In this study, the oils of açaí, passion fruit, pequi, and guava were submitted to physicochemical analysis to investigate their potential application in the food industry. Gas chromatography associated with mass spectroscopy showed that oleic and linoleic acids are mainly responsible for the nutritional quality of açaí, passion fruit, pequi, and guava oils, which exhibited 46.71%, 38.11%, 43.78%, and 35.69% of the former fatty acid, and 18.93%, 47.64%, 20.90%, and 44.72% of the latter, respectively. The atherogenicity index of the oils varied from 0.11 to 0.65, while the thrombogenicity index was 0.93 for açaí, 0.35 for guava, and 0.3 for passion fruit oils, but 1.39 for pequi oil, suggesting that the use of the first three oils may lead to a low incidence of coronary heart disease. Thermogravimetry showed that all tested oils were thermally stable above 180 °C; therefore, they can be considered resistant to cooking and frying temperatures. In general, the results of this study highlight possible applications of these oils in the food industry, either in natura or in typical food production processes.

Cyclins are a family of proteins characterized by possessing a cyclin box domain that mediates binding to cyclin dependent kinases (CDKs) partners. In this study, the search for a partner cyclin of the PHO85-1 CDK retrieved PCL-1 an ortholog of yeast Pcls (for Pho85 cyclins) that performs functions common to Pcls belonging to different cyclin families. We show here that PCL-1, as a typical cyclin, is involved in cell cycle control and cell progression. In addition, PCL-1 regulates glycogen metabolism; Δpcl-1 cells accumulate higher glycogen levels than wild-type cells and the glycogen synthase (GSN) enzyme is less phosphorylated and, therefore, more active in the mutant cells. Together with PHO85-1, PCL-1 phosphorylates in vitro GSN at the Ser636 amino acid residue. Modeling studies identified PHO85-1 and PCL-1 as a CDK/cyclin complex, with a conserved intermolecular region stabilized by hydrophobic and polar interactions. PCL-1 is also involved in calcium and NaCl stress response. Δpcl-1 cells are sensitive to high NaCl concentration; on the contrary, they grow better and overexpress calcium responsive genes under high calcium chloride concentration compared to the wild-type strain. The expression of the calcium-responsive CRZ-1 transcription factor is modulated by PCL-1, and this transcription factor seems to be less phosphorylated in Δpcl-1 cells since exhibits nuclear location in these cells in the absence of calcium. Our results show that PCL-1 locates at different cell regions suggesting that it may determine its activity by controlling its intracellular location and reveal an interesting functional divergence between yeast and filamentous fungus cyclins.

The Programa Institucional de Bolsas de Iniciação à Docência (PIBID) of the Ministry of Education aims to promote the integration between basic education and higher education, inserting future professionals in the daily routine of the Brazilian public education network. Since its implementation in 2007, studies have been defending the need for this program to train future education professionals, since contributes to the development of pedagogical praxis based on reflected teaching action. As undergraduate students are the main agents of this program, this study has the research question: How do students evaluate PIBID? Thus, it aims to analyze the perception of students of undergraduate courses at the Universidade Federal da Bahia about the contributions of PIBID to teacher training. It is a case study, with a qualitative approach, which uses semi-structured interviews with, as a research resource. Eighteen scholarship students were interviewed, linked to 9 courses, from day and night shifts, offered by UFBA. From the content analysis, it was possible to identify that the students expressed the relevance of PIBID as a necessary public policy for the training of future teachers. On the other hand, they recognized that despite being a pedagogical innovation with significant impacts on teacher training and professional development, the contingency of resources, on the part of the federal government, imposes challenges for the development of the program.

Despite the documented effects on human and animal health, particles smaller than 0.1 µm in diameter found in soils, sediments, and the atmosphere remain unregulated. Yet, cerium and titanium oxide nanoparticles associated with traffic increase mortality, cause behavioral changes, and inhibit the growth in amphibians. Mites of the genus Hannemania spend their early stages in the soil before becoming exclusive parasites of amphibians. Unlike other mites, Hannemania is found inside the epidermis of amphibians, thus facilitating the intake of particles, and leading to direct and chronic exposure. To better understand this exposure path, we sampled amphibians hosting mites in a river potentially polluted by traffic sources. Particles collected from mites were studied by scanning electron microscopy and Raman spectroscopy while sediment samples were analyzed for total metal content by portable X-ray fluorescence. Our results indicate that sediment samples showed significant correlations between elements (Zr, Mn, Ti, Nb, Fe) often associated with components in catalytic converters and a level of Zr that exceeded the local geochemical background, thus suggesting an anthropic origin. Furthermore, particles adhered to mites exhibited the characteristic Raman vibrational modes of ceria (CeO2, 465 cm−1), ceria-zirconia (CeO2-ZrO2, 149, 251, and 314 cm−1), and rutile (TiO2, 602 cm−1), pointing out to the deterioration of catalytic converters as the most likely source. This research highlights both the importance of unregulated catalytic converters as a source of ultrafine Ce-Ti particle pollution and the role of sub-cutaneous mites as a vector of these particles for amphibian exposure.

The Milky Way Bulge extra-tidal star survey is a spectroscopic survey with the goal of identifying stripped globular cluster stars from inner Galaxy clusters. In this way, an indication of the fraction of metal-poor bulge stars that originated from globular clusters can be determined. We observed and analyzed stars in and around BH 261, an understudied globular cluster in the bulge. From seven giants within the tidal radius of the cluster, we measured an average heliocentric radial velocity of 〈RV〉 = −61 ± 2.6 km s ^−1 with a radial velocity dispersion of 〈 σ 〉 = 6.1 ± 1.9 km s ^−1 . The large velocity dispersion may have arisen from tidal heating in the cluster’s orbit about the Galactic center, or because BH 261 has a high dynamical mass as well as a high mass-to-light ratio. From spectra of five giants, we measure an average metallicity of 〈[Fe/H]〉 = −1.1 ± 0.2 dex. We also spectroscopically confirm an RR Lyrae star in BH 261, which yields a distance to the cluster of 7.1 ± 0.4 kpc. Stars with 3D velocities and metallicities consistent with BH 261 reaching to ∼0.°5 from the cluster are identified. A handful of these stars are also consistent with the spatial distribution of potential debris from models focusing on the most recent disruption of the cluster.

The storage lesions and the irradiation of blood cellular components for medical procedures in blood banks are events that may induce nanochanges in the membrane of red blood cells (RBCs). Alterations, such as the formation of pores and vesicles, reduce flexibility and compromise the overall erythrocyte integrity. This review discusses the alterations on erythrocytic lipid membrane bilayer through their characterization by confocal scanning microscopy, Raman, scanning electron microscopy, and atomic force microscopy techniques. The interrelated experimental results may address and shed light on the correlation of biomechanical and biochemical transformations induced in the membrane and cytoskeleton of stored and gamma-irradiated RBC. To highlight the main advantages of combining these experimental techniques simultaneously or sequentially, we discuss how those outcomes observed at micro- and nanoscale cell levels are useful as biomarkers of cell aging and storage damage.

Orchiectomy is performed in some species using only sedation and local anesthesia to decrease the expense of performing the procedure using general anesthesia. The objective of this study was to determine if dogs can be castrated safely and painlessly by using only sedation and local anesthesia. After dogs were sedated with intramuscularly administered xylazine (1 mg/kg) and subanesthetic ketamine (1 mg/kg), testes and skin were anesthetized with 2% lidocaine (6 mg/kg, total dose) buffered with sodium bicarbonate. Systolic, diastolic, and mean blood pressures; heart and respiratory rates; and movement scores were determined before and during surgery when manipulations were most likely to cause pain. No dog reacted substantially to injection of the combination of sedatives, and no dog reacted noticeably to injection of lidocaine. During surgery, the average heart rate was reduced from baseline by 40–60 beats per minute, and the average respiratory rate was reduced by ~10 breaths per minute. An overall reduction of arterial blood pressures was observed. All but one dog moved purposely in response to a toe pinch at the end of surgery. We found that sedating dogs intramuscularly with xylazine and a sub-anesthetic dose of ketamine and administering lidocaine at the incision site and intratesticularly allowed dogs to be castrated humanely and avoided the expense of general anesthesia and the need for hospitalization.

Colorectal cancer (CRC) is a common malignancy with 1. 8 million cases in 2018. Autophagy helps to maintain an adequate cancer microenvironment in order to provide nutritional supplement under adverse conditions such as starvation and hypoxia. Additionally, most of the cases of CRC are unresponsive to chemotherapy, representing a significant challenge for cancer therapy. Recently, autophagy induced by therapy has been shown as a unique mechanism of resistance to anticancer drugs. In this regard, long non-coding RNAs (lncRNAs) analysis are important for cancer detection, progression, diagnosis, therapy response, and prognostic values. With increasing development of quantitative detection techniques, lncRNAs derived from patients' non-invasive samples (i.e., blood, stools, and urine) has become into a novel approach in precision oncology. Tumorspecific GAS5, HOTAIR, H19, and MALAT are novels CRC related lncRNAs detected in patients. Nonetheless, the effect and mechanism of lncRNAs in cancer autophagy and chemoresistance have not been extensively characterized. Chemoresistance and autophagy are relevant for cancer treatment and lncRNAs play a pivotal role in resistance acquisition for several drugs. LncRNAs such as HAGLROS, KCNQ1OT1, and H19 are examples of lncRNAs related to chemoresistance leaded by autophagy. Finally, clinical implications of lncRNAs in CRC are relevant, since they have been associated with tumor differentiation, tumor size, histological grade, histological types, Dukes staging, degree of differentiation, lymph node metastasis, distant metastasis, recurrent free survival, and overall survival (OS).

The objective of this study was to evaluate the effect of cutting age on the production, chemical composition, degradation kinetics and anatomy of Leucena (Leucaena leucocephala). The experimental design was a completely randomized design with a factorial arrangement 2x4 (two types of tissues and four cutting ages) for the production data of dry matter and 3x4 (three degrees of degrees of digestion of tissues and four cutting ages). Observed effect (P < 0.05) for the total production of DM of Leucena in function of different ages. The tissue types grain and non-grain showed maximum production at 70 days of age, with production of 2333,00 and 716.60 kg DM ha-1, respectively. The parameters of degradation of DM evaluated decreased significantly with the increase in the maturity of the plant, in the same way the chemical composition presented behavior inherent to the advance of age. The effective degradability DM also decreased with the increase in the rate of passage (2, 5, and 8% h-1). The highest rate of degradation (c) was obtained for 30 days. With the advance in plant maturity increases the proportion of vascular tissue lignificad influencing parameters of ruminal degradation of Leucena. The ages assessed influenced the chemical composition of the Leucena (P < 0.05), where the levels of dry matter, crude protein, acid detergent fiber and lignin and ash showed increasing linear behavior. The cutting age of 70 days offers an optimal point regarding the proportion of anatomical tissues correlated with the degradation and chemical composition of the Leucena.

Nanodiamonds have been studied for several biomedical applications due to their inherent biocompatibility and low cytotoxicity. Recent investigations have shown perspectives in using fluorescent nanodiamonds as nanothermometers because of their optical properties’ dependence on temperature. Easy and accurate localized temperature sensing is essential in a wide variety of scientific fields. Our work demonstrated how the fluorescence spectrum of high-pressure high-temperature fluorescent nanodiamonds of three different sizes: 35 nm, 70 nm and 100 nm, changes with temperature within an important biological temperature range (25 °C to 60 °C). Taking advantage of this phenomenon, we obtained nanothermic scales (NS) from the zero phonon lines (ZPL) of the NV0 and NV− colour centres. In particular, the 100 nm-sized features the more intense fluorescence spectra whose linear dependence with temperature achieved 0.98 R2 data representation values for both NV0 and NV−. This model predicts temperature for all used nanodiamonds with sensitivities ranging from 5.73% °C−1 to 6.994% °C−1 (NV0) and from 4.14% °C−1 to 6.475% °C−1 (NV−). Furthermore, the non-cytotoxic interaction with HeLa cells tested in our study enables the potential use of fluorescence nanodiamonds to measure temperatures in similar nano and microcellular aqueous environments with a simple spectroscopic setup.

Hepatocellular carcinoma (HCC) ranks fifth in occurrence and second in mortality of all cancers. The development of effective therapies for HCC is urgently needed. Anticancer drugs targeted to the liver-specific asialoglycoprotein receptors (ASGPRs) are viewed as a promising potential treatment for HCC. ASGPRs facilitate the recognition and endocytosis of molecules, and possibly vehicles with galactose end groups, by the liver. In this study, bovine serum albumin (BSA) was conjugated with lactose using a thermal treatment. The formation of lactosylated BSA (BSA-Lac) was confirmed by a change of the chemical structure, increased molecular mass, and Ricinus communis lectin recognition. Subsequently, the low-crosslinking BSA-Lac nanoparticles (LC BSA-Lac NPs) and high-crosslinking BSA-Lac nanoparticles (HC BSA-Lac NPs) were synthesized. These nanoparticles presented spherical shapes with a size distribution of 560 ± 18.0 nm and 539 ± 9.0 nm, as well as an estimated surface charge of −26 ± 0.15 mV and −24 ± 0.45 mV, respectively. Both BSA-Lac NPs were selectively recognized by ASGPRs as shown by biorecognition, competition, and inhibition assays using an in vitro model of HCC. This justifies pursuing the strategy of using BSA-Lac NPs as potential drug nanovehicles with selective direction toward hepatocellular carcinoma.

ABSTRACT Objective: The objective of this study was to determine the factors associated with the persistence of pain in patients operated on for lumbar stenosis. Method: One hundred and fifty-three patients were studied, divided into two groups: 1) Patients with persistent pain in varying degrees, 2) Patients without pain. Age, sex, affected levels, comorbidities, surgical risk, and type of surgical procedure were evaluated. Results: There were 108 patients in the group with pain and 45 in the group without pain. In the group with pain, there were 28 patients with diabetes mellitus, 31 smokers, and 28 alcohol-dependent patients, with a significant difference of p = 0.001 and an RR = 1.1. A simple widening procedure was performed in 48 patients in the group with pain and 12 patients in the group without pain, with RR = 0.8, and widening plus instrumentation was performed in 7 patients in both the with and without pain groups. Conclusion: The indication of a surgical procedure in patients with spinal stenosis must take many factors into account in addition to clinical factors and the segments affected, since these factors impact patient prognosis. In the multivariate analysis, the variable most closely associated with persistent pain was the procedure performed. Level of Evidence III; Case-control study.g

ABSTRACT Objective: To evaluate the direct costs of transforaminal lumbar interbody fusion (TLIF) and minimally invasive surgery (MIS) or open technique (OPEN). Methods: The present study is descriptive and retrospective. Sixteen patients with degenerative spinal pathology operated on with the TLIF MIS technique and TLIF OPEN were included over a 13-month period. Days of hospital stay, blood loss, surgical time, medical care and costs were compared. Results: The mean number of days of hospital stay was 6.7 ± 4.3 days with TLIF MIS and 11.1 ± 6.5 days with TLIF OPEN. The blood loss was 307 ± 81.6 ml (range 200400 ml) with TLIF MIS and 803 ± 701.3 ml (range 200-1800 ml) with TLIF OPEN. The surgical time was 320 ± 92.6 minutes (range 210-500 minutes) in TLIF MIS and 372 ± 95.2 minutes (280-540 minutes) in TLIF OPEN. Conclusions: The difference in surgical costs and time between the two procedures was not statistically significant. There was less bleeding during the TLIF-MIS surgery, as well as a correlation between shorter days of hospital stay proportional to bleeding and surgical time, which translates into a reduction in the cost of these items. Level of Evidence III; Analysis based on alternatives and limited costs.

VEGETABLE oils ,FOOD industry ,CORONARY disease ,PASSION fruit ,MANUFACTURING processes ,LINOLEIC acid ,PALMITIC acid - Abstract
[ABSTRACT FROM AUTHOR]

BACKGROUND: The necessity to manufacture scaffolds with superior capabilities of biocompatibility and biodegradability has led to the production of extracellular matrix (ECM) scaffolds. Among their advantages, they allow better cell colonization, which enables its successful integration into the hosted tissue, surrounding the area to be repaired and their formulations facilitate placing it into irregular shapes. The ECM from porcine urinary bladder (pUBM) comprises proteins, proteoglycans and glycosaminoglycans which provide support and enable signals to the cells. These properties make it an excellent option to produce hydrogels that can be used in regenerative medicine. OBJECTIVE: The goal of this study was to assess the biocompatibility of an ECM hydrogel derived from the porcine urinary bladder (pUBMh) in vitro using fibroblasts, macrophages, and adipose-derived mesenchymal stem cells (AD-MCSs), as well as biocompatibility in vivo using Wistar rats. METHODS: Effects upon cells proliferation/viability was measured using MTT assay, cytotoxic effects were analyzed by quantifying lactate dehydrogenase release and the Live/Dead Cell Imaging assay. Macrophage activation was assessed by quantification of IL-6, IL-10, IL-12p70, MCP-1, and TNF-α using a microsphere-based cytometric bead array. For in vivo analysis, Wistar rats were inoculated into the dorsal sub-dermis with pUBMh. The specimens were sacrificed at 24 h after inoculation for histological study. RESULTS: The pUBMh obtained showed good consistency and absence of cell debris. The biocompatibility tests in vitro revealed that the pUBMh promoted cell proliferation and it is not cytotoxic on the three tested cell lines and induces the production of pro-inflammatory cytokines on macrophages, mainly TNF-α and MCP-1. In vivo, pUBMh exhibited fibroblast-like cell recruitment, without tissue damage or inflammation. CONCLUSION: The results show that pUBMh allows cell proliferation without cytotoxic effects and can be considered an excellent biomaterial for tissue engineering.

Purpose: The purpose of this study was to evaluate research from Brazilian postgraduate students who provide evidence of effectiveness for Nursing Interventions Classification (NIC). Methods: We conducted a literature review study of thesis and dissertations available in the Brazilian Digital Library of Dissertations and Theses (D/T) in May 2021 regardless of the year they were conducted. In those studies that did not utilize the NIC in the effectiveness evaluation, the cross‐mapping methodology was employed between NIC and the interventions used by the authors of the studies. Results: Using a systematic process, we identified 91 studies. Twenty‐seven met a priori inclusion and exclusion criteria. We found an increase in studies that focused on nursing interventions in the last 10 years (n = 19), a large proportion of clinical trials (n = 16), and the majority of articles from the Southeast region of Brazil (n = 20). The areas of focus were adult and elderly care, and with a special interest in the behavioral domain (n = 11). Two sensitivity criteria were identified in all D/T (n = 27), and each study presented evidence of effectiveness of a minimum of three criteria simultaneously. Conclusions: Based on the effectiveness criteria, the Brazilian scientific production in postgraduate programs carried out by nurses provides evidence of the effectiveness for NIC nursing interventions. Implications for nursing practice: It is recommended to conduct further research that uses the NIC in the planning, conduct, and evaluation of interventions, based on effectiveness criteria of nursing sensitivity. In addition, PCL-1 regulates glycogen metabolism; Δ

The proper registration of cases of deaths and their analysis contribute to give visibility to the real gaps and demands of health services. The monitoring and the analysis of the Infant Mortality Rate (IMR) and its characteristics are paramount for the development of health policies and interventions aimed at the promotion and protection of child health. Thus, we aimed to characterize neonatal deaths recorded in Palmas, Tocantins, Brazil, in the period from 1999 to 2018, according to sex, weight and age of newborns, maternal age, and municipality of occurrence. There were 800 deaths of children aged up to 27 days of life in the municipality of Palmas from 1999 to 2018, with a statistical difference in the proportion of neonatal deaths according to the occurrence of the event, gender, and weight, with a higher proportion of early neonatal deaths when their occurrence was in Palmas. Although the municipality of Palmas has recorded advances in infant mortality indicators, specifically in the neonatal component, considering that Brazil proposes to reduce newborn mortality to a maximum of five per thousand live births by 2030, advances will be necessary to qualify perinatal care in the region in order to avoid preventable infant deaths. Thus, an urgent demand is the improvement of prenatal and maternal-infant care services in the municipality in order to favor maternal and infant indicators.

IMPORTANCE: Astronauts returning from long-duration spaceflight experience ocular remodeling related to cephalad fluid shifts induced by microgravity. It is hypothesized that the absence of diurnal reductions in intracranial pressure in microgravity creates a low but persistent pressure gradient at the posterior aspect of the eye, which results in ocular remodeling and space-associated neuro-ocular syndrome (SANS) over many months. OBJECTIVE: To determine whether partial reintroduction of footward fluid shifts during simulated microgravity via lower body negative pressure (LBNP) during sleep attenuates choroid engorgement, an early marker of ocular remodeling related to SANS. DESIGN, SETTING, AND PARTICIPANTS: Between May 2019 and February 2020, participants with no major cardiovascular, kidney, or ophthalmic disease completed 3 days of supine (0°) bed rest with and 3 days without 8 hours of nightly LBNP in a randomized, crossover design. This single-center investigation took place at the UT Southwestern Medical Center. All analyses were conducted blinded to condition and time point. INTERVENTIONS: Eight hours of nightly LBNP (−20 mm Hg) vs no LBNP. MAIN OUTCOMES AND MEASURES: The primary outcome was the change in choroid area and volume after 3 days of bed rest measured by optical coherence tomography. RESULTS: Of 10 participants, 5 were female, the mean (SD) age was 29 (9) years, and the age range was 18 to 55 years. Central venous pressure increased from the seated to supine position (mean [SD], seated: −2.3 [2.0] vs supine: 6.9 [2.0] mm Hg; P
