Cowpea (Vigna unguiculata) is a vital crop in sub-Saharan Africa, but the legume pod borer (LPB), Maruca vitrata, can cause over 80% yield losses. Natural resistance to this lepidopteran pest is absent in cowpea germplasm, and insecticides are ineffective due to the pest’s cryptic behavior. To address this, a genetically modified (GM) cowpea expressing the cry1Ab protein from Bacillus thuringiensis (Bt) was developed, providing complete LPB resistance. This Bt cowpea, commercialized as Sampea 20-T in Nigeria, was recently approved in Ghana as Songotra T. To evaluate its performance and the financial returns of its cultivation, field trials were conducted across multiple locations in northern Ghana to compare it to the non-transgenic Songotra control and two commercial cultivars, Kirkhouse-Benga and Wang-Kae. Songotra T exhibited protection against LPB infestations and damage, achieving a grain yield of 2534 kg/ha compared to 1414–1757 kg/ha for the other entries. As expected, non-LPB pest infestations and damage were similar across all entries. Economic analysis revealed that Songotra T had the highest return on investment (464%), outperforming the other tested cultivars (214%). These results demonstrate the potential of GM crops to enhance yields and profitability for resource-poor farmers, underscoring the value of biotechnology for addressing critical agricultural challenges.

Black flies play a prominent role in public health and the epidemiology of parasitic diseases of humans, domesticated and wild animals. Correct identification and a comprehensive survey are required to identify vector and pest species and thus understand their biological attributes which play a vital role in the monitoring program. DNA barcoding is an established molecular tool that provides rapid and accurate species identification. Our study strengthens the molecular database for black flies in Malaysia by adding 59 cytochrome c oxidase I sequences for 22 species, of which 14 are included for the first time. These sequences, combined with those in public databases, represent a total of 338 sequences for 52 Malaysian species, nearly 50% of which were collected from type localities. At the subgeneric level, barcode gap analysis most accurately identified species in the subgenus Nevermannia (92%), followed by Simulium s. l. (91%), and Gomphostilbia (81%). The remaining sequences were ambiguous and could not be distinguished from those of nearest neighbour species due to an overlap in genetic divergence and low genetic diversity, especially between insular species. Tree analyses indicate that certain species had incomplete lineage sorting and low mitochondrial signals. Possible cryptic species were indicated in the Simulium (Gomphostilbia) batoense and S. (G.) epistum species groups. Species delimitations were consistent with morphological identifications except in large species groups such as the S. (G.) asakoae, S. (G.) batoense, S. (G.) epistum, and S. (Simulium) melanopus groups. The use of type specimens or specimens collected from type localities (topotypes) in barcoding is strongly recommended for reference sequences to increase the reliability of the molecular database.

This study employs macrodata on 42 African countries to examine whether remittances and financial development (including its sub-components of access, depth and efficiency) contribute to the equalisation of incomes across the continent. Robust evidence based on the dynamic GMM estimator shows that: (i) remittances heighten income inequality in Africa, (ii) Africa’s financial system is not potent enough for propelling remittances towards the equalisation of incomes, and (iii) vis-à-vis financial access and depth, inefficiencies characterising Africa’s financial institutions is the main reason remittances contribute to the widening of the income disparity gap. Nonetheless, the optimism which we provide by way of threshold analysis shows that channelling resources into the development of Africa’s financial sector could yield shared income distribution dividends. In particular, efforts should be made to achieve a minimum threshold of 23.05 (index) for financial access, and 3.02 (index) for that of financial institutions efficiency if Africa’s financial sector is to repackage remittances towards the equalisation of incomes. A few policy recommendations are provided in the end.

Experimental studies were conducted to evaluate 16 advanced breeding lines of cowpea (Vigna unguiculata (L) Walp) for genetic variability, heritability and correlation between maturity and yield related traits. The genotypes exhibited significant (P < 0.05) differences for the eight traits evaluated viz., number of days to 50 % flowering, number of days to 90 % pod maturity, plant height at maturity, number of pods per plant, number of seeds per pod, pod yield, grain yield and hundred seed weight. SARI-3-11-100, SARI-6-2-6, SARVX-09-004 and IT07K-299-6 had grain yields of 1.99 ± 0.30, 1.88 ± 0.20, 1.95 ± 0.30 and 1.91 ± 0.20 t/ha, respectively, which were significantly higher than the check (Songotra) (1.68 ± 0.01 t/ha). In addition, SARI-5-5-5 matured significantly earlier than the check but no significant difference was observed for grain yield. The higher value of phenotypic component compared to the corresponding genotypic component for all the traits suggest that there was an environmental influence on the performance of the genotypes. Hence, the need for multi-location evaluation of the promising lines for onward release if found stable. The information provided in this study, can be exploited in cowpea breeding program.

Abstract Inherited retinal dystrophies (IRDs) are a clinically and genetically heterogeneous group of Mendelian disorders primarily affecting photoreceptor cells. The same IRD-causing variant may lead to different retinal symptoms, demonstrating pleiotropic phenotype traits influenced by both underlying genetic and environmental factors. In the present study, we identified four unrelated IRD families with the HK1 p.E851K variant, which was previously reported to cause autosomal dominant retinitis pigmentosa (RP), and described their detailed clinical phenotypes. Interestingly, we found that in addition to RP, this particular variant can also cause dominant macular dystrophy and cone-rod dystrophy, which primarily affect cone photoreceptors instead of rods. Our results identified pleiotropic effects for an IRD-causing variant and provide more insights into the involvement of a hexokinase in retinal pathogenesis.

BACKGROUND:Maternal and Child health remains at the core of global health priorities transcending the Millennium Development Goals into the current era of Sustainable Development Goals. Most low and middle-income countries including Ghana are yet to achieve the required levels of reduction in child and maternal mortality. This paper analysed the trends and the associated risk factors of stillbirths in a district hospital located in an impoverished and remote region of Ghana. METHODS:Retrospective hospital maternal records on all deliveries conducted in the Navrongo War Memorial hospital from 2003-2013 were retrieved and analysed. Descriptive and inferential statistics were used to summarise trends in stillbirths while the generalized linear estimation logistic regression is used to determine socio-demographic, maternal and neonatal factors associated with stillbirths. RESULTS:A total of 16,670 deliveries were analysed over the study period. Stillbirth rate was 3.4% of all births. There was an overall decline in stillbirth rate over the study period as stillbirths declined from 4.2% in 2003 to 2.1% in 2013. Female neonates were less likely to be stillborn (Adjusted Odds ratio = 0.62 and 95%CI [0.46, 0.84]; p = 0.002) compared to male neonates; neonates with low birth weight (4.02 [2.92, 5.53]) and extreme low birth weight (18.9 [10.9, 32.4]) were at a higher risk of still birth (p

Background: Anaemia in pregnancy is associated with adverse obstetric outcomes. When detected early in pregnancy, it can be treated; however, information on its prevalence and associated factors is limited in rural Ghana. Aim: The aim of this study was to determine the prevalence and maternal factors associated with anaemia in pregnancy at first antenatal care (ANC) visits. Setting: The study was conducted in the Navrongo War Memorial Hospital, a secondary referral facility in the Kassena-Nankana district in rural northern Ghana. Methods: A retrospective analysis of antenatal clinic records of pregnant women collected from January to December 2014. All pregnant women initiating antenatal clinic, who had initial haemoglobin (Hb) levels measured, were included in the study. Logistic regression analyses were carried out to determine factors associated with anaemia at the initiation of ANC. Results: We analysed data from 506 women with median Hb of 11.1 g/dL (IQR 7.31–13.8). The median gestational age at booking was 14 weeks (5–36 weeks). The prevalence of anaemia was 42.7%, with 95% confidence interval (CI) [38.4–47.1], and was high among teenage mothers (52% [34.9–67.8]), mothers who booked in the third trimester (55% [33.6–74.7]) and grand multiparous women (58% [30.7–81.6]). Factors associated with anaemia included grand multiparity (odds ratio [OR] = 1.94 with 95% CI [1.58–2.46]), booking during the third trimester (OR = 2.06 [1.78–2.21]) and mother who were underweight compared to those with normal weight (OR = 3.17 [1.19–8.32]). Conclusion: Burden of anaemia in pregnancy is still high in rural northern Ghana. We advocate further strengthening of the primary health care system to improve early access to ANC delivery.

Little is known about the humoral immune response against DNA prime-recombinant adenovirus 5 (rAd5) boost HIV vaccine among HIV-infected patients on long-term suppressive antiretroviral therapy (ART). Previous studies emphasized cellular immune responses; however, current research suggests both cellular and humoral responses are likely required for a successful therapeutic vaccine. Thus, we aimed to understand antibody response and function induced by vaccination of ART-treated HIV-1-infected patients with immune recovery. All subjects participated in EraMune 02, an open-label randomized clinical trial of ART intensification followed by a six plasmid DNA prime (envA, envB, envC, gagB, polB, nefB) and rAd5 boost HIV vaccine with matching inserts. Antibody binding levels were determined with a recently developed microarray approach. We also analyzed neutralization efficiency and antibody-dependent cellular cytotoxicity (ADCC). We found that the DNA prime-rAd5 boost vaccine induced a significant cross-clade HIV-specific antibody response, which correlated with antibody neutralization efficiency. However, despite the increase in antibody binding levels, the vaccine did not significantly stimulate neutralization or ADCC responses. This finding was also reflected by a lack of change in total CD4+ cell associated HIV DNA in those who received the vaccine. Our results have important implications for further therapeutic vaccine design and administration, especially in HIV-1 infected patients, as boosting of preexisting antibody responses are unlikely to lead to clearance of latent proviruses in the HIV reservoir.

Thousands of antibodies have been generated and screened in functional neutralization assays, and antibodies associated with cross-strain neutralization and passive protection in primates, have been identified. To facilitate this type of discovery, a high throughput-screening tool is needed to accurately classify mAbs, and their antigen targets. In this study, we analyzed and evaluated a prototype microarray chip comprised of the HIV-1 recombinant proteins gp140, gp120, gp41, and several membrane proximal external region peptides. Vaccines based on Plasmodium falciparum apical membrane antigen 1 (AMA1) have failed due to extensive polymorphism in AMA1. To assess the strain-specificity of antibody responses to malaria infection and AMA1 vaccination, we designed protein and peptide microarrays representing hundreds of unique AMA1 variants. Following clinical malaria episodes, children had short-lived, sequence-independent increases in average whole-protein seroreactivity, as well as strain-specific responses to peptides representing diverse epitopes. Vaccination resulted in dramatically increased seroreactivity to all 263 AMA1 whole-protein variants. High-density peptide analysis revealed that vaccinated children had increases in seroreactivity to four distinct epitopes that exceeded responses to natural infection. A single amino acid change was critical to seroreactivity to peptides in a region of AMA1 associated with strain-specific vaccine efficacy. Antibody measurements using whole antigens may be biased towards conserved, immunodominant epitopes. Peptide microarrays may help to identify immunogenic epitopes, define correlates of vaccine protection, and measure strain-specific vaccine-induced antibodies.

This study examines the contingency effect of infrastructure (disaggregated into physical and digital) in the relationship between foreign aid and inclusive growth in Sub‐Saharan Africa (SSA). We employ macro data for 41 SSA countries and the dynamic system GMM estimator for the empirical analysis. We find that (i) foreign aid promotes inclusive growth in SSA, (ii) whereas transport infrastructure enhances inclusive growth, energy infrastructure, sanitation infrastructure, and digital infrastructure are statistically insignificant, and (iii) only transport infrastructure amplifies the inclusive growth‐enhancing effect of foreign aid. Across the digital and physical infrastructure domains, we find that the contingency effect of the latter is rather remarkable. Our threshold analysis also indicates that for digital infrastructure and transport infrastructure to condition complementary policies to foster inclusive growth in SSA, minimum thresholds of 22% and 57.8% are required. We conclude that comprehensive transport infrastructural development is key if foreign aid is to enhance inclusive growth in SSA. [ABSTRACT FROM AUTHOR]

The second messenger hydrogen peroxide transduces changes in the cellular redox state by reversibly oxidizing protein cysteine residues to sulfenic acid. This signaling event regulates many cellular processes but has never been shown to occur in the brain. Here, we report that hydrogen peroxide heightens endocannabinoid signaling in brain neurons through sulfenylation of cysteines C201 and C208 in monoacylglycerol lipase (MGL), a serine hydrolase that deactivates the endocannabinoid 2-arachidonoyl-sn-glycerol (2-AG) in nerve terminals. The results suggest that MGL sulfenylation may provide a presynaptic control point for 2-AG-mediated endocannabinoid signaling.
