Forty-five Horsfield’s tortoises ( Testudo horsfieldii; syn. Agrionemys horfieldii, Russian tortoise) belonging to different owners had decreased appetite and respiratory issues. Twenty-nine tortoises had epiphora, dyspnea, and white necrotic diphtheroid oral plaques (group G1). Ten of the remaining 16 tortoises had serious dehydration, appetite disorder, and depression (G2). The last 6 tortoises had only decreased appetite and moderate conjunctival discharge (G3). During the physical examination of all 45 tortoises, a cytologic sample and an oral swab for herpesvirus and Mycoplasma agassizii PCR testing were taken. In 20 of 29 specimens from G1, in 8 of 16 from G2, and 0 of 6 from G3, the cytologic exam revealed intranuclear acidophilic inclusion bodies, multinucleate cellular syncytia, and further abnormalities caused by herpesviral infection. Moreover, all 45 tested subjects were found to be positive for testudinid herpesvirus 1; 2 were positive for M. agassizii. This prospective study suggests that Horsfield’s tortoises with such signs would benefit from this screening procedure, given that it was effective in a significant proportion of infected and symptomatic animals, and no negative effects were seen.

Testicular tumours are the most common neoplasms in male dogs accounting for approximately 90% of all tumours affecting the genitourinary tract. Gray-scale ultrasonography in combination with colour and power Doppler imaging has been well accepted as an accurate technique for assessing scrotal lesions and vascularization of the testis. Colour Doppler sensitivity for low blood flows appears promising in the study of testicular disorders. The aim of this study was to assess if colour and power Doppler ultrasound is a good tool for the investigation of testicular lesions in dogs, to report the sonographic features of lesions and to measure colour and power Doppler parameters such as resistive index (RI), pulsatility index (PI), hypovascularization and hypervascularization (VI) determining if they can be used to distinguish testicular neoplasms from the wide spectrum of non-neoplastic pathological findings. In this study, 50 male dogs of various breeds, aged between 7 and 14 years, presented with testicular disorders were selected. RI and PI were calculated. Mean RI values for neoplastic, inflammatory and degenerative lesions were 0.54, 0.45 and 0.58, respectively. Mean PI values were 0.62, 0.55 and 0.63, respectively. Hypovascularization and hypervascularization of the lesion were evaluated throughout the vascularity index (VI). Vascular signals in neoplasms were significantly intensified around and inside the mass if compared with those measured during inflammatory and degenerative lesions. VI markedly increased in solid tumours. Pathological testes were removed; macroscopical, histological and immunoistochemical evaluations were carried out. Colour Doppler showed increased intralesional and peripheral flows in all neoplastic lesions analysed. No flows were detected around cysts.

In swine production, the economic gain in the farrowing room corresponds to the number of weaned piglets/sow per year. Pre-weaning mortality is at least 11–13% of total losses within one herd, considering a previous 7–8% of the stillbirths. Piglet crushing by the sow contributes significantly to the overall piglet mortality. It is caused by multiple factors related to the sow, the piglet and due to modern swine husbandry along the last 50 years. This study aimed to compare three different designs of farrowing crates during the first three days of life in relation to piglet-crushing mortality in intensive herd. One hundred fifty-eight sows with spontaneous deliveries were housed in the following farrowing crates groups: group A (n = 51) farrowing within conventional crates, group B (n = 47) farrowing within the slide cage and group C (n = 60) in the up and down designed, for a total of 2487 live born piglets. This study aimed to investigate the preservability and viability of the rabbit spermatozoa diluted in a new semen extender Formula® in comparison with Lepus® at 17 °C of storage. The main characteristic of the new extender formulation is the use of an enzymatic agent associated to a polysaccharide as energy source precursor, added with gentamycin. During eight trials, ejaculates from 70 bucks were collected and diluted at 1:10 ratio with both the extenders, after 24 h of storage the semen doses were used for the artificial insemination (AI). Aliquots of the semen doses for each trial were stored at 17 °C, the total and progressive motility were checked at 0, 4, 12, 18, 24, 36, 48, 60, 72, 84, 96, 108 h of storage. Gestational diabetes mellitus (GDM) is a rare disease in dogs that depends from an endogenous insulin supply that is insufficient to meet the tissues demands for the action of diabetogenic molecules like progesterone (P4), estradiol, growth hormone (GH), placental lactogen and placental cytokines. This report describes for the first time a case of GDM complicated by ketoacidosis treated with the P4 receptor blocker aglepristone. In our case aglepristone seemed to increase insulin sensitivity, helping in the resolution of ketoacidosis and of diabetes mellitus. Aglepristone may represent an effective and well tolerated therapeutic option in case of GDM.

A two-years old male mixed breed dog weighing 11 kg was brought to the Veterinary Teaching Hospital by its owner because he noticed a pigmented skin zone with a mass lateral to the penis. The patient was in good clinical condition. On clinical examination, only one testicle was found within the scrotum, and a mass lateral to the right site of the penis was present; upon palpation, the mass was presumed to be a testicle. An ultrasound examination confirmed the presence of an ectopic testicle in that position. The dog had normal fertility and testosterone levels were normal. The patient underwent bilateral orchiectomy and was discharged the following day. The peculiarity of this case is the presence of two independent scrota, an ectopic testicle and its adnexa passing through the abdominal wall, was not supposed to be due a migration along the typical male gonad path during descent.

The physical and gynecological examination of a Holstein heifer single-born with a disorder of sexual development showed anatomical abnormalities such as the presence of a scrotum and 2 vulvae and an anal sphincter that was positioned on the right side of the body. Also, an early pregnancy was diagnosed. Cytogenetic and hormone analysis was requested, and the animal showed normal female metaphases (60,XX) and hormonal profiles. However, in gross anatomy and histological examinations, a structure compatible with a penis, the absence of a uterine body, 2 exophytic structures, and a septum in the vagina were detected.

The royal python (Python regius) is commonly bred in captivity. To have a successful breeding, accurate monitoring of the reproductive activity is necessary. The use of non-invasive monitoring methods in exotics is important in order to minimize stress. For this purpose, ultrasound has been anecdotally used to monitor royal python reproductive activity. However, there is limited information regarding the reproductive cycle of this species. The aim of the present study is to monitor the female reproductive cycle of the royal python using ultrasonography and gonadal steroid metabolite measurements in the faeces. The reproductive activity of one hundred twenty-nine adult female P. regius was examined during two consecutive years. We performed brief scans on non-anaesthetized snakes using a portable ultrasound system and a 10-12 MHz linear array transducer (MyLab™ 30 Gold, Esaote). Ultrasound features, dimension and echogenicity of the reproductive structures were determined. During the second reproductive cycle, the hormonal profiles of 30 animals were also evaluated, with a monthly collection of faecal samples. These samples were classified according to reproductive stage, and the mean faecal progesterone and 17β-estradiol levels were calculated using the results from an enzyme-linked immunosorbent assay (ELISA). Progesterone levels increased during the reproductive cycle. estradiol levels showed greater variability, although they appeared to increase before coupling when compared to the levels between coupling and egg laying. The present study suggests that it is possible to identify different phases in the female royal python reproductive cycle: anovulatory phase, transition, folliculogenesis and embryogenesis. Ultrasound is also useful for identifying follicular regression or slugs. Gonadal steroid metabolite measurements from the faeces could help integrate the reproductive information. The use of ultrasonography in addition with the steroid metabolite measurement in the faeces gives an accurate picture of ovarian activity in captive adult female royal pythons.

Abnormalities of the canine prostate gland, in particular, benign hyperplasia and cysts, are frequent in dogs over 5 years of age. Treatment strategies for dogs with prostatic cysts include ultrasound-guided percutaneous drainage with alcohol sclerotherapy, reduction surgery, and omentalization. The aim of this study was to evaluate the use of platelet-rich plasma (PRP) for the treatment of prostatic cysts in dogs. Ten mixed breed adult male dogs were included in the study. Percutaneous drainage was performed prior to ultrasound-guided PRP injection with a 21G spinal needle. The PRP dose contained half of the solution drained from the cyst. Follow-ups were performed 6, 12, 24, and 60 d after treatment. Ten prostatic cysts were successfully treated with PRP after the ultrasound-guided drainage of the cystic cavities. No side effects were observed during the procedures. After the introduction of PRP, cysts detected by ultrasound appeared hypoechoic. On the sixth day, the hypoechoic appearance was maintained and margins were still identifiable. At 24-day and 60-day follow-ups, cysts were no longer detectable by ultrasound. One of the ten dogs had to be euthanized because of the presence of a malignant mesenchymal neoplasm of the oral cavity (fibroblastic sarcoma) and histological examination of serial sections of the prostate gland was performed. Histologic evaluation 1 year after treatment showed a regression of the cystic lesion injected with PRP. The therapy, which uses PRP injection in combination with ultrasound-guided drainage, can be considered a safe and effective strategy for the treatment of prostatic cysts in dogs.Les anomalies de la prostate chez le chien, plus particulièrement l’hyperplasie bénigne et les kystes, sont fréquentes chez les chiens âgés de 5 ans ou plus. Les stratégies de traitement pour les chiens avec des kystes prostatiques incluent le drainage transcutané par échographie guidée avec sclérothérapie à l’alcool, chirurgie de réduction, et omentalisation. Le but de la présente étude était d’évaluer l’utilisation de plasma enrichi de plaquettes (PEP) pour le traitement de kystes prostatiques chez des chiens. Dix chiens mâles adultes de race mélangée ont été inclus dans l’étude. Le drainage transcutané a été effectué avant l’injection par échographie guidée de PEP avec une aiguille spinale de 21G. La dose de PEP contenait la moitié de la solution drainée du kyste. Des suivis ont été faits à 6, 12, 24, et 60 j suivant le traitement. Dix kystes prostatiques ont été traités avec succès avec le PEP suite au drainage des cavités kystiques par échographie guidée. Aucun effet secondaire ne fut observé durant les procédures. Après l’introduction du PEP, les kystes détestés par échographie apparaissaient hypoéchogène. Au sixième jour, l’apparence hypoéchogène était maintenue et les bordures étaient toujours identifiables. Lors des suivis aux jours 24 et 60, les kystes n’étaient plus détectables par échographie. Un des dix chiens dû être euthanasié à cause de la présence d’un néoplasme mésenchymateux malin de la cavité orale (sarcome fibroblastique) et l’examen histologique de sections sériées de la prostate a été effectué. L’évaluation histologique 1 an après le traitement a montré une régression de la lésion kystique injectée avec du PEP. La thérapie, qui utilise l’injection de PEP en combinaison avec un drainage par échographie guidée, peut être considérée comme une stratégie sécuritaire et efficace pour le traitement de kystes prostatiques chez le chien(Traduit par Docteur Serge Messier).

In the present study the Authors developed a new modified boar semen extender for short-term liquid storage, based on the use of amikacin sulphate and fructose rather than gentamicin and glucose. The new extender (ME-S) was evaluated and compared in vitro to commercial ones (CRONOS ™ , TRIXcell ™ ) and to a modified extender designated for long term storage (ME-L) for progressive motility. Contents In the last decade, progenitor cells isolated from dissociated endometrial tissue have been the subject of many studies in several animal species. Recently, endometrial cells showing characteristics of mesenchymal stem cells (MSC) have been demonstrated in human, pig and cow uterine tissue samples. The aim of this study was the isolation and characterization of stromal cells from the endometrium of healthy bitches, a tissue that after elective surgery is routinely discarded. Multipotent stromal cells could be isolated from all bitches enrolled in the study (n = 7). The multipotency of cells was demonstrated by their capacity to differentiate into adipocytic, osteocytic and chondrocytic lineages. Clonogenicity and cell proliferation ability were also tested. Furthermore, gene expression analysis by RT-PCR was used to compare the expression of a set of genes (CD44, CD29, CD34, CD45, CD90, CD13, CD133, CD73, CD31 CD105, Oct4) with adipose tissue-derived MSC. Stromal cells isolated from uterine endometrium showed similar morphology, ability of subculture and plasticity, and also expressed a panel of genes comparable with adipose tissue-derived MSC. These data suggest that endometrial stromal cells fulfil the basic criteria proposed by the “Mesenchymal and Tissue Stem Cell Committee of the International Society for Cellular Therapy” for the identification of mesenchymal stem cells. Although endometrial mesenchymal stem cells (EnMSC) showed a lower replicative ability in comparison with adipose tissue-derived MSC, they could be considered a cell therapeutic agent alternative to adipose tissue or bone marrow-derived MSC in dog.

Prostate cancer is one of the most important malignancies in men. In old men the frequency of prostate cancer at necroscopy has been reported to exceed 40%. Dogs are the only large mammals other than humans with a significant incidence of spontaneous prostate cancer. Adenocarcinoma, transitional cell carcinoma and undifferentiated carcinoma are the most common histological type but the precise cell of origin in dog is not known. The incidence of prostatic carcinoma in dogs is low (0.2% - 0.6%). Prostatic carcinomas occur in sexually intact and neutered dogs and the risk increase in castrated dogs associated to pulmonary and bone metastases. The castration does not initiate the development of prostatic carcinoma in dog but does favour tumour progression. In men the early stage detection of prostate cancer can offer various therapies as radical prostatectomy, radial therapy, thermal ablation, anti-androgen therapy, chemotherapy. In dogs the diagnosis is often in advanced stage of the cancer and the survival time for dogs with prostate cancer is poor. The median time reported is 30 days after diagnosis. In this study we reported three cases of prostatic carcinoma in intact sexually dogs and one in a neutered dog. The sexually intact subjects were older (mean age = 10.5 years) and they had prostatic adenocarcinoma (PCA). The interval between castration and onset of prostatic problems was 3 years. All the dogs showed dysuria, macroscopic hematuria, dyschezia and ataxia. All dogs have been euthanized in order to relieve pain and suffering.

Standardbred maiden mares are generally stressed and in poor physical condition because of their incorrect management at the end of their racing careers. The purpose of this study was to identify an objective assessment that, similar to body condition score (BCS) determination, is easy to measure and able to confirm or improve fattening status assessment, as well as to ascertain whether a relationship with reproduction efficiency exists in subjects destined for a first-time insemination program. The authors assessed 29 Standardbred maiden mares (7 ± 2 years old) during the breeding season. On January 15 (day 0), the same operator performed the following on all subjects: the first gynecological and ultrasound examination, a BCS assessment (range 0 to 5), and an adiposity objective assessment (i.e., measurement of fat thickness by ultrasound scan). Cystic endometrial hyperplasia-pyometra complex is the most frequent and important endometrial disorder encountered in bitches. The pathogenesis of the disease is related to the activity of progesterone [Feldman and Nelson, Canine and Feline Endocrinology and Reproduction (1996) W.B. Saunders, Philadelphia]. Cystic endometrial hyperplasia (CEH) is an abnormal response of the bitch's uterus to ovarian hormones [De Bosschere et al. Theriogenology (2001) 55, 1509]. CEH is considered by many authors to be an exaggerated response of the uterus to chronic progestational stimulation during the luteal phase of the oestrous cycle, causing an abnormal accumulation of fluid within the endometrial glands and uterine lumen (De Bosschere et al. 2001). The resulting lesions of pyometra are due to the interaction between bacteria and hormones. The aim of this study was to evaluate if transabdominal uterine ultrasonography can be a useful and reliable diagnostic method to confirm Dow's [Veterinary Record (1958) 70, 1102] and De Bosschere's histopathological classification of CEH-pyometra complex. The study was carried out on 45 bitches with pyometra, 10 purebreds and 35 crossbreeds, 1-15 years old, 20% of which had whelped at least once. None of these animals had received exogenous oestrogen or progesterone treatment. On admission the 45 animals were in the luteal phase of the oestrus cycle. Clinical signs, blood parameters, uterine ultrasonography, bacterial swabs and uterine histopathological results were recorded. Results suggest that ultrasonographic examination is a useful and reliable tool for the diagnosis of cystic endometrial hyperplasia.

The rupture of a pregnant uterus is a rare complication of pregnancy, which can result in the death of the mother and the fetus. This life threating condition generally occurs during late pregnancy, it can be asymptomatic and scarred uteruses have been reported as incidental findings during abdominal surgery. This clinical case describes a queen with vulvar bleeding during the early stage of pregnancy. Wall laceration of a pregnant uterus was diagnosed by means of an ultrasound examination. During ovariohysterectomy two uterine ruptures, fragments of the great omentum were found sequestering embryonic tissue. Histopathology showed that the uterine wall was torn in the same place where two embryonic implantations were found and residues of embryonic tissue were observed. This clinical case shows that when a traumatic injury is reported, the mandatory abdomen investigations should evaluate the condition of the uterus and determine the early stage of a pregnancy.

The aims of the study were to identify microbial flora in boar semen under field conditions in northern Italy, to investigate antibiotic resistance and sensitivity of isolated bacteria, and to evaluate elimination of bacteria after storage in two types of extenders added with different antibiotics (amikacin vs gentamicin). A total of 60 boars were collected in 13 pig farms. Bacteriological and mycological investigations were performed immediately on raw semen samples, then at 48 and 120 h of storage on semen diluted randomly in a new short-term modified extender (ME-S) or in a commercial one (CRONOSTM). Bacterial contamination was found in 63% of raw semen samples and different bacterial species were isolated: E.coli, Serratia marcescens, Staphylococcus epidermidis and aureus, Proteus spp., Streptococcus spp. and Pseudomonas aeruginosa. E. coli was the most isolated contaminant (53%); Pseudomonas aeruginosa was found only in one semen sample. Sexual development in mammals is based on a complicated and delicate network of genes and hormones that have to collaborate in a precise manner. The dark side of this pathway is represented by pathological conditions, wherein sexual development does not occur properly either in the XX and the XY background. Among them a conundrum is represented by the XX individuals with at least a partial testis differentiation even in absence of SRY. This particular condition is present in various mammals including the dog. Seven dogs characterized by XX karyotype, absence of SRY gene, and testicular tissue development were analysed by Array-CGH. In two cases the array-CGH analysis detected an interstitial heterozygous duplication of chromosome 9. The duplication contained the SOX9 coding region. In this work we provide for the first time a causative mutation for the XXSR condition in the dog. Moreover this report supports the idea that the dog represents a good animal model for the study of XXSR condition caused by abnormalities in the SOX9 locus.

A five-year-old female cat weighing 3 kg was presented by the owner after noticing a large pink, bilobed mass protruding through the vulva during labour. The cat was in good condition, with appropriate lactation, and the newborn kittens were nursing normally. The uterus was not reverted or invaginated at examination, and there was rupture of the mesovarium, mesometrium and uterine-vaginal connection around the cervix. Manual reduction of the prolapsed uterus was not possible because of torn ligaments. A coeliotomy was performed to remove the ovaries, and the apex of the uterine horns was passed by the vaginal route. The remaining part of the mesometrium was disconnected, and the prolapsed uterus was removed. The queen and kittens were discharged from the hospital on the second day after surgery. An unusual feature of this case is that the prolapse was complete, without eversion of any part of the uterus through a vaginal tear.

In the first days after birth is common to see weight loss in puppies that should not exceed 10% of body weight at birth. The main causes of weight loss are urine and meconium issued followed by non-recovery of fluids expelled. The aims of the study were to check and outline growth curves in boxer breed during the first two week of life. In our study between 3th and 5th day after birth puppies retrieved the weight and got back a gain that leads to double in two weeks. We observed a mean weight loss of 11.26 g (2.27%) between the first two days of life, subsequently they recovered the birth weight on day four. In our study puppies that regained the birth weight earlier were the puppies with higher weight at birth and were the heaviest at day 13.

Contrast-enhanced ultrasound with sulphur hexafluoride microbubbles was performed in seven healthy dogs without a history of reproductive pathology and with histologically confirmed normal testes and in 42 dogs with chronic scrotal anomalies. All dogs underwent orchiectomy and histological examination. Enhancement patterns and perfusion parameters (peak intensity and regional blood flow) of testes of healthy dogs and testes with chronic lesions were compared. Fourteen non-pathologic and 60 pathologic testes were considered. Forty testes were neoplastic (24 interstitial cell tumours, 9 seminomas, 7 Sertoli cell tumours), 20 were non-neoplastic (16 testicular degenerations, 2 chronic orchitis, 1 testicular atrophy, 1 interstitial cell hyperplasia). In healthy dogs, the contrast medium flow had a rapid homogeneous wash-in and wash-out, with a short peak phase. With contrast ultrasound, testes that were inhomogeneous with a hyperenhancing pattern were associated with neoplasia (sensitivity: 87.5%, specificity: 100%). Lesions with persistent inner vessels and a hypo-to-isoechoic background were significantly associated with seminomas (sensitivity: 77.8%, specificity: 100%). Testes with non-neoplastic lesions were characterized by a scant/moderate homogeneous enhancement. Perfusion parameters were higher in neoplastic lesions. Contrast ultrasound was a feasible diagnostic tool in the assessment of testicular lesions, with hyperenhancement being an important feature in the diagnosis of malignancy.

Two operators attempted to detect pregnancy ultrasonographically in 196 sows daily from 15 to 25 days after insemination; 20 unbred sows were also investigated. The probe was applied transcutaneously on the right abdominal wall near the last three mammary glands. During each examination, the embryos were visualised and their transverse and longitudinal dimensions were measured. Pregnancy was confirmed by an ultrasonographic detection of embryos five days after the first ultrasound diagnosis and finally 30 to 32 days after insemination. The accuracy of diagnosis was less than 83 per cent on days 15, 16 and 17 but improved to more than 90 per cent from day 18 onwards. The uterine echotexture was studied in seven sows at oestrus and 15, 17, 19, 21, 23 and 25 days after insemination. The echotexture was more homogeneous from days 15 to 25 after insemination than at oestrus.

Insulin resistance (IR) in dogs is suspected when hyperglycemia is present despite administration of insulin doses greater than 1.0 to 1.5 UI/kg. IR is caused by increases in counter regulatory hormones concentrations (glucagon, glucocorticoids, catecholamines and growth hormone). This study was conducted to investigate the use of aglepristone (RU 46534), a P4 receptor antagonist, for the treatment of IR diabetes mellitus in bitches during the luteal phase. All animals were treated with porcine insulin zinc suspension (Caninsulin) and aglepristone (Alizin) 10 mg/kg subcutaneously at day 1, 2, 9 and 17 from diagnosis. At day 5, no significant variation in glycemia was shown. At day 12 and 20, serum glucose concentrations were significant lower (p < 0.05). From day 12 the insulin dose was reduced to 0.8 IU BID. Insulin was reduced in the following weeks and glycemia was controlled.

A six-year-old intact female Lagotto Romagnolo was referred with a two-day history of purulent vulvar discharge associated with fever, lethargy, polyuria, polydipsia and signs of abdominal pain. Abdominal ultra - sound revealed a grass awn foreign body in the vaginal fornix. Culture swabs obtained from the vagina revealed the presence of Staphylococcus epidermidis as the preponderant organism. Ovariohysterectomy was performed, and the presence of the grass awn was confirmed. A chronic-active vaginitis was found at histological examina - tion. The dog recovered with resolution of all clinical signs. Differential diagnoses for acute vulvar discharge in bitches should include retention of vaginal foreign bodies. To the authors' knowledge, this is the first reported case of a grass awn foreign body in the vaginal fornix of a dog.

Male pseudohermaphroditism is a sex differentiation disorder in which the gonads are testes and the genital ducts are incompletely masculinized. An 8 years old dog with normal male karyotype was referred for examination of external genitalia abnormalities. Adjacent to the vulva subcutaneous undescended testes were observed. The histology of the gonads revealed a Leydig and Sertoli cell neoplasia. The contemporaneous presence of testicular tissue, vulva, male karyotype were compatible with a male pseudohermaphrodite (MPH) condition.
