Cell lines were established from AV-PR.Cre (n=3) and AV-PRL.Cre (n=5) SCLC tumors and analyzed by deep sequencing of the sgRNA target regions in Trp53, Rb1 and Rbl2. Cancer cells have a characteristic metabolism, mostly caused by alterations in signal transduction networks rather than mutations in metabolic enzymes. For metabolic drugs to be cancer-selective, signaling alterations need to be identified that confer a druggable vulnerability. Here, we demonstrate that many tumor cells with an acquired cancer drug resistance exhibit increased sensitivity to mechanistically distinct inhibitors of cancer metabolism. We demonstrate that this metabolic vulnerability is driven by mTORC1, which promotes resistance to chemotherapy and targeted cancer drugs, but simultaneously suppresses autophagy. We show that autophagy is essential for tumor cells to cope with therapeutic perturbation of metabolism and that mTORC1-mediated suppression of autophagy is required and sufficient for generating a metabolic vulnerability leading to energy crisis and apoptosis. Our study links mTOR-induced cancer drug resistance to autophagy defects as a cause of a metabolic liability and opens a therapeutic window for the treatment of otherwise therapy-refractory tumor patients., mTORC1 is a key mediator of drug resistance and also regulates autophagy. In this study, the authors demonstrate that cancer cells with acquired drug resistance exibit metabolic vulnerabilities mediated by high levels of mTORC1 and the consequent inhibition of autophagy.

Ziel dieser Arbeit war die Aufdeckung eines möglichen Zusammenhangs zwischen dem MDR1-Genotyp und dem Auftreten von Arzneimittelunverträglichkeiten nach der Anwendung von Profender® beim Hund. Solche Arzneimittelunverträglichkeiten wurden unserem Institut in den letzten Jahren im Rahmen der MDR1-Diagnostik häufig gemeldet. Durch eine retrospektive Auswertung dieser gesammelten Fälle ist es gelungen den vermuteten Zusammenhang zum MDR1-Genotyp der betroffenen Tiere aufzudecken und durch weitere in vitro und in vivo Versuche zu bestätigen. Durch Transportversuche an spezifischen Zelllinien konnte eine Interaktion von dem in Profender® enthaltenen Emodepsid mit dem MDR1-Transporter des Hundes belegt werden. Anschließende Versuche an MDR1-defekten Mäusen zeigten deutlich eine vermehrte Gehirnpenetration von Emodepsid beim Fehlen von MDR1 in der Blut-Hirn-Schranke und eine damit verbundene Neurotoxizität in vivo. Besonders auffällig waren in diesem Zusammenhang Störungen der Muskelkoordination, welche sich beim Hund hauptsächlich als Zittern äußerten. Auch Ataxie trat häufig auf. Koordinationsstörungen konnten auch bei der Maus beobachtet und mit Hilfe des gewählten Models (Rotarod) zuverlässig quantifiziert werden. Der direkte Vergleich mit den MDR1-intakten Wildtyp-Tieren, in denen keinerlei Symptome auftraten, belegte einen Zusammenhang zwischen Emodepsid-induzierter Neurotoxizität und dem Fehlen von MDR1 in der Blut-Hirn-Schranke. Die beobachteten Symptome weisen dabei sowohl bei Hunden als auch bei Mäusen auf eine Übererregung des zentralen Nervensystems bzw. eine gestörte Balance zwischen erregenden und hemmenden Strukturen hin, wobei der Toxizitätsmechanismus zum jetzigen Zeitpunkt nicht genau bekannt ist. Wahrscheinlich ist ein komplexes Vergiftungsgeschehen mit Beteiligung mehrerer Neurotransmittersysteme.Obwohl bisher keiner der uns bekannten Fälle letal verlief und alle Symptome auch ohne Behandlung vollständig wieder abklangen, wurde dennoch oft von einem langandauernden und/oder schwerwiegenden Verlauf berichtet. In der tierärztlichen Praxis empfiehlt sich daher vor der Anwendung von Profender® beim Hund ein Test auf das Vorliegen des MDR1-Defekts, um die Applikation bei MDR1-defekten Hunden mit besonderer Sorgfalt durchzuführen. Bestehen dabei Zweifel an der korrekten Umsetzung durch den Besitzer, sollte ggf. auf ein anderes Präparat zurückgegriffen werden., The aim of the present study was to find a possible relationship between the MDR1-genotype and drug intolerances after the administration of Profender® in dogs. Drug intolerances after administration of Profender® in dogs were reported to our institute pretty often in the last few years in the context of MDR1-genotyping. In a retrospective evaluation of those cases we were able to detect the supposed relationship between the MDR1-genotype of the affected dogs, which was confirmed in further in vitro and in vivo experiments. Transport studies in MDR1-expressing cell lines proved an interaction of emodepside (contained in Profender®) and the canine MDR1-transporter. Subsequent studies in MDR1-deficient mice showed a clear increase of emodepside brain penetration and correlated neurotoxicity in the absence of MDR1 in the blood-brain barrier in vivo.This neurotoxicity mainly led to tremor and ataxia in affected dogs. Also the mice showed ataxia, which was reliably quantified with the model used in the present study (rotarod setup). As no symptoms were observed in the corresponding MDR1-intact wildtype mouse strain, an association between the absence of MDR1 in the blood-brain barrier and Emodepside-induced neurotoxicity can be drawn. The observed symptoms in dogs and mice suggest an over-excitation of the central nervous system respectively an imbalance of excitation and inhibition. However the mechanism of toxicity remains unclear. Most likely this mechanism is very complex and involves different neurotransmitter systems. Although none of the drug intolerances reported to our institute was lethal and all symptoms resolved completely without treatment, many cases showed a long lasting and/or severe course. Therefore, MDR1-genotyping is recommended before the use of Profender® in the veterinary practice to perform drug administration with particular care in MDR1-deficient (MDR1-/-) dogs. If a correct drug administration cannot be guaranteed by the owner another type of formulation might be a better choice.
