Abstract

Background: Clinical observations suggest that malignant astrocytomas may arise from well-differentiated, low-grade tumors that have undergone anaplastic progression or may develop de novo. Mutations that alter the function of the p53 gene product are thought to play a critical role in astrocytoma tumorigenesis. The authors studied the pattern of mutant p53 expression in astrocytomas to define its role in the formation of malignant tumors by these different pathways. Methods. Tissues from 44 astrocytomas corresponding to Grades I-IV of the World Health Organization (WHO) classification were analyzed for the presence of mutations in exons 5, 7, and 8 of the p53 gene using single strand conformation polymorphism (SSCP) and sequence analysis of DNA amplified by the polymerase chain reaction. Immunostaining for mutant p53 proteins was performed on tissues fixed in formaldehyde solution and embedded in paraffin; the tissues were from these 44 astrocytomas and another 103 astrocytomas obtained from archival material. Results. Tumors with mutant p53 genes were reliably identified by immunostaining for mutant p53 proteins. A higher percentage of astrocytomas of histologic Grades II-IV stained positively for p53 than were identified by mutational analysis. Abstract

A Low Cost Micro-computer based Local Area Network for medical office automation is described which makes use of an array of multiple and different personal computers interconnected by a local area network. Each computer on the network functions as fully potent workstations for data entry and report generation. The network allows each workstation complete access to the entire database. Additionally, designated computers may serve as access ports for remote terminals. Through “Gateways” the network may serve as a front end for a large mainframe, or may interface with another network. The system provides for the medical office environment the expandability and flexibility of a multi-terminal mainframe system at a far lower cost without sacrifice of performance.

36. Dibutyryl Cyclic Adenosine 3',5'-Monophosphate: Its Role in Regulation of Cat Brain Extracellular Fluid

Abstract

• The role of dibutyryl cyclic adenosine 3',5'-monophosphate (db cAMP) in influencing the water content of the cat's gray and white matter was evaluated by intracortical injection of 20 μL of 2 x 10-1 to 2 x 10-6M solution of db cAMP. Cholera toxin, a stimulator of adenylate cyclase, also was tested. Concentrations of db cAMP less than 10-3M failed to produce significant change in brain water content, while concentrations greater than 10-2M produced 31.5% ± 8% and 17.3% ± 3.6% increases in white matter volume, respectively (P <.05). Cholera toxin did not increase brain water levels. These results are discussed in relation to the pathophysiology of brain edema.
