1. The Politeness Strategies of Thai Undergraduates in an Instant Messaging Application

Abstract

The aim of this study is twofold: to analyze politeness strategies in the online conversations of Thai students, and to suggest how this analysis can be applicable to pedagogical practice. A corpus of a 21-month instant online conversation among students and teachers has been analyzed. Throughout the time of data collection, the teachers continually encouraged students to use English and French to promote the use of foreign languages that they were learning. The result of statistical analysis showed the relation between speech acts and politeness strategies used in the data. It could be claimed that the students' language proficiency governed their politeness strategies. As a result, the authors proposed a modification of Brown & Levinson's (1978) Weightiness formula for non-native speakers of English as: W[subscript x] = LP(S) × [D(S,H) + P(H,S) + R[subscript x]]. The results also showed that emoticons were used as redressive actions in politeness. Further, interlanguage pragmatics in the data were discussed based on linguistic competency and socio-cultural norms in the participants' L1. The results suggest that teachers and curriculum developers could better understand students' communication behaviors and language competency through computer-mediated communication. Finally, we offer suggestions to promote online communication in the context of active learning.

Abstract

Cilia defects lead to scoliosis in zebrafish, but the underlying pathogenic mechanisms are poorly understood and may diverge depending on the mutated gene. Here, we dissected the mechanisms of scoliosis onset in a zebrafish mutant for the rpgrip1l gene encoding a ciliary transition zone protein. rpgrip1l mutant fish developed scoliosis with near-total penetrance but asynchronous onset in juveniles. Taking advantage of this asynchrony, we found that curvature onset was preceded by ventricle dilations and was concomitant to the perturbation of Reissner fiber polymerization and to the loss of multiciliated tufts around the subcommissural organ. Rescue experiments showed that Rpgrip1l was exclusively required in foxj1a-expressing cells to prevent axis curvature. Genetic interactions investigations ruled out Urp1/2 levels as a main driver of scoliosis in rpgrip1 mutants. Transcriptomic and proteomic studies identified neuroinflammation associated with increased Annexin levels as a potential mechanism of scoliosis development in rpgrip1l juveniles. Investigating the cell types associated with annexin2 over-expression, we uncovered astrogliosis, arising in glial cells surrounding the diencephalic and rhombencephalic ventricles just before scoliosis onset and increasing with time in severity. Anti-inflammatory drug treatment reduced scoliosis penetrance and severity and this correlated with reduced astrogliosis and macrophage/microglia enrichment around the diencephalic ventricle. Mutation of the cep290 gene encoding another transition zone protein also associated astrogliosis with scoliosis. Thus, we propose astrogliosis induced by perturbed ventricular homeostasis and associated with immune cell activation as a novel pathogenic mechanism of zebrafish scoliosis caused by cilia dysfunction.

Abstract

Abstract Xenopus provides a simple and efficient model system to study nephrogenesis and explore the mechanisms causing renal developmental defects in human. Hnf1b (hepatocyte nuclear factor 1 homeobox b), a gene whose mutations are the most commonly identified genetic cause of developmental kidney disease, is required for the acquisition of a proximo-intermediate nephron segment in Xenopus as well as in mouse. Genetic networks involved in Hnf1b expression during kidney development remain poorly understood. We decided to explore the transcriptional regulation of Hnf1b in the developing Xenopus pronephros and mammalian renal cells. Using phylogenetic footprinting, we identified an evolutionary conserved sequence (CNS1) located several kilobases (kb) upstream the Hnf1b transcription start and harboring epigenomic marks characteristics of a distal enhancer in embryonic and adult renal cells in mammals. By means of functional expression assays in Xenopus and mammalian renal cell lines we showed that CNS1 displays enhancer activity in renal tissue. Using CRISPR/cas9 editing in Xenopus tropicalis, we demonstrated the in vivo functional relevance of CNS1 in driving hnf1b expression in the pronephros. We further showed the importance of Pax8-CNS1 interaction for CNS1 enhancer activity allowing us to conclude that Hnf1b is a direct target of Pax8. Our work identified for the first time a Hnf1b renal specific enhancer and may open important perspectives into the diagnosis for congenital kidney anomalies in human, as well as modeling HNF1B-related diseases.

Abstract

BackgroundThe success of the CRISPR/Cas9 genome editing technique depends on the choice of the guide RNA sequence, which is facilitated by various websites. Despite the importance and popularity of these algorithms, it is unclear to which extent their predictions are in agreement with actual measurements.ResultsWe conduct the first independent evaluation of CRISPR/Cas9 predictions. To this end, we collect data from eight SpCas9 off-target studies and compare them with the sites predicted by popular algorithms. We identify problems in one implementation but found that sequence-based off-target predictions are very reliable, identifying most off-targets with mutation rates superior to 0.1 %, while the number of false positives can be largely reduced with a cutoff on the off-target score. We also evaluate on-target efficiency prediction algorithms against available datasets. The correlation between the predictions and the guide activity varied considerably, especially for zebrafish. Together with novel data from our labs, we find that the optimal on-target efficiency prediction model strongly depends on whether the guide RNA is expressed from a U6 promoter or transcribed in vitro. We further demonstrate that the best predictions can significantly reduce the time spent on guide screening.ConclusionsTo make these guidelines easily accessible to anyone planning a CRISPR genome editing experiment, we built a new website ( http://crispor.org ) that predicts off-targets and helps select and clone efficient guide sequences for more than 120 genomes using different Cas9 proteins and the eight efficiency scoring systems evaluated here.

Abstract

LEAFY plant-specific transcription factors, which are key regulators of flower meristem identity and floral patterning, also contribute to meristem activity. Notably, in some legumes, LFY orthologs such as Medicago truncatula SINGLE LEAFLET (SGL1) are essential in maintaining an undifferentiated and proliferating fate required for leaflet formation. This function contrasts with most other species, in which leaf dissection depends on the reactivation of KNOTTED-like class I homeobox genes (KNOXI). KNOXI and SGL1 genes appear to induce leaf complexity through conserved downstream genes such as the meristematic and boundary CUP-SHAPED COTYLEDON genes. Here, we compare in M. truncatula the function of SGL1 with that of the Arabidopsis thaliana KNOXI gene, SHOOT MERISTEMLESS (AtSTM). Our data show that AtSTM can substitute for SGL1 to form complex leaves when ectopically expressed in M. truncatula. The shared function between AtSTM and SGL1 extended to the major contribution of SGL1 during floral development as ectopic AtSTM expression could promote floral organ identity gene expression in sgl1 flowers and restore sepal shape and petal formation. Together, our work reveals a function for AtSTM in floral organ identity and a higher level of interchangeability between meristematic and floral identity functions for the AtSTM and SGL1 transcription factors than previously thought.

Abstract

Based on a new approach to the detection of radio transients associated with extensive air showers induced by ultra high energy cosmic rays, the experimental apparatus CODALEMA is in operation, measuring about 1 event per day corresponding to an energy threshold ~ 5. 10^16 eV. Its performance makes possible for the first time the study of radio-signal features on an event-by-event basis. The sampling of the magnitude of the electric field along a 600 meters axis is analyzed. It shows that the electric field lateral spread is around 250 m (FWHM). Abstract

We have measured the gamma-ray fluxes of the blazars Mrk 421 and Mrk 501 in the energy range between 50 and 350 GeV (1.2 to 8.3 x 10^25 Hz). The detector, called CELESTE, used first 40, then 53 heliostats of the former solar facility "Themis" in the French Pyrenees to collect Cherenkov light generated in atmospheric particle cascades. The signal from Mrk 421 is often strong. We compare its flux with previously published multi-wavelength studies and infer that we are straddling the high energy peak of the spectral energy distribution. The signal from Mrk 501 in 2000 was weak (3.4 sigma). We obtain an upper limit on the flux from 1ES 1426+428 of less than half that of the Crab flux near 100 GeV. Taking advantage of recent technical progress which has overcome some of the difficulties encountered in the 1960's in the radio detection of extensive air showers induced by ultra high energy cosmic rays (UHECR), a new experimental apparatus (CODALEMA) has been built and operated. We will present the characteristics of this device and the analysis techniques that have been developed for observing electrical transients associated with cosmic rays. We find a collection of events for which both time and arrival direction coincidences between particle and radio signals are observed. The counting rate corresponds to shower energies $\geq 5\times 10^{16}$ eV. Firm evidence for a radio emission counterpart of cosmic ray air showers is presented. By the use of an antenna array set up in coincidence with ground particle detectors, we find a collection of events for which both time and arrival direction coincidences between particle and radio signals are observed. Abstract

We present the characteristics and performance of a demonstration experiment devoted to the observation of ultra high- energy cosmic ray extensive air showers using a radiodetection technique. In a first step, one antenna narrowed band filtered acting as trigger, with a 4 $\sigma$ threshold above sky background-level, was used to tag any radio transient in coincidence on the antenna array. Recently, the addition of 4 particle detectors has allowed us to observe cosmic ray events in coincidence with antennas.

Abstract

We have converted the former solar electrical plant THEMIS (French Pyrenees) into an atmospheric Cherenkov detector called CELESTE, which records gamma rays above 30 GeV (7E24 Hz). Here we present the first sub-100 GeV detection by a ground based telescope of a gamma ray source, the Crab nebula, in the energy region between satellite measurements and imaging atmospheric Cherenkov telescopes. At our analysis threshold energy of 60 +/- 20 GeV we measure a gamma ray rate of 6.1 +/- 0.8 per minute. Allowing for 30% systematic uncertainties and a 30% error on the energy scale yields an integral gamma ray flux of I(E>60 GeV) = 6.2^{+5.3}_{-2.3} E-6 photons m^-2 s^-1. The analysis methods used to obtain the gamma ray signal from the raw data are detailed. In addition, we determine the upper limit for pulsed emission to be <12% of the Crab flux at the 99% confidence level, in the same energy range. Our result indicates that if the power law observed by EGRET is attenuated by a cutoff of form e^{-E/E_0} then E_0 < 26 GeV. This is the lowest energy probed by a Cherenkov detector and leaves only a narrow range unexplored beyond the energy range studied by EGRET., Comment: 34 pages, accepted by the Astrophysical Journal

The mean IOP was reduced by 27.4%, from 22.2 mmHg to 16.1 mmHg. In subgroup analyses, the mean IOP was significantly reduced from baseline, irrespective of whether previous treatment was monotherapy or combination therapy, and preserved or PF therapy. Physicians mostly (88.1%) reported the IOP-lowering efficacy of PF FC bimatoprost 0.03%/timolol 0.5% to be as expected or better than expected. Switching to PF FC bimatoprost 0.03%/timolol 0.5% resulted in reductions from baseline in the number of patients reporting ocular symptoms. Adverse events were reported by 6.2% of patients, the most common being eye irritation (1.6%) and eye pruritus (1.0%). Physicians reported treatment compliance as better or unchanged compared with prior treatment in almost all patients (93.9%). Most patients were expected to continue PF FC bimatoprost 0.03%/timolol 0.5% after the end of the study.Conclusion: Switching to PF FC bimatoprost 0.03%/timolol 0.5% was associated with significant IOP reductions from baseline over 12 weeks. Adverse events were uncommon, and compliance was high compared with previous therapy. PF FC bimatoprost 0.03%/timolol 0.5% may be a suitable treatment for patients with inadequately controlled IOP or who are sensitive to preservatives. This multicenter, prospective, open-label, observational study was designed to investigate the efficacy and tolerability of bimatoprost 0.01% in routine clinical practice.Methods: Data were collected from 10,337 patients with primary open-angle glaucoma or ocular hypertension attending 1334 centers in Germany. The primary efficacy outcome was mean change in IOP in each eye from baseline to 10–14 weeks after initiation of bimatoprost 0.01%. Target IOP, prior therapies, additional treatments, and adverse events were also assessed. All treatment decisions were at the physicians’ discretion.Results: Bimatoprost 0.01% significantly lowered mean IOP from baseline by –4.1 mmHg (P < 0.0001) in all patients after a mean of 10.45 weeks. In patients without previous treatment, bimatoprost 0.01% reduced mean IOP from baseline by –6.5 mmHg (P < 0.0001). Bimatoprost 0.01% also significantly reduced IOP in patients previously treated with monotherapy of β-blockers, prostaglandin analogs, carbonic anhydrase inhibitors or bimatoprost 0.03%. No adverse events were reported by 93.9% of patients during treatment with bimatoprost 0.01%; the most commonly reported adverse events were eye irritation (2.0%), ocular hyperemia (1.4%), and conjunctival hyperemia (1.2%). Physicians and patients rated tolerability and adherence as high, and most patients said they would continue with bimatoprost 0.01% treatment.Conclusion: Bimatoprost 0.01% can produce additional IOP-lowering effects when used in routine clinical practice in patients who have received prior therapy, in addition to lowering IOP in previously untreated patients. A high rate of continuation of therapy with bimatoprost 0.01% was observed in patients who switched from a variety of different medications. Abstract

Rhizobia and legumes are able to interact in a symbiotic way leading to the development of root nodules. Within nodules, rhizobia fix nitrogen for the benefit of the plant. These interactions are efficient because spectacularly high densities of nitrogen fixing rhizobia are maintained in the plant cells. DNF2, a Medicago truncatula gene has been described as required for nitrogen fixation, bacteroid's persistence and to prevent defense-like reactions in the nodules. This manuscript shows that a Rhizobium mutant unable to differentiate is not sufficient to trigger defense-like reactions in this organ. Furthermore, we show that the requirement of DNF2 for effective symbiosis can be overcome by permissive growth conditions. The dnf2 knockout mutants grown in vitro on agarose or Phytagel as gelling agents are able to produce nodules fixing nitrogen with the same efficiency as the wild-type. However, when agarose medium is supplemented with the plant defense elicitor ulvan, the dnf2 mutant recovers the fix- phenotype. Together, our data show that plant growth conditions impact the gene requirement for symbiotic nitrogen fixation and suggest that they influence the symbiotic suppression of defense reactions in nodules.

Abstract

Three months of data taken by the Engineering Array Surface Detector of the Pierre Auger Observatory have been analyzed with the aim of setting a procedure for the study of showers at E_0 ~ 10^18 eV. We present the results, concerning: (i) the selection procedure and the check of its efficiency; (ii) the event rate and its stability; (iii) the characteristics of such events (i.e., angular direction, energy estimator S(1000), rise time vs core distance, correlation with pressure) showing their consistency with the expectations. Abstract

The Pierre Auger Southern Observatory in Argentina has begun taking data as it is being developed up to a final enclosed area of 3000 square kilometres. A key aspect of the project is to provide information on the origin of the highest energy cosmic rays through understanding the arrival directions of those particles. To avoid claims of a spurious anisotropy detection because trials have not been properly accounted, the Auger Collaboration has agreed 'a priori' to the analysis prescription presented here. It specifies the following: 1. The accumulation time for the (future) data set to be analyzed. 2. The anisotropy 'targets', each assigned a chance probability level. 3. The analysis procedure for each trial. A positive result will be claimed for any target search only if its chance probability is less than its assigned level. The levels are chosen so that the total chance probability for one or more positive results is 0.001. Exploratory searches beyond this prescription will be encouraged, but the Auger Collaboration will not assign any confidence level to anisotropies that may be discovered that way. Any such discovery would identify a good target for a prescription to be used with a subsequent Auger data set., 4 pages

Abstract

The absolute calibration of an air fluorescence detector (FD) is an important element in correctly determining the energy of detected cosmic rays. The absolute calibration relates the flux of photons of a given wavelength at the detector aperture to the electronic signal recorded by the FD data acquisition system. For the Auger FDs, the primary absolute calibration method uses a diffusive surface which is placed in front of a telescope aperture to uniformly illuminate the telescope field of view with a known light signal. This single-wavelength measurement (375 nm) will be made at intervals of several months until the stability of the telescopes is determined. The relative wavelength dependence of the calibration is determined through independent measurements. The error in absolute calibration at a single wavelength is estimated to be less than 10%. Two other absolute calibration methods are used to provide an independent verification of the primary measurement. The stability of the calibration with time is monitored nightly by a relative calibration system. In this paper we will provide descriptions of the absolute and relative calibration methods used by the Auger air fluorescence observatory. Abstract

The Pierre Auger Observatory is a``hybrid'' UHECR detector. The surface detector (SD) and air fluorescence detector (FD) of the observatory are designed for observation of cosmic ray showers in coincidence, with a 10% duty cycle. The resulting data are expected to be superior in quality to those of either the SD or FD operating individually. Hybrid operation, triggering, data acquisition, and event reconstruction were successfully demonstrated during the prototype phase of the project. This paper focuses on 75 hybrid events recorded during a four month period of running with the prototype detectors in late 2001 and early 2002. The geometric technique for reconstruction of the hybrid events is described and its advantages over the traditional FD-only method are demonstrated.
