The integration of Knowledge Management (KM) in various fields, including academia and business, has been fostered by the promotion of Information and Communication Technologies (ICTs). The aim of this study was, on the one hand, to understand the key processes of KM in students of the Faculty of Education and the School of Engineering in a Spanish university, and, on the other hand, to evaluate the extent to which ICTs serve as support to these processes. To this end, the designed and validated MAINGC questionnaire was administered to 200 students who entered university in the 2018-2019 academic year. Fourteen ICT tools and four dimensions of KM were analyzed: information management, transformation of information into knowledge, management of the resulting learning, and ICT tools for KM support. The results showed differentiated patterns of technology use and perceptions of KM according to sex, age and degree. Indeed, education degree students reported higher perceptions of competence in KM processes compared to engineering students. Participants' responses regarding ICT tools suggested that the use of these tools has not yet been fully integrated into KM processes and revealed that both groups of students showed different patterns of use within the academic environment.

The problem of maximizing the range of a propeller-driven aircraft in a level flight cruise is analyzed within the framework of optimal control. The specific fuel consumption and propeller efficiency of its propulsive system are characterized by functions of the velocity and engine power (full model), in contrast to previous works, where they were considered to be constant. To conduct the study, a notional Piper Cherokee PA-28 is selected as representative of light aircraft, defining both the airplane and mission features. Two simplified models are also derived: the Von Mises model, with constant specific fuel consumption and propeller efficiency, and the Parget and Ardema model, defined by constant specific fuel consumption and propeller efficiency depending on the velocity. The problem is solved numerically by means of a direct transcription method. Since the optimal problems of the Von Mises and Parget and Ardema models are singular, it is necessary to incorporate a regularization term. Such a numerical algorithm is validated against the analytical solution given by the Breguet formulation. In this context, the velocity and mass (state variables), the power throttle (control), and the best range are determined. The full model provides a maximum range of 1492 km. The differences between the Von Mises and Parget and Ardema models are about 24 km and 1 km, respectively. A non-optimal steady cruise is also analyzed, providing a significant reduction in the flight time, with a decrease of about 2% of the range. The evolution of the state variables and control in the steady cruise, however, separates from the full model. On the other hand, the Parget and Ardema model almost reproduces the full model results, leading to a clear image of the physics involved: the best range comes from maximizing the product of the propeller and aerodynamic efficiencies with respect to the velocity, which determines the optimal arc.

Despite over two decades of intense research into bioelectrochemical systems (BESs), their practical implementation remains unrealized, partly due to the low performance of bioelectrodes. With the introduction of additive manufacturing techniques, the development of a new generation of bioelectrodes with custom-shaped geometries using conductive composites has become feasible. This study examines the potential of using two conductive composites, Poly-lactic acid (PLA) and thermoplastic polyurethane (TPU), for 3D-printed electrodes. Electrochemical characterization reveals that TPU has a charge transfer resistance approximately two orders of magnitude higher than PLA, rendering it unsuitable for bioelectrodes. The presence of triangular patterns enhances the performance of planar electrodes, with optimal results observed for PLA-based electrodes with surface pattern depths between 0.6 and 1.4 mm. Additionally, electrodeposition (ED) of graphene oxide (GO) further improves performance across all cases. During the subsequent biotic start-up, patterned PLA electrodes with a depth of 1.4 mm exhibit higher current. However, these 3D-printed electrodes exhibit degradation after 56 days of operation.

Spinal cord stimulation (SCS) treatment for chronic pain relies on the activation of primary sensory fibres ascending to the brain in the dorsal columns. While the efficacy of SCS has been demonstrated, the precise mechanism of action and nature of the fibres activated by stimulation remain largely unexplored. Our investigation in humans with chronic neuropathic pain undergoing SCS therapy, found that post-synaptic dorsal column (PSDC) fibres can be activated synaptically by the primary afferents recruited by stimulation, and axonically by the stimulation pulses directly. Synaptic activation occurred in 9 of the 14 patients analysed and depended on the vertebral level of stimulation. A clear difference in conduction velocities between the primary afferents and the PSDC fibres were observed. Identification of PSDC fibre activation in humans emphasises the need for further investigation into the role they play in pain relief and the sensory response sensation (paraesthesia) experienced by patients undergoing SCS.

IntroductionCoagulopathy associated with isolated traumatic brain injury (C-iTBI) is a frequent complication associated with poor outcomes, primarily due to its role in the development or progression of haemorrhagic brain lesions. The independent risk factors for its onset are age, severity of traumatic brain injury (TBI), volume of fluids administered during resuscitation, and pre-injury use of antithrombotic drugs. Although the pathophysiology of C-iTBI has not been fully elucidated, two distinct stages have been identified: an initial hypocoagulable phase that begins within the first 24 h, dominated by platelet dysfunction and hyperfibrinolysis, followed by a hypercoagulable state that generally starts 72 h after the trauma. The aim of this study was to design an acronym as a mnemonic device to provide clinicians with an auxiliary tool in the treatment of this complication.MethodsA narrative analysis was performed in which intensive care physicians were asked to list the key factors related to C-iTBI. The initial sample was comprised of 33 respondents. Respondents who were not physicians, not currently working in or with experience in coagulopathy were excluded. Interviews were conducted for a month until the sample was saturated. Each participant was asked a single question: Can you identify a factor associated with coagulopathy in patients with TBI? Factors identified by respondents were then submitted to a quality check based on published studies and proven evidence. Because all the factors identified had strong support in the literature, none was eliminated. An acronym was then developed to create the mnemonic device.Results and conclusionEleven factors were identified: cerebral computed tomography, oral anticoagulant & antiplatelet use, arterial blood pressure (Hypotension), goal-directed haemostatic therapy, use fluids cautiously, low calcium levels, anaemia-transfusion, temperature, international normalised ratio (INR), oral antithrombotic reversal, normal acid–base status, forming the acronym “Coagulation.” This acronym is a simple mnemonic device, easy to apply for anyone facing the challenge of treating patients of moderate or severe TBI on a daily basis.

Abstract Background The intersection between poverty and mental health is clear. Period poverty, understood as the lack of access to menstrual products, has been gaining attention especially among low and middle-income countries as an overlooked aspect of gendered poverty. Less is known about the incidence of period poverty in high-income countries and its association with mental health. The purpose of this study is to examine this association in a representative sample of young women living in an urban setting in southern Europe. Methods This is a cross-sectional study. Data were obtained from a representative survey of individuals aged 15 to 34 in the city of Barcelona (Spain), with a sample group of 647 young women. Subjects were selected through a systematic stratified random sampling method. A proportional quota sampling was used. The information was registered using CAPI data collection method. Period poverty was measured by a combination of three questions about the lack of access or misuse of menstrual products for economic reasons. The GHQ-12 was used to measure the risk of poor mental health. The analysis was carried out using multivariable logistic regression. This study explores the possibility of utilising electrochemically assisted anaerobic digestion supplemented with carbon-based materials to stimulate methanogenesis. Two different carbonaceous materials—commercial activated carbon (AC), and pyrolysed argan (PA, derived from argan shells)—were employed as supplements, with cheese whey (CW) being used as the substrate. Methane production slightly increased in the electrochemically assisted digesters, potentially translating into a 2–4% increase in the output of industrial digesters. In addition, reactors supplemented with PA also exhibited better production rates (496–508 L·kgVS−1), although there was no observed improvement in the quantity of biogas at the end of the biodegradability experiment. In contrast, when commercial AC was used as the supplement, the start-up phase was accelerated (5 days), although methane productivity decreased (273–352 L·kgVS−1). These observations were supported by microbiological analyses, demonstrating that the reactors with the poorest performance (those supplemented with AC) experienced the most significant decrease in both archaeal and bacterial populations.

To design architectural spaces that not only respond to the basic needs of users, but also seek their emotional well-being, it is necessary for the architecture students to have a special sensitivity and be aware of the different sensations that their designs should and can evoke. To achieve this competence without exploring real spaces, Immersive Virtual Reality technology offers an important contribution to the field of architecture. The purpose of this research is to determine if the sensations perceived in virtual architectural spaces by students are similar to the real ones and to determine the characteristics of this technology that allow a better perception of sensations. Six architectural modules were designed to be walked through and experienced at real scale using a Head Mounted Display by 22 students of the first and fifth year of studies of Architecture career in Peru. An ad-hoc questionnaire allowed to know the perceived sensations and the benefits of the tool. The results obtained showed that the perception of sensations of the fifth year students is a little closer to those expressed by a group of seven experts compared to that of the first year students and that the students consider the characteristics of accessibility, real scale of the space and the possibility of going through and looking at the space in all directions are those that have given more realism to the experience and therefore better perception of the space, while the characteristics of natural light and shadows, construction materials and external environment have been less valued in the realism of the experience. It is concluded that the sensory experimentation in architectural spaces modelled realistically in virtual environments allows the perception of sensations very similar to those that the architect seeks to convey initially.

IntroductionMajor trauma is one of the major health care problems facing modern society, trauma systems require careful planning to achieve an ideal level of coverage for the population. The Patient Blood Management Program is an integrated and global strategy to provide patient care that aims to assess and address, when possible, the etiology of blood abnormalities rather than transfuse without treating the underlying cause. We aimed to describe the factors that are associated with the clinical decision to transfuse polytraumatized patients admitted to the Intensive Care Unit (ICU).MethodWe performed a cross sectional multicenter study of patients admitted to ICUs for trauma in 14 Spanish hospitals from September 2020 to December 2021.ResultsA total of 69 patients were treated in the emergency room due to polytrauma, 46% of them were considered serious in the initial triage. Thirty were caused by a fall from considerable height (43.47%), followed by 39 patients admitted due to trac accidents (56.52%). The location of the trauma was mainly cranioencephalic, followed by thoracic trauma. Of the 69 patients, 25 received a blood transfusion during their ICU stay (36.23%).DiscussionNo significant differences were observed between transfused and non-transfused patients, except for the severity scales, where transfused patients have a higher score on all the scales assessed in the ICU except for the Revised Trauma Score. As we can see, the incidence of kidney failure was also different between the groups analyzed, reaching 44.00% in transfused patients and 13.64% in the group of patients without blood transfusion, p = 0.005. In this sense, 92.00% of the transfusions performed were inadequate according to the criteria of Hb in blood prior to the decision to transfuse (Hb < 9). Our data support the need to consider clinical practice guidelines regarding blood transfusion and its practices.

We report the discovery of COOL J0335−1927, a quasar at z = 3.27 lensed into three images with a maximum separation of 23.″3 by a galaxy cluster at z = 0.4178. To date, this is the highest redshift wide-separation lensed quasar known. In addition, COOL J0335−1927 shows several strong intervening absorbers visible in the spectra of all three quasar images with varying equivalent widths. The quasar also shows mini-broad line absorption. We construct a parametric strong gravitational lens model using ground-based imaging, constrained by the redshift and positions of the quasar images as well as the positions of three other multiply imaged background galaxies. Using our best-fit lens model, we calculate the predicted time delays between the three quasar images to be Δ t _AB = ${499}_{-146}^{+141}$ (stat) and Δ t _AC = $-{127}_{-17}^{+83}$ (stat) days. Folding in systematic uncertainties, the model-predicted time delays are within the ranges 240 < Δ t _AB < 700 and −300 < Δ t _AC < −30. We also present g -band photometry from archival Dark Energy Camera Legacy Survey and Pan-STARRS imaging, and new multi-epoch observations obtained between 2022 September 18 UT and 2023 February 22 UT, which demonstrate significant variability in the quasar and will eventually enable the measurement of the time delay between the three quasar images. The currently available light curves are consistent with the model-predicted time delays. This is the fifth paper from the COOL-LAMPS collaboration.

Microbial Electrolysis Cells are devices capable of converting the organic fraction present in the wastewaters into hydrogen. Integrating this relatively new technology into wastewater treatment plants can improve the energy balance and result in significant savings in greenhouse gases emissions. However, there are not many studies available in the scientific literature on the carbon footprint of these systems. This paper compares carbon footprint of a wastewater treatment plant located in South Spain, to the carbon footprint of this same plant in which the aerobic treatment is partially replaced by a Microbial Electrolysis Cell. The carbon footprint attributed to the construction of the plants was similar in both cases. However, the wastewater treatment plant with the Microbial Electrolysis Cell system would allow mitigating up to 2,700 t CO2-equivalents, which represents a 42% saving in greenhouse gas emissions compared to the existing wastewater treatment plants.

Abstract Background The low cost of 16S rRNA gene sequencing facilitates population-scale molecular epidemiological studies. Existing computational algorithms can resolve 16S rRNA gene sequences into high-resolution amplicon sequence variants (ASVs), which represent consistent labels comparable across studies. Assigning these ASVs to species-level taxonomy strengthens the ecological and/or clinical relevance of 16S rRNA gene-based microbiota studies and further facilitates data comparison across studies. Results To achieve this, we developed a broadly applicable method for constructing high-resolution training sets based on the phylogenic relationships among microbes found in a habitat of interest. When used with the naïve Bayesian Ribosomal Database Project (RDP) Classifier, this training set achieved species/supraspecies-level taxonomic assignment of 16S rRNA gene-derived ASVs. The key steps for generating such a training set are (1) constructing an accurate and comprehensive phylogenetic-based, habitat-specific database; (2) compiling multiple 16S rRNA gene sequences to represent the natural sequence variability of each taxon in the database; (3) trimming the training set to match the sequenced regions, if necessary; and (4) placing species sharing closely related sequences into a training-set-specific supraspecies taxonomic level to preserve subgenus-level resolution. As proof of principle, we developed a V1–V3 region training set for the bacterial microbiota of the human aerodigestive tract using the full-length 16S rRNA gene reference sequences compiled in our expanded Human Oral Microbiome Database ( eHOMD ). We also overcame technical limitations to successfully use Illumina sequences for the 16S rRNA gene V1–V3 region, the most informative segment for classifying bacteria native to the human aerodigestive tract. Finally, we generated a full-length eHOMD 16S rRNA gene training set, which we used in conjunction with an independent PacBio single molecule, real-time (SMRT)-sequenced sinonasal dataset to validate the representation of species in our training set. This also established the effectiveness of a full-length training set for assigning taxonomy of long-read 16S rRNA gene datasets. Conclusion Here, we present a systematic approach for constructing a phylogeny-based, high-resolution, habitat-specific training set that permits species/supraspecies-level taxonomic assignment to short- and long-read 16S rRNA gene-derived ASVs. This advancement enhances the ecological and/or clinical relevance of 16S rRNA gene-based microbiota studies. Video Abstract

ABSTRACT Dolosigranulum pigrum is positively associated with indicators of health in multiple epidemiological studies of human nasal microbiota. Knowledge of the basic biology of D. pigrum is a prerequisite for evaluating its potential for future therapeutic use; however, such data are very limited. To gain insight into D. pigrum’s chromosomal structure, pangenome, and genomic stability, we compared the genomes of 28 D. pigrum strains that were collected across 20 years. Phylogenomic analysis showed closely related strains circulating over this period and closure of 19 genomes revealed highly conserved chromosomal synteny. Gene clusters involved in the mobilome and in defense against mobile genetic elements (MGEs) were enriched in the accessory genome versus the core genome. A systematic analysis for MGEs identified the first candidate D. pigrum prophage and insertion sequence. A systematic analysis for genetic elements that limit the spread of MGEs, including restriction modification (RM), CRISPR-Cas, and deity-named defense systems, revealed strain-level diversity in host defense systems that localized to specific genomic sites, including one RM system hot spot. Analysis of CRISPR spacers pointed to a wealth of MGEs against which D. pigrum defends itself. These results reveal a role for horizontal gene transfer and mobile genetic elements in strain diversification while highlighting that in D. pigrum this occurs within the context of a highly stable chromosomal organization protected by a variety of defense mechanisms. IMPORTANCE Dolosigranulum pigrum is a candidate beneficial bacterium with potential for future therapeutic use. This is based on its positive associations with characteristics of health in multiple studies of human nasal microbiota across the span of human life. For example, high levels of D. pigrum nasal colonization in adults predicts the absence of Staphylococcus aureus nasal colonization. Also, D. pigrum nasal colonization in young children is associated with healthy control groups in studies of middle ear infections. Our analysis of 28 genomes revealed a remarkable stability of D. pigrum strains colonizing people in the United States across a 20-year span. We subsequently identified factors that can influence this stability, including genomic stability, phage predators, the role of MGEs in strain-level variation, and defenses against MGEs. Finally, these D. pigrum strains also lacked predicted virulence factors. Overall, these findings add additional support to the potential for D. pigrum as a therapeutic bacterium.

Intra-operative electrode placement for sacral neuromodulation (SNM) relies on visual observation of motor contractions alone, lacking complete information on neural activation from stimulation. This study aimed to determine whether electrophysiological responses can be recorded directly from the S3 sacral nerve during therapeutic SNM in patients with fecal incontinence, and to characterize such responses in order to better understand the mechanism of action (MOA) and whether stimulation is subject to changes in posture. Eleven patients undergoing SNM were prospectively recruited. A bespoke stimulating and recording system was connected (both intraoperatively and postoperatively) to externalized SNM leads, and electrophysiological responses to monopolar current sweeps on each electrode were recorded and analyzed. The nature and thresholds of muscle contractions (intraoperatively) and patient-reported stimulation perception were recorded. We identified both neural responses (evoked compound action potentials) as well as myoelectric responses (far-field potentials from muscle activation). We identified large myelinated fibers (conduction velocity: 36–60 m/s) in 5/11 patients, correlating with patient-reported stimulation perception, and smaller myelinated fibers (conduction velocity

La Cuenca de Cañadón Asfalto, ubicada entre el Macizo Nordpatagónico y la Cuenca del Golfo San Jorge, presenta una importante columna de sedimentos jurásicos y cretácicos acumulados en hemigrábenes pertenecientes a un sistema de rift formado durante la fragmentación de Gondwana. La Cuenca de Cañadón Asfalto incluye varios depocentros sedimentarios denominados Gastre, Gan Gan, Fossatti, Valle de General Racedo, Gorro Frigio y Paso de Indios, limitados por bloques basamentales en zonas de transferencia. La columna sedimentaria jurásica y cretácica puede dividirse en tres megasecuencias principales (J 1, J 2 y K) separadas entre sí por discordancias de significado regional que se apoyan sobre un basamento de metamorfitas y granitoides del Paleozoico. La Megasecuencia J 1 está constituida por la Formación Las Leoneras, la Formación Lonco Trapial y la Formación Cañadón Asfalto, y su edad se extiende entre el HettangianoSinemuriano?/Pliensbachiano y el Bajociano tardío. Se acumuló en ambientes fluvial, lacustre y volcánico durante los estadíos incipiente a juvenil del rift. La Megasecuencia J 2 incluye sedimentos aluviales y lacustres continentales correspondientes a la Formación Cañadón Calcáreo, de edad oxfordiana-kimmeridgiana, que se depositaron en depocentros parcialmente coincidentes con los anteriores durante una reactivación tectónica multiepisódica del rift. Durante el Tithoniano tardío – Hauteriviano existe un importante hiato estratigráfico. Durante este lapso se generaron esfuerzos transpresivos-transtensivos con rotación de bloques a escala regional, que originaron levantamientos de grandes áreas de la cuenca y la subsecuente erosión de zonas extensas, aunque el entorno tectónico y su correspondiente campo de esfuerzos se encuentran aún en discusión. El posterior colapso de estas áreas elevadas, con un control tafrogénico inicial y una posterior subsidencia termal, permitió el desarrollo del espacio disponible para la acumulación de la Megasecuencia K, formada por los niveles continentales del Grupo Chubut del Barremiano-Campaniano, al cual le siguieron los depósitos marinos y litorales de las formaciones Paso del Sapo y Lefipán del CampanianoDaniano, representativos de la primera transgresión atlántica en la cuenca. Como consecuencia de los procesos detallados más arriba, una marcada discordancia angular separa a la base de la Megasecuencia K de todos los depósitos más antiguos de la cuenca. Posteriormente, durante el Paleoceno tardío-Eoceno, se produjo un episodio tectónico compresivo en el margen occidental de la cuenca, seguido por la acumulación del Complejo Volcánico y Piroclástico del Río Chubut Medio en ambientes de calderas asociadas a un vulcanismo de retro-arco. En el Oligoceno?- Mioceno a Reciente se produjo el levantamiento y la inversión tectónica de la mayor parte de la Cuenca de Cañadón Asfalto, representado por fallamiento generalizado y por plegamiento localizado de intensidad y estilo variables. En este momento se desarrollaron pequeñas cuencas intermontanas limitadas por fallas.

The Bahía Blanca Estuary (38° 50' S, 62° 30' W) presents salt marshes where interactions between the local main plant and the dominant crab generate some very characteristics salt pans. These pans alter the normal water circulation and condition its trajectory generating an erosive process. The removed sediment is then exported to the main estuary through the creeks that evolve from those biological-physical interactions. To study it, a conceptual model is proposed, based on the laws deduced from observation of these phenomena in the field, and then verified with measurable data within macroscale time units. The objective of this article is to model how the interaction between the crab C. granulata and the plant S. perennis modifies the landscape of the salt marsh and influences the loss of sediment with a Cellular Automata Model. Originally developed to study the effect of the interaction plant - crab in the formation of tidal creeks, the model copies the basic laws that dominate the problem based on purely biological factors. Finally, the volume of sediment that is removed and how it varies in time are evaluated. The model results a very good tool to integrate a large quantity of data collected recently and to be able to extract conclusions on processes that have a very slow dynamics. Additionally, it could reproduce faithfully the salt marsh landscape product of the plants - crab interaction.

BackgroundThe effect of spinal cord stimulation (SCS) amplitude on the activation of dorsal column fibres has been widely studied through the recording of Evoked Compound Action Potentials (ECAPs), the sum of all action potentials elicited by an electrical stimulus applied to the fibres. ECAP amplitude grows linearly with stimulus current after a threshold, and a larger ECAP results in a stronger stimulus sensation for patients. This study investigates the effect of stimulus frequency on both the ECAP amplitude as well as the perceived stimulus sensation in patients undergoing SCS therapy for chronic back and/or leg pain.MethodsPatients suffering with chronic neuropathic lower-back and/or lower-limb pain undergoing an epidural SCS trial were recruited. Patients were implanted according to standard practice, having two 8-contact leads (8 mm inter-electrode spacing) which overlapped 2–4 contacts around the T9/T10 interspace. Both lead together thus spanning about three vertebral levels. Neurophysiological recordings were taken during the patient’s trial phase at two routine follow-ups using a custom external stimulator capable of recording ECAPs in real-time from all non-stimulating contacts. Stimulation was performed at various vertebral levels, varying the frequency (ranging from 2 to 455 Hz) while all other stimulating variables were kept constant. During the experiments subjects were asked to rate the stimulation-induced sensation (paraesthesia) on a scale from 0 to 10.ResultsFrequency response curves showed an inverse relationship between stimulation sensation strength and ECAP amplitude, with higher frequencies generating smaller ECAPs but stronger stimulation-induced paraesthesia (at constant stimulation amplitude). Both relationships followed logarithmic trends against stimulus frequency meaning that the effects on ECAP amplitude and sensation are larger for smaller frequencies.ConclusionThis work supports the hypothesis that SCS-induced paraesthesia is conveyed through both frequency coding and population coding, fitting known psychophysics of tactile sensory information processing. The inverse relationship between ECAP amplitude and sensation for increasing frequencies at fixed stimulus amplitude questions common assumptions of monotonic relationships between ECAP amplitude and sensation strength.

Introduction: A growing evidence suggests that immune dysregulation and thrombotic phenomena are key features in the pathophysiology of COVID-19. Apart from antivirals and respiratory support, anticoagulants, corticoids and immunomodulators are increasingly being prescribed, especially for more severe cases. We describe the clinical outcome of a large cohort of patients preferentially treated with glucocorticoids and interleukin inhibitors. Methods: Single center and retrospective case series. Adult patients admitted with COVID-19 related respiratory insufficiency were included. Patients who died within 2 days after admission and those testing positive but asymptomatic were excluded. We defined two study periods: from March 3rd to March 31 st, 2020 (beginning of epidemic until peak of incidence) and April 1 st to May 7 th, 2020 (second half of epidemic). The majority of patients received respiratory support, combinations of antimicrobials, anticoagulants, corticoids and interleukin inhibitors. Antivirals were preferentially given in the first period. The clinical outcome (death and ventilator dependency) of both periods was compared. Results: From March 3 rd to May 7 th, 685 patients were included for analysis (58.4% males, mean age 68.9 years). ABSTRACT Multiple epidemiological studies identify Dolosigranulum pigrum as a candidate beneficial bacterium based on its positive association with health, including negative associations with nasal/nasopharyngeal colonization by the pathogenic species Staphylococcus aureus and Streptococcus pneumoniae. Using a multipronged approach to gain new insights into D. pigrum function, we observed phenotypic interactions and predictions of genomic capacity that support the idea of a role for microbe-microbe interactions involving D. pigrum in shaping the composition of human nasal microbiota. We identified in vivo community-level and in vitro phenotypic cooperation by specific nasal Corynebacterium species. Also, D. pigrum inhibited S. aureus growth in vitro, whereas robust inhibition of S. pneumoniae required both D. pigrum and a nasal Corynebacterium together. D. pigrum l-lactic acid production was insufficient to account for these inhibitions. Genomic analysis of 11 strains revealed that D. pigrum has a small genome (average 1.86 Mb) and multiple predicted auxotrophies consistent with D. pigrum relying on its human host and on cocolonizing bacteria for key nutrients. Further, the accessory genome of D. pigrum harbored a diverse repertoire of biosynthetic gene clusters, some of which may have a role in microbe-microbe interactions. These new insights into D. pigrum’s functions advance the field from compositional analysis to genomic and phenotypic experimentation on a potentially beneficial bacterial resident of the human upper respiratory tract and lay the foundation for future animal and clinical experiments. IMPORTANCE Staphylococcus aureus and Streptococcus pneumoniae infections cause significant morbidity and mortality in humans. For both, nasal colonization is a risk factor for infection. Studies of nasal microbiota identify Dolosigranulum pigrum as a benign bacterium present when adults are free of S. aureus or when children are free of S. pneumoniae. Here, we validated these in vivo associations with functional assays. We found that D. pigrum inhibited S. aureus in vitro and, together with a specific nasal Corynebacterium species, also inhibited S. pneumoniae. Furthermore, genomic analysis of D. pigrum indicated that it must obtain key nutrients from other nasal bacteria or from humans. These phenotypic interactions support the idea of a role for microbe-microbe interactions in shaping the composition of human nasal microbiota and implicate D. pigrum as a mutualist of humans. These findings support the feasibility of future development of microbe-targeted interventions to reshape nasal microbiota composition to exclude S. aureus and/or S. pneumoniae.

Background In extant ecosystems, complex networks of ecological interactions between organisms can be readily studied. In contrast, understanding of such interactions in ecosystems of the geologic past is incomplete. Specifically, in past terrestrial ecosystems we know comparatively little about plant biotic interactions besides saprotrophy, herbivory, mycorrhizal associations, and oviposition. Due to taphonomic biases, epiphyte communities are particularly rare in the plant-fossil record, despite their prominence in modern ecosystems. Accordingly, little is known about how terrestrial epiphyte communities have changed across geologic time. Here, we describe a tiny in situ fossil epiphyte community that sheds light on plant-animal and plant-plant interactions more than 50 million years ago. Methods A single silicified Todea (Osmundaceae) rhizome from a new locality of the early Eocene (ca. 52 Ma) Tufolitas Laguna del Hunco (Patagonia, Argentina) was studied in serial thin sections using light microscopy. The community of organisms colonizing the tissues of the rhizome was characterized by identifying the organisms and mapping and quantifying their distribution. A 200 × 200 µm grid was superimposed onto the rhizome cross section, and the colonizers present at each node of the grid were tallied. Results Preserved in situ, this community offers a rare window onto aspects of ancient ecosystems usually lost to time and taphonomic processes. The community is surprisingly diverse and includes the first fossilized leafy liverworts in South America, also marking the only fossil record of leafy bryophyte epiphytes outside of amber deposits; as well as several types of fungal hyphae and spores; microsclerotia with possible affinities in several ascomycete families; and evidence for oribatid mites. Discussion The community associated with the Patagonian rhizome enriches our understanding of terrestrial epiphyte communities in the distant past and adds to a growing body of literature on osmundaceous rhizomes as important hosts for component communities in ancient ecosystems, just as they are today. Because osmundaceous rhizomes represent an ecological niche that has remained virtually unchanged over time and space and are abundant in the fossil record, they provide a paleoecological model system that could be used to explore epiphyte community structure through time.

Three-dimensional printing could provide flexibility in the design of a new generation of electrodes to be used in microbial electrochemical technologies (MET). In this work, we demonstrate the feasibility of using polylactic acid (PLA)/graphene—a common 3D-printing material—to build custom bioelectrodes. We also show that a suitable activation procedure is crucial to achieve an acceptable electrochemical performance (plain PLA/graphene bioanodes produce negligible amounts of current). Activation with acetone and dimethylformamide resulted in current densities similar to those typically observed in bioanodes built with more conventional materials (about 5 Am−2). In addition, the electrodes thus activated favored the proliferation of electroactive bacteria.

Rapid freehand drawings are of great importance in the early years of university studies of architecture, because both the physical characteristics of spaces and their sensory characteristics can be communicated through them. In order to draw architectural spaces, it is necessary to have the ability to visualize and manipulate them mentally, which leads us to the concept of spatial skills; but it also requires a development of spatial perception to express them in the drawings. The purpose of this research is to analyze the improvement of spatial skills through the full-scale sketching of architectural spaces in virtual immersive environments and to analyze spatial perception in reference to the capture of spatial sensations in virtual immersive environments. Spatial skills training was created based on the freehand drawing of architectural spaces using Head Mounted Displays (HMD) and registered the spatial sensations experienced also using HMD, but only in previously modeled realistic spaces. It was found that the training significantly improved orientation, rotation and visualization, and that the sensory journey and experimentation of architectural spaces realistically modeled in immersive virtual reality environments allows for the same sensations that the designer initially sought to convey.

Abstract Altered nutrient cycles and consumer populations are among the top anthropogenic influences on ecosystems. However, studies on the simultaneous impacts of human-driven environmental alterations on ecosystem functions, and the overall change in system multifunctionality are scarce. We used estuarine tidal flats to study the effects of changes in herbivore density and nutrient availability on benthic microalgae (diversity, abundance and biomass) and ecosystem functions (N2-fixation, denitrification, extracellular polymeric substances -EPS- as a proxy for sediment cohesiveness, sediment water content as a proxy of water retention capacity and sediment organic matter). We found consistent strong impacts of modified herbivory and weak effects of increased nutrient availability on the abundance, biomass and diversity of benthic microalgae. However, the effects on specific ecosystem functions were disparate. Some functions were independently affected by nutrient addition (N2-fixation), modified herbivory (sediment organic matter and water content), or their interaction (denitrification), while others were not affected (EPS). Overall system multifunction remained invariant despite changes in specific functions. This study reveals that anthropogenic pressures can induce decoupled effects between community structure and specific ecosystem functions. Our results highlight the need to address several ecosystem functions simultaneously for better ecosystem characterization and management.

We present a comprehensive study about the technical aspects of the application of focused ion beam (FIB) to the study of cuticles and compressions of fossil leaves. The technique allowed us to cross section and image fossil coalified plant remains with a spatial resolution within the 10 nm range, far higher than any other method employed so far. At various stages of the milling process, we observed significant gallium redeposition on the trench walls, particularly pronounced in the case of cavities. These highly unwanted artefacts can be greatly reduced, but not wholly eliminated, by lowering either or both the beam current and acceleration voltage; nevertheless, great care is needed when interpreting cross-sectional images.

This paper presents a path planner solution that makes it possible to autonomously explore underground mines with aerial robots (typically multicopters). In these environments the operations may be limited by many factors like the lack of external navigation signals, the narrow passages and the absence of radio communications. The designed path planner is defined as a simple and highly computationally efficient algorithm that, only relying on a laser imaging detection and ranging (LIDAR) sensor with Simultaneous localization and mapping (SLAM) capability, permits the exploration of a set of single-level mining tunnels. It performs dynamic planning based on exploration vectors, a novel variant of the open sector method with reinforced filtering. The algorithm incorporates global awareness and obstacle avoidance modules. The first one prevents the possibility of getting trapped in a loop, whereas the second one facilitates the navigation along narrow tunnels. The performance of the proposed solution has been tested in different study cases with a Hardware-in-the-loop (HIL) simulator developed for this purpose. In all situations the path planner logic performed as expected and the used routing was optimal. Furthermore, the path efficiency, measured in terms of traveled distance and used time, was high when compared with an ideal reference case. The result is a very fast, real-time, and static memory capable algorithm, which implemented on the proposed architecture presents a feasible solution for the autonomous exploration of underground mines.

ABSTRACT The expanded Human Oral Microbiome Database (eHOMD) is a comprehensive microbiome database for sites along the human aerodigestive tract that revealed new insights into the nostril microbiome. The eHOMD provides well-curated 16S rRNA gene reference sequences linked to available genomes and enables assignment of species-level taxonomy to most next-generation sequences derived from diverse aerodigestive tract sites, including the nasal passages, sinuses, throat, esophagus, and mouth. Using minimum entropy decomposition coupled with the RDP Classifier and our eHOMD V1-V3 training set, we reanalyzed 16S rRNA V1-V3 sequences from the nostrils of 210 Human Microbiome Project participants at the species level, revealing four key insights. First, we discovered that Lawsonella clevelandensis, a recently named bacterium, and Neisseriaceae [G-1] HMT-174, a previously unrecognized bacterium, are common in adult nostrils. Second, just 19 species accounted for 90% of the total sequences from all participants. Third, 1 of these 19 species belonged to a currently uncultivated genus. Fourth, for 94% of the participants, 2 to 10 species constituted 90% of their sequences, indicating that the nostril microbiome may be represented by limited consortia. These insights highlight the strengths of the nostril microbiome as a model system for studying interspecies interactions and microbiome function. Also, in this cohort, three common nasal species (Dolosigranulum pigrum and two Corynebacterium species) showed positive differential abundance when the pathobiont Staphylococcus aureus was absent, generating hypotheses regarding colonization resistance. By facilitating species-level taxonomic assignment to microbes from the human aerodigestive tract, the eHOMD is a vital resource enhancing clinical relevance of microbiome studies. IMPORTANCE The eHOMD (http://www.ehomd.org) is a valuable resource for researchers, from basic to clinical, who study the microbiomes and the individual microbes in body sites in the human aerodigestive tract, which includes the nasal passages, sinuses, throat, esophagus, and mouth, and the lower respiratory tract, in health and disease. The eHOMD is an actively curated, web-based, open-access resource. eHOMD provides the following: (i) species-level taxonomy based on grouping 16S rRNA gene sequences at 98.5% identity, (ii) a systematic naming scheme for unnamed and/or uncultivated microbial taxa, (iii) reference genomes to facilitate metagenomic, metatranscriptomic, and proteomic studies and (iv) convenient cross-links to other databases (e.g., PubMed and Entrez). By facilitating the assignment of species names to sequences, the eHOMD is a vital resource for enhancing the clinical relevance of 16S rRNA gene-based microbiome studies, as well as metagenomic studies.
