Pulmonary fibrosis is a sequelae of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) infection that currently lacks effective preventative or therapeutic measures. Post-viral lung fibrosis due to SARS-CoV-2 has been shown to be progressive on selected patients using imaging studies. Persistent infiltration of macrophages and monocytes, a main feature of SARS-CoV-2 pulmonary fibrosis, and long-lived circulating inflammatory monocytes might be driving factors promoting the profibrotic milieu in the lung. The upstream signal(s) that regulates the presence of these immune cells (despite complete viral clearance) remains to be explored. Current data indicate that much of the stimulating signals are localized in the lungs. However, an ongoing low-grade systemic inflammation in long Coronavirus Disease 2019 (COVID-19) symptoms suggests that certain non-pulmonary regulators such as epigenetic changes in hematopoietic stem cells might be critical to the chronic inflammatory response. Since nearly one-third of the world population have been infected, a timely understanding of the underlying pathogenesis leading to tissue remodeling is required. Herein, we review the potential pathogenic mechanisms driving lung fibrosis following SARS-CoV-2 infection based upon available studies and our preliminary findings (Graphical abstract). However, no studies have assessed acute changes in IL-6 concentration after consecutive days of extreme and long-term exercise.Objective: This study aimed to assess acute changes in serum IL-6 concentration during four days of long-distance walking.Methods: This prospective observational study assessed 25 athletes (aged 44.8 ± 9.1 years), who covered a total of 251 km in four days. Blood samples were collected daily to assess serum IL-6 concentrations. Repeated-measures analysis of variance (with Bonferroni’s post hoc test) and the Kruskal–Wallis H-test (with Dunn’s post hoc test) were used to investigate the differences between the measures.Results: The serum IL-6 concentrations were higher on the four days of walking (1st day: 26.8 ± 14.8; 2nd day: 14 ± 7.4; 3rd day: 9.4 ± 10.8; 4th day: 4.5 ± 0.2 pg/mL) when compared to pre-walk values (pre-walk: 2.2 ± 2.1 pg/mL; p < 0.001). On the first day, there was a tenfold increase compared to the pre-walk value.Conclusion: The inflammatory response increased the serum concentration of IL-6 after four days of exercise. Desenvolveram-se estudos sobre flebotomíneos em área de leishmaniose tegumentar, fazenda Boa Sorte, Município de Corguinho, Mato Grosso do Sul, Brasil, com vistas a incriminar vetor dessa parasitose. No início dos estudos, encontravam-se bem preservados vários tipos da cobertura vegetal primitiva, com predomínio de cerrado e cerradão, denominado localmente de "croa". Decorridos quatro meses, parte significativa da "croa" e do cerrado foi queimada para transformação em áreas de pastagens. Durante julho/1991 a junho/93, realizaram-se coletas semanais das 18:00 às 6:00 horas, com armadilha CDC (Center on Disease Control), em floresta-galeria, floresta de encostas, cerrado, "croa", peridomicílio (chiqueiro e poleiro) e no interior de uma tulha; coletas mensais com armadilha de Shannon das 18:00 às 24:00 horas em floresta-galeria e "croa". De junho/91 a setembro de 1992, capturas mensais com isca humana, por 24 horas, em floresta-galeria. Investigou-se infecção natural por flagelados em flebotomíneos coletados com armadilha de Shannon e isca humana. As coletas com CDC resultaram 24 espécies de Lutzomyia e duas de Brumptomyia. A "croa" foi o ambiente que mais contribuiu com espécimens e que apresentou a maior diversidade, juntamente com a floresta de encostas. Nas coletas com CDC, L. whitmani revelou-se a mais abundante, índice de abundância padronizado = 0,991; porém, esteve muito pouco representada no interior do anexo domiciliar; apresentou prevalência de 96,0% nas armadilhas de Shannon e isca humana, respectivamente com 3.265 e 516 espécimens. Sua maior freqüência deu-se em épocas frias e secas. Dotada de atividade quase que exclusivamente noturna, exibiu pico de ocorrência das 18:00 às 19:00 horas. A taxa de infecção natural por flagelados, em 680 fêmeas de flebotomíneos dissecadas, foi de 0,15% e, entre 613 fêmeas de L. whitmani, de 0,16%. Com base em seu comportamento, L. whitmani foi incriminada como provável vetora da leishmaniose tegumentar na área. A segunda espécie mais abundante, L. lenti, não demonstrou antropofilia. Apresenta-se também a fauna flebotomínica por ambientes.

Abstract Background Programmed cell death (apoptosis) has been implicated in tumor development and may affect the metastatic potential of tumor cells. The role of bcl-2, a proto-oncogene that inhibits apoptosis, has been studied in several malignancies, including cutaneous melanoma (CM). The purpose of this study was to evaluate the immunohistochemical expression of bcl-2 in 35 regional lymph node, 28 subcutaneous and 17 visceral CM metastases, correlating the findings with patient survival. Methods In a historical cohort study patient survival was correlated with the expression of bcl-2 in regional lymph node, subcutaneous and visceral metastases of CM. Eighty slides containing surgical specimens from 50 patients diagnosed with stage III and IV CM, 28 male (56%) and 22 female (44%), were analyzed. Mean age at diagnosis was 43 years (16–74 years; median = 42 years). Mean Breslow depth was 5.01 mm (0.4–27.5 mm). The slides were submitted to immunohistochemical reaction using anti-bcl-2 monoclonal antibody and classified according to the degree of staining (< 5%; 5 to 50%; or > 50% of tumor cells stained). The relationship between bcl-2 protein expression and survival for each type of metastasis, gender and age at initial diagnosis was analyzed. Results Mean overall survival was 33.9 months after the diagnosis of the initial metastatic lesion (range: 0 to 131 months). Twenty-four out of 50 patients (48%) had died from CM by the end of the study period. bcl-2 expression was detected in 74.3, 85.7 and 82.4% of lymph node, subcutaneous and visceral metastases, respectively. After univariate and multivariate analyses, no correlation was found between positive bcl-2 expression and overall survival for the types of metastases evaluated. Conclusion The immunohistochemical expression of bcl-2 in metastasis alone is not a prognostic marker for CM.

Polycrystalline CuIn 1 − x Al x Se 2 (CIASe) thin films have been prepared on bare and Mo-coated substrates by selenization of evaporated Cu–In–Al precursors layers with 0≤ x ≤0.3 and thicknesses below 1.4 µm. X-ray diffraction measurements show several peaks around the (112) reflection corresponding to CIASe phases with different Al contents. This has been attributed to the existence of a depth compositional gradient in the layers with higher Al contents due to a diffusion of Al towards the back contact region. Raman scattering measurements directly performed with the excitation laser spot on both the front and back surfaces from absorbers that were previously mechanically removed from the Mo coated substrates confirm the existence of complex CIASe absorbers with graded Al content, which leads to a corresponding grading of the absorber bandgap. Enrichment of Al content at the back contact region is also accompanied by a decrease of the grain size as shown by cross-sectional Scanning Electron Microscopy (SEM). The best CIASe based solar cells have J sc , V oc , FF and efficiencies of 29.3 mA/cm 2 , 430 mV , 51.1% and 6.5%, respectively, for an overall Al/(In+Al) relative content x ~0.2. Comparison of these values with those from equivalent devices made without Al show a significant increase of the device efficiency. These data confirm the existence of a significant improvement of the device performance because of the increased absorber bandgap in the CIASe quaternary alloys. [ABSTRACT FROM AUTHOR]

Cu 2 ZnSnS 4 (CZTS) solar cells commonly undergo post-annealing treatments to enhance their performance. However, multiple concurrent effects, including Cu-Zn disorder, Na diffusion from the back contact, CZTS grain boundary passivation, and elemental interdiffusion at the CdS/CZTS interface, can occur simultaneously during post-annealing, making it challenging to isolate their specific influence on device performance. To address this complexity, we have utilized Cu-Zn-disordered CZTS absorbers to eliminate the influence of Cu-Zn disorder in this study. Post-annealing experiments conducted at 120 to 300 °C in nitrogen and air atmospheres aim to decouple and assess the impact of different annealing conditions on the performance of CZTS solar cells. Our findings suggest that the optimal post-annealing might be a combined effect of two distinct processes: CZTS absorber surface passivation occurring at 325 °C and CdS crystallization at 225 °C. Objective: The most commonly used methods for bone age (BA) reading were described in the Caucasian population decades ago. However, there are secular trends in skeletal maturation and different BA patterns between ethnic groups. Automated BA reading makes updating references easier and more precise than human reading. The objective of the present study was to present automated BA reference curves according to chronological age and gender in the Mexican population and compare the maturation tempo with that of other populations., Methods: The study included 923 healthy participants aged 5 to 18 years between 2017 and 2018. A hand radio-graph was analyzed using BoneXpert software to obtain the automated BA reading according to Greulich and Pyle (G&P) and Tanner-Whitehouse 2 (TW2) references. We constructed reference curves using the average difference between the BA and chronological age according to sex and age., Results: The G&P and TW2 automated reference curves showed that Mexican boys exhibit delays in BA during middle childhood by 0.5 to 0.7 (95% confidence interval &lsqb;CI], -0.9 to -0.2) years; however, they demonstrate an advanced BA of up to 1.1 (95% CI, 0.8 to 1.4) years at the end of puberty. Mexican girls exhibited a delay in BA by 0.3 to 0.6 (95% CI, -0.9 to -0.1) years before puberty and an advanced BA of up to 0.9 (95% CI, 0.7 to 1.2) years at the end of puberty., Conclusion: Mexican children aged <10 years exhibited a delay in skeletal maturity, followed by an advanced BA by approximately 1 year at the end of puberty. Lung fibrosis is an unabated wound healing response characterized by the loss and aberrant function of lung epithelial cells. Herein, we report that extracellular Clusterin promoted epithelial cell apoptosis whereas intracellular Clusterin maintained epithelium viability during lung repair. Unlike normal and COPD lungs, IPF lungs were characterized by significantly increased extracellular Clusterin whereas the inverse was evident for intracellular Clusterin. In vitro and in vivo studies demonstrated that extracellular Clusterin promoted epithelial cell apoptosis while intercellular Clusterin modulated the expression of the DNA repair proteins, MSH2, MSH6, OGG1 and BRCA1. The fibrotic response in Clusterin deficient (CLU-/-) mice persisted after bleomycin and it was associated with increased DNA damage, reduced DNA repair responses, and elevated cellular senescence. Remarkably, this pattern mirrored that observed in IPF lung tissues. Together, our results show that cellular localization of Clusterin leads to divergent effects on epithelial cell regeneration and lung repair during fibrosis.

Selenium (Se) deficiency has been associated with lowered resistance to mastitis in dairy cattle. However, little published data exists on the effect of Se supplementation before calving on udder health of pastured dairy heifers. Further, the relative efficacy of injectable barium selenate and oral organic Se for improving udder health in cows has not previously been tested. The objectives of this study were to determine the effects of precalving Se supplementation and type of supplementation on the blood activity of glutathione peroxidase and measures of udder health immediately after calving and during the first month of lactation in pastured dairy heifers. One hundred forty pregnant Chilean Holstein-Friesian heifers were fed a basal diet containing, on average, 0.15 mg of Se/kg of dry matter. One month before predicted calving, heifers were allocated to 1 of 3 groups. Group 1 (n=49) received no supplementary Se, group 2 (n=46) received a single subcutaneous injection of Se (1 mg/kg of live weight, as barium selenate), and group 3 (n=45) was fed Se yeast (3 mg/heifer/d until calving). Heifers supplemented with barium selenate had a higher glutathione peroxidase activity from 14 d in milk onwards. Selenium supplementation, irrespective of source, tended to reduce the prevalence of intramammary infection (IMI) and decrease the prevalence of quarters with high somatic cell count (SCC) at calving. Overall, Se supplementation did not result in a reduction of the incidence of new IMI or clinical mastitis or in decreased SCC during the balance of the first month of lactation. However, in pasture-based heifers injected with barium selenate before calving, and fed diets with 1.3 and 2.5 mg of Se/d precalving and during lactation, respectively, no cases of clinical mastitis were observed in the first month of lactation., (Copyright © 2010 American Dairy Science Association. Five multiparous Holstein cows in midlactation that were fitted with ruminal and duodenal cannulas were used in a 3 x 5 incomplete Latin square. The objective of this study was to examine the effects on nutrient digestion of wheat processing and method of tallow addition to the diets of lactating dairy cows. Diets consisted of 45% forage and 55% concentrate, and each diet contained 20% wheat and 2% tallow (as-fed basis). Treatments were dry-rolled wheat with tallow added to the concentrate, steam-rolled wheat with tallow added to the concentrate, and steam-rolled wheat with tallow added first to the wheat. The dry matter intake; digestion of starch, fiber, and fatty acids; ammonia N concentration; and molar proportions of volatile fatty acids in ruminal fluid were not affected by treatments. The apparent digestibility in the total tract of organic matter and nitrogenous compounds was significantly higher for the steam-rolled treatment with tallow added first to the wheat. Mean ruminal fluid pH was similar across treatments; however, cows fed the diet containing steam-rolled wheat with tallow added first to the wheat had the smallest pH change from 0 to 2 h postfeeding. Milk yield did not differ, regardless of cow diet. Method of tallow addition had marked effects on the apparent digestibility of organic matter and N in the total tract of lactating dairy cows.
