We aimed to examine the effects of brain ischemia-reperfusion (IR) especially on serum parameters or liver enzymes, free radicals, cytokines, oxidatively damaged DNA, spermidine/spermine N-1-acetyltransferase (SSAT). The effects of addition of putrescine on IR will be evaluated in terms of inflammation and oxidant-antioxidant balance in liver.The study was conducted on 46 male Albino Wistar rats weighing 200–250 g. The rats were grouped into: 1-Sham group (n = 6). 2-IR group (n = 8): The carotid arteries were ligated for 30-min and reperfusion was achieved for 30-min under general anesthesia. 3-Ischemia + putrescine + reperfusion group (IPR) (n = 8): Unlike the IR group, a single dose of 250 μmol kg−1 putrescine was given by gavage at the beginning of reperfusion. In putrescine treatment groups in addition to the procedures performed in the IR group a total of 4 doses of 250 μmol kg−1 putrescine were given at 12-h intervals, with the first dose immediately after 30-min reperfusion (4-IR+putrescine group (IR+P1) (n = 8)); 3 h after the 30-min reperfusion (5-IR+putrescine group (IR+P2) (n = 8)); 6 h after the 30-min reperfusion (6-IR+putrescine group (IR+P3) (n = 8)). ALT, AST, ATP, NO, SSAT, 8-OHdG levels were analyzed in the serum, and liver samples. NF-κB and IL-6 levels were analyzed in the liver samples.Brain IR causes inflammatory, oxidative and DNA damage in the liver, and putrescine supplementation through gavage reduces liver damage by showing anti-inflammatory and antioxidant effects.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most common malignant tumors of the pancreas. Preclinical studies show that it evades the immune system with immune checkpoints and promotes tumor development. V-domain Ig suppressor of T cell activation (VISTA) is a new immune-check point from the B7 family and is highly expressed in cancer cells. Overexpression of toll like receptor 4 (TLR4) in pancreatic adenocarcinoma is associated with inducing tumorigenesis, tumor growth and resistance to chemotherapy. Naloxone is an opioid and inhibits TLR4-ligand association. In this study, we investigated the connection of TLR4 and downstream pathway with immune-check point VISTA in pancreatic cancer proliferation. We first collected pancreatic cancer-related datasets using the GEPIA2 and UALCAN databases. Based on the data obtained the effect of different concentrations and incubation times of Naloxone were used on PANC-1 cells proliferation. Combination of Naloxone and VISTA-siRNA were applied, and effect of both Naloxone and combined reatment on TLR4, Interleukin 1 receptor associated kinase 4 (IRAK4) and VISTA gene expression were analysed in pancreatic cancer cells. As a result of analysis with Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR), gene expression levels of TLR4, IRAK4 and VISTA were significantly suppressed and cell proliferation was significantly reduced. We found that administration of Naloxone and VISTA-siRNA in combination to PDAC cells suppressed signaling. Therefore, we considered that the relationship between VISTA and TLR4 signaling pathways and the other possible associated signal molecules may be an important marker in determining the response of immune checkpoint inhibitors in cancer treatment.

Objectives We aimed to determine the differences between normozoospermic and oligozoospermic individuals according to levels of spermatid-specific thioredoxin reductase 3 (SPTRXR3/STRX3/TXNDC8/TXNRD3) and testis expressed protein 101 (TEX-101), and to evaluate the correlations between spermiogram data and biochemical parameters. Methods The study was carried out at the Andrology Laboratory of Medicine Faculty of Selcuk University. Two groups were designed: Group 1: Normozoospermia (n=40, sperm concentration ≥ 15 million/mL), Group 2: Oligozoospermia; (n=40, sperm concentration Results TEX-101 protein levels were significantly different in normozoospermia (2.12 ± 0.08 ng/mL) compared to oligozoospermia (1.55 ± 0.04 ng/mL). SPTRXR3 levels (6.98 ± 0.46 ng/mL) were higher in oligozoospermia than normozoospermia (3.07 ± 0.35 ng/mL). Both TEX-101 and SPTRXR3 levels were correlated statistically with most of the spermiogram parameters. Conclusions High SPTRXR3 and low TEX-101 levels may be a biomarker in evaluation of male infertility. The relations between spermiogram parameters indicates that results present a new clinical approach in biology of oligozoospermic male.

OBJECTIVES: This research was aimed to find out the effects of 3',4'-dihydroxyflavonol (DiOHF) on apoptosis, DNA damage, and tumor necrosis factor-α (TNF-α) levels in the frontal cortex of rats with induced experimental brain ischemi reperfusion. MATERIALS AND METHODS: A total of 38 Wistar albino male rats were used. Groups were created as 1-Sham; 2-Ischemia-reperfusion (I/R); 3-I/R + DiOHF (10 mg/kg); 4-Ischemia + DiOHF + reperfusion; 5-DiOHF + I/R. I/R was performed by carotid artery ligation for 30 min in anesthesized animals. Following experimental applications, blood samples were taken from anesthetized rats to obtain erythrocyte and plasma. Later, the rats were killed by cervical dislocation, and frontal cortex samples were taken and stored at − 80° C for the analysis. RESULTS: In the ischemic frontal cortex tissue sections degenerate neuron numbers, Terminal deoxynucleotidyl transferase-dUTP nick end labeling (TUNEL) positive cell ratio and caspase-3 positive cell ratio increased. Malondialdehyde, TNF-α, and 8-OHdG levels were increased in both plasma and tissue in ischemia group, whereas tissue and erythrocyte glutathione levels were significantly suppressed. However, these values were significantly reversed by DiOHF treatment. CONCLUSION: The results of the study showed that I/R significantly increased apoptosis, TNF-α, and DNA damage in rats with brain I/R. However, 10 mg/kg intraperitoneal DiOHF treatment improved deterioted parameters.

Background: Serum thymus and activation-regulated chemokine (TARC) and periostin are reliable biomarkers in eosinophilic asthma. Objective: This study was carried out to determine the use of periostin and TARC as biomarkers in asthma and to compare the superiority of one over the other, especially in asthma with an eosinophilic phenotype. Methods: The study was conducted with 87 patients with asthma and 42 healthy control subjects. Patients with asthma were also divided into eosinophilic and non-eosinophilic phenotypes. A pulmonary function test was performed in all the participants, and serum and induced sputum TARC, periostin concentrations, eosinophils, and total immunoglobulin E values were examined. Results: TARC and periostin levels were significantly higher in the asthma group than in the control group (p < 0.001). The serum TARC level in the eosinophilic group was significantly higher than in the non-eosinophilic and control groups (p < 0.001). The induced sputum TARC level was significantly higher in the non-eosinophilic group than in the control group (p < 0.001). The TARC and periostin levels of the patients were evaluated by using receiver operator characteristic analysis. The cutoff value for TARC was determined to be 1415.39 ng/L; likewise, the cutoff value for periostin was 107.60 ng/L. The present study detected that serum levels of TARC correlated to serum levels of periostin (r = 0.54; p = 0.032). Furthermore, when evaluating correlations between serum and sputum levels, there was a correlation detected between TARC and periostin in serum, whereas this correlation was stronger in sputum: r = 0.66, p = 0.020; and r = 0.62, p = 0.028, respectively. Conclusion: Serum and sputum TARC and periostin may contribute for monitoring the improvement of patients, particularly those with asthma. Furthermore, TARC was a more reliable biomarker than periostin for patients with eosinophilic asthma.

In recent years, free fatty acid binding proteins (FABPs) are implicated in spermatogenesis and sperm morphology. FABPs are members of the intracellular lipid-binding protein family; they exhibit tissue specific expression like the FABP9/PERF15 (Perforated15) male germ cell-specific fatty acid linkage-protein.The aim of the study was to assess the levels of seminal FABP-9 in normozoospermic and oligozoospermic men, and the possible relations between seminal FABP-9 levels and semen parameters.Research was carried out on 60 male volunteers who were admitted to Selcuk University Faculty of Medicine of Andrology Laboratory. Normozoospermic individuals (n = 30) were identified as Group 1, and Oligozoospermic individuals (n = 30) were identified as Group 2. The semen samples were collected in sterile plastic containers. Sperm parameters were assessed according to Kruger's criteria. Seminal plasma FABP-9 levels were analyzed by ELISA method. Outcomes were statistically evaluated at 0.05 significance level with SPSS (22.0). The Receiver Operating Characteristic (ROC) curve was used to evaluate the performance of FABP-9 levels as compared to that of the concentration and motility data of the sperm. FABP-9 levels were significantly higher in normozoospermic individuals (3.41 ± 1.64 ng/mL) than in oligozoospermic individuals (1.99 ± 0.78 ng/mL). There were significant correlations between FABP-9 levels and sperm concentration, total sperm count, motility, progressive motility, immobility, Total Progressive Motil Sperm Count (TPMSC), head anomaly, and teratozoospermia index.We suggest that FABP-9 level is an important biomarker, and low levels of semen FABP-9 may impact the fertility status based on the ROC findings.

Background: Naphthoquinones have protective effects through different mechanism against to human malignancies including pancreatic cancer. One of these mechanisms is to avoid reactive oxygen species (ROS) production. Changes in enzymatic (superoxide dismutases, catalase [CAT], ascorbate peroxidase, glutathione peroxidase, and glutathione reductase) antioxidant systems, such as formation of an oxidative biomarker (H2O2, malondialdehyde, ischemic modified albümin, etc.) have a critical role in ROS mechanism. According to this knowledge, we evaluated the anticancer and antioxidant activity of the juglone. Objectives: The aim of this study was to investigate the cytotoxic activity of juglone and to investigate its effect on antioxidant activity in BxPC-3 and PANC-1 pancreatic cancer cell lines. Materials and Methods: The cytotoxic activities of juglone on cell viability were investigated on pancreatic cancer cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell viability test. One of the antioxidant enzymes, CAT activities and antioxidant status marker reduced glutathione (GSH) levels were measured by spectrophotometric analysis. We compared the groups as juglone treatment and control groups (no treatment) at different hours (24th, 48th and 72th h). Results:Juglone reduced the cell viability of human pancreatic cancer cells in a concentration-dependent manner. Juglone supplementation showed an antiproliferative effect toward pancreatic cancer cell lines at IC50 values. The IC50 of juglone on BxPC-3 and PANC-1 pancreatic cell line was 21.05 μM and 21.25 μM, respectively. Furthermore, juglone had a significantly higher degree of enzymatic activity to compensate the oxidative stress. CAT activity was found a significant increase compared to the control group at 24, 48, and 72 h in PANC-1 cells; it was found a significant increase at 72 h in BxPC-3 cell line. Reduced glutathione level is decreased at 24 and 48 h while at 72 h GSH level is increased in BxPC-3 cell line after juglone treatment compared to the control group. In PANC-1 cell line, GSH level is increased at 24 and 48 h, but it was decreased at 72 h compared to the control group. Conclusions: Our results indicate that juglone can be a potent anticancer molecule and may prove essential in pancreatic cancer therapy. Juglone may play a central role in antioxidant system defense in pancreatic cells.

This study aimed to determine the effect of 3',4'-Dihydroxyflavonol (DiOHF) on lipid peroxidation, DNA damage and inflammation in ovarian ischaemia (I)-reperfusion (R) injury. This study was performed on 44 Wistar-albino female rats. Groups were designed as Control; Sham; I/R (the left ovary was ligated for 2 h and then reperfused for 2 h); I/R + DiOHF (after 2 h ischaemia and 2 h reperfusion, 30 mg/kg of DiOHF was given intraperitoneally and reperfusion was allowed for 2 h more); I + DiOHF + R (after 2 h I, 30 mg/kg of DiOHF was given at the beginning of 2 h reperfusion); DiOHF + I/R (2 h after DiOHF administration, the left ovary was ligated for 2 h and then reperfused for 2 h). Blood and ovarian tissue samples were analysed for GSH, MDA, 8-OHdG, SOD, and IL-6. Ovarian tissue was examined histopathologically. Ovarian I/R has led to inflammation and oxidative damage. However, DiOHF activated the antioxidant system and prevented DNA damage induced by I/R in ovarian tissue. Vascularisation, oedema, and inflammation also occurred in ovarian tissue in I/R group. The results of this study indicated that I/R led to disturbance of the oxidant/antioxidant system balance and increased DNA damage; however, DiOHF supplementation prevented DNA damage, lipid peroxidation and inflammation by increasing the antioxidant system in ovarian I/R injury in rats. However, in potential I/R situations, DiOHF application appears to be beneficial in reducing inflammation, oxidant injury, and DNA damage, and in activating the antioxidant system. Increased DNA injury and inflammatory response (IL-6) and structural impairment were treated by administration of intraperitoneal (DiOHF) which strongly stimulated the antioxidant system, inhibited antioxidant activities, prevented DNA damage and inflammation process.What are the implications of these findings for clinical practice and/or further research? This study's strength is that it is the first research demonstrates the prevention of DNA damage in ovarian I/R by DiOHF supplementation. This flavonoid (DiOHF) may be used for treatment in different ovarian ischaemia/reperfusion.

The study aimed to investigate the role of magnesium sulfate prophylaxis in nephrotoxicity caused by colistin. Thirty Wistar Albino rats were divided into four groups: control, colistin, magnesium (Mg), and Mg + colistin. The drugs were administered to the groups for seven days. Urea-creatinine values were measured at the beginning (T0) and end (T1) of the study. Malondialdehyde (MDA) levels were measured in plasma and kidney tissue, glutathione (GSH) levels were analyzed in the erythrocyte and kidney tissues. At the end of the study, the semiquantitative score (SQS) was calculated by the histopathological examination of the kidneys. Urea values significantly decreased in Mg and Mg + colistin groups compared to the baseline (p = 0.013 and p = 0.001). At the time of T1, these groups had significantly lower urea values than the colistin and control groups. Creatinine value was significantly increased in the colistin group compared to baseline (p = 0.005), the creatinine value in the colistin group was significantly higher than the Mg + colistin group (p = 0.011). Around 15% of productive couples in the world are infertile. Recent years, biochemical mechanisms leads to male infertility are started to research. Redox regulation and oxidative stress (OS) show importance in the pathogenesis of infertility in male. Malondialdehyde (MDA) and Glutathione (GSH) are biochemical indicatives of sperm damage by reactive oxygen species (ROS). In addition, sperm are coated with a thick glycocalyx rich in sialic acids. It is aimed to determine and evaluate the differences between normozoospermic and oligozoospermic individuals according to sialic acid, MDA and GSH concentrations and correlations between spermyogram and these parameters.This study was carried out on seminal plasma of individuals who admitted to Selcuk University Faculty of Medicine IVF Unit Andrology Laboratory. The groups were divided into two as normozoospermics (n=30, sperm concentration≥15million/mL), and oligozoospermics (n=30, sperm concentration15million/mL). Spermyogram were evaluated regarding WHO (2010) Kruger criteria. GSH, MDA and sialic acid concentrations were analyzed in seminal plasma. Diagnostic performance of sialic acid has been determined with ROC curve analysis.Sialic acid levels were significantly lower in Normozoospermic than Oligozoospermic individuals (p0.0001), MDA and GSH levels were not differ significantly in both groups (p0.05). Sialic acid correlated significantly with most of the spermyogram findings. When diagnostic performance of sialic acid was evaluated, the cut off value of sialic acid found as 4.175nmol/mL by ROC curve.High seminal plasma sialic acid levels may be used as a biomarker and sialic acid is important determinant in oligozoospermia.

The purpose of this study was to investigate the effects of zinc and melatonin administration on interleukin-6, lipid peroxidation parameters, and element metabolism in DMBA-induced breast cancer in female rats. A total of 42 recently weaned Wistar rats were divided into 5 groups as follows: control (group 1), DMBA control (group 2), DMBA + zinc (group 3), DMBA + melatonin (group 4), and DMBA + melatonin and zinc (group 5). Malondialdehyde (MDA) and glutathione (GSH) levels in breast tissue and blood samples were determined via spectrophotometric methods. In addition, iron, magnesium, zinc, and copper levels in serum samples were determined by atomic emission, and plasma interleukin-6 levels were determined by ELISA method. BackgroundA recently discovered hormone, irisin is accepted to be significantly involved in the regulation of body weight. Thyroid functions may be, directly or indirectly, associated with irisin.AimThe aim of the present study is to determine the effect of experimental thyroid dysfunction on irisin levels in rats.MethodsThe study registered 40 adult male Sprague-Dawley rats, which were allocated to groups as follows: 1. Control; 2. Hypothyroidism induced by injection of 10 mg/kg/day intraperitoneal propylthiouracil (PTU) for 3 weeks; 3. Hypothyroidism (PTU 2 weeks) + L-thyroxin (1.5 mg/kg/day for 1 week); 4. Hyperthyroidism induced in rats by 3-week thyroxin (0.3 mg/kg/day); 5. Hyperthyroidism + PTU. At the end of the study, blood samples were collected to quantify free triiodothyronine (FT3), free triiodothyronine (FT4) and irisin levels.ResultsFT3and FT4levels were reduced in hypothyroidism and were significantly elevated in hyperthyroidism (p < 0.001). Irisin values, on the other hand, were found to be elevated in both hypothyroidism and hyperthyroidism groups (p < 0.001).ConclusionThe results of the study suggest that irisin values increase in thyroid dysfunction, hypo- and hyperthyroidism, and that when hypothyroidism is corrected by thyroxin administration and hyperthyroidism by PTU injection, plasma irisin values go back to normal.

A molecule released primarily from the fat cells adiposities; adiponectin has important effects on the regulation of body weight.AimThe present study aimed to explore the effects of experimental thyroid dysfunction and its treatment on nesfatin-1 and adiponectin levels in rats.MethodsThe study included 40 adult male Sprague-Dawley rats which were grouped as follows: (1) control; (2) hypothyroidism [hypothyroidism was induced by intraperitoneal injection of 10 mg/kg/day propylthiouracil (PTU) for 3 weeks]; (3) hypothyroidism + thyroxine group [after hypothyroidism was induced by 2-week PTU injection, they were treated with high-dose L-thyroxine (1.5 mg/kg/day) for 1 week]; (4) hyperthyroidism [hyperthyroidism was induced by 3-weeks’ thyroxine injection (0.3 mg/kg/day)]; (5) hyperthyroidism + PTU (after hyperthyroidism was induced by 2-weeks’ thyroxine injection, the animals were given 10 mg/kg/day PTU for 1 week). Ankaferd blood stopper (ABS) is a medical product that is used in several injuries, dental operations, prevention of minor or major bleeding after spontaneous or surgerical operations and have anti-microbial, anti-inflamatory, anti-thrombin, anti-platelet, anti-atherosclerotic, anti-oxidants effects. The present study is aimed to evaluate the effects of ABS on 8-hydroxy-2?-deoxyguanosine (8-OHdG), superoxide dismutase (SOD), myeloperoxidase (MPO) levels over pleural adhesions in rabbits with pulmonary parenchymal damage.16 New Zelland species rabbits were divided in two groups such as control (n7) and study group (n7). One rabbit in each group died during anesthesia. In both groups, we performed wedge resections in equal size to the left lower lobes of all rabbits. No interventions were made on control group, whereas 5 puff’s (1 cc) ABS was performed to the resection area at study group. Tube thoracostomy that performed both groups were terminated postoperatively at 6th hour after drainage and air leakages follow up. Rabbits were sacrificed with anesthetics at postoperative 8th day. Lung tissues were collected for analyzing of 8-OHdG, SOD, MPO. The 8-OHdG levels were respectively 2.010.39 ng/ml in control group and, 0.380.12 ng/ml in study. The differences between study and control group were statistically important group (p0.001). SOD and MPO levels did not show any statistically importance in the groups. As a conclusion, we can say that oxidative DNA damage prevented by ABS.

Amaç: Sunulan çalışmanın amacı, hemodiyaliz ve sağlıklı kişilerde serum kadmiyum (Cd), total triiyodotironin (T3), total tiroksin (T4) ve adrenokor- tikotropik hormon (ACTH) düzeyleri arasında istatistiksel olarak farklılık ve de gruplar arasında bu eser element ile tiroit profilleri ve ACTH arasında korelasyon olup olmadığını değerlendirmekti. Gereç ve Yöntem: Çalışmaya 2-16 yıldır haftada en az üç kez diyalize giren, her seansı en az 4 saat olan 47 hemodiyaliz hastası (hemodiyaliz grubu) ve 23 sağlıklı kişi (kontrol grubu) dahil edildi. Hemodiyaliz grubunda kan numuneleri diyaliz öncesi ve sonrası alındı. Kontrol grubunun kan numuneleri 10 saatlik açlık sonrası alındı. Bul- gular: Total T3 (p0.001) düzeyleri kontrol grubunda diyaliz öncesi ve diyaliz sonrasına göre daha yüksekti. Total T3 (p0.05) düzeyleri diyaliz öncesi diya- liz sonrasına göre daha düşüktü. Kontrol grubunda Total T4 düzeyleri yüksek, Cd düzeyleri ise düşük tespit edildi. Bu parametreler, diyaliz öncesine göre ise, istatistiksel açıdan önemli farklılık (sırasıyla p0.001, p0.01) gösterdi. ACTH düzeyleri bakımından gruplar arasında önemli bir farklılık bulunmadı. Tartışma: Hemodiyaliz hastalarında Cd düzeylerinin ölçülmesinin önemli ola- bileceği kanısındayız. Ancak Cd, tiroit profilleri ve ACTH düzeyleri arasında önemli bir ilişki bulamadık., Aim: The aim of the present study is to determine whether there are statisti- cally differences between hemodialysis patients and control (healthy) groups according to level of serum cadmium (Cd), total triiodothyronine (T3) and total thyroxine (T4) and adrenocorticotropin hormone (ACTH), and besides whether there are any correlation coefficients of trace element with thyroid profiles, ACTH in these groups. Material and Methods:The study included 47 hemodialysis patient who were dialyzed three times a week and each session was at least 4 hours. The duration of dialysis range were 2-16 yr. This gro- up called as a Hemodialyis group. Blood samples were taken before (pre- hemodialysis) and after (post-hemodialysis) hemodialysis session. Control group included 23 healthy volunteers with no medical problem. In control group, samples were taken after 10 hour fasting. Results: Levels of total T3 were higher in group of control than the levels of pre-hemodialysis and post-hemodialysis (p≤0.001). Total T3 levels were lower in pre-hemodialysis (p≤0.05) than those of post-hemodialysis. The highest level of total T4 and lowest level of Cd were in control group and these parameters were statisti- cally different from the pre-hemodialysis (respectively p≤0.001, p≤0.01). We did not determine any significant differences between all groups according to data of ACTH levels. Discussion: We suggest that analyzing the levels of Cd may be useful for the hemodialysis patients. But, we did not determine a significant association between Cd and thyroid profiles and ACTH levels.
