The changes in the plankton biomass structure in relation to nutrient inputs were studied in the Gulf of Venice (Northern Adriatic Sea), an area characterized by a very marked trophic state variability. The investigation was carried out at two stations, in March, May and July 2005 and 2006, considering the whole water column. The size structure (from picoplankton to mesozooplankton) of both autotrophs and heterotrophs was analysed. Signals of diluted waters and nutrient inputs were more marked in 2005 than in 2006. In 2005, the total plankton biomass was almost double (87 ± 37 μg·C·l−1) that in 2006 (44 ± 26 μg·C·l−1). The variations were determined mainly by phytoplankton, with a 70% decrease, and a shift from a community dominated by microphytoplankton (49 ± 12%) in 2005 to one dominated by bacteria (43 ± 11%) in 2006 was observed. The relationship between the heterotrophic (H) and autotrophic (A) biomass indicated a rapid decline of the H/A ratio with increasing phytoplankton biomass. This study, although temporally limited, is consistent with the results reported for other marine environments and it seems to confirm the importance of nutrient inputs in structuring the biomass of plankton community.

Halomonas glaciei isolated from frazil ice in the Ross Sea (Antarctica) during austral summer 2003 was phenotypically characterized and its capability of degrading organic matter was tested. We evaluated specific bacterial growth rates (mu) to understand at which temperatures bacterial growth shows a linear and direct relationship with the available substrate (4-22 degrees C) and afterwards we tested H. glaciei growth curves and degradative potential at 0, 10 and 37 degrees C using two different media (one enriched and one depleted in PO(4)). The strain grew exponentially only at 10 degrees C. The fastest hydrolysis rates were expressed by enzymes aimed at polysaccharide degradation (alpha-D-glucosidase, beta-D-glucosidase and beta-D-galactosidase) while alkaline phosphatase and aminopeptidase activities were rather low. Our data suggest a preferential demand for carbon derived from carbohydrates rather than from proteins: ectoenzyme activities transformed into carbon mobilization from organic polymers, showed that the total carbon potentially released from polysaccharides can be almost one order of magnitude higher than the protein carbon mobilization. Principal component analysis of the enzyme affinity separated the six experimental conditions, highlighting how different physical (temperature) and chemical (PO(4) enrichment or depletion) features actively lead to a differentiation in the efficiency of the ectoenzymes produced, resulting in preferential degradation of diverse kinds of organic substrates.

A 1-year cycle of physico-chemical measurements was conducted in the Marano and Grado Lagoon (Adriatic Sea, Italy) with the aim to assess its trophic state. Some selected variables (nutrients, chlorophyll a , dissolved oxygen, temperature and salinity) were used to describe the system within the application of the Water Framework Directive (WFD) and to compare it with other Mediterranean environments. From an ecological point of view the Marano and Grado Lagoon is one of the best conserved wetlands in the whole Mediterranean area and is protected by several European environmental pieces of legislation. However, the neighbouring mainland hosts several socio-economic activities which can affect its water quality. The results obtained in this work highlighted considerable spatial and temporal variability of physico-chemical parameters. Specifically, a large amount of anthropogenic nutrients, mainly in form of nitrogen (N), was carried from inland through river discharges in the Marano basin (western sector). However, these inputs were offset by the water exchange with the adjacent open sea, which exerted a dynamic dilution effect, and induced low water residence time thus limiting nutrients enrichment and their accumulation for long time periods. The Molar Redfield ratio (DIN/SRP) was always higher than the optimum value of 16 (in some cases it was up to 2000), thus suggesting that the system is phosphorus (P) limited, as confirmed by low chlorophyll a content, rare algal blooms and good water oxygenation. Trophic state indices and threshold values (EEA method, Carlson Trophic Index, TRIX and ASSETS) were applied. A broad range of classes was found (from oligotrophic to hypertrophic depending on water bodies, method applied and period of the year), thus suggesting that an over simplification of this complex ecosystem should be avoided. Further studies will be conducted in order to define the Lagoon’s ecological status sensu WFD and the adequate management implications.

In the summers of 2000, 2001 and 2002, large amounts of sticky mucilaginous material aggregated to form masses of impressive dimensions over large areas of the Adriatic Sea, particularly in its northern part. Aggregates differing in size were sampled by SCUBA divers and submitted to chemical (nutrient and organic matter concentrations) and biological analysis (virus, bacteria and phytoplankton abundances and bacterial metabolism). Suspended and sinking mucilaginous aggregates were biota-rich environments where the abundance of planktonic organisms and the concentration of nutrients were orders of magnitude higher than in the surrounding seawater. The embedded phytoplankton was mostly composed of diatoms, but the dinoflagellate Gonyaulax fragilis, previously reported in association with the aggregates, was also present. A variety of processes occurred within the mucilaginous aggregates which resulted in the transformation of the organic matter composition and lability and contributed to a partial degradation of mucilage. For an efficient degradation of mucilage, several conditions are necessary: high bacterial abundance and activity and an efficient recycling of nutrients within the aggregates. Most of these conditions, appear to change depending on the type and age of the aggregate. During the first phase of aggregation (cobwebs and ribbons), bacterial activities addressed the degradation of organic matter, particularly that of the nitrogen fraction. The degradation products were rapidly taken up by bacteria, supporting an increase in their abundance and production. In aged mucilage (clouds), the degradation processes decreased and the bacterial metabolism suggested the presence of new organic labile compounds probably due to phytoplankton production. On the basis of our results, stringers, generally considered the first step of the aggregation process, seemed to be the result of a mechanical disruption of other types of aggregates.

Spatial and temporal changes in phytoplankton production and bacterial C demand were investigated at four stations in the Northern Adriatic Sea over 3 years. The effect of the Po River plume was observed at the western stations; in particular, the northernmost one (B06) showed the highest values of primary production, both as hourly peaks (up to 14 mg C m− 3 h− 1) and daily water column integrated values (up to 740 mg C m− 2 day− 1), the southern station (C04) was only sporadically influenced and did not differ significantly from the easternmost ones (C12 and B13), where the lowest TPP values were recorded (around 1 mg C m− 3 h− 1). In this study the first in situ data are reported on short-term phytoplankton C extra cellular release in the Northern Adriatic Sea. At every station a considerable percentage of primary production (PER > 20% as an average, with peaks of up to 70%) was released as dissolved organic carbon. In particular, an association of fairly high PER (> 10%) and specific production (Pb > 10 mg C mg chl− 1 h− 1) was observed from spring to summer, when the mucilage phenomenon usually starts. This result might suggest the presence of an uncoupling between photosynthesis and growth, probably related with nutrient availability, which would be responsible for a high production of extra cellular organic carbon. Phytoplankton primary production and bacterial carbon production were closely related and bacterial C production accounted, on average, for a higher percentage of primary production than the values typically reported in the literature on aquatic environments. The flow of organic matter from phytoplankton to bacteria seems to satisfy the bacterial carbon demand during most of the spring and summer, at least in the upper water layers. However, during the summer, there is evidence that BCD sometimes exceeds the amount of C produced by phytoplankton. Neither phytoplankton nor bacterial production showed significant differences over the relevant years, and their absolute values did not change when comparing periods with or without mucilage. However, there were indications of an uncoupling between phytoplankton photosynthesis and growth and of a shift from an autotrophic to a heterotrophic metabolism, especially during the spring and summer period when mucilage might occur.

Little is known concerning environmental factors that may control the distribution of virioplankton on large spatial scales. In previous studies workers reported high viral levels in eutrophic systems and suggested that the trophic state is a possible driving force controlling the spatial distribution of viruses. In order to test this hypothesis, we determined the distribution of viral abundance and bacterial abundance and the virus-to-bacterium ratio in a wide area covering the entire Adriatic basin (Mediterranean Sea). To gather additional information on factors controlling viral distribution on a large scale, functional microbial parameters (exoenzymatic activities, bacterial production and turnover) were related to trophic gradients. At large spatial scales, viral distribution was independent of autotrophic biomass and all other environmental parameters. We concluded that in contrast to what was previously hypothesized, changing trophic conditions do not directly affect virioplankton distribution. Since virus distribution was coupled with bacterial turnover times, our results suggest that viral abundance depends on bacterial activity and on host cell abundance.

Although the temporal and spatial variability of virioplankton in the northernmost part of the Adriatic Sea has been repeatedly explored suggesting that viruses constitute an extremely dynamic component of the plankton community and hypothesizing their importance in marine food webs and mucilage events, there is still no information about viral replication rates. Hence, the contribution of viruses to bacterial mortality and the cycling of organic matter in this part of the Adriatic basin are still not fully comprehended. Assessment of the role of viral lysis requires a robust means of estimating viral production. Since, up to now, none of the available methods evolved to a state of a standard yet, in this preliminary study 3 different experimental approaches were simultaneously assayed (viral production estimated by radiotracer incorporation method [TdR], dilution technique for the estimate of viral production in already infected bacteria [DIL] and serial dilution method in manipulated phage-host assemblage [SER]). The present study provided the first evidence of viral production rates in this study area, that resulted in comprising between ∼ 3.5–15 × 10 8 viruses L − 1 h − 1 and critically faced up the results obtained by different techniques with the consideration that they suffer from different biases. Based on TdR and DIL viral proliferation estimates, viral lysis was responsible for the loss of 54 to 95% of the bacterial standing stock, while the viral-induced mortality by SER (325% d − 1 ) was likely consistently overestimated. These results indicate that viral lysis is a significant factor for prokaryotic mortality suggesting its implication as an important pathway for the cycling of dissolved organic matter in the Gulf of Trieste.

During the 2006 Italian Antarctic expedition a diel sampling was performed close to Cape Hallett (Ross Sea) during the Austral summer. Under-ice seawater samples (∼4 m) were collected every 2 h for 28 h in order to estimate prokaryotic processes' variability and community structure dynamics. Prokaryotic and viral abundances, exoenzymatic activities (β-glucosidase, chitinase, lipase, alkaline phosphatase and leucine aminopeptidase), prokaryotic carbon production ( 3 H -leucine incorporation) and community structure (Denaturing Gradient Gel Electrophoresis – DGGE fingerprints) were analysed. Results showed that the diel variability of the prokaryotic activity followed a variation in salinity, probably as a consequence of the periodical thawing of sea ice (driven by solar radiation and air temperature cycles), while negligible variation in viral and prokaryotic abundances occurred. The Bacterial and Archaeal community structures underwent an Operational Taxonomic Units (OTUs) temporal shift from the beginning to the end of the sampling, while Flavobacteria -specific primers highlighted high variations in this group possibly related to sea ice melting and substrate release.

The prokaryotic community, both in terms of abundance and activity (exo-enzymatic hydrolysis of proteins, polysaccharides and phosphorylated molecules and leucine uptake), was investigated seasonally for a 3-year period (2004-2006) in the Gulf of Venice (northern Adriatic Sea). By focusing on spatial and temporal variability, the prokaryote dynamics showed significant variations on a horizontal and seasonal scale, but no substantial differences were observed among years. The basin-scale variability was mainly influenced by allochthonous inputs from the Po river and the Venice Lagoon, which were the main source of nutrients, acting as a 'bottom up' control on prokaryotes. On a seasonal scale, all the microbial parameters (except the polysaccharide degradation) showed significant variations following the temperature fluctuations. The annual rate of change was very low for all the chemical, physical and biological parameters and only the abundance and phosphatase activity of the prokaryotes differed significantly among years.
