4. Effect of Kinases in Extracellular Vesicles from HIV-1-Infected Cells on Bystander Cells.

Abstract

As of 2023, there were 39.9 million people living with Human Immunodeficiency Virus type 1 (HIV-1). Although great strides have been made in treatment options for HIV-1, and our understanding of the HIV-1 life cycle has vastly improved since the start of this global health crisis, a functional cure remains elusive. One of the main barriers to a cure is latency, which allows the virus to persist despite combined antiretroviral therapy (cART). Recently, we have found that exosomes, which are small, membrane-enclosed particles released by virtually all cell types and known to mediate intercellular communication, caused an increase in RNA Polymerase II loading onto the HIV-1 promoter. This resulted in the production of both short- and long-length viral transcripts in infected cells under cART. This current study examines the effects of exosome-associated kinases on bystander cells. The phospho-kinase profiling of exosomes revealed differences in the kinase payload of exosomes derived from uninfected and HIV-1-infected cells, with CDK10, GSK3β, and MAPK8 having the largest concentration differences. These kinases were shown to be biologically active and capable of phosphorylating substrates, and they modulated changes in the cell cycle dynamics of exposed cells. Given the relevance of such effects for the immune response, our results implicate exosome-associated kinases as new possible key contributors to HIV-1 pathogenesis that affect bystander cells. Additional file 1: Fig. S1. Evaluation of average HUT102 EV sizes via NanoTracking Analysis (NTA). EVs from HUT102 cell supernatants were isolated via differential ultracentrifugation. Peak size of 2 k, 10 k, and 100 k EVs from HUT102 cells was analyzed by NTA. Fig. S2. Evaluation of protein cargo associated to distinct EV populations from ATL-16 cells. EVs from the HTLV-1 infected cell line, ATL-16, were isolated via differential ultracentrifugation at speeds of 2000 × g, 10,000 × g, and 100,000 × g, as described previously for HUT102 cells, resulting in 2 k, 10 k, and 100 k ATL-16 EVs. Proteins were evaluated via Western blot analysis for viral, cellular, and EV markers. Fig. S3. Validation of siRNA treatment on HTVL-1 infected cells. HTLV-1 infected cells (HUT102; Donor cells) were treated with scrambled (lane 1), CD45 (lane 2), and ICAM-1 (lane 3) siRNA to suppress translation of the target transcripts. 48. Generation of 3D Neurospheres for Modeling HIV-1 Infection and Stem Cell EV Analysis

Abstract

Human Immunodeficiency Virus Type 1 (HIV-1) is a single stranded RNA retrovirus that affects 36.9 million people globally. Patients infected with HIV-1 may develop neurological symptoms as a result of central nervous system (CNS) cell damage. To better understand viral impact on the brain, we adapted an in vitro model of the CNS, 3D neurospheres, composed of multiple cell types. Here, we wanted to demonstrate HIV-1 infection of neurospheres, as well as explore the potential of stem cell (iPSC and MSC) EVs to protect neurospheres from cellular damage. Therefore, we infected neurospheres with HIV-1, treated with cART, and probed for HIV-1 marker proteins pr55, p24, and nef. We also verified the presence of astrocytes, microglia, and neurons in the neurosphere after infection through western blotting for cellular markers. To further investigate the potential neuroprotective effects of EVs, we induced cellular damage through irradiation in the neurospheres and observed increased differentiation of astrocytes and microglia and adherence of cells after EV treatment. Lastly, we tested for MMP proteins important for the degradation of the extracellular matrix and found that MMP9 was present in iPSC EVs, suggesting a mechanism by which the EVs may penetrate the neurosphere and enter cells. Overall, we show that neurospheres may be a potential future platform for more comprehensive in vitro CNS HIV-1 research and that stem cell EVs may exert neuroprotective effects on damaged cells., Journal of Student-Scientists' Research, Vol. 1 (2019)

Abstract

The Phoenix Lander, first of NASA's Mars Scout missions, arrived at the Red Planet on May 25, 2008. From the moment the lander separated from its interplanetary cruise stage shortly before entry, the spacecraft could no longer communicate directly with Earth, and was instead entirely dependent on UHF relay communications via an international network of orbiting Mars spacecraft, including NASA's 2001 Mars Odyssey (ODY) and Mars Reconnaissance Orbiter (MRO) spacecraft, as well as ESA's Mars Express (MEX) spacecraft. All three orbiters captured critical event telemetry and/or tracking data during Phoenix Entry, Descent and Landing. During the Phoenix surface mission, ODY and MRO provided command and telemetry services, far surpassing the original data return requirements. The availability of MEX as a backup relay asset enhanced the robustness of the surface relay plan. In addition to telecommunications services, Doppler tracking observables acquired on the UHF link yielded an accurate position for the Phoenix landing site.
