Abstract

Food addiction is characterized by a loss of behavioral control over food intake and is associated with obesity and other eating disorders. The mechanisms underlying this behavioral disorder are largely unknown. We aimed to investigate the changes in miRNA expression promoted by food addiction in animals and humans and their involvement in the mechanisms underlying the behavioral hallmarks of this disorder. We found sharp similitudes between miRNA signatures in the medial prefrontal cortex (mPFC) of our animal cohort and circulating miRNA levels in our human cohort, which allowed us to identify several miRNAs of potential interest in the development of this disorder. Tough decoy (TuD) inhibition of miRNA-29c-3p in the mouse mPFC promoted persistence of the response and enhanced vulnerability to developing food addiction, whereas miRNA-665-3p inhibition promoted compulsion-like behavior and also enhanced food addiction vulnerability. In contrast, we found that miRNA-137-3p inhibition in the mPFC did not lead to the development of food addiction. Therefore, miRNA-29c3p and miRNA-665-3p could be acting as protective factors with regard to food addiction. 7. nf-core/proteinfold: a bioinformatics best-practice analysis pipeline for protein 3D structure prediction

Abstract

The advances in deep learning frameworks have revolutionised protein studies and contributed to unprecedented accurate predictions of protein structures. The release of AlphaFold2 has materialised this major breakthrough. AlphaFold2 has also paved the way towards the development of a new generation of publicly available prediction tools whose combination constitutes a powerful toolkit for the systematic study of unknown proteomes. Despite their obvious usefulness, the deployment of these new methods across the research community remains hampered by technical hurdles. The main challenges are the dependency of each prediction method on a wealth of external software and sequence databases as well as the lack of a workflow framework that could orchestrate the execution of such tools in a scalable, portable and reproducible manner under well-defined standards. These issues can be specifically addressed by Nextflow together with the nf-core community initiative. Here we present nf-core/proteinfold (https://github.com/nf-core/proteinfold), a Nextflow pipeline developed according to nf-core guidelines, which simplifies the use of state-of-the-art techniques in protein structure modelling. These best-practice guidelines ensure that the pipeline is properly optimised for execution on the major cloud providers as well as HPC infrastructures. We foresee that this development endeavour will have a significant impact on a variety of biological analyses based on protein structures by granting access to an open-source, community-developed resource to obtain protein folds.

Abstract

Down syndrome (DS) individuals present increased risk for Alzheimer's disease (AD) neuropathology and AD-type dementia. Here, we investigated the use of green tea extracts containing (-)-epigallocatechin-3-gallate (EGCG), as co-adjuvant to enhance the effects of environmental enrichment (EE) in Ts65Dn mice, a segmental trisomy model of DS that partially mimics DS/AD pathology, at the age of initiation of cognitive decline. Classical repeated measures ANOVA showed that combined EE-EGCG treatment was more efficient than FE or EGCG alone to improve specific spatial learning related variables. Using principal component analysis (PCA) we found that several spatial learning parameters contributed similarly to a first PC and explained a large proportion of the variance among groups, thus representing a composite learning measure. This PC1 revealed that EGCG or FE alone had no significant effect. However, combined FE-EGCG significantly ameliorated learning alterations of middle age Ts65Dn mice. Interestingly. PCA revealed an increased variability along learning sessions with good and poor learners in Ts65Dn, and this stratification did not disappear upon treatments. Our results suggest that combining EE and EGCG represents a viable therapeutic approach for amelioration of age-related cognitive decline in DS, although its efficacy may vary across individuals., Peer Reviewed, Postprint (published version)
