Background and Aim: In females of various species and experimental animals, iron (Fe) status in follicular fluid (FF) is associated with local physiological reproductive events related to follicle development, steroidogenesis, and oocyte maturation. However, these mechanisms remain unknown. This study aimed to determine and compare the intrafollicular and plasma concentrations of Fe, ferritin (Ferr), and transferrin (TRF) in cycling mares. Materials and Methods: Sixty ovaries were collected during the breeding season from 30 clinically normal mares raised for slaughterhouse meat production. Blood samples were collected before slaughter. Follicles were classified into three categories according to size: Small (20–30 mm; n = 20), medium (≥31–40 mm; n = 20), and large (≥41 mm; n = 20). The FF samples, after collection, were immediately taken to the laboratory for processing and were centrifuged, and the Fe and Ferr concentrations in the supernatant and plasma were determined by spectrophotometry. Results: Although intrafollicular Fe and Ferr were similar to plasma, TRF was significantly higher in FF than in systemic circulation (p < 0.05). Follicular development does not modify the status of Fe in the mare. Conclusion: Based on this evidence, it is possible that the acquisition of this molecule possibly originated from a local de novo source, whereas their diffusion through ultrafiltration does not play a relevant role. These results provide new scientific insights into the status of follicle Fe, suggesting its involvement in normal ovarian functions in mares.

Lactation in dairy cows implies comprehensive endocrine and metabolic changes including a systemic electrolytic reaction. Previous studies have rarely considered these specific demands due to the influence of lactation periods. Therefore, this study aimed to assess the effects of early, middle, and late lactation phases on the dynamic changes in serum concentrations of progesterone (P4), 17β-oestradiol (E2), cortisol, and some electrolytes (Ca++, Mg++, Na+, K+, Cl−, Pi) and biochemical parameters (alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), creatine kinase (CK), total bilirubin, urea, and iron (Fe++) in 10 Holstein, 10 Brown Swiss, and 10 Modicana multiparous healthy dairy cows (4.2 ± 1.7 years of age) sampled at 60-day intervals throughout lactation. Lactation induced significant changes in the concentrations of P4, which peaked at >120–180 days, decreased at >240–300 days, and increased again after 300 days. Cortisol showed an opposite trend to P4, with concentrations progressively decreasing, except for the phase between >240 and 300 days, and a steep drop at >300 days compared to previous phases. Na+ concentrations showed the lowest values at 0–60 d and the highest ones at >180–240 days, whereas Mg++ showed the highest values at >60–120 d and the lowest at >300 d. Significant correlations were found between P4 with cortisol, Cl− and K+, and cortisol with Ca++ and LDH. Significant differences in average concentrations of AST, ALT, LDH, Ca++, Mg++, and Fe++ were observed among different dairy cow breeds. Understanding the dynamic changes in hormone levels, electrolytes, and biochemical parameters during different lactation phases, while considering breed differences in dairy cows, is crucial for improving herd health management and milk production in commercial dairy farms.

Cattle productivity, whether in terms of meat yield or milk production, is intricately regulated by a multitude of factors. Among them, hormone concentrations play a significant role, reflecting the complex interplay between endocrine regulation and physiological processes that ultimately determine the efficiency and yield of production. High concentrations of 5-hydroxytriptamine (5-HT) are associated with a reduced metabolic load at the onset of lactation and a lower milk yield. Thyroid hormones (THs) and cortisol also affect several metabolic pathways, including carbohydrate, protein, and lipid metabolism. The aim of this study was to assess if milk or meat aptitudes and age influence circulating 5-HT, THs, and cortisol concentrations, investigating the possible interactions among these parameters. The research was performed on 46 healthy cows of three different breeds. Enzyme-linked immunosorbent assay (ELISA) methods were used to quantify circulating 5-HT and cortisol concentrations, and an immunochemiluminescent analyzer was used for THs. For parameters exhibiting non-normal distributions, an ANCOVA model using age, aptitude, and their interaction as fixed factors was applied. Significant lower T3 concentrations were recorded in dairy cows than in meat cows. Moreover, T4 significantly decreased with advancing age both in cows aimed at milk and meat production. Lastly, T4 was positively correlated with T3 and 5-HT in meat production-oriented cows.

Horse transport is considered a cause of stress in animals and is known to affect the 5-HT concentrations in both the brain and other tissues. The aim of this research was to investigate the effect of horse transportation and slaughter stress on plasma serotonin’s concentration and the expression levels of the related 5-HT1B and 5-HT2A receptors in PBMCs. Furthermore, the IL-12 levels and a variety of blood parameters, including triglycerides, total cholesterol, glucose, aspartate aminotransferase, creatine phosphokinase, lactate dehydrogenase, were also considered. This research was carried out on 32 horses submitted to short road transport of 40 km to slaughter. Blood samples were collected in baseline conditions (T0) and 24 h later, after they were slaughtered (T1). The results showed a significantly increased expression of 5-HT1B and 5-HT2A receptors and a significantly decreased expression of IL-12 in PBMCs at T1 vs. T0. Furthermore, a significant increase in cortisol and glucose concentrations, and LDH activity was observed at T1. In contrast, a significantly lower circulating 5-HT concentration was observed at T1 vs. T0. These results indicate that the stress induced by transport and slaughter stimuli led to the serotoninergic system’s activation, suggesting that the expression of serotonin receptors could be used as a pivotal marker of stress, with potential applications for the improvement of elective protocols to observe the guidelines relating to transported horses.

Background and Aim: Toxoplasma gondii is a global zoonotic parasite infecting virtually all warm-blooded species, although a species-specific variability is evident referring to symptoms frame. Both the success of T. gondii and the outcome of infection depend on a delicate balance between host cellular pathways and the evasion or modulation strategies elicited by the parasite. The hormonal and molecular mechanisms involved in this delicate host-parasite balance are still unclear, especially when considering intermediate host species other than mouse. This study aimed to assess any correlation between T. gondii infection and selected molecular and hormonal factors involved in responses to infection in susceptible species such as swine. Moreover, blood counts and hematochemical assays (glucose, total cholesterol, and triglycerides dosage) were performed to evaluate the overall health condition of animals. Materials and Methods: Toxoplasmosis was diagnosed by enzyme-linked immunosorbent assay for antibodies determination and real-time polymerase chain reaction (RT-PCR) for T. gondii DNA detection. Target genes coding for key factors of cell responses to T. gondii infection were selected, and their transcription was assessed in various tissues by quantitative RT-PCR. 17-β estradiol concentrations were assessed by fluorimetric enzyme-linked immunoassay and the AIA- 360 automated immunoassay analyzer. Blood count and hematochemical analyses were performed by a blood cell counter and a spectrophotometer, respectively. Results: The present research highlighted significant differences among infected and uninfected swine (control group) for both transcription profiles of some of the molecular factors considered and 17-β estradiol concentrations. Referring to the assessed hematological and biochemical parameters, no statistically significant differences were observed in infected swine compared to the control group. Conclusion: Our results contribute to the enrichment of data available about the subject and could be useful for a deeper knowledge of the interaction between this parasite and its hosts. However, more aspects are still unclear, such as the effective response of downstream molecules from the same pathways to the variation of factors observed in this study either assessing how the same factors respond to Toxoplasma gondii infection in other host speciesand further analyses should be performed on other host species.

Recent acquisitions on human-animal interaction and on its use in complementary medicine are reported. The contribution of horses to one health concept and the effects of equine-assisted activity and therapy (EAAT) are evaluated, also in the light of need to monitor stress conditions in therapeutic horses. The role of hypothalamus-pituitary-adrenal axis’ hormones and of total and free iodothyronines in stress coping of horses during therapeutic riding is critically discussed

IntroductionIntegrating by-products into livestock diet represents a great opportunity for implementing the concept of circular economy while reducing feed costs. Olive cake (OC) is considered an agro-industrial waste, but the high content of valuable metabolites makes it a promising feed integration. Therefore, this study investigated the effect of OC integration in beef cattle diet on different blood parameters.MethodsForty-eight young growing fattening Limousines-−24 bulls (body weight 350 ± 15 kg) and 24 heifers (280 ± 10 kg)—, aged 240 ± 20 days, were randomly allocated to 1 of 3 dietary treatments: concentrate at 0% (Control group: CTR), 10% (Low-olive cake group: L-OC), or 15% (High-olive cake group: H-OC) of OC inclusion. Blood samples and body weights were collected before administrating the supplemented diet (0 d), at the end of the stocker growing phase (56 d), and at the end of the fattening (147 d). After being slaughtered, animal carcasses were weighted. A linear regression model was fitted for each blood parameter with the 0 d as covariate and diet, time, sex, diet × time, and diet × sex as fixed effects.ResultsIn males, body weight was highest in CTR, but carcass weight was similar in all the groups. All the blood parameters were within physiological ranges, independently from the animal diet. CTR group showed the highest alanine aminotransferase (ALT, P = 0.0027) and creatine kinase (P = 0.0119), whereas total bilirubin (P = 0.0023) was higher in H-OC than in CTR. Moreover, ALT was highest in CTR at 56 d, becoming similar in all the groups at 147 d (P = 0.0280). Instead, the increase observed in total cholesterol from 56 to 147 d was lower in H-OC compared with CTR and L-OC (P = 0.0451). A significant effect of diet × sex interaction was observed on triglycerides, urea, liver enzymes, and insulin. These data support the OC inclusion of up to 15% of the concentrate with no detrimental effect on beef cattle metabolic status.DiscussionIn conclusion, OC can be considered as a component in beef diet giving an opportunity to improve agriculture sustainability.

Agro-industrial by-products incorporated into livestock feed formulations can positively impact feed costs and promote a circular bio-economy. Italy produces significant amounts of olive cake (OC), a by-product of olive oil extraction, with the potential for incorporation into bovine diets. However, information on its effects on endocrine responses in growing beef calves is lacking. Forty-eight Limousines randomly allocated to dietary treatment (control or 10%-OC or 15%-OC inclusion) were segregated according to sex and body weight. Serum concentrations of TSH, thyroid hormones, and cortisol were measured on day 0, day 56, and at the end of the trial on day 147. Circulating TSH, total (T3, T4) and free (fT3, fT4) iodothyronines, and cortisol concentrations were all within the normal physiological ranges, with no significant effect imparted by diet. However, the diet × time interaction was significant for T3. The cortisol, T3, T4, and fT4 registered on day 147 were higher than those of day 56, and cortisol was higher in heifers than bulls. Final body weight was positively correlated with TSH and T3 and negatively with cortisol concentration. These findings suggest that the inclusion of OC at levels up to 15% in growing/finishing beef diets had no adverse effects on the calves’ thyroid and cortical status.

Thyroid hormones physiologically regulate the semen quality, by altering testosterone concentrations, and different seminal parameters, as well as sperm motility, viability and semen volume. Serum thyroxine (T4), free thyroxine (fT4), thyroid-stimulating hormone (TSH) and testosterone (T) concentrations were measured in 26 adult Labrador Retriever dogs (n. 20 normospermic and n. 6 azoospermic groups), aged 3 ± 0.5 years old, to determine their specific references, by taking into account the related conventional and kinematic sperm parameters and clinical ultrasound evaluations. The mean values of T4, fT4, TSH and T concentrations, as soon as those of sperm parameters and clinical evaluations of normospermic group were in line with dog’s physiological range of literature data. Normospermic group showed significant correlations between T4 and T (r = −0.681; p < .01), sperm progressive motility (%) (r = −0.623; p < .01), and sperm non-progressive motility (%) (r = 0.625; p < .02). The azoospermic group showed higher T4, fT4, TSH and lower T concentrations, compared to normospermic group, and a positive correlation between T4 and T (r = 0.8548; p < .046). The use of breed-specific hormonal ranges and sperm parameters will improve the knowledge of their interaction in Labrador Retriever dogs, adding a new segment of scientific literature.Highlights Thyroxine (T4), free thyroxine (fT4), thyroid stimulating hormone (TSH) and testosterone (T) concentrations were measured in male dogs. The aim of the present study was to assess and compare the effect of increasing exercise levels of thirty-six trained Dutch Warmblood horses fed a isoenergetic high versus low fiber content on the circulating Thyroid hormones (THs): triiodothyronine (T3), thyroxine (T4), free triiodothyronine (fT3) and free thyroxine (fT4) concentrations. The training schedule was carried out in three consecutive phases, according to three exercise levels (light, medium and medium-heavy) performed on trainingsmill. Each phase lasted 4 weeks. At week 4 of each phase a standardized exercise test (SET) on treadmill was performed. Blood samples were collected at rest (baseline values), and after 15, 30 and 60 min after SET, according to different phases. Horses were randomly distinguished in two groups of 18 subjects. One group of horses was fed a high fiber diet (HF group) and one group was fed a low fiber diet (LF group). Results showed that in basal conditions diet influenced significantly fT3 values, with the highest concentration in HF, and fT4 values with the highest concentrations in LF group. The time influenced significantly THs in both groups, with the highest values at June for T3, and at October for T4, fT3 and fT4. A significant correlation between T4 and fT4 was observed in basal conditions. In exercise conditions, diet influenced only fT4 values, with thehighest concentrations in LF group. The time influenced significantlyT4 andfT4 concentrations, with the highest values at 15 min for fT4 and at 30 min for T4 values and the lowest values at 60 min for both hormones. Significant correlations between T3 and fT3, as well as T4 and fT4 were observed in exercise conditions. The evaluation of THs output, according to diet contents, could mirror a quantitative and qualitative interplay between thyroid function and increasing exercise level of trained horses.

The review discusses the hormonal changes during exercise stress. The exercise generally produces a rise of adrenaline (A), noradrenaline (NA), adrenocorticotropic hormone (ACTH), cortisol, glucagon, growth hormone, arginine vasopressine, etc., and a drop of insulin. The hormonal events during reestablishment of homeostasis due to exercise stress can be divided into a catabolic phase, with decreased tolerance of effort, and reversible biochemical, hormonal and immunological changes, and an anabolic phase, with a higher adaptive capacity, and enhanced performance. The two main hormonal axes activated in the catabolic phase are sympathetic–adrenal–medullary system and hypothalamic-pituitary-adrenal (HPA) axis, while in the anabolic phase, growth hormone-insulin-like factor I axis, and gonadal axes. The hormonal responses during exercise and recovery can be regarded as regulatory and integrated endocrine responses. The increase of catecholamines and ACTH is dependent on the intensity of exercise; a marked increase in plasma A occurs during exercises with high emotional content. The response of cortisol is correlated with the duration of exercise, while the effect of exercise duration on β-endorphin changes is highly dependent on the type of exercise performed. Cortisol and β-endorphin changes usually occur in phase, but not during exercises with high emotional content. Glucocorticoids and iodothyronines are involved in meeting immediate energy demands, and a model of functional interactions between HPA axis and hypothalamic-pituitary-thyroid axis during exercise stress is proposed. A modulation of coping responses to different energy demanding physical activities required for sport activities could be hypothesized. This review supports the proposed regulation of hypophysiotropic TRHergic neurons as metabolic integrators during exercise stress. Many hormonal systems (ghrelin, leptin, glucose, insulin, and cortisol) are activated to control substrate mobilizations and utilization. The cardiovascular homeostasis, the fluid and electrolyte balance during exercise are highly dependent on vasoactive hormones (antidiuretic hormone, atrial natriuretic peptide, renin–angiotensin–aldosterone, and prostaglandins) control.

Background and Aim: During the physiological growing, thyroid and proteoglycan glycosaminoglycan (GAG) changes dynamically occur, according to genetic and non-genetic factors. The purpose of this research was to compare the effects of early postnatal development (10 days) until 210 days of life on the triiodothyronine (T3), thyroxine (T4), the relative T4:T3 ratio, and GAGs profile, and to define the different reference intervals of the calf's development through the various growing phases. Materials and Methods: The effect of growing on total thyroid hormones and GAG profiles was studied from 10 days to 210 days of age in 64 clinically healthy Brown calves, 30 males and 34 females. Several studies have demonstrated that in woman the sex hormones such as estrogen (E2) and progesterone (P4) influence iron (Fe) regulation, contributing to variations in Fe parameters along the menstrual cycle. These mechanisms based on the regulation of hepcidin (Hepc) which limits Fe availability during the cycle, remain poorly characterized in healthy mares. The objective of this study was to establish the relationship between Hepc, Fe, ferritin (Ferr), and the primary ovarian hormones E2 and P4 in cycling Purebred Spanish mares. Blood samples were taken from 31 Purebred Spanish mares day −5, on day 0, day +5 and day +16 of the cycle. Fe and Ferr significantly increased and Hepc decreased during pre- and ovulatory periods. The secretion peak of estradiol-17β (E2) was reached on day 0 and progesterone (P4) between days +5 and +16. Fe and Ferr were positively correlated (r = 0.57). Fe and Ferr were negatively correlated with Hepc (r = −0.72 and r = −0.02, respectively). E2 and P4 were negatively and positively correlated with Hepc (r = −0.753 and r = 0.54, respectively). In cycling Purebred Spanish mares there is a measurable relationship between steroid hormones and systemic Fe metabolism. Estrogenic dominance in the pre- and ovulatory period allows for a more effective iron status, mediated by hepcidin inhibition. However, P4 during the luteal phase substantially reduces serum Fe and iron stores, possibly related to Hepc stimulation. Future research is required to clarify the relationship between steroid hormones and iron metabolism at the molecular level in equids.

In some species, female steroid hormones modify the profile of acute phase proteins (APPs) during the estrous cycle and pregnancy, according to the ovulation, embryonic implantation and placental development; however, nowadays there's no experimental evidence for equine species. Objectives of this study were: to compare the serum amyloid A (SAA), haptoglobin (Hp) and C-reactive protein (CRP) concentrations between cyclic and pregnant mares, and to analyze the influence of estradiol-17β (E2) during estrous cycle or estrone sulfate (E1) during pregnancy, and progesterone (P4) on these proteins to assess their potential role to identify the cyclicity or pregnancy in Spanish mares. Blood samples were taken from 20 Purebred Spanish mares on the day of ovulation (day 0), on days +5 and +16 post-ovulation, and then, monthly during the whole pregnancy. SAA, Hp and CRP did not change between day 0, +5 and +16 post-ovulation days. P4 concentrations were significantly higher on day +16 than on days +5 and 0; and E2 concentrations were significantly higher on day 0 than day +5. On the other hand, pregnancy was characterized by a progressive increase in the Hp, variable modifications of E1 and P4 concentrations, without changes in SAA and CRP. The absence of significant differences in the APPs between days 0, +5 and +16, suggested that these proteins cannot be used as biomarkers of diagnosis of heat or pregnancy in Spanish mares, at least early, since the Hp later increases during the gestation. Nevertheless, it is possible to use them for comparative purposes with other equine breeds, as supervisor instrument of health status in breeding females as diagnostic tools to monitor pregnancy's development and/or subclinical reproductive inflammations, that could lead to the early embryonic death.

In social animals, recognizing conspecifics and distinguishing them from other animal species is certainly important. We hypothesize, as demonstrated in other species of ungulates, that horses are able to discriminate between the faces of conspecifics and the faces of other domestic species (cattle, sheep, donkeys and pigs). Our hypothesis was tested by studying inter-and intra-specific visual discrimination abilities in horses through a two-way instrumental conditioning task (discrimination and reversal learning), using two-dimensional images of faces as discriminative stimuli and food as a positive reward. Our results indicate that 8 out of 10 horses were able to distinguish between two-dimensional images of the faces of horses and images showing the faces of other species. A similar performance was obtained in the reversal task. The horses' ability to learn by discrimination is therefore comparable to other ungulates. Horses also showed the ability to learn a reversal task. However, these results were obtained regardless of the images the tested horses were exposed to. We therefore conclude that horses can discriminate between two dimensional images of conspecifics and two dimensional images of different species, however in our study, they were not able to make further subcategories within each of the two categories. Despite the fact that two dimensional images of animals could be treated differently from two dimensional images of non-social stimuli, our results beg the question as to whether a two-dimensional image can replace the real animal in cognitive tests.

The study examined the effects of different stages of pregnancy and lactation on thyroid, insulin, and glucose responses in dairy cows. In the present study, 30 dairy cows (10 Holstein, 10 Simmental, and 10 Brown) at 30 ± 20 d of lactation were randomly selected and blood samples were collected once every 60 d for one year to measure circulating thyroid stimulating hormone (TSH), total and free triiodothyronines (T3, fT3) and thyroxines (T4, fT4), insulin, and glucose. Pregnant cows showed higher T4 (p = 0.010) and insulin (p = 0.046) concentrations at >180 d than at >60–120 d of pregnancy and in nonpregnant cows. Along the lactation phase, circulating T4 concentrations showed a biphasic trend, decreasing from 0–60 d to >60–120 d phase, which showed the lowest values, and then increasing until the end of lactation (>300 d) (p = 0.016). Glucose showed the highest concentrations at the start of lactation (0–120 days) and the lowest values at the end (p = 0.008). The monitoring of thyroid hormones, insulin, and glucose changes represents an important tool to evaluate the anabolic and/or catabolic adaptation in response to functional periods in dairy cows, which can potentially predispose the cows to an excessive negative energy balance and related metabolic issues.

This study aimed to determine the thyroid and lipid profiles in 30 Nicastrese goats, along different physiological periods: before mating (nonpregnant goats), during the whole pregnancy (pregnant goats), and during postpartum and early lactation (milking goats). Blood samples were collected monthly from March 2020 to January 2021. Serum thyroid-stimulating hormone (TSH), total and free triiodothyronine (T3, fT3), and thyroxine (T4, fT4) concentrations were measured using immunoenzymatic assay kits and serum lipid panels (triglyceride (TG) and total cholesterol (tCho)) by enzymatic colorimetric method; very-low-density lipoprotein cholesterol (VLDL Cho) was calculated. Pregnant and milking goats showed the lower T3 (p < 0.0002) and T4 (p < 0.0005) concentrations, with lower BCS (p < 0.001) only in pregnant ones. Milking goats showed tCho (p < 0.006) concentrations lower than nonpregnant ones, and TG and VLDL Cho (p < 0.001) lower than both pregnant and nonpregnant goats. T4:T3 and T3:fT3 were significantly and positively correlated in both pregnancy and lactation. Under similar environmental, nutrition, and management conditions, different physiological phases play a significant role in the thyroid and lipid profiles in Nicastrese goats. These endocrine and metabolic resources could contribute to the knowledge useful for the salvage of this endangered, local, native breed.

In cycling females, the periovulatory period is characterized by stimulation of the hypothalamic pituitary adrenal (HPA) axis. The aim of present study was to analyze the pattern and interrelationships among adrenocorticotropic hormone (ACTH), cortisol (CORT), aldosterone (ALD) and electrolytes (sodium—Na+, potassium—K+ and chloride—Cl−) during periovulatory period in cycling mares. Venous blood samples were obtained daily from a total of 23 Purebred Spanish broodmares, aged 7.09 ± 2.5 years, from day −5 to day +5 of estrous cycle, considering day 0, the day of ovulation. Plasma ACTH was measured by a fluorescent immunoassay kit, serum CORT and ALD by means of a competitive ELISA immunoassay, and plasma Na+, K+ and Cl− were quantified by an analyzer with selective electrodes for the three ions. ACTH showed higher concentrations at day 0 compared to days −5 to −1 and +1 to +3 (p < 0.05). CORT showed higher concentrations at day 0 compared to days −5 to −2 and +1 to +5 (p < 0.05). ALD showed higher concentrations at day 0 compared to days −5 to −2 (p < 0.05) and +2 (p < 0.05). Na+ and Cl− showed higher concentrations at day 0, compared to day −5 and +5. K+ showed lower concentrations at day 0 compared to day +1 (p < 0.05). The significant correlations obtained between ACTH and CORT (r = 0.20) and between ACTH and ALD (r = 0.32) suggest that although ACTH may have an effect both on CORT and ALD, there are other very important determinants that could be considered. Hence, it is possible to presume that the pituitary adrenocortical response and ALD may be involved in the ovulatory mechanisms without a direct relation with electrolyte pattern.

In some species, catecholamines in follicular fluid (FF) are related to local physiological events responsible for the regulation of ovarian functions and oocyte maturation. The aim of the present study was to determine and compare intrafollicular and systemic concentrations of dopamine (DA), noradrenaline (NA) and adrenaline (AD) in cycling mares. Sixty ovaries were collected during breeding season from 30 mares raised for slaughterhouse meat production, with clinically normal reproductive tracts, were evaluated. Blood samples were collected prior to slaughter. Follicles were classified into three categories in relation to size: small (20–30 mm; n = 20), medium (≥31–40 mm; n = 20) and large (≥41 mm; n = 20). Follicular fluid (FF) samples were extracted from each follicle. Intrafollicular DA, NA and AD concentrations were significantly higher than the systemic concentrations (p < 0.05). Intrafollicular DA concentrations were higher in medium than small and large follicles (p < 0.05). Intrafollicular NA concentrations were higher in small than medium and large follicles (p < 0.05). Intrafollicular AD concentrations were higher in large than small and medium follicles (p < 0.05). Follicle diameter was significantly and negatively correlated with NA and AD (p < 0.05). A significant correlation of the same hormone concentration in FF and in systemic fluid was observed (p < 0.05). In summary, the FF can serve as an intraovarian catecholamine-storing compartment, with the ability to release neurotransmitters in a regulated way. These results provide novel insights into the neuronal nature of the follicle, suggesting the involvement of catecholamines in normal ovarian functions in mares.

Since in the mare and other animal species such as bitches and cats, the endometrial cell pattern varies depending on the phase of the estrous cycle, the aim of this study was to describe and quantify the endometrial cytological (EC) findings in cycling jennies. EC of eight nonpregnant jennies by cytobrush (CB) at diestrus (day 1 and day 14) and estrous (day 21) were evaluated. All slides were stained with Wright´s stain and microscopically examined at both 400× and 1000× magnification. Seven high-power fields (400×) were assessed in each smear and the endometrial epithelial cells and neutrophils (PMNs) were counted. Endometrial epithelial cells were classified as intact, distorted or fragmented and, on the basis of the presence of dense groups, in monolayer or single clusters. Cytoplasmic characteristics, such as vacuolation or streaming and size, form, position of nuclear characteristics, including karyorrhexis, were recorded. Background aspect, as clear, proteinaceous, or debris, was also considered. In general, sampling by CB provided a yield of cells and clumped endometrial epithelial cells in many smears, being more abundant in estrus than early and late diestrus. Individual endometrial epithelial cells, during estrous, presented a columnar morphology, ciliated or not ciliated and basal nuclei. During diestrus phase, endometrial epithelial cells presented a more cuboidal ciliated or not ciliated morphology. Moderate amount of proteinacious material and red blood cells (RBC) was also observed. Non variation in the percentage of PMNs during diestrus was obtained, but lower and segmented PMNs in CB smears were shown in estrous. This study provides new insights on the physiological changes of endometrial epithelial cells in cycling jennies during the estrus cycle. The CB technique represents a suitable and adequate method for endometrial evaluation, taking into account cytological and/or cytopathological purposes also in jennies.

The hypothesis of this study was to investigate if the presence of corpus luteum (CL) in one ovary could modify the hormonal content of follicular fluid (FF) in the follicles. Sixty ovaries were taken after the slaughter of 30 clinically healthy mares. In relation to the sizes, the follicles were classified into three different categories, as small (20–30 mm), medium (31–40 mm) and large (≥41 mm). Blood samples were collected from the jugular vein of mares before their slaughter, and then the FF samplings were extracted from each single follicle. The ovaries that were collected were classified into two groups, according to the presence (CL-bearing) or absence (non-CL-bearing) of CL. The serum and FF samples were analysed for progesterone (P4), oestradiol-17β (E2), testosterone (T), androstenedione (A4) and dehydroepiandrosterone (DHEA). Intrafollicular P4 concentrations in large follicles of CL-bearing groups were lower than for non-CL-bearing ones. Intrafollicular E2 concentrations increased with the increase of the follicle diameter in both groups, CL-bearing and non-CL-bearing. However, in the FF with a large and medium follicle size, E2 concentrations were significantly higher in non-CL-bearing groups than in CL-bearing groups. T and A4 significantly increased in the large and medium follicle sizes when compared to the small follicle sizes in both groups, but higher concentrations in the non-CL-bearing group were obtained. Intrafollicular DHEA significantly decreased with the increase of the follicular diameter in both groups. Steroid hormones in FF dynamically changed, according to the presence or not of CL in the ovary. This study brings new knowledge on the role of the CL in the follicular hormonal composition in mares.

Aim: The purpose of the research was to test and validate an innovative and safe chemiluminescence method to evaluate sexual hormones in serum and plasma samples of Tursiops truncatus. Materials and Methods: The research was performed on 9 bottlenose dolphins entertained in Oltremare and in Zoomarine aquatic parks, sampled by the tail vein or from the ventral one and an ultrasound monitoring, throughout a 6 months period. Blood samples were analyzed using a chemiluminescence method. Data obtained were compared to radioimmunoassay and enzyme immuno assay reference data, with the purpose to test and validate this method, through the calculation of the coefficient of variability, and its reliability on serum and plasma samples. A one-way analysis of variance was applied to test the effect of time on serum and plasma hormonal changes. Results: Mean concentrations of estradiol-17β in serum were equal to 149.07±6.82 pmol/L, and in plasma equal to 159.14±12.99 pmol/L; mean values of progesterone in serum were equal to 0.69±0.05 pmol/L, and in plasma equal to 0.64±0.05 pmol/L; mean values of testosterone in serum were equal to 44.43±14.42 nmol/L, and in plasma equal to 48.99±11.20 nmol/L. Conclusion: It would be interesting to widen the investigations on a larger number of subjects, in which the relationship between the concentrations of free and binding steroid hormones, with the dosing of binding proteins, would define the physiological ranges of reference in the T. truncatus.

North-eastern Sicily is an area with iodine deficiency disorders occurring in both humans and animals. The aim of this study was to test the influences of an endemic goitre area on iodothyronine ranges and their pattern in adult ovine and caprine specimens stabled in different locations of Sicily, taking into account the different sexes. A total of 48 Comisana sheep and 51 Maltese goats was studied. The sheep included 10 females and 6 males stabled in a non-endemic goitre area (farm A: control group), and 16 females and 16 males stabled in an endemic goitre area (farm B: observational group). The goats included 6 females and 13 males stabled in a non-endemic goitre area (farm A: control group), and 16 females and 16 males stabled in an endemic goitre area (Farm B: observational group). The results showed lower T3 and higher fT4 (P< 0,0001), ki so bili nastanjeni na kmetiji B v primerjavi s kmetijo A. V primerjavi s kmetijo A so pri kozah, ki so bile nastanjene na kmetiji B, ugotovili višje ravni prostega T3 (p < 0,0001), pri samcih na kmetiji B nižje ravni T4 (p < 0,0001), pri samicah pa višjo raven prostega T4 (p < 0,0001). Ugotovljene so bile statistično značilne razlike med skupinami tako glede na spol kot glede na endemičnost področja na ravni vezanih in prostih jodotironinov pri ovcah in kozah. Pridobljeni podatki so pokazali, da območja z endemično golšavostjo pri ljudeh nimajo nujno enakega učinka pri ovcah in kozah, za katere se zdi, da so sposobne ustvariti prilagoditveno strategijo, zaradi katere se ne pokažejo klinični znaki bolezni ščitnice. Nutrition plays a leading role that most influences thyroid response and energetic metabolism. Aim was to compare the effect of diet on thyroid and lipid status in guide dogs during a 12-weeks training period. Eight Labrador Retrievers were divided into two groups homogeneous for sex, age, body weight, and Body Condition Score (BCS) and fed two commercial diets one, HPF, characterized by low-carbohydrate/high-protein/high-fat (29%:39%:19% as-fed) and the other, LPF, by high-carbohydrate/low-protein/low-fat (50%:24%:12% as-fed) content. The serum thriiodothyronine (T3), thyroxine (T4), cholesterol (CHOL), triglycerides (TAGs) and non-esterified fatty acids (NEFA) were determined at Day 0, 28, 56, and 84, before the daily training. Statistical model included the effects of Diet (HPF vs. LPF) and Time (Day 0 to Day 84), and their interaction. In the HPF group, Diet significantly (p < 0.01) increased T4, CHOL, and TAGs and decreased NEFA. In both groups, Time significantly (p < 0.05) increased T4 and TAGs, CHOL at Day 28, and NEFA at Day 56. The interaction did not influence serum hormones and lipid pattern. The adjustments in thyroid and lipid responses to moderate exercise in HPF group were driven mainly by the nutrient composition of the diet in relation to the involvement of metabolic homeostasis.
