Abstract

Ewing sarcoma (ES) is characterized by EWS::FLI1 or EWS::ERG fusion proteins. Knowing that ion channels are involved in tumorigenesis, this work aimed to study the involvement of the KCNN1gene, which encodes the SK1 potassium channel, in ES development. Bioinformatics analyses from databases were used to study KCNN1expression in patients and cell lines. Molecular approaches and in vitro assays were used to study the transcriptional regulation of KCNN1and its involvement in the regulation of ES cell proliferation. KCNN1is overexpressed in ES patient biopsies, and its expression is inversely correlated with patient survival. EWS::FLI1, like EWS::ERG, promotes KCNN1and SK1 expression, binding to GGAA microsatellites near the promoter of KCNN1isoforms. KCNN1is involved in the regulation of ES cell proliferation, with its silencing being associated with a slowing of the cell cycle, and its expression modulates membrane potential and therefore calcium flux. These results highlight that KCNN1is a direct target of EWS::FLI1 and EWS::ERG and demonstrate that KCNN1is involved in the regulation of intracellular calcium activity and ES cell proliferation, making it a promising therapeutic target in ES.

18. Why Pedagogy Matters: The Importance of Teaching in a Standards-Based Environment

Abstract

The goal of the standards movement has been to improve student outcomes for all children regardless of their backgrounds or risk factors. The focus has primarily been on the instructional, program or performance standards. Paramount importance has been placed on what children will do to demonstrate that they have learned. While important, there is another ingredient in achieving positive student outcomes. What teachers do and how they do it is critically important and has a profound impact on the quality of the educational experience for children. This paper presents the seminal work of the Center For Research On Education, Diversity and Excellence (CREDE), one of the twelve federally funded research centers on education. Its findings, summarized in five critical elements of effective pedagogy, demonstrate that when consistently implemented the result is greater student outcomes across the curriculum regardless of age, and higher academic test scores regardless of the student population. Application in early childhood education settings is also discussed.

This study describes and examines the experiences of displaced manufacturing workers as they lose jobs and retrain in community college settings. The analysis included perceptions of community college services and programming designed to help the workers transition into retraining programs, along with those factors that they identified as contributing to or inhibiting success. Abstract

Technology can facilitate a pedagogy-based on neo-Vygotskian sociocultural theory, and on developmentally appropriate instruction. This article reviews the basic principles of such a pedagogy, and then presents a detailed case study of one day's instruction in a preschool classroom, that is richly infused with a system employing digital photography, computer image manipulation, and color printing. This system situates instruction in the socio-cultural nexus of the students and their families and facilitates the effective pedagogy by the rapid and appealing communication and stimulation provided by technology.

This book is designed to introduce teachers to digital technology and its uses in the classroom as a new teaching tool that can create curriculum, document learning, provide instant feedback, and share student growth with parents. The book's introduction details the ease in which digital technology can be incorporated into the curriculum. Following the introduction, eight chapters are presented which correspond to the curriculum areas of: (1) language; (2) storytelling/drama; (3) emerging literacy; (4) social studies; (5) physical development; (6) mathematics; (7) sensory exploration; and (8) science. Each chapter begins with an introduction to the curriculum area, its importance, how it is learned, and how that learning can be facilitated through the use of digital technology. The chapters present sample lesson plans that list plan objectives, detailed descriptions of the project, and extension opportunities for small groups, independent explorations, one-on-one instruction, and family involvement. Abstract

Various soil health indicators that measure a chemically defined fraction of nitrogen (N) or a process related to N cycling have been proposed to quantify the potential to supply N to crops, a key soil function. We evaluated five N indicators (total soil N, autoclavable citrate extractable N, water‐extractable organic N, potentially mineralizable N, and N‐acetyl‐β‐D‐glucosaminidase activity) at 124 sites with long‐term experiments across North America evaluating a variety of managements. We found that 59%–81% of the variation in N indicators was among sites, with indicator values decreasing with temperature and increasing with precipitation and clay content. The N indicators increased from 6%–39% in response to decreasing tillage, cover cropping, retaining residue, and applying organic sources of nutrients. Overall, increasing the quantity of organic inputs, whether from increased residue retention, cover cropping, or rotations with higher biomass, resulted in higher values of the N indicators. Although N indicators responded to management in similar ways, the analysis cost and availability of testing laboratories is highly variable. Further, given the strong relationships of the N indicators with carbon (C) indicators, measuring soil organic C along with 24‐h potential C mineralization could be used as a proxy for N supply instead of measuring potentially mineralizable N or any other N indicator directly. Nitrogen indicators of soil health vary predictably across North America with climate and soil texture.The response to soil health practices is similar across all indicators.There are strong correlations among nitrogen indicators, but also between carbon and nitrogen indicators.Soil organic carbon and 24‐h carbon mineralization potential can be used to capture nitrogen cycling.

37. Self‐regenerating black medic cover crop provides agronomic benefits at low nitrogen.

Abstract

A black medic (Medicago lupulina L.) cover crop is able to regenerate from seed annually and produce biomass at the end of each growing season, but its long‐term effectiveness on crop productivity, water use efficiency (WUE), and soil nutrient status within a no‐till cropping system is unclear. A field experiment was established in 2003 in Saskatchewan in a 3‐yr crop rotation [flax (Linum usitatissimum L.)–oat (Avena sativa L.)–wheat (Triticum aestivum L.)]. Treatments included cover crop (black medic or no medic), and N fertilizer (20, 60, and 100% of recommended N) arranged in a split‐split plot design. Over 10 yr, medic aboveground fall biomass averaged 625 kg ha–1 (range 0–1,868 kg ha–1) and was greatest at the lowest N rate, 879 kg ha–1. Medic increased grain yield at 20% N fertilizer; no effect was observed at higher N rates. Medic increased tiller density and kernel weight at 20% N, indicating that medic positively influenced the crop throughout the entire life cycle. Medic presence did not affect grain N or P status. Medic did not affect level of available soil N in fall but consistently increased level of soil available P. This extended to the 30–60 cm soil depth in the 100% vs. lower N rates, suggesting medic roots may have influenced P cycling. In oat stubble, medic increased spring soil water and WUE. In conclusion, black medic improved crop productivity at the low N rate but improved available soil P at all N rates, warranting further research. Core Ideas: This is the first North American study to evaluate long‐term effects of medic in a no‐till system.Medic had its greatest effect at 20% vs. 60 and 100% recommended fertilizer N.At the 20% N rate, medic increased grain yield (14%), grain N and P, and WUE.Medic presence increased soil P concentration at the 100% N rate and at deeper soil depths. Abstract

Glioblastoma (GBM) is the most deadly type of malignant brain tumor, despite extensive molecular analyses of GBM cells. In recent years, the tumor microenvironment (TME) has been recognized as an important player and therapeutic target in GBM. However, there is a need for a full and integrated understanding of the different cellular and molecular components involved in the GBM TME and their interactions for the development of more efficient therapies. In this review, we provide a comprehensive report of the GBM TME, which assembles the contributions of physicians and translational researchers working on brain tumor pathology and therapy in France. We propose a holistic view of the subject by delineating the specific features of the GBM TME at the cellular, molecular, and therapeutic levels.
