O presente trabalho teve como objetivo a produção enzimática de um éster com atividade antioxidante (galato de isoamila) por esterificação de ácido gálico e álcool isoamílico em meio de solvente (terc-butanol). Oito lipases comerciais de diferentes fontes (microbianas e animal) foram empregadas na forma livre (extratos brutos) como biocatalisadores. As reações foram conduzidas empregando 0,25 mol L-1 de cada reagente (ácido gálico e álcool isoamílico), agitação mecânica de 240 rpm, 40°C, 4 h de reação e 1000 unidades de atividade enzimática baseada na hidrólise da emulsão de azeite de oliva. Os máximos valores de porcentagem de conversão foram obtidos para as preparações de lipase de Pseudomonas fluorescens – LPF (41,5 ± 5,1%) e de pâncreas suíno – LPP (38,4 ± 2,3%), a preparação de lipase testada com menor custo. A produtividade para estes sistemas de reação foi da ordem de 25 µmoles de ácido gálico consumido h-1 U-1. Estes revelam que a aplicação de uma lipase de baixo custo (LPP) pode ser promissora na produção de ésteres com propriedades antioxidantes, uma importante classe de compostos empregados na área de alimentos e cosméticos.

The present study aims to produce a mycelium-bound lipase of the fungus Rhizopus oryzae CCT3759 by submerged fermentation in order to be applied as biocatalyst in the hydrolysis of different vegetable oils. Optimal cultivation conditions have been achieved in a medium containing olive oil as inducer for 72 h of fermentation, thus obtaining 30.5 g/L of dry biomass concentration and hydrolytic activity of 389.1 U/g, which corresponds to a total lipase activity around of 12,000 U/L. Maximum hydrolytic activity was observed at pH 6.0 and 40 °C. Kinetic parameters concerning apparent Michaelis–Menten constant (Km = 50.5 mM) and maximum reaction rate (Vmax = 815.4 µmol/g min) have been determined in olive oil emulsion hydrolysis. Thermal stability tests revealed that the enzyme retained 75% of its initial activity after 4 h at 50 °C, whose thermal inactivation constant (Kd) and half-life (t1/2) was 0.073 h−1 and 9.4 h, respectively. The effect of biocatalyst concentration, expressed as activity units—U (200 and 400 U), on the hydrolysis of vegetable oils was investigated under fixed conditions: oil/buffer mass ratio of 25% (m/m), 100 mM buffer sodium phosphate pH 6.0, 40 °C and the mechanical stirring frequency of 600 rpm. As expected, increasing the initial activity from 200 to 400 U leads to higher values of initial reaction rates and hydrolysis percentage. However, initial reaction rate values were similar for six different vegetable oils due to the high accessibility of the lipase to the substrate under such experimental conditions. A complete hydrolysis of olive, cottonseed, sunflower and canola oils has been achieved after 26–30 h of reaction using 400 U of activity. These results suggest a promising application of the produced biocatalyst in the production of free fatty acids, an important class of compounds for oleochemical industries.

Alternative strategies are required to develop the optimized production of fatty acids using biocatalysis; molecular docking and response surface methodology are efficient tools to achieve this goal. In the present study, we demonstrate a novel and robust methodology for the sustainable production of fatty acids from Moringa oleifera Lam oil using lipase-catalyzed hydrolysis (without the presence of emulsifiers or buffer solutions). Seven commercial lipases from Candida rugosa (CRL), Burkholderia cepacia (BCL), Thermomyces lanuginosus (TLL), Rhizopus niveus (RNL), Pseudomonas fluorescens (PFL), Mucor javanicus (MJL), and porcine pancreas (PPL) were used as biocatalysts. Initial screening showed that CRL had the highest hydrolytic activity (hydrolysis degree of 81%). Molecular docking analysis contributed to the experimental results, showing that CRL displays more stable binding free energy with oleic acid (C18:1), which is the fatty acid of highest concentration in Moringa oleifera Lam oil. To evaluate and optimize the hydrolysis process, response surface methodology (RSM) was used. The effect of temperature, mass ratio oil:water, and hydrolytic activity on enzymatic hydrolysis was evaluated by central composite design using RSM. Under the optimized conditions (temperature of 37 °C, mass ratio oil:water of 25%, and hydrolytic activity of 550 U goil -1 ), the maximum hydrolysis degree (100%) was achieved. The present study provides a robust method for the enzymatic hydrolysis of different oils for efficient and sustainable fatty acid production.

— The current preference of costumers for natural and healthy products is increasing the employment of biotechnological processes that use enzymes, and the synthesis of esters is an example of this change. However, enzymes are high-cost product, which stimulates research in finding solutions that make them more economically attractive, like immobilization. This work aimed to use different protocols for immobilizing lipase and its application in ester synthesis. The results showed that Pseudomonas fluorences lipase (AKL) was the most efficient for immobilization among other studied lipases (Pseudomonas fluorences lipase (AKL), Pseudomonas cepacia lipase (PSL), Hog Pancreas lipase (PHL), Pancreas Porcinas lipase (PPL), and Mucor Javanicus lipase (MJL)), with hydrolytic activity of 3323.6 U/g. Both immobilization methods (physical adsorption and entrapment) showed promising results towards hydrolytic activity. The best immobilization by adsorption was obtained using AKL onto PHB (polyhydroxybutyrate), with 698.61 U/g of hydrolytic activity. For entrapment, AKL also presented the best result, with 247.30 U/g of hydrolytic activity. For the synthesis of ester, after a 60 h-reaction using the immobilized derivatives by physical adsorption, the esterification yield was 74.26 %. In terms of hydrolytic activity, the employed protocols were very promising and encourage the continuity of this study towards the optimization of processes using industrial lipases.

Here, we present a biorefinery perspective for biolubricant production with the valorisation of solid waste from Moringa oleifera Lam oil extraction (WMO). To achieve this, WMO, silica (SiO2), and a hybrid (SiO2–WMO) were prepared and employed as supports for the immobilisation of Candida rugosa lipase (CRL) via interfacial adsorption. The matrixes and immobilised biocatalysts were characterised by FTIR and AFM. The SiO2–WMO matrix was the most suitable, providing an immobilisation yield of 86 %. In the optimised conditions of esterification (40 °C, equimolar acid/alcohol ratio, and 22.6 mg/g of immobilised protein loading) in solvent-free medium, SiO2–WMO–CRL presented the best performance (94 % conversion) and retained its original activity after eight cycles. Furthermore, isoamyl esters exhibited physicochemical properties similar to those of commercial lubricants and could favourably be used as biolubricants in hydraulic machines and equipment. This pioneering work provides the opportunity for effective and cleaner production of biolubricants under a zero-waste Moringa oleifera Lam biorefinery approach.

— The continual search for alternative environmentally cleaner technologies in industrial processes has led to an increase in the use of enzymatic processes globally. However, due to their physical characteristics they require immobilization in order to remain effective. The objective of this study was to investigate the immobilization of the biotechnologically important and commercially available Candida rugosa lipase (CRL) by physical interfacial adsorption onto a number of matrices to act as biocatalysts. Five different types of support were tested: i) macroporous silica (synthetic inorganic), ii) polyhydroxybutyrate (natural organic), iii) polypropylene (synthetic organic), iv) polymethacrylate (synthetic organic), and v) polystyrene-divinylbenzene (synthetic organic). Results generated during this study showed that from the group of materials tested, polystyrene-divinylbenzene gave the best results with the highest amount of immobilized protein (8.10 ± 0.31 mg/g) and a good immobilization yield (90.35% ± 1.53%). The efficiency of protein immobilization was found to be highest when carried out at pH4.5, which is close to the isoelectric point of the enzyme.

This work aims to study the immobilization of Candida rugosa lipase (CRL) onto corn straw residue. For this purpose, chemical, morphological, and textural characteristics of the corn straw; immobilization process by adsorption; and immobilized enzyme activity and storage stability were evaluated. The corn straw presented isoelectric point of 7.0, surface with hydroxyl bands being favorable to the immobilization process. An irregular surface was also observed with fibers and pores, which are mesoporous and macroporous, characteristics that demonstrate efficiency in mass transfer mechanisms. Upon immobilization, it was observed that adsorption velocity is proportional to the square of the available adsorption sites (pseudo-second-order), and that the immobilization occurs in monolayers (Langmuir isotherm). The adsorption process was favorable and considered as a chemical adsorption mechanism. After immobilization, the optimum temperature increased, the optimum pH reduced, and the affinity of the biocatalyst for the substrate decreased. Corn straw derivative demonstrated good thermal stability. Regarding storage stability, there was approximately 12% loss of activity after 60 days of storage at 4 °C. Considering that no treatment was applied to the corn straw, this result is satisfactory and shows good affinity between this support and CRL.

A indústria açucareira tem como principal subproduto o melaço que é produzido na proporção de 40 a 60 quilos por tonelada de cana-de-açúcar processada. O melaço oriundo de uma tonelada de cana-de-açúcar processada pode produzir cerca de 12 litros de etanol, além do açúcar fabricado. Deste todo modo, constata-se que, na maioria dos países latino-americanos com produção açucareira, o melaço pode constituir uma fonte de etanol relevante e precursora para o atendimento das necessidades internas de combustível (HORTA NOGUEIRA, 2004). Na busca por alternativas para aperfeiçoar o processo de produção do etanol nas usinas, este trabalho visou mostrar a influência da clarificação e esterilização no preparo do melaço a ser empregado no processo fermentativo, determinando assim condições ótimas para a produção de etanol. Foram realizados três tipos de tratamentos no mosto para que a fermentação ocorresse: T1- Melaço não clarificado e não estéril. T2- Melaço estéril e não clarificado. T3- Melaço estéril e clarificado. O tratamento T3 foi o que apresentou maior produtividade e rendimento, devido ao procedimento de esterilização e clarificação que se mostraram influentes no processo fermentativo.

Os micro-organismos endofiticos sao fungos e bacterias encontrados no interior de tecidos e orgaos saudaveis das plantas e que nao causam danos aparentes aos hospedeiros durante o ciclo vital in vivo. Os fungos endofiticos sao capazes de produzir substâncias de interesse biotecnologico, sendo que algumas enzimas possuem aplicacoes em processos industriais. A especie Annona crassiflora , conhecida popularmente como marolo ou pinha do cerrado, e uma arvore nativa e exclusiva do cerrado brasileiro, amplamente difundido em todo o bioma. E encontrada e abrangem os estados de Sao Paulo, Minas Gerais, Bahia, Mato Grosso do Sul, Mato Grosso e Tocantins. Nesse contexto, o presente trabalho avaliou o potencial biocatalitico de enzimas produzidas por fungos endofiticos isolados de Annona crassiflora aplicadas em reacoes de biotransformacao.

Lipase from Candida rugosa (CRL) was immobilized via adsorption on niobium pentoxide. The best condition for adsorption was pH 6, 150 rpm and enzyme/support ratio of 8000 U gsupport −1. The kinetic study resulted in a better fit for pseudo-first order kinetics, and Langmuir isotherm best fit the data. Adsorption thermodynamics showed that the adsorption was favorable, and it approached to physical adsorption. After immobilization process, the optimum pH and the affinity of the biocatalyst for the substrate decreased, and the optimum temperature increased. Niobium pentoxide immobilized derivative (IDNb) presented suitable thermal stability, preserving more than 50% of its residual activity after 60 min of incubation at 60 °C. After 60 days of storage at 4 °C, the recorded loss of activity for IDNb was 45%.

Biodiesel production was investigated by hydroesterification using enzymatic catalysis in both hydrolysis and esterification reactions. Soybean oil resulting from food frying processes (WCO) was used as feedstock. Lipase from Geotrichum candidum was produced by submerged fermentation and used without prior purification as biocatalyst in WCO hydrolysis, which was optimized using a factorial design. A complete hydrolysis of WCO (44.1% mass fraction) was obtained after 80 min of reaction at 40 °C, performed in an emulsifier-free system with no buffer (40 °C and 900 rpm). In a subsequent step, FFA obtained from the hydrolysis reaction were esterified (40 °C, 200 rpm) using ethanol as acyl-acceptor (1:1.5) and Pseudomonas fluorescens lipase (PFL) immobilized on styrene-divinylbenzene (SDB) as biocatalyst (15% m/v). This biocatalyst was recycled for 6 cycles with no significant activity losses. The biodiesel composition was found by gas chromatography, and proton nuclear magnetic resonance (1H NMR) spectra and Fourier transform infrared spectroscopy (FTIR) were used to verify the quality and purity of biodiesel. Enzymatic biodiesel achieved FAEE content of 94.1 ± 0.5% and no traces of monoacyglycerol (MAG), diacylglycerol (DAG), triacylglycerol (TAG) or free glycerol were observed, indicating that the hydroesterification route produced high-quality biodiesel, despite the fact that WCO was used as feedstock.

Para suprir a crescente demanda mundial energetica, em um contexto de grandes preocupacoes sobre as questoes ambientais e a escassez das reservas dos combustiveis fosseis e, alem do interesse pela independencia energetica, investe-se na pesquisa e em formas alternativas de energia. Dentre essas alternativas, o biodiesel destaca-se por ser um combustivel renovavel produzido utilizando-se fontes naturais. As analises da materia-prima, como a densidade, a viscosidade e a massa molar media, foram realizadas previamente ao processo de sintese. Este trabalho consiste na producao de biodiesel a partir de oleo de acai empregando lipase comercial imobilizada em suporte de baixo custo. A sintese de biodiesel alcancou um rendimento de 51,49% em 12h de reacao.

The present study reports the improved enzymatic synthesis of ethyl valerate (green apple flavor) by esterification reaction of ethanol and valeric acid in heptane medium. Lipase from Thermomyces lanuginosus (TLL) was immobilized by physical adsorption on polyhydroxybutyrate (PHB) particles and used as a potential biocatalyst. The effect of certain parameters that influence the ester synthesis was evaluated by factorial design. The experimental conditions that maximized the synthesis of ethyl valerate were 30.5°C, 18% m/v of biocatalyst (TLL-PHB), absence of molecular sieves, agitation of 234 rpm, and 1,000 mM of each reactant (ethanol and valeric acid). Under these conditions, conversion percentage ≈92% after 105 min of reaction was observed. Soluble TLL was also used as biocatalyst and the highest conversion was of 82% after 120 min of reaction. Esterification reaction performed in a solvent-free system exhibited conversion of 13% after 45 min of reaction catalyzed by immobilized lipase, while the soluble lipase did not exhibit catalytic activity. The synthesis of the ester was confirmed by Fourier transform infrared spectroscopy and gas chromatography-mass spectrometry analyses. After six consecutive cycles of ethyl valerate synthesis, the prepared biocatalyst retained ≈86% of its original activity.

Simultaneous enzymatic hydrolysis and anaerobic biodegradation of lipid-rich wastewater from poultry industry with porcine pancreatic lipase at different concentrations (from 1.0 to 3.0 g L−1) were performed. The efficiency of the enzymatic pretreatment was measured by the Chemical Oxygen Demand (COD) removal and formation of methane. All samples pretreated with lipase showed a positive effect on the COD removal and formation of methane. After 30 days of anaerobic biodegradation the methane production varied from 569 ± 95 to 1,101 ± 10 mL for crude wastewater and pretreated at 3.0 g L−1 enzyme, respectively. COD removal of wastewater supplemented at different enzyme concentrations was found to be threefold higher than crude wastewater. The use of lipases seems to be a promising alternative for treating lipid-rich wastewaters such as those from the poultry industry.

A low-cost commercially available lipase preparation from animal source was used to perform enzymatic hydrolysis pretreatment of lipid-rich wastewater from dairy industries. The hydrolysis was carried for different incubation time (4–24 h) and the pretreatment efficiency was verified by running comparative biodegradability tests (crude and treated effluents). All pretreated assays showed higher reaction rate in relation to crude wastewater assay, which was confirmed by the increased levels of biogas production and higher organic matter (COD) and color removal. The pretreatment was optimized for 12 h hydrolysis time, enabling high-biogas formation (445 ± 29 mL) and organic matter removal (78.2%). Promising results in relation to the biogas formation (354 ± 34 mL) were also found when hydrolysis and biodegradation steps were carried out simultaneously. The use of enzymatic treatment seemed to be a very promising alternative for treating wastewaters having high-fat contents, such as those from the dairy industry.

The objective of this study was to evaluate the immobilization of lipase on a chitosan support by physical adsorption, aiming at its application in hydrolytic and synthetic reactions. Two types of chitosan (flakes and porous) were used for immobilizing lipase from a microbial source (Candida rugosa) and animal cells (porcine pancreas). The best results for recovery of total activity after immobilization were obtained for microbial lipase and porous chitosan beads. This set was selected for further immobilization studies, including full characterization of the immobilized derivative in aqueous and organic media. In aqueous medium, the operational and thermal stabilities of this preparation were quantified. In organic medium, the direct synthesis of n-butyl butyrate in organic solvent was chosen as a model reaction. The influence of several parameters, such as temperature, initial butyric acid concentration and amount of enzyme in the reaction system, was analyzed. Production of n-butyl butyrate was optimized and an ester yield response equation was obtained, making it possible to predict ester yields from known values of the three main factors. Use of this immobilized preparation was extended to the direct esterification of a large range of carboxylic acids (from C2 to C12) with a variety of alcohols (from C2 to C10).

Microbial lipase from Candida rugosa immobilized into porous chitosan beads was tested for esterification selectivity with butanol and different organic acids (C2–C12), and butyric acid and different aliphatic alcohols (C2–C10). After 24 h, the acids tested achieved conversions of about 40–45%. Acetic acid was the only exception, and in this case butanol was not consumed. Different alcohols led to butyric acid conversions >40%, except for ethanol, in which case butyric acid was converted only 26%. The system’s butanol and butyric acid were selected for a detailed study by employing an experimental design. The influence of temperature, initial catalyst concentration, and acid:alcohol molar ratio on the formation of butyl butyrate was simultaneously investigated, employing a 23 full factorial design. The range studied was 37–50°C for temperature (X1), 1.25–2.5% (w/v) for the catalyst concentration (X2), and 1 and 2 for the acid:alcohol molar ratio (X3). Catalyst concentration (X2) was found to be the most significant factor and its influence was positive. Maximum ester yield (83%) could be obtained when working at the lowest level for temperature (37°C), highest level for lipase concentration (2.5% [w/v]), and center level of acid:alcohol molar ratio (1.5). The immobilized lipase was also used repeatedly in batch esterification reactions of butanol with butyric acid, revealing a half-life of 86 h.

Lecitase Ultra (a quimeric fosfolipase commercialized by Novozymes) has been immobilized via two different strategies: mild covalent attachment on cyanogen bromide agarose beads and interfacial activation on octyl-agarose beads. Both immobilized preparations have been submitted to different individual or cascade chemical modifications (amination, glutaraldehyde or 2,4,6-trinitrobenzensulfonic acid (TNBS) modification) in order to check the effect of these modifications on the catalytic features of the immobilized enzymes (including stability and substrate specificity under different conditions). The first point to be remarked is that the immobilization strongly affects the enzyme catalytic features: octyl-Lecitase was more active versus p-nitrophenylbutyrate but less active versus methyl phenylacetate than the covalent preparations. Moreover, the effects of the chemical modifications strongly depend on the immobilization strategy used. For example, using one immobilization protocol a modification improves activity, while for the other immobiled enzyme is even negative. Most of the modifications presented a positive effect on some enzyme properties under certain conditions, although in certain cases that modification presented a negative effect under other conditions. For example, glutaraldehyde modification of immobilized or modified and aminated enzyme permitted to improve enzyme stability of both immobilized enzymes at pH 7 and 9 (around a 10-fold), but only the aminated enzyme improved the enzyme stability at pH 5 by glutaraldehyde treatment. This occurred even though some intermolecular crosslinking could be detected via SDS-PAGE. Amination improved the stability of octyl-Lecitase, while it reduced the stability of the covalent preparation. The search for an inexpensive support has motivated our group to undertake this work dealing with the use of chitosan as matrix for immobilizing lipase. In addition to its low cost, chitosan has several advantages for use as a support, including its lack of toxicity and chemical reactivity, allowing easy fixation of enzymes. In this article, we describe the immobilization of Candida rugosa lipase onto porous chitosan beads for the enzymatic hydrolysis of olive oil. The binding of the lipase onto the support was performed by physical adsorption using hexane as the dispersion medium. A comparative study between free and immobilized lipase was conducted in terms of pH, temperature, and thermal stability. A slightly lower value for optimum pH (6.0) was found for the immobilized form in comparison with that attained for the soluble lipase (7.0). The optimum reaction temperature shifted from 37 degrees C for the free lipase to 50 degrees C for the chitosan lipase. The patterns of heat stability indicated that the immobilization process tends to stabilize the enzyme. The half-life of the soluble free lipase at 55 degrees C was equal to 0.71 h (Kd = 0.98 h(-1)), whereas for the immobilized lipase it was 1.10 h (Kd = 0.63 h(-1)). Kinetics was tested at 37 degrees C following the hydrolysis of olive oil and obeys the Michaelis-Menten type of rate equation. The Km was 0.15 mM and the Vmax was 51 micromol/(min x mg), which were lower than for free lipase, suggesting that the apparent affinity toward the substrate changes and that the activity of the immobilized lipase decreases during the course of immobilization.

Three different approaches for lipase catalysed synthesis of citronellyl acetate by a commercial available immobilized lipase have been studied: a) direct esterification reaction of citronellol with acetic acid; b) alcoholysis of butyl acetate with citronellol and c) transesterification of citronellyl butyrate with butylacetate. Heptane was used as solvent for all the experiments. The extent of reaction occurred in the order alcoholysis>transesterification>esterification. Substrate partitioning between the immobilization support and the organic medium seems to greatly influence the catalytic performance of this enzyme preparation. Production of citronellyl acetate, was found to be dependent on the partition coefficient of the acyl donor which account the greatest value for the acetic acid.

O objetivo deste trabalho foi a produção de biodiesel via rota enzimática, utilizando resíduo gorduroso de fritura como substrato e o planejamento fatorial como ferramenta de análise. O planejamento fatorial foi empregado para avaliar os parâmetros da reação de transesterificação, quanto à razão molar óleo:etanol (1:3; 1:4,7; 1:9; 1:13,3 e 1:15) e temperatura (30, 34, 45, 56 e 60ºC). A reação foi conduzida em shaker, usando 20 g do meio reaciona e 2 g do derivado imobilizado de lipase durante 15h. Os diferentes ensaios foram avaliados por cromatografia gasosa (verificação do teor de ésteres de etila) e viscosímetro, mostrando uma produção de 67,9% de ésteres de etila para a razão molar óleo:etanol de 1:4,7 e 34ºC de temperatura; e uma viscosidade de 3,82 cP para a razão molar óleo:etanol de 1:3 e 45ºC de temperatura, sendo estes os melhores resultados, indicando que a variável que interferiu significativamente no rendimento da reação foi a temperatura.

Lipids-rich wastewater was partial hydrolyzed with porcine pancreas lipase and the efficiency of the enzymatic pretreatment was verified by the comparative biodegradability tests (crude and treated wastewater). Alternatively, simultaneous run was carried out in which hydrolysis and digestion was performed in the same reactor. Wastewater from dairy industries and low cost lipase preparation at two concentrations (0.05 and 0.5% w.v-1) were used. All the samples pretreated with enzyme showed a positive effect on organic matter removal (Chemical Oxygen Demand-COD) and formation of methane. The best results were obtained when hydrolysis and biodegradation were performed simultaneously, attaining high COD and color removal independent of the lipase concentration. The enzymatic treatment considerably improved the anaerobic operational conditions and the effluent quality (lower content of suspended solids and less turbidity). Thus, the use of enzymes such as lipase seemed to be a very promising alternative for treating the wastewaters having high fat and grease contents, such as those from the dairy industry.O presente trabalho teve como objetivo o pré-tratamento de efluente da indústria de laticínios na hidrólise de lipídeos, empregando lipase de fonte de células animais de baixo custo disponível comercialmente (pâncreas de porco) na formação de gás metano por biodegradabilidade anaeróbia empregando diferentes concentrações de lipase (0,05 e 0,5 % w.v-1). A utilização de lipase no pré-tratamento do efluente acelerou a hidrólise de lipídeos e, conseqüentemente, auxiliou o tratamento biológico resultando na redução da matéria orgânica em termos de Demanda Química de Oxigênio (DQO), cor e sólidos em suspensão como lipídeos. Os melhores resultados em termos de remoção de DQO e cor foram obtidos quando a hidrólise e biodigestão foram realizadas simultaneamente, independente da concentração de lipase empregada. Estes resultados mostram que a hidrólise e biodigestão podem ser realizadas simultaneamente empregando baixa concentração de lipase (0,05% w.v-1), contribuindo para a redução de custo e tempo no processo de biodegradação aneróbia.

Chitin was functionalized with hexamethylenediamine followed by glutaraldehyde activation, and its capacity to bind Candida rugosa lipase was investigated. The loading of 250 units g(-1) support showed to be effective, resulting in a uniform enzyme fixation with high catalytic activity. Both free and immobilized lipases were characterized by determining the activity profile as a function of pH, temperature, and thermal stability. For the immobilized lipase, the influence of the reaction temperature and substrate polarity in nonconventional biocatalysis was also analyzed. Production of butyl esters was found to be dependent on the substrate partition coefficient, which accounts the greatest value for the system butanol and butyric acid. The highest enzyme activity was found for the system butanol and caprylic acid at a reaction temperature of 40 degrees C. Under such conditions, the operational stability tests indicated that a small enzyme deactivation occurs after 12 batches, revealing a biocatalyst half-life of 426.7 h.

The search for an inexpensive support has motivated our group to undertake this work dealing with the use of chitosan as matrix for immobilizing lipase. In addition to its low cost, chitosan has several advantages for use as a support, including its lack of toxicity and chemical reactivity, allowing easy fixation of enzymes. In this article, we describe the immobilization of Candida rugosa lipase onto porous chitosan beads for the enzymatic hydrolysis of olive oil. The binding of the lipase onto the support was performed by physical adsorption using hexane as the dispersion medium. A comparative study between free and immobilized lipase was conducted in terms of pH, temperature, and thermal stability. A slightly lower value for optimum pH (6.0) was found for the immobilized form in comparison with that attained for the soluble lipase (7.0). The optimum reaction temperature shifted from 37°C for the free lipase to 50°C for the chitosan lipase. The patterns of heat stability indicated that the immobilization process tends to stabilize the enzyme. The half-life of the soluble free lipase at 55°C was equal to 0.71 h (K d = 0.98h-1), whereas for the immobilized lipase it was 1.10 h (K d = 0.63h-1). Kinetics was tested at 37°C following the hydrolysis of olive oil and obeys the Michaelis- Menten type of rate equation. The K m was 0.15 mM and the V max was 51 µmol/ (min-mg), which were lower than for free lipase, suggesting that the apparent affinity toward the substrate changes and that the activity of the immobilized lipase decreases during the course of immobilization.

The enzymatic synthesis of selected low molecular weight esters such as acetate esters by direct esterification using acetic acid as acyl donor usually display low yields. The acetic acid changes the polarity of the reaction medium, which in this turn modifies the partitioning of water between the solid phase (enzyme preparation) and the liquid phase (substrate), resulting in its accumulation on the enzyme solid phase. This may reduce the enzyme´s local pH. Therefore, the enzyme active site is modified and the reaction became nearly impossible. Our previous work showed that there is a negative relationship between enzyme activity and substrate partition coefficient (Ps); that is, the higher the substrate partition coefficient the lower the amount of product formed. This work investigated the feasibility of minimizing this inhibition by replacing the esterification reaction for alcoholysis reaction using several acetate esters. This approach enhanced the reaction yields to 46%, which is about 3 times higher than that one obtained in the esterification route.

O soro resultante da produção de queijo, por possui alta carga orgânica e composição rica em lactose, proteínas, lipídios e minerais, apresenta potencial para uma série de aplicações mais nobres e ambientalmente favoráveis do que o seu despejo como rejeito. Dentre as aplicações possíveis do soro, encontram-se a sua utilização direta como substrato para produção de enzimas e, de forma indireta, na geração de energia, como, por exemplo, na produção de biogás por digestão anaeróbia. Neste último caso, a presença de lipídios pode limitar o processo. Portanto, destaca-se a importância das lipases na atuação da etapa de hidrólise da digestão anaeróbia, contribuindo para o processo de geração de energia. Dentro deste contexto, o presente estudo teve como objetivo avaliar a aplicação do soro de queijo na produção de lipases ligadas ao micélio e na geração de biogás. Para tal propósito, foram avaliados os fungos filamentosos Penicillium janthinellum CCT3162, Penicillium italicum AT4421 e Mucor circinelloides URM 4182, conhecidos por serem bons produtores de lipase ligada ao micélio. O estudo foi iniciado pela determinação das condições otimizadas da hidrólise do lipídio presente no soro, avaliando o efeito da linhagem fúngica, do carregamento de lipase (na forma de células íntegras), pH do meio e temperatura, por meio de planejamento experimental utilizando o método de Taguchi, com matriz L9 saturada. O fungo M. circinelloides foi selecionado como a fonte mais eficiente da enzima lipase, empregando soro de queijo como meio de cultivo. A suplementação do soro foi também avaliada, mas os aditivos utilizados (azeite de oliva, peptona e sais) não proporcionaram aumento significativo na atividade hidrolítica da lipase. A principal lipase gerada foi produzida a partir do meio de cultivo composto por soro de queijo integral, sem nenhum aditivo, por um período de incubação de 120 h. Algumas propriedades da lipase obtida foram pH ótimo de 7,5, temperatura ótima no intervalo de 40 a 60 °C e tempo de meia-vida com a relação à estocagem de 26,8 dias. Foram também analisados os parâmetros cinéticos da lipase quanto ao Km (115,74 mmol L-1 ) e Vmax (186,92 µmol g-1 min-1 ). O presente trabalho teve como objetivo a obtenção de ésteres com propriedades lubrificantes a partir de matérias-primas renováveis, como a biomassa microbiana, e um subproduto da indústria sucroalcooleira (óleo fúsel), avaliando a eficiência de catalisadores heterogêneos (bioquímicos e químicos) por meio de duas rotas catalíticas: 1) transesterificação do óleo microalgal empregando a lipase de Burkholderia cepacia (LBC) imobilizada em Nb2O5 e 2) transesterificação direta da biomassa microbiana catalisada por H3PMo12O40 impregnado em Nb2O5. O trabalho foi desenvolvido em cinco etapas. Inicialmente, a microalga Dunaliella salina foi cultivada em fotobiorreator de coluna de bolhas com capacidade de 10 L visando obtenção de material lipídico suficiente para as reações. Foram realizados 15 cultivos, obtendo-se um volume total de 150 L, correspondente a 172,5 g de biomassa e 32,76 g de material lipídico. Em seguida foram preparados os catalisadores propostos por protocolos já estabelecidos, sendo obtido para o catalisador químico acidez superficial de 5,644 mmol H+ g -1 e para o catalisador bioquímico elevada atividade hidrolítica (1400 ± 103 U g-1 ). Na terceira etapa, as reações de transesterificação por catálise bioquímica, empregando o óleo microalgal e diferentes aceptores do grupo acila (alcoóis etílico, butílico e isoamílico) óleo fúsel simulado e óleo fúsel foram conduzidas em reatores de vidro com capacidade de 50 mL, contendo 10 gramas de meio reacional à 45 °C e agitação magnética (150 rpm), durante um período máximo de 120 h, empregando iso-octano como solvente. Como reação controle foram efetuados testes com óleo de macaúba nas mesmas condições reacionais. A transesterificação direta da biomassa microalgal, empregando como catalisador H3PMo12O40 impregnado em Nb2O5 foi a quarta etapa do trabalho, em que as reações foram conduzidas em reator pressurizado (Parr Série 5500), a 250 °C, agitação de 300 rpm e razão molar 1:180 material lipídico/aceptor do grupo acila. Finalmente, os produtos obtidos foram purificados e quantificados por cromatografia gasosa, RMN 1H, viscosimetria e teor de glicerídeos. Os resultados obtidos demonstraram o potencial da microalga Dunaliella salina na produção de biolubrificantes, visto que ambos os catalisadores (químico e bioquímico) atuaram de forma eficiente convertendo os ácidos graxos presentes no óleo microalgal nos ésteres alquílicos correspondentes. A estabilidade à oxidação é uma das propriedades mais importantes do biodiesel pois afeta a sua estabilidade durante o período de estocagem influenciando no desempenho do motor. As pesquisas atuais tendem a desenvolver aditivos antioxidantes para melhorar sua estabilidade. Algumas plantas exibem propriedades antioxidantes devido à sua composição química e que podem auxiliar nessa estabilização. O objetivo deste trabalho foi estudar a estabilidade termo oxidativa do biodiesel de soja e do sebo bovino puro e aditivado com folhas e extrato das folhas de acerola, graviola, manga e pitanga frente à luz, a 60º C e a baixas temperaturas por 0, 168, 504, 1176 e 1848 h de estocagem de forma a avaliar o seu enquadramento dentro das normas ANP, ASTM, EN. As folhas usadas como aditivo antioxidante foram moídas num moinho de facas a 10 mesh e os extratos das folhas foram obtidos pelo processo Soxhlet. O biodiesel de soja foi obtido pela rota etílica e o de sebo bovino pela metílica. A estabilidade termo oxidativa foi monitorada determinando-se o índice de acidez, a viscosidade cinemática, a densidade, por espectroscopia no infravermelho com Transformada de Fourier e por termogravimetria. Os resultados mostraram que o biodiesel de soja e do sebo bovino é estável frente à luz após 1848 h. O efeito antioxidante das folhas sobre o biodiesel de soja após 168 h de estocagem a 60º C segue a seguinte ordem: folhas de graviola > folhas de pitanga > folhas de manga > folhas de acerola, enquanto para o extrato das folhas tem-se: extrato das folhas de acerola> extrato das folhas de graviola>extrato das folhas de manga>extrato das folhas de pitanga, após 504h. Para o biodiesel de sebo bovino as folhas apresentaram a ordem: folhas de pitanga>folhas de acerola >folhas de manga>folhas de graviola, e para os extratos a ordem: extratos da folha de manga > extratos da folha de graviola >extratos da folha de pitanga>extratos da folha de acerola. O butil hidroxitolueno (BHT), antioxidante sintético, apresentou maior efeito antioxidante sobre o biodiesel de soja em relação às aditivações com folhas e seus extratos após 1176h. No biodiesel de sebo bovino, a atividade do BHT foi semelhante aos extratos das folhas de acerola, graviola e pitanga após 504h e o de manga após 1176h. A ordem da estabilidade do biodiesel de soja aditivado frente a baixas temperaturas foi: extrato de pitanga > extrato de manga > extrato de acerola > extrato de graviola. Os resultados dos testes em baixa temperatura indicam que o biodiesel de soja aditivado ou não pode ser usado em qualquer região do país para todas as estações do ano. O efeito dos extratos das folhas na estabilidade do biodiesel frente a baixas temperaturas foi: extratos das folhas de manga>extratos de acerola>extratos de graviola>extratos de pitanga. O biodiesel de sebo bovino puro apresenta menor estabilidade frente a baixas temperaturas em relação ao biodiesel de soja. Os resultados obtidos do IA, viscosidade cinemática, densidade, FTIR e TGA indicam que as folhas e extratos das folhas das frutas estudadas podem atuar como aditivos antioxidantes na estabilidade oxidativa do biodiesel. Oxidation stability is one of the most important properties of biodiesel because it affects their stability during the storage period influence on engine performance. Current research tend to develop antioxidant additives to improve their stability. Some plants have antioxidant properties due to their chemical composition and may assist in this stabilization. The aim of this study was to oxidative term stability of soybean biodiesel and pure beef tallow and spiked with leaves and extracts of the leaves of cherry, soursop, mango and cherry front light, 60 ° C and low temperatures for 0, 168, 504, 1176 and 1848 h of storage in order to evaluate its framework within the ANP, ASTM, EN. The leaves used as an antioxidant additive were ground in a knife mill to 10 mesh and extracts of leaves were obtained by Soxhlet process. Soybean biodiesel was obtained by the ethyl route and beef tallow by methyl. The term oxidative stability was monitored by determining the acid number, kinematic viscosity, density, by infrared spectroscopy with Fourier Transform and thermogravimetry. The results showed that the biodiesel soya and tallow is stable against the light for 1848 h. The antioxidant effect of leaves on soybean biodiesel after 168 h of storage at 60 ° C follows the following order: leaves of soursop> sheets of cherry> mango leaves> acerola leaves while for the leaves of the statement we have: extract leaves acerola> extract of the leaves of soursop> extract of mango leaves> extract of cherry leaves after 504h. For the leaves beef tallow biodiesel showed the order: leaves of cherry> leaves acerola> mango leaves> soursop leaves and extracts the order: Mango leaf extracts> soursop leaf extracts> leaf extracts of cherry> acerola leaf extracts. The butylated hydroxytoluene (BHT), synthetic antioxidant, showed higher antioxidant effect on soybean biodiesel in relation to additivations with leaves and their extracts after 1176h. In beef tallow biodiesel BHT activity was similar to the extracts of the leaves of cherry, soursop and cherry after 504h and the sleeve after 1176h. The order of stability of soy biodiesel additive against low temperatures was: cherry extract> mango extract> acerola extract> soursop extract. The results of the tests indicate that low temperature soybean biodiesel additive or may not be used in any region of the country for all seasons. The effect of the extracts of leaves in front biodiesel stability at low temperatures was: extracts of mango leaves> acerola extracts> soursop extracts> cherry extracts. Biodiesel pure beef tallow has lower stability in low temperatures to soy biodiesel. The results of the AI, kinematic viscosity, density, FTIR and TGA show that the extracts of the leaves and fruits studied sheets can act as antioxidant additives in the oxidative stability of biodiesel.

Este trabalho teve como objetivo sintetizar o palmitato de isopropila, éster emoliente, empregando como biocatalisador lipases microbianas imobilizadas em partículas de poli(estireno-co-divinilbenzeno) (STY-DVB-M) obtidas por meio da técnica de polimerização em suspensão e magnetizadas por co-precipitação de íons de Fe +2 e Fe +3 em meio básico. Inicialmente, a influência da concentração do agente de suspensão e da agitação na distribuição granulométrica do polímero sintetizado foi avaliada por planejamento experimental 2 2, com triplicata no ponto central. As polimerizações que resultaram nas maiores quantidades de partículas com tamanhos apropriados para utilização como suporte para imobilização (entre 80 e 24 mesh), foram obtidas empregando as seguintes condições experimentais: 1% de agente de suspensão (PVA) e 400 rpm de agitação. O suporte obtido foi utilizado para imobilizar a lipase de Candida rugosa (LCR) e lipase de Penicillium camemberti (LG) via adsorção física e os biocatalisadores resultantes aplicados em reações de esterificação do ácido palmítico com isopropanol em meio heptano. Para cada biocatalisador foi adotado um planejamento experimental estrela rotacional 22, com triplicata no ponto central para avaliar a influência da concentração de biocatalisador (% m/v) e da razão molar (ácido:álcool) no rendimento de esterificação. Nas condições otimizadas em 12 h de reação foram obtidos 75,60 e 88,53% de rendimento de esterificação, respectivamente, para a LCR e LG imobilizada em STY-DVB-M. A quantidade de água formada durante o bioprocesso não foi considerada fator relevante para interferir no progresso da síntese. O biocatalisador obtido pela LG imobilizada em STY-DVB-M foi empregado em biorreator de tanque agitado (280 mL), na condição ótima, em um experimento com ampliação de escala, atingindo 85,68% de rendimento em 12 horas de reação. No entanto, foi observado cisalhamento do suporte em função da agitação mecânica, optando-se pela realização do bioprocesso em biorreator de leito fixo (dimensões: 11 mm de diâmetro interno x 16,6 mm de comprimento) com recirculação do meio reacional (1,5 mL.min-1). A operação do sistema nesta configuração foi impossibilitada pela evaporação e/ou percolação do solvente. Assim, foram realizadas reações em reator de leito fixo em meio isento de solvente nas seguintes razões molares: 1:3, 1:4 e 1:6 (ácido:álcool), cujos melhores resultados foram obtidos na razão molar de 1:4, apresentando 56% de rendimento, 215,88 g.L-1 do palmitato de isopropila e produtividade de 26,87 g.L-1.h-1, decorridas 8h de reação com vazão de reciclo de 1,5 mLmin-1. O emprego de peneira molecular no reservatório do substrato proporcionou um acréscimo de aproximadamente 7% na formação do éster. Essa mesma condição experimental (1:4 em meio isento de solvente) foi testada em reator de tanque agitado, atingindo produtividade de 33,40 g.L-1.h-1 em 8h de reação. No entanto, em função da desvantagem do cisalhamento do suporte constatada neste tipo de biorreator, concluiu-se que o biorreator de leito fixo foi a configuração mais adequada, para a condução do bioprocesso estudado, apresentando viabilidade do processo enzimático empregando lipase imobilizada em suporte híbrido magnetizado para a síntese do palmitato de isopropila. The aim of this study was to synthesize isopropyl palmitate, an emollient ester, using as biocatalyst microbial lipase immobilized on magnetic copolymer. The support was prepared by suspension polymerization technique using styrene and divinylbenzene monomers in the presence of magnetite particles synthesized by co-precipitation of Fe+2 and Fe+3. The influence of the suspending agent concentration and the mechanical agitation on the granulometric distribution of the synthesized polymer was assessed by a 22 factorial design with triplicate at central point. The statistical analysis indicated that the optimal experimental conditions to obtain the largest amount of particles with suitable size (between 80 and 24 mesh) to be used as immobilizing support were attained at 1% suspending agent (PVA) at 400 rpm stirring. The resulting support was used to immobilize Candida rugosa lipase (LCR) and Penicillium camemberti lipase (LG) by physical adsorption and applied in the esterification reaction of palmitic acid with isopropyl alcohol in presence of solvent (heptane). For each biocatalyst a central composite 22 experimental design with triplicate at central point was performed to determine the influence of biocatalyst concentration (% m/v) and molar ratio (acid: alcohol) on the reaction yield. Under the optimal conditions higher performance was achieved by LG immobilized on STY-DVB-M (88.53%) than LCR immobilized on STYDVB-M (75.60%) with shaking at 12 hours reaction. For both reactions the amount of water formed as byproduct was not found to be a relevant factor that interferes in the synthesis progress. The most activity immobilized biocatalyst (LG STY-DVB-M) was used in agitated tank bioreactor (280 mL), under optimized conditions, in an experiment with scale-up, reaching 85.68% yield in 12 hours. However, it has been observed that the support was low mechanical shear and therefore an attempted was made to run the reaction in a packed bed bioreactor (dimensions: 11 mm internal diameter x 16,6 mm length) with recirculation of the reaction medium (1,5 mL.min-1). The system could not be operated under this configuration because of solvent evaporation and/or percolation. Thereby, the reactions were carried out in packed bed reactor using solvent-free medium under the following molar ratios: 1:3, 1:4 and 1:6 (acid: alcohol). The best performance (56% yield, 215.88 g L-1 of isopropyl palmitate and 26.87 gL-1.h-1 of productivity) was attained running the reactor with substrate at molar ratio of acid to alcohol of 1:4 for 8 hours with recycle rate of 1,5 mL.min-1. The use of a molecular sieve in the substrate reservoir resulted in an increase of approximately 7% of ester formation. By comparison, esterification run under the same experimental condition (1:4 in solvent-free medium) was carried out in a stirred tank reactor and slight higher productivity (33.40 g.L-1.h-1) was achieved. However, due the shear limitation of the support, it was concluded that the most suitable configuration for this bioprocess is the packed bed bioreactor, thereby showing that the enzymatic process is feasible using lipase immobilized on a magnetized hybrid support for isopropyl palmitate synthesis.

Recentemente, tem havido indícios de que o ser humano passou a compreender de forma mais profunda que o meio ambiente e o responsável pelo oferecimento das condições essenciais a sua sobrevivência e evolução, e a busca por alternativas energéticas mais adequadas a essa nova forma de pensar e viver tem sido parte integrante da solução para os problemas socioambientais que se mostram cada vez mais urgentes. A produção de biodiesel tem sido tema de pesquisas intensivas e o aumento no conhecimento sobre os efeitos danosos ao ambiente do uso de combustíveis fosseis como fonte de energia tem motivado a busca da viabilização do uso de biocombustíveis. O presente trabalho teve como objetivo aprimorar a tecnologia de síntese enzimática de biodiesel pela rota etílica. Utilizou-se como proposta a transesterificação do óleo de babaçu com etanol mediada pela enzima lipase em fluxo continuo empregando reator de leito fluidizado, visando aumentar a produtividade do sistema, bem como obter biodiesel com propriedades adequadas a sua utilização como biocombustível. Todos os experimentos foram realizados na ausência de solventes a 50 °C, utilizando como catalisadores a lipase microbiana de Burkholderia cepacia imobilizada em matriz de SiO2-PVA e Candida antarctica adsorvida em resina de metil-metacrilato (NovozymR 435). O reator de leito fluidizado (coluna de vidro: 12 mm × 375 mm) com recirculação foi construído e as características hidrodinâmicas foram determinadas por meio da distribuição do tempo de residência (DTR) usando teste de traçador. Para ambos biocatalisadores foram investigados os efeitos da razão molar óleo e etanol (1:5 a 1:12), tempo espacial (6 a 14 h) e carregamento de lipase (3 a 23 gramas) no rendimento de transesterificação e produtividade. Grande excesso de etanol (razão molar 1:12) e a incorporação de uma coluna empacotada com LewatitR GF202 para remover o glicerol formado como subproduto foram condições essenciais para manter uma operação estável do reator para ambos os biocatalisadores, apesar dos diferentes desempenhos obtidos. Empregando B. cepacia imobilizada em SiO2-PVA, a eficiência do reator foi limitada mesmo utilizando elevado carregamento de lipase (22,3 g). Valores máximos de rendimento de transesterificação [(53,0 } 4,8)%] e produtividade [(9,3 } 0,8)×10-6 molester/gcat/min] foram alcançados no tempo espacial de 9,3 h, revelando um tempo de meia-vida do biocatalisador de 18 dias. A viabilidade do FBR foi demonstrada usando NovozymR 435 e um desempenho superior foi alcançado com apenas 5 gramas de biocatalisador e tempo especial de 8 h, fornecendo rendimentos médios de transesterificação de (98,1 } 2,4)% e produtividade de (9,9 } 0,2)×10-5 molester/gcat/min. Nessas condições as propriedades da amostra de biodiesel foram: teor de éster de 97,1%; massa especifica a 20 °C de 872 kg/m3; viscosidade cinemática a 40 °C de 4,86 mm2/s; 1,4% em massa de monoacilglicerol e 1,0% em massa de diacilglicerol. Outros parâmetros foram estimados por modelos empíricos, visando a predição da qualidade do biodiesel do óleo de babaçu: 64 para o número de cetano; 257 para o índice de saponificação; 17 gI2/100g para o índice de iodo; e 8,5 °C negativos para o ponto de entupimento de filtro a frio. Os resultados obtidos foram bastante satisfatórios e contribuíram para o estabelecimento de condições reacionais e operacionais para a produção enzimática de biodiesel em reator de leito fluidizado, utilizando etanol e óleo de babaçu como materiais de partida em meio isento de solventes. Recently there has been evidence that humans began to understand more deeply that the environment is responsible for providing the essential conditions to their survival and evolution, and the search for suitable energy alternatives to this new way of thinking and living has been an integral part of the solution to the environmental problems that are increasingly urgent. In this context, the production of biodiesel has been the subject of intensive research, because although the prospects of oil shortages have been postponed several times due to the discovery of new reserves, issues related to political and social instability of major producer countries and the increase in knowledge about the harmful effects to the environment from the use of fossil fuels as an energy source have motivated the search for the feasibility of the use of biofuels. The present study aimed at improving the enzymatic technology to produce biodiesel by ethanolysis. The transesterification of babassu oil with ethanol mediated by lipase in a fluidized bed reactor (FBR) under continuous run was used as a proposal to increase the productivity of the system as well as to obtain biodiesel having suitable properties to be used as a biofuel. All experiments were carried out in solvent-free system at 50°C and microbial lipases from Burkholderia cepacia immobilized on SiO2-PVA hybrid matrix and Candida antarctica adsorbed on methyl-methacrylate resin (NovozymR 435) were used as biocatalysts. The FBR system (glass column: 12 mm × 375 mm) with recirculation was assembled and its hydrodynamic characteristics, such as the mean residence time and variance, were evaluated through the residence time distribution (RTD) employing a pulse input. For both biocatalysts the effects of oil-to-ethanol molar ratio (1:5 to 1:12), space time (6 to 14 h) and lipase loading (3 to 23 grams) in the transesterification yield and productivity were investigated. Great excess of ethanol (molar ratio oil-to-ethanol of 1:12) and the incorporation of a column packed with LewatitR GF202 to remove the glycerol formed as a byproduct were found to be essential to maintain a stable reactor operation for both biocatalysts, although different performances were attained. Using B. cepacia lipase immobilized on SiO2-PVA the reactor efficiency was limited even for high lipase loading (22.3 g). Maximum transesterification yield [(53.0 } 4.8)%] and productivities [(9.3 } 0.8)×10-6 molester/gcat/min] were achieved at space time of 9.3 h, revealing a biocatalyst half-life of 18 days. The feasibility of the FBR was demonstrated using NovozymR 435 and enhanced performance was attained using only 5 grams of biocatalyst at 8 h space time, which provided transesterification yield of 98.1 % and productivity of (9.9 } 0.2)×10-5 molester/gcat/min. Under such conditions the biodiesel properties were as follows: ester content of 97.1%, density at 20 °C of 872 kg/m3, kinematic viscosity at 40 °C of 4.86 mm2/s, 1.4 wt% of monoacylglycerol and 1.0 wt% of diacylglycerol. Other parameters were estimated by empirical models in order to predict the quality of biodiesel from babassu oil: 64 for the cetane number; 257 for the index of saponification, 17 g I2/100g for index iodine, and - 8.5 ° C for the cold filter plugging point. The results obtained were quite satisfactory and have contributed to the establishment of operating conditions for the enzymatic production of biodiesel in a fluidized bed reactor using ethanol and babassu oil as starting materials in a solvent-free reaction medium.

O presente projeto teve como objetivo selecionar componentes orgânicos para obtenção de compósitos híbridos pela técnica sol-gel utilizando como precursor tetraetilortossilicato (TEOS) visando avaliar o efeito do agente orgânico na evolução do sol para gel e nas propriedades finais dos suportes para imobilizar a lipase microbiana de Burkholderia cepacia. Foram testados três compostos orgânicos ?-ciclodextrina (?CD), carboximetilcelulose (CMC) e hidroxietilcelulose (HEC) e os resultados obtidos foram comparados com o comportamento já estabelecido para o polivinilálcool (PVA), tanto em termos morfológicos como atividade catalítica em meio aquoso (hidrólise de triacilgliceróis) e meio orgânico (síntese de éster de cadeia curta e longa). A motivação do estudo está diretamente relacionada com a elevada afinidade do suporte híbrido SiO2-PVA pelo glicerol, sendo uma das limitações do uso deste suporte para imobilizar lipases com finalidade de aplicação em reações de transesterificação conduzidas em fluxo contínuo. As características morfológicas e estruturais dos suportes resultantes foram determinadas empregando técnicas convencionais, indicando similaridade das propriedades estruturais, fornecendo suportes com áreas superficiais entre 361,6 a 529,7 m2 g-1 e morfologia amorfa. A diferença marcante entre os suportes foi verificada quanto a capacidade de adsorção do subproduto da reação de transesterificação, sendo verificado que a substituição do polivinilálcool por ?-ciclodextrina reduziu em até 50% a habilidade do suporte hibrido em absorver glicerol. Os componentes derivados de celulose apresentaram resultados similares aos obtidos com o polivinilálcool. Em termos de afinidade pelo glicerol os resultados obtidos permitiram a classificação dos suportes híbridos na seguinte ordem crescente: SiO2-CMC > SiO2-PVA > SiO2-HEC > SiO2- ?CD. Quanto a afinidade dos suportes para imobilização da lipase de B. cepacia, os valores de atividade hidrolítica dos derivados imobilizados, nas condições adotadas, revelaram que todos os suportes tiveram o mesmo tipo de interação com a enzima resultando em valores de recuperação de atividade na faixa de 56 - 63%, valores próximos aos normalmente obtidos pela matriz de SiO2-PVA (68%). Os parâmetros cinéticos (Km e Vmax) determinados indicaram que a lipase imobilizada nos diferentes tipos de suporte, apresentou afinidade semelhante pelo substrato azeite de oliva. Os valores determinados de Km e Vmax da lipase de B. cepacia na forma livre (Km = 410 mM e Vmax =12391 Ug-1) mostraram que independente do suporte, a imobilização reduziu aproximadamente 60% da afinidade da enzima pelo substrato. Em meio orgânico, tanto as reações de esterificação como de transesterificação mediadas pelos derivados imobilizados apresentaram desempenho similar com atividade de esterificação da ordem de 118 ?M.g-1min-1 e rendimentos de formação de ésteres etílicos da ordem de 98,30%. De uma forma geral, os resultados obtidos foram promissores e permitiram selecionar o composto orgânico ?CD para substituir com vantagens o polivinilálcool no preparo de matrizes hibridas para imobilização da enzima lipase. O menor poder hidrofílico desta matriz em relação às outras matrizes testadas pode ser creditado a estrutura cíclica da ?CD que reduziu a capacidade do suporte SiO2-?CD em adsorver glicerol, principal subproduto da reação de transesterificação. Testes em processos conduzidos em fluxo contínuo são sugeridos para comprovação da eficiência dessa matriz. The objective of this work was to select organic compounds for preparing hybrid composites by the sol-gel technique using tetraethylorthosilicate (TEOS) as precursor aiming at evaluating the effect of the organic agent in the transformation of the sol into gel and in the final properties of the matrices for immobilizing lipase from Burkholderia cepacia. Three organic components ?-cyclodextrin (?CD), carboxymethylcellulose (CMC) and hydroxyethylcellulose (HEC) were tested. The organic component polyvinyl alcohol (PVA) was used as control taking into consideration its morphological properties and the catalytic activity of the immobilized derivative in aqueous medium (triacylglycerols hydrolysis) and organic medium (synthesis of short and long chain esters). The motivation of this work was directly related with the high affinity of the matrix polysiloxane-polyvinyl alcohol (SiO2- PVA) by the glycerol, limiting the use of the immobilized derivative in the transesterification reactions running on a continuous flow. The morphological and structure properties of the resulting matrices was determined using conventional techniques, showing similar structure properties, with surface area values between 361.6 and 529.7 m2g-1 and amorphous morphology. The main difference among the matrices were observed regarding their ability to adsorb glycerol, the main byproduct of the transesterification reaction. It was verify that the replacement of the polyvinyl alcohol by ?-cyclodextrin reduced in about 50% the hybrid composite capacity to adsorb glycerol. On the other hand, the cellulose components showed similar results as the polyvinyl alcohol. Considering as an evaluated parameter the glycerol affinity, the achieved results allowed classifying the hybrid composites as follows: SiO2- CMC > SiO2-PVA > SiO2-HEC > SiO2-?CD. The performance of the hybrid composites for immobilizing lipase from B. cepacia, through the hydrolytic activity reveal that all the matrices show the same interaction with the lipase, resulting in activity recovered values between 56-63%, which is close to the value attained by SiO2-PVA matrix (68%). The kinetic parameters (Km and Vmax) values determined for free lipase (Km = 410 and Vmax = 12391 Ug-1), showed that independely of the immobilization support the affinity of the enzyme for the substrate was reduced by 60%. In organic medium, all the immobilized derivatives showed similar behavior on both esterification and transesterification reactions attaining esterification activity of 118 ?M.g-1min-1 and ethyl ester yields of 98.30%. Overall, the results were promising and allowed selecting the organic component ?CD to replace, with advantages, the use of PVA for preparing hybrid composites. The lower hydrophilic capacity of this matrix compared with the others matrices can be explained by the cyclic structure of the ?CD that reduced the capacity of the SiO2-?CD matrix in the glycerol adsorption, the main byproduct of the transesterification reaction. Assays carried out in packed bed reactor running on a continuous basis are propose to confirm the efficiency of this matrix.

O efeito de diferentes agentes antioxidantes naturais lipossolúveis (óleo de copaíba, manteiga de tucumã, manteiga de cacau, óleo de buriti e especiarias como orégano, tomilho branco, alecrim, sálvia sclarea, cravo talo e cananga) foi determinado na síntese enzimática de monoglicerídeos (MAG) a partir da glicerólise do óleo de babaçu, tomando como parâmetro de comparação os resultados alcançados na reação isenta de estratégia antioxidante (controle negativo) e sob atmosfera inerte (controle positivo). As reações foram conduzidas em reatores operando em regime descontínuo e contínuo usando a lipase de Burkholderia cepacia imobilizada em SiO2-PVA como catalisador. O melhor desempenho em reatores descontínuos foi obtido usando N2 no meio reacional (60 % MAG) seguido do óleo de buriti (57,6 % MAG) e manteiga de cacau (56,6 % MAG). Entretanto, a incorporação do óleo de buriti no meio reacional alterou o perfil de formação de monoglicerídeos, ocasionando elevada concentração de monoleina. Tomando por base esses resultados, selecionou-se a manteiga de cacau como agente antioxidante para a condução dos testes em fluxo contínuo. Nesse contexto, visando estabelecer maior homogeneidade do meio reacional e alcançar rendimentos elevados de MAGs, foram realizados testes em reator de leito fixo em fluxo contínuo, sob atmosfera inerte, variando-se a proporção entre óleo e glicerol no meio de alimentação. Os resultados obtidos nesta série de experimentos demonstraram forte influencia da razão molar dos materiais de partida na síntese de MAGs e o melhor desempenho foi obtido empregando a razão molar intermediária (1:9), que proporcionou a formação de 31,5 % de MAGs e produtividade de 52,2 mgMAGs/gamostra.h. Testes empregando manteiga de cacau como agente antioxidante foram efetuados e desempenhos similares daqueles obtidos em atmosfera inerte foram obtidos, confirmando o efeito benéfico da utilização da manteiga de cacau no meio de alimentação como agente antioxidante. Isto sugere que a manteiga de cacau inibiu ou reduziu a oxidação do óleo de babaçu durante a síntese de MAGs, indicando a possibilidade de substituição da atmosfera inerte (N2) na reação. Análises espectrofotométricas dos produtos de oxidação primários (hidroperóxidos e dienos conjugados) e secundários (aldeídos e cetonas) validaram a eficiência da manteiga de cacau como agente antioxidante da matéria-prima lipídica durante todo o processo contínuo. The effects of several natural antioxidants (copaiba oil, buriti oil, cocoa butter, tucuman butter, oregano, white thyme, rosemary, salvia sclarea, carnation stem and cananga) were assessed in the enzymatic synthesis of monoglycerides (MAG) from the glycerolysis of babassu oil. The reactions were catalyzed by Burkholderia cepacia lipase immobilized on SiO2-PVA and the assays carried out in batch and continuous runs. Results were compared with those attained in the control reactions (without any strategy to avoid oxidation and under inert atmosphere), and the best approach was tested in a continuous packed bed reactor. The best performance was achieved using N2 in the reaction medium (60 % of MAG) followed by buriti oil (57.6 % of MAG) and cocoa butter (56.6 % of MAG), preventing the oxidation of babassu oil in batch reaction. However, the incorporation of buriti oil in the medium influenced the MAG profile, leading to the largest formation of monoolein, unlike other runs. Based on these results, cocoa butter was chosen as antioxidant agent to carry out the assays under continuous flow. Therefore, aiming to establish greater homogeneous reaction media and to attain high MAGs yields, assays were performed in packed bed reactor under continuous flow, in inert atmosphere, varying the proportional of babassu oil and glycerol in the feed media. The results obtained in this set of runs demonstrated a strong influence of molar ratio between starting materials in the synthesis of MAGs and the best performance was achieved using the intermediate molar ratio (1:9), which afforded the formation of 31.5% of MAGs and productivity of 52.2 mgMAGs/gsample.h. Tests using cocoa butter as antioxidant were made and similar results were attained, confirming the beneficial effect of using cocoa butter in the feed medium as an antioxidant agent. This suggests that the cocoa butter inhibited or reduced babassu oil oxidation during synthesis of MAGs, indicating the possibility of replacing the inert atmosphere (N2) in the reaction. Spectrophotometric analysis of the primary oxidation products (conjugated dienes and hydroperoxides) and secondary (aldehydes and ketones) validated the efficiency of cocoa butter as an antioxidant agent of lipid a continuous process. Thereby, among the nine tested antioxidant agents, cocoa butter was the most effective in both systems, since it not interfere in the MAG profile and also reduced the cost of the process.

O desenvolvimento sustentável é atualmente um dos maiores focos das pesquisas no mundo em virtude dos impactos ambientais, tais como aquecimento global, geração de resíduos e emissão de gases poluentes causados pelo uso de combustíveis fósseis. Deste modo, pesquisas têm sido concentradas em tecnologias que permitam a substituição de refinarias à base de petróleo por biorrefinarias que utilizam matérias-primas renováveis. Neste contexto, os fungos filamentosos surgem como um recurso promissor no desenvolvimento de novos produtos sustentáveis, entre os quais os fungos pertencentes ao filo Zigomiceto, contribuem significativamente para esse desenvolvimento e estão sendo extensivamente estudados para a aplicação em biorrefinarias, com destaque especial para os fungos do gênero Mucor. Esse gênero, particularmente da espécie Mucor circinelloides, é um potencial produtor da enzima lipase e de biomassa com quantidades significativas de lipídios (single cell oil) o que permite sua exploração no processo de produção de biodiesel. Desta forma, o objetivo deste trabalho foi explorar importantes aplicações biotecnológicas da linhagem de Mucor circinelloides URM 4182, tanto para produção de lipases quanto de lipídios. O trabalho experimental realizado consolidou dados da potencialidade do fungo para produção de lipase ligada ao micélio e sua utilização como biocatalisador para obtenção de biodiesel partir de óleos vegetais alternativos, como andiroba, coco, macaúba, palma e pinhão manso. Nas condições estudadas, foram obtidos rendimentos de transesterificação entre 88% a 97% e as conversões mais elevadas foram alcançadas para os óleos láuricos sugerindo seletividade da lipase para ácidos de cadeia curta. A potencialidade da linhagem Mucor circinelloides URM 4182 foi comprovada na obtenção de lipídios em cultivos efetuados em biorreator utilizando glicose e milhocina, respectivamente, como fontes de carbono e de nutrientes alternativos aos suplementos sintéticos. Nessas condições, valores médios de produtividade biomassa (3,10 ± 0,01 g/L/dia) contendo (31 ±0,01 % m/m) de lipídios, correspondendo produtividade lipídica de (0,97 ± 0,01 g/L.dia) foram alcançados. A composição do óleo microbiano revelou elevados teores dos ácidos graxos saturados palmítico (C16:0) e esteárico (C18:0) e dos insaturados oleico (C18:1) e linoleico (C18:2), que são os ácidos graxos semelhantes aos dos óleos vegetais convencionais utilizados para a produção de biodiesel. Além disso, foram verificados teores consideráveis do ácido gama-linolênico (GLA - C18:3), que tem relevante importância nas indústrias farmacêutica e de alimentos. A esterificação e transesterificação simultâneas do óleo microbiano ou diretamente da biomassa celular com etanol mediada por catalisadores heterogêneos bioquímicos e químicos forneceram amostras de biodiesel com elevados teores de ésteres de etila (> 97%) que atendem as normas de qualidade para uso como biocombustível. Sustainable development is now the major research focus in the world because of the environmental impacts, such as global warming, waste generation and greenhouse gas emissions caused by the use of fossil fuels. Thus, research has been focused on technologies that enables the replacement of petroleum based refineries by biorefinery based on renewable raw materials. In this context, filamentous fungi emerge as a promising resource in the development of new sustainable products, including the fungi belonging to the phylum Zigomicete which contribute significantly to this development and are being extensively studied for using in biorefineries, with particular emphasis on the Mucor sp. fungus genus. This genus, particularly from the species of Mucor circinelloides, is a potential producer of the enzyme lipase and biomass having significant amounts of lipids (single cell oil) which allows its exploitation in the biodiesel production. Thus, the aim of this study was to explore important biotechnological applications of a Brazilian strain of Mucor circinelloides URM 4182 for both production of lipases and lipids. The experimental work consolidated this fungus capability to produce mycelium bound lipase and its use as biocatalysts for biodiesel production from alternative vegetable oils such as andiroba, coconut, macaw palm, palm and jatropha. The attained transesterification yields were in the range from 88 to 97%, and the highest conversions were achieved for lauric oils suggesting that M. circinelloides lipase has high selectivity for short chain fatty acids. The capability of this strain was also proven to produce lipids at cultivation conditions established in this work using glucose and corn steep liquor, respectively, as carbon sources and alternative nutrients to the mineral supplements. Under these conditions, average values for biomass productivity (3.10 ± 0.01 g/L/day) containing high lipis levels (31.2 ± 0.01% m/m) and lipid productivity of 0.97 ± 0.01 g/L.dia were achieved. The composition of microbial oil revealed high levels of saturated fatty acids such as palmitic acid (C16:0) and stearic (C18:0) and unsaturated oleic (C18:1) and linoleic (C18:2), which are fatty acids similar to conventional vegetable oils used for the production of biodiesel. In addition, considerable levels of gamma-linolenic acid (GLA-C18:3) were also verified which has relevant importance in the pharmaceutical and food industries. The simultaneous esterification and transesterification from microbial oil or direct fungal biomass with ethanol mediated by heterogeneous catalysts (biochemical and chemical) provided samples with high levels of ethyl esters (> 97%) that meet the quality standards for using as biofuel.

O objetivo deste trabalho foi contribuir no desenvolvimento de técnicas para obtenção de biocatalisadores imobilizados de baixo custo e alto poder catalítico para ser empregado na síntese de biodiesel a partir de óleos vegetais pela rota etílica. Baseia-se na utilização de células íntegras de fungos filamentosos com elevada atividade lipolítica (glicerol éster hidrolase - E.C. 3.1.1.3) imobilizadas em suporte adequado. O trabalho experimental foi desenvolvido em diferentes etapas e iniciado pela triagem de linhagens de fungos adquiridas de banco de cultura nacional, com habilidade de produzir lipase intracelular em cultivo submerso. Mediante a avaliação da atividade lipolítica do micélio, foram selecionadas as linhagens dos fungos: Rhizopus oryzae (3231 e 4692), Mucor circinelloides (4140 e 4182) e Penicillium citrinum (4216) e efetuados testes de imobilização por adsorção física (in situ e extra-situ) da biomassa produzida, utilizando os seguintes suportes: espuma de poliuretano, Celite e polihidroxibutirato. Nas condições testadas, o melhor desempenho foi alcançado pela matriz de poliuretano que permitiu a retenção de elevada concentração de biomassa celular para todas as linhagens selecionadas, exceto para fungo de P.citrinum (4216). Testes adicionais de imobilização da biomassa celular na espuma de poliuretano foram efetuados, complementando desta forma, as propriedades físicas e morfológicas desse suporte e do sistema imobilizado. Os resultados indicaram que espumas de poliuretano cortadas em cubos de 6 mm foram mais adequadas para promover completa adsorção da biomassa celular produzida pelos fungos selecionados, em especial do fungo M. circinelloides 4182, que apresentou elevado poder catalítico na reação de transesterificação. Em virtude desse fato, as condições de cultivo dessa linhagem foram estudadas, mediante variação das fontes de carbono e nitrogênio, aeração e inóculo. O biocatalisador obtido nas melhores condições (atividade lipolítica= 65 U/g) foi utilizado na etanólise do óleo de babaçu e o processo foi otimizado por meio de um delineamento fatorial, avaliando-se a influência da temperatura (33 - 47C) e da razão molar entre óleo de babaçu e etanol (1:4,8 - 1:13,2) no rendimento de transesterificação. As condições otimizadas preditas pelo planejamento fatorial foram: temperatura de 35°C e razão molar etanol/óleo de 6:1, obtendo-se rendimento de 90,5% empregando 20% m/m de biocatalisador e terc-butanol como solvente. A estabilidade operacional do biocatalisador foi avaliada em reator operando em regime de bateladas consecutivas e em regime contínuo. Resultados satisfatórios foram obtidos em ambos os sistemas, entretanto o sistema contínuo evitou a dessorção do micélio do suporte, revelando tempo de meia-vida do biocatalisador superior a 20 dias. Finalmente, a influência de óleos vegetais contendo diferentes composições em ácidos graxos (andiroba, macaúba, pinhão manso e palma) foi avaliada na reação de etanólise, sendo constatado a preferência do biocatalisador na transesterificação de ácidos graxos de cadeia média, presentes nos óleos de babaçu e macaúba. De forma geral, os resultados obtidos foram promissores e demonstraram a potencialidade de células íntegras de fungos filamentosos imobilizadas na síntese de biodiesel, com destaque para o desempenho obtido pelo fungo Mucor circinelloides 4182. The objective of this work was contributed in developing techniques to obtain immobilized biocatalysts with low cost and high catalytic activity to be applied in the biodiesel synthesis by ethanol route. It is based on the utilization of filamentous fungi whole cells with high lipase activity (glycerol ester hydrolase - E.C.3.1.1.3) immobilized in suitable support. The experimental work was developed in different ways and started by screening fungi strains acquired from National Culture Collection which has the ability to produce intracellular lipase in submerse cultivation. Through lipase activity the fungi strains: Rhizopus oryzae (3231 and 4692), Mucor circinelloides (4140 e 4182) and Penicillium citrinum (4216) were selected. Tests to evaluate the biomass immobilization by physical adsorption (in situ and extra-situ) using the supports: polyurethane foam, Celite and polyhidroxybutyrate were carried out. The best performance was attained by polyurethane matrix that allowed attain high biomass concentration for all strain fungi, except for P. citrinum (4216). Additional tests were performed to complement the physical and morphological properties of support and immobilized system. Results indicated that polyurethane foam cut in cubes 6-mm were suitable to provide full adsorption of biomass from all selected fungi strains, specially strain of M. circinelloides 4182 displayed high catalytic activity in the transesterification reaction. Consequently, cultivation conditions of this strain were studied, evaluating the carbon and nitrogen sources, aeration and inoculum level. The biocatalyst obtained under the best conditions (lipase activity = 65 U/g) was used in the ethanolysis of babassu oil and the process was optimized by experimental design. The influence of temperature (33 - 47?C) and ethanol to oil molar ratio (4.8:1 - 13.2:1) were investigated in the transesterification yield. Under the optimized conditions (35°C and oil to ethanol molar ratio 6:1) transesterification yield of 90.5% was obtained employing 20% wt. of biocatalyst and tert-butanol as solvent. The biocatalyst operational stability was assessed under consecutive batch runs and continuous system. Satisfactory results were attained in both systems, however, the continuous system avoided cells desorption from support, revealing biocatalyst half life higher than 20 days. Finally, the influence of vegetable oils with different fatty acid compositions (babassu, andiroba, macauba, Jatropha, palm) was studied in the ethanolysis reaction, being evidenced the specificity of the biocatalyst to transesterify medium chain fatty acids as presented in babassu and macauba oils. The results obtained were promise and demonstrated the potential of filamentous fungi whole cells immobilized as biocatalysts in the biodiesel synthesis, with prominence to the performance of Mucor circinelloides 4182 strain.

O presente trabalho teve como objetivo estudar a síntese do biodiesel por transesterificação etílica de diferentes matérias-primas lipídicas empregando catalisadores heterogêneos (químico e bioquímico). Para cumprir com os objetivos propostos foram selecionadas matérias-primas lipídicas de baixo impacto no setor alimentício, entre as quais destacam-se: óleos vegetais (andiroba, babaçu, macaúba, palma e pinhão manso) e gordura residual (sebo bovino) e catalisadores de comprovada potencialidade como óxido de nióbio impregnado com sódio (químico) e a lipase de Burkholderia cepacia imobilizada em suporte híbrido sílicaalcool polivinílico (bioquímico). O trabalho foi desenvolvido em três etapas. Inicialmente, foram determinadas as propriedades físico-químicas das diferentes matérias-primas lipídicas, algumas ainda pouco exploradas, para verificar se apresentavam potencial para serem utilizadas na reação de transesterificação. Em seguida foram preparados os catalisadores propostos por protocolos já estabelecidos, sendo obtido para o catalisador químico elevado teor de sódio impregnado no óxido de nióbio (25,43 ± 0,29%) e para catalisador bioquímico elevada atividade hidrolítica (1814 ??281 U/g). Na segunda etapa, as reações de transesterificação foram conduzidas em regime de batelada em condições adequadas para cada catalisador em termos de temperatura, tempo e proporção de catalisador. Na terceira etapa, os produtos obtidos foram purificados e quantificados por cromatografia gasosa, RMN 1H, viscosimetria e análise termogravimétrica (TGA). O conjunto de dados obtidos demonstrou que a formação de ésteres etílicos a partir das diferentes matérias-primas é viável para os catalisadores testados. Ambos os catalisadores (químico e bioquímico) atuaram de forma eficiente convertendo os ácidos graxos presentes nas matérias-primas lipídicas nos ésteres etílicos correspondentes e apresentaram elevada estabilidade em bateladas consecutivas, com destaque para o catalisador bioquímico que revelou um tempo de meiavida de 290 h. Entretanto, a qualidade da matéria-prima lipídica interferiu a atuação dos catalisadores de maneira diferenciada. Enquanto, o catalisador químico foi sensível a presença de níveis elevados de acidez, como o constatado no óleo de macaúba, a atuação do catalisador bioquímico sofreu influência da presença de peróxidos indicativo de oxidação apresentada pelo óleo de andiroba. Com exceção dos óleos de macaúba e andiroba que apresentaram qualidade comprometida, todas as matérias-primas lipídicas originaram amostras de biodiesel com características adequadas para serem usadas como combustível, incluindo valores de viscosidade entre 3,9 a 6,0 (cSt) que atendem as especificações estabelecidas na ASTM 6751- 02. Apesar do desempenho similar dos catalisadores testados, a via química foi superior em termos de produtividade em relação à via bioquímica. Entretanto, essa baixa produtividade pode ser incrementada utilizando métodos não convencionais de aquecimento, como por exemplo, irradiação de micro-ondas e ultrassom. Os resultados obtidos neste trabalho demonstram ainda que os catalisadores heterogêneos testados possuem potencial para substituir os sistemas homogêneos normalmente empregados na síntese do biodiesel. Essa substituição oferece vantagens, que podem propiciar um aumento considerável nas perspectivas de sustentabilidade sócio-ambiental de todo o processo de produção. The present work aimed at studying the biodiesel synthesis by transesterification reaction from several lipidic feedstocks via ethyl route employing heterogeneous catalysts (chemical and biochemical). To attain the proposed objectives non-edible feedstock having low impact in the food segment, among which stand out: vegetable oils (andiroba, babassu, macauba, palm and Jatropha curcas) and residual fat (beef tallow) and potential catalysts as niobium oxide impregnated with sodium (chemical) and lipase from Burkholderia cepacia immobilized on silica-polyvinyl alcohol matrix (biochemical) were previously selected. The work was developed in three steps. Initially, the physico-chemical properties of the different lipidic feedstocks were investigated, some of them still little explored, to identify their potential as reactants in the transesterification reactions. Then the proposed catalysts were prepared by protocols already established, being obtained for the chemical catalyst high level of sodium impregnated in the niobium oxide (25.43 ± 0.29%) and for the biochemical catalyst high hydrolytic activity (1,814± 281 U/g). In the second step, the transesterification reactions were carried out in batch reactors under appropriated conditions for each catalyst in terms of temperature, time and catalyst proportion. In the third step, the obtained products were purified and quantified by gas chromatography, 1H NMR spectroscopy, viscosimetry and thermogravimetric analysis (TGA). The dataset obtained demonstrated that the formation of ethyl esters from the different feedstocks was feasible for the tested catalysts. Both catalysts (chemical and biochemical) were efficient in converting all fatty acids present in the lipidic feedstock into the corresponding ethyl esters and showed high stability under consecutive batch runs, with emphasis for the biochemical catalyst with a half-life time of 290 h. However, the poor quality of the feedstocks strong affected the performance of the catalysts in a different way. While the chemical catalyst was sensitive to high acidity levels, as verified in the macauba oil, the biochemical catalyst performance was influenced by the presence of peroxides indicating oxidation as showed in the andiroba oil. Except for these oils, all the other feedstocks originated biodiesel samples with appropriate characteristics to be used as fuel, including viscosity values between 3.9 to 6.0 (cSt) that are in accordance with specifications recommended by ASTM 6751-02. Even though the catalysts showed similar performances, the chemical route gave higher productivity than that attained by biochemical route. However, such lower productivity can be increased using non conventional heating systems as for instance, micro-wave irradiation and ultrasound. The results obtained in this work demonstrated that the selected heterogeneous catalysts possess potential to replace the homogeneous systems usually employed in the biodiesel synthesis. Such replacement offers advantages that can provided a considerable increase in the perspectives to attain an environmental sustainability of process as a whole.

O presente projeto teve como objetivo estudar a aplicação de campos eletromagnéticos de alta freqüência (micro-ondas) em processos enzimáticos, particularmente na síntese enzimática de biodiesel a partir da etanólise de óleos vegetais em meio isento de solvente. A enzima indicada para esse tipo de processo - lipase (EC 3.1.1.3) foi usada na forma imobilizada e os ensaios foram realizados utilizando óleos vegetais de baixo custo (babaçu e palma) e etanol como fontes de matérias-primas. Para cada óleo vegetal foi usada uma específica preparação de lipase, tomando por base resultados anteriormente obtidos. Para o óleo de babaçu foi utilizada a lipase de Burkholderia cepacia-PS, enquanto para o óleo de palma foi empregada a lipase de Pseudomonas fluorescens-AK. O trabalho experimental foi desenvolvido em três etapas. Na primeira etapa, a lipase de Burkholderia cepacia-PS foi imobilizada em dois suportes não comerciais, um inorgânico (Nb2O5) e outro inorgânicoorgânico (híbrido, SiO2-PVA) visando determinar o suporte mais adequado para imobilizar esta específica preparação de lipase. Os derivados imobilizados resultantes foram caracterizados quanto suas propriedades bioquímicas (pH e temperatura), cinéticas (Km e Vmax); estabilidade térmica (60 ?C) e posteriormente utilizados na reação de interesse. Os resultados obtidos indicaram que o derivado imobilizado na matriz híbrida (SiO2-PVA) foi mais estável termicamente e mais eficiente para mediar a reação de etanólise do óleo de babaçu, alcançando 90,1% de rendimento em 24 h. Esta matriz também foi utilizada para imobilizar a lipase de Pseudomonas fluorescens-AK. Na segunda etapa do trabalho, foram realizados testes de estabilidade térmica das lipases imobilizadas em SiO2-PVA no reator de micro-ondas (Discover/University-Wave, Cem Corporation). Os testes foram realizados a 60?C em meio orgânico, revelando tempos de meia-vida da ordem de 5,63 e 8,22 h, respectivamente para Burkholderia cepacia e Pseudomonas fluorescens. Na última etapa, buscou-se por meio da utilização de um método estatístico, determinar as condições adequadas de transesterificação para cada óleo vegetal. Esta etapa foi realizada por meio de uma matriz de planejamento fatorial, avaliando simultaneamente a influência da temperatura e razão molar na síntese do biodiesel. Para a etanólise do óleo de babaçu, a formação dos ésteres etílicos foi fortemente influenciada pelas variáveis quadráticas (X12, X22), razão molar e temperatura, respectivamente. O modelo matemático proposto permitiu prever as condições necessárias que favorecem o alcance de elevados rendimentos da produção de biodiesel, sendo a reação maximizada (100%) em 10h de reação para meios reacionais constituídos de uma razão molar de 1:12 (óleo/etanol) a 50 ºC. Para a etanólise do óleo de palma, a formação dos ésteres etílicos foi fortemente influenciada pelas variáveis lineares (X1, X2), razão molar e temperatura, respectivamente. O modelo matemático proposto indicou que os níveis baixos temperatura (43°C) e razão molar 1:8 (óleo/etanol) conduzem a uma conversão de 97% dos ácidos graxos presentes no óleo de palma em ésteres etílicos em apenas 12 h de reação. Os resultados obtidos indicam que o desenvolvimento de reações enzimáticas assistidas por micro-ondas constitui um procedimento potencial para a produção de biodiesel tendo em vista que foi possível diminuir os tempos de reação, demonstrando a viabilidade do processo enzimático proposto para ambos óleos vegetais, com vantagens adicionais em relação ao processo submetido ao aquecimento convencional. The objective of this project was to study the application of electromagnetic fields of high frequency (microwave) in enzymatic process, especially in the synthesis of biodiesel by the ethanolysis of vegetal oils in a solvent free system. The indicated enzyme for this process - lipase (EC 3.1.1.3) was used in an immobilized form and the assays were carried out using low cost vegetal oils (babassu oil and palm oil) and ethanol as raw material sources. For each vegetal oil a specific lipase preparation was used based on previous results. For babassu oil lipase from Burkholderia cepacia-PS was used while for palm oil lipase from Pseudomonas fluorescens-AK. The experimental work was developed in three steps. In the first, lipase from Burkholderia cepacia-PS was immobilized on two non-commercial matrixes, as inorganic matrix (niobium oxide, Nb2O5) and hybrid matrix (polysiloxane-polyvinyl alcohol, SiO2- PVA) aiming to determine the most suitable matrix to immobilize this specific preparation of lipase. The resulting immobilized derivatives were analyzed regarding their biochemical properties (pH and temperature), kinetic properties (Km and Vmax) and thermal stability (60oC) following their applications in the reaction proposed. The results obtained indicated that the immobilized derivate on hybrid matrix (SiO2-PVA) was more thermally stable and efficient to mediate the ethanolysis reaction of babassu oil, reaching a yield of 90.1% in 24 h. This matrix was also used to immobilize lipase from Pseudomonas fluorescens-AK. In the second stage of work, thermal stability tests of the immobilized lipases on SiO2-PVA under microwave irradiation were performed using a microwave reactor (Discover/University-Wave, Cem Corporation). The tests were performed at 60oC in organic medium, revealing half-life times in the order of 5.65 and 8.22h, respectively for Burkholderia cepacia and Pseudomonas fluorescens. In the last stage, conditions to carry out the transesterification for each vegetable oil were determined, using a statistic method. This stage was conducted by a factorial design matrix, attaining the influence of temperature and molar ratio in biodiesel synthesis. For the ethanolysis of babassu oil, the formation of ethyl esters was strongly influenced by quadratic variables (X12, X22), molar ratio and temperature, respectively. The statistical model allowed predicting the adequate conditions that could reach high yields of biodiesel production, maximizing the reaction (100%) in 10h for reaction media consisting of a molar ratio of 1:12 (oil/ethanol) at 50oC. For ethanolysis of palm oil, the formation of ethyl ester was highly influenced by linear variables (X1, X2), molar ratio and temperature, respectively. The proposed statistic model indicated that the low levels of temperature (43oC) and ratio molar 1:8 (oil/ethanol) conduct to 97% conversion of fatty acid present on palm oil in ethyl esters in 12h of reaction. The obtained results indicated that the development of enzymatic reactions conducted by microwaves constitutes a potential procedure for the production of biodiesel in view that it was possible to reduce the reaction time, demonstrating the viability of the considered enzymatic process for both vegetal oils, with advantages adds in relation to the submitted process to the conventional heating.
