IntroductionPlanting trees in urban areas can mitigate some of the emissions generated in cities by carbon sequestration (annual uptake of CO2 through the process of photosynthesis) and carbon storage (amount of carbon stored in the tree's biomass throughout its lifespan). The aim of this study was to calculate the carbon footprint from nursery production to final establishment of different tree species grown for planting in urban parks in a northern European context.Material and methodsThe analysis included a cradle-to-gate approach and investigated the amount of carbon the adult trees needed to sequester in order to compensate for initial carbon emissions and which temporal perspectives are of concern. Greenhouse gas emissions were estimated based on an inventory of consumption of fuels, energy, materials and other production inputs during cultivation, delivery, planting and establishment of three different tree species in three different locations in Sweden. The tree species considered in the analysis (Salix alba, Quercus rubra, Pinus sylvestris) were selected due to significant differences in their growth rates. Salix alba is a competitive strategist in resource-rich habitats, and is proficient at converting these resources into vigorous growth. Pinus sylvestris is a pronounced stress strategist with good ability to handle resource-limited habitats, and invests in traits accordingly, resulting in significantly slower development. Quercus rubra has its main distribution in cool and moderately resource-rich habitats, but has relatively high stress tolerance and can be considered intermediate between the other two species in terms of growth rate.Results and discussionThe results showed that within 16 years of planting, all species in all three cities, except Pinus sylvestris planted in Umeå, compensated for initial carbon emissions, i.e. showed net absorption of CO2 after emissions from cultivation, delivery, planting and establishment of the trees had been deducted. There was a clear link between the time by which compensation of initial carbon emissions was achieved and growth rate of the different species, with the fast-growing Salix alba showing the best results. The single largest source of emissions among all activities carried out during cultivation, delivery and planting of all species, regardless of the city in which they were planted, was fuel consumption during tree planting.

Neurons with internal memory have been proposed for biological and bio-inspired neural networks, adding important functionality. We introduce an internal time-limited charge-based memory into a III–V nanowire (NW) based optoelectronic neural node circuit designed for handling optical signals in a neural network. The new circuit can receive inhibiting and exciting light signals, store them, perform a non-linear evaluation, and emit a light signal. Using experimental values from the performance of individual III–V NWs we create a realistic computational model of the complete artificial neural node circuit. We then create a flexible neural network simulation that uses these circuits as neuronal nodes and light for communication between the nodes. This model can simulate combinations of nodes with different hardware derived memory properties and variable interconnects. Using the full model, we simulate the hardware implementation for two types of neural networks. First, we show that intentional variations in the memory decay time of the nodes can significantly improve the performance of a reservoir network. Second, we simulate the implementation in an anatomically constrained functioning model of the central complex network of the insect brain and find that it resolves an important functionality of the network even with significant variations in the node performance. Our work demonstrates the advantages of an internal memory in a concrete, nanophotonic neural node. The use of variable memory time constants in neural nodes is a general hardware derived feature and could be used in a broad range of implementations.

Vertical InAs/InGaAs nanowire MOSFETs are fabricated in a gate-last fabrication process, which allows gate-lengths down to 25 nm and accurate gate-alignment. These devices demonstrate high performance with transconductance of 2.4 mS/μm, high on-current, and off-current below 1 nA/μm. An in-depth analysis of the heterostructure MOSFETs are obtained by systematically varying the gate-length and gate location. Further analysis is done by using virtual source modeling. The injection velocities and transistor metrics are correlated with a quasi-ballistic 1-D MOSFET model. Based on our analysis, the observed performance improvements are related to the optimized gate-length, high injection velocity due to asymmetric scattering, and low access resistance.

We present a method for estimating the trap distributions on each of the surfaces in a multi-gate MOSFET. We perform I-V hysteresis measurements on InGaAs Tri-gate MOSFETs with various channel widths (25, 60, and 100 nm) from which top surface and side wall trap distributions are determined. We show that the total trap distribution of a device can be expressed as a linear combination of the top surface and side wall trap distributions. The results show that the minimum trap density of the top InGaAs (100) surface is smaller than that of the {110} side walls by almost an order of magnitude. Since the nanowire constituting the channel in these devices is selectively regrown, rather than etched out, the different trap distributions can be explained by the specific surface chemistries of two surfaces.

Actual motor competence (MC), perceived motor competence (PMC), and health-related fitness (HRF) exhibit a dynamic and reciprocal relationship in child populations, but little is known about the nature of these relationships in young adulthood. The purpose of the study was to assess these relationships in a sample of college-aged males. A total of 55 participants enrolled in an undergraduate Kinesiology course completed the study. Perceived motor competence (PMC) was assessed with the Physical Self-Perception Profile questionnaire; MC was assessed using maximum throw and kick speed and maximum jump distance; HRF was assessed with a two-minute push-up test, two-minute sit-up test, and the Multistage 20-m Shuttle Run Test. Pearson’s bivariate correlations were calculated to assess relationships among PMC total score, MC scores, and HRF scores. Two separate indices were calculated to create composite total MC and total HRF scores used for subsequent analyses. Significant correlations were found between PMC total score, MC index, and HRF index. Multiple linear regressions were used for analyzing predictive measures for HRF and PMC scores. From the two regression models, significance varied among total MC scores, PMC scores, and HRF individual measures. These findings may suggest that relationships among MC, HRF, and PMC strengthen over developmental time in young adult males.

In this paper, InAs/GaSb nanowire tunnel field-effect transistors (TFETs) are studied theoretically and experimentally. A 2-band 1-D analytic tunneling model is used to calculate the on- and off-current levels of nanowire TFETs with staggered source/channel band alignment. Experimental results from lateral InAs/GaSb are shown, as well as first results on integration of vertical InAs/GaSb nanowire TFETs on Si substrates.

The purpose of the study was to examine the changes in peak oxygen consumption ( V ˙O2peak) and running economy (RE) following four-weeks of high intensity training and concurrent strength and conditioning during the off-season in collegiate female field hockey players. Fourteen female student-athletes (age 19.29 ± 0.91 years) were divided into two training groups, matched from baseline V ˙O2peak: High Intensity Training (HITrun; n = 8) and High Intensity Interval Training (HIIT; n = 6). Participants completed 12 training sessions. HITrun consisted of 30 min of high-intensity running, while HIIT consisted of a series of whole-body high intensity Tabata-style intervals (75–85% of age predicted maximum heart rate) for a total of four minutes. In addition to the interval training, the off-season training included six resistance training sessions, three team practices, and concluded with a team scrimmage. V ˙O2peak was measured pre- and post-training to determine the effectiveness of the training program. A two-way mixed (group × time) ANOVA showed a main effect of time with a statistically significant difference in V ˙O2peak from pre- to post-testing, F(1, 12) = 12.657, p = 0.004, partial η2 = 0.041. Average (±SD) V ˙O2peak increased from 44.64 ± 3.74 to 47.35 ± 3.16 mL·kg−1·min−1 for HIIT group and increased from 45.39 ± 2.80 to 48.22 ± 2.42 mL·kg−1·min−1 for HITrun group. Given the similar improvement in aerobic power, coaches and training staff may find the time saving element of HIIT-type conditioning programs attractive.

Abstract Estimating emissions and removals from forest degradation is important, yet challenging, for many countries. This paper reports results from analysis of country reporting (to the United Nations Framework Convention on Climate Change and also to several climate finance initiatives) and key take-aways from a south-south exchange workshop among 17 countries with forest mitigation programmes. During the workshop discussions it became clear that, where forest degradation is a major source of emissions, governments want to include it when reporting on their mitigation efforts. However, challenges to accurately estimating emissions from degradation relate to defining forest degradation and setting the scope for estimating carbon stock changes; to detecting and monitoring degradation using earth observation data; and to estimating associated emissions and removals from field observation results. The paper concludes that recent and ongoing investments into data and analysis methods have helped improve forest degradation estimation, but further methodological work and continued effort will be needed.

Abstract α7 nicotinic acetylcholine receptors (nAChRs) are homopentameric ligand-gated ion channels with critical roles in the nervous system. Recent studies have resolved and functionally annotated closed, open, and desensitized states of these receptors, providing insight into ion permeation and lipid binding. However, the process by which α7 nAChRs transition between states remains unclear. To understand gating and lipid modulation, we generated two ensembles of molecular dynamics simulations of apo α7 nAChRs, with or without cholesterol. Using symmetry-adapted Markov state modeling, we developed a five-state gating model. Free energies recapitulated functional behavior, with the closed state dominating in absence of agonist. Open-to-nonconducting transition rates corresponded to experimental open durations. Cholesterol relatively stabilized the desensitized state, and reduced open-desensitized barriers. These results establish plausible asymmetric transition pathways between states, define lipid modulation effects on the α7 nAChR conformational cycle, and provide an ensemble of structural models applicable to rational design of lipidic pharmaceuticals.

Abstract ρ-type γ-aminobutyric acid-A (GABAA) receptors are widely distributed in the retina and brain, and are potential drug targets for the treatment of visual, sleep and cognitive disorders. Endogenous neuroactive steroids including β-estradiol and pregnenolone sulfate negatively modulate the function of ρ1 GABAA receptors, but their inhibitory mechanisms are not clear. By combining five cryo-EM structures with electrophysiology and molecular dynamics simulations, we characterize binding sites and negative modulation mechanisms of β-estradiol and pregnenolone sulfate at the human ρ1 GABAA receptor. β-estradiol binds in a pocket at the interface between extracellular and transmembrane domains, apparently specific to the ρ subfamily, and disturbs allosteric conformational transitions linking GABA binding to pore opening. In contrast, pregnenolone sulfate binds inside the pore to block ion permeation, with a preference for activated structures. These results illuminate contrasting mechanisms of ρ1 inhibition by two different neuroactive steroids, with potential implications for subtype-specific gating and pharmacological design.

Abstract Asylum seekers’ traumatic experiences in combination with discrimination, social isolation, and exclusion in the host country leads to low adherence from health and integration initiatives. Along with their inability to seek health care and physical inactivity, this situation increases their mental illness and, most importantly, decreases their well-being. In fact, the lack of well-being (e.g., life satisfaction) is a better marker of mortality and morbidity than the presence of mental illness. In this context, one of the major single determinants of well-being is character, a dimension of personality that stands for self-regulation, adaptation, and intentional conscious behavior (i.e., goals and values). Host countries often implement integration initiatives including activities aiming to attenuate mental illness, but only a handful are evaluated and reported, with even fewer addressing character development, increases in life satisfaction, or adherence. Our aim was to evaluate the integration initiative “Health for Everyone—Sport, Culture, and Integration”, a 10-week physical activity community project. Specifically, we investigated changes in life satisfaction and character traits (i.e., Self-Directedness, Cooperativeness, and Self-Transcendence) and if these variables, at baseline, predicted adherence and changes in physiological health (i.e., cardiorespiratory fitness, skeletal muscle mass, body fat mass, and visceral fat). Participants (n = 269) answered (pre and post measurements) the Satisfaction with Life Scale, the Short Character Inventory, and undertook physiological tests. In addition, their attendance to the physical activity sessions was registered throughout the project (i.e., adherence). Participants showed no significant increases in Self-Directedness, Cooperativeness, or life satisfaction, but significant decreases in Self-Transcendence. Moreover, higher life satisfaction and lower Self-Transcendence at baseline predicted higher adherence to the activity. However, neither character traits nor life satisfaction predicted changes in physiological health. We argue that low frequency physical activity initiatives may improve this population’s physical health because participants probably have a sedentary life and low levels of physical health due to their asylum conditions (e.g., unemployment, low income, poor housing and social network). Furthermore, physical activity per se may not improve the well-being of asylum seekers. Hence, promoting well-being and character development might require person-centered initiatives focusing on the whole individual in order to fit programmes to the needs and life situation of this population.

Abstract Loss-of-function mutations of the CFTR gene cause the life-shortening genetic disease cystic fibrosis (CF), whereas overactivity of CFTR may lead to secretory diarrhea and polycystic kidney disease. While effective drugs targeting the CFTR protein have been developed for the treatment of CF, little progress has been made for diseases caused by hyper-activated CFTR. Here, we solve the cryo-EM structure of CFTR in complex with CFTRinh-172 (Inh-172), a CFTR gating inhibitor with promising potency and efficacy. We find that Inh-172 binds inside the pore of CFTR, interacting with amino acid residues from transmembrane segments (TMs) 1, 6, 8, 9, and 12 through mostly hydrophobic interactions and a salt bridge. Substitution of these residues lowers the apparent affinity of Inh-172. The inhibitor-bound structure reveals re-orientations of the extracellular segment of TMs 1, 8, and 12, supporting an allosteric modulation mechanism involving post-binding conformational changes. This allosteric inhibitory mechanism readily explains our observations that pig CFTR, which preserves all the amino acid residues involved in Inh-172 binding, exhibits a much-reduced sensitivity to Inh-172 and that the apparent affinity of Inh-172 is altered by the CF drug ivacaftor (i.e., VX-770) which enhances CFTR’s activity through binding to a site also comprising TM8.

Fluorophore-coupled bile acids (BA) represent an important tool for intravital analysis of BA flux in animal models of cholestatic diseases. However, addition of a fluorophore to a BA may alter transport properties. We developed and validated a 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole-coupled taurocholic acid (3β-NBD-TCA) as a probe for intravital analysis of BA homeostasis. We compared transport of 3β-NBD-TCA to [3H]-TCA in HEK293 cells stably expressing the mouse hepatic or renal BA carriers mNtcp or mAsbt, respectively. We also studied distribution kinetics intravitally in livers and kidneys of anesthetized wildtype and mOatp1a/1b cluster knockout mice (OatpKO) with and without administration of the Ntcp inhibitor Myrcludex B and the ASBT inhibitor AS0369. In vitro, 3b-NBD-TCA and [3H]-TCA showed comparable concentration- and time-dependent transport via mNtcp and mAsbt as well as similar inhibition kinetics for Myrcludex B and AS0369. Intravital analysis in the livers of wildtype and OatpKO mice revealed contribution of both mNtcp and mOatp1a/1b in the 3β-NBD-TCA uptake from the sinusoidal blood into hepatocytes. Combined deletion of mOatp1a/1b and inhibition of mNtcp by Myrcludex B blocked the uptake of 3β-NBD-TCA from sinusoidal blood into hepatocytes. This led to an increase of 3β-NBD-TCA signal in the systemic circulation including renal capillaries, followed by strong enrichment in a subpopulation of proximal renal tubular epithelial cells (TEC). The enrichment of 3β-NBD-TCA in TEC was strongly reduced by the systemic ASBT inhibitor AS0369. NBD-coupled TCA has similar transport kinetics as [3H]-TCA and can be used as a tool to study hepatorenal BA transport.

Background: Standard dosages of analgesic and sedative drugs are given to intensive care patients. The resulting range of blood concentrations and corresponding clinical responses need to be better examined. The purpose of this study was to describe daily dosages, measured blood concentrations, and clinical responses in critically ill patients. The purpose was also to contribute to establishing whole blood concentration reference values of the drugs investigated. Methods: A descriptive study of prospectively collected data from 302 admissions to a general intensive care unit (ICU) at a university hospital. Ten drugs (clonidine, fentanyl, morphine, dexmedetomidine, ketamine, ketobemidone, midazolam, paracetamol, propofol, and thiopental) were investigated, and daily dosages recorded. Blood samples were collected twice daily, and drug concentrations were measured. Clinical responses were registered using Richmond agitation-sedation scale (RASS) and Numeric rating scale (NRS). Results: Drug dosages were within recommended dose ranges. Blood concentrations for all 10 drugs showed a wide variation within the cohort, but only 3% were above therapeutic interval where clonidine (57 of 122) and midazolam (38 of 122) dominated. RASS and NRS were not correlated to drug concentrations. Conclusion: Using recommended dose intervals for analgesic and sedative drugs in the ICU setting combined with regular monitoring of clinical responses such as RASS and NRS leads to 97% of concentrations being below the upper limit in the therapeutic interval. This study contributes to whole blood drug concentration reference values regarding these 10 drugs.

The rise of open science and the absence of a global dedicated data repository for molecular dynamics (MD) simulations has led to the accumulation of MD files in generalist data repositories, constituting the dark matter of MD — data that is technically accessible, but neither indexed, curated, or easily searchable. Leveraging an original search strategy, we found and indexed about 250,000 files and 2000 datasets from Zenodo, Figshare and Open Science Framework. With a focus on files produced by the Gromacs MD software, we illustrate the potential offered by the mining of publicly available MD data. We identified systems with specific molecular composition and were able to characterize essential parameters of MD simulation such as temperature and simulation length, and could identify model resolution, such as all-atom and coarse-grain. Based on this analysis, we inferred metadata to propose a search engine prototype to explore the MD data. To continue in this direction, we call on the community to pursue the effort of sharing MD data, and to report and standardize metadata to reuse this valuable matter.

Abstract The NACHT-, leucine-rich-repeat-, and pyrin domain-containing protein 3 (NLRP3) is a critical intracellular inflammasome sensor and an important clinical target against inflammation-driven human diseases. Recent studies have elucidated its transition from a closed cage to an activated disk-like inflammasome, but the intermediate activation mechanism remains elusive. Here we report the cryo-electron microscopy structure of NLRP3, which forms an open octamer and undergoes a ~ 90° hinge rotation at the NACHT domain. Mutations on open octamer’s interfaces reduce IL-1β signaling, highlighting its essential role in NLRP3 activation/inflammasome assembly. The centrosomal NIMA-related kinase 7 (NEK7) disrupts large NLRP3 oligomers and forms NEK7/NLRP3 monomers/dimers which is a critical step preceding the assembly of the disk-like inflammasome. These data demonstrate an oligomeric cooperative activation of NLRP3 and provide insight into its inflammasome assembly mechanism.
