Extravasation of T cells from the bloodstream into inflamed tissues requires interactions between T cells and vascular endothelial cells, a necessary step that allows T cells to exert their effector function during the immune response to pathogens and to sterile insults. This cellular cross talk involves adhesion molecules on both the vascular endothelium and the T cells themselves that function as receptor-ligand pairs to slow down circulating T cells. These will eventually extravasate into sites of inflammation when they receive the correct chemokine signals. Accumulation of T cells within the vascular wall can lead to vessel thickening and vascular disease, whereas T-cell extravasation into the myocardium often leads to cardiac chronic inflammation and adverse cardiac remodeling, hallmarks of heart failure. On the flip side, T-cell trafficking is required for pathogen clearance and to promote tissue repair after injury resulting from cardiac ischemia. Thus, a better understanding of the central players mediating these interactions may help develop novel therapeutics to modulate vascular and cardiac inflammation. Here, we review the most recent literature on pathways that regulate T-cell transendothelial migration, the last step leading to T-cell infiltration into tissues and organs in the context of vascular and cardiac inflammation. We discuss new potential avenues to therapeutically modulate these pathways to enhance or prevent immune cell infiltration in cardiovascular disease.

Introduction: Diastolic dysfunction in Heart Failure with Preserved Ejection Fraction (HFpEF) is associated with T cell systemic inflammation and downregulation of myocardial unfolded protein response (UPR) genes. The T cell UPR can modulate T cell effector function and immune responses, yet this is largely unexplored in HFpEF. Hypothesis: We hypothesized T cells contribute to cardiometabolic HFpEF and that metabolic and nitrosative stress-induced alterations of the T cell UPR modulate T cell activation and diastolic function. Methods: Male C57/BL6 (wild-type, WT) or T cell receptor alpha-deficient ( Tcra-/-) were fed a high-fat diet (HFD) and L-NAME for 1, 3, and 5 weeks, or standard chow (STD). We assessed diastolic function by invasive hemodynamic analyses. Immune cells were characterized in the heart and spleen by flow cytometry, and cardiac pathology was assessed by histology. The UPR gene expression was evaluated in splenic CD4 + T cells over time by qPCR. Results: Unlike WT mice, Tcra-/- mice fed HFD/L-NAME for 5 weeks did not develop diastolic dysfunction or cardiomyocyte hypertrophy, demonstrating a critical role for T cells in experimental cardiometabolic HFpEF. Indeed, cardiac CD4 + T cells were increased in WT mice fed HFD/L-NAME for 5 weeks compared to STD, concordant with an expansion of splenic CD44 hi CD62l lo interferon-gamma (IFNγ) + effector cells. Strikingly, splenic T cells also experienced downregulation of the UPR proteins activating transcription factor 4 ( Atf4 ), Atf6 , and X box-binding protein 1 ( Xbp1s/u ). We found that the Atf4 arm of the T cell UPR is the earliest responder to HFD/L-NAME, with observed downregulation of Atf4 and its downstream effector C/EBP homologous protein ( Chop ) as early as 3 weeks, prior to onset of diastolic dysfunction, followed by the subsequent downregulation of Atf6 and Xbp1s/u . Conclusions: T cells contribute to diastolic dysfunction and cardiomyocyte hypertrophy in experimental cardiometabolic HFpEF. Early modulation of T cell Atf4 precedes diastolic dysfunction, enhanced IFNγ + effector T cell activation, and global T cell UPR downregulation. Ongoing studies are focused on understanding the functional consequences of impaired T cell UPR in cardiometabolic HFpEF.

SUMMARYLyme disease is a tick-borne disease caused by bacteria of the genus Borrelia. The disease can initially manifest as an erythema migrans rash and, if able to evade the host immune defenses, can progress into a secondary stage chronic disease with debilitating physical or neurological manifestations1,2. The host factors that modulate susceptibility for Lyme disease have remained mostly unknown. Here we show a novel host defense mechanism against Lyme disease in humans. Using epidemiological and genetic data from FinnGen, we identify a common missense variant at the gene encoding for Secretoglobin family 1D member 2 (SCGB1D2) protein that increases the susceptibility for Lyme disease. The genetic variant changes proline at position 53 to leucine and is predicted as deleterious. Consequently, we validate the dysfunction of this protein variant using live Borrelia burgdorferi (Bb). Recombinant reference SCGB1D2 protein inhibits the growth of Bb twice as effectively as the recombinant SCGB1D2 P53L deleterious missense variant. Together, these data suggest that SCGB1D2 is a host defense factor present in the skin, sweat, and other secretions which protects against Bb infection. This finding provides a novel therapeutic avenue for drug development to prevent and treat Lyme disease.

Recombinant adeno-associated virus (rAAV) vectors are increasingly popular gene delivery tools in biological systems. They are safe and lead to high-level, long-term transgene expression. rAAV are available in multiple serotypes, natural or engineered, which enable targeting to a wide array of tissues and cell types. In addition, rAAVs are relatively easily produced in a well-equipped lab or obtained from a viral vector core facility. Unfortunately, there is no standardization of quality control assays beyond titering and purity assessments. Next-generation sequencing (NGS) can be used to identify rAAV preparations. Because the rAAV genome is single stranded, previous studies have assumed that rAAV genomes must be converted to double strands before NGS. We demonstrate that rAAV DNA extracts exist primarily as double-stranded species. We hypothesize that these molecules form from the natural base pairing of complementary [+] and [−] strands after DNA extraction and show that rAAV DNA extracts are sufficient templates for downstream NGS without the labor-intensive double-stranding step. Here, we provide a detailed protocol for the simple and rapid NGS of rAAV genomes from DNA extracts. With this protocol, users can quickly confirm the identity of an rAAV preparation and detect the presence of contaminating rAAV DNA. In addition, we share custom Python scripts that allow users to accurately determine the serotype and detect Cre-independent DNA recombination events in rAAV containing Lox sites within minutes. We have used these scripts to analyze more than 100 rAAV preparations. Although we focused on the detection of cross-contaminating rAAV DNA and recombination events, our Python scripts can be customized to detect other sequences or events, such as reverse packaging of plasmid backbone or DNA from the packaging cell line. We find that the NGS of rAAV DNA extracts, termed viral genome sequencing, is a simple and powerful method for rAAV validation.

Background Borrelia burgdorferi is a bacterial spirochete that can cause Lyme disease after infecting a susceptible host. Immune responses to the bacteria are highly variable and host specific. The murine substrain, C3H/HeJ, is one of the most frequently utilized mouse models for Lyme disease. In this study, we sought to investigate the correlation of age with onset and severity of dermatitis, both in C3H/HeJ mice infected with B. burgdorferi as well as humans who have had a diagnosis of Lyme disease. Methods Female C3H/HeJ mice aged 6-8 weeks, 1 year, or 2 years were infected intraperitoneally with 105 B. burgdorferi. Dermatitis of the tail was evaluated by gross examination and histology. Human data via electronic health records of 342,499 Finnish individuals was tested and analyzed for associations between Lyme disease and atopic dermatitis. Results Dermatitis worsened over the course of untreated infection, with ulceration, hemorrhaging, flaking, hair loss, and dark lesions as well as spongiosis and acanthosis. These features of dermatitis were present in infected mice after 1 year of age. This relationship among Lyme disease, atopic dermatitis, and host age seen in the C3H/HeJ mouse model is consistent with a large pool (342,499) of human epidemiological data from Finland. We identified 5,248 individuals with Lyme disease and 17,233 with atopic dermatitis in FinnGen. Retrospective analysis shows Lyme disease is associated with atopic dermatitis (OR = 1.91 [1.68 -2.37], P < 2e−16). More visits due to Lyme disease complications (3 or more visits versus 1 visit) were associated with atopic dermatitis (OR = 2.19 [1.35-3.55], P = 0.0014) and risk of developing atopic dermatitis over time (HR=2.26 [1.54-3.95], P = 0.0017). Conclusion Data from mice and humans reveal a novel relationship among Lyme disease, age, and atopic dermatitis. Through defined pathological scoring, we demonstrate the onset of murine atopic dermatitis with B. burgdorferi infection, which is further exacerbated by host age at time of infection. In humans, a diagnosis of Lyme disease in FinnGen was associated with atopic dermatitis and further research is warranted to establish causation.

Calculus is typically one of the first college courses encountered by science, technology, engineering, and mathematics (STEM) majors. Calculus often presents major challenges affecting STEM student persistence, particularly for students from groups historically underrepresented in STEM. For life sciences majors, calculus courses may not offer content that is relevant to biological systems or connect with students' interests in biology. We developed a transformative approach to teaching college-level math, using a dynamical systems perspective that focuses first on demonstrating why students need math to understand living systems, followed by providing quantitative and computational skills, including concepts from calculus, that students need to build and analyze mathematical models representing these systems. We found that students who complete these new math courses perform better in subsequent science courses than their counterparts who take traditional calculus courses. We also provide evidence that the new math curriculum positively impacts students' academic performance, with data that show narrowing of the achievement gap, based on students' math grades, between student subgroups in the new math courses. Moreover, our results indicate that students' interest in the concepts and skills critical to the quantitative preparation of 21st-century life sciences majors increases after completing the new contextualized math curriculum.

Gupta et al. investigate forces generated during cell division in confining epithelial monolayers. In addition to finding that cells generate forces during mitotic rounding, they find that cells generate protrusive forces along the division axis that drive elongation and outward forces that facilitate postdivision spreading., Epithelial cells undergo striking morphological changes during division to ensure proper segregation of genetic and cytoplasmic materials. These morphological changes occur despite dividing cells being mechanically restricted by neighboring cells, indicating the need for extracellular force generation. Beyond driving cell division itself, forces associated with division have been implicated in tissue-scale processes, including development, tissue growth, migration, and epidermal stratification. While forces generated by mitotic rounding are well understood, forces generated after rounding remain unknown. Here, we identify two distinct stages of division force generation that follow rounding: (1) Protrusive forces along the division axis that drive division elongation, and (2) outward forces that facilitate postdivision spreading. Cytokinetic ring contraction of the dividing cell, but not activity of neighboring cells, generates extracellular forces that propel division elongation and contribute to chromosome segregation. Forces from division elongation are observed in epithelia across many model organisms. This article explores how research helps construct a certain type of ‘gender’ knowledge that arises from, informs and reinforces ‘instrumentalist’ gendered policy perspectives on development of the Global South. It uses a case study of research funded under the ESRC-DFID Joint Fund for Poverty Alleviation which awarded 122 grants amounting to £66.2 million (around US$88 million) between 2005 and 2015. From a systematic review of the awards a typology of gender inclusion and exclusion was constructed that found 60% of all awards mentioned gender or included some level of gender analysis. The subsequent synthesis of the evidence suggested that in only 30% of all awards was the gendered knowledge produced central to the study and/or focused on better understanding gender roles, relations and identities. Applying a Feminist Institutionalist lens, the study highlights how institutional ideas around gender are reflected in the funding call specifications, and in turn influence how researchers ‘engendered’ their research, and the type of gendered knowledge produced. It finds much of the new gendered knowledge produced out of the Joint Fund emerged from non-gender focused research often produced by non-gender specialists. It suggests that as gender becomes mainstreamed into research, and as more researchers ‘do gender’, so research may become, conversely, less ‘gendered’. The ‘new’ gender knowledge produced may then merely evidence existing institutional policy positions rather than advance the policy agenda.

Neutralizing activity found in the blood following intramuscular vaccination protects against systemic disease from SARS-CoV-2 infection. Mild breakthrough mucosal infections in vaccinated individuals can contribute to transmission chains of infection, and as such the contribution of intramuscular vaccination to mucosal immunity is important to understand. We assessed vesicular stomatitis virus (VSV) SARS-CoV-2 SPIKE protein neutralization activities as well as anti-viral spike and RBD IgG and IgA isotype antibody binding, in saliva from individuals who had recovered from SARS-CoV-2 infection and people who received immunization. The various vaccination strategies deployed globally showed key differences in the extent of neutralizing activity that could be measured. Even in subjects with significant peak activity 2–4 weeks following a second dose of mRNA vaccination, this neutralizing activity was transient and significantly reduced within 3–6 months. Among adenoviral vectors, ChAdOx1 elicited significantly more salivary neutralizing activity and antibody levels compared to Ad26.S. A large range of salivary neutralizing activity is induced following different intramuscular SARS-COV-2 vaccination strategies. All were orders of magnitude lower than neutralization levels observed in blood, and it remains to be determined if any of the observed neutralizing activity in the saliva can neutralize virus in the upper respiratory tract upon exposure. Based on studies where high levels of mucosal IgA antibodies were induced in response to stimulation with aerosol intranasal vaccines, such vaccines could be important in lowering transmission of SARS-COV-2. Lyme disease, caused by the bacteria Borrelia burgdorferi, is the most common and rapidly growing vector-borne infectious disease in the United States and Europe. High variability in disease burden among Lyme patients suggests that individual immune responses may be key drivers of clinical presentation and patient outcomes. Use of high resolution flow-based immunosorbent profiling revealed that a subset of Lyme patients with persistent symptoms were producing high concentrations of IgE specific to B. burgdorferi. Comparing C57B/6 mice, which are tolerant to B. burgdorferi, and C3H/HeJ mice, which are susceptible to disease, we find high levels of IgE specific for B. burgdorferi in C3H/HeJ but not C57B/6 mice. Furthermore, IgE was found to target Borrelia peptidoglycan in both acute and chronic infection models. Histologic analysis of mouse Lyme arthritic ankle tissue showed mast cells, which release highly immunogenic effectors upon activation by bound IgE, degranulating at significantly higher rates compared to uninfected controls. Forced mast cell degranulation exacerbated Lyme arthritis in infected mice. This data suggests that a subset of Lyme patients with persistent symptoms may have developed an allergic response to conserved bacterial antigens from a B. burgdorferi infection, as opposed to an autoimmune type response. Inclusion of IgE reactivity in diagnostic testing and examination of pathological immune responses to bacterial antigens could assist clinicians in patient care and effective treatments. In this article, we focus on how neoliberal performance metrics impact on non-traditional students at a modern university in England. We argue that the introduction of ‘quality assurance’ measures, (such as the National Student Survey and the Teaching Excellence Framework) are driven by an ideology which purports to have student’s best interests at heart by raising teaching standards, focusing on graduate employability and wider participation, but in fact works to discourage critical pedagogic practices that would allow for more democratic and dialogic spaces of learning. This article presents findings from one multi-modal qualitative case study at a particular higher education institution in London, where many of the students originate from socially and economically deprived areas and frequently come from ethnic minority groups. We argue that the radical space of the classroom provides a unique opportunity for students to move into collective and empathetic modes of learning that yield both normative measures of ‘success’ as well as more transformative outcomes. We maintain that critical pedagogies work to disrupt the neoliberal narrative that champions individual success and the student-as-consumer model, and by doing so, helps to redress the persistent inequalities that non-traditional students face in UK higher education settings.

BackgroundAs higher education institutions strive to effectively support an increasingly diverse student body, they will be called upon to provide their faculty with tools to teach more inclusively, especially in science, technology, engineering, and mathematics (STEM) classrooms where recruitment and retention of students from underrepresented and disadvantaged groups present long-standing challenges. Pedagogical training approaches to creating inclusive classrooms involve interventions that raise awareness of student and instructor social identities and explore barriers to learning, such as implicit bias, microaggressions, stereotype threat, and fixed mindset. Such efforts should focus on embracing diversity as an asset leveraged to benefit all students in their learning. In this paper, we describe the impact of multiday, off-campus immersion workshops designed to impart faculty with these tools. Based on analysis of workshop participant data, we report the resulting changes in faculty knowledge of factors affecting classroom climate and student success in STEM, attitudes about students, and motivation to adopt new teaching practices aimed at fostering equitable and culturally responsive learning environments.ResultsKey findings indicate that attendees (1) increased their knowledge of social identities and the barriers to learning in STEM classrooms, particularly those faced by students from underrepresented groups in STEM or socioeconomically challenged backgrounds; (2) changed their attitudes about students’ abilities as science majors, shifting away from a fixed-mindset perspective in which characteristics, such as intelligence, are perceived as innate and unalterable; and (3) modified their teaching approaches to promote inclusivity and cultural responsiveness.ConclusionFaculty members, who are linchpins in the evolution of college classrooms into settings that provide students with equitable opportunities to succeed academically in STEM, can benefit from participating in immersion workshops structured to support their awareness of issues affecting classroom culture related to race/ethnicity, LGBTQ status, religious affiliation, ability, socioeconomic status, and other social identities that contribute to disparities in STEM achievement and persistence.

Hyperosmolarity has been used as a means to improve yields in various processes such as antibody production in hybridoma cells and retroviral and adenoviral production systems. In this study we demonstrate that treatment of AAVpro 293T packaging cells with the sugars sucrose or sorbitol during adeno-associated virus (AAV) production improves the yields of AAV2 and rAAV2-retro serotypes 1.8-fold and 1.5-fold, respectively. Using an iodixanol gradient centrifugation strategy followed by a column-based buffer exchange, we show that the sugar supplements are not carried over to the final viral preparation and have no effect on the purity or infectivity of the virus. Given the wide availability, low cost, and ease of use of these sugars, we believe that this strategy can be easily adopted by vector cores and laboratories to improve yields of their AAV preparations.

Organ renewal is governed by the dynamics of cell division, differentiation and loss. To study these dynamics in real time, we present a platform for extended live imaging of the adult Drosophila midgut, a premier genetic model for stem-cell-based organs. A window cut into a living animal allows the midgut to be imaged while intact and physiologically functioning. This approach prolongs imaging sessions to 12–16 hr and yields movies that document cell and tissue dynamics at vivid spatiotemporal resolution. By applying a pipeline for movie processing and analysis, we uncover new and intriguing cell behaviors: that mitotic stem cells dynamically re-orient, that daughter cells use slow kinetics of Notch activation to reach a fate-specifying threshold, and that enterocytes extrude via ratcheted constriction of a junctional ring. By enabling real-time study of midgut phenomena that were previously inaccessible, our platform opens a new realm for dynamic understanding of adult organ renewal.

London’s Soho, situated in the urban heart of the city has long been understood as both a cosmopolitan and diverse space where transgression and deviance, particularly in relation to the sex industry and sexual commerce, are constitutive of this area. Drawing on three years of ethnographic fieldwork, we add to some of the existing debates on sexual spaces in Soho by documenting the changes to the social/sexual landscape of sex shops in this area, and look to geographers interested in the spatial politics of gender and sexuality to understand the importance of this particular place. Looking at two particular sex shops in Soho, we argue that the spatial practices in this very specific part of the city encourage a disruption of traditional hierarchies that often govern gender and sexed practices, and invite women, LGBTQ and kink communities to inhabit more inclusive spaces of sexual citizenship

The themed section consists of articles that explore the relationship between power and space in relation to gender and sexuality by looking at processes of transgression, subversion or expansion of normative spatial practices and narratives. Using a theoretical framework that draws out power and space within a more specific context of feminist and queer literature, the articles explore the possibility to transgress, subvert or expand norms at the interstices of spatial boundaries beyond traditional binaries and hierarchies. Collectively, the articles call for a continued theoretical and methodological focus into the importance of looking at everyday sites of struggles and resistance in the crevasses, the liminal zones of space. The transgression of spatialized norms of sexuality and gender present a transformative potential that should be recognized for its political significance but, we argue, with caution as heteronormative and heteropatriarchal norms too often remain de rigueur in a neoliberal context.

In her chapter on the UK Independence Party (UKIP), Erin Sanders-McDonagh explores the views of female UKIP supporters and adds to existing literature on the importance of nativism within the populist radical right (PRR). Nativism has often been seen as a key factor in determining electoral support for PRR parties, and women in this sample are clearly attracted to UKIP as a result of their adherence to nativist ideas. Drawing on in-depth interview data, she argues that women who support UKIP interpolate nativism in highly gendered ways—with Muslim women specifically seen to be a threat to ‘British’ culture and values. She looks at the shift from the 2014 UKIP Manifesto to the 2017 Manifesto, drawing attention to the ways in which Islamophobic sentiments creep in. Sanders-McDonagh highlights the consonance in the official discourses of UKIP and the anti-Muslim expressions in the interview data and argues that examining these shifts in political ideologies can reveal important aspects of the nature of gendered political dynamics.

This paper draws on ethnographic research conducted in Amsterdam, exploring the ways in which women tourists engage with public sex performances, drawing comparisons between these types of shows and other types of pornographic materials. Empirical data collected in Amsterdam, focusing on women's visual consumption of public sex performances at a well-known tourist sexual theatres that features live sex (including vaginal penetrative sex and oral sexual encounters for/by men/women, as well as masturbation and other highly sexualized acts), suggests that sex shows are positioned as legitimate sexual entertainment for men, women, and couples, and that a wide range of tourist women from different backgrounds visit these shows in substantial numbers. Botryllus schlosseri is a colonial ascidian with a natural ability to anastomose with another colony to form a vascular and hematopoietic chimera. In order to fuse, two individuals must share at least one allele at the highly polymorphic fuhc locus. Otherwise, a blood-based inflammatory response will occur resulting in a melanin scar at the sites of interaction. The single-locus genetic control of allorecognition makes B. schlosseri an attractive model to study the underlying molecular mechanisms. Over the past decade, several candidate genes involved in allorecognition have been identified, but how they ultimately contribute to allorecognition outcome remains poorly understood. Here, we report our initial molecular characterization of a recently identified candidate allodeterminant called Botryllus histocompatibility factor (bhf). bhf, both on a DNA and protein level, is the least polymorphic protein in the fuhc locus studied so far and, unlike other known allorecognition determinants, does not appear to be under any form of balancing or directional selection. Additionally, we identified a second isoform through mRNA-Seq and an EST assembly library which is missing exon 3, resulting in a C-terminally truncated form. We report via whole-mount fluorescent in situ hybridization that a subset of cells co-express bhf and cfuhc(sec). Finally, we observed BHF's localization in HEK293T at the cytoplasmic side of the plasma membrane in addition to the nucleus via a nuclear localization signal. Given the localization data thus far, we hypothesize that BHF may function as a scaffolding protein in a complex with other Botryllus proteins, rather than functioning as an allorecognition determinant.

Purpose – The purpose of this paper is to explore dirty work sites within an academic context. Working with particular “unloved” groups (Fielding, 1993) can present a number of challenges to researchers, and if professional boundaries are not carefully maintained, researchers can be seen as “dirty workers” within an academic context. Design/methodology/approach – The paper draws a qualitative research project that explores women's involvement with nationalist movements in the UK. Findings – Researching “unloved” groups, and in particular racist organizations, presents a number of potential emotional and professional, and can render researchers “dirty workers” if clear professional boundaries are not maintained. Originality/value – Examining academia and some academic research as a dirty work site adds to existing literature (Kreiner et al., 2006) that suggests any occupation can have a “dirty work” element that must be negotiated. This article will explore the gentrification of Soho, reflecting on ethnographic research undertaken in the area over the past fifteen months, to argue that the recent social, political, and economic changes in Soho must be understood in relation to private, marketized and globalized neoliberal capitalist forces. We argue that the changes to the area result in a heavily-weighted form of gentrification that works to actively and knowingly sanitize the city, removing ‘undesirable’ people and venues from the area. As such, we propose to define this process as ‘hegemonic gentrification’, and distinguish this from other forms of gentrification in order to understand the different processes that underpin these specific changes, and more broadly, it allows us to problematize these changes as regards to the ‘right to the city’, and to expand current understandings in a way that allows for a more nuanced analysis of urban gentrification and its impacts within neolibreral capitalism.

The dismantling of the welfare state across the United Kingdom (and indeed a number of other Western industrialised democracies, such as Canada and the United States) and the reductions to welfare provisions and entitlements are having a detrimental impact on women's equality and safety. Towers and Walby argue that the recent cuts to welfare provision in the United Kingdom, particularly for women's services, could lead to increased levels of violence for women and girls. This paper makes the argument that female victims of domestic abuse experience violence on two levels: first, at the intimate/personal level through their relationship with an abuser and, second, at a structural level, through the state failing to provide adequate protection and provision for women who have experienced violence in intimate relationships. Using a specific example of post-violence community services delivered to both the children of women who have experienced domestic violence and the women themselves, this paper draws on empirical research carried out in 2010–2011 with London-based third-sector and public sector organisations delivering the Against Violence and Abuse Project ‘Community Group Programme’. We argue that the lack of services for women involved in, or exiting, a violent relationship can amount to state-sanctioned violence, if funding is withheld, or indeed, stretched to breaking point.
