Abstract

The critical current densities and hysteresis losses of bronze-processed Nb3Sn superconducting wires with an Nb-1at%Ta alloy core were measured. The experiment was conducted to determine the impact of adding manganese, nickel, silicon, germanium, boron, zinc and aluminum to the Cu-5at%Sn bronze matrix. The wires design were of the central-copper-stabilizer type and were also tin plated to raise its critical current densities.

Abstract

[Abstract] Phase diagram of binary system CTAB (cetyltrimethylammonium bromide) : OIPh (o-iodophenol) was investigated by DSC, TG methods along with polarizing-microscopic and visual observations. Owing to the large difference in cohesive energy and volatility between the two component substances, the phase behavior is rather complex particularly in the CTAB-rich region. It was found that CTAB and OIPh produce 1: 3 intermolecular complex in addition to the 1: 1 complex already reported. Both of the complexes undergo incongruent melting at about 305 K and 365 K, respectively. Formation of another 2: 1 complex in the high temperature region is proposed. The 2: 1 complex produces solid solution of mesomorphic nature in a limited composition range with the high-temperature metastable phase of CTAB. Thermodynamics of formation of the 1: 1 complex in the crystalline state is discussed.

Abstract

New platinum complexes, (-)-(R)-2-aminomethylpyrrolidine(1,1- cyclobutanedicarboxylato)platinum(II) monohydrate (DWA2114R) and its enantiomeric isomer, (+)-(S)-2-aminomethylpyrrolidine(1,1- cyclobutanedicarboxylato)platinum(II) monohydrate (DWA2114S), were compared in their antitumour effects and nephrotoxicity-inducing activities. Both compounds were effective against the murine tumours L1210 and Colon 26 by i.p. injection of 20-100 mg kg-1. While DWA2114S showed marked increases in blood urea nitrogen (BUN) and urinary protein and sugar in BDF1 mice treated i.p. at the maximum tolerated dose, DWA2114R showed no increases in these parameters. To clarify the difference of nephrotoxicity between the isomers, tissue distribution was examined. Renal Pt concentration in DWA2114S-treated mice was more than 5-fold higher compared with that in DWA2114R-treated mice 2h after i.p. injection of 80 mg kg-1. However, there were no such marked differences in the lung, liver, heart, spleen and plasma. The low content of Pt in the kidneys of DWA2114R-treated mice could explain its lower nephrotoxicity. The in vitro experiments for uptake of the drugs into the cultured normal rat kidney cells and fresh splenocytes revealed that the Pt amount in the cells treated with DWA2114S, especially in the kidney cells, was much higher than DWA2114R.

37. Stimulation of calcium mobilization but not proliferation by bombesin and tachykinin neuropeptides in human small cell lung cancer cells

Abstract

The tachykinin family of neuropeptides, including substance P and neurokinins A and B, induce a transient increase in intracellular free calcium concentration in human small cell lung carcinoma (SCLC) cells, as measured with a calcium indicator fura-2. The effects are dose dependent and even greater than that of bombesin at equimolar concentrations in these cells. The tachykinins, like bombesin, induce calcium mobilization mainly from intracellular store(s). None of the peptides, however, shows a stimulatory effect on DNA synthesis. In addition, exogenously applied bombesin does not stimulate DNA synthesis at any concentration tested. We also examined the effects of a recently reported bombesin antagonist [D-Arg1, D-Phe5, D-Trp7,9, Leu11]substance P in SCLC cells, and compared them to those in Swiss 3T3 fibroblasts in which the mitogenic effect of bombesin is well characterized. The antagonist at 10(-5) M completely abolishes the Ca2+-mobilizing effect of 10(-7) M bombesin in SCLC cells, and that of 10(-9) M but not 10(-7) M bombesin in Swiss 3T3 cells. The antagonist at this concentration effectively inhibits the mitogenic action of bombesin (10(-9) M) in Swiss 3T3 cells; however, much higher doses (approximately 10(-4) M) are needed to inhibit DNA synthesis in SCLC cells. Moreover, the antagonist inhibits DNA synthesis in bombesin/gastrin-releasing peptide-nonproducing cells with a similar dose dependency as in producing cells. These results indicate that bombesin/gastrin-releasing peptide and other calcium mobilizing peptides do not always act as a growth factor in SCLC cells, and that the bombesin antagonist could inhibit growth of SCLC cells through a mechanism other than bombesin antagonism.

学位の種類: 博士（医学）. 報告番号: 乙第1209号. 学位記番号: 新大院博（医）乙第1126号. 学位授与年月日: 平成4年1月22日, 日本大腸肛門病学会雑誌. 1992, 45(2), 138-153., 食餌中の脂肪と繊維が腸内嫌気性菌に及ぼす影響と、大腸菌の発癌過程における経時的腸内嫌気性菌数の変動についてラットを用いて実験的に検討した。ラットは生後8週齢ウイスター系ラット40匹を使用し、I群: 高脂肪高繊維食、II群: 高脂肪無繊維食、III群: 無脂肪高繊維食、IV群: 無脂肪無繊維食を投与した。餌料投与開始5週後より各群を5匹づつDMH投与群 (Ia、IIa、IIIa、IVa) と対照群 (Ib、IIb、IIIb、IVb) に分け、餌料投与開始後4週目 (前期)、11週目 (中期)、24週目 (後期) の便中嫌気性菌の定量を行い、30週目に屠殺剖検し大腸腫瘍の検索を行い以下の結論を得た。繊維には糞便単位重量あたりの嫌気性菌数を減少させる作用が、脂肪には増加作用が認められた。DMH投与群は、IIIa 群以外では後期 (担癌状態) において腸内嫌気性菌数は対照群に比べ低値を示した。大腸発癌の過程において脂肪は initiation 効果を強めることで発癌に促進的に作用し、食物繊維は promotion 効果を弱めることで腫瘍発育に抑制的に作用すると考えられた。, This study was undertaken to examine the effect of fat and fibers contained in diets on intestinal flora and tumor induction in rats. A total of 40 male Wister rats were maintained with one of 4 different diets: Group 1 with high fat and high fiber diet, Group II high fat and fiberless diet, Group III with high fiber and fatless diet and Group IV with diet without fat and fiber. After feeding different diets for 5 weeks, these groups were further subdivided in accordance with the type of carcinogens. 1, 2-dimethyhydrazine (20 mg/kg/week) or EDTA, administered over 10 consecutive weeks. Anaerobic bacteria were evaluated at 4, 11 and 24 weeks after initiation of feeding the diets. All rats were killed and autopsied at 30 feeding weeks. The value of anaerobic bacteria was high in rats fed high fat diets and low in those fed high fiber diets. Compared with control group, anaerobic bacteria indicated a rather low value in cancer-bearing rats except for Group III. High fat diets were accompanied by a high risk for total tumor induction and diets without fat by a low risk, but Group IV without fat and fibers indicated an almost identical cancer volume with Group 1 fed high fat and high fiber diet. These results suggest that dietary fat increase anaerobic bacteria and promotes carcinogenesis, but, on the other hand, dietary fiber decrease anaerobic bacteria resulting in a suppressive effect on cancer growth in rats., 博士（医学）, 新潟大学
