The objective of this study is to examine the invasiveness of hemostasis by non-thermal plasma (NTP) compared with hemostasis by thermal coagulation (TC). The inflammation recovery process after hemostasis by TC and NTP was compared by using histological methods and nuclear medical molecular imaging. The necrotic areas in the NTP group disappeared after 5 days, whereas they remained 15 days after hemostasis in the TC group. The accumulation of 2-deoxy-2-[F-18] fluoro-D-glucopyranose (F-18-FDG), which reflects the existence of inflammatory cells, was higher in the TC group than in the NTP group on day 15. Thus, this study indicates that hemostasis by NTP is less inflammatory than TC. This report is the first to evaluate inflammation that occurred after hemostasis with medical devices noninvasively.

第26回宇宙利用シンポジウム(2010年1月25日-26日, 宇宙航空研究開発機構宇宙科学研究本部相模原キャンパス), The Twenty-sixth Space Utilization Symposium (January 25-26, 2010: ISAS/JAXA Sagamihara, Japan), An experiment was successfully conducted at'Kibo' module of International Space Station (ISS) from November 14, 2008 to March 28, 2009. In this research project, a focus was made on biological effects of high-radiation background among various factors of space environments. Human lymphoblastoid TK6 cells were frozen and sent to ISS and finally brought back to earth; mainly preserved as frozen in ISS and receiving a total 72 mSv. In addition, we tried to elucidate influences of microgravity on such radiation effects by incubating them at 37C under microG or 1G for 8 days and then refreezing them in ISS. The mutational assays for the non-incubated flight-cells demonstrated the following effects: i) ca.2.3-fold induction of thymidine kinase-deficient (TK(sup -)) mutations over the level of ground-control cell, ii) a suppression (ca.56%) of TK(sup -) mutations, induced by a challenging dose of 2 Gy X-rays and iii) a more efficient repair of DNA double-strand breaks created by I-SceI expression (ca.1.9- and ca.1.8-fold due to non-homologous end joining and homologous recombination, respectively). The characterization (LOH analysis) of the TK(sup -) mutants obtained in the above assays also supports the possibility that frozen cells can record damage accumulated during space travel and subsequently express the genetic and cellular responses when grown on earth. 第25回宇宙利用シンポジウム(2009年1月14日-15日, 宇宙航空研究開発機構宇宙科学研究本部相模原キャンパス), The Twenty-fifth Space Utilization Symposium (January 14-15, 2009: ISAS/JAXA Sagamihara, Japan), This paper is aimed to introduce how our developing methodologies for both loss of heterozygosity (LOH) analysis and DNA double-strand break (DSB) repair estimation are applied to detect the biological effects of space radiation. Our space experiments using human cultured cells in Kibou Facility of International Space Station (ISS) are planned to detect mutagenic effects (i), combined effects of radiation with micro gravity (ii), and radioadaptive response (iii). After cell recovery to the earth, we plan to do the following approaches for the above three biological effects: i) an LOH analysis for the mutants induced after the incubation of recovered frozen-cells exposed to low-dose space radiation during the 3 months preservation in ISS, ii) an examination for the existence of combined effects by comparing the mutation inductions between the cells incubated in ISS for ca. １ week under 1 G and those incubated under micro G, and iii) an estimation of the radioadaptive response from determination of the repair efficiencies of DSB introduced at the specific site of chromosome in the recovered cell., 形態: カラー図版あり, 資料番号: AA0064297067
