3. Does Size Matter? Animal, Living and Non-Living Classification, Implications for Teaching

- Abstract
An empirical investigation of elementary school teacher candidates on classification activities dealing with animate and inanimate objects in terms of being living or non-living demonstrates that as the size of the objects increases, subjects are more likely to classify them correctly as either being an animal or having living characteristics. Despite a variety of misconceptions having an impact on the results, size magnitude is shown to play a significant role on proper classification. The subjects' performance on these activities at an advanced stage of their preparation suggests that their factual and procedural knowledge are deficient due to a lack of opportunities for conceptual development of the items tested. The identification of the role of size on the proper classification of objects in the activities bears significantly on the science curricular structure at the elementary school level. As the results of this study indicate, both pre-service elementary school teachers and by extension their prospective students need longer practice dealing with living and non-living classification activities, particularly in tasks where the microscopic features of matter can be investigated so that the proclivity to regard size as the defining characteristic is effectively addressed. more...

5. Impact of Guided Inquiry with Simulations on Knowledge of Electricity and Wave Phenomena

The impact of inquiry-based instruction on the improvement of content knowledge of electricity and wave properties in two studies was the subject of this investigation. Groups of pre-college and undergraduate students performed a series of tasks designed to determine the effectiveness of exploratory/investigative approaches to the study of wave properties in fostering content retention and conceptual change, as well as in comparison with traditionally performed experimental activities. The results show that a guided inquiry-based method using simulations significantly improves content performance on electricity and wave motion, although not in optics; in particular, it helps students to visualize charge exchanges and electric neutrality, the shape of electric fields, types of waves and their characteristics, and the effects of the transmitting medium on the speed of a wave. Additionally, its exploratory nature is superior to the confirmatory laboratory experience, both in content retention and in facilitating the incorporation of perceptual features that help learners deal with documented challenging properties of waves. more...

Treatment of tumors with Toca 511, a gamma retroviral replicating vector encoding cytosine deaminase, followed by 5-fluorocytosine (5-FC) kills tumors by local production of 5-fluorouracil (5-FU). In brain tumor models, this treatment induces systemic anti-tumor immune responses and long-term immune-mediated survival. Phase 1 Toca 511 and Toca FC (extended-release 5-FC) clinical trials in patients with recurrent high-grade glioma show durable complete responses and promising survival data compared to historic controls. The work described herein served to expand on our earlier findings in two models of metastatic colorectal carcinoma (mCRC). Intravenous (i.v.) delivery of Toca 511 resulted in substantial tumor-selective uptake of vector into metastatic lesions. Subsequent treatment with 5-FC resulted in tumor shrinkage, improved survival, and immune memory against future rechallenge with the same CT26 CRC cell line. Similar results were seen in a brain metastasis model of mCRC. Of note, 5-FC treatment resulted in a significant decrease in myeloid-derived suppressor cells (MDSCs) in mCRC tumors in both the liver and brain. BackgroundToca 511 (vocimagene amiretrorepvec) is a retroviral replicating vector encoding an optimized yeast cytosine deaminase (CD). Tumor-selective expression of CD converts the prodrug, 5-fluorocytosine (5-FC), into the active chemotherapeutic, 5-fluorouracil (5-FU). This therapeutic approach is being tested in a randomized phase II/III trial in recurrent glioblastoma and anaplastic astrocytoma (NCT0241416). The aim of this study was to identify the immune cell subsets contributing to antitumor immune responses following treatment with 5-FC in Toca 511-expressing gliomas in a syngeneic mouse model.MethodsFlow cytometry was utilized to monitor and characterize the immune cell infiltrate in subcutaneous Tu-2449 gliomas in B6C3F1 mice treated with Toca 511 and 5-FC.ResultsTumor-bearing animals treated with Toca 511 and 5-FC display alterations in immune cell populations within the tumor that result in antitumor immune protection. Attenuated immune subsets were exclusive to immunosuppressive cells of myeloid origin. Depletion of immunosuppressive cells temporally preceded a second event which included expansion of T cells which were polarized away from Th2 and Th17 in the CD4+ T cell compartment with concomitant expansion of interferon gamma-expressing CD8+ T cells. Immune alterations correlated with clearance of Tu-2449 subcutaneous tumors and T cell-dependent protection from future tumor challenge.ConclusionsTreatment with Toca 511 and 5-FC has a concentrated effect at the site of the tumor which causes direct tumor cell death and alterations in immune cell infiltrate, resulting in a tumor microenvironment that is more permissive to establishment of a T cell mediated antitumor immune response. more...

Wigandia ecuadoriensis , a member of the Namaceae family, is a source of metabolites and has been traditionally used as an anti-inflammatory. This work aimed to determine the total phenolic content (TPC), total flavonoid content (TFC), antioxidant effect, inhibition of α-glucosidase and cholinesterase enzymes (AChE, BChE), and antibacterial activity of the methanolic extract (ME) and subfractions of Wigandia ecuadoriensis. The findings revealed that ME and its subfractions exhibited significant antioxidant capacity, with the ethyl acetate fraction being the most active, displaying an IC
50 of 17.66 µg/mL against the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 10.31 µg/mL against 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS). This activity was attributed to its high total phenolic content (357.47 mg GAE/g). Furthermore, W. ecuadoriensis fractions showed marked antimicrobial properties against human pathogen strains with Minimum Bactericidal Concentration (MBC) values of 1.56–6.25 mg/mL for S. aureus , E. faecalis and E. coli. Furthermore, aqueous fraction exhibited slight inhibition of acetylcholinesterase (IC50 : 915.98 µg/mL) and butyrylcholinesterase (IC50 : 380.42 µg/mL). Interestingly, EF showed the greatest inhibitory effect of α-glucosidase (IC50 : 38.44 µg/mL) which is more potent than the control used, acarbose (IC50 : 179.07 µg/mL). UHPLC-QTOF-MS analysis identified forty compounds, including phenolic acids, flavonoids, saponins, terpenes, and fatty acyls. As far as we know, this is the first study to evaluate the chemical composition and biological potential of W. ecuadoriensis. Our results provide the first evidence to the chemical knowledge of the species W. ecuadoriensis and demonstrate its bioactive potential as an interesting source of secondary metabolites with possible beneficial properties for health. Toca 511 (vocimagene amiretrorepvec), a nonlytic, amphotropic retroviral replicating vector (RRV), encodes and delivers a functionally optimized yeast cytosine deaminase (CD) gene to tumors. In orthotopic glioma models treated with Toca 511 and 5-fluorocytosine (5-FC) the CD enzyme within infected cells converts 5-FC to 5-fluorouracil (5-FU), resulting in tumor killing. Toca 511, delivered locally either by intratumoral injection or by injection into the resection bed, in combination with subsequent oral extended-release 5-FC (Toca FC), is under clinical investigation in patients with recurrent high-grade glioma (HGG). If feasible, intravenous administration of vectors is less invasive, can easily be repeated if desired, and may be applicable to other tumor types. Here, we present preclinical data that support the development of an intravenous administration protocol. First we show that intravenous administration of Toca 511 in a preclinical model did not lead to widespread or uncontrolled replication of the RVV. No, or low, viral DNA was found in the blood and most of the tissues examined 180 days after Toca 511 administration. We also show that RRV administered intravenously leads to efficient infection and spread of the vector carrying the green fluorescent protein (GFP)-encoding gene (Toca GFP) through tumors in both immune-competent and immune-compromised animal models. However, initial vector localization within the tumor appeared to depend on the mode of administration. Long-term survival was observed in immune-competent mice when Toca 511 was administered intravenously or intracranially in combination with 5-FC treatment, and this combination was well tolerated in the preclinical models. Enhanced survival could also be achieved in animals with preexisting immune response to vector, supporting the potential for repeated administration. On the basis of these and other supporting data, a clinical trial investigating intravenous administration of Toca 511 in patients with recurrent HGG is currently open and enrolling. more...

Patients with the most common and aggressive form of high-grade glioma, glioblastoma multiforme, have poor prognosis and few treatment options. In 2 immunocompetent mouse brain tumor models (CT26-BALB/c and Tu-2449-B6C3F1), we showed that a nonlytic retroviral replicating vector (Toca 511) stably delivers an optimized cytosine deaminase prodrug activating gene to the tumor lesion and leads to long-term survival after treatment with 5-fluorocytosine (5-FC). Survival benefit is dose dependent for both vector and 5-FC, and as few as 4 cycles of 5-FC dosing after Toca 511 therapy provides significant survival advantage. In the virally permissive CT26-BALB/c model, spread of Toca 511 to other tissues, particularly lymphoid tissues, is detectable by polymerase chain reaction (PCR) over a wide range of levels. In the Tu-2449-B6C3F1 model, Toca 511 PCR signal in nontumor tissues is much lower, spread is not always observed, and when observed, is mainly detected in lymphoid tissues at low levels. The difference in vector genome spread correlates with a more effective antiviral restriction element, APOBEC3, present in the B6C3F1 mice. Despite these differences, neither strain showed signs of treatment-related toxicity. These data support the concept that, in immunocompetent animals, a replicating retroviral vector carrying a prodrug activating gene (Toca 511) can spread through a tumor mass, leading to selective elimination of the tumor after prodrug administration, without local or systemic pathology. 49. Graphical Representations and the Perception of Motion: Integrating Isomorphism through Kinesthesia into Physics Instruction

- Abstract
The incorporation of engaging and meaningful learning experiences is essential for the enhancement of critical thinking and the development of scientific literacy. The study engaged several groups of students in activities designed to elicit their understanding of a graphical representation of motion, and to determine the kinesthetic effect of using a computer-interfaced motion detector in two isomorphic tasks. (I) A forensic context provided an abstracted determination of their ability to describe the motion of an intruder from the data collected by a sensor, and displayed as a graph; (II) An analysis of the motion of two pendulums, a real and a virtual one (as a simulation). An investigation of students' analysis of graphical representations of motion also emphasized the transferability of similar concepts from hypothetical/virtual scenarios to an experimental task to determine the impact of kinesthesia on performance. The results indicate that the features associated with subjects' own movements, and those of other real objects are statistically and cognitively significant in their improvements on the analysis of graphical representations of motion. The findings have broad implications for the use of inquiry-based and technology-rich contexts in the teaching of science. more...

Mannan oligosaccharides (MOS) have shown to stimulate immune response in different fish species, but the results may appear contradictory and have not been tested in conjunction with vaccination. We hypothesized that dietary MOS supplementation could enhance efficacy and immune responses after immunization with a novel hydrogen peroxide‐inactivated vaccine. Nile Tilapia, Oreochromis niloticus, fingerlings were distributed into four groups (supplemented or not, vaccinated or not) and were used to determine immune and hematological parameters and somatic indexes. For the bacterial challenge and vaccine efficacy test, fish were assigned to five experimental groups. Twenty‐eight days after the feeding trial and vaccination, the groups were challenged intraperitoneally and monitored for 21 days. The relative percent survival of the groups basal diet‐vaccinated and MOS diet‐vaccinated were 98.1% and 100.0%, respectively. The dietary inclusion of MOS after vaccination for 28 days had no influence on vaccine efficacy, which could be explained because of the high efficacy of the vaccine itself. Therefore, we suggest that the contribution of MOS supplementation to immune responses after vaccination in fish would be greater for vaccines with lower efficacy, or when tested for heterologous challenges or for crossed protection.
