1. Catalysing virtual collaboration : The experience of the remote TIES working groups

Abstract

During the COVID-19 pandemic, the idea of collaboration and scientific exchange between members of the scientific community was enhanced by technology. Virtual meetings and work platforms have become common resources to continue generating research, partially replacing instances of joint in-person work before, during or after a conference. The idea of teleworking played a fundamental role in remote collaboration groups within The International Statistical Society (TIES), a community of interdisciplinary scientists such as statisticians, mathematicians, meteorologists, and biologists, among others working on quantitative methods to enhance solutions to environmental problems. In 2021 the Society launched three working groups with the aim of improving networking across the Society's members and develop creative collaboration, while advancing statistical and computational methods motivated by real-world driven applications in environmental research. Here, we provide insights from this virtual collaborative initiative.

2. Time Series Clustering using the Total Variation Distance with Applications in Oceanography

Abstract

An algorithm for determining stationary periods for time series of random sea waves is proposed in this work. This is a problem in which changes between stationary sea states are usually slow and segmentation procedures based on change-point detection frequently give poor results. The method is based on a new procedure for time series clustering, built on the use of the total variation distance between normalized spectra as a measure of dissimilarity. The oscillatory behavior of the series is thus considered the central characteristic for classification purposes. The proposed algorithm is compared to several other methods which are also based on features extracted from the original series and the results show that its performance is comparable to the best methods available and in some tests it performs better. This clustering method may be of independent interest.

Abstract

This document illustrates the technical layout and the expected performance of the Micro Vertex Detector (MVD) of the PANDA experiment. The MVD will detect charged particles as close as possible to the interaction zone. Design criteria and the optimisation process as well as the technical solutions chosen are discussed and the results of this process are subjected to extensive Monte Carlo physics studies. Abstract

This document describes the technical layout and the expected performance of the Straw Tube Tracker (STT), the main tracking detector of the PANDA target spectrometer. The STT encloses a Micro-Vertex-Detector (MVD) for the inner tracking and is followed in beam direction by a set of GEM-stations. The tasks of the STT are the measurement of the particle momentum from the reconstructed trajectory and the measurement of the specific energy-loss for a particle identification. Dedicated simulations with full analysis studies of certain proton-antiproton reactions, identified as being benchmark tests for the whole PANDA scientific program, have been performed to test the STT layout and performance. 5. The impact of herbicide tolerant oilseed rape in selected agro-ecosystems

Abstract

A range of field studies of cross pollination between herbicide tolerant and conventional oilseed rape crops and plots were conducted to demonstrate the effects of variety, distance, pollen source and sink size and intervening crop on levels of outcrossing. Experiments investigating the influence of variety on outcrossing showed that hybrid oilseed rape varieties containing high proportions of male sterile plants (varietal associations) were pollinated at higher frequencies than standard fully fertile varieties. Studies conducted using various sizes of genetically modified herbicide tolerant (GMHT) pollen sources showed that small GMHT feral populations cross pollinated with crops in close proximity and the levels of contamination obtained depended on the genotype of the conventional crop. Long range cross pollination of male sterile oilseed rape receptor plots showed that pollination events were measurable at up to 600m from the large GMHT pollen source. Results from a study of cross pollination between mixed populations of GMHT oilseed rape plants and conventional varieties also demonstrated that a hybrid rape type (varietal association) was cross pollinated at considerably higher levels than an open pollinated and a fully restored hybrid variety. Evidence gathered in the experiment suggested that, over a wide range of initial GMHT contamination rates, the final proportion of GMHT seed in the total population was a constant fraction of the initial contamination rate. Outcrossing data was used to compare negative exponential and inverse power law models for their fit to describe the observed relationship between cross pollination and distance from source. Results showed that the inverse power law provided a better fit of the data. This demonstrated that dispersal described by the inverse power law was more likely to lead to cross pollination at both near and large distances from the pollen source compared to the negative exponential model. The consequences of the likely ecological behaviour of GMHT traits resulting from the dispersal curves for regulation and risk assessment are discussed. The effect of the herbicides used in herbicide tolerant and conventional oilseed rape on weed populations were compared in a single season. Results suggested that the herbicides have different activity spectra thus resulting in a variety of surviving weed species in HT treatments. The change in active ingredient and the timing of herbicide application in HT winter oilseed rape crops will likely cause a change in the weed species that are being controlled or those that escape treatment. Levels of weed biomass recorded prior to harvest of the oilseed rape crop showed that there may be differences between treatments in terms of the quantity of seed returned to the seedbank from the range of weed species present. Limited data on the behaviour of herbicide tolerant volunteers showed that single and putative double-tolerant plants were as susceptible as conventional oilseed rape volunteers to normal selective herbicides used in cereal crops. Data from a number of elements of the studies on GM contamination rates, weed control, and seed bank estimates were used to develop a simple population projection model. The model used a Markov process to examine the fate of volunteer and feral populations of oilseed rape comprising a mixture of conventional and herbicide tolerant types. Results from the model indicated that the prevalence of the GMHT trait in the weed or feral population was more sensitive to the efficacy of control practices used in the rotation than the levels of cross-pollination and competition between the herbicide tolerant and conventional varieties in the mixed population. Thus, although the cross-pollination studies suggested that varietal associations are more likely to be cross-pollinated by GM pollen than fully fertile varieties, the projection model suggested that resulting differences in the prevalence of the GM trait in volunteer and feral populations may not be very large. Abstract

The coronavirus disease of 2019 (COVID-19) pandemic launched an unprecedented global effort to rapidly develop vaccines to stem the spread of the novel severe acute respiratory syndrome coronavirus (SARS-CoV-2). Messenger ribonucleic acid (mRNA) vaccines were developed quickly by companies that were actively developing mRNA therapeutics and vaccines for other indications, leading to two mRNA vaccines being not only the first SARS-CoV-2 vaccines to be approved for emergency use but also the first mRNA drugs to gain emergency use authorization and to eventually gain full approval. This was possible partly because mRNA sequences can be altered to encode nearly any protein without significantly altering its chemical properties, allowing the drug substance to be a modular component of the drug product. Lipid nanoparticle (LNP) technology required to protect the ribonucleic acid (RNA) and mediate delivery into the cytoplasm of cells is likewise modular, as are technologies and infrastructure required to encapsulate the RNA into the LNP. This enabled the rapid adaptation of the technology to a new target. Upon the coattails of the clinical success of mRNA vaccines, this modularity will pave the way for future RNA medicines for cancer, gene therapy, and RNA engineered cell therapies. In this review, trends in the publication records and clinical trial registrations are tallied to show the sharp intensification in preclinical and clinical research for RNA medicines. Demand for the manufacturing of both the RNA drug substance (DS) and the LNP drug product (DP) has already been strained, causing shortages of the vaccine, and the rise in development and translation of other mRNA drugs in the coming years will exacerbate this strain. To estimate demand for DP manufacturing, the dosing requirements for the preclinical and clinical studies of the two approved mRNA vaccines were examined. To understand the current state of mRNA-LNP production, current methods and technologies are reviewed, as are current and announced global capacities for commercial manufacturing. Finally, a vision is rationalized for how emerging technologies such as self-amplifying mRNA, microfluidic production, and trends toward integrated and distributed manufacturing will shape the future of RNA manufacturing and unlock the potential for an RNA medicine revolution.

12. ‘To Be Shut Up’: New Evidence for the Development of Quarantine Regulations in Early-Tudor England

Abstract

Summary On 13 January 1518, a series of ordinances to regulate plague outbreaks were proclaimed in London. These have previously been thought to have been the first set of quarantine measures issued in England; part of an ambitious new social policy led by the King’s chief minister, Cardinal Thomas Wolsey, and intended to bring England in line with the leading Renaissance states in Europe. The discovery of two previously unknown documents from the archives of St George’s College, Windsor—which predate the London ordinances—requires a reconsideration of this narrative. The article argues that demands for plague quarantine regulations were driven primarily by Henry VIII’s personal and pronounced fear of infection and considers the factors that may have led to such a fear. In doing so, it introduces new evidence for the death of Henry’s grandmother, Elizabeth Woodville, and explores the ways in which medical ideas travelled across Europe.

Abstract

The regioselectivity of the Friedel-Crafts-type cyclisation of a range of γ-aryl-β-benzoylamido acids, bearing oxy substituents at the C(3)- and C(4)-positions of the γ-aryl ring, has been investigated. In all of the cases examined (with 3,4-dimethoxy, 3,4-methylenedioxy and 3-hydroxy-4-methoxy substituents) the Lewis acid promoted cyclisation proceeds with exclusive regioselectivity for attack at the C(6)-position rather than at the C(2)-position, and furnishes the corresponding N - and O -protected 3-amino-6,7-dihydroxy-1-tetralone derivatives. This inherent regioselectivity can be overturned by the regioselective introduction of chlorine as a blocking group for the C(6)-position; subsequent Lewis acid promoted cyclisation then proceeds with exclusive regioselectivity for attack at the C(2)-position to deliver the corresponding N - and O -protected 3-amino-5-chloro-7,8-dihydroxy-1-tetralone derivative. These complementary cyclisation protocols represent useful methods for the preparation of these benzo-fused carbocyclic ring systems, which are the functionalised AB-rings of a range of morphinan alkaloids.

Abstract

Lipid nanoparticles (LNPs) are established in the biopharmaceutical industry for efficient encapsulation and cytosolic delivery of nucleic acids for potential therapeutics, with several formulations in clinical trials. The advantages of LNPs can also be applied in basic research and discovery with a microfluidic method of preparation now commercially available that allows preparations to be scaled down to quantities appropriate for cell culture. These preparations conserve expensive nucleic acids while maintaining the particle characteristics that have made LNPs successful in later stages of genetic medicine development. Additionally, this method and the resulting LNPs are seamlessly scalable to quantities appropriate for in vivo models and development of nucleic acid therapeutics.The present work describes the methodology for preparing LNPs loaded with siRNA, mRNA or plasmids using a commercially available microfluidic instrument and an accompanying transfection kit. Guidelines for application to cultured cells in a well-plate format are also provided.

Abstract

Conjugate addition of enantiopure lithium ( R )- N -benzyl- N -(α-methylbenzyl)amide to a range of ω-aryl-α,β-unsaturated esters gives the corresponding ω-aryl-β-amino esters as single diastereoisomers in high yields. Friedel–Crafts cyclisation of the pendant carbonyl group onto the ω-aryl ring then gives a range of 3-amino-substituted 1-indanones, 1-tetralones, and 1-benzosuberones, representing an efficient and short protocol for the preparation of these benzo-fused carbocycles in enantiopure form.

37. Anaesthetic management of a patient with eosinophilic granulomatosis with polyangiitis for internal fixation of a fractured femur.

Abstract

Eosinophilic granulomatosis with polyangiitis (EGPA) is a rare disease with an estimated annual incidence of 0.5–6.8 per million. It is characterised by necrotising vasculitis with multiorgan eosinophilic infiltration. Pulmonary manifestations are the most common presentation of EGPA, and cardiac complications are the most common cause of death. Anaesthetic management of EGPA is challenging due to perioperative pulmonary complications, multiorgan involvement and greater risk of cholinesterase enzyme deficiency. We are reporting the anaesthetic management of a 58-year- old woman, diagnosed with EGPA 3 years ago, who underwent urgent intramedullary nail insertion for a femur fracture. The anaesthetic technique comprised femoral nerve block and spinal anaesthesia, thereby avoiding (1) the need for upper airway manipulation, (2) potential adverse effects of anticholinesterase drugs (for reversal of neuromuscular blockade) and (3) histamine release associated with morphine administration perioperatively. Abstract

The Cascade arc is the warm-slab subduction zone global end member, where a broad variety of primitive magmas with highly variable slab fluid signatures have erupted in close spatial and temporal proximity. A number of petrogenetic models have been proposed to explain the occurrence of such diverse magmas, but the source(s) of these magmas and the timing of fluid addition to the sub-arc mantle remain controversial. We present uranium-series isotope data ( 238 U– 230 Th– 226 Ra) for eighteen mafic lavas from the Three Sisters region of the central Oregon Cascades, and for a further six lavas from the rear-arc Newberry Volcano. The majority of these samples have geochemical characteristics (e.g. Nb/Zr, Ba/Zr, 87 Sr/ 86 Sr) consistent with previously described calc-alkaline basalts from this region of the arc, and indicative of limited fluid involvement at some stage in their genesis. Trace element and long-lived radiogenic isotope modeling suggests that this fluid was derived from dehydration of subducting sediments, and was added to an enriched, garnet-bearing mantle wedge source. The trace element systematics of the lavas are consistent with small degree ( 230 Th/ 238 U) and ( 226 Ra/ 230 Th) ≥ 1, similar to values measured in fresh MORB and other parts of the arc. Results of a dynamic melting model support the interpretation that these lavas are small degree melts of an asthenospheric source, and do not allow for a lithospheric mantle source. However, the U-series data do not permit us to determine whether fluid addition was the trigger for melting, or whether the lavas were generated from a secular equilibrium source that had experienced fluid addition > 350 ka prior to melting. Regardless, modern fluid input is limited and melting is dominantly occurring in response to upwelling and decompression.

Abstract

The Corixidae of New Zealand braided river beds were previously assigned to two species Sigara potamius and S. limnochares on the relative size of the strigil, a comb-like structure attached to the sixth abdominal tergite of males. Further collecting has rejected this taxonomic distinction. Phylogenetic analysis of mitochondrial DNA from a range of populations has revealed four distinct clades, none of which relates to currently accepted species boundaries, and it has not been possible to identify morphological traits correlating to these four clades. Consequently, only the species S. potamius is retained and applied to all populations, and S. limnochares becomes its junior synonym. Analysis of genetic variation indicates that variation within this species originated during the Pliocene.

Abstract

Historically displays used three colorants in an additive system. The CIE chromaticity diagram adequately illustrated color capability. Modern displays are not constrained by this additive architecture and the diagram can fail in its purpose. The “gamut rings” diagram provides a solution to illustrate color capability that directly correlates to color gamut volume.

Abstract

African buffalo were introduced into a wildlife conservancy in the southeast of Zimbabwe in an effortto increase the conservancy's economic viability, which is primarily based on eco-tourism. The buffalo were infected with SAT serotypes (SAT-1, SAT-2 and SAT-3) of foot and mouth disease (FMD) virus, and in order to isolate the conservancy and prevent the transmission of FMD to adjacent populations of domestic livestock, the conservancy was surrounded by a double-fence system, 1.8 m in height. The intention was to prevent the movement of both wildlife and domestic animals across the perimeter. However, two years after the buffalo were introduced, FMD occurred in cattle farmed just outside of the conservancy. Using serological and molecular diagnostic tests, epidemiological investigations showed that it was most likely that antelope (impala or kudu), infected through contact with the buffalo herd within the conservancy, had jumped over the fence and transmitted the virus to the cattle.
