Abstract

The Middle Eastern prevalence of sickle cell anemia, a genetic disorder that affects red blood cells, necessitates additional research. On a molecular level, we sought to identify and sort the oral microbiota of healthy individuals and those with sickle cell anemia. Furthermore, it is crucial to comprehend how changes in the genetic makeup of the oral microbiota impact the state of sickle cell anemia. Using next-generation sequencing, the 16S rRNA amplicon was examined using saliva samples from 36 individuals with sickle cell anemia and healthy individuals. These samples were obtained from sickle cell anemia patients (18 samples) and healthy control participants (controls, 18 samples). Various analyses are conducted using bioinformatic techniques to identify distinct species and their relative abundance. Streptococcus, followed by Fusobacterium nucleatum, Prevotella, and Veillonella were the most prevalent genera of bacteria in the saliva of the SCA and non-SCA individuals according to our findings. Rothia mucilaginosa, Prevotella scoposa, and Veillonella dispar species were the dominant species in both sickle cell anemia and non-sickle cell anemia subjects. Streptococcus salivarius, Actinomyces graevenitzii, Actinomyces odontolyticus, and Actinomyces georgiae spp. were the most prevalent bacterial spp. in the studied SCA cases. The sequencing of the 16S rRNA gene yielded relative abundance values that were visualized through a heatmap analysis. Alterations in the oral microflora's constitution can significantly affect the susceptibility of sickle cell anemia patients to develop more severe health complications. Abstract

Kiwifruit vine decline syndrome (KVDS) is an emerging challenge related to various factors, including water stagnation, soil compaction, root hypoxia, imbalanced soil redox potential, soil nutrient imbalance and fungal pathogens, yet its definitive causes remain unconfirmed. This study compared soils and roots of healthy plants (CTRL group) and plants affected by KVDS (KVDS group) at a kiwifruit orchard (Actinidia chinensis cv. 'Zesy002') in Sermoneta (Lazio region), Italy. Our findings indicate that soil redox potential, a parameter related to soil oxidation, and soil macroporosity were significantly lower in the KVDS soils. The water table was also found to be higher in the KVDS block of the orchard, demonstrating a higher soil water content. This suggests less aeration and more water content in KVDS‐affected soils compared to the control. Root analyses showed that KVDS led to browning and decomposition of rhizodermis, decay of the central stele, detachment of the cortex, reduced cell size and the presence of starch granules in the parenchyma, with significant variations in root morphometric parameters. To mitigate KVDS, sustainable agronomic practices, including precision irrigation, green manure, compost addition and water drainage root pruning, were carried out on some KVDS‐symptomatic vines (SUST group). Molecular analyses revealed the presence of 10 fungal species from treated root plants, with Paraphaeosphaeria michotii, Fusarium oxysporum and Ilyonectria vredenhoekensis being frequently isolated from symptomatic KVDS‐affected plants but less from SUST plants. Also, Beauveria bassiana and Bacillus amyloliquefaciens, showed an effect against I. vredenhoekensis and P. michotii, and may be suitable candidates as biocontrol agents. The adoption of sustainable agronomic practices resulted in significantly more healthy plants (46% increase) by the end of the trial, demonstrating that a sustainable soil use and water management at kiwifruit orchards can mitigate KVDS through restoration of natural growth conditions. Abstract

Essential oils (EOs), extracted from aromatic plants, are interesting natural products and represent an important part of the traditional pharmacopeia. The use of some EOs as alternative antimicrobial and pharmaceutical agents has attracted considerable interest recently. Most of the EOs and their single constituents have been reported to inhibit several phytopathogens, human pathogens, and insects as well as their effective uses in food and pharmaceutical industries. The current review discussed the chemical composition and bioactivity of some important EOs extracted from some Mediterranean plants and their principal bioactive single constituents. Information has been furnished on the mechanisms, mode of actions, and factors affecting the bioactivity of some single constituents from different Mediterranean plant EOs. The current review gives an insight into some common plant EOs belonging to Lamiaceae, Apiaceae, Rutaceae, and Verbenaceae families commonly growing in Mediterranean region. Further information has been provided about the medical uses of some EOs for several human diseases covering the pharmacological effects (anti-inflammatory, antioxidant, and anticarcinogenic). The antimicrobial effects have been also considered in the current review. Although plant EOs are considered promising natural alternatives for many chemical drugs, they still need more specific research for wide application especially in food and pharmaceutical industries.
