ABSTRACTBackground: Understanding the mechanism of sliding induced skin injuries on artificial turf is important to define preventive measures. Recent findings revealed that high peak loads on the knee and thigh during impact of a sliding tackle are critical for inducing skin injuries. Unfortunately, skin failure data under combined impact load is lacking.Methods: In this study the load tolerance of skin to impact on both dry and wet artificial turf and natural grass is investigated by developing an ex-vivomodel, a biaxial load applicator and a loading protocol.Results: The critical shear-normal stress combination at which skin breakdown occurred on dry artificial turf was 38 and 20 N.cm−2.  Skin damage on wet artificial turf firstly was noticed at a shear-normal stress combination of 55 and 20 N.cm−2.  On natural grass, skin breakdown only occurred at a combined shear and normal stress of 130 and 125  N.cm−2.  The thickness of the stratum corneum after impact strongly correlated to the applied mechanical load during impact on dry artificial turf.Conclusion: In contrast to natural grass, skin damage on dry artificial turf is strongly related to the magnitude of the impact load. Wetting of the artificial turf system improves the load tolerance of skin to impact.

Pharmacogenomics is a fast-growing field of investigations that aims to further elucidate the inherited nature of interindividual differences in drug disposition and effects, with the ultimate goal of providing a stronger scientific basis for selecting the optimal drug therapy. Providing the right drug for the right patient is an important problem in the treatment of cancer. This is mainly due to the lack of information about the sensitivity of the tumor for a specific treatment modality, such as either chemotherapy or radiation treatment. This presentation highlights two approaches to identify responsiveness to treatment. Both approaches are based on the identification of expression profiles. The first approach concentrates on drug resistance and the second on the signaling pathways leading up to the death of the cell. Both approaches provide expression profiles; however, the more dynamic expression profiling as used to determine the signaling in damage cells promises to be a better determinant for the pharmacogenomic changes in expression profiles and, consequently, a potential better determinant for drug efficacy. SUMMARYObjectives: To extend the hypothesis that late auditory EP shifts represent pathophysiologcial markers in schizophrenia. Methods: Early negative (±100 µs) and late positive (>300 ms) auditory oddball and CNV responses are topographically compared in 3 medicated schizophrenic subtypes.Results: Only late cortical responses differentiate between paranoid, residual and disorganised schizophrenia. Core features of the deficit residual state encompass low-voltage N2P3responses and missing central initial CNV components. Both paranoid and disorganised schizophrenics show a distinctive reactivity of the frontal cortex. Paranoid schizophrenia is characterized by an extensive frontal spread of the task-related P3response. Disorganised schizophrenia shows a specifically increased frontal CNV component indicating a nonspecific arousal reaction.Conclusion: Combinations of late auditory EP-patterns fairly fit with the cognitive/behavioral status observed in schizophrenia spectrum disorders.

Keratins are a group of cytoskeletal proteins that are found in human epidermis and other stratified squamous epithelia. Several different types of keratins have been described. Keratin 10 (K10) is a keratin that is expressed in well differentiated, suprabasal keratinocytes, and keratin 6 (K6) is a keratin which is associated with hyperproliferation. Psoriasis is a chronic inflammatory skin disease, and besides inflammation, disturbed differentiation and hyperproliferation are its hallmarks. In order to study the hyperproliferation associated keratinization in both well differentiated and poorly differentiated keratinocytes, and in order to assess the proliferative activity of all K10 and K6 subpopulations, simultaneous assessment of K6, K10, and DNA content is required. So far, a triple staining protocol had not been available. In the present study, we established a novel protocol for simultaneous measurement of K6, K10, and DNA content, which enables the characterization of the proliferative activity of several cellular subpopulations in epidermis. From 16 patients with psoriasis and from 15 healthy volunteers, punch biopsies were obtained. After preparation of single cell suspensions, cells were stained with the anti-keratin 10 IgG1-isotype monoclonal antibody RKSE60, with the anti-keratin 6 IgG2a-isotype monoclonal antibody LHK6B, and with the DNA fluorochrome TO-PRO-3 iodide. Isotype specific secondary antibodies conjugated with phycoerythrein (PE) and fluorescein isothiocyanate (FITC) were used as the second step in the staining procedure. Controls were measured omitting the primary antibodies, and gates were set in order to differentiate between the K10 and K6 subpopulations. Samples from both psoriatic patients and healthy volunteers were than measured. Owing to the IgG specificity of RKSE60 and LHK6B, no cross-reactivity was observed between these antibodies. The triple staining with RKSE60, LHK6B, and TO-PRO-3 iodide showed subpopulations of K10 expressing cells, K10/K6 co-expressing cells, and K6 only expressing cells. There was a significant difference in the proportion of K6 expression and K10/K6 co-expression between psoriatic and normal skin. Moreover, the proliferative activity of these subpopulations could be quantified by this protocol. We concluded that a triple staining protocol for the assessment of K6, K10, and DNA content, using the monoclonal antibodies LHK6B, RKSE60, and TO-PRO-3 iodide, supplies reliable and reproducible data for cellular studies on these keratins and for studying the proliferative activity of the subpopulations of these keratins in epidermis. Moreover, the present study showed that with respect to the proportion of K6, significant differences are present between psoriatic and healthy human skin.Cytometry (Comm. Clin. Normal rat kidney (NRK) fibroblasts are immortalized cells that are strictly dependent on externally added growth factors for proliferation. When cultured in the presence of epidermal growth factor (EGF) as the only growth stimulating hormone, these cells have a normal phenotype and undergo density-dependent growth inhibition. It has been postulated that this density-arrest results from a decrease of EGF receptor levels below a threshold level which makes these cells unresponsive to stimulation by EGF. In the present study, we show that NRK cells, made quiescent by serum-deprivation at submaximum density, are mitogenically still responsive to EGF, but show enhanced mitogenic stimulation after 8 hr pre-treatment with either transforming growth factor β (TGFβ) or retinoic acid (RA), while prostaglandin F2α (PGF2α) and bradykinin (BK) enhance the mitogenic stimulation by EGF only slightly under these conditions. Addition of TGFβ or RA results in an increase of both ¹²⁵I-EGF-binding capacity and EGF receptor mRNA levels. Using flow cytometric analysis, we show that pre-treatment with TGFβ or RA increases the percentage of cells entering the cell cycle as a function of time. Furthermore, pre-treatment of the cells with TGFβ or RA increases the rate of mitogen-activated protein kinase (MAPK) phosphorylation by EGF. PGF2α and BK also increase EGF receptor levels, but only with delayed kinetics. These results show that already in serum-deprived quiescent NRK cells, EGF receptor levels limit EGF-induced mitogenic stimulation. In many cell types cytoplasmic alkalization is an early marker for cell activation. An amiloride-sensitive Na+/H+ exchanger is an important regulator of this process. However, in keratinocytes the existence of a Na+/H+ exchanger nor a proliferation-associated increase in intracellular pH (pHi) has been demonstrated.
The aim of this study was to investigate whether or not keratinocytes, derived from the BALB/MK cell line, contain a Na+/H+ exchanger and whether cytoplasmic alkalization is proliferation-associated in these cells. This mouse keratinocyte cell line can easily be switched between a proliferative and a quiescent state under defined culture conditions. The novel pH-sensitive dye seminaphthorhodafluor (SNARF)-calcein proved to be very suitable for flow cytometric pHi measurements in BALB/MK cells. Initial measurements of the pHi using a cocktail of the established fluorochromes 2',7'-bis(carboxyethyl)-5,6-carboxyfluorescein (BCECF) and SNARF-1 failed because of the differential uptake and binding kinetics of these pH-sensitive dyes.
Using SNARF-calcein we were able to show proliferation to be associated with increased pHi. However, culture conditions were critical for these measurements. Our data indicate that the Na+/H+ exchanger is involved in this process, since acid load and pHi-recovery experiments showed the alkalization to be amiloride-sensitive.

The article discusses the outcome of the endeavor of photographer Zed Nelson on the emerging acceptance for artificial enhancement of physical appearances. It states that the result of his endeavor was his documentary book "Love Me." It mentions that Nelson has photographed several surgically removed body parts and artificial beauty, and has showed the emergence of beauty pageants after the American women earned the right to vote. It also mentions that the media plays a role in this emergence.
