients allocated 1:1 from April 11 to July 06, 2020. Colchicine regimen was 0.5 mg thrice daily for 5 days, then 0.5 mg twice daily for 5 days. The first dose was 1.0 mg whether body weight was ≥ 80 kg.EndpointsThe primary endpoints were the need for supplemental oxygen; time of hospitalization; need for admission and length of stay in intensive care units; and death rate and causes of mortality. As secondary endpoints, we assessed: serum C-reactive protein, serum Lactate dehydrogenase and relation neutrophil to lymphocyte of peripheral blood samples from day zero to day 7; the number, type, and severity of adverse events; frequency of interruption of the study protocol due to adverse events; and frequency of QT interval above 450 ms.ResultsThirty-five patients (18 for Placebo and 17 for Colchicine) completed the study. Both groups were comparable in terms of demographic, clinical and laboratory data at baseline. Median (and interquartile range) time of need for supplemental oxygen was 3.0 (1.5-6.5) days for the Colchicine group and 7.0 (3.0-8.5) days for Placebo group (p = 0.02). Median (IQR) time of hospitalization was 6.0 (4.0-8.5) days for the Colchicine group and 8.5 (5.5-11.0) days for Placebo group (p = 0.03). At day 2, 53% vs 83% of patients maintained the need for supplemental oxygen, while at day 7 the values were 6% vs 39%, in the Colchicine and Placebo groups, respectively (log rank; p = 0.01). Hospitalization was maintained for 53% vs 78% of patients at day 5 and 6% vs 17% at day 10, for the Colchicine and Placebo groups, respectively (log rank; p = 0.01). One patient per group needed admission to ICU. No recruited patient died. At day 4, patients of Colchicine group presented significant reduction of serum C-reactive protein compared to baseline (p < 0.001). The majority of adverse events were mild and did not lead to patient withdrawal. Diarrhea was more frequent in the Colchicine group (p = 0.17). Cardiac adverse events were absent.DiscussionThe use of colchicine reduced the length of supplemental oxygen therapy and the length of hospitalization. Clinical improvement was in parallel with a reduction on serum levels of C-reactive protein. The drug was safe and well tolerated. Colchicine may be considered a beneficial and not expensive option for COVID-19 treatment. Clinical trials with larger numbers of patients should be conducted to further evaluate the efficacy and safety of colchicine as an adjunctive therapy for hospitalized patients with moderate to severe COVID-19.

Respiratory tract infection can lead to diseases such as pneumonia, bronchospasm, bronchiolitis and pleural effusion, and consequently to hospitalization. The aim of this study was to investigate the presence of viruses in hospitalized children with respiratory tract infection and to assess the correlation between lower respiratory tract infection (LRTI) and the viruses studied. We screened children from 0 to 6 years of age with respiratory tract infection. From May 2004 to September 2005, a total of 272 nasopharyngeal aspirate samples were collected from hospitalized children in Sao Jose do Rio Preto, Brazil, and stored with trizol at –80°C until testing by RT-PCR. The distribution of LRTI in the study population was: 49.63% (135/272) pneumonia, 23.18% (63/272) bronchospasm, 16.17% (44/272) bronchiolitis, and 11.02% (30/272) pleural effusion. A viral infection was found in 54.41% (148/272) of the samples, as follows: in 79 (29%) RSV, in 63 (23%) HRV, in 14 (5.14%) PIV3, in 9 (5%) HMPV, in 8 (2.9%) PIV1, in 4 (1.4%) FLUB, in 3 (1.1%) FLUA and 1 (0.4%) PIV2. Furthermore, there was a significant correlation between viral infection and bronchiolitis (43/44 cases: 97.8%; p≤0.001) and RSV infection and bronchiolitis (30/44 cases: 68.2%; p≤0.001). Finally, this study has confirmed that not only is RSV the most important virus in association with bronchilitis in infants, but also can be frequently detected in children between 2 and 5 years of age hospitalized for ARI in this region. O vírus sincicial respiratório humano (HRSV) é frequente causa de infecção respiratória aguda (IRA) em crianças. Em estudo anterior detectamos HRSV (por RTPCR em tempo real) em 15% das crianças com hipertrofia amigdaliana crônica sem sintomas de IRA, indicando que essas crianças podem ser portadoras silenciosas e saudáveis. Dados preliminares do laboratório incluem a detecção de proteínas estruturais do HRSV em células linfomononucleares de tonsilas e de linfonodos, incluindo linfócitos T CD4+ e B. O presente estudo visou a investigar os efeitos da infecção por HRSV especificamente em linfócitos B. Nosso objetivo foi analisar o impacto do HRSV na viabilidade celular e funcionalidade de linfócitos B infectados com HRSV. Confirmamos a presença de infecção natural em linfócitos B de tonsilas humanas, inclusive com formação de estruturas indicativas de corpos de inclusão. HRSV também infecta in vitro linfócitos B purificados de tonsilas humanas, também com formação de corpos de inclusão. Em seguida, estudamos a infecção por HRSV na linhagem de linfócitos B Jeko-1, e constatamos que ela é susceptível ao HRSV, com a presença de estruturas semelhantes a corpos de inclusão. A porcentagem de Jeko-1 infectada por HRSV determinada por citometria de fluxo 24h e 240h após a infecção foi de 40% e 4%, respectivamente. A infecção por RSV não reduziu significantemente a viabilidade de células Jeko-1. Os dados indicam que embora haja acúmulo de RNA viral nas células, isso não se reflete em aumento correspondente no sobrenadante. De modo inusitado, a dosagem por real-time PCR dos RNAs mensageiros virais mostrou ausência quase completa de RNAm para a proteína L. A secreção de citocinas no sobrenadante de células Jeko-1 infectadas HRSV mostrou ser diferente de células inoculadas com mock, especialmente evidenciado por aumento na secreção de IL-6, IL-10 e TNF-α nas células infectadas. O presente modelo in vitro de infecção de linhagem de célula B por HRSV ratifica a capacidade do HRSV de infectar linfócitos B, com formação de fábricas virais, porém com permissividade comprometida, favorecendo uma infecção pouco produtiva e de longa duração. Human respiratory syncytial virus (HRSV) is a frequent cause of acute respiratory infection (ARI) in children. In a previous study, we detected HRSV (by realtime RT-PCR) in 15% of children with chronic tonsillar hypertrophy without ARI symptoms, indicating that these children may be silent healthy carriers. Preliminary laboratory data evidenced detection of HRSV structural proteins in lymphomononuclear cells of tonsils and lymph nodes, including CD4+ T and B lymphocytes. The present study aimed to investigate the effects of HRSV infection specifically on B lymphocytes, the impact of HRSV on viability and functionality of HRSV-infected B lymphocytes. We confirmed the presence of natural infection in human tonsil B lymphocytes, including formation of structures indicative of inclusion bodies. HRSV also infects in vitro purified B lymphocytes from human tonsils, also with formation of inclusion bodies. Next, we studied the HRSV infection in the Jeko-1 B lymphocyte lineage, and found that it is susceptible to HRSV, with the presence of inclusion body-like structures. The percentage of HRSV-infected Jeko-1 determined by flow cytometry 24h and 240h after infection was 40% and 4%, respectively. RSV infection did not significantly reduce the viability of Jeko-1 cells. The data indicate that although there is accumulation of viral RNA in the cells, this is not reflected in a corresponding increase of virus RNA progeny in the supernatant. Unexpectedly, the real-time PCR measurement of viral messenger RNAs showed almost complete absence of RNAm for the L protein. The secretion of cytokines in the supernatant of HRSV infected Jeko-1 cells showed to be different from mock inoculated cells, especially evidenced by increased secretion of IL-6, IL-10 and TNF-α in infected cells. The present in vitro model of B cell lineage infection by HRSV confirms the ability of HRSV to infect B lymphocytes, with the formation of viral factories, but with compromised permissiveness, favoring an unproductive and long-lasting infection.

O metapneumovírus humano (HMPV) é um importante agente causador de infecções respiratórias agudas (IRA), com tropismo para o epitélio respiratório e elevado impacto em morbidade e mortalidade na infância em todo o mundo. Porém, pouco se sabe sobre sua patogênese, especialmente em infecções de tecidos não epiteliais. Estudos prévios mostraram o genoma de HMPV em tecidos tonsilares de crianças com hipertrofia adenoamigdaliana. Nosso objetivo geral foi estudar a infecção por HMPV em tecidos de tonsilas de crianças sem sintomas de IRA, identificar as células-alvo e confirmar a presença de replicação de HMPV nesses tecidos, que podem ser fontes de disseminação do vírus na comunidade. Para tanto, foi realizada RT-PCR para o genoma do HMPV em tecidos linfoides utilizando o kit Taqman® Universal PCR Master Mix (Applied Biosystem). Citometria de fluxo foi usada para quantificar diferentes imunofenótipos de células infectadas e a localização in situ foi feita em cortes de tecido embebidos em parafina e analisados por imuno-histoquímica usando o método de SIMPLE (serial immuno-peroxidase labeling and erasing). Além das células epiteliais, HMPV foi detectado em células linfomononucleares nos centros germinativos, na zona do manto e nas áreas interfoliculares dos tecidos tonsilares. As células-alvo infectadas por HMPV foram identificadas e a análise imunofenotípica mostrou que células naturalmente infectadas foram positivas para marcadores CD3, CD4, CD8, CD20, CD68, CD123 e CD14. Observou-se que células linfoides foram mais frequentemente infectadas em amígdalas, enquanto células mieloides foram mais frequentemente infectadas em adenoides. Um dos principais achados deste estudo foi alta frequência de macrófagos M2 (CD206 + CD80 +), e não M1, infectados em ambos os tecidos. Além de detectar macrófagos positivos para HMPV in vivo, foi demonstrada a suscetibilidade à infecção pelo HMPV em linhagem celular de macrófagos. HMPV infeccioso foi recuperado de amígdalas humanas em cultura de células, induzindo efeito citopático caracterizado com formação de sincícios, arredondamento celular e extensa ruptura da monocamada, o que foi confirmado por imunocitoquímica. O presente estudo mostra pela primeira vez que o HMPV infecta tonsilas hiperplásicas de crianças assintomáticas, que podem assim ser potenciais reservatórios do vírus, um achado significante para a epidemiologia do vírus. Human metapneumovirus (HMPV) is an important causative agent of acute respiratory infections (ARI), which tropism for the respiratory epithelium is known and a leading cause of childhood morbidity and mortality worldwide. However, little is known about the pathogenesis of HMPV, especially infections of tissues other than the airways epithelia. We have previously reported that HMPV genome was detected in tonsillar tissue taken from children with hyperplastic palatine tonsils and adenoids. In the present study, we analyzed HMPV infection in tonsillar tissues from asymptomatic children, identify target cells and investigate HMPV replication in these tissues, raising the possibility that hyperplastic tonsils may be sources of HMPV for the community. To achieve our goal, RT-PCR assay was performed to detect genome HMPV in lymphoid tissues using Taqman® Universal PCR Master Mix Kit (Applied Biosystem). We used flow cytometry to quantify infected phenotypes and in situ localization was performed in sliced tissues embedded in paraffin to perform immunohistochemistry followed by SIMPLE (serial immuno-peroxidase labeling and erasing). In addition to epithelial cells, we were also able to detect HMPV in germinal centers, mantle zone and in interfolicular region. Moreover, serial immunohistochemical stainings showed that naturally infected cells were positive for CD3, CD4, CD8, CD20, CD68, CD123 and CD14 markers. We observed that lymphoid cells were the most frequently infected cell types in palatine tonsils, whereas HMPV positive myeloid cells were more frequently detected in adenoids. One of the main findings of this study was the high frequency of M2 and not M1 infected macrophages (CD206+CD80+) in both tissues. In addition to detect HMPV positive macrophages in vivo, the susceptibility to HMPV infection was demonstrated. Infectious HMPV from human tonsils was recovered in cell culture with visible cytopathic effect, characterized by syncytia formation, cell rounding and extensive monolayer disruption, which was confirmed by immunocytochemistry. The present study shows for the first time that HMPV infects hyperplastic tonsils of asymptomatic children, which may thus be potential reservoirs of the virus, a significant finding for the epidemiology of the virus.

A infecção pelo vírus da chikungunya (CHIKV) em humanos é geralmente caracterizada por um breve viremia, seguida de um rápido clearence viral. Apesar do quase imediato controle viral sistêmico, estudos sugerem a persistência do CHIKV em humanos e primatas não-humanos em reservatório imunológicos. A fase crônica da infecção por CHIKV e pelo vírus do Ross River (RRV), alfavírus artritogênicos, possui características similares à desregulação da reposta imune observada na artrite reumatoide (AR). O desenvolvimento de uma poliartralgia persistente em número considerável de pacientes possui um grande impacto social e econômico. Assim, potencialmente, a permanência viral pode estar relacionada com a manutenção da resposta inflamatória exacerbada por longos períodos. Sabe-se que a migração de células T CD4+ para a cápsula articular ocorre durante a primeira semana de infecção, e correlaciona-se negativamente com o pior prognóstico da chikungunya. Contudo, pouco se sabe sobre o papel dos subtipos das células T na fisiopatologia da doença. Uma vez que não existiam modelos de camundongos que permitissem o estudo da artrite crônica induzida pelo CHIKV, nosso objetivo foi desenvolver e caracterizar a resposta de células T e persistência viral em um modelo animal crônico. Portanto, em nosso estudo, desenvolvemos um novo modelo animal para o estudo da chikungunya crônica, que é capaz de recapitular os sinais comumente observados na artrite crônica da chikungunya em humanos. Os nossos resultados demonstram a importância das células T CD4+ para a patogenia da doença crônica da chikungunya em camundongos, especificamente células Th17, dados corroborados em humanos. Alguns dados sugerem que a infecção por CHIKV regula negativamente a produção de citocinas essenciais na regulação de resposta de células Th17, tais como, IL-10, IL-27 e IL-35. Adicionalmente, nossos resultados demonstraram que, de forma dependente da expressão de IFITM3, os macrófagos e células sinoviais são reservatórios virais e passíveis a replicação viral de alfavírus artritogênicos. Macrófagos infectados mantém o microambiente extremamente inflamatório através da produção de citocinas inflamatórias como o IL-6, e quimiocinas como CCL5 e CCL20, estimulando um maior recrutamento de infiltrado de células T na sinovias infectadas com o CHIKV, mesmo após o controle viral sistêmico. Em conclusão, esses dados demonstram que no nosso modelo animal a artrite crônica induzida por CHIKV é dependente da sinalização de IL-17 pelas células Th17, e que a persistência viral em macrófagos sinoviais mantém o microambiente inflamatório propicio para a manutenção do recrutamento das células T para a região sinovial. Chikungunya virus (CHIKV) infections in humans are usually characterized by brief viremia followed by rapid viral clearance. Despite the almost immediate systemic viral control, studies suggest the persistence of CHIKV in humans and non-human primates in immunological reservoirs. The chronic phase of infection by CHIKV and by Ross River virus (RRV), arthritogenic alphaviruses, has characteristics similar to the dysregulation of the immune response seen in rheumatoid arthritis (RA). The development of persistent polyarthralgia in a considerable number of patients has a significant social and economic impact. Thus, potentially, viral permanence may be related to the maintenance of the exacerbated inflammatory response for long periods. It was previously demonstrated that the migration of CD4+ T cells to the joint capsule occurs during the first week of infection, and it correlates with the worst prognosis of chikungunya. However, little is known about the role of T cell subtypes in the pathophysiology of the disease. Since there were no mouse models that would allow the study of CHIKV-induced chronic arthritis, we aimed to develop and characterize a new chronic animal model, followed by its T cell response and viral persistence. Therefore, in our study, we developed a new animal model for the chronic chikungunya understanding. Our model can induce the signs commonly seen in chronic chikungunya arthritis in humans. Our results demonstrate the importance of CD4+ T cells for the pathogenesis of chronic chikungunya disease in mice, specifically Th17 cells, data corroborated in humans. Some data suggest that CHIKV infection negatively regulates the production of essential cytokines in the regulation of Th17 cell response, such as IL-10, IL-27, and IL-35. Additionally, our results demonstrated that macrophages and synovial cells, in an IFITM3 expression-dependent manner, are viral reservoirs and susceptible to viral replication of arthritogenic alphaviruses. Infected macrophages maintain the extremely inflammatory microenvironment through the inflammatory cytokines production such as IL-6, and chemokines such as CCL5 and CCL20, stimulating greater recruitment of T cell infiltrate in CHIKV-infected synovia, even after systemic viral control. In conclusion, these data demonstrate that in our animal model CHIKV-induced chronic arthritis is dependent on IL-17 signaling by Th17 cells and that viral persistence in synovial macrophages maintains the inflammatory microenvironment conducive to the maintenance of T cell recruitment to the synovial region.

No presente estudo investigamos dois aspectos pouco estudados dos mecanismos de transmissão vertical de vírus Zika (ZIKV) e Chikungunya (CHIKV) durante gestação e lactação. O ZIKV é conhecidamente teratogênico e capaz de infectar diversas células na placenta humana, inclusive trofoblastos, células intrinsecamente resistentes a infecções virais por vários mecanismos, que incluem a expressão de microRNAs (miRNAs) regulatórios de respostas antivirais. A susceptibilidade dessas células a ZIKV e CHIKV e o impacto das infecções na expressão de miRNAs da célula hospedeira foi estudado por sequenciamento de nova geração (NGS). O repertório de miRNAs em células infectadas permitiu fazer a predição computacional de vias negativamente e positivamente reguladas pelas infecções virais. Identificamos o fator de regulação de CREB3 (CREBRF) como alvo em potencial de diversos miRNAs positivamente regulados, o que tem impacto na indução de autofagia. A validação experimental foi feita pelo silenciamento com RNAs de interferência (RNAi) para silenciar CREBRF, resultando em impacto significativamente distintos nas infecções por ZIKV e CHIKV: a replicação de ZIKV aumentou, enquanto a de CHIKV sofreu redução, indicando que a regulação desses miRNAs pode ter efeito pró ou antiviral, dependendo de mecanismos de interação vírus-célula distintos entre os dois. Há diversos relatos de transmissão vertical de CHIKV, mas pouco se sabe sobre rotas e mecanismos de transmissão. No presente estudo foi desenvolvido um modelo experimental de infecção por CHIKV, usando camundongos fêmeas da linhagem BALB/c infectadas durante a gestação e lactação, o que possibilitou a confirmação da transmissão transplacentária em 8,4% dos fetos analisados, com indução de efeitos teratogênicos graves. Tendo em vista que a maior parte dos animais nascidos e amamentados por fêmeas infectadas por CHIKV sobrevivem, foi avaliado o perfil de resposta imune desses animais ao longo da vida adulta contra esse vírus. Os resultados mostraram que os camundongos amamentados em fêmeas infectadas têm resposta imune humoral e celular ativas contra o vírus na idade adulta. Mais importante, esses animais são mais resistentes à infecção experimental por CHIKV 90 dias após o nascimento, indicando que a resposta imune induzida pela amamentação é efetiva e de longa duração. Observamos que as glândulas mamárias das fêmeas infectadas são sítios de replicação para CHIKV, e que há a transmissão de antígenos virais para os neonatos via leite. Esses resultados são inéditos, e criam as bases para futuras investigações sobre mecanismos biológicos básicos da transmissão vertical de ZIKV e CHIKV, bem como para abordagens translacionais em gestantes e neonatos infectados por esses agentes. In the present study, we investigated two unexplored aspects of the Zika virus (ZIKV) and Chikungunya virus (CHIKV) vertical transmission mechanisms during pregnancy and lactation. ZIKV is known to be teratogenic and able to infect several cells in the human placenta, including trophoblasts, which are cells intrinsically resistant to viruses by several mechanisms. The expression of microRNAs (miRNAs) that regulate antiviral responses is very important to sustain this resistance. The susceptibility of these cells to ZIKV and CHIKV and the impact of changes in the expression of host miRNAs was studied by new generation sequencing (NGS) in this work. Using the repertoire of miRNAs in infected cells, we carried out computational prediction of negatively and positively regulated pathways during viral infections. We identified the CREB3 regulatory factor (CREBRF) as a potential target of several up-regulated miRNAs, what has an impact on autophagy induction. The experimental validation was performed by silencing CREBRF with interference RNAs (RNAi). Interestingly, we identified distinct impacts of CREBRF silencing on ZIKV and CHIKV infections: the replication of ZIKV increased, while CHIKV suffered a significant reduction in its genome replication. These results indicate that these miRNAs can regulate pro or antiviral pathways, based on the distinct virus-cell interactions between them. There are several reports of vertical transmission of CHIKV, but little is known about routes and mechanisms of transmission. In the present study, an experimental model of CHIKV infection was developed, using female mice of the BALB/c strain infected during pregnancy and lactation, which enabled the confirmation of transplacental transmission in 8.4% of the analyzed fetuses, with induction of teratogenic effects. Taking advantage that most animals born and breastfed by CHIKV infected females survive, the profile of immune response of these animals throughout adulthood against this virus was evaluated. The results showed that mice breastfed in infected females develop active humoral and cellular immune responses against the virus in adulthood. More importantly, these animals are more resistant to experimental CHIKV infection 90 days after birth, indicating that the response induced by breastfeeding is effective and long lasting. We observed that the mice mammary glands are sites of replication for CHIKV, and the transmission of viral antigens to neonates via milk occurs. These results will be very important for future investigations about the basic biological mechanisms of vertical transmission of ZIKV and CHIKV, as well as for translational approaches in pregnant women and newborns infected by these agents.

O vírus Oropouche (OROV) pertence à ordem Bunyavirales, família Peribunyaviridae, gênero Orthobunyavirus, e está entre as mais frequentes causas de arbovirose no Brasil. OROV é endêmico da região Amazônica, onde já causou mais de meio milhão de casos em surtos publicados, no entanto recentemente tem ganhado atenção devido à ocorrência de infecções em seres humanos e primatas não humanos fora daquela região. OROV é transmitido pela picada do vetor Culicoides paraensis e causa doença febril aguda. OROV tem genoma composto por três segmentos de RNA (L, M e S) que codificam as proteínas estruturais, bem como duas não estruturais: NSm e NSs. A proteína NSs é um fator de virulência reconhecido em alguns vírus do gênero Orthobunyavirus, mas quase nada se sabe sobre funções da proteína NSm. Evidências obtidas com outros vírus da família mostram que NSm tem papel em processos de montagem e morfogênese de partículas virais. Todavia, não existem conhecimentos sobre funções da NSm de OROV. A partir do desenvolvimento deste trabalho foi possível conhecer que a proteína NSm de OROV não é essencial à produção de progênie viral, mas está envolvida com a montagem do vírus pela via canônica no complexo de Golgi. A ausência de NSm modifica o recrutamento de organelas envolvidas em fábricas virais, com diminuição da área dessas fábricas, atrasando a produção de progênie infecciosa durante a fase exponencial de replicação. Outra importante descoberta é que NSm está envolvida na atenuação da virulência de OROV in vivo, já que na sua ausência OROV é altamente letal em modelo murino. O contrário foi observado para o vírus deletado da proteína NSs, que é menos letal que o OROVwt. Mais ainda, OROV deletado simultaneamente de NSm e NSs é quase completamente atenuado. OROV sem NSs e NSm infectam células do sangue periférico humano das linhagens mielóide, de modo similar ao OROVwt, mas com produção alterada da citocina TNF-? sugerindo que essas proteínas não estruturais estão envolvidas na patogenicidade de OROV. A expressão heteróloga de NSm demonstra que NSm sozinha tem distribuição diferente da NSm do vírus nativo, indicando que a expressão de elementos da poli-proteína M viral é essencial para o seu direcionamento final. Em conjunto, essas observações mostram que NSm de OROV está envolvida com a montagem de fábricas virais e com a morfogênese viral, e tanto NSm quanto NSs estão relacionadas com a patogenicidade do vírus em modelo murino. O mutante de OROV com deleção de ambas as proteínas é significantemente atenuado, e pode ser uma importante ferramenta para o desenvolvimento de vacina. Esses dados inéditos contribuem para o conhecimento de OROV e podem auxiliar na descoberta de possíveis alvos terapêuticos. Oropouche virus is classified in Bunyavirales order, family Peribunyaviridae, genus Orthobunyavirus and it\'s one of the most frequent causes of arboviruses in Brazil. OROV is endemic in the Amazon region, where it infected half million cases in documented outbreaks. However, it\'s recently gaining more attention due to an occurrence in humans and primates outside this region. OROV is transmitted by the bite of Culicoides paraensis vector and causes a febrile disease. Its genome is composed of three negative RNA strands (L, M and S) that codes the structural and non-structural viral proteins: NSm and NSs. NSs is a known virulence factor in the Orthobunyavirus genus, but nearly nothing is known about NSm function. Evidence obtained with other orthobunyaviruses showed that NSm is involved with viral particle assembly and morphogenesis. But there are not information\'s about OROVs NSm function. With the development of this work it was possible to discover that NSm protein it is not essential for progeny production, but it is involved with the canonical assembly pathway in Golgi complex. The absence of NSm modifies organelles recruitment to viral factories, diminishing viral factories area, affecting progeny production in the exponential growth phase in the replication curve. Another important discovery is that NSm is a virulence attenuator factor since the OROV NSm mutant is highly pathogenic in a mouse model. The opposite was observed in OROV NSs mutant, being less lethal compared to OROV wild type. Moreover, OROV mutant lacking both non-structural proteins infect human peripheral blood cells from myelocytic lineage, in similar ways of OROV wild-type, but with altered TNF-a cytokine production, suggesting that these proteins are involved with OROV pathogenicity. Heterologous NSm expression demonstrated that its coding region by itself it is not functional, indicating that is necessary to express elements from the M polyprotein to the correct destination. Taking together, these observations show that OROVs NSm is involved with viral factories assembly and viral morphogenesis, and both NSm and NSs are related to pathogenicity in a mouse model. OROV mutant lacking both non-structural proteins its highly attenuated in vivo, and it can be an essential tool to develop a vaccine to this virus. These new data contribute to the discovery of possible therapeutic targets in treating this virus.

As infecções virais agudas do trato respiratório inferior representam importante problema de saúde pública no mundo e são responsáveis por grande número de óbitos em lactentes jovens, em países em desenvolvimento. Realizamos estudo observacional retrospectivo de coorte, que teve como objetivo, investigar a epidemiologia e os fatores associados à gravidade dos quadros de infecções virais agudas de vias aéreas inferiores em crianças de < 6 anos de idade, hospitalizadas no Hospital e Pronto Socorro Delphina Rinaldi Abdel Aziz em Manaus. Incluímos 146 crianças < 6 anos de idade, hospitalizadas por infecções virais agudas de vias aéreas inferiores no HPS Delphina Rinaldi Abdel Aziz, em Manaus, no Setor de Pediatria, no período de 1 de abril de 2017 a 31 de agosto de 2018, com diagnóstico clínico e com isolamento viral positivo em aspirado de nasofaringe. Coletamos todos os dados em prontuário eletrônico através de fichas de avaliação específicas. As crianças foram divididas em 2 grupos, de acordo com a gravidade da doença classificada conforme o escore de Gagliardi (doença moderada, com escore de Gagliardi 0-4; n= 66 (45,2%) e doença grave, com escore 5-7; n= 80 (54,8%)). Observamos maior número de casos de infecções virais agudas de vias aéreas inferiores nos meses com maior precipitação pluviométrica. O vírus sincicial respiratório foi o mais prevalente (n=103; 70,3%). O único fator de risco de gravidade da doença encontrado foi a presença de coinfecção viral (RR 1,59; IC95% 1,18- 2,15). Não houve associação da gravidade da doença com o tipo de vírus nem com a presença de comorbidades. Os pacientes com doença grave tiveram maior tempo de ventilação mecânica em comparação àqueles com doença moderada (mediana de 2 dias vs. 0; p=0,01). Doze pacientes (8,2%) morreram, todos com doença grave. Os fatores de risco para óbito foram evolução para choque (RR 10,53; IC 95% 2,40-46,17) e necessidade de uso de drogas vasoativas (RR 10,86; IC95% 2,48-47,63). No Brasil os vírus Zika e Chikungunya vem causando enormes epidemias com grande morbidade e significante mortalidade, tornou-se importante desenvolver métodos diagnósticos eficazes para discrimar suas infecções. No presente trabalho, desenvolvemos ensaios imunoenzimáticos utilizando células de cultura, aedes albopictus pseudo C6/36, infectadas e utilizadas como antígeno viral (EIA-ICC), para diagnósticos de infecções provocadas pelos vírus Zika (ZIKV) e Chikungunya (CHIKV). As placas contendo células infectadas tiveram suas antigenicidades verificadas nos ensaios utilizando anticorpos policlonais oriundo de camundongos hiperimunizados com ZIKV, CHIKV e outros flavivirus e alphavirus. O EIAICC ZIKV detectou anticorpos murinos de forma homotípica, porém reações cruzadas foram encontradas com Dengue 1 (DENV 1), Saint Louis Vírus (SLEV), Rocio Vírus (ROCV) e West Nilo Vírus (WNV). O EIA-ICC CHIKV também detectou anticorpos murinos de forma homotípica e reações cruzadas foram observadas com os Mayaro Vírus (MAYV) e Mucambo Vírus (MUCV). Resultados comparativos de 39 amostras foram realizados no EIA-ICC ZIKV, que demonstrou sensibilidade de 87,5% (IgM) e 100% (IgG) e especificidade de 95,65% (IgM) e 83,33% (IgG). O EIA-ICC CHIKV, teste comparativos entre 59 amostras, demostrou sensibilidade de 64,7% (IgM) e 92,3% (IgG) e especificidade de 40% (IgM) e 96,96% (IgG). Os EIA-ICCs também foram usados para diagnósticos das arboviroses ZIKV e CHIKV em 174 soros de pacientes com doença febril aguda, no EIA-ICC ZIKV foi encontrado 8,13% de IgM, 20,34% IgG e 6,97% de IgG e IgM na mesmo soro, no EIA-ICC CHIKV encontramos IgG em 8,13% dos soros e IgG em 16,86%, soros com anticorpos IgG e IgM nos mesmos foram vistos em 1,74%. Também encontramos em 11 soros anticorpos tanto para ZIKV quanto para CHIKV. Portanto o EIA-ICC ZIKV mostrou para detecção de IgM e IgG, adequadamente sensível e específico para ser validado em estudos com maiores números de amostras e ser aplicados no diagnóstico. It\'s important to develop effective diagnostic methods to discriminate against their infections. In the present study, we developed immunoenzymatic assays using cultured cells from aedes albopictus C6 / 36 infected and used as viral antigen (EIA-ICC), for diagnosis of infections caused by the Zika (ZIKV) and Chikungunya (CHIKV) viruses. Plates containing infected cells had their antigenicities verified in the assays using polyclonal antibodies from mice hyperimmunized with ZIKV, CHIKV and other flaviviruses and alphaviruses. The EIA-ICC ZIKV detected murine antibodies in a homotypic manner, but cross-reactions were found with Dengue 1 virus (DENV 1), Saint Louis virus (SLEV), Rocio virus (ROCV) and West Nile virus (WNV). Rinovírus (RV) é freqüentemente detectado nos tecidos tonsilares e nas secreções de nasofaringe de pacientes com doença adenotonsilar crônica, sem sintomas de infecção respiratória aguda (IRA). O objetivo deste estudo foi investigar a infecção por rinovírus em tonsilas humanas, com base em dois aspectos: infecção in vivo de células linfóides de tonsilas humanas naturalmente infectadas; e infecção ex vivo de células dissociadas desses tecidos inoculadas com rinovírus, visando a contribuir para elucidar possíveis mecanismos de infecção em amígdalas palatinas e adenóides humanas. De um total de 104 pacientes com doenças adenotonsilar crônicas, 21.1% (22/104) e 42.3% (44/104) apresentaram respectivamente amígdalas palatinas e adenóides positivas para RV por PCR. A replicação viral foi confirmada por hibridização in situ com sonda para intermediário replicativo nas regiões internas e externas aos folículos linfóides de amígdalas e adenóides, bem como em porções do epitélio ciliado de adenoides, e apenas raramente nas células epiteliais escamosas de tonsilas palatinas. A presença e distribuição de proteína estrutural do capsídeo viral foi detectada por imunohistoquímica (IHQ), utilizando anticorpos contra proteínas estruturais virais VP1 e VP2 nas tonsilas positivas para RV por qPCR. Os resultados indicaram marcação positiva tanto na superfície (epitélio), quanto em regiões extrafoliculares e centros germinativos. Em seguida, foi possível verificar a co-localização da marcação positiva da proteína estrutural VP2 de RV com marcadores linfocitários de membrana. Células CD4 + e CD20 + apresentaram marcação positiva para VP2 verificada usando estratégia de \'sequential immuno-peroxidase labelling and erasing\' (SIMPLE). Culturas primárias de células linfomononucleares (CLMN) de tonsilas sabidamente negativas para RV por PCR, foram infectadas in vitro, com RV (MOI=1). A replicação de RV foi titulada por TCID50, mostrando aumento inicial (24 h) e subsequente queda após 48 horas. Por IF observamos que os fenótipos de CLMN infectadas com RV in vitro foram células T CD4 + e B, mas também com células CD8 +, CD56 + e CD33 +. RV não infectou células CD123 +. RV foi isolado em WI- 38 e HeLa a partir de tecidos e secreções de nasofaringe de pacientes com hipertrofia tonsilar sem sintomas de infecção respiratória aguda. Nossos resultados confirmam que tonsilas de pacientes sem sintomas respiratórios agudos podem ser reservatórios de RV, que infecta não somente epitélio, mas também CLMN (frequentemente linfócitos T CD4 + e linfócitos B). A detecção de RNA intermediário replicativo e proteínas estruturais VP1 e VP2 nas tonsilas hipertróficas, além do isolamento de vírus infeccioso a partir de tecidos e secreções nasofaríngeas, classificam tonsilas hipertróficas como sítios de infecção e replicação de RV, e sugerem que esses indivíduos hipertróficos são portadores assintomáticos de RV persistente, e podem ser importantes fontes de transmissão de RV na comunidade. The chronic adenotonsillar diseases are frequent otorhinolaryngologic conditions caused by chronic inflammation of adenoids and palatine tonsils. Rhinovirus (RV) is highly frequently detected in secretions, and tonsillar tissues from patients experience chronic tonsillar hypertrophy without symptoms of ARI, and our goal is to full understanding of viral infections in hypertrophic tonsillar tissues by RV. Of 104 enrolled patients with adenotonsillar chronic diseases, 21.1% (22/104) and 42.3% (44/104) had palatine tonsils and adenoids positive for RV by qPCR, respectively. RV Viral replication was confirmed by in situ hybridizations. Minus-strand RNA were detected in all tested samples (7 tonsils and 9 adenoids), and positive reactions were seen inside and outside of lymphoid follicles from tonsils and adenoids, in the ciliated epithelium of the adenoids and rarely in positive squamous epithelium cells from tonsils. The presence of viral structural protein VP1 and VP2 was detected within and outside of the lymphoid follicles from tonsils and adenoids, and also in epithelial cells from adenoids by immunohistochemistry (IHC). Later, by sequential immuno-peroxidase labelling and erasing protocol (SIMPLE), we saw co-localization of RV VP2 capsid protein staining with CD4 positive cells and CD20 positive cells. We confirmed that RV could infect primary culture of tonsilar mononuclear cells (TMNCs). Additionally, intracellular replication of RV in TMNCs, measured by TCID50 in HeLa cells, had an initial increase in the first 24 hours, and dropped at 48 hours post infection. Immunolocalization staining with anti-RV and TMNCs surface markers indicated that phenotypes of susceptible cells were T-cells both CD4+ and CD20+, but also, we saw co-localization of VP-2 protein with CD8+ cells, CD56+ cells and CD33+ cells. RV-16 couldn\'t infect CD123+ cell in our experiments. Finally, we were able to recover 4 rhinoviruses by inoculating WI-38 fibroblast cells and HeLa cells, confirming by the cytopathic effect and immunofluorescence positive staining with anti-VP1 antibody. Taken together, our results indicate that tonsils and adenoids of patients without ARI may be reservoirs of replicating human rhinovirus, infecting manly Tcells CD4+ and CD20+ B-cells. The high-frequency detection of RNA (-) and VP1 expression in tissues from patients with chronic adenotonsillar diseases, plus the isolation of infectious viral particles, suggests that these detected agents replicate in the adenotonsillar tissues and this specific sites may be important sources of transmission of RV in the community.

A degradação lisossomal de proteínas de membrana endocitadas ocorre por meio do direcionamento destas proteínas para vesículas intralumenais (ILVs), formadas no lúmen dos corpos multivesiculares (MVBs), e subsequente fusão dos MVBs com lisossomos. Apesar de sua importância na degradação de proteínas transmembrana, os MVBs possuem outra importante função, a de produzir e liberar vesículas extracelulares (EVs). Neste processo os MVBs não se fundem com lisossomos, mas sim com a membrana plasmática o que resulta na liberação das vesículas residentes no interior dos MVBs para o meio extracelular. Diversas proteínas participam do direcionamento de cargas para os MVBs. Os estudos que delinearam a via de tráfego mediada por AP1 concentraram-se nos complexos contendo a subunidade ?1 que medeia o transporte de proteínas entre a rede trans-Golgi (TGN) e endossomos. Contudo, o genoma humano codifica uma segunda isoforma desta subunidade, denominada ?2, e evidências presentes na literatura e também observadas por nosso grupo indicam que AP1?2 pode regular uma via de tráfego distinta da via classicamente atribuída a AP1. Utilizando ensaios de uptake de EGF em condições onde foi realizado o KD de ?1 ou ?2, foi observado que o silenciamento de ?2 prejudica a degradação de EGF internalizado por seu receptor. Efeito também observado para o próprio receptor de EGF (EGFR) em ensaios de biotinilação da superfície celular. Demonstrando que a degradação lisossomal do complexo EGF-EGFR pela via canônica dos MVBs requer o complexo AP1?2, mas não AP1?1. Em conjunto com este estudo também foi analisado o mecanismo molecular de direcionamento da proteína Nef do HIV-1 para os MVBs associados a liberação de EVs. A proteína Nef do HIV é determinante na modulação do ambiente intracelular favorecendo a replicação do vírus e progressão à AIDS. Nef é ativamente secretado em EVs e sua liberação pode levar a apoptose de células vizinhas aceptoras dessas vesículas. Nef também medeia a redução dos níveis de CD4 e moléculas de MHC-I em EVs. Ainda não é conhecido o mecanismo molecular utilizado por Nef para ser exportado em EVs, mas sabe-se que Nef interage fisicamente com a proteína II acessória da maquinaria ESCRT, Alix, importante no processo de formação das ILVs e seleção das cargas que serão internalizadas nos MVBs. EVs coletadas de células HeLa e linfócitos T CD4+ silenciados para Alix demostraram reduções significativas na liberação de Nef. Estes resultados indicam que Nef requer Alix para sua eficiente liberação em EVs. Lysosomal degradation of endocytosed membrane proteins occurs through the targeting of these proteins to intraluminal vesicles (ILVs), formed in the multivesicular bodies (MVBs) lumen, and the subsequent fusion of MVBs with lysosomes. Despite its importance in the degradation of transmembrane proteins, MVBs have another important function, the production and release of extracellular vesicles (EVs). In this process, the MVBs do not fuse with lysosomes, but fuse with the plasma membrane resulting in the release of these vesicles that reside within MVBs to the extracellular environment. Several proteins regulate the targeting of cargo to MVBs. Studies that delineated the functions of AP1 in protein trafficking, focused on complexes containing the ?1 subunit, which mediates transport between trans-Golgi network (TGN) and endosomes. However, the human genome encodes a second isoform of this subunit, named ?2. Evidences from the literature, as well as results from our research group, indicate that AP1?2 regulates transport pathways that are distinct from the pathways classically attributed to AP1. By performing EGF-uptake assays under ?1 or ?2 knockdown (KD) conditions, it was observed that ?2 is required for degradation of internalized EGF, effect also observed for the EGF receptor (EGFR) using cell surface biotinylation assays. These results demonstrate that the lysosomal degradation of the EGFEGFR complexes via the canonical MVBs pathway requires the AP1?2 complex, but not AP1?1. In parallel with this study, we also analyzed the molecular mechanism of HIV-1 Nef targeting to MVBs associated with the EVs release. Nef is an important determinant in the modulation of the intracellular environment for efficient HIV replication and progression to AIDS. Nef is actively secreted via EVs and its release may lead to apoptosis of bystander acceptor cells. Moreover, Nef reduces the levels of CD4 and MHC-I molecules in EVs. Despite the importance of Nef release in EVs, the molecular mechanism used by Nef to be exported via EVs is unknown. Nef physically interacts with the ESCRT machinery accessory protein Alix, an important player in the process of ILVs formation and cargo selection. EVs released from HeLa cells and CD4+ T lymphocytes under Alix KD conditions demonstrated a significant IV reduction in Nef release via EVs. These results indicate that Nef requires Alix for its efficient release in EVs.

O vírus da Zika (ZIKV) é um arbovírus emergente da família Flaviviridae, do gênero Flavivirus transmitido por mosquitos Aedes. Apesar da sua importância emergente na saúde pública, ainda pouco se conhece sobre os mecanismos moleculares envolvidos no ciclo replicativo do ZIKV em célula humanas. Assim, o objetivo geral deste estudo foi caracterizar a distribuição subcelular do ZIKV na célula hospedeira e elucidar fatores celulares que regulam o tráfego intracelular de proteínas envolvidos nesses processos. Mais especificamente, determinar os compartimentos celulares que servem de plataforma de montagem para o ZIKV. Além disso, também verificar se o funcionamento da maquinaria Endosomal Sorting Complexes Required for Transport (ESCRT) é requerido no ciclo replicativo de ZIKV. Para identificar a localização subcelular do ZIKV, foram utilizados diferentes marcadores celulares, e, de acordo com os resultados, foi demonstrado que com 3 horas pós infecção (h. p. i.) ocorre colocalização de proteínas do ZIKV com um marcador de endossomo primário, enquanto que com 15h p.i. já é possível detectar proteínas virais no Retículo Endoplasmático (RE). Subsequentemente, com 27h p.i. o ZIKV direciona-se para o complexo de Golgi. Juntos, esses resultados indicam o direcionamento do ZIKV através da via secretória ao longo do tempo. Além disso, foi testado o envolvimento da maquinaria dos ESCRTs por meio do silenciamento da expressão da proteína TSG101 de ESCRT-I em células infectadas com ZIKV. Os resultados obtidos, sugerem que ESCRT-I tem participação importante na replicação do ZIKV, ocorrendo a diminuição dos títulos virais quando TSG101 é depletada da célula. Em conjunto, os resultados permitem concluir que ao longo da infecção o ZIKV encontrase associado aos compartimentos da via secretória inicial (RE e complexo de Golgi), e que a proteína TSG101 de ESCRT-I exerce papel importante na replicação viral. Sendo assim, esse estudo possibilitou um melhor entendimento sobre a dinâmica de replicação do ZIKV em células humanas. Zika virus (ZIKV) is an arbovirus of the Flaviviridae family, of the genus Flavivirus that is transmitted by Aedes mosquitoes. Despite its emerging importance in public health, little is known about the molecular mechanisms involved in the replicative cycle of ZIKV in human cells. Thus, the general objective of this study was to characterize the subcellular distribution of the ZIKV in the host cell and to elucidate cellular factors that regulate the intracellular trafficking of proteins involved in these processes. More specifically, to determine the cellular compartments that serve as assembly platforms for the ZIKV. In addition, the study aimed to verify if the functioning of the Endosomal Sorting Complexes Required for Transport (ESCRT) machinery is required in the replicative cycle of ZIKV. In order to identify the subcellular localization of ZIKV, different intracellular markers were used, and, according to the results, it was demonstrated that at 3 hours post infection (h. p. i.) ZIKV proteins colocalize with an early endosome marker, whereas within 15h p.i. it is already possible to detect newlysynthesized viral proteins in the endoplasmic reticulum (ER). Subsequently, within 27h p.i., the ZIKV is directed to the Golgi complex. Together, these results delineate the targeting of ZIKV proteins through the secretory pathway over time. In addition, the involvement of the ESCRT machinery was tested by knocking down the expression of ESCRT-I protein TSG101 in ZIKV-infected cells. The results obtained suggest that ESCRT-I plays an important role in ZIKV replication, with viral titers decreasing when TSG101 levels are depleted in the cell. Together, the results allow us to conclude that ZIKV is associated with the initial secretory pathways (RE and Golgi complex) throughout the infection, and that the ESCRT-I TSG101 protein plays an important role in viral replication. Thus, this study contributes to a better understanding of the dynamics of ZIKV replication in human cells.

Human activities have being responsible for the global environmental changes, resulting in an increase number of incident of vector- and rodent-borne diseases worldwide. Rodents and arthropods-borne viruses are important globally emerging and re-emerging viruses and most of them are RNA viruses. Efficient and early diagnosis of these infections are very important to prevent their spread, to improve clinical management of the patients, as wells to protect livestock and domestic animals. Currently, available diagnostic methods such as immunoassays, polymerase chain reaction and virus isolation can detect only one or few viruses in a single assay. The DNA microarray platform has emerged as diagnostic tool suitable for high throughput screening of pathogenic agents. The aim of this study was to develop a DNA microarray platform (RoboArboVirusChip) for detecting rodent- and arthropod-borne viruses, which belong to seven families: Bunyaviridae (genera Orthobunyavirus, Nairovirus and Phlebovirus), Flaviviridae (genus Flavivirus), Togaviridae (genus Alphavirus), Reoviridae (genera Orbivirus, Seadornavirus and Coltvivirus), Rhabdoviridae (genera Vesiculovirus and Ephemerovirus), and Asfarviridae (genus Asfarvirus). Specific oligonucleotide probes of 60-mer (n=4209) targeting 412 virus species and generic probes of 25-35-mer (n=87) targeting viruses at the genus level were designed. A total of 17 reference viruses belonging to the Bunyaviridae, Flaviviridae, Rhabdoviridae and Togaviridae families were used to standardize RoboArboVirusChip. All reference viruses were specifically detected without any cross hybridization; however, the generic probes were not able to identify the viruses at the genus level. The RoboArboVirusChip was able to specifically identify four viruses contained in three different mixtures: i) virus of different families, ii) virus of the Flavivirus genus, and iii) the Dengue virus (DENV) serotypes. The four DENV serotypes were use to evaluate the sensitivity of the RoboArboVirusChip, which was able to detect a minimum of 25 RNA copies/mL of the viruses, confirming its high sensitivity. The applicability of the RoboArboVirusChip to detect viruses in clinical samples was tested with serum samples obtained from dengue suspected cases (four positive cases and 40 negative cases). DENV was detected in the four positive serum samples, while in the 40 negative serum samples, it was not detected any virus. The results obtained in this study suggest that the RoboArboVirusChip platform could be a useful tool for early diagnosis of robovirus and arbovirus infections during epidemic outbreaks, helping in the rapid implementation of disease containment strategies As atividades humanas têm sido responsável por mudanças ambientais globais, resultando num aumento do número de casos de doenças transmitidas por vetores e roedores em todo o mundo. Os vírus transmitidos por roedores e artrópodes são vírus emergentes e re-emergentes de importância global, sendo que a maioria deles são vírus de RNA. O diagnóstico eficiente e precoce dessas infecções são muito importantes para evitar a sua propagação, para melhorar o manejo clínico dos pacientes e também, para proteger o gado e os animais domésticos. Atualmente, os métodos de diagnóstico disponíveis, tais como os imunoensaios, a reação em cadeia da polimerase e o isolamento viral podem detectar apenas um ou poucos vírus em um único ensaio. A plataforma de microarranjo de DNA tem surgido como uma ferramenta de diagnóstico apropriada para o monitoramento em larga escala de agentes patogênicos. O objetivo deste estudo foi desenvolver uma plataforma de microarranjo de DNA (RoboArboVirusChip) para a detecção de vírus transmitidos por roedores e artrópodes, os quais pertencem a sete famílias: Bunyaviridae (gêneros Orthobunyavirus, Nairovirus e Phlebovírus), Flaviviridae (gênero Flavivirus), Togaviridae (gênero Alphavirus), Reoviridae (gênero Orbivirus, Seadornavirus e Coltvivirus), Rhabdoviridae (géneros Vesiculovirus e Ephemerovirus), e Asfarviridae (gênero Asfarvirus). Sondas oligonucleotídicas de 60-mer (n=4209) específicas contra 412 espécies virais, e sondas genéricas de 25-35-mer (n=87) para detecção de vírus a nível do gênero foram desenhados. Um total de 17 vírus de referência, pertencentes às famílias Bunyaviridae, Flaviviridae, Rhabdoviridae e Togaviridae foram utilizados para padronizar o RoboArboVirusChip. Todos os vírus de referência foram detectados especificamente sem apresentação de hibridação cruzada, porem as sondas genéricas não foram capazes de detectar os vírus a nível do gênero. O RoboArboVirusChip foi capaz de identificar especificamente quatro vírus contidos em diferentes misturas: i) vírus de diferentes famílias, ii) vírus pertencentes ao gênero a Flavivirus, e iii) os sorotipos do vírus da dengue (DENV). Os quatro sorotipos do DENV foram utilizados para determinar a sensibilidade do RoboArboVirusChip, o qual foi capaz de detectar um mínimo de 25 copias de RNA/mL. A aplicabilidade do RoboArboVirusChip para detectar vírus em amostras clínicas foi avaliada testando amostras de soro de pacientes com suspeita de dengue (quatro casos positivos e 40 casos negativos). Os resultados obtidos neste estudo sugerem que o RoboArboVirusChip poderá ser uma ferramenta útil para o diagnóstico precoce da infecção causada por robovírus e arbovírus, auxiliando na rápida implementação de estratégias de contenção das doenças causadas por esses vírus

O vírus sincicial respiratório humano (HRSV) é o mais frequente agente patogênico da família Paramyxoviridae. Apesar de sua grande importância e impacto em saúde pública, alguns aspectos demandam elucidação. Entre eles, estão os mecanismos de tráfego intracelular de proteínas virais para o sitio de montagem. Baseado nisso, fizemos um estudo de imunofluorescência tentando contribuir para o entendimento da participação da via secretória no tráfego de proteínas estruturais de HRSV que não são glicosiladas: proteínas de matriz (M) e de nucleocapsídeo (N). Pudemos observar que essas proteínas seguem rota similar àquelas que são glicosiladas no Golgi, como a proteína de fusão (F). Ademais, as proteínas M e N, além de colocalizarem com proteínas celulares da via secretória, tais como trans-Golgi network-46 (TGN46) e sorting nexin-2 (SNX2), também influem no recrutamento de proteínas celulares para os corpos de inclusão virais, como mostrado no caso da proteína Glut1. Os dados indicam que proteínas M e N de HRSV seguem pela via endocítica inicial, acumulam-se em corpos de inclusão que seriam fábricas virais e, no caso de TGN46, podem ser incorporadas aos vírus em brotamento Human respiratory syncytial virus (HRSV) is the most relevant cause of respiratory infection in children worldwide. Despite its importance in public health, some aspects of the mechanisms of the trafficking of viral structural proteins remain unclear. In the present study, immunofluorescence was used to understand how the virus matrix (M) and nucleocapsid (N) proteins, which are non-glycosylated , are addressed to inclusion bodies in Hep-2 cells (MOI=3). M and N proteins followed similar intracellular trafficking routes as compared to the glycosylated fusion (F) viral protein. Moreover, M and N proteins colocalized with two key elements of the secretory pathway: trans-Golgi network- 46 (TGN46) and sorting nexin-2 (SNX2). Viral proteins M and N appear to be involved in the recruitment of cell proteins at the formation of virus inclusion bodies, as shown for Glucose Transporter Type 1 (Glut1). The data suggest that HRSV M and N proteins follow the secretory pathway, initiating in early endosomes, as indicated by the co-localization with TGN46 and SNX2. In addition, these host cell proteins accumulate in inclusion bodies that are viral factories, and can be part of budding viral progeny. Therefore, HRSV M and N proteins, even though they are not glycosylated, take advantage of the secretory pathway to reach virus inclusion bodies. Confocal images suggest that SNX2, which is known for its membrane-deforming properties, could play a pivotal role in HRSV budding

O Vírus da Imunodeficiência Humana (HIV) é o agente etiológico da Síndrome da Imunodeficiência Adquirida (AIDS). A AIDS é uma doença de distribuição mundial, e estima-se que existam atualmente pelo menos 36,9 milhões de pessoas infectadas com o vírus. Durante o seu ciclo replicativo, o HIV promove diversas alterações na fisiologia da célula hospedeira a fim de promover sua sobrevivência e potencializar a replicação. A rápida progressão da infecção pelo HIV-1 em humanos e em modelos animais está intimamente ligada à função da proteína acessória Nef. Dentre as diversas ações de Nef está a regulação negativa de proteínas importantes na resposta imunológica, como o receptor CD4. Sabe-se que esta ação resulta da indução da degradação de CD4 em lisossomos, mas os mecanismos moleculares envolvidos ainda são totalmente elucidados. Nef forma um complexo tripartite com a cauda citosólica de CD4 e a proteína adaptadora 2 (AP-2), em vesículas revestidas por clatrina nascentes, induzindo a internalização e degradação lisossomal de CD4. Pesquisas anteriores demonstraram que o direcionamento de CD4 aos lisossomos por Nef envolve a entrada do receptor na via dos corpos multivesiculares (MVBs), por um mecanismo atípico, pois, embora não necessite da ubiquitinação de carga, depende da ação de proteínas que compõem os ESCRTs (Endosomal Sorting Complexes Required for Transport) e da ação de Alix, uma proteína acessória da maquinaria ESCRT. Já foi reportado que Nef interage com subunidades dos complexos AP-1, AP-2, AP-3 e Nef não parece interagir com subunidades de AP-4 e AP-5. Entretanto, o papel da interação de Nef com AP-1 e AP-3 na regulação negativa de CD4 ainda não está totalmente elucidado. Ademais, AP-1, AP-2 e AP-3 são potencialmente heterogêneos devido à existência de isoformas múltiplas das subunidades codificadas por diferentes genes. Todavia, existem poucos estudos para demonstrar se as diferentes combinações de isoformas dos APs são formadas e se possuem propriedades funcionais distintas. O presente trabalho procurou identificar e caracterizar fatores celulares envolvidos na regulação do tráfego intracelular de proteínas no processo de regulação negativa de CD4 induzido por Nef. Mais especificamente, este estudo buscou caracterizar a participação do complexo AP-1 na modulação negativa de CD4 por Nef de HIV-1, através do estudo funcional das duas isoformas de ?-adaptina, subunidades de AP-1. Utilizando a técnica de Pull-down demonstramos que Nef é capaz de interagir com ?2. Além disso, nossos dados de Imunoblot indicaram que a proteína ?2-adaptina, e não ?1-adaptina, é necessária no processo de degradação lisossomal de CD4 por Nef e que esta participação é conservada para degradação de CD4 por Nef de diferentes cepas virais. Ademais, por citometria de fluxo, o silenciamento de ?2, e não de ?1, compromete a diminuição dos níveis de CD4 por Nef da membrana plasmática. A análise por imunofluorêsncia indireta também revelou que a diminuição dos níveis de ?2 impede a redistribuição de CD4 por Nef para regiões perinucleares, acarretando no acúmulo de CD4, retirados por Nef da membrana plasmática, em endossomos primários. A depleção de ?1A, outra subunidade de AP-1, acarretou na diminuição dos níveis celulares de ?2 e ?1, bem como, no comprometimento da eficiente degradação de CD4 por Nef. Além disso, foi possível observar que, ao perturbar a maquinaria ESCRT via super-expressão de HRS (uma subunidade do complexo ESCRT-0), ocorreu um acumulo de ?2 em endossomos dilatados contendo HRS-GFP, nos quais também detectou-se CD4 que foi internalizado por Nef. Em conjunto, os resultados indicam que ?2-adaptina é uma importante molécula para o direcionamento de CD4 por Nef para a via ESCRT/MVB, mostrando ser uma proteína relevante no sistema endo-lisossomal. Ademais, os resultados indicaram que as isoformas ?-adaptinas não só possuem funções distintas, mas também parecem compor complexos AP-1 com diferentes funções celulares, já que apenas a variante AP-1 contendo ?2, mas não ?1, participa da regulação negativa de CD4 por Nef. Estes estudos contribuem para o melhor entendimento dos mecanismos moleculares envolvidos na atividade de Nef, que poderão também ajudar na melhor compreensão da patogênese do HIV e da síndrome relacionada. Em adição, este trabalho contribui para o entendimento de processos fundamentais da regulação do tráfego de proteínas transmembrana no sistema endo-lisossomal. The Human Immunodeficiency Virus (HIV) is the etiologic agent of Acquired Immunodeficiency Syndrome (AIDS). AIDS is a disease which has a global distribution, and it is estimated that there are currently at least 36.9 million people infected with the virus. During the replication cycle, HIV promotes several changes in the physiology of the host cell to promote their survival and enhance replication. The fast progression of HIV-1 in humans and animal models is closely linked to the function of an accessory protein Nef. Among several actions of Nef, one is the most important is the down-regulation of proteins from the immune response, such as the CD4 receptor. It is known that this action causes CD4 degradation in lysosome, but the molecular mechanisms are still incompletely understood. Nef forms a tripartite complex with the cytosolic tail of the CD4 and adapter protein 2 (AP-2) in clathrin-coated vesicles, inducing CD4 internalization and lysosome degradation. Previous research has demonstrated that CD4 target to lysosomes by Nef involves targeting of this receptor to multivesicular bodies (MVBs) pathway by an atypical mechanism because, although not need charging ubiquitination, depends on the proteins from ESCRTs (Endosomal Sorting Complexes Required for Transport) machinery and the action of Alix, an accessory protein ESCRT machinery. It has been reported that Nef interacts with subunits of AP- 1, AP-2, AP-3 complexes and Nef does not appear to interact with AP-4 and AP-5 subunits. However, the role of Nef interaction with AP-1 or AP-3 in CD4 down-regulation is poorly understood. Furthermore, AP-1, AP-2 and AP-3 are potentially heterogeneous due to the existence of multiple subunits isoforms encoded by different genes. However, there are few studies to demonstrate if the different combinations of APs isoforms are form and if they have distinct functional properties. This study aim to identify and characterize cellular factors involved on CD4 down-modulation induced by Nef from HIV-1. More specifically, this study aimed to characterize the involvement of AP-1 complex in the down-regulation of CD4 by Nef HIV-1 through the functional study of the two isoforms of ?-adaptins, AP-1 subunits. By pull-down technique, we showed that Nef is able to interact with ?2. In addition, our data from immunoblots indicated that ?2- adaptin, not ?1-adaptin, is required in Nef-mediated targeting of CD4 to lysosomes and the ?2 participation in this process is conserved by Nef from different viral strains. Furthermore, by flow cytometry assay, ?2 depletion, but not ?1 depletion, compromises the reduction of surface CD4 levels induced by Nef. Immunofluorescence microscopy analysis also revealed that ?2 depletion impairs the redistribution of CD4 by Nef to juxtanuclear region, resulting in CD4 accumulation in primary endosomes. Knockdown of ?1A, another subunit of AP-1, resulted in decreased cellular levels of ?1 and ?2 and, compromising the efficient CD4 degradation by Nef. Moreover, upon artificially stabilizing ESCRT-I in early endosomes, via overexpression of HRS, internalized CD4 accumulates in enlarged HRS-GFP positive endosomes, where co-localize with ?2. Together, the results indicate that ?2-adaptin is a molecule that is essential for CD4 targeting by Nef to ESCRT/MVB pathway, being an important protein in the endo-lysosomal system. Furthermore, the results indicate that ?-adaptins isoforms not only have different functions, but also seem to compose AP-1 complex with distinct cell functions, and only the AP-1 variant comprising ?2, but not ?1, acts in the CD4 down-regulation induced by Nef. These studies contribute to a better understanding on the molecular mechanisms involved in Nef activities, which may also help to improve the understanding of the HIV pathogenesis and the related syndrome. In addition, this work contributes with the understanding of primordial process regulation on intracellular trafficking of transmembrane proteins.

O Vírus Sincicial respiratório (HRSV) é um patógeno de grande importância para crianças. É o maior causador de infecções respiratórias agudas e também um dos principais causadores de internações e mortes de crianças menores de 5 anos. Cepas de HRSV com mutações no sítio de ligação do Palivizumab vêm apresentando resistência ao medicamento. Pouco se sabe sobre a prevalência destas mutações em amostras clínicas, apesar de sua potencial importância na patogênese viral. Além disso, existem poucos dados sobre a evolução molecular da proteína F do HRSV, sobretudo no Brasil. O presente estudo tem como objetivo caracterizar fenotipicamente através de ensaios de microneutralização in vitro, cepas oriundas de crianças não tratadas com Palivizumab circulantes em 2013 apresentando mutações na proteína F, mais especificamente relacionadas a potencial resistência nos epítopos de ligação do Palivizumab. Como resultado deste trabalho todas as amostras testadas foram neutralizadas pelo Palivizumab, concluindo-se que as mutações encontradas não conferem resistência ao monoclonal. Respiratory Syncytial Virus (HRSV) is a pathogen of great importance for children. It is the major cause of acute respiratory infections and also one of the main causes of hospitalizations and deaths of children under 5 years. Strains of HRSV with mutations in the binding site of Palivizumab have been showing resistance to the drug. Little is known about the prevalence of these mutations in clinical samples, despite their potential importance in viral pathology. In addition, there is little data on the molecular evolution of HRSV F protein, especially in Brazil. The present study aims to characterize phenotypically by in vitro microneutralization assays, strains from children not treated with Palivizumab circulating in 2013 showing mutations in F protein, more specifically related to potential resistance in the binding epitopes of Palivizumab. As result of this study all samples tested were neutralized by Palivizumab, concluding that the mutations found did not confer resistance to the monoclonal.

O arbovírus Oropouche (OROV), da família Peribunyaviridae, gênero Orthobunyavirus, é importante causador de doença febril nas Américas Central e do Sul. Como outros Orthobunyavirus, OROV é envelopado, com genoma compreendido por três segmentos de RNA de fita simples de polaridade negativa, nomeados de acordo com o seu tamanho, como S (small), M (médio) e L (large). O segmento S codifica a proteína estrutural do nucleocapsideo N e, em uma fase de leitura alternativa, a proteína não estrutural NSs. Apesar da patogênese de OROV ser pouco conhecida, dados obtidos com modelos experimentais murinos indicam que a apoptose desempenha um papel central na morte de células infectadas. Portanto, embora não seja essencial à replicação de OROV, NSs pode desempenhar papeis importantes para a patogênese. Nosso grupo demonstrou previamente que OROV induz apoptose em células HeLa pela via intrínseca, e esse efeito requer a síntese de proteínas virais. No presente estudo objetivamos investigar o papel da proteína NSs de OROV na apoptose induzida em células HeLa. Nós encontramos que a superexpressão da proteína NSs de OROV induz apoptose em diferentes linhagens de células humanas (Glioblastoma, Huh7 e HCE) 24 horas após transfecção. NSs induz apoptose pela via intrínseca, com liberação de citocromo c da mitocôndria, ativação de caspases 9 e 3, e fragmentação de cromatina. Curiosamente, a proteína NSs também induz apoptose pela via extrínseca, confirmada pela detecção de caspase 8 ativada. Um clone infeccioso do OROV sem a proteína NSs (OROVdelNSs) também não induz apoptose em células HeLa até 36 horas pós-infecção, confirmando a importância da proteína NSs para a indução de morte celular por apoptose. A predição da estrutura da proteína NSs revelou a presença de uma sequência de treze aminoácidos na região C-terminal da proteína, que tem similaridade como receptor de TNF. Ensaio de PCR array indicou que a proteína NSs de OROV causa aumento na quantidade de transcritos de TNF em células HeLa. The arbovirus Oropouche (OROV), of the family Peribunyaviridae, genus Orthobunyavirus, is an important cause of febrile disease in Central and South America. Like other Orthobunyaviruses, OROV is enveloped, with a genome comprised of three segments of single-stranded RNA of negative polarity, named according to their sizes as S (small), M (medium) and L (large). The S segment encodes the structural nucleocapsid N protein and, on an alternative reading frame, the NSs nonstructural protein. Although the pathogenesis of OROV is poorly understood, data obtained from murine experimental models indicate that apoptosis plays a central role in killing infected cells. Therefore, while not essential for the replication of OROV, NSs may play important roles in the pathogenesis. Our group has previously demonstrated that OROV induces apoptosis in HeLa cells via the intrinsic pathway, and this effect requires the synthesis of viral proteins. The present study aimed to investigate the role of OROV protein NSs in apoptosis induced in HeLa cells. We found that overexpression of the NSs protein induces apoptosis in different human cell lines (glioblastoma neuroblastoma and human corneal epithelium) 24 hours after transfection. NSs induces apoptosis by the intrinsic pathway, with release of cytochrome c from mitochondria, activation of caspases 9 and 3, and chromatin fragmentation. Interestingly, the NSs protein also induces activation of caspase 8, an element of the extrinsic pathway of apoptosis. An OROV infectious clone lacking the NSs protein (OROVdelNSs) does not induce apoptosis in HeLa cells up to 36 hours postinfection, confirming the importance of the NSs protein for the induction of cell death by apoptosis. The prediction of the structure of the NSs protein revealed the presence of a sequence of thirteen amino acids in the C-terminal region of the protein, which has similarity as a TNF receptor, and this could shed light into a possible mechanism of apoptosis to be validated by experiments silencing target genes in Hela cells.

O vírus Oropouche (OROV) é um arbovírus que ocorre na região amazônica causando surtos de doenças febris agudas e que, ocasionalmente, podem ser associados a meningoencefalite. Aproximadamente 500.000 casos de Oropouche teriam ocorrido no Brasil. Entretanto, não existe vacina contra o OROV. O objetivo deste trabalho foi desenvolver um modelo animal de infecção por OROV para estudar a patogênese da doença e um modelo para testar candidatas vacinais. Protótipo vacinal utilizando a proteína recombinante do nucleocapsídeo (N) de OROV (NrOROV), que é o principal antígeno viral, foi usado como potencial candidato para vacina. Neste estudo utilizou-se um modelo animal em camundongos Balb/c de 12 semanas de idade, inoculados intracerebralmente com 8x105 PFU de OROV, capaz de induzir 100% de letalidade após o terceiro dia da infecção. Altos títulos virais foram encontrados no cérebro e na medula espinhal dos animais. Surpreendentemente, 12 e 24 horas pós-infecção foi possível detectar vírus no fígado e baço (3 Log10 PFU/g) dos camundongos. Com este modelo foram testados os candidatos vacinais. Grupos de camundongos foram imunizados 3 vezes com OROV, OROV e FCA, NrOROV, NrOROV e FCA, NrOROV, Poli I:C e Montanide ISA 720. Após 3 imunizações, os animais foram desafiados com 10 LD50 de OROV e observados por 20 dias. Os animais imunizados com NrOROV e adjuvantes, não foram capazes de produzir anticorpos neutralizantes e adquirir imunidade protetora contra OROV enquanto que os imunizados com OROV apresentaram altos níveis de anticorpos neutralizantes e completa proteção in vivo. Ainda, os anticorpos produzidos pelos animais imunizados permitiram estudar o ciclo de replicação celular do OROV utilizando imunofluorescência. Oropouche (OROV) is an arbovirus that occurs in the South American, Amazon region, producing outbreaks of acute febrile illness occasionally associated to meningoencephalitis. Approximately 500,000 cases of Oropouche have been reported in Brazil in the last 60 years. However, there is no available vaccine for OROV. We show here the development of an animal model of OROV suitable for studies on pathogenesis and vaccine testing. A vaccine prototype based on recombinant OROV nucleocapsid protein (NrOROV), an important viral antigen, was evaluated in the animal model. Initialy, we observed that all 12-week-old Balb/c mice inoculated intracerebrally with 8x105 PFU died after the third day of infection. Surprisingly, OROV genome was detectable in the liver as early as 12 hours post infection (pi) and in the spleen at 24 hours pi at 3 log10 PFU/g. Besides, high viral titers were found in brain and spinal cord. To test the NrOROV as a vaccine candidate, animals divided in 5 groups were immunized subcutaneously 3 times, two weeks apart with either OROV, OROV and Freud complete Adjuvant (FCA), NrOROV, NrOROV and FCA, NrOROV and Poly I:C and Montanide ISA 720. The experiment also included a group of naïve animals. After the third immunization, the animals were challenged with 10LD50 by intracerebral route and followed for 20 days. The animals immunized with NrOROV and adjuvants developed specific antibodies that were not able to neutralize the virus or confer protective immunity against OROV. Nevertheless, mice immunized with OROV showed high levels of neutralizing and protective antibodies. Despite the discouraging results with NrOROV as a vaccine, the mouse model is suitable to study pathogenesis, and to test other vaccines for OROV.

A glicoproteína gp120 do HIV apresenta domínios conservados, dentre os quais está o C4. Este domínio está envolvido no reconhecimento da molécula CD4 na superfície das células alvo, e anticorpos dirigidos contra ele neutralizam o vírus. Em trabalho anterior, este domínio foi introduzido dentro da região globular da proteína fibra do adenovírus. Como a fibra adenoviral é estimulante do sistema imune, decidimos testar a capacidade de indução de anticorpos anti-C4 por essa proteína modificada. Assim, foram construídos vetores plasmídicos e adenovirais portando o gene da região globular da fibra adenoviral contendo o domínio C4, que usados na imunização de camundongos induziram anticorpos capazes de reconhecer a proteína gp120. Também construímos um vetor baculoviral expressando essa proteína híbrida, que purificada por HPLC, foi utilizada para imunizar camundongos que também produziram anticorpos capazes de reconhecer a proteína gp120. Nossos dados sugerem que a região globular da fibra adenoviral é uma boa plataforma para a exposição de epítopos de imunização. HIV glycoprotein gp120 has conserved domains, one of them being the C4 domain. This region is involved in the recognition of the CD4 marker in target cells and antibodies that recognize this domain can block HIV infection. Previously, the C4 domain was introduced in the adenovirus fiber knob. As the adenovirus fiber stimulates de immune system, we decided to test the production of anti-C4 antibodies by this hybrid protein. We constructed plasmid and adenovirus vectors carrying the fiber knob modified with the C4 domain. Immunization of mice with these vectors showed the production of specific antibodies that recognized de gp120 glycoprotein. Also, we constructed a baculovirus vector expressing the hybrid protein, which was purified by HPLC. Mice immunized with this protein also produced antibodies capable of recognizing gp120. Our data suggest that the fiber knob is a good carrier protein for epitope immunization.

O vírus Oropouche pertence à família Bunyaviridae, gênero Orthobunyavirus, sorogrupo Simbu, e é segunda causa mais freqüente de arbovirose febril no Brasil. Estima-se que mais de meio milhão de casos de febre do Oropouche tenham ocorrido no Brasil nos últimos 30 anos, havendo também ocorrências no Panamá, Peru, Suriname e Trinidad. Epidemias de febre do Oropouche têm sido registradas quase que exclusivamente na Amazônia. Porém, com o aquecimento global do planeta, desmatamentos e conseqüente redistribuição de insetos vetores e animais reservatórios, há risco de disseminação de vírus Oropouche para outras regiões do Brasil e da América do Sul. A patogenia da infecção por Oropouche não é bem entendida. Modelo em roedores, usando inoculação intracerebral, tem sido descrito, mas não com uso da via subcutânea, que mais se assemelha à rota natural da infecção. O objetivo deste trabalho foi estabelecer e caracterizar um modelo experimental de infecção com Oropouche, usando inoculação subcutânea em hamster, a fim de contribuir para o entendimento da patogenia do mesmo. Oropouche da linhagem BeAn19991, passado em cérebro de camundongo recém-nascido, foi inoculado (106,25 TCID50/100?L) por via subcutânea na coxa de hamsters sírios com 2-3 semanas de idade. Em torno de 3 dias alguns animais desenvolveram doença com sinais clínicos brandos caracterizados por: eriçamento dos pêlos e agressividade, outros com características mais graves: perda de peso, tremores sugestivos de calafrios, letargia e paralisia. Os animais foram sacrificados 1, 3, 5, 8 e 11 dias pós-inoculação ou quando eram encontrados em estado agônico. Baço, cérebro, coração, fígado, músculo e sangue foram colhidos para titulação viral, histologia e imunohistoquímica. Infiltrado inflamatório estava presente no cérebro, principalmente em torno dos vasos, meninges e discretamente no coração. O vírus Oropouche foi detectado no tecido cerebral (107,23 TCID50/g), no fígado (106,67TCID50/g) e no sangue (106 TCID50/g). Antígeno de Oropouche foi encontrado, difusamente no fígado, associado com hepatócitos e em neurônios de diversos locais do cérebro. Este modelo reproduz uma infecção sistêmica por Oropouche e pode tornar-se útil no estudo da patogênese, bem como para testar drogas antivirais e possíveis candidatos à vacina. Oropouche virus belongs to the family Bunyaviridae, genus Orthobunyavirus, serogroup Simbu and is the second most frequent arboviral febrile illness in Brazil. There are estimates of more than half million cases of Oropouche fever in Brazil in the past 30 years, with cases registered in Panama, Peru, Suriname and Trinidad. Oropouche fever has been registered almost exclusively in the Amazon, but with the global warming, deforestation and the consequent redistribution of vectors and reservoir animals, the risk of Oropouche virus dissemination to others areas of Brazil and South America increases. However, the pathogenesis of Oropouche infection is not well understood. Rodent models using intracerebral inoculation have been described, but no attempts to use a route that more closely resembles the natural route of infection have been published. This study was conducted to establish and characterize an experimental model of infection with Oropouche, using subcutaneous inoculation of hamsters, in order to contribute to the understanding of Oropouche pathogenesis. We have established an experimental model through subcutaneous inoculation of hamsters in order to study pathogenesis. Suckling mouse brain passaged Oropouche strain BeAn19991 was inoculated (106,25 TCID50/100?L) subcutaneously in the thigh of syrian hamsters 2-3 weeks old. Around day 3 animals developed disease characterized by lethargy, paralysis, chill-like shaking and ruffled fur. Animals were sacrificed on days 1, 3, 5, 8 and 11 post-inoculation or whenever found to be agonic. Brain, heart, liver, spleen, muscle and blood were harvested for virus titration, histology and Oropouche immunohistochemistry. Inflammatory infiltrate was present in the brain, mostly as perivascular cuffs, meninges and some in the heart. Oropouche titers were 107,23 TCID50/g of tissue in brain 106,67 TCID50/g in liver, and 106 TCID50/g in blood. Oropouche antigen was detected diffusely distributed in the liver in association with hepatocytes, and in neurons in several regions of the brain. This model reproduces systemic Oropouche infection and may become useful in pathogenesis studies, as well as to test antiviral drugs and possible vaccine candidates.

As infecções humanas com Leishmania spp dão origem a um amplo espectro de manifestações clínicas que vão desde lesões tegumentares até compromisso visceral. O fenótipo da doença está intimamente relacionado com o tipo de resposta do hospedeiro mamífero à infecção do parasito e à variabilidade genética entre as espécies. Neste contexto, a identificação e caracterização de genes que regulam a virulência no fenótipo de Leishmania são essenciais para a compreensão dos mecanismos patogênicos do hospedeiro-parasita. Estudos anteriores realizados no laboratório mostraram a atenuação da virulência através da superexpressão do SL RNA em transfectantes de Leishmania (L.) major e Leishmania (V.) braziliensis em modelo murino. Para entender as mudanças fenotípicas encontradas em Leishmania (L.) major, a análise do perfil de expressão de genes permitiu identificar seis genes down-regulados e up-regulados, que poderiam contribuir individualmente com um padrão de virulência. Seis genes foram selecionados, os quais estão diferencialmente expressos entre as linhagens controle e transfectante. A obrigatoriedade de realização do Teste de Ácidos Nucleicos (NAT) na triagem laboratorial dos candidatos à doação de sangue, permite o aumento da segurança transfusional, uma vez que o teste detecta precocemente o material genético dos vírus transmissíveis por transfusão. Com a introdução do NAT na prática diária dos serviços de hemoterapia brasileiros, foi também preconizada a utilização do controle interno de qualidade (CQI) como um parâmetro adicional para liberação de resultados com maior confiabilidade. Este manual técnico tem como objetivo subsidiar os serviços de hemoterapia na implantação e análise do CQI-NAT, auxiliando nas ações de melhoria contínua dos processos. Para tanto, foram implantados e validados no laboratório NAT, do hemocentro de Ribeirão Preto, nove procedimentos operacionais que contemplam todo o fluxo de produção das amostras para CQI-NAT, desde sua validação com testes de homogeneidade e estabilidade, definição dos limites dos gráficos de controle e utilização na rotina laboratorial, possibilitando a identificação de erros sistemáticos ou aleatórios que possam prejudicar os resultados da rotina. Os Vesiculovirus são um gênero de vírus de RNA da família Rhabdoviridae que inclui os sorotipos Carajás, Cocal, Marabá, Piry, Alagoas e Indiana. Estes são causadores de estomatite vesicular em ruminantes e doença febril humana no Brasil. As vesiculoviroses e suas epidemiologias são pouco conhecidas em seres humanos. Ainda, os Vesiculovirus (VSV) são pouco diagnosticados no homem e em animais pela escassez de métodos laboratoriais diagnósticos. Por isso, objetivamos neste trabalho desenvolver e testar uma RT-PCR em tempo real, pelo método SYBR Green, visando à detecção de VSV brasileiros. Primers que amplificam parte do gene da proteína G dos VSV foram utilizados no teste o qual mostrou-se capaz de detectar genomas dos VSV Piry, Indiana, Alagoas e Carajás. O método foi usado para testar amostras séricas de pacientes com doença febril aguda, de bovinos, de equinos e de macerados de artrópodos. A RT-PCR em tempo real mostrou-se 100 vezes mais sensível que a RT-PCR convencional para Vesiculovirus e também, permitiu detectar até 10 cópias de RNA do vírus Piry. Ainda, a RT-PCR em tempo real para Vesiculovirus mostrou-se capaz de diagnosticar e quantificar o VSV Alagoas nas amostras séricas de bovinos e de equinos. Portanto, a RT-PCR em tempo real desenvolvida neste trabalho, provavelmente, será muito útil no diagnóstico e também, em futuras pesquisas, que permitirão ampliar o conhecimento epidemiológico, ainda pouco conhecido, sobre os Vesiculovirus. The Vesiculovirus is a Rhabdoviridae family genre of RNA virus that includes serotypes Carajás, Cocal, Maraba, Piry, Alagoas and Indiana. These are causes of vesicular stomatitis in ruminants and human febrile illness in Brazil. The vesiculoviroses and its epidemiology are little known in humans. Still, Vesiculovirus (VSV) are poorly diagnosed in humans and laboratory animals by the lack of diagnostic methods. Therefore, we proposed in this work to develop and test a real-time RT-PCR by SYBR Green method, focusing on the detection of Brazilian VSV. Primers that amplify part of the VSV G protein gene were used in the test which proved capable of detecting genomes of VSV Piry, Indiana, Alagoas and Carajás. The method was used to test serum samples from patients with acute febrile disease, cattle, horses and macerated arthropods. Real time RT-PCR showed to be 100 times more sensitive than conventional RT-PCR for Vesiculovirus and also was possible to detect up to 10 RNA copies of the Piry virus. Also, the real-time RT-PCR for Vesiculovirus proved able to diagnose and quantify Alagoas VSV in serum samples from cattle and horses. Therefore, the real-time RT-PCR developed in this work will probably be very useful in the diagnosis and in future research, which will increase the epidemiological knowledge, as it is still little known about the Vesiculovirus.

A dengue é uma doença infecciosa causada pelo vírus da dengue (DENV) e transmitida principalmente pela picada de mosquitos Aedes aegypti. A dengue é a doença viral transmitida por artrópodes de maior importância em saúde pública, afetando principalmente a países tropicais e subtropicais do mundo. As epidemias de dengue têm aumentado consideravelmente nos últimos anos em todo o mundo e as fronteiras de circulação do vírus vem se expandido constantemente. Assim, estudos de diferentes aspectos da doença e do vírus são de grande importância para aperfeiçoar os conhecimentos sobre esta ameaça. Neste sentido, é importante que algumas análises, como as filogenéticas e evolutivas dos vírus sejam realizadas para identificação dos genótipos circulantes, a origem dos mesmos, o relacionamento com outros subtipos e a evolução sofrida ao longo do tempo. Este estudo teve por objetivo analisar o relacionamento filogenético e evolutivo dos DENV isolados em Ribeirão Preto entre 2010 e 2011. Amostras de soro (n=79) de pacientes com dengue estocadas a -80ºC foram inoculadas em células C6/36 para tentativa de isolamento viral, o qual foi confirmado a partir de 39 amostras por imunofluorescência indireta e/ou RT-PCR em tempo real. Sequenciamento de parte do gene da proteína viral NS5 ou do gene da proteína E mostrou que 25 pertenciam ao DENV-1, seis ao DENV-2 e oito ao DENV-3. Para as análises filogenéticas e evolutivas, o gene da proteína E de 15 DENV-1, quatro DENV-2 e um DENV-3 foi sequenciado. Estas análises foram realizadas também utilizando toda a região codificadora de dois DENV-1, um DENV-2 e um DENV-3. As análises mostraram que todos os vírus introduzidos em Ribeirão Preto foram provenientes de vírus originados no Estado do Rio de Janeiro. Duas linhagens de DENV-1, uma de DENV-2 e uma de DENV-3 circularam em Ribeirão Preto entre 2010 e 2011. O relacionamento filogenético dos vírus foi similar independentemente do uso da sequência do gene da proteína E ou de toda a região codificadora. Dengue is the viral disease transmitted by arthropods of greater importance in public health, particularly affecting the tropical and subtropical countries of the world. Dengue epidemics have increased considerably in recent years throughout the world and the borders of virus circulation have been expanding constantly. Thus studies of different aspects of the disease and the virus are of great importance to improve knowledge about this threat. In this sense, it is important that some analyzes such as phylogeny and evolution be carried out to identify the circulating genotypes, their origin, their relationship with other subtypes and evolution along the time. This study aimed to analyze the phylogenetic and evolutionary relationships of DENV isolated in Ribeirão Preto between 2010 and 2011. Serum samples (n = 79) of dengue patients stored at -80°C were inoculated into C6/36 cells for virus isolation attempts, which was confirmed from 39 samples by indirect immunofluorescence and/or real-time RT-PCR. Sequencing of part of the viral NS5 gene protein or the E gene protein showed that 25 belonged to DENV-1, six to DENV-2 and eight to DENV-3. For the phylogenetic and evolutionary analyzes, the E gene protein of 15 DENV-1, four DENV-2 and one DENV-3 was sequenced. These analyzes were also carried out with the entire coding region of two DENV-1, one DENV-2 and one DENV-3. The analysis showed that all viruses were introduced in Ribeirão Preto from viruses originated in the state of Rio de Janeiro. Two lineages of DENV-1, one of DENV-2 and one of DENV-3 circulated between 2010 and 2011. The phylogenetic relationship of these viruses was similar regardless of the use of the E gene protein or the entire coding region sequences.

A dengue é uma doença infecciosa com grandes taxas de morbimortalidade, causada pelo vírus da dengue (DENV). Segundo a Organização Mundial de Saúde, cerca de 50 a 100 milhões de pessoas são infectadas anualmente em mais de 100 países tropicais e subtropicais de todos os continentes. O espectro clínico da infecção pelo DENV pode incluir formas assintomáticas ou sintomaticas que variam desde uma febre indeterminada e autolimitada, passando pela febre clássica da dengue (FD) até quadros graves denominados febre hemorrágica da dengue/síndrome do choque da dengue (FHD/SCD). Recentemente, ocorreu um dramático aumento do número de casos de FHD/SCD nas Américas, e este aumento coincidiu com a introdução do dengue sorotipo 3, genótipo III. No presente trabalho, objetivou-se a clonagem e a expressão das proteínas NS1 e NS3 do vírus da dengue tipo 3. As proteínas NS1 e NS3 do DENV-3 foram clonadas e expressas com sucesso em sistema procarioto. A amplificação dos genes das proteínas NS1 e NS3 foi realizada por RT-PCR, o qual gerou amplicons de cerca de 1050 e 1850 pb, respectivamente. Em seguida, os genes foram clonados por inserção dos amplicons no vetor plasmidial pCR-XL. Os genes de NS1 e NS3 foram subclonados no vetor de expressão pQE-30 através de sítios de restrição para as enzimas BamHI e HindIII. A expressão proteica foi obtida em sistema procarioto utilizando a cepa BL21(DE3) de E. coli, resultando em proteínas de 45 e 70 kDa as quais foram confirmadas por análises em Western blot utilizando como anticorpo primário fluido ascítico imune de camundongos e soro de pacientes com dengue. Estas proteínas virais podem ser utilizadas para estudos relacionados à patogênese, replicação e mecanismos de escape do sistema imune do DENV, além disso, podem ser potencias antígenos em métodos de diagnóstico. Dengue is an infectious disease with high morbidity and mortality rates caused by dengue virus (DENV). According to the World Health Organization, about 50 to 100 million people are infected annually in more than 100 tropical and subtropical countries from all continents. The clinical spectrum of DENV infection can includes asymptomatic or symptomatic forms ranging from undetermined and self-limited fever, through dengue fever (DF) to severe disease called dengue hemorrhagic fever/dengue shock syndrome (DHF/DSS). Recently, there has been a dramatic increase in the number of cases of DHF/DSS in the Americas, and this increase coincided with the introduction of dengue virus type 3 (DENV-3), genotype III. The present study aimed to clone and express NS1 and NS3 proteins of DENV-3. The NS1 and NS3 proteins of DENV-3 was successfully cloned and expressed in a prokaryotic system. Amplification of NS1 and NS3 genes was carried out by RT-PCR, which yielded amplicons of approximately 1050 and 1850 bp, respectively. Then, the genes were cloned by inserting the amplicons into the plasmid vector pCR-XL. NS1 and NS3 genes were subcloned into the expression vector pQE-30 through the restriction sites for BamHI and HindIII enzymes. The protein expression was obtained in a prokaryotic system using the strain BL21 (DE3) of E. coli, resulting in 45 and 70 kDa proteins, which were confirmed by Western blot analysis using immune mouse ascitic fluid and serum of patients with dengue as primary antibody. These viral proteins can be used to study the pathogenesis, mechanisms of replication and immune escape of DENV, moreover, can be potential antigens in diagnostic methods.

O estudo dos protozoários é importante por diversos motivos, entre eles sua diversidade, sua importância evolucionária e impacto na saúde pública. Muitos aspectos tornam o estudo desses seres vivo ainda mais fascinantes, como por exemplo, fenômenos como o trans-splincing e a presença de um vírus em algumas espécies de leishmanias. A proteína U5-15k é considerada de grande importância fazendo parte do spliciossoma. Neste trabalho tivemos o objetivo de iniciar a caracterização desta proteína empregando ferramentas de bioinformática para analisar sua sequencia de aminoácidos, sua estrutura secundária e modelar sua estrutura por homologia, também foi possível otimizar sua expressão, purificação, caracterizar sua auto clivagem e fazer estudos biofísicos de Espalhamento Dinâmico de Luz e Espectroscopia de Dicroísmo Circular. O Leishmania RNA vírus 1-4 (LRV1-4) é um vírus da família Totiviridae de capsídeo icosaédrico que codifica duas proteínas (proteína capsidial e RNA polimerase). Sendo pouco estudado tivemos o objetivo de iniciar a caracterização molecular deste vírus com objetivos futuros de empreender estudos estruturais e funcionais. Não foi possível obter expressão recombinante em nenhuma das duas proteínas virais, por isso se optou por se estabelecer um novo protocolo de lise celular e purificação com o qual se obteve imagens inéditas de Microscopia Eletrônica de Varredura, na qual o vírus é purificado ainda envolto em material genético. The study of protozoa is important for several reasons, including their diversity, their evolutionary importance and public health impact. Many aspects make the protozoas study even more exciting, for example, phenomena such as trans-splincing and the presence of virus in some species of Leishmania. The U5-15k protein is considered of great importance as part of the spliciossome machinery. In this work we intended to characterize this protein , this using bioinformatics tools to analyze its amino acid sequence, its secondary structure and its structure by homology modeling, it was possible to optimize expression, purification, characterization and make biophysical studies of their self cleavage of Dynamic Light Scattering and Spectroscopy of Circular Dichroism. The Leishmania RNA virus 1-4 (LRV1-4) is a virus belonging to the Totiviridae family with an icosahedral capsid structure that encodes two proteins (a capsid major protein and an RNA polymerase). It was not possible to obtain the recombinant expression of any of the two viral proteins, so it was decided to establish a new protocol for cell lysis and purification with which it was obtained unprecedented images of Transmission electron microscopy, in which the virus is further purified wrap on genetic material.

Leishmaniavirus (LRV) é um vírus de RNA de fita dupla que infecta persistentemente algumas espécies de Leishmania, principalmente os da subespécie Viannia. Embora o impacto na saúde pública da leishmaniose nos trópicos seja significativa, muito pouco se sabe sobre os papéis possíveis, se houver, que LRV pode ter sobre a biologia da Leishmania, como por exemplo na infectividade e patogenicidade. Este estudo foi conduzido para avaliar a replicação de LRV1-4 durante o ciclo de crescimento de Leishmania (V.) guyanensis da cepa M4147, que é naturalmente infectada com LRV1-4 (M4147/LRV1-4). Cultivo de M4147/LRV1-4 em meio sólido permitiu o isolamento de um clone sem LRV1-4 (nomeado clone 40), que foi utilizado como controle negativo. Não houve diferença entre as curvas de crescimento de M4147/LRV1-4 e clone 40. A carga viral de LRV1-4 determinada por PCR em tempo real mostrou que não houve replicação viral durante a fase exponencial de crescimento do M4147/LRV1-4 no estado líquido, mas somente após a fase estacionária de Leishmania. Houve uma diminuição progressiva da carga de LRV 1-4 ao longo do tempo durante as passagens de M4147/LRV1-4 na cultura axênica. Comparação de infectividade de macrófagos peritoneais in vitro mostrou que o percentual de macrófagos infectados foi significativamente maior para M4147/LRV1-4 (51%) do que para o clone 40 (28%) em 96 horas após a infecção, mas não antes desse tempo. Não houve diferença significativa no número de amastigotas por macrófago infectado. O teste de patogenicidade em coxim plantar de hamster infectados com M4147/LRV1-4 e com o Clone 40 não apresentaram diferenças no tamanho da lesão. LRV 1-4 replica-se lentamente em Leishmania (V) guyanensis, sem efeito aparente sobre o ciclo de crescimento do parasita ou de sua patogenicidade na pata de hamster, porem sua presença aumentou a porcentagem de macrófagos peritoneais infectados in vitro, sem alteração da carga parasitária. While the public health impact of leishmaniasis in the tropics is significant, very little is known about possible roles, if any, that LRV may have on the biology of Leishmania, their infectivity and pathogenicity. These studies were conducted to measure replication of LRV1-4 during the growth cycle of Leishmania (V.) guyanensis of the strain M4147, which is naturally infected with LRV1-4 (M4147/LRV1-4). Growing of M4147/LRV1-4 on solid médium permitted isolation of one clone without LRV1-4 (named clone 40), which was used as a negative control. There was no difference between growth curves of M4147/LRV1-4 and clone 40. LRV1-4 load determined by real-time PCR did not increase significantly during the exponential phase of growth of M4147/LRV1-4 in liquid, but only after the stationary phase was reached. There was a Progressive decrease in the load of LRV 1-4 over time during passages of M4147/LRV1-4 in axenic culture. Comparison of peritoneal macrophage infectivity in vitro showed that the percentage of infected macrophages was significantly higher for M4147/LRV1-4 (51%) than for clone 40 (28%) at 96 hours post infection, but not before that time. No significant differences in numbers of amastigotes per infected macrophage were noted. The pathogenicity assay in hamster footpad infected with M4147/LRV1-4 or clone 40 showed no differences in size of lesion and parasite load. LRV1-4 seems to replicate slowly in Leishmania (V.) guyanensis, without apparent effect on the parasite growth cycle or its pathogenicity in the hamster footpad, but its presence increases the ffequency of mouse peritoneal macrophage in vitro infection by the parasite, without change in the parasite load.

O vírus sincicial respiratório humano (VSRH) é o agente viral mais freqüentemente relacionado a doenças do trato respiratório inferior em crianças abaixo de um ano de idade. Analíse da varibilidade antigênica e gênica mostraram que o VSRH pode ser divido em dois grupos: A e B. O vírus é um membro do gênero Pneumovirus pertencente a família Paramyxoviridea, e possui três principais proteínas que são: glicoproteina F (fusão), glicoproteina G (adesão), glicoproteina SH (pequena proteína hidrofóbica). A proteína F é responsável pela fusão da célula ao vírus, enquanto a proteína G tem papel fundamental na replicação do vírus, porém a função da proteína SH, ainda não está bem definida, estudos recentes mostram-na como responsável por inibir a sinalização do fator de necrose tumoral alfa (TNF-a). Neste estudo foram colhidas amostras de 965 crianças, entre os anos de 2004 e 2005, dentre as quais 424 foram positivas. 117 amostras foram seqüenciadas a proteína SH e G e comparadas com amostras que circularam mundialmente. A analíse filogenética mostrou uma baixa variabilidade entre os genótipos estudados tanto do grupo A quanto do B. The human respiratory syncytial virus (HRSV) is the major cause of lawer respiratory tract infections in infantis, young children and elderly. Analysis of the antigenic and genetic variability has shown that there are two groups of the virus HRSV, A and B. The virus (HRSV) is a member of the genus pneumovirus in the paramyxoviridae family. The virus encodes three membrane-bound glicoproteins, namely the fusion (F) attachment (G) and small hydrophobic (SH) proteins. The F mediates fusion of the virus and cell membranes and the G proteins is involved in virus attachment. The biological properties of the F and G glicoproteins and role that they play during virus replication relatively well understood, however the functional significance of the SH protein during replication remains unclear, although recent study shown that it can inhibit TNF-alpha. In this study, HRSV strains were isolated from nasopharyngeal aspirates collected from 965 children between 2004 and 2005, yielding 424 positive samples. We sequenced the small hydrophobic protein (SH) gene and protein (G) of 117 samples and compared them with other viruses identified worldwide. The phylogenetic analysis showed a low genetic variably among the isolates but allowed us to classify the viruses into different genotypes for the A and B HRSV strains.

O bocavirus humano (HBoV) é um parvovirus recentemente identificado em associação com a presença de sintomas de infecção do trato respiratório. Esse vírus possui um genoma de aproximadamente 5217 nucleotídeos que contém 3 open reading frames que codificam 4 proteínas (NS1, NP-1, VP-1 e VP-2). HBoV tem sido detectado em amostras respiratórias de diversas partes do mundo, incluindo Austrália, América do Norte, Europa, Ásia e África, o que sugere uma distribuição global desse vírus. Entretanto, nenhum estudo longitudinal de HBoV em amostras respiratórias foi realizado na América Latina. Dessa forma, nós realizamos um estudo prospectivo de HBoV em lavados nasofaríngeos (LFNs) coletados de pacientes com sintomas de infecção do trato respiratório (IRA) atendidos em um hospital universitário de Ribeirão Preto, SP e em um hospital universitário de Salvador, BA no período entre 2005 a 2007. 1288 LFNs de 1217 pacientes foram encaminhados ao laboratório de virologia e foram testados por PCR para HBoV. Desses pacientes, 962 eram menores de 5 anos e 177 eram maiores de 5 anos. Além disso, também foram analisados 50 LFNs de crianças menores de 5 anos que não tinham sintomas respiratórios. Todas as amostras positivas para HBoV foram testadas para todos os outros vírus respiratórios, incluindo o vírus sincicial respiratório (HRSV), rinovirus humano (HRV), influenza humano (HFLU), metapneumovirus humano (HMPV), parainfluenza humano (HPIV), coronavirus humano (HCoV) e adenovirus humano (HAdV). A carga viral de HBoV foi determinada por PCR em tempo real em todas as amostras positivas e o genoma completo de 19 amostras de HBoV foi seqüenciado. Com intuito, de fazer um levantamento sorológico e determinar sítios replicativos de HBoV, nós ainda clonamos e expressamos em S. cerevisae (Y258) o gene de VP2, que codifica uma das proteínas do capsídeo viral. A prevalência desse vírus foi de 4,8% em crianças menores de cinco anos e de 1% em pacientes maiores de cinco anos. HBoV não foi detectado em crianças sem sintomas. Dos 259 pacientes analisados em 2005, 25 (10%) foram positivos para HBoV. Esse vírus circulou mais frequentemente em abril, mês de maior incidência do HRSV. Em 2006, HBoV foi detectado em apenas 10 LFNs de 334 (3%) amostras testadas, sem qualquer pico de freqüência. Em 2007 HBoV foi detectado em 13 de 552 (2%) amostras, com uma freqüência de detecção um pouco maior em junho e julho. Os sintomas mais comumente observados foram rinorréia, tosse, febre e chiado, que foram observados geralmente em mais de 50% dos casos positivos para HBoV. Não houve uma diferença significativa na prevalência desses sintomas entre as crianças positivas e negativas para HBoV. Entretanto, foi observada uma maior freqüência de diarréia entre as crianças com esse vírus. Nesse estudo também foi documentado uma alta freqüência de co-infecções virais entre os pacientes com HBoV. Os vírus mais frequentemente associados com o bocavirus humano foram: HRSV, HRV e HAdV. Além disso, foi detectado uma maior carga viral media e uma maior freqüência de diarréia nos 15 pacientes com infecção exclusiva por HBoV do que nos pacientes com co-infecção. Esses resultados mostraram que HBoV pode alcançar títulos enormes (tão grandes como1014/mL) em LFNs de pacientes com sintomas respiratórios e que isso é associado a de diarréia. O seqüenciamento do genoma inteiro de HBoV realizado nesse estudo indica que a divergência genômica entre as amostras desse vírus é muito pequena. Como conclusão, nós demonstramos que HBoV circula e é detectado em associação com sintomas de infecção respiratória e diarréia no Brasil. Novos estudos, com um longo acompanhamento em diferentes populações serão necessários para determinar a sazonalidade e o real impacto clínico de HBoV em nosso país. Human bocavirus (HBoV) is a parvovirus recently identified in association with respiratory tract infections. HBoV 5217 nt genome contains 3 open reading frames encoding four proteins (NS1, NP-1, VP-1 and VP-2). HBoV has been reported in respiratory samples from children in several parts of the world (including Australia, North America, Europe, Asia, and Africa), suggesting that the virus circulates worldwide. However, no longitudinal studies of HBoV in respiratory samples have been reported in Latin America. We report a prospective study of HBoV in nasopharyngeal aspirates (NPAs) collected from patients seen for acute respiratory tract infections (ARI) at the University of Sao Paulo Hospital in Ribeirao Preto, southeast Brazil and at the University Hospital in Salvador, Brazil. 1288 NPAs from 1217 patients was submitted to the virology lab for respiratory virus detection from 2005 to 2007 and were screened for HBoV by polymerase chain reaction (PCR), whom 962 were under 5 years of age and 177 were older than 5 years. In addition, NPAs from 50 children under 12 years without IRA was also tested to HBoV for PCR. All samples positive of HBoV was tested for others respiratory virus, including the human respiratory syncitial virus (HRSV), human rhinovirus (HRV), human influenza (HFLU), human metapneumovirus (HMPV), human parainfluenza virus (HPIV), human coronavirus (HCoV) and human adenovirus (HAdV). These samples had their HBoV viral load determined by real time PCR and the viral entire genome of nineteen HBoV sample was sequenced. We also cloned and expressed in S. cerevisae (Y258) the gene of VP2, one protein of viral capside. The prevalence of this virus was of 4,8% in children under 5 years and 1% in adults, both with IRA. HBoV was not found on the patients without symptoms. In 2005, of the 259 patients tested, 25 (10%) were positive for HBoV. Interestingly, the virus circulated more frequently in April, the month of peak activity of respiratory HRSV. In 2006 HBoV was detected in only 10 NPAs out of 334 samples (3%) tested, without any notable peak of frequency. In 2007 HBoV was detected in 13 out of 552 (2%) tested samples with little higher frequency of detection in June an July. Rhinorrhea, cough, and wheezing were observed in more than 50% of the HBoV-positive children, and no obvious respiratory clinical differences were noted between HBoV-positive and negative children. However, was noted a higher frequency of diarrhea on HBoV-positive patients. In this study was also observed a larger frequency (71%) of viral coinfections between the HBoV-positive patients. The respiratory viruses more frequently associated with human bocavirus were: HRSV, HRV and HAdV. Interestingly, on the 15 HBoV-alone patients was observed a higher viral load and a higher prevalence of diarrhea than HBoV-coinfection patients. These results showed that this virus can reach enormous titles (like 1014) in NPAs from patients with respiratory infection symptoms and this is associated with diahhrea. The entire genome sequencing of HBoV of our study indicates that the genetic divergence between the HBoV lineages is small. In conclusion, we demonstrated that HBoV circulates and is detected in association with respiratory symptoms and diarrhea in Brazil. Long term surveillance will be needed to determine whether or not an HBoV season occurs and what is the real clinical impact of this virus in our country.

INTRODUÇÃO: As infecções respiratórias agudas de etiologia viral são um fator de morbimortalidade em todo o mundo. Devido a novas epidemias por vírus respiratórios e avanços no diagnóstico, em especial, com técnicas moleculares, novos agentes têm sido identificados nos últimos anos. Os vírus respiratórios (VR) são responsáveis por cerca de 5% das infecções nosocomiais. Em adultos internados em Unidade de Terapia Intensiva a presença de infecção por vírus respiratórios é reportada em poucas publicações. Os objetivos deste estudo foram avaliar a prevalência de infecções por vírus respiratórios em pacientes internados em Unidade de Terapia Intensiva, a freqüência de infecções virais de origem nosocomial, os fatores de risco associados e seu impacto na morbimortalidade destes pacientes. MÉTODOS: No período de maio de 2003 a junho de 2004, amostras de lavado de nasofaringe foram coletadas de pacientes internados em unidade de terapia intensiva do Hospital das Clínicas da Faculdade de Medicina da Universidade de São Paulo, independentemente de sintomas respiratórios. O diagnóstico de vírus respiratórios (Vírus Respiratório Sincicial, vírus da Influenza A e B, parainfluenza, adenovírus, rinovírus, coronavírus e metapneumovírus) foi feito pelas técnicas de imunofluorescência direta e reação em cadeia pela polimerase. Dados clínico-epidemiológicos foram prospectivamente coletados em ficha específica. RESULTADOS: Foram avaliados pacientes em 228 internações, sendo o diagnóstico de 57 infecções por vírus respiratórios estabelecido em cinqüenta e duas admissões (23%). Rinovírus foi diagnosticado em 22 casos, coronavírus em 15, adenovirus em sete, influenza A em seis, vírus sincicial respiratório em cinco, Parainfluenza em um e metapneumovirus em um caso. Trinta e quatro episódios (59,6%) de infecções virais foram considerados de origem nosocomial. Foram investigados os fatores de risco associados à ocorrência de infecções por VR, à necessidade de ventilação mecânica e ao óbito na UTI. Na análise univariada a ocorrência de infecção por vírus respiratório associou-se à presença de quadro respiratório à admissão na UTI, à presença de co-morbidades (hipertensão arterial sistêmica e AIDS), à gravidade do quadro clínico admissional definido pelo valor de APACHE e ao uso de ventilação mecânica invasiva. Na análise multivariada, através de regressão logística, as variáveis que permaneceram significantemente associadas a ocorrência de infecção por VR foram a idade (OR 0,96), a hipertensão arterial (OR 3,95), a presença de quadros respiratórios à admissão na UTI (OR 2,22) e o valor de APACHE (OR 1,06). Os fatores de risco associados à necessidade de ventilação mecânica invasiva foram as infecções por vírus respiratórios (OR 1,98), o tempo de internação na UTI (OR 1,16), e valor do APACHE (OR 1,07). Os fatores de risco para óbito nesta série foram doença cardíaca ou neoplasia, infecções fúngicas, uso de ventilação mecânica, e o valor de APACHE na admissão. Não houve associação entre o diagnóstico de infecção por vírus respiratórios e a ocorrência de óbito (p=0,118). CONCLUSÃO: As infecções virais respiratórias foram freqüentes em pacientes adultos internados em Unidade de Terapia Intensiva, sendo a maioria de origem nosocomial. Os pacientes com diagnóstico de infecção por vírus respiratórios tiveram maior necessidade de ventilação mecânica nesta série. O diagnóstico das viroses respiratórias deveria ser insistentemente buscado em pacientes com quadros respiratórios à admissão para que medidas de controle da transmissão nosocomial fossem implementadas. A associação com hipertensão arterial mereceria ser objeto de novos estudos. INTRODUCTION: Acute respiratory infections of viral etiology are a factor of morbidity and mortality in the all world. Advances in the diagnosis, in special, with molecular techniques, new agents have been identified in the last years. Respiratory viruses (RV) are responsible for about 5% of the nosocomial infections. Few studies have addressed the incidence, morbidity and mortality of viral respiratory infection in adults admitted in Intensive Care Units (ICU). We evaluated the prevalence of viral respiratory infections in ICU adult patients, the ratio of these infections that were of nosocomial origin, the risk factors and the impact of viral respiratory infections in the morbidity and mortality of these patients. METHODS: From May 2003 to June 2004, nasopharyngeal aspirates were taken twice a week, irrespective of respiratory symptoms, from 228 patients admitted at the ICU of the Hospital of Clinics, Faculty of Medical Sciences, University of São Paulo. Respiratory viruses (Respiratory Syncytial Virus, influenza virus, parainfluenza virus, adenovirus, rhinovirus, coronaviruses and metapneumovirus) were diagnosed by direct immunofluorescent assay (DFA) or polymerase chain reaction. (PCR). Medical and epidemiological data were prospectively collected. RESULTS: Fifty seven RV was diagnosed in 52 of the 228 ICU admissions (23%). Rhinovirus was the RV more frequently diagnosed (22 cases), followed by Coronaviruses (15 cases), Adenoviruses (7 cases), Influenza A viruses (6 cases), Respiratory Syncytial Virus (5 cases), Parainfluenza virus (one case) and Metapneumovirus (one case). Thirty and four episodes (59.6%) were considered of nosocomial origin. We evaluated the risk factors associated with the occurrence of RV infections, the need of invasive mechanical ventilation and death at the ICU. Univariate analysis showed that RV infections were associated with respiratory tract involvement at admission, some comorbidities (arterial hypertension and AIDS), to APACHE score at admission, and to the need of invasive mechanical ventilation. In multivariate analysis, age (OR 0.96), arterial hypertension (OR 3.95), respiratory tract involvement at admission (OR 2.22) and APACHE value (OR 1.06) were significantly associated with the occurrence of RV infection. Risk factors associated with the need of invasive mechanical ventilation were RV infections (OR 1.98), longer time at the ICU (OR 1.16) and APACHE value (OR 1.07). Death at the ICU was significantly associated with heart disease or neoplasia, fungal infection, mechanical ventilation and APACHE value. RV infection was not associated with ICU death (p=0.118). CONCLUSIONS: Respiratory Virus infections were frequent in adult ICU patients in the present series; the majority of them were of nosocomial origin. Patients with RV infections were more likely to need mechanical ventilation during ICU admission. Diagnosis of RV infections should be included in the diagnostic assessment of ICU patients, especially those with respiratory tract involvement at admission. This policy will certainly favor the implementation of preventive measures to control nosocomial transmission. The association with arterial hypertension deserves further studies.

Reis, AD Incidence of respiratory illness by human respiratory syncytial virus in a cohort of newborn in Sao Paulo city: comparison of techniques and genetic diversity. In this study we analyzed a cohort of newborn children from 3 public hospitals from the city of Sao Paulo, aiming to evaluate the incidence and the genetic diversity of human respiratory sincicial virus. 270 healthy newborn children fulfilling inclusion criteria were enrolled from December 2002 to April 2005. Average follow up time was 113.5 month. 173 out of 270 newborns had at least one episode of respiratory illness; 92 presenting a single episode of disease and 81 presenting 2 or more episodes. 316 samples were obtained, and HRSV was detected in 36 samples (11.3%), with an incidence rate of 9.84/1000 children/month. Polimerase Chain Reaction (PCR) was the most sensitive technique, detecting 35 samples (11.1%), followed by immunofluorescence (25 samples, 7.9%) and viral isolation (20, 6.7%) (p
