Hybrid intravascular near-infrared fluorescence (NIRF)-ultrasound imaging (IVUS) aims to improve the inspection of vascular pathology using fluorescent agents with specificity to biomarkers of atherosclerosis. Two novel intravascular NIRF-IVUS systems with miniaturized imaging catheters were developed and characterized. NIRF-IVUS imaging of preclinical animal models in vivo and human coronary arteries ex vivo demonstrated the detection of atherosclerotic lesions., Hybride Bildgebung aus intravaskulärer Nahinfrarot-Fluoreszenz (NIRF) und Ultraschall (IVUS) bestrebt die Untersuchung von vaskulärer Pathologie mittels Atherosklerose-spezifischen Fluoreszenzfarbstoffen zu verbessern. Zwei neuartige intravaskuläre NIRF-IVUS-Systeme mit miniaturisierten Bildgebungskathetern wurden entwickelt und charakterisiert. Mittels NIRF-IVUS Bildgebung konnten atherosklerotische Läsionen im lebenden Tiermodell und in explantierten humanen Koronararterien detektiert werden.

Photonic microscopy emerged as an indispensable tool for biomedical research by investigating structural, molecular, and functional properties of life on the microscale. However, a restricted wealth of information, toxic labeling, or shallow imaging still impede a comprehensive examination. This thesis concerns the development and application of a novel hybrid optoacoustic and multiphoton microscope that attempts to overcome previous limitations by a unique potential of studying living specimens label-free, at so far inaccessible depth, and via multi-modal contrast., Photonische Mikroskopie entwickelte sich zu einem unverzichtbaren Werkzeug für die strukturelle, molekulare und funktionelle Erforschung von biologischen Proben. Dennoch ist eine umfassende Untersuchung durch eine beschränkte Informationsfülle, toxische Markierungsstrategien und geringe Eindringtiefe limitiert. Diese Arbeit widmet sich der Weiterentwicklung und Anwendung eines neuartigen hybriden Mikroskops, welches durch ein einzigartiges Potential der multimodalen, markierungsfreien und tiefen Bildgebung bisherige Limitierungen überwindet.

Amongst the optical imaging modalities, optoacoustic tomography (OAT) is a promising fast evolving non-invasive imaging method for high resolution mapping of optical absorption in tissues. One of the major difficulties in quantitatively resolving molecular probes in tissue is the effect of light attenuation and inhomogeneous illumination which creates biases. In this work I present a modified version of the recently introduced “interpolated model matrix inversion” scheme for optoacoustic tomography to correct for inhomogeneous surface illumination. Additionally, I show methods which can significantly reduce the biasing effect of light attenuation. The performance of the employed algorithms is presented in simulations, as well as phantom and in an animal studies., Unter den optischen bildgebenden Verfahren ist Optoakustische Tomographie eine vielversprechende sich schnell weiterentwickelnde Modalität, welche nicht invasiv hoch aufgelöste Bilder optisch absorbierender Gewebestrukturen liefert. Für die Quantifizierung molekularer Proben stellen die Fehler, die durch inhomogene Oberflächenbeleuchtung und durch die Lichtabschwächung erzeugt werden ein großes Problem dar. In dieser Arbeit stelle ich eine erweiterte Version des kürzlich veröffentlichten “interpolated model matrix inversion” Verfahrens zur Korrektur von inhomogener Oberflächenbeleuchtung vor. Zusätzlich stelle ich Methoden vor, welche in der Lage sind den Einfluss der Lichtabschwächung signifikant zu verringern. Die Performanz der vorgestellten Algorithmen wird anhand von Simulationen, Phantom- und Tierstudien evaluiert.
