In vitro culture growth rate of Populus alba upon using nano metal-based-oxides such as hematite (Fe2O3 NPs), zinc oxide (ZnO NPs), and manganese oxide (Mn2O3 NPs) nanoparticles as micronutrients.

Gazania (L.) is one of wild ornamental plant which has special architectures. It is a small genus of 16 species, belonging to the family Asteraceae (Formerly: Compositae). It is low-growing perennial herb, grow in clumps and can also form dense carpets covering large areas. Gazania has multiple genera and species with mutli-petals colors. The chlorophyll pigmentation was measured in leaves and petals of these specimens which show high significant difference. Genomics is the study of genomes, and it can be used to conduct research on genome evolution, genomic variations, gene regulation, and important biological mechanisms using detailed genome sequence information. The diversity of ornamental plants, as well as the slow progress of genome research on ornamental plants for a long time. RAPD-PCR was applied in the present study to estimate the genetic variation in these samples which resulted in a total polymorphism percentage of 42.36%. This polymorphism percentage lead to the variation in essential physiological pathways in Gazania spp and reflected on the multivariate petals color. Resumo Gazania (L.) é uma planta ornamental silvestre que possui arquiteturas especiais. É um pequeno gênero de 16 espécies, pertencente à família Asteraceae (Anteriormente: Compositae). É uma erva perene de baixo crescimento, cresce em tufos e também pode formar tapetes densos que cobrem grandes áreas. Gazania tem vários gêneros e espécies com cores de múltiplas pétalas. A pigmentação da clorofila foi medida nas folhas e pétalas desses espécimes que apresentam alta diferença significativa. Genômica é o estudo de genomas e pode ser usada para conduzir pesquisas sobre a evolução do genoma, variações genômicas, regulação gênica e mecanismos biológicos importantes, usando informações detalhadas da sequência do genoma. A diversidade das plantas ornamentais, assim como o lento progresso da pesquisa do genoma de plantas ornamentais há muito tempo. O RAPD-PCR foi aplicado no presente estudo para estimar a variação genética nessas amostras que resultou em uma porcentagem total de polimorfismo de 42,36%. Este percentual de polimorfismo leva à variação nas vias fisiológicas essenciais em Gazania spp e reflete na cor multivariada das pétalas.

The utilization of chitosan nanoparticles is a novel technique for gene transformation into plant tissues. It takes a few minutes to transform gene to plant. UidA gene was detected in Escherichia coli (K12 strain) using polymerase chain reaction analysis by UidA-specific primers. The gene was transformed into the explants of two different plant species (Solanum tuberosum and Paulownia tomentosa). These plants have different natures as crop and woody plants respectively. Therefore, they have different abilities to express the UidA gene. The gene is expressed into blue color in plant tissues due to the formation and expression of the GUS enzyme. The transformation of the UidA gene was detected morphologically by the formation of blue color; and molecular using PCR. Chitosan nanoparticles were characterized by UV/Visible spectroscope and photographing with a transmission electron microscope (TEM). As a result of this research, it is suggested that chitosan nanoparticles be used in gene transformation into plant tissues. Because it is safe, quick, and inexpensive, as well as biocompatible and biodegradable.

A biotic stress due to sodium azidemay inhibit bacterial growth, but also can induce some physiological reactions and some fatty acids production. Escherichia coli is Gram-negative bacteria carries Acetyl-CoA gene which is translated into Acetyl-CoA carboxylase (ACC) “a biotin-dependent enzyme” that catalyzes the irreversible carboxylation of acetyl-CoA to produce malonyl-CoA. This compound is the first step in fatty acid production. Different concentrations of sodium azide mutagen were applied to E. coli to estimate the variation in Acetyl-CoA gene sequence comparing to control. The sequencing results were subjected to TCOFFEE tool in Jalview software to assess the multiple sequence alignment. As a measure for gene variation due to mutation treatment, we estimated the fatty acids produced from the mutant bacterial strains comparing to control. The GC-MS profile of the fatty acids concluded that both control and 250 µg/ml mutant bacteria showed the highest concentrations of some fatty acids, where 125 µg/ml mutant bacteria showed complete inhibition in fatty acid production. Besides, RAPD-PCR molecular marker was performed to estimate the general genetic variation in the mutant E. coli comparing to control. Sodium azide treatment resulted in a total polymorphism percentage of 77.97%. This percent explained the high genetic variation in the total genome content of azide-mutant E. coli comparing to control. The net results from this work that in a certain concentration of sodium azide treatment (250 µg/ml), the production of some essential fatty acids increased.

Fenugreek is a globally important legume that is widely cultivated for its therapeutic benefits in most parts of the world. Seeds on the other hand have a poor germination and growth rate when exposed to salinity. The effect of ultrasonic exposure period on germination and early seedling behaviors of fenugreek seeds under salt stress was investigated in a laboratory experiment. During germination and early seedling stages, all tests were conducted at 40 kHz in a water bath ultrasonic device with two durations (10 and 20 min) under salinity stress using different concentrations of NaCl (0, 1000, 3000, and 5000 mg/l). The results revealed a substantial decrease in germination percentage, all growth criteria, with increasing NaCl concentration and a significant increase in biomass produced by the Fenugreek (total soluble protein, total soluble carbohydrate, and proline), all of which are thought to be mechanisms for salinity resistance. Ultrasonication of fenugreek seeds for 10 and 20 min has a significant impact on seed germination, early seedling development and biochemical constituents under normal and stress conditions. The genetic stability of fenugreek DNA content was affected by these different treatments. This variation was estimated by RAPD-PCR molecular marker, and resulted in a total polymorphism percentage of 49.72% from all the primers. All these different treatments caused variation in the physiological responses and DNA content. This variation enhanced with more ultrasonic and salt treatments. Hence, these stresses can be used for enhancing the variable metabolic processes in fenugreek plant and stimulate its medicinal properties.

Lepidium sativum is an edible medicinal plant from the family Brassicaceae. It holds many uses in folk medicine, and the bioactivities activities originate from its constituents. So, it is essential to estimate the different physiological compounds in this plant. It is also essential to determine which organ in the plant could contain these active compounds. So, it is necessary to evaluate the growth conditions of this plant. In this study, L. sativum is grown in different conditions (with water only in Petri-dish, in pots with peat-moss soil and on jars with Murashige and Skoog (MS) medium with tissue culture technique). From the results obtained, it was clear that cultivation in peat-moss soil supplement the plant with more pigmentation concentration (but not for a long time) and much protein content. In comparison, cultivation on MS media provides much protein content. In all parameters, seeds germinated with water only have the lowest value. Genetically, RAPD-PCR molecular marker illustrates that there is no genetic stability in L. sativum grew on different cultivation conditions. The polymorphism percentage was 54.71%. So, there is genetic variation in response to different cultivation conditions.

Zea mays is an important crop and an essential source of fatty acids. Hence, increasing and adding new fatty acids led to the enhancement of these properties. Transformation of external Acetyl-CoA gene (Aco) can enhance fatty acid components, as ACo is expressed into Acetyl-CoA carboxylase (ACCase) enzyme, which is the first essential step in the fatty acid production process. Chitosan nanoparticles are safe and fast polymer nanoparticles that are applied for gene transformation. Conventional PCR was performed for the detection of the ACo gene in both transgenic and nontransgenic maize lines. The results confirm the presence of the gene in the transgenic lines and absence in non-transgenic lines. The Gas chromatography-mass spectrometry (GC-MS) analysis for fatty acid contents in transgenic and non-transgenic maize lines showed an increase in fatty acid contents in transgenic lines compared to non-transgenic ones. Besides, the transgenic maize’s lines produced extra new fatty acids not found in non-transgenic ones.

Lettuce (Lactuca spp.) is an annual and self-pollinating crop that belongs to the Asteraceae (Compositae) family. It is one of the most globally essential commercial vegetable crops, used in salads and sandwiches. The lettuce leaves are used to make a cigarette without nicotine. Seeds and stems contain edible oil and dried latex. Gene banks have conserved a large pool of lettuce's genetic resources, including wild Lactuca species with the same chromosome numbers 2n = 2x = 18. Lactuca species vary greatly in terms of geographical distribution and morpho-agronomic characteristics. By crossing commercial varieties with locally adapted varieties, novel alleles can be introduced, increasing genetic diversity and making preselection for desirable traits easier. Lettuce breeders and geneticists' main objectives are to improve lettuce for various desirable traits, including tolerance to abiotic and biotic stress and high yield. These targets accomplished with modern genomics tools together with traditional breeding methods. This chapter discusses lettuce conservation and biodiversity, stages of lettuce breeding, agriculture practices, and conventional breeding techniques and their restrictions. It also includes modern plant breeding tools and marker-assisted breeding, editing of genome, and genetic engineering.

The chemical mutagens, namely sodium azide and guanidine hydrochloride (30, 40 and 50 mM) for each were applied for the in vitro culture of Gardenia jasminoides Ellis. Eight morphological traits were evaluated to calculate the result of different concentrations of mutagens compared to control. In sodium azide, all morphological traits decreased compared to control, except the number of branches and leaves and leaf length, which increases compared to control. Unlike guanidine hydrochloride, all the morphological traits are enhanced in mutants rather than control. The polymorphism percentage resulted from genetic variation varied (57.71% in the case of sodium azide and 67.59% in guanidine hydrochloride). Hence, guanidine hydrochloride is a more powerful mutagen and causes more genetic variations and instability than sodium azide, as was reflected in morphological parameters (i.e., fresh weight, shoot length, root length, number of lateral roots, number of leaves, numbers of branches, leaf length, and leaf width) and physiological traits (i.e., chlorophyll pigmentation and carotenoids). Sargassum is a common macroalgae found across the world, especially in the warm temperate regions. It is also known as brown seaweed. Sargassum is known for its medicinal and industrial applications. In the present study Sargassam samples were collected from three different coastal regions of Egypt in the Northern coast, Marsa Matrough and Hurghada. In addition to morphological parameters of identification, the collected species were differentiated and further characterized by Inter Simple Sequence Repeat (ISSR), and 18 S rDNA techniques. This led to the identification of four different species, namely Sargassum subrepandum var. rueppellii, S. platycarpum, S. latifolium, S. subrepandum var. massauense from the areas of collection. Four different ISSR primers used enabled differentiation of total polymorphism to the order of 33.24% among them. The phylogenetic relationship resulted in similarity between Sargassum subrepandum var. rueppellii and S. subrepandum var. massauense.

This work has been focused on the synthesis of the raw materials iron oxide nanoparticles; hematite (α -Fe2O3) from Iron (II) chloride tetra hydrate (FeCl2-4H2O) and iron (III) chloride hexahydrate (FeCl3-6H2O) using Solid State Chemical Reaction technique. Nanostructure powders were characterized by X-ray diffraction (XRD), scanning electron microscope (SEM), energy dispersive X-ray spectroscopy (EDX), and UV-visible spectroscopy. XRD confirmed the formation of crystalline α -Fe2O3 nanostructured andits average crystallite size increased from ~ 12 to 28 nm with increasing annealing temperature from 200 up to 80℃ while SEM confirmed the morphology and the purity of the sample was evaluated from the energy dispersive spectrum (EDS). Moreover, strain decreased with increasing annealing temperature. UV-visible characterization indicated the existence of both direct and indirect band gap in the samples. All The annealed samples showed the direct band gap at ~ 2.21 eV. However, the indirect band gap increased from 1.6 to 1.94 eV when the annealing temperature increased from 200 up to 700℃ and remained almost the same for sample annealed at 800℃. The observed values of optical band gaps were in close agreement with the reported values. Our results indicated that the annealing would give rise to a good crystalline α -Fe2O3 nanoparticles with reduced strain.

Transgenic plants are becoming a more powerful tool in modern biotechnology. Genetic engineering was used in biotech-derived products to create genetically modified (GM) plants. Plant bioreactor systems have proven to be extremely effective in the production of disease resistance plants. The onion (Allium cepa, L.) is a common, important perennial vegetable crop grown in Egypt for food and economic value. Onions are susceptible to a variety of fungal infections and diseases. Aspergillus niger is a common onion phytopathogen that causes diseases such as black mould (or black rot), which is a major issue, particularly when exporting onions. A.niger grows between the bulb's outer (dead, flaky) skin and the first fleshy scales, which become water-soaked. Thionin genes produce thionin proteins, which have antimicrobial properties against a variety of phytopathogens, including A. niger. Chitosan nanoparticles act as a carrier for the thionin gene, which allows A. cepa to resist infection by A. niger. Transgenic A. cepa has a high level of resistance to fungal infection. Transgenic A. cepa had a 27 % weight inhibition compared to non-transgenic one, which had a 69 % inhibition. The expressed thionin protein has a 52 % inhibitory effect on A. niger spore germination. All of these findings supported thionin protein's antifungal activity as an antimicrobial peptide. Furthermore, the data presented here demonstrated the efficacy of chitosan nanoparticles in gene transformation. The present study describes the benefits of producing transgenic onion resistance to black rot diseases.

Paulownia tomentosa (Thunb.) Steud. is a very important hard woody plant, an extremely fast-growing tree and produce timber. Therefore, there is a demand to produce transgenic Paulownia plant resistant to bacterial infection. Microbial infection (especially bacterial one) is serious sever and cause a loss in plant productivity as they bear upon the character and amount of plant product. Two phytopathogenic bacteria were chosen to consider their effect on Paulownia tomentosa. These two bacterial species were Erwinia carotovora and Pseudomonas aeruginosa. Two thionin genes (AT1G12660 and AT1G12663) were selected. They produce antimicrobial peptides to resist this bacterial infection. Chitosan nanoparticle is a novel technology in genetic transformation into plant tissues. Chitosan nanoparticles were used in a ratio of 1:1 with the plasmid DNA carrying thionin genes independently. Characterization for chitosan nanoparticles was applied to determine the conditions of genetic transformation. The new transgenic P. tomentosa lines produced are partially resistant to these two bacterial infections compared to non-transgenic lines. The inhibitory percentage in the transgenic lines ranged from 8 to 21% wherein the non-transgenic the inhibitory percentage of P. tomentosa leaves ranged from 53-24%. Likewise, it is noticed that is Paulownia tomentosa less infectious than Erwinia carotovora. In conclusion, I recommend using chitosan nanoparticle is an excellent way for gene transformation into plant tissues. Also, manipulate the idea of using thionin as antimicrobial genes to resist bacterial infection for different plant species.

Background Lactic acid bacteria (LAB) could be used for bio-production of lactic acid (LA) from wastes of dairy industries. This study aimed to produce LA using isolated and identified LAB capable of withstanding high salt concentration of salted cheese whey and adopting immobilization technique in repeated batch fermentation process. Results Seventy four isolates of LAB were isolated from salted cheese whey and examined for lactic acid production. The superior isolates were biochemically and molecularly identified as Enterococcus faecalis, Enterococcus faecium, and Enterococcus hirae. Then the best of them, Enterococcus faecalis, Enterococcus hirae and dual of them besides Lacticaseibacillus casei were immobilized by sodium alginate 2% in entrapped cells. Repeated batch fermentation was executed for LA production from the mixture of salted whey and whey permeate (1:1) using immobilized strains during static state fermentation under optimum conditions (4% inoculum size in mixture contained 5% sucrose and 0.5% calcium carbonate and incubation at 37 °C). The potent bacterial strain was Enterococcus faecalis which gave the maximum LA production of 36.95 g/l with a yield of 81% after 36 h incubation at 37 °C in presence of 5% sugar. Conclusion Immobilized cells exhibited good mechanical strength during repetitive fermentations and could be used in repetitive batch cultures for more than 126 days.

Soil salinity is a growing threat all over the world due to its toxic effect to reduce soil fertility and water uptake in the crops. An average of 418 million ha soil is saline in nature. Various climatic, geomorphic and rainfall pattern causes which involved in saline soil formation. To reduce the toxic effect proper management of saline soil is required. Irrigation water also a major concern regarding soil salinity management. Saline irrigation water enhances and maintains the severity soil salinity. Crop production aspects root zone salinity provides a strong negative impact on soil fertility. Salinity causes the reduction in nutrient ion, and water uptake has a significant negative effect on crop yields. Soil and water salinity interactions and their influence on crop growth and management of salinity are deliberated in this chapter.

Populus alba is a deciduous large woody tree. The plant was introduced to shooting, then rooting multiplication on MS media. This work's design depends on two factors (media power and sucrose concentrations) with different concentrations resulted in 12 treatments. There were nine responses measured in the new individuals. Some morphological parameters had a significant difference, such as shoot length, leaf and root numbers. Where in the physiological measurements, all the parameters measured had significant differences. RAPD-PCR molecular marker was applied to assess the variation in the new individuals' genetic stability from treatments with the two factors. Four RAPD primers gave reproducible bands and resulted in a total polymorphism percentage of 38.33%. The new individuals resulted from the tissue culture technique were identical to the mother plant and each other. However, in this work, the use of different media power and sucrose concentrations resulted in different values in morphological, physiological responses, and P. alba's molecular variation.

Microalgae have received continued attention as a potential source for biofuel production. However, the lack of suitable strains that provide a lipid-rich biomass and tolerate harsh condition inhibits their industrial application. This report describes an effort to transform Synechocystis sp. with genes encoding acetyl-CoA carboxylase (ACC), a key regulatory enzyme in the lipogenesis pathway, from the white mustard plant (Sinapis alba) and the bacterium Escherichia coli DH5α using chitosan nanoparticles. Although a recombinant plasmid encoding S. alba ACC failed to express, successful transformation was achieved with a recombinant plasmid encoding E. coli DH5α ACC. The successful transformant, Synechocystis sp. PAK13, exhibited increased ACC expression compared with its wild-type parent (11.8 vs. 7.2 ng), which significantly increased its lipid content (by 3.6-fold). Synechocystis sp. PAK13 also exhibited a significant (20%) reduction in photosynthetic pigments, a 1.52-fold higher glucose content and a 3.5-fold lower sucrose content than the wild-type. In conclusion, this report introduces a useful strategy to overexpress the ACC gene in microalgae, creating strains with improved lipid production that are suited to industrial applications.

Organic farming could be an all-encompassing generation administration framework that empowers and improves agroecosystem wellbeing, counting biodiversity, natural cycles, and soil biological activity. It stresses the role of management activities in preference to the use of off-farm data, considering that regional conditions require locally adapted systems. This can be achieved using agronomic, biological, and mechanical methods, in equal share to synthetic materials, to carry out any specific role inside the organization. Organic farming is still only a small industry, which represents only 2% of global food sales. However, it is growing in importance in the world. It is hard to get information due to lack of official statistics and the level of confidentiality of systems of organic produce. Soil practices such as crop rotations, organic fertilizers, symbiotic associations, cover crops, inter-cropping, and minimum tillage are central to organic practices. The static arrangements of soil are achieved by soil fauna and vegetation. Besides, cycling of nutrients and energy is enhanced by increasing the retentive abilities of the soil for nutrients and water.

أصبحت مشکلة البطالة وانخفاض الدخول الحقیقیة من أهم المشاکل الملحة فی الإقتصاد المصری والتی یتعین مواجهتها لما لها من آثار سلبیة علی الإقتصاد والأمان الإجتماعی، إضافة إلی کونها أحد مظاهر أهدار الموارد المتمثلة فی القوی البشریة العاطلة الباحثة عن العمل وعدم توظیفها بما یساهم فی تحقیق نمو أکبر، وعلیه استهدف هذا البحث تحدید دور بعض المشاریع الغذائیة الصغیرة فی زیادة الدخل وتوفیر فرص العمل بمحافظة الفیوم وذلک من خلال عدة أهداف فرعیة وهی دراسة تطور المشاریع الصغیرة الممولة بمحافظة الفیوم، والتعرف علی قدرة بعض المشاریع الغذائیة الصغیرة محل الدراسة فی الحد من مشکلة البطالة وقیاس مستوى دخول العاملین. وقیاس العائد لتلک المشاریع. بالاضافة إلى التقدیر القیاسی لدوال الإنتاج والتکالیف لتحدید مدى کفاءة تلک المشاریع فی تحقیق التنمیة الإقتصادیة بمحافظة الفیوم. والتعرف علی أهم المشکلات والمعوقات التی تواجه تلک المشاریع محل الدراسة ومحاولة إیجاد الحلول والأسالیب اللازمة لحلها. وقد توصل البحث إلى عدة نتائج أهمها:- أن إجمالی عدد المشاریع الممولة وعدد المستفیدین من الذکور وإجمالی القروض بمحافظة الفیوم تتسم بعدم الاستقرار النسبی فی حین یتسم عدد المستفیدین من الإناث وتکلفة فرصة العمل بالاستقرار النسبی نظرا لانخفاض معامل الاختلاف النسبی والبالغ 48.84% ، 24.78% على التوالی. - بلغت قیمة المرونة الإنتاجیة الإجمالیة لمشاریع تصنیع الجبن البیضاء ومنشآت التخلیل نحو 1.28 ، 1.18 على التوالی ویعنی ذلک أن زیادة الموارد الإجمالیة بالدالة المقدرة بنسبة 10% تؤدی إلى زیادة فی إنتاج الجبن البیضاء والمخللات بنسبة 12.8 ، 11.8% على التوالی. وهذه النتیجة توجه بزیادة حجم تلک المشاریع وتوجه بإعادة مزج عناصر الإنتاج المستخدمة فی العملیة الإنتاجیة بما یحقق التولیفة المثلى منها والاستخدام الکفء لها للانتقال إلى مرحلة الإنتاج الاقتصادیة.- ویتضح تفوق حجم الانتاج الأمثل، وحجم الإنتاج المعظم للربح على حجم الإنتاج الراهن لمنشآت تصنیع الجبن البیضاء بمقدار 0.75 ، 2.44 طن على التوالی. کما یتضح تفوق حجم الانتاج الأمثل، وحجم الإنتاج المعظم للربح على حجم الإنتاج الراهن لمنشآت التخلیل بمقدار 5.02 ، 31.53 طن على التوالی.- أن نسبة إجمالی العائد إلى إجمالی التکالیف لمشاریع تصنیع الجبن البیضاء ومنشآت التخلیل حوالی 1.59 ، 1.36 على التوالی، وأن العائد على الجنیه المستثمر بلغ حوالی 0.59 ، 0.36 جنیه على التوالی. وبلغت نسبة التشغیل حوالی 0.63 ، 0.74 على التوالی ویعبر انخفاضها عن الواحد الصحیح على مدى اربحیة المشروع الإنتاجی.- وتبرز مشاریع تصنیع الجبن البیضاء والتخلیل أهمیتها فى استیعاب العمالة، کعامل جوهری ذا أهمیة فی النشاط الزراعی والمجتمع الریفی، حیث یعمل بها حوالى 124 ، 252 فرد على التوالی جمیعهم من ابناء القریة التی توجد بها هذه المنشآت ومن أبرز العوامل الکامنة وراء ارتفاع قدرة هذه الصناعات على استیعاب العمالة هو انخفاض تکلفة فرصة العمل نسبیا بتلک المنشآت حیث قدرة تکلفة إتاحة فرصة العمل بنحو 8.35 ، 16.12 ألف جنیه على التوالی، کما بلغت نسبة متوسط إجمالی الأجور إلى متوسط إجمالی التکالیف المتغیرة بالمشروع نحو 26.6%، 35.4% على التوالی وبذلک تمتاز هذه المشاریع بارتفاع المساهمة النسبیة للأجور فی التکالیف المتغیرة، وبذلک تصبح هذه الصناعات قادرة على امتصاص العمالة الزراعیة الفائضة ، کما أن إقامة مثل هذه المشاریع فی المجتمعات الریفیة یؤدى إلى خلق فرص توظیف من ناحیة، وإلى زیادة دخول العمال الزراعیین من ناحیة أخرى

Aspergillus niger is a dangerous pathogen for many plants. It is a major cause of the destruction, rotting and decomposition of plant tissues. Toxicity caused by A. niger can be inhibited by mutation decreasing the destructive effect on plants. An 18S rDNA molecular tool was used to identify A. niger strains. Sodium azide (NaN3) is a chemical mutagen that disturbs fungal enzymatic activity and causes microbial production of cellulose-degrading enzymes, decreasing mycotoxin production. Different concentrations of sodium azide were used to treat A. niger (30, 40 and 50 µM). The study was designed on two levels: the first level concerned the mutant A. niger’s mode of action: the higher the mutagen concentration, the lower the growth diameter and spore counts. The mutant A. niger’s total proteins and flavonoids were reduced when compared to control. RAPD-PCR showed genetic variation in the genetic content of mutant fungi compared to control resulting in a polymorphism percentage of 78.56%. The second level included the effect of these mutants on two plants (onion and maize). The greater the increase in mutant concentration, the greater the increase in the plants’ morphological and physiological behavior. In conclusion, mutation reduced fungal activity and strengthened plant resistance.

Cavendish bananas ʻGrand Nainʼ are commercially important cultivars. It belongs to the Cavendish subgroup of the AAA banana cultivar group. These cultivars face the problem of quick rotting due to infection with different phytopathogens. Gene transformation technique was used to improve the cultivar productivity and resistance to infection. Thionin genes (AT1G12660 and AT1G12663) were isolated from Arabidopsis thaliana. Thionin genes (thio-60 and thio-63) are antimicrobial genes producing antimicrobial proteins which inhibit fungal infection. These two thionin genes were integrated into the pEGAD vector. Then transformed into the tested banana cultivar via chitosan nanoparticles using the shoot tip culture technique. The chitosan nanoparticles are efficient, rapid and safe transformation technique. The resulted transgenic banana lines were partially resistant to infection by two different fungal species: Fusarium solani and Fusarium equiseti. These fungi cause rotting for the non-transgenic lines compared to the transgenic lines which resist the rotting infection due to the expressed inhibitory thionin protein. Transformation of thionin into the transgenic plants were confirmed by conventional PCR.
